Description

  

  Verfahren zur Herstellung von Bis-thiosemicarbazonen    Es wurde bereits vorgeschlagen, basisch     substituierte     Bis-4-alkyl-thiosemicarbazone des Diacetyls der allge  meinen Formel

    worin R .den Methyl- oder Äthylrest oder die Gruppe  NRR die Pyrrolidino-, Piperidino- oder     2-Methyl-          piperidino-Gruppe    bedeuten, herzustellen. Diese Ver  bindungen weisen eine Wirkung gegen Geflügelcoccidiose  (Eimeria tenella) in vivo- auf, wie in Nature Bd. 206,  1340 (1965) nachträglich     bestätigt    wurde.  



  Es wurde nun gefunden, dass spezielle unsymme  trisch basisch substituierte Bis-thiosemicarbazone gegen  über strukturell ganz ähnlich gebauten Verbindungen  eine überraschend grössere chemotherapeutische Breite       aufweisen    und daher hervorragend für die. Anwendung  gegen Geflügel coccidiose geeignet sind.  



  Die Erfindung betrifft daher ein Verfahren zur  Herstellung von basisch substituierten     Bis-thio-semi-          carbazonen    der Formel I

    worin R eine Diäthylamino- oder Piperidino-Gruppe  bedeutet, das dadurch     gekennzeichnet    ist, dass man ein  Monothiosemicarbazon der Formel II

    mit dem Senföl der Formel III  S=C=N-CH2-CH2-B (III)  umsetzt, wobei jeweils einer der Reste A und B die  Gruppe

  Die erhaltenen Basen kann man mit Säuren in ihre  Säureadditionssalze und die Salze .durch Behandlung  mit Basen in die freien Basen überführen.  



  Es sind bereits substituierte Bis-thiosemicarbazone  von Diketonen bekannt, vgl. österr. Patentschrift Num  mer 225 715 sowie Nature Bd. 206, S. 1340 ff (1965).  



  Die Verfahrensprodukte des erfindungsgemässen  Verfahrens     wurden        jedoch    bisher nicht     beschrieben;     unsymmetrisch basisch substituierte     Diacetyl-bis-thio-          semicarbazone    sind bisher nicht bekanntgeworden.  



  Der besondere     Vorteil    der     Verfahrensprodukte    liegt  darin, dass sie auf Grund ihrer vorteilhaften Eigen  schaften (geringe     Toxizität)    zur Routineprophylaxe bei  der     Geflügelcoccidiose        vorzüglich        geeignet    sind;     während     dies bei den bisher     bekannten    basisch substituierten     Bis-          thiosemicarbazonen    nicht der Fall ist (vgl. Nature,     Bd.     206, S. 1341 rechte Spalte,     Zeilen    30-34).

        Die als Ausgangsstoffe herangezogenen Senföle sind  2-Diäthylaminoäthyl-, 2-Piperidinoäthyl- und  3-Pyrrolidino-propylsenföl.  Die     Senföle    werden in bekannter Weise aus den  Aminen durch Umsetzung mit     Schwefelkohlenstoff        und     anschliessender     Oxydation    der dabei in     erster    Stufe ent  stehenden Dithiocarbamate mit Natriumchlorid (NaClO2)  dargestellt.  



  Die neuen     Verfahrenserzeugnisse    können als Heil  mittel Verwendung finden. Insbesondere eignen sie sich  durch ihren günstigen chemotherapeutischen     Index    für  die Therapie und Prophylaxe der Geflügelcoccidiose  und können als Zusatz zum Trinkwasser oder Futter  zur Anwendung kommen.     Beispielsweise    wird eine Ver  abreichung der     Verfahrenserzeugnisse    von 0,0025 bis  0,01 ö im Futter bei sicherer Wirkung über     einen    Zeit  raum von einigen Wochen gut vertragen.

   Die im folgen  den aufgeführten Tabellen 2 (Toxizität) und 1 (Wirkung)  zeigen die Überlegenheit der Verfahrenserzeugnisse 1  und II gegenüber einer strukturähnlichen unsymme  trischen Verbindung (III) und dem symmetrisch, ba  sisch substituierten Bis-thio-semicarbazon IV, wie sich  aus dem Index beim Sektionsbefund ergibt.  



  Gemäss Tabellen 1 und 2 wurden die Substanzen im       Trinkwasserversuch    auf ihre     Verträglichkeit    und thera  peutische Wirkung gegenüber einer Coccidieninfektion  bei Küken geprüft.  



  Bei den therapeutischen Trinkwasserversuchen  wurde so vorgegangen, dass ein Kollektiv mehrerer Tage  alter Küken (weisse Leghorn) mit je 100 000 versporten  Oocysten (Eimeria tenella) mittels Kropfsonde infiziert  wurden. Danach wurden die     gleichmässig    infizierten  Tiere in einzelne Gruppen von meist 10 Küken unter  teilt.

   Diese Gruppen wurden entweder gemeinsam be  handelt, wie in der     Tabelle    angegeben, oder dienten als  infizierte, nicht behandelte Kontrollgruppen.     Die    Infek  tion war so     eingestellt,    dass sämtliche infizierten Kon  trolltiere im Beobachtungszeitraum einer     Blinddarm-          coccidiose    erlagen; diese Ergebnisse sind daher nicht  einzeln aufgeführt.     Die    Behandlung     begann        unmittelbar     nach der     Infektion    für 48     .Stunden;    die Lösung     wurde     einmal erneuert.

   Während der Zeit von 2 Tagen konn  ten die Versuchstiere das mit dem     Präparat    versetzte         Trinkwasser    ad libitum     aufnehmen.    Während des Ver  suches wurden die Tiere auf Ooc.-Ausscheidung unter  sucht, und es wurde     festgestellt,    ob etwa aufgetretene  Todesfälle auf Coccidiose zurückzuführen waren. oder  auf andere Ursachen. Am Ende des Versuches wurde  ein Sektion der überlebenden Tiere durchgeführt,     und     es wurde der Schweregrad des Befalls festgestellt, der  numerisch in Werten von 0-8 ausgedrückt wird. Bei  den in den Tabellen aufgeführten Werten     handelt    es  sich um     die    Mittelwerte des jeweiligen Kollektivs.  



  Bei den Verträglichkeitsversuchen in     Tabelle    2  wurde so     vorgegangen,    dass die nicht infizierten     Tiere     die zu     prüfende        Substanz    mit dem     Trinkwasser        in    glei  cher Weise     erhielten,    wobei unter diesen Versuchsbe  dingungen das mit dem Präparat versetzte Trinkwasser  den     Tieren    5 Tage lang zur Verfügung stand. Die bei  den     Verträglichkeitsversuchen    verwendeten Küken ge  hörten der gleichen Rasse an wie die Tiere der thera  peutischen Versuche.

   Die Versuchsdauer     betrug    minde  stens 3 Wochen, so dass eine genügend lange Beob  achtungszeit gegeben war. Die erste Verabreichung des  mit Präparat versetzten     Trinkwassers    geschah zu Beginn       :dieser    Versuchsperiode. Die in den Tabellen aufge  führten     Überlebensraten    wurden am Ende der     3wöchi-          gen        Beobachtungsperiode    ermittelt.

Dies geht einer  seits aus dem Sektionsbefund gemäss Tabelle 1 hervor,  anderseits ist die geringe Toxizität (Tabelle 2)     besonders     im Hinblick auf     die    Verbindung IV bei den nicht  infizierten Tieren deutlich.  



  Da die Verfahrensprodukte I und II eine höhere  Coccidiosewirkung besitzen und, wie aus den obigen  Tabellen ersichtlich, eine wesentlich bessere Verträglich  keit zeigen, sind sie     hinsichtlich    des chemotherapeu  tischen Indexes den Verbindungen III und IV erheblich  überlegen.  



  <I>Beispiel 1</I>  27,2 g (0,1 Mol)     Diacetyl-mono-(2-diäthylamino-          äthylthiosemicarbazon)-mono-hydrazon    werden in 80 cm3  Äthanol durch Erwärmen gelöst und 17,0 g (0,1 Mol)  3-Pyrrolidinopropyl-senföl zugegeben und das ganze  3 Stunden am Dampfbad unter Rückfluss erhitzt. Die  Reaktionslösung wird unter     vermindertem    Druck auf  '/;; des Gesamtvolumens     eingedampft    und das Endpro  dukt durch Zugabe von Äther zur Abscheidung ge  bracht. Es wird abgesaugt, aus Äthanol umkristallisiert,  abgesaugt, mit     kaltem    Äthanol und Äther gewaschen  und am Dampfbad getrocknet.

   Man     erhält    35 g = 79  der Theorie  Diacetyl-mono-(2-diäthylaminoäthylthiosemi       carbazon)-mono-(3-pyrrolidinopropylthio-          semicarbazon).     



  Die Substanz schmilzt bei 150 C.  



  Das als Ausgangsstoff verwendete     Diacetyl-mono-          (2-diäthylaminoäthylthiosemicarbazon)    -mono-hydrazon  wurde dargestellt durch langsames Zutropfen (etwa  1 Stunde) einer Lösung von 15,8 g (0,1 Mol)     2-Diäthyl-          aminoäthyl-senföl    (Kp.2mm 70-72  C) in 25 cm3 Äthanol  zu einer siedenden Lösung von 11,4 g (0,1 Mol)     Di-          acetyl-dihydrazon    (Schmp. l64  C) in 150 cm3 Äthanol.

    Ist alles eingetragen, wird noch 15 Minuten unter Rück  fluss gekocht, abgekühlt und von kleinen Anteilen aus  fallender Bis Verbindung     abgesaugt.    Das Filtrat wird  unter vermindertem Druck auf     1/.1    des Gesamtvolumens  eingedampft und das gewünschte Produkt durch Zugabe  von Äther zur Abscheidung gebracht. Es wird abge  saugt, aus Benzol umkristallisiert, abgesaugt, mit kaltem  Äthanol und Äther gewaschen und im Exsikkator ge  trocknet. Man erhält 20 g = 73 % der Theorie     Diacetyl-          mono    - (2 -. diäthylaminoäthylthiosemicarbazon) - mono     -          hydrazon    als gelbliches Kristallpulver vom Schmelzpunkt  115 bis 117 C.

      Das .als Ausgangsstoff verwendete       Diacetyl-mono-(2-diäthylaminoäthylthiosemi-          carbazon)-mono-hydrazon     kann auch durch 2stündiges Kochen einer     Lösung    von  11,5 g (0,1 Mol) Diacetyl-mono-hydrazon-mono-oxim  in 50 cm3 Äthanol und 19,0 g (0,1 Mol)     2-Diäthyl-          aminoäthylthiosemicarbazid    in 70 cm3 Äthanol und  22 cm3 wässriger konz. Salzsäure hergestellt werden.  Nach Abkühlen der Reaktionslösung wird durch por  tionsweise Zugabe von verdünntem Ammoniak bis  pH 7,5 die Verbindung ausgeschieden, abgesaugt, mit  Wasser, kaltem     Äthanol    und Äther gewaschen und  getrocknet. Ausbeute<B>70%</B> der Theorie, Schmelzpunkt  130 bis 132  C.  



  <I>Beispiel 2</I>  Durch Umsetzung von 28,4 g (0,1 Mol)     Diacetyl-          mono    - (3 - pyrrolidinopropylthiosemicarbazon) -     mono-          hydrazon    und 15,8 g (0,1 Mol)     2-Diäthylaminoäthyl-          senföl    unter gleichen Bedingungen, wie in Beispiel 3  beschrieben, erhält man nach analoger Aufarbeitungs  weise 37 g = 84 % der Theorie  Diacetyl-mono-(2-diäthylaminoäthylthiosemi       carbazon)-mono-(3-pyrrolidinopropylthio-          semicarbazon)     vom Schmelzpunkt     149-151'C.     



  Das als Ausgangsstoff verwendete     Diacetyl-mono-          (3-pyrrolidino-propylthiosemicarbazon)-mono-hydrazon     wurde durch Umsetzung von 17,0 g (0,1 Mol)     3-          Pyrrolidinopropyl-senföl    (Kp.1mm 87-90  C) und 11,4 g  (0,1 Mol) Diacetyl-dihydrazon wie in Beispiel 1 be  schriebenen Weise dargestellt. Man ,erhält 22 g = 76  der Theorie als gelbliches Kristallpulver vom Schmelz  punkt 127-128  C.  



  Das als Ausgangsstoff verwendete     Diacetyl-mono-          (3-pyrrolidinopropylthiosemicarbazon)    -mono - hydrazon  kann auch durch 2stündiges Kochen einer Lösung von  11,5 g (0,1 Mol) Diacetyl-mono-hydrazon-mono-oxim  und 20,2 g (0,1 Mol)     3-Pyrrolidinopropylthiosemi-          carbazid    wie in Beispiel 1 angegeben hergestellt wer  den. Ausbeute<B>67%</B> der Theorie, Schmelzpunkt 12,6  bis 127  C.  



  <I>Beispiel 3</I>       Diacetyl-mono-(2-piperidinoäthylthiosemicarbazon)-          mono-(3-pyrrolidinopropylthiosemicarbazon     28,2 g (0,1 Mol)     Diacetyl-mono-(3-pyrrolidino-          propylthiosemicarbazon)-mono-hydrazon    und 17,0 g  (0,1     Mol)        2-Piperidinoäthyl-senföl        (Kp.i        mm    84-86 C)  werden unter     gleichen        Bedingungen    wie in Beispiel 1  und 2 beschrieben umgesetzt und isoliert.

        .Man erhält 35 g = 77 % der Theorie     Diacetyl-          mono    - (2 - piperidinoäthylthiosemicarbazon) - mono -     (3-          pyrrolidinopropylthiosemicarbazon)    in Form von gelb  lichen Kristallen vom Zersetzungspunkt     224-225     C.

  There are already substituted bis-thiosemicarbazones of diketones known, cf. Austrian patent number 225 715 and Nature Vol. 206, pp. 1340 ff (1965).



  However, the products of the process according to the invention have not yet been described; Unsymmetrically basic substituted diacetyl-bis-thiosemicarbazones have not yet become known.



  The particular advantage of the products of the process is that, due to their advantageous properties (low toxicity), they are excellently suited for routine prophylaxis in poultry coccidiosis; while this is not the case with the previously known basic substituted bisthiosemicarbazones (cf. Nature, Vol. 206, p. 1341, right column, lines 30-34).

        The mustard oils used as starting materials are 2-diethylaminoethyl, 2-piperidinoethyl and 3-pyrrolidino-propyl mustard oil. The mustard oils are prepared in a known manner from the amines by reaction with carbon disulfide and subsequent oxidation of the dithiocarbamates that arise in the first stage with sodium chloride (NaClO2).



  The new process products can be used as medicinal products. Due to their favorable chemotherapeutic index, they are particularly suitable for the therapy and prophylaxis of poultry coccidiosis and can be used as an additive to drinking water or feed. For example, an administration of the process products from 0.0025 to 0.01 ö in the feed is well tolerated over a period of a few weeks with a sure effect.

   The following tables 2 (toxicity) and 1 (effect) show the superiority of process products 1 and II over a structurally similar asymmetrical compound (III) and the symmetrically, basic substituted bis-thio-semicarbazone IV, as can be seen from the Index at the autopsy results.



  According to Tables 1 and 2, the substances were tested in a drinking water experiment for their tolerance and therapeutic effect against coccidial infection in chicks.



  In the therapeutic drinking water experiments, the procedure was that a collective of several days old chicks (white Leghorn) were infected with 100,000 oocysts (Eimeria tenella) transported using a goiter tube. The animals that were evenly infected were then divided into individual groups of mostly 10 chicks.

   These groups were either treated together, as indicated in the table, or served as infected, untreated control groups. The infection was set in such a way that all infected control animals succumbed to appendicitis coccidiosis during the observation period; these results are therefore not listed individually. Treatment started immediately after infection for 48 hours; the solution was renewed once.

   During the period of 2 days, the test animals were able to ingest the drinking water mixed with the preparation ad libitum. During the experiment, the animals were examined for Ooc. Excretion, and it was determined whether any deaths that occurred were due to coccidiosis. or other causes.
