Verfahren zur Herstellung von substituierten Diphenylalkenylaminen
Die Erfindung bezieht sich auf ein Verfahren zur Herstellung von substituierten Diphenylalkenylaminen.
Die eRindungsgemäss erhältlichen DiphenylaLkenyl- amine haben die allgemeine Formel I des Formelblat- tes, in der A eine gerade oder verzweigte Alkylengruppe mit höchstens 6 Kohlenstoffatomen, B eine Alkylamino-oder Dialkylaminogruppe oder eine gesät- tigte einkernige heterocyclische Gruppe mit einem Fünf-oder Sechsring, die an der Kohlenwasserstoffkette A mittels eines im-Heteroring befindlichen Stickstoffatomes gebunden ist, und R ein Wasserstoffatom oder eine Alkylgruppe darstellt. Die Verbindungen können in ihre nicht-toxische Salze übergeführt werden. Beispiele von heterocyclischen Gruppen, die durch das Symbol B dargestellt werden können, sind 1-Pyrrolidyl, 1-Piperazinyl, Piperidino, Morpholino und Thiomorpholino.
Die Diphenylalkenylamine nach Formel I haben therapeutische Bedeutung insbesondere als Analgeticum. Es werden Verbindungen bevorzugt, in denen A die Alkylenkette

und B eine Dialkylaminogruppe und R Wasserstoff oder eine Methylgruppe darstellt, während die Trifluormethylgruppe sich in Ortho-oder Parastellung (vorzugsweise in letzterer) am Phenylring zu der Ami noalkenylgruppe befindet. Wertvolle Verbindungen gemäss der Erfindung sind N. N-Dimethyl-4-phenyl4-(p-trifluor-methylphenyl)but-3-enylamin, N.
N-Dimeth yl-4-phenyl-$ (o-trif Luormethylphenyl) but-3-enylamin, und N. N-Dimethyl¯2-methyl-3-(o-trifLuormethylphenyl)- prop-2-enylamin,, sowie ihre nichttoxischen Salze. Besonders gute Eigenschaften zeigt die erstgenannte Verbin- dung.
Für therapeutische. Zwecke lassen sich die Diphenylalkenylamine gemäss Formel I als solche oder in Form ihrer nicht-toxischen Salze benutzen, d. h. Salze, welche dem tierischen Organismus nicht schädlich sind, wenn sie in therapeutischen Dosen verabreicht werden, wie die Hydrohalogenide, Oxalate, Tartrate, Fumarate, Zitrate und Acetate. Diese Salze können aus den freien Basen nach an sich bekannten Methoden hergestellt werden, z. B., indem äquivalente Mengen Base und Säure in einem inerten organischen Lösungsmittel gemischt werden und das Lösungsmittel anschliessend verdampft wird, oder im verwendeten Medium nicht oder schwer lösliche Salze filtriert werden.
Die Diphenylalkenylamine nach Formel I werden gemäss der Erfindung durch Dehydratation einer Verbindung der Formel II erhalten. Die Dehydratation lässt sich z. B. mit Schwefelsäure oder einer aromatischen Sulfonsäure, z. B. p. Toluolsulfonsäure durchfüh- ren.
Verbindungen der Formel II sind z. B. erhältlich, indem man auf ein lentsprechendes Keton ein OrganoW maignasiumhalogenid einwirken lässt und den erhaltenen Komplex hydrolysiert.
Beispiel 1
Ein Gemisch aus 6, 75 g N. N-Dimethyl-4-phenyl 4-hydroxy-4-(o-trifluormethylphenyl) butylamin und 20 cm3 20"/piger Schwefelsäure erhitzt man eine Stunde zum Sieden am Rückfluss. Das Gemisch wird alkalisch gemacht und mit Diäthyläther extrahiert. Die ätherische Lösung wird über Natriumsulfat getrocknet.
Nachdem die festen Stoffe abfiltriert sind, wird eine ätherische Salzsäurelösung zu dem Filtrat gegeben. Das gefällte Hydrochlorid wird abfiltriert und aus einem Athanol/Athergemisch umkristallisiert. Dabei werden 6 g N. N-Dimethyl-4-phenyl-4-(o-trifluormethylphenyl)- but-3-enylaminhydrochlorid, Schmelzpunkt 154-156 erhalten. Der Schmelzpunkt steigt bei weiterem Umkristallisieren zu 155-157 C an.

Beispiel 2
Man wendet das Verfahren nach Beispiel 1 an, geht jedoch von 3, 37 g N. N-Dimethyl-4-hydroxy 4-phenyl-4-(m-trifluormethylphenyl) butylamin aus.
Man erhält dann 2, 45 g des N. N-Dimethyl-4-phenyl 4-(m-trifluormethylphenyl) but-3-enylamins, Schmelzpunkt 139-140 C.

Beispiel 3
Aus N. N-Dimethyl-4-hydroxy-4-phenyl-4- (p-tri- fluormethylphenyl) butylamin erhält man nach dem Verfahren gemäss Beispiel 1 2, 05 g des N. N-Dimethyl 4-phenyl-4- (p-trifluormethylphenyl) but-3-enylaminoxalats. Schmelzpunkt 177-177, 5¯ C.

Beispiel 4
Aus 3, 0 g N. N-Dimethyl-3-hydroxy-3-phenyl-3- (m- trifluormethylpheny1) propylaminhydrochlorid werden durch Anwendung des Verfahrens nach Beispiel 1 2, 5 g des N. N-Dimethyl-3-phenyl-3-(m-trifluormethyl- phenyl) prop-2-enylaminoxalats erhalten. Der Schmelzpunkt war nach Umkristallisieren aus einem Äthanol/ Athergemisch 161-163 C.

Aus 6, 74 g N. N-Dimethyl-2-methyl-3-hydroxy 3-phenyl-3-(o-trifluormethylphenyl) propylamin erhielt man durch Anwendung des Verfahrens nach Beispiel 1 6, 5 g des N. N-Dimethyl-2-methyl-3-phenyl-3-(o-trifluormethylphenyl) prop-2-enylaminoxalats. Der Schmelzpunkt war nach Umkristallisieren aus Aceton 129-130, 5 C.

Aus 7, 59 g 1-Morpholino-2-methyl-3-hydroxy3-phenyl-3-(o-trifluormethylphenyl) propan wurden durch Anwendung des Verfahrens nach Beispiel 1 6, 0 g des l-Morpholino-2-methyl-3-phenyl-3-(o-tri fluormethylphenyl) prop-2-enoxalats erhalten. Der Schmelzpunkt war nach Umkristallisieren aus Aceton 178-179, 5 C.

Aus 7, 6 g 1-Morpholino-2-methyl-3-hydroxy 3-phenyl-3- (m-trifluormethylphenyl) propan wurden durch Anwendung des Verfahrens nach Beispiel 1 4, 7 g des 1-Morpholino-2-methyl-3-phenyl-3- (m-tri- fluormethylphenyl) prop-2-enoxalats erhalten. Der Schmelzpunkt war nach Umkristallisieren aus einem Athanol/Athergemisch 182-184 C.

Beispiel 8
Aus N-Methyl-4-hydroxy-4-phenyl-4-(o-trifluorme- thylphenyl) butylamin wurde durch Anwendung des Verfahrens nach Beispiel 1 das N-methyl-4-phenyl- 4- (o-tdfluormethylphenyl) but-3-enylaminoxalat erhalten.
Pharmazeutische Präparate, die einen oder mehrere der therapeutischen Wirkstoffe nach Formel I oder ihre nicht-toxischen Salze zusammen mit einem pharmakologisch zulässigen Träger enthalten, können z. B. in Form von Tabletten (einschl. Tabletten mit verzö- gerter Wirkung), Kapseln, injizierbaren L¯sungen, Pastillen oder Zerstäubungspräparaten zum Inhalieren hergestellt werden. Die gemeinten Präparate können in bekannter Weise hergestellt werden, erforderlichenfalls unter Anwendung auf diesem Gebiete bekannter Ver dünnungsmittel. Die Menge des Wirkstoffes in dem pharmazeutischen Präparat kann variiert werden, ist jedoch so zu wÏhlen, dass dem Patienten täglich 25-200 mg verabreicht werden können ohne dass ihm das Beschwerden macht.
Bims solche Menge eignet sich im allgemeinen für klinische Zwecke und wird vorzugsweise in Form von Tabletten verabreicht, welche 100 mg des Wirkstoffes enthalten.
