Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung einer neuen Gruppe von substituierten Propionaniliden mit lokalanästhetischer Wirkung.
Die erfindungsgemäss hergestellten substituierten Propionanilide werden durch die folgende allgemeine Formel wiedergegeben :

worin R niederes Alkyl oder Cyclohexyl und Y Phenyl, o-Halogenphenyl, o-Tolyl oder 2, 6-Xylyl bedeuten.
Es wurde gefunden, dass diese substituierten Propionanilide als Lokalanästhetika brauchbar sind. Im allgemeinen sind die Säureadditionssalze, wie z. B. das Hydrochlorid, Hydrobromid, Sulfat, Phosphat, Maleat und Citrat, in denjenigen Fällen besonders brauchbar, in denen Lösungen der Verbindungen verwendet werden sollen.
Beispielsweise sind 3-n-Butylamino-2'-chlor-2-methyl-propionanilid-hydrochlorid, 3-n-Butylamino-2-meth- yl-2'6'-propion-xylidid-hydrochlorid, 3-Isobutylamino-2methyl-o-propion-toluidid-hydrochlorid und 2'-Chlor3-cyclohexylamino-2-methyl-propionanilid-hydrochlorid besonders vorteilhaft für die Verwendung in der Infil trations-und Leitungsanästhesie für zahnärztliche und allgemeine chirurgische Zwecke, und sie sind für die örtliche Anästhesie von zugänglichen Schleimhäuten brauchbar.
Das erfindungsgemässe Verfahren wird durch die folgende Gleichung wiedergegeben :

Als Ausgangsmaterial I können Amine, wie z. B.
Methylamin, Athylamin, n-oder Isopropylamin, n-oder Isobutylamin und Cyclohexylamin, verwendet werden.
Als zweites Ausgangsmaterial II können Methacrylanilid, 2'-Chlor-methacrylanilid, 2'-Brommethacryl- anilid, o-Methacrylotoluidid und 2', 6'-Methacryloxyl- idid verwendet werden. Die Reaktion kann zweckmässig ausgeführt werden, indem man einen grossen Überschuss des Amins I, vorzugsweise ca. 3 Mol I, mit einem Mol des Methacrylanilids II mischt. Das Gemisch wird dann während einer gewissen Zeit bei Zimmertemperatur stehengelassen. Alkohole, wie z. B. Methanol und Athanol, können erforderlichenfalls als Reaktions- lösungsmittel verwendet werden. Das Endprodukt III kann aus dem Reaktionsgemisch in bekannter Weise gewonnen werden.
Beispielsweise kann das Produkt, nachdem die überschüssige Menge des Ausgangsamins aus dem Reaktionsgemisch entfernt worden ist, mit Benzol extrahiert werden. Das Produkt kann durch Destillation der Base unter vermindertem Druck weiter gereinigt werden. Die gereinigte Base kann mit pharmazeutisch verwendbaren Säuren in die Säureadditionssalze übergeführt werden.
Das Ausgangsmethacrylanilid II kann beispielsweise erhalten werden, indem man 2-Brom-2-methyl-propion- anilid oder seine vorstehend definierten Derivate in einem hochsiedenden, basischen Lösungsmittel, wie z. B. Chinolin, erhitzt.
Beispiel 1
16, 1 g (0, 1 Mol) Methacrylanilid wurden mit 17, 7 g (0, 3 Mol) n-Propylamin gemischt, und das Gemisch wurde 7 Tage lang bei Zimmertemperatur stehengelassen. Dann wurde die überschüssige Menge n-Propyl-' amin durch Destillation unter vermindertem Druck entfernt. Der Rückstand wurde mit Benzol extrahiert, und die Benzollösung wurde mit 10"/oiger Salzsäure extra- hiert. Der Extrakt wurde unter Eiskühlung mit Natriumhydroxyd alkalisch gemacht. Das resultierende bl wurde wieder mit Benzol extrahiert, und der Extrakt wurde mit wasserfreiem Natriumsulfat getrocknet.
Die Benzollösung wurde eingedampft, um Benzol zu entfernen, und das zur ckbleibende ol wurde unter ver mindertem Druck destilliert. Das Ol wurde in Benzol gelöst, und trockenes Chlorwasserstoffgas wurde durch die Lösung geleitet. Das Hydrochlorid wurde aus Methanol-Ather umkristallisiert, wodurch 12, 0 g 2-Methyl 3-n-propylamino-propionanilid-hydrochlorid als farblose Platten erhalten wurden.

Beispiel 2 16, 1 g Methacrylanilid und 22, 0 g n-Butylamin wurden wie in Beispiel 1 beschrieben behandelt, wodurch 15, 2 g 3-n-Butylamino-2-methyl-propionanilid-hydro- cMorid als farblose Schuppen erhalten wurden.

16, 1 g Methacrylanilid und 30, 0 g Cyclohexylamin wurden wie in Beispiel 1 beschrieben behandelt, wodurch 14, 4 g 3-Cyclohexylamino-2-methyl-propionani- lid-hydrochlorid als farblose Schuppen erhalten wurden.

Beispiel 4 17, 5 g o-Methacrylotoluidid wurden mit einer methanolischen Lösung gemischt, die 9, 3 g Methylamin enthielt, und das Gemisch wurde 7 Tage lang bei Zimmertemperatur stehengelassen. Dann wurden das Methanol und das überschüssige Methylamin durch Destillation unter vermindertem Druck entfernt. Der Rückstand wurde wie in Beispiel 1 beschrieben behandelt, und 15*8 g 3-Methylamino-2-methyl-o-propiontoluidid- hydrochlorid wurden als farblose feine Nadeln erhalten.

The present invention relates to a process for the preparation of a new group of substituted propionanilides with a local anesthetic effect.
The substituted propionanilides prepared according to the invention are represented by the following general formula:
