Verfahren zur Herstellung von neuen Acridanderivaten
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen Acridanderivaten und ihren Säureadditionssalzen.
Es wurde überraschenderweise gefunden, dass die bisher nicht bekannten Acridanderivate der Formel I,

in welcher R1 und R2 unabhängig voneinander Wasser- stoff oder niedere Alkylreste, oder zusammen einen Polymethylenrest, und R3 und R4 unabhängig voneinander Wasserstoff, Halogenatome bis Atomnummer 35, Trifluormethylreste oder niedere Alkyl-oder Alkoxyreste bedeuten, R5 den Phenylrest, einen Biphenylyl-oder Phenoxyphenylrest oder einen Benzyl- oder Diphenylalkylrest, deren Benzolkerne in allen Fällen höchstens je drei Substituenten aus der durch Halogenatome, Trifluormethylreste, Nitrogruppen, Aminogruppen und nie dere Alkyls Alkoxy-, Alkylamino-, Dialkylamino- und Alkanoylaminoreste gebildeten Gruppe enthalten können,
oder einen gegebenenfalls durch Halogenatome, niedere Alkyl- oder Alkoxyreste substituiertem Pyridylrest, R6 einen niederen Alkylrest, und A einen Alkylidenoder Alkylenrest mit 1 bis 5 Kohlenstoffatomen bedeutet, und ihre Additionssalze mit anorganischen und organischen Säuren wertvolle pharmakologische Eigenschaften, insbesondere eine vorzügliche antivirale und tumorhemmende Wirksamkeit mit günstigem therapeutischem Index besitzen. Die antivirale Wirksamkeit lässt sich im Tierversuch, z. B. an der Maus, gegenüber dem Columbia-SK-Virus nachweisen, während die tumorhemmende Wirksamkeit in Tierversuchen, z. B. am induzierten Dimethylbenzanthracen-Hautcarcinom der Maus, festgestellt wurde.
Die Tierversuche charakterisieren die Verbindungen der Formel I und ihre Säureadditionssalze als Wirkstoffe zur Behandlung von Viruskrankheiten, wie Encephalitis, Encephalomyelitis und anderen, sowie zur Behandlung von Neoplasien. Überdies zeigen die Verbindungen der allgemeinen Formel I und ihre Säureadditionssalze eine mildle, aspirinartige, analgetische Wirksamkeit.
In den Verbindungen der Formel I und den entsprechenden, weiter unten genannten Ausgangsstoffen sind R1 und R2 unabhängig voneinander, beispielsweise durch Wasserstoff, Methyl-, Äthyl- oder n-Propylreste, Isopropyl-, n-Butyl- oder Isobutylreste verkörpert oder bilden zusammen z. B. den Tetramethylen-, Pentamethylen oder Hexamethylenrest. R3 und R4 sind unabhängig voneinander, beispielsweise Wasserstoff, Fluor, Chlor- oder Bromatome, Trifluormethyl-, Methyl-, Äthyl-, n-Propyl-, Isopropyl-, Methoxy-, Athoxy-, n Propoxy- oder Isopropoxyreste. R5 ist z.

R6 ist z.B. ein Methyl-, Äthyl-, n-Propyl-, Isopropyl-, n-Butyl-, Isobutyl-, sek.-Butyl-, n-Pentyl-, Isopentyl- oder n-Hexylrest.
A ist als Alkylidenrest z. B. ein Methylen-, Äth- yliden-, Propyliden-, Isopropyliden-, Butyliden-, Iso butyliden-oder Pentylidenrest, und als Alkylenrest z. B. ein Sithylen-, Propylen-, Äthyl äthyl en-, Trimethylen-, Tetramethylen- oder Pentamethylenrest.
Die neuen Verbindungen der Formel I erhält man, indem man auf eine Verbindung der Formel II,

Niedere Oxoalkane, wie Formaldehyd und Acetaldehyd, werden z. B. in Gegenwart von katalytisch aktiviertem Wasserstoff in einem inerten organischen Lösungsmittel mit Verbindungen der Formel II in Reaktion gebracht, Formaldehyd kann auch gelöst in Ameisensäure in der Wärme eingesetzt werden.

in welcher Rr, R2, R3 und R4 die oben angegebene Bedeutung haben, mit Halogenalkanoylhalogeniden, Alkan sulfonyioxyalkanoylhalogeniden oder Arylsulfonyloxyalkanoylhalogeniden in inerten organischen Lösungsmitteln, wie z. B. Benzol oder Toluol, in der Wärme leicht herstellbar. Einzelne Vertreter dieses Verbindungstypus, z.B. das 10-Chloracetyl-, das 10-(3-Chlor-pro pionyl)- und das lO-(5-Jod-valeryl)-acridan, sind bekannt. Eine Anzahl substituierter Acridane der Formel V ist bekannt, und weitere sind analog den bekannten herstellbar.
Amine der Formel IV sind in grosser Zahl bekannt.
Die nach erfindungsgemässen Verfahren erhaltenen Verbindungen der Formel I werden gewünschtenfalls anschliessend in üblicher Weise in ihre Additionssalze mit anorganischen und organischen Säuren übergeführt.
Beispielsweise versetzt man eine Lösung einer Verbindung der Formel I in einem organischen Lösungsmittel, wie Methanol, Äthanol oder Äther, mit der als Salzkomponente gewünschten Säure oder einer Lösung derselben und trennt das ausgefallene Salz ab. Zur Verwendung als Arzneistoffe können anstelle freier Basen nichttoxische Salze hergestellt werden, das heisst, Salze mit solchen Säuren, deren Anionen bei den in Frage kommenden Dosierungen pharmakologisch annehmbar sind, das heisst keine toxischen Wirkungen ausüben.
Ferner ist es von Vorteil, wenn die als Arzneistoffe zu verwendenden Salze gut kristallisierbar und nicht oder wenig hygroskopisch sind. Zur Salzbildung mit Verbindungen der Formel I können z. B. die Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphor säune, Methansulfonsäure, Äthansulfonsäure, PHydroxy- äthansulfonsäure, Essigsäure, Milchsäure, Oxalsäure, Bernsteinsäure, Fumarsäure, Maleinsäure, Apfelsäure, Weinsäure, Zitronensäure, Benzoesäure, Salicylsäure, Phenylessigsäure, Mandelsäure und Embonsäure verwendet werden.
Die neuen Verbindungen der Formel I und ihre pharmazeutisch annehmbaren Salze können peroral, rektal und parenteral verabreicht werden. Die täglichen Dosen der freien Basen oder von pharmazeutisch annehmbaren Salzen derselben bewegen sich zwischen 50 und 5000 mg für erwachsene Patienten. Geeignete Doseneinheitsformen, wie Dragees, Tabletten, Suppositorien oder Ampullen, enthalten vorzugsweise 10 bis 500 mg eines erfindungsgemässen Wirkstoffs oder eines pharmazeutisch annehmbaren Salzes desselben. Ferner kommt auch die Anwendung entsprechender Mengen von nicht einzeldosierten Applikationsformen, wie Sirups, Sprays, Aerosole, Salben oder Puder, in Betracht.
Doseneinheitsformen für die perorale Anwendung enthalten als Wirkstoff vorzugsweise zwischen 5 und 90 % einer Verbindung der Formel I oder eines pharmazeutisch annehmbaren Salzes einer solchen. Zu ihrer Herstellung kombiniert man den Wirkstoff z. B. mit festen, pulverförmigen Trägerstoffen, wie Lactose, Saccharose, Sorbit, Mannit; Stärken, wie Kartoffelstärke, Mais stärke oder Amylopektin, ferner Laminariapulver oder Citruspulpenpulver; Cellulosederivaten oder Gelatine, gegebenenfalls unter Zusatz von Gleitmitteln, wie Magnesium-oder Calciumstearat oder Polyäthylenglykolen (Carbowaxen) von geeigneten Molekulargewichten zu Tabletten oder zu Dragee-Kernen. Letztere überzieht man beispielsweise mit konzentrierten Zuckerlösungen, welche z.
B. noch arabischen Gummi, Talk und/oder Titandioxyd enthalten können, oder mit einem in leichtflüchtigen organischen Lösungsmitteln oder Lösungsmittelgemischen gelösten Lack. Diesen Überzügen können Farbstoffe zugefügt werden, z. B. zur Kennzeichnung verschiedener Wirkstoffdosen.
Als Doseneinheitsformen für die rektale Anwendung kommen z. B. Suppositorien, welche aus einer Kombination eines Wirkstoffs oder eines geeigneten Salzes desselben mit einer Neutralfettgrundlage bestehen, oder auch Gelatine-Rektalkapseln, welche eine Kombination des Wirkstoffs oder eines geeigneten Salzes desselben mit Polyäthylenglykolen (Carbowaxen) von geeignetem Molekulargewicht enthalten, in Betracht.
Ampullen zur parenteralen, insbesondere intramuskulären Verabreichung enthalten vorzugsweise ein wasserlösliches Salz eines Wirkstoffs in einer Konzentration von vorzugsweise 0,5 bis 10 %, gegebenenfalls zusammen mit geeigneten Stabilisierungsmitteln und Puffersubstanzen, in wässriger Lösung.
Die folgenden Vorschriften sollen die Herstellung von Tabletten und Dragees näher erläutern: a) 1000 g Wirkstoff, z. B. 10-[N-(3, 4-Dichlorbenzyl)- glycyl]-9, 9-dimethyl-acridan-hydrochlorid, werden mit 550,0 g Lactose und 292,0 g Kartoffelstärke vermischt, die Mischung mit einer alkoholischen Lösung von 8 g Gelatine befeuchtet und durch ein Sieb granuliert. Nach dem Trocknen mischt man 60,0 g Kartoffelstärke, 60,0 g Talk, 10,0 g Magnesiumstearat und 20,0 g kolloidales Siliciumdioxyd zu und presst die Mischung zu 10000 Tabletten von je 200 mg Gewicht und 100 mg Wirkstoffgehalt (Hydrochlorid), die gewünschtenfalls mit Teilkerben zur feineren Anpassung der Dosierung versehen sein können. b) Aus 1000 g Wirkstoff, z.
B. 10-[N-(3,4-Dichlor- phenyl)-glycyi]-acridan, 379,0 g Lactose und der alkoho- lischen Lösung von 6,0 g Gelatine stellt man ein Granulat her, das man nach dem Trocknen mit 10,0 g kolloidalem Siliciumdioxyd, 40,0 g Talk, 60,0 g Kartoffelstärke und 0,5 g Magnesiumstearat mischt und zu 10 000 Dragie-Kernen presst. Diese werden anschliessend mit einem konzentrierten Sirup aus 533,5 g kristallisierter Saccharose, 20,0 g Schellack, 75,0 g arabischen Gummi, 250,0 g Talk, 20,0 g kolloidalem Siliciumdioxyd und 1,5 g Farbstoff überzogen und getrocknet. Die erhaltenen Dragees wiegen je 240 mg und enthalten je 100 mg Wirkstoff.
Im nachfolgenden Beispiel sind die Temperaturen in Celsiusgraden angebeben.
Beispiel 4,0 g (0,009 Mol) 10 10-[N-(2,4-Dichlorbenzyl)-glycyl]- ! 9,9-dimethyl-acridan (freigesetzt aus dem Hydrochlorid, Smp. 2350, unter Zersetzung) werden in 1,6 g (0,030 Mol) 85 % iger Ameisensäure gelöst und mit 2 ml (0,020 Mol) einer 40 % igen Formalinlösung versetzt.
Unter Rühren bei Raumtemperatur tritt heftiges Aufschäumen ein. Nach dem Nachlassen der Gasentwicklung wird noch während 8 Stunden in einem Ölbad von 100 bis 1100 erhitzt. Nach dem Abkühlen versetzt man mit 3 ml 5n Salzsäure und dampft unter vermindertem Druck ein. Der Rückstand wird mit 100 ml Methylenchlorid versetzt und aus dem Hydrochlorid des 10-[N (2,4-Dichlorbenzyl)-N-methylglycyl]-9,9-dimethyl-acridan mit 20 ml 2n Natronlauge die entsprechende Base freigesetzt. Die Methylenchloridlösung wird mit dreimal 50 ml Wasser gewaschen, über Calciumchlorid getrocknet und eingedampft. Der Rückstand (3,9 g) wird in 100 ml Äther gelöst und mit 3 ml 3n ätherischer Salzsäurelösung ins Hydrochlorid übergeführt.
Das ausge- fallene Hydrochlorid wird abgenutscht und aus Methylien- chlorid-Hexan umkristallisiert. Man erhält so 3,0 g 10- [N-(2 ,4-Dichlorbenzyl)-N-methyl- glycyl] -9 ,9-dimethyl-acridan Hydrochlorid, Smp. 204 bis 2050, unter Zersetzung.

The antiviral effectiveness can be tested on animals, e.g. B. on the mouse, against the Columbia SK virus, while the anti-tumor activity in animal experiments, z. B. on the induced dimethylbenzanthracene skin carcinoma of the mouse.
The animal experiments characterize the compounds of the formula I and their acid addition salts as active ingredients for the treatment of viral diseases such as encephalitis, encephalomyelitis and others, and for the treatment of neoplasms. In addition, the compounds of general formula I and their acid addition salts show a mild, aspirin-like, analgesic activity.

B. benzene or toluene, easily produced in the heat. Individual representatives of this type of connection, e.g. 10-chloroacetyl-, 10- (3-chloro-propionyl) - and 10- (5-iodo-valeryl) acridane are known. A number of substituted acridans of the formula V are known, and others can be prepared analogously to the known.
Amines of the formula IV are known in large numbers.
The compounds of the formula I obtained by the process according to the invention are, if desired, then converted into their addition salts with inorganic and organic acids in the customary manner.
For example, a solution of a compound of the formula I in an organic solvent, such as methanol, ethanol or ether, is mixed with the acid or a solution thereof desired as the salt component and the precipitated salt is separated off. For use as medicinal substances, non-toxic salts can be prepared instead of free bases, that is to say salts with acids whose anions are pharmacologically acceptable at the dosages in question, that is to say do not have any toxic effects.
It is also advantageous if the salts to be used as medicinal substances are readily crystallizable and have little or no hygroscopic properties. For salt formation with compounds of formula I, for. B. hydrochloric acid, hydrobromic acid, sulfuric acid, phosphoric acid, methanesulphonic acid, ethanesulphonic acid, PHydroxyethanesulphonic acid, acetic acid, lactic acid, oxalic acid, succinic acid, fumaric acid, maleic acid, malic acid, tartaric acid, citric acid, benzoic acid and emicylic acid, phenylic acid are used.
The new compounds of the formula I and their pharmaceutically acceptable salts can be administered orally, rectally and parenterally. The daily doses of the free bases or pharmaceutically acceptable salts thereof range between 50 and 5000 mg for adult patients. Suitable dosage unit forms, such as dragees, tablets, suppositories or ampoules, preferably contain 10 to 500 mg of an active ingredient according to the invention or a pharmaceutically acceptable salt thereof. Furthermore, the use of corresponding amounts of non-metered application forms, such as syrups, sprays, aerosols, ointments or powders, is also possible.
Unit dosage forms for oral use contain as active ingredient between 5 and 90% of a compound of the formula I or a pharmaceutically acceptable salt thereof. To produce them, combining the active ingredient z. B. with solid, powdery carriers such as lactose, sucrose, sorbitol, mannitol; Starches, such as potato starch, corn starch or amylopectin, also laminaria powder or citrus pulp powder; Cellulose derivatives or gelatin, optionally with the addition of lubricants, such as magnesium or calcium stearate or polyethylene glycols (carbowaxene) of suitable molecular weights to form tablets or dragee cores. The latter is coated, for example, with concentrated sugar solutions, which z.
B. can contain arabic gum, talc and / or titanium dioxide, or with a paint dissolved in volatile organic solvents or solvent mixtures. Dyes can be added to these coatings, e.g. B. to identify different drug doses.
As unit dosage forms for rectal use, for. B. suppositories, which consist of a combination of an active ingredient or a suitable salt thereof with a neutral fat base, or gelatin rectal capsules, which contain a combination of the active ingredient or a suitable salt thereof with polyethylene glycols (carbowaxene) of suitable molecular weight, into consideration.
Ampoules for parenteral, in particular intramuscular, administration preferably contain a water-soluble salt of an active ingredient in a concentration of preferably 0.5 to 10%, optionally together with suitable stabilizers and buffer substances, in aqueous solution.
The following rules are intended to explain the manufacture of tablets and coated tablets in more detail: a) 1000 g of active ingredient, e.g. B. 10- [N- (3, 4-dichlorobenzyl) - glycyl] -9, 9-dimethyl-acridan hydrochloride, are mixed with 550.0 g of lactose and 292.0 g of potato starch, the mixture with an alcoholic solution of 8 g of gelatin moistened and granulated through a sieve. After drying, 60.0 g of potato starch, 60.0 g of talc, 10.0 g of magnesium stearate and 20.0 g of colloidal silicon dioxide are mixed in and the mixture is pressed into 10,000 tablets each weighing 200 mg and containing 100 mg of active ingredient (hydrochloride), which, if desired, can be provided with partial notches for finer adjustment of the dosage. b) From 1000 g of active ingredient, e.g.
B. 10- [N- (3,4-dichlorophenyl) -glycyi] -acridane, 379.0 g of lactose and the alcoholic solution of 6.0 g of gelatine are made into granules which are dried mixed with 10.0 g of colloidal silicon dioxide, 40.0 g of talc, 60.0 g of potato starch and 0.5 g of magnesium stearate and pressed to 10,000 dragie kernels. These are then coated with a concentrated syrup of 533.5 g of crystallized sucrose, 20.0 g of shellac, 75.0 g of gum arabic, 250.0 g of talc, 20.0 g of colloidal silicon dioxide and 1.5 g of dye and dried. The coated tablets each weigh 240 mg and each contain 100 mg of active ingredient.
In the following example the temperatures are given in degrees Celsius.
Example 4.0 g (0.009 mol) 10 10- [N- (2,4-dichlorobenzyl) -glycyl] -! 9,9-dimethyl-acridane (released from the hydrochloride, m.p. 2350, with decomposition) are dissolved in 1.6 g (0.030 mol) of 85% formic acid, and 2 ml (0.020 mol) of a 40% formalin solution are added.
Vigorous foaming occurs with stirring at room temperature. After the evolution of gas has subsided, the mixture is heated in an oil bath from 100 to 1100 for 8 hours. After cooling, 3 ml of 5N hydrochloric acid are added and the mixture is evaporated under reduced pressure. 100 ml of methylene chloride are added to the residue, and the corresponding base is released from the hydrochloride of 10- [N (2,4-dichlorobenzyl) -N-methylglycyl] -9,9-dimethyl acridane with 20 ml of 2N sodium hydroxide solution. The methylene chloride solution is washed three times with 50 ml of water, dried over calcium chloride and evaporated. The residue (3.9 g) is dissolved in 100 ml of ether and converted into the hydrochloride with 3 ml of 3N ethereal hydrochloric acid solution.
The precipitated hydrochloride is filtered off with suction and recrystallized from methylene chloride-hexane. 3.0 g of 10- [N- (2,4-dichlorobenzyl) -N-methyl-glycyl] -9,9-dimethyl-acridane hydrochloride, melting point 204 to 2050, are obtained with decomposition.
