  Procédé de préparation de diphénylacétamides
 La présente invention a pour objet un procédé de préparation de nouveaux   diphénylacétamides    de formule :

 dans laquelle R est un radical alcoyle inférieur, de préférence de moins de 5 atomes de carbone (par exemple méthyle, éthyle,   propyle,      iisopropyle    et   butyle)    ;   R'est    un radical alcoyle inférieur (par exemple méthyle, éthyle,
   propyle,      butyle et hexyle)    ou   alcènyle    inférieur (par exemple allyle,   méthallyle,      2-butènyle    et   2-hexènyle)    ;

   R"est de l'hydrogène, un halogène (fluor, chlore, brome ou iode), un radical   halométhyle,    qui peut   êtremono-halométhyle    (par exemple chlorométhyle) ou   polyhalométhyle    (par exemple   trifluorométhyle), alcoyle inférieur    (par exemple méthyle et éthyle), alcoxy inférieur (par exemple   méthoxy    et   éthoxy),    hydroxy, nitro, amino cyano, alcanoyloxy inférieur (par exemple   acétoxy)    ou phényle.

   Y est un radical alcoylène inférieur de 2 à 8 atomes de carbone, et peut notamment être un radical éthylène propylène, butylène ou   hexydene,    à chaîne droite ou ramifiée ;   Y'est    un radical alcoylène de moins de 8 atomes de carbone, tel que méthylène, éthylène, propylène, butylène ou   hexylène,    à chaîne droite ou   ramifiée, R"'est    un groupe aromatique, tel que phényle ou phényle substitué par un reste R", méthylèneoxy-R'ou méthylèneoxy-R"ou encore   thiènyle,    furyle ou   pyridyle.    Les composés préférés sont ceux dans lesquels R est éthyle, R" est de l'hydrogène,   R"'est phényle et p-amino-phényle,   
Y est éthylène, Y'est méthylène.



   Les bases libres de formule ci-dessus sont   transfo-    mables en sels   d'addiction    d'acide, dont on peut mentionner les sels d'acides minéraux, tels que les   halogèn-    hydrates par exemple chlorhydrates,   bromhydrates    et iodhydrates), les sulfates, les phosphates et les sels d'acides organiques, tels que citrates, tartrates, oxalates, ascorbates,   méthanesulfonates et succinates.

   (Bien    entendu, on   utilise des acides pharmacologiquement    acceptables lorsque le sol est destiné à l'emploi thérapeutique.)
 Les bases libres de formule ai-dessus peuvent être également transformées en sels quaternaires par traitement de la base avec un ester   alcoylique    inférieur (par exemple chlorure de méthyle, bromure de méthyle, bromure d'éthyle, sulfate de   diméthyle    et nitrate d'éthyle) ou avec des esters aralcoyliques (par exemple chlorure de   benzyle)    et d'autres esters similaires d'acides inorganiques.



   Conformément à l'invention, on prépare les composés de formule ai-dessus en faisant réagir le   diphényl-    acétamide correspondant avec un composé de formule

<tb>  dans   laquelle R"'et Y'sont    tels que définis ci-dessus, et un hydracide   halogéné    anhydre alcoolique et on traite le composé intermédiaire avec un agent hydrogénant, par exemple un borohydrure aloalin, pour obtenir le produit final.



   Le   a-alcoxy-N-alcoyl-N-(alcoylaminoalcoyl)-diphényl-    acétamide peut être préparé par la suite de réactions suivante : on fait réagir un chlorure de benzylalcoylaminoalcoyle avec une alcoylamine ; on fait réagir la N-ben  zyl-N,      N'-dalcoylalcoylènediamne    ainsi obtenue avec un chlorure   d'a-chloro-diphénylacétyle, puis avec    un   alca-    nol inférieur, et on hydrogène   le produit résultant, obtc-    nant ainsi   l'&alpha;-alcoxy-N-alcoyl-N-(alcoylaminoalcoyl)-di-      phénylacétamide    employé pour ta préparation du produit final.



   Il est bien connu que, dans le cas des composés à structure asymétrique, le dédoublement en isomères d et 1 peut donner naissance à d'autres produits pharmaceu  tiquement    actifs. Les composés de formule ai-dessus sont utiles comme analgésiques. Ils peuvent être administrés par voie orale ou intramusculaire de la même manière que le chlorhydrate de   mépélidine,    pour combattre la douleur.



   Dans les exemples qui suivent toutes les températures sont données en degrés centigrades.



  Exemple   1   
Chlorhydrate de 2-Úthoxy-N-{2-[(¯-hydroxyphÚnÚthyl) mÚthyl-amino] Úthyl } -N-mÚthyl-2,2-diphÚnylacÚtamide (a) Préparation de la   N-benzyl-N, N'-diméthyl-   
 Úthyl¯ne-diamine:
 A une solution refroidie de   500 g    de monométhylamine aqueuse à 40%, on ajoute 258 g de chlorhydrate de chlorure de   2-(N-benzyl-N-méthyl-amino)-éthyle.    On agite le mélange pendant une heure à la température ordinaire, puis on le chauffe Ó 80-90¯ pendant 3 heures, on le   refroidmt et    on le traite avec 200 g d'hydroxyde de sodium, ajouté en plusieurs portions. On extrait le mélange 3 fois avec des portions de 600ml d'éther et,   après avoFir réurn    les extraits éthérés, on les sèche sur du carbonate de potassium.

   Après évaporation du solvant, on distille le résidu et on obtient 137 g de   N-benzyl-N,   
N'-dimÚthyl-Úthyl¯nediamine,   bouXant à environ    80  820/0,    5 mm.



  (b) Préparation du chlorhydrate de N-[2-(N'benzyl   N'-methylamino) ethylj-2-ethoxy-N-
 méthyl-2, 2-diphénylacétamide   
 On ajoute goutte à goutte une solution de 175 g de
Famine de la partie (a) dans 500ml de chloroforme à une solution de 261 g de chlorure d'a-chloro-diphénylacétyle dans 1000 ml de chloroforme, en maintenant la   température à 20-250.    On chauffe le mélange à reflux pendant une heure, puis on le traite goutte à goutte avec 500ml d'alcool absolu, tout en distillant 1400 ml de solvant. On traite le résidu avec 1500 ml d'alcool absolu et on le chauffe à reflux pendant 8 heures. On distille la majeure partie du solvant à la pression atmosphérique et le restant   (500 ml)    sous pression réduite.

   Après dilu  tion    avec 1500 ml d'éther, le résidu fournit environ 427 g de chlorhydrate de   N- [2- (benzylméthylaunino) éthyl]-2-    Úthoxy-N-mÚthyl-2, 2-diphÚnyl-acÚtamide, ayant un point de fusion d'environ   159-160 .    Après recristallisation dans un mélange d'acétonitrile et d'éther, le produit purifié fond à environ   162-164 .   



  (c) Préparation du chlorhydrate de 2-Úthoxy-N
 mÚthyl-N-(2-mÚthylaminoÚthyl)
 2,   2-diphenylacetamide   
 On traite une solution chaude de 35, 0 g de chlorhydrate de   N- [2- (benzylméthylamino)-éthyl]-2-éthoxy-N-    mÚthyl-2, 2-diphÚnylacÚtamide de la partie (b) avec une suspension de 3 g de charbon   paHadtié    à   5 ouzo    et on place le tout dans un appareil de Parr sous 3, 86 kg/cm2 d'hy  drogène.    L'hydrogénation est terminée en environ 30 mn.



  Le produit cristallise dans la solution. On chauffe le mélange et on le dilue avec 200 ml d'alcool absolu pour dissoudre le produit, on filtre et on dilue le filtrat   refrodd    avec environ   300 ml d'éther, obtenant    ainsi un solide cristallin pesant environ 28, 1   g ;    p. f. environ 200  201 .    Par   reciwstallisation dans    230 mi d'alcool absolu, on obtient environ 24, 7 g de chlorhydrate de   2-éthoxy-N-      méthyl-N- (2-méthylaminoéthyl)-2, 2-diphényl-acétamide,    p. f. environ   202-2030.   



  (d) PrÚparation du 2-Úthoxy-N-mÚthyl-N-(2-mÚthyl
 amino-Úthyl)-2,2-diphÚnylacÚtamide
   On traite    une   suspension    de 25, 0 g   de chlorhydrate    de   2-éthoxy-N-méthyl-N- (2-méthyl-aminoéthyl)-2, 2-diphé-      nylacétamide,    de la partie (c) dans   200    ml d'eau avec une solution de 3, 0g d'hydroxyde de sodium dans 30ml d'eau. On extrait à l'éther la base libérée et on sèche l'extrait sur du sulfate de magnésium. Après évaporation du solvant, on   obtient environ 19, 5g    de   2-éthoxy-N-      méthyl-N-    (2-mÚthyl-amino-Úthyl)-2,   2-diphénylacétamide,    p. f. environ   45-48 .   



  (e) Préparation du chlorhydrate de 2-Úthoxy-N   méthyl-N-(2-méthylphénacylamino)-éthyl-   
 2,   2-diphénylacétamide   
 On chauffe à reflux un mélange de 12, 0 g de 2-étho   xy-N-méthyl-N- (2-méthylanjino-éthyl)-2, 2-diphénylacé-    tamise de la partie   (d)    et de 5, 7 g   d'a-chloro-acétophé-    none dans   250 nd    de xylène pendant 15 mn et on refroidit le mélange.

   On filtre pour séparer la matière solide formée et on traite le filtrat avec 4 mi de gaz chlorhydrique   alcooli'que SN, obtenant ainsi    8, 2g d'un produit fondant à   175-180 .    On suspend cette matière dans   80 mol    d'eau, on filtre et on sèche, obtenant ainsi 7, 0g de chlorhy  drate de 2-éthoxy-N-méthyl-N- [2- (méthyl-phénacylamino)    Úthyl]-2, 2-diphÚnylacÚtamide, p. f. environ   187-188 .   



  (f) PrÚparation du chlorhydrate de 2-Úthoxy-N-{2
 [(¯-hydroxyphÚnÚthyl)mÚthyl-amino]Úthyl}-N
 mÚthyl-2,2-diphÚnylacÚtamide
 On traite une suspension de 4, 1 g de chlorhydrate   de 2-éthoxy-N-méthyl-N- [2- (méthylphénacylamino) éthyl]-    2,   2-diphénylacEtamide,    de la partie (e), dans 20 ml   d'éthanol à 50  /o    avec une solution de 0, 4 g d'hydroxyde de sodium dans 30 ml d'alcool Ó 95%. Ensuite, on ajoute 0,   4      de barohydrurc    de sodium et on agite lentement le mélange pendant 10 mn. On extrait le   mé-    lange agité 3 fois avec des portions de 100 ml d'éther et on sèche les extraits réunis sur du sulfate de magnésium.

   Ensuite on filtre la solution et on traite le filtrat avec   2 ml    de gaz chlorhydrique alcoolique, obtenant ainsi environ 3,5 g de chlorhydrate de 2-Úthoxy-N-{[(¯-hy-droxyphÚnÚthyl)mÚthylamino]Úthyl}-N-mÚthyl-2,2-diphÚnylacÚtamide, ayant un point de fusion d'environ 162  1640.   



   Le groupe hydroxyle de ce   produit peut etre acylé    de la manière suivante : (a) Chlorhydrate de 2-Úthoxy-N-{2-[(¯-acÚtoxyphÚnÚ
 thyl)mÚthylamino]Úthyl}-N-mÚthyl
 2,2-diphÚnylacÚtamide
 On chauffe à reflux un mélange de   5,    0 g de chlorhy  drate    de   2-éthoxy-N- {2- [ (p-hydroxyphénéthyl)-méthyl-      atmino] éthyl}-N-méthyl-2, 2-diphényl-acétamide préparé    dans l'exemple 1,   30ml d'anhydride acétique    et 10ml de pyridine pendant 30mn.

   Après refroidissement, on   étend le mélange à 300ml avec    de l'éther anhydre, et on obtient le chlorhydrate de 2-6thoxy-N- {2   [      (-acétoxy-      phénéthyl) méthylamino] éthyl}-N-méthyl-2, 2-diphénylacé- tamide cristallin.    



  (b) Chlorhydrate de 2-Úthoxy-N-{2-[(¯-propionyl
 oxyphÚnÚthyl)mÚthylamino]Úthyl}-N-mÚthyl
 2,2-diphÚnylacÚtamide
 On traite un mélange de 4, 8 g de chlorhydrate de 2 Úthoxy-N-{-2[(¯-hydroxyphÚnÚthyl)-mÚthylamino]Úthyl}
N-mÚthyl-2,2-diphÚnylacÚtamide de l'exemple 1 et de 100   nA    de chloroforme avec 1, 0 g de chlorure de   propio-      nyle. Ensuite on chauffe le mélange à    reflux pendant 30 mn, on le refroidit et on le dilue avec 200 ml d'éther, obtenant ainsi 4, 5 g du produit désiré.



  Exemple 2
 Chlorhydrate de   2-ethoxy-N-methyl-N      ? y-       a-phenylpropylamino) ethylJ-2, 2-diphenylacetamide   
 On traite un mélange de   2-éthoxy-N-méthyl-N-(2-    méthylaminoéthyl)-2,   2-diphénylacétamide préparé    dans l'exemple   1, partie (d)    et de   3-chloro-propiophénone    comme décrit dans l'exemple 1, partie (e). Ensuite, on fait rÚagir le produit rÚsultant comme dÚcrit dans l'exemple 1, partie   (f).   



  Exemple 3
 Chlorhydrate de 2-Úthoxy-N-{2-[(¯-hydroxyphÚnÚthyl)
 mÚthylamino]Úthyl}-N-mÚthyl-2,2-bis
 (o-chlorophÚnyl)acÚtamide
 On traite une solution de 175 g de l'amine de   l'exem-    ple 1, partie   (a)    comme décrit dans l'exemple 1, partie (b), sauf que l'on remplace le chlorure   d'a-chloro-diphé-      nylacétyle par 159g de chlotrufe    de   2-chloro-2,    2-bis (o  chloro-phényl)-2-acétyle.    On procède ensuite comme décrit dans l'exemple 1, partie (c), (d),   (e)    et   (f).   




  



  Process for preparing diphenylacetamides
 The present invention relates to a process for the preparation of novel diphenylacetamides of formula:

 in which R is a lower alkyl radical, preferably of less than 5 carbon atoms (eg methyl, ethyl, propyl, isopropyl and butyl); R is a lower alkyl radical (for example methyl, ethyl,
   propyl, butyl and hexyl) or lower alkenyl (eg allyl, methallyl, 2-butenyl and 2-hexenyl);

   R "is hydrogen, halogen (fluorine, chlorine, bromine or iodine), halomethyl radical, which may be mono-halomethyl (eg chloromethyl) or polyhalomethyl (eg trifluoromethyl), lower alkyl (eg methyl and ethyl) ), lower alkoxy (eg methoxy and ethoxy), hydroxy, nitro, amino cyano, lower alkanoyloxy (eg acetoxy) or phenyl.

   Y is a lower alkylene radical of 2 to 8 carbon atoms, and can in particular be an ethylene propylene, butylene or hexydene radical, with a straight or branched chain; Y is an alkylene radical of less than 8 carbon atoms, such as methylene, ethylene, propylene, butylene or hexylene, straight or branched chain, R "'is an aromatic group, such as phenyl or phenyl substituted by a radical R ", methyleneoxy-R ′ or methyleneoxy-R" or also thienyl, furyl or pyridyl. <tb> wherein R "'and Y' are as defined above, and an alcoholic anhydrous halogenated hydracid and the intermediate compound is treated with a hydrogenating agent, for example an aloaline borohydride, to obtain the final product.



   Α-Alkoxy-N-alkyl-N- (alkylaminoalkyl) -diphenylacetamide can be prepared by the following reactions: reacting a benzylalkylaminoalkyl chloride with an alkyllamine; the N-ben zyl-N, N'-dalkylalkylenediamne thus obtained is reacted with an α-chloro-diphenylacetyl chloride, then with a lower alkanol, and the resulting product is hydrogenated, thereby obtaining α -alkoxy-N-alkyl-N- (alkylaminoalkyl) -di- phenylacetamide used in the preparation of the final product.



   It is well known that, in the case of compounds with an asymmetric structure, the resolution into d and 1 isomers can give rise to other pharmaceu tically active products. The compounds of formula α above are useful as analgesics. They can be administered orally or intramuscularly in the same way as mepelidine hydrochloride, to combat pain.



   In the examples which follow all the temperatures are given in degrees centigrade.



  Example 1
2-Úthoxy-N- {2 - [(¯-hydroxyphÚnÚthyl) mÚthyl-amino] Úthyl} -N-mÚthyl-2,2-diphÚnylacÚtamide hydrochloride (a) Preparation of N-benzyl-N, N'-dimethyl-
 Úthylene-diamine:
 To a cooled solution of 500 g of 40% aqueous monomethylamine was added 258 g of 2- (N-benzyl-N-methyl-amino) -ethyl chloride hydrochloride. The mixture was stirred for one hour at room temperature, then heated to 80-90¯ for 3 hours, cooled and treated with 200 g of sodium hydroxide, added in several portions. The mixture was extracted 3 times with 600 ml portions of ether and, after removing the ethereal extracts, dried over potassium carbonate.

   After evaporation of the solvent, the residue is distilled off and 137 g of N-benzyl-N are obtained,
N'-dimÚthyl-Úthyl¯nediamine, bouXant to about 80820/0.5 mm.



  (b) Preparation of N- [2- (N'benzyl N'-methylamino) ethylj-2-ethoxy-N- hydrochloride
 methyl-2, 2-diphenylacetamide
 A solution of 175 g of
Famine part (a) in 500ml of chloroform to a solution of 261g of α-chloro-diphenylacetyl chloride in 1000ml of chloroform, maintaining the temperature at 20-250. The mixture is heated under reflux for one hour, then it is treated dropwise with 500 ml of absolute alcohol, while distilling 1400 ml of solvent. The residue is treated with 1500 ml of absolute alcohol and heated under reflux for 8 hours. Most of the solvent is distilled off at atmospheric pressure and the remainder (500 ml) under reduced pressure.

   After dilution with 1500 ml of ether, the residue gives about 427 g of N- [2- (benzylmethylaunino) ethyl] -2- Úthoxy-N-mÚthyl-2, 2-diphÚnyl-acÚtamide hydrochloride, having a point of fusion of about 159-160. After recrystallization from a mixture of acetonitrile and ether, the purified product melts at about 162-164.



  (c) Preparation of 2-Úthoxy-N hydrochloride
 methyl-N- (2-methylaminoÚthyl)
 2, 2-diphenylacetamide
 A hot solution of 35.0 g of N- [2- (benzylmethylamino) -ethyl] -2-ethoxy-N-mÚthyl-2, 2-diphÚnylacÚtamide from part (b) is treated with a suspension of 3 g of carbon packed with 5 ouzo and the whole is placed in a Parr apparatus under 3.86 kg / cm2 of hydrogen. The hydrogenation is completed in about 30 minutes.



  The product crystallizes in solution. The mixture is heated and diluted with 200 ml of absolute alcohol to dissolve the product, filtered and the refrodd filtrate is diluted with about 300 ml of ether, thus obtaining a crystalline solid weighing about 28.1 g; p. f. about 200,201. By re-installing in 230 ml of absolute alcohol, approximately 24.7 g of 2-ethoxy-N-methyl-N- (2-methylaminoethyl) -2, 2-diphenyl-acetamide hydrochloride is obtained, p. f. approximately 202-2030.



  (d) Preparation of 2-Úthoxy-N-mÚthyl-N- (2-mÚthyl
 amino-Úthyl) -2,2-diphÚnylacÚtamide
   A suspension of 25.0 g of 2-ethoxy-N-methyl-N- (2-methyl-aminoethyl) -2, 2-diphenylacetamide hydrochloride of part (c) in 200 ml of water is treated. with a solution of 3.0g of sodium hydroxide in 30ml of water. The liberated base is extracted with ether and the extract is dried over magnesium sulfate. After evaporation of the solvent, approximately 19.5g of 2-ethoxy-N-methyl-N- (2-mÚthyl-amino-Úthyl) -2, 2-diphenylacetamide, p. f. about 45-48.



  (e) Preparation of 2-Úthoxy-N methyl-N- (2-methylphenacylamino) -ethyl- hydrochloride
 2, 2-diphenylacetamide
 A mixture of 12.0 g of 2-etho xy-N-methyl-N- (2-methylanjino-ethyl) -2, 2-diphenylacean-sieve from part (d) and 5.7 g is refluxed. α-chloro-acetophenone in 250 nd xylene for 15 min and the mixture cooled.

   It is filtered to separate the solid material formed and the filtrate is treated with 4 ml of alcoholic hydrochloric gas SN, thus obtaining 8.2 g of a product melting at 175-180. This material is suspended in 80 mol of water, filtered and dried, thus obtaining 7.0 g of 2-ethoxy-N-methyl-N- [2- (methyl-phenacylamino) Úthyl] -2, 2 hydrochloride. -diphÚnylacÚtamide, p. f. about 187-188.



  (f) Preparation of 2-Úthoxy-N- {2 hydrochloride
 [(¯-hydroxyphÚnÚthyl) mÚthyl-amino] Úthyl} -N
 methyl-2,2-diphÚnylacÚtamide
 A suspension of 4.1 g of 2-ethoxy-N-methyl-N- [2- (methylphenacylamino) ethyl] - 2, 2-diphenylacEtamide hydrochloride, from part (e), is treated in 20 ml of ethanol at 50% with a solution of 0.4 g of sodium hydroxide in 30 ml of 95% alcohol. Then 0.4 of sodium barohydride is added and the mixture is stirred slowly for 10 min. The stirred mixture was extracted 3 times with 100 ml portions of ether and the combined extracts dried over magnesium sulfate.

 2,2-diphÚnylacÚtamide
 A mixture of 4.8 g of 2 Úthoxy-N - {- 2 [(¯-hydroxyphÚnÚthyl) -mÚthylamino] Úthyl} hydrochloride is treated.
