Verfahren zur Herstellung von (a-Alkylidenacyl)-phenyl-verbindungen
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von (a-Alkylidenacyl)-phenoxyund (a-Alkylidenacyl)-phenyl-mercapto-Derivaten der Monocarbonsäuren, in welchen der Monocarbonsäureteil von einer aliphatischen, aliphatisch-aromatischen oder einer aromatischen Monocarbonsäure abgeleitet ist.

veranschaulicht werden, worin jeder der Substituenten R, R1, R2 ist Wasserstoff, Halogen oder Halogenmethyl, Hydroxyl, niederaliphatische, niederaliphatische-oxyoder niederaliphatische-thio-Gruppe, mit gerader oder verzweigter gesättigter oder ungesättigter und substituierter oder unsubstituierter Kette, wobei als Substituenten Alkyl-, Amino-, Halogen-methyl-, Carboxyl- oder substituiertes Carboxyl-, Cyano-, Hydroxyl-, Alkylthio-, Arylthio-, Arylsulfonyl-, Alkylsulfonyl oder Nitrogruppen sein können; Rt, R2 ist ferner eine alicyclische substituierte oder unsubstituierte Gruppe, wobei die Substituent-Gruppen dieselben sind wie oben für die aliphatische Gruppe beschrieben;
eine Aryloder Aryloxy-oder Arylthio-Oruppe, insbesondere Phenyl, Phenoxy oder Phenylthio, worin der Aryl (Phenyl)-Teil unsubstituiert sein kann oder einen Substituenten gebunden an einen oder mehrere seiner Kohlenstoffatome aufweisen kann, welche Kohlenstoffatome gewählt sind aus ! niedrigen, geraden oder verzweigten Ketten, Alkyl, Alkoxy, Alkylthio, Hydroxyl, Halogen oder Halogen-methyl;
eine arylaliphatische Gruppe, insbesondere mononuklear-arylaliphatische, vorteilhafterweise Phenylalkyl, welcher mittels eines Sauerstoftoder Schwefelatoms an die Gruppe

R3, R4, R5 und R6 ist Wasserstoff, Halogen oder Halogenmethyl, niederaliphatische, niederaliphatische oxy oder niederaliphatische-thio mit gerader oder verzweigter, unsubstituierte oder mit einem Substituenten substituierte Kette, wobei der Substituent von oben beschriebenem Typus ist, und worin die niederaliphatische-oxy- oder niederaliphatische-thio-Gruppe vorteilhafterweise aus folgenden Gruppen besteht:
Niederalkoxy oder Niederalkylthio, unsubstituierte oder substituierte, wie z. B.
Carboxyalkoxy, Carboxyalkylthio und ähnliche; Aliphatischsulfonyl, insbesondere ein Alkylsulfonyl; Hydroxy, Nitro, Amino, Carboxy oder substituiertes Carboxy, insbesondere Carbamoyl und N-substituiertes Carbamoyl; Aryl, insbesondere Phenyl, unsubstituiertes oder substituiertes, wie oben beschrieben, zwecks Bindung an Aryl-(Phenyl) Teile; und wobei R5 und R4 und/oder R5 und R6 zusätzlich verkettet sind, unter Bildung, mit den Ringkohlenstoffen, an welchen sie gebunden sind, eines 5oder 6gliedrigen carbocyclischen Ringes.
A bedeutet Sauerstoff oder Schwefel, welcher in das Sulfoxyd oder Sulfon oxydiert werden kann.
B ist eine divalente, aliphatische, aromatische oder aliphatisch-aromatische Gruppe, vorteilhafterweise eine verzweigte oder gerade Kette einer niederaliphatischen Gruppe, welche Sauerstoff oder Schwefelatome, als Teil der aliphatischen Kette enthalten kann, eine Phenylniederalkyl- oder eine Phenylgruppe.
In den oben angeführten Definitionen und den Ansprüchen, umfasst der Ausdruck Halogen, Chlor, Brom, Jod, Fluor und Halogenmethyl, insbesondere Trichloromethyl, Trifluoromethyl und ähnliche.
Das erfindungsgemässe Verfahren zur Herstellung oben genannter (u-Alkylidenacyl)-phenyl-Verbindungen und deren Salze, Ester und Amide sind dadurch gekennzeichnet, dass man eine a, a-Dialiphatischacetyl-phe- nyl-Verbindung bromiert, in welcher Verbindung aer Phenylring an einen organischen Carbonsäureteil mittels eines divalenten Atoms, Sauerstoff oder Schwefel weiter gebunden ist und in welcher der Phenylring 1, 2, 3 oder 4 weitere Substituenten aufweist, dann die resultierende Verbindung mit einem Dehydrohalogenisierungsmittel behandelt und eine Verbindung aus dem Ausgangsmaterial erhält, an welchem der Acyl-Teil in einen a-Alkyliden- - aliphatischacyl - Teil konvertiert wurde.
Falls somit als Ausgangsmaterial ein gesättigtes Acylphenoxy- oder gesättigtes Acylphenylmercaptoderivat, mit zwei Methylgruppen und einem Wasserstoff, der an den zur Carbonylgruppe der Acylfunktion anliegendem Kohlenstoff gebunden ist, verwendet wird, wird die ungesättigte Acylgruppe durch Bromierung der gesättigten Acylgruppe und dann durch Entfernung des Bromwasserstoffs, gebildet; die Doppelbindung wird somit durch Behandlung mit einem Dehydrohalogenierungsmittel. wie Silberacetat oder Silberfluorid in Benzol oder Lithiumcnlorid in Dimethylformamid, eingeführt.
Falls eine der Methylgruppen im oben angeführten Ausgangsmaterial durch Athyl oder ein höheres Alkyl ersetzt wird, wird die ungesättigte Bindung hauptsächlich zwischen der höheren Alkylgruppe und dem Kohlenstoff in u-Stellung zur Carbonylfunktion, erzeugt. Im Falle alle Methylgruppen im oben angeführten Ausgangsmaterial durch eine höhere Alkylgruppe ersetzt sind, erscheint die Doppelbindung hauptsächlich zwischen dem sx-Kohlenstoff und der Alkylgruppe mit einer grö sseren Anzahl an Kohlenstoffatomen.
Gegenstand der vorliegenden Erfindung ist auch die Herstellung von Salzen, Estern und Amiden. Zur Herstellung der letzteren wird ein Amin der Struktur -NR;Ra verwendet, worin R7 und Rs Wasserstoff oder eine aliphatische, unsubstituierte oder substituierte Gruppe oder aromatische, unsubstituierte oder substituierte Gruppe ist, oder bei Verbindung mit dem Stickstoffatom, an welches sie gekettet sind, unter Bildung eines heterocyclischen Rings, für Pyrrolidyl, Morpholinyl oder Piperazinyl stehen können.
Die erfindungsgemäss hergestellten Verbindungen und insbesondere Verbindungen mit der oben veranschaulichten Struktur, worin mindestens eine niederaliphatische oder aromatische Gruppe an den a-Alkylidenacylteil der Verbindung gebunden ist, so wie ihre Derivate und Produkte, aus welchen sie hervorgegangen sind, besitzen nach Verabreichung diuretische, natriuretische, chloruretische und saluretische Eigenschaften ; sie sind deshalb nützlich bei der Behandlung von vielen Leiden, welche von einer übermässigen Retention der Elektrolyte, insbesondere Natriumchlorid oder Natriumund Chloridionen stammen. Die erfindungsgemäss erzeugten Verbindungen sind auch bei der Behandlung von Hypertension, der von einem Herzschlaganfall herrührt, Ödem und andern krankhaften Zuständen, welche mit der Elektrolyte und Flüssigkeitsretention verbunden sind.
Diese Verbindungen können auch verwendet werden zur Erhöhung des Urinabflusses. Eine aussergewöhnlich gute Wirksamkeit zeigt (a-Alkylidenacyl)phenoxyessigsäure und (a - Alkylidenacyl)-phenylmer- captoessigsäureverbindungen, in welchen der a-Alkylidenteil ein niederaliphatischer ist, und in welchen mindestens ein Substituent, vorteilhafterweise ein Halogen oder halogenähnlicher oder ein Niederalkylsubstituent, an den Phenylring gekettet ist.
Pharmakologische Studien dieses erfindungsgemäss erzeugten Produktes zeigen, dass sie unter den diuretischen Mitteln die einzigartige Eigenschaft, ein 2- bis 5mal grössere Ausscheidung an Elektrolyte, im Verhältnis zu den bekannten diuretischen Mitteln, zu verursachen, besitzen. Während somit die meisten bekannten diuretischen Mittel eine Schwelle oder Höhe in der Menge des Elektrolyten, den sie auszuscheiden vermögen, erreichen, und welche Schwelle nicht überschritten werden kann auch mit der Erhöhung der Dosis des Mittels, sind die erfindungsgemäss hergestellten Verbindungen imstande, die Ausscheidung einer 2- bis 5oder mehrfachen Menge dieses Schwellenwertes zu verursachen.
Während eine einfache Dosis einer erfindungsgemäss hergestellten Verbindung die Ausscheidung einer sehr grossen Menge an Elektrolyt verursacht, insbesondere falls die Verbindung intravenös verabreicht wird, fällt die Aktivität des Produktes auf etwa den Ausscheidungsschwellenwert z. B. des diuretischen Mittels vom Thiazid-Typus ab. Auf diese Weise ist es möglich, mittels der erfindungsgemäss erzeugten Produkte eine sehr schnelle und merkliche Entfernung an Elektrolyten zu erzielen, ohne dabei i eine Gefahr eines Elektrolytflüssig- keitentzuges zu Iaufen.
Das oben beschriebene Verfahren wird durch nachfolgende Beispiele näher erläutert.
Beispiel 1
4-Methacryloylphenoxy-essigsäure
35,6 g (0,16 Mol) von 4-Isobutyrylphenoxy-essigsäure werden zu 125 ml Eisessigsäure bei Zimmertemperatur zugegeben. Danach werden 25,7 g (0,16 Mol) Brom in 30 ml Eisessigsäure tropfenweise zu dem Reaktionsgemisch bei 25 C unter Rühren und innerhalb von einer Stunde zugefügt. Das Rühren wird während einer weitern Stunde fortgesetzt und dann wird das Gemisch in ein Gemisch von 300 g Eis und 500 ml Wasser geschüttet. Der Feststoff, welcher sich ausscheidet, wird auf einem Filter gesammelt, gewaschen und aus Benzol rekristallisiert; es werden 33 g von 4-(2 Bromisobutyryl)-phenoxy-essigsäure mit F = 144 bis 1450 C erhalten.
Analyse berechnet für Cr2H13BrO4 : Br, 26,54, gefunden: Br, 26,57.
12 g (0,04 Mol) der oben erhaltenen bromierten Verbindung werden in 800 ml Benzol gelöst und mit 15 g (0,09 Mol) Silberacetat versetzt. Das Gemisch wird gerührt und während 4 Stunden bei Rückfluss behandelt und dann gekühlt. Danach werden 150 ml Wasser und 15 ml konzentrierter Chlorwasserstoffsäure zugesetzt, wonach die Silbersalze ausfallen und durch Filtrieren entfernt werden. Das Benzol wird dann auf ein kleines Volumen abgedampft und mit Hexan verdünnt; der Feststoff wird separiert und aus Benzol kristallisiert und liefert 4,1 g von 4-Methacryloylphenoxy-essigsäure, mit F = 124,5 bis 126,50 C.
Analyse für C12H1204:
Berechnet: C 65,45; H 5,49, neutrales Äquivalent 220,2,
Gefunden: C 65,65; H 5,59, neutrales Äquivalent 221,7.
Die als Ausgangsmaterial verwendete 4-Isobutyrylphenoxy-essigsäure wird wie folgt erhalten:
160 g (1,2 Mol) von gepulvertem Aluminiumchlorid und 200 ml Schwefelkohlenstoff werden in einer 1 Liter-, mit einem Rührer, Tropftrichter, Rückflusskühler und in das Innere reichenden Thermometer versehenen, bHalsflasche gefüllt. Danach werden 61 g (0,4 Mol) Phenoxy-essigsäure portionenweise unter Rühren zugefügt und dann 53,5 g (0,5 Mol) Isobutyrylchlorid unter Rühren, innerhalb einer Zeitspanne von 0,5 Stunden bei etwa 22 bis 260 C zutropfen gelassen.
Nach Rühren während einer Stunde bei Zimmertemperatur, wird die Reaktionsflasche in ein Wasserbad gesetzt und die Temperatur bei 500 C während 3 Stunden aufrechterhalten, Der Schwefelkohlenstoff wird dann abdekantiert und der zurückgebliebene Aluminiumkomplex wird zu einem Gemisch von 500 g Eis und 125 ml konzentrierter Chlorwasserstoffsäure zugesetzt. Das sich gebildete gelbe Ö1 wird separiert und liefert 51,6 g an 4-Isobutyrylphenoxy-essigsäure mit Kp = 185-1900 C bei 1 mm Hg.
Beispiel 2
4-Metharyloyl-3-chlorophenoxy-essigsäure
Dieses Produkt wurde im wesentlichen mit Hilfe derselben Methode wie der in Beispiel 1 beschriebenen, erzeugt, unter Verwendung folgender Materialien: 3-Chloro-4-isobutyrylphenoxy- g 0,0379 Mol esagsäure 10,17 g 0,0379 Mol Eisessigsäure 250 ml Brom 6,34 g Eisessigsäure 30 ml
Man erhielt 4-(2-Bromoisobutyryl)-3-chloropihenoxy- essigsäure.
Analyse für Cz2Hl2BrClO4 : Berechnet: C 42,94 H 3,61 Br 23,81, Gefunden: C 43,33 H 3,78 Br 23,22.
Das oben genannte Produkt wurde dann gemäss dem in Beispiel 1 beschriebenen Verfahren unter Verwendung der folgenden Substanzen dehydrohalogeniert: 4-(2-Bromoisobutyryl)-3 -chloro- phenoxy-essigsäure 5 g 0,0149 Mol Benzol 200 ml Silberacetat 5 g 0,02990 Mol Wasser 150 ml konzentrierte Chlorwasserstoffsäure 15 ml
Das niedergeschlagene rohe Produkt wurde auf einem Filter gesammelt und liefert 2,8 g mit F = 125 bis 1270 C. Nach vier Rekristallisationen aus Benzol erhielt man 1,05 g von 4-Methacryloyl-3-chlorophenoxyessigsäure mit F = 128 bis 1290 C (Korrig.).
Analyse für C12H11ClO4: Berechnet: C 56,54 H 4,35 C1 13,93, Gefunden: C 56,31 H 4,45 C1 14,10.
Beispiel 3
2-Methacryloyl-5-chlorophenoxy-essigsäure
10 g (0,039 Mol) 2-Isobutyryl-5-chlorophenoxyessigsäure werden in 45 ml Essigsäure gelöst und es wird 6,25 g (0,039 Mol) in 20 ml Essigsäure gelöstem Brom zu der gerührten Lösung, indem Masse, indem sich das Brom entfernt, zugegeben. Dieser Vorgang dauert etwa 15 bis 20 Minuten. Das Reaktionsgemisch wird in 300 ml Wasser unter Rühren geschüttelt und der Feststoff, der ausfällt, wird aus einem 3:1-Gemisch von Hexan und Benzol kristallisiert; es werden 9,5 g 2-(2 Bromoisobutyryl)-5-chlorophenoxy-essigsäure mit F = 109-1110 C erhalten.
Analyse für C12H12BrC10: Berechnet: C 42,95 H 3,60 Br 23,81 C1 10,56, Gefunden: C 42,89 H 3,85 Br 23,59 C1 10,71.
4,3 g (0,0128 Mol) 2-(2-Bromoisobutyryl)-5-chlorophenoxy-essigsäure werden in 65 ml trockenem Benzol gelöst und 4,3 g Silberfluorid zugesetzt. Das Gemisch wird gerührt, während 10 Minuten erhitzt; danach gibt man 50 ml Wasser hinzu, kühlt das Gemisch hernach auf 200 C und säuert es an mit 6n Chlorwasserstoffsäure. Die ausgefallenen Silbersalze werden mittels einer Nutsche entfernt und die Benzolschicht von der wässrigen Schicht auf dem Filtrat separiert und mit einem gleichwertigen Athervolumen verdünnt. Diese Lösung wird mit mehreren Portionen einer 10% eigen Natriumbicarbonatlösung extrahiert und der wässrige Extrakt dann mit Chlorwasserstoffsäure angesäuert; man erhält ein Öl, das allmählich erstarrt.
Nach Separieren, trockenem und 4maligem Kristallisieren aus einem Gemisch von Benzol und Hexan erhält man 0,8 g 2-Methacryloyl-5-chlorophenoxy-essigsäure F = 100 bis 1010 C.
Analyse für C12H11C10: Berechnet: C 56,59 H 4,35 C1 13,93, Gefunden: C 56,65 H 4,58 C1 13,81.
Beispiel 4 4-(2-Athylidenpropionyl)-3-chlorphenoxy-essigsäure
110 g (0,037 Mol) 4-(2-Methylbutyryl)-3-chlorophenoxy-essigsäure in 200 ml Essigsäure gelöst und 6,0 g (0,037 Mol) Brom in 50 ml Essigsäure werden tropfenweise unter Rühren innerhalb von 10 bis 15 Minuten zugegeben. Die Reaktion wird durch Zusetzen von 2 Tropfen 48 % iger Bromwasserstoffsäure initiiert.
Das Reaktionsgemisch wird dann zu einem Liter Wasser, welches ein wenig Natriumbisulfit enthielt, gegeben und der sich separierte Feststoff gesammelt, mit Wasser ge waschen und an der Luft bei 65 C getrocknet. Nach Kristallisieren aus Benzol, erhält man 10,9 g 4-(2 Bromo-2-methylbutyryl)-3-chlorophenoxy-essigsäure, F = 123-124 C.
Analyse für C13H14BrClO4: Berechnet: C 44,66 H 4,04 Br 22,86 C1 10,09.
14 g (0,04 Mol) der oben erhaltenen Bromverbindung, 5,1 g (0,12 Mol) Lithiumchlorid und Dimethylformamid wurden vermischt und bei 80900 C während 4 Stunden erhitzt. Das Gemisch wird dann in 500 ml Wasser geschüttet. Der sich separierte Feststoff wird zweimal aus Natriumbicarbonat niedergeschlagen und dann aus Benzol kristallisiert und liefert 4,5 g 4-(2 Äthylidenpropionyl)-3-chlorophenoxy-essigsäure, F = 114-116 C.
Beispiel 5
4-(2-Äthylidenbutyryl)-3-chlorophenoxy-essigsäure
Zu einer Lösung von 10,64 g (0,0374 Mol) 4-(2 Äthylbutyryl)-3-chlorophenoxy-essigsäure in 200 ml Essigsäure werden unter Rühren 2 Tropfen 48 % iger Bromwasserstoffsäure zugesetzt und anschliessend 6,0 g (0,0374 Mol) Brom in 50 ml Essigsäure zutropfen gelassen. Nach Beendigung dieser Operation wird das Gemisch 15 Minuten gerührt und dann in 1 Liter Wasser, welches 2 g Natriumbisulfit enthielt, geschüttet.
Der sich separierte Feststoff wird auf einem Filter gesammelt, mit Wasser gewaschen, an der Luft getrocknet und aus 55 ml Benzol kristallisiert; man erhält 10,16 g 4-(2-Äthyl-2-bromobutyryl)-3-chlorophenoxy-essigsäure , F 130131 C.
Analyse Für C14H16BrClO4: Berechnet: C 46,23 H 4,44 Br 21,98 Cl 9,75, Gefunden: C 46,56 H 4,72 Br 21,91 Cl 9,72.
6 g (0,0185 Mol) des erhaltenen Bromketons werden in 400 ml heissem Menthol gelöst. Zu dieser Lösung gibt man dann 7 g (0,042 Mol) gepulvertes Silberacetat langsam und unter mechanischem Rühren, zu.
Das Gemisch wird während etwa 5,5 Stunden erhitzt und dann mit 6n Chlorwasserstoffsäure angesäuert. Das sich gebildete Silbersalz wird durch Filtrieren entfernt, die Benzol schicht separiert und 2g Darco-Aktivkohle zugefügt, worauf man dann das Gemisch bei 25 bis 30 C während 48 Stunden hält. Die Lösung wird dann filtriert, mit Äther verdünnt und mit 5 % iger Natriumbicarbonatlösung extrahiert. Der wässrige Extrakt wird mit Norit-Aktivkohle behandelt und angesäuert, liefert 4-(2-Äthylidenbutyryl)-3-chlorophenoxy-essigsäure, welche nach Trocknen an der Luft bei 650 C und Rekristallisieren aus Benzol bei 118-119 C schmilzt; man erhält 2,9 g.
Analyse für C14H1ClO4: Berechnet: C 59,47 H 5,35 C1 12,54, Gefunden: C 59,73 H 5,12 C1 12,50.
Das folgende Beispiel beschreibt eine alternative Methode zur Herstellung obiger Verbindung.
Beispiel 6
4-(2-Äthyldenbutyryl)-3-chlorophenoxy-essigsäure 8,3 g (0,0228 Mol) des gemäss Beispiel 5 hergestellten Bromketons, wird in 60 ml Dimethylformamid gelöst und die Lösung mit 2,9 g (0,0684 Mol) wasserfreiem Lithiumchlorid versetzt. Das Gemisch wird auf einem Dampfbad unter gelegentlichem Schütteln während 2 Stunden erhitzt, gekühlt und in 1 Liter kaltes Wasser geschüttet. Der Feststoff, der sich separiert, wird durch Filtrieren gesammelt, mit 500 ml Wasser gewaschen und dann in verdünnter Bicarbonatlösung gelöst. Die Lösung wird anschliessend mit Norit-Aktivkohle geschüttelt, frei von Feststoff und filtriert und angesäuert. Der sich separierte Feststoff wird an der Luft getrocknet und liefert 5 g 4-(2-Athylidenbutyryl)-3- chlorphenoxy-essigsäure, F = 119-1200 C.
Beispiel 7
2,3,-Dichloro-4-(2-äthylidenbutyryl) phenoxy-essigsäure
Gemäss dem in Beispiel 1 beschriebenen Verfahren wird aus 2,3 -Dichloro-4-(2-äthylbutyryl) -phenoxy-essig- säure die Verbindung 2,3-Dichloro-4-(2-bromo-2-äthyl- butyryl)-phenoxy-essigsäure, mit F = 151,5152,50 C und aus dieser das erwünschte Produkt 2,3-Dichloro-4- (2-äthylidenbutyryl)-phenoxy-essigsäure, mit F = 124 bis 125,50 C erhalten.
Analyse für C14H14Cl2O4: Berechnet: C 53,02 H 4,45 C1 22,36, Gefunden: C 53,28 H 4,43 Cl 22,34.
Beispiel 8 4-(2-Athylidenbutyryl)-2-äthyl-3-chloro- phenoxy-essigsäure
Gemäss dem in Beispiel 1 beschriebenen Verfahren wird 4-(2-Äthylbutyryl)-2-methyl-3-chlorophenoxy-essigsäure, mit F = 143-1440 C bromiert, wobei die Verbindung 4-(2-Bromo-2-äthylbutyryl)-2-methyl-3-chlorophenoxy-essigsäure, mit F = 136-137 C erhalten wird. Aus dieser wird dann durch Dehydrohalogenierung 4-(2-Äthyldenbutyryl)-2-methyl-3-chlorophenoxyessigsäure, mit F = 132,5l33,50 C erzeugt.
Analyse für C1H17ClO4: Berechnet: C 60,71 H 5,77 Cl 11,95, Gefunden: C 60,76 H 5,83 Cl 11,83.
Beispiel 9
4-(2-Äthyldenbutyryl)-2,3-dimethylphenoxy essigsäure
Gemäss dem in Beispiel 1 beschriebenen Verfahren wird 4-(2-Sithylbutyryl)-2, 3-dimethylphenoxy-essigsäure, mit F = 97-980 C mit Brom zu 4-(Bromo-2-äthyl butyryl)-2,3-dimethylphenoxy-essigsäure, mit F = 117 bis 1180 C umgewandelt und aus dieser Verbindung wird 4-(2-Äthylidenbutyryl)-2, 3-dimethylphenoxy-essig- säure mit F = 103 bis 1040 C hergestellt.

3-Chloro-4-senecioylphenoxy-essigsäure
Aus 3-Chloro-4-isovaleryl-phenoxy-essigsäure wurde gemäss Verfahren von Beispiel 1 3-Chloro-4-(2-bromisovaleryl)-phenoxy-essigsäure erzeugt.
12,3 g (0,035 Mol) dieser oben genannten Verbindung werden zu einer Lösung von 23,6 g (0,4 Mol) Trimethylamin in 150 ml absolutem Äthanol zugesetzt.
Die Lösung wird in einem mit Glas ausgekleideten Autoklav verschlossen und bei 800 C während 18 Stunden erhitzt. Nach Kühlen und Öffnen des Autoklaven wurde festgestellt, dass sich das Reaktionsgemisch aus einer rotbraunen Lösung und aus einem wei ssen kristallinen Niederschlag aus Trimethylamin-hydrobromid zusammensetzt. Der Feststoff wird durch Filtrieren entfernt und das Filtrat durch Abdampfen auf einem Dampfbad, auf ein kleines Volumen konzentriert.
Nach Zufügen des Rückstandes in Wasser bildet sich eine trübe Lösung des Trimethylaminsalzes des Produktes. Nach Ansäuern mit 6n Chlorwasserstoffsäure bildet sich ein fester Niederschlag, den man durch Filtrieren sammelt und hernach mit Natriumbicarbonat löst.
Die so erhaltene Lösung schüttelt man mit Entfärbungsholzkohle, filtriert und säuert mit Chlorwasserstoffsäure an. Es scheiden sich 7,3 g eines dunklen, bernsteinfarbigen Stoffes mit F = 120 bis 1300 C aus, welcher nach Kristallisieren aus einem Gemisch von 50 ml Cyclohexan und 70 ml Benzol und dann aus Benzol 1,4 g 3-Chloro 4-senecioyl-phenoxy-essigsäure, F = 134-136 C erhitzt.

3-Methyl-4-(1-isopropyliden-2-methylbutyryl) phenoxy-essigsäure
5 g 3-Methyl-4-(2 - isopropyl -3 - methylbutyryl)phenoxy-essigsäure werden in 40 ml Essigsäure gelöst und die Lösung auf 500 C erhitzt; zu dieser Lösung werden darauf einige Tropfen einer Lösung bestehend aus 2,56 g (0,016 Mol) Brom in 20 ml Essigsäure zugesetzt. Die sich gebildete Bromfarbe bleibt während 30 Minuten bestehen, wonach ein halber ml 48 % ige Bromwasserstoffsäure zugesetzt wird, was die rote Farbe des Broms langsam zum Verschwinden bringt. Man lässt dann das Gemisch auf 20 bis 250 C abkühlen und gibt die Hälfte der restlichen Bromlösung innerhalb von 30 Minuten zu. Nach weiterem 2stündigem Stehen bei 20 bis 300 C wird das Reaktionsgemisch, welches wiederum hellgelb geworden ist, auf 500 C erhitzt und der Rest der Bromlösung innerhalb von 30 Minuten zugegeben.
Darauf lässt man das so erhaltene Gemisch während 17 Stunden bei 20 bis 250 C stehen und schüttet es dann in Wasser, welches ein wenig Natriumbisulfit enthält. Der sich niedergeschlagene Feststoff wird durch Filtrieren gesammelt, gut mit Wasser gewaschen und liefert 6,3 g 3-Methyl-4-(2-bromo-2-isopropyl-3-methylbutyryl)-phenoxy-essigsäure, mit einem Schmelzpunkt unter Zersetzen, von etwa 140-1410 C. 3-chloro-4-senecioylphenoxy acetic acid
From 3-chloro-4-isovaleryl-phenoxy-acetic acid, 3-chloro-4- (2-bromoisovaleryl) -phenoxy-acetic acid was produced according to the method of Example 1.
12.3 g (0.035 mol) of the above-mentioned compound are added to a solution of 23.6 g (0.4 mol) of trimethylamine in 150 ml of absolute ethanol.
The solution is sealed in a glass-lined autoclave and heated at 800 ° C. for 18 hours. 3-methyl-4- (1-isopropylidene-2-methylbutyryl) phenoxy-acetic acid
5 g of 3-methyl-4- (2 - isopropyl -3 - methylbutyryl) phenoxy-acetic acid are dissolved in 40 ml of acetic acid and the solution is heated to 500 ° C .; A few drops of a solution consisting of 2.56 g (0.016 mol) of bromine in 20 ml of acetic acid are then added to this solution. The resulting bromine color persists for 30 minutes, after which half a ml of 48% hydrobromic acid is added which slowly causes the red color of the bromine to disappear. The mixture is then allowed to cool to 20 to 250 ° C. and half of the remaining bromine solution is added over the course of 30 minutes. 5.2 g (0.0141 mol) of the bromine compound prepared above and 3.59 g (0.0411 mol) of lithium bromide are added to 34 ml of methylformamide and the mixture is heated to 80 to 90 ° C. for 5 hours.
