Verfahren zur Herstellung eines neuen Glucosides
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Epipodophyllotoxin-benzyliden-; S-D-glu- cosid (Formel I).
Erfindungsgemäss gelangt man zu Epipodophyl lotoxin-benzyliden-iS-D-glucosid, indem man 4'-Demethyl -epipodophyllotoxin-ss-D-benzyliden-glucosid (Formel II) mit Diazomethan methyliert.
Erfindungsgemäss kann das angeführte Verfahren so ausgeführt werden, dass man 4'-Demethyl-epipodophyl lotoxin--D-benzyliden-glucosid in einem inerten Lösungsmittel, wie z. B. Dioxan, dem etwas Methanol zugegeben wird, löst und die Lösung bei 0 bis 30 , vorzugsweise bei Zimmertemperatur, im Dunkeln mit einer Lösung von Diazomethan in Äther versetzt. Der Verlauf der Methylierung wird dünnschichtchromatographisch verfolgt. Zur Aufarbeitung entfernt man das Lösungs- mittel im Vakuum und reinigt das erhaltene Material mittels Chromatographie.
Das als Ausgangsstoff verwendete, bisher unbekannte 4'-Demethyl-epipodophyllotoxin--D-benzyliden-glucosid kann wie folgt erhalten werden : 4'-Demethyl-epipodophyllotoxin (Formel III) wird mit Carbobenzoxychlorid zu 4'-Carbobenzoxy-4'-demethyl-epipodophyllotoxin (Formel IV) umgesetzt (siehe Beispiel 1), letzteres mit x-Acetobromglucose in Gegenwart von Zinkoxyd umgesetzt, aus dem entstandenen Tetra-O-acetyl-4'-carbobenzoxy-4'-demethyl-epipodo- phyllotoxin--D-glucosid (Formel V) die Carbobenzoxygruppe entfernt (siehe Beispiel 2), das auf diese Weise erhaltene Tetra-O-acetyl-4'-demethyl-epipodophyllotoxin- -e-D-glucosid (Formel Vl)
durch Methanolyse in Gegenwart von Zinkacetat entacetyliert (siehe Beispiel 3) und das so erhaltene 4'-Demethyl-epipodophyllotoxin-3-D- -glucosid (Formel VII) schliesslich mit Benzaldehyd in Gegenwart eines Katalysators, z. B. wasserfreies Zinkchlorid, umgesetzt (siehe Beispiel 4).
Epipodophyllotoxin-benzyliden--D-glucosid besitzt in vitro eine hohe cytostatische Wirkung an Mastocytomzellen und Fibroblastenkulturen. Ferner zeichnet sich die genannte Verbindung durch eine starke Wirksamkeit gegenüber experimentellen Tumoren, insbesondere der Mäuseleukämie L-1210, aus.
Die neue Verbindung kann als Arzneimittel allein oder in entsprechenden Arzneiformen für orale, enterale oder parenterale Verabreichung verwendet werden.
Zwecks Herstellung geeigneter Arzneiformen wird diese mit organischen oder anorganischen, pharmakologisch indifferenten Hilfsstoffen verarbeitet. Als Hilfsstoffe werden verwendet z. B. für Tabletten und Dragées :
Milchzucker, Stärke, Talk, Stearinsäure usw. für Sirupe :
Rohrzucker-, Invertzucker-, Glucoselösungen u. a. für Injektionspräparate :
Wasser, Alkohole, Glycerin, pflanzliche Öle und dgl. für Suppositorien : natürliche oder gehärtete Öle, Wachse u. a. mehr.
Zudem können die Zubereitungen geeignete Konser vierungs-, Stabilisierungs-, Netzmittel, Lösungsvermittler, Süss-und Farbstoffe, Aromantien usw. enthalten.
In den nachfolgenden Beispielen, welche die Ausführung des Verfahrens erläutern, den Umfang der Erfindung aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden. Die Schmelzbzw. Zersetzungspunkte sind auf dem Kofler-Block bestimmt.

Beispiel 1
4'-Carbobenzoxy-4'-demethyl-epipodophyllotoxin
60 g staubfein pulverisiertes 4'-Demethyl-epipodophyllotoxin werden in 1000 ml wasserfreiem Äthylen- chlorid suspendiert und nach Versetzen mit 19 ml abs.
Pyridin auf-10 abgekühlt. Unter Rühren und Feuchtigkeitsausschluss wird bei-10 innert 2, 5 Stunden eine Lösung von 34 g Chlorameisensäure-benzylester in 100 ml Äthylenchlorid zugetropft und danach eine weitere halbe Stunde reagieren gelassen. Anschliessend wird die Reaktionslösung mit Wasser gewaschen, die organische Phase über Natriumsulfat getrocknet, im Vakuum eingedampft und der Rückstand im Hochvakuum bei 70-80 getrock- net. Kristallisation des Rohproduktes aus Aceton-Äther und dann zweimal aus Methanol liefert 4'-Carbobenzoxy -4'-demethyl-epipodophyllotoxin, v.
Doppelschmelzpunkt 117-119 /202-205 . Durch Trocknen im Hochvakuum zuerst bei 95-110 und dann bei 130 oder Kristallisation aus Aceton-Äther wird die lösungsmittelfreie Form vom Smp. 201-204, [α]21D = -43,9 (c = 0, 535) in CHC13, erhalten.
Beispiel 2 Tetra-O-aceryS-4'-demetllyl-e pipodophyllotoxin-l- -D-glucosid
8, 56 g 4'-Carbobenzoxy-4'-demethyl-epipodophyllo- toxin werden in 55 ml Acetonitril bei 65 bis zur Lösung verrührt, 3, 92 g trockenes Zinkoxyd zugesetzt und nach Zugabe von 20, 0 g a-Acetobromglucose unter Feuchtigkeitsausschluss bei 65 verrührt. Nach je 10 Minuten wird eine Probe entnommen und im Dünnschichtchroma- togramm (Kieselgel/Chloroform + 10% Aceton) untersucht.
Nach vollständigem Umsatz der Acetobromglucose (ca. 40-60 Minuten nach Beginn der Reaktion) kühlt man auf Zimmertemperatur ab, verdünnt mit 50 ml Chloroform, filtriert nicht umgesetztes Zinkoxyd ab und dampft das Filtrat im Vakuum bei 40 auf ein Volumen von ca. 20 ml ein. Dieses Konzentrat wird in 250 ml Chloroform gelöst, die Chloroformphase zweimal mit je 300 ml Wasser-Methanol (9 : 1) gewaschen und die Chloroformschicht nach Trocknung über Natriumsulfat im Vakuum eingedampft.
Den getrockneten Rückstand extrahiert man viermal mit je 70 ml siedend-heissem Äthanol-Wasser-1 : 3-Gemisch. Den Äthanol-unlöslichen Anteil chromatographiert man an der 50fachen Menge Kieselgel mit Chloroform-Aceton (95 : 5) als Elutionsmittel.
Die Glucosidfraktionen werden vereinigt und ergeben nach Trocknung im Hochvakuum bei 80 , 7 g Tetra-O- -acetyl-4'-carbobenzoxy-4'-demethyl-epipodophyllotoxin -, 3-D-glucosid. Zur Abspaltung des Carbobenzoxyrestes werden 7 g dieser Verbindung in 250 ml Äthanol-Aceton- -4 : 1 gelöst, mit 10 ml Eisessig und 2 g Palladium-Kohle (mit 10% Pd) versetzt und bei 20 hydriert. Danach filtriert man vom Katalysator ab, wäscht diesen mit warmem Chloroform-Methanol-Gemisch (1 : 1) nach und dampft das Filtrat im Vakuum ein. Den Rückstand kristallisiert man aus Methanol und dann mehrmals aus Äthanol.
Tetra-O-acetyl-4'-demethyl-epipodophyllotoxin- --D-glucosid kristallisiert in feinen Nadeln vom Smp.
224-227 , [a] 2lD = 61, 0 (c = 0, 356) in Chloroform.
Beispiel 3 4'-Demethyl-e pipodoph yllotoxin--D-gl ucosid
2, 0 g des nach Beispiel 2 erhaltenen Tetra-0-acetyl- -4'-demethyl-epipodophyllotoxin-0-D-glucosids und 1 g wasserfreies Zinkacetat werden in 30 ml absolutem Methanol 25 Stunden unter Rückfluss erwärmt. Anschliessend wird der entstandene weisse Niederschlag durch Zusatz von wenigen ml Eisessig und leichtem Erwärmen in Lösung gebracht, das Lösungsmittel im Vakuum bei 40 entfernt und der Rückstand in 50 ml Chloroform- Butanol (4 : 1) aufgenommen.
Die organische Phase wäscht man zweimal mit je 10 ml Wasser, dampft nach Trocknung über Natriumsulfat im Vakuum ein und chromatographiert den Rückstand an Kieselgel. Isopropyl acetat-Methanol (9 : 1), wassergesättigt, eluiert zuerst unpolare Anteile, dann reines 4'-Demethyl-epipodophyllotoxin--D-glucosid.
Die einzelnen Fraktionen werden im Dünnschichtchromatogramm auf Kieselgelplatten mit dem Fliessmittel Isopropylacetat-Methanol (8 : 1), was sergesättigt. untersucht und die Glucosidfraktionen vereinigt und zweimal aus Methanol kristallisiert. 4'-De- methyl-epipodophyllotoxin-ss-D-glucosid schmilzt bei 222 bis 230 , eine andere Modifikation bei 262-264 , [ap'o = -88 (c = 0, 507) in Methanol.
Beispiel 4 4'-Demethyl-e pipodophyllotoxin--D-ben yliden-gf ocosid
I g getrocknetes 4'-Demethyl-epipodophyllotoxin-p-D- -glucosid wird in 20 ml reinem Benzaldehyd gelöst und nach Zugabe von 0, 5g wasserfreiem Zinkchlorid unter Feuchtigkeitsausschluss während 5-6 Stunden bei 20 auf der Maschine geschüttelt. Das Fortschreiten der Reaktion wird laufend mittels Dünnschichtchromatographie verfolgt. Dazu eignen sich Kieselgelplatten u. Chloroform mit 6% Methanol als Fliessmittel.
Zur Sichtbarmachung der Substanzen besprüht man die Platten mit einer lSOigen Lösung von Cer (IV)-ammoniumnitrat in 50% iger Schwefelsäure und erwärmt anschliessend auf 100-120 . Zur Aufarbeitung des Kondensationsproduktes versetzt man die klare, rotbraun gefärbte Reaktionslösung mit Chloroform und schüttelt mit Wasser aus. Die wässrige Phase wird 2mal mit Chloroform nachextrahiert.
Man vereinigt sämtliche Chloroformphasen, wäscht 2mal mit Wasser nach, trocknet über Natriumsulfat und verdampft die Lösungsmittel im Vakuum. Der anfallende ölige Rückstand wird zu Entfernung von immer noch anhaftendem Benzaldehyd mit Pentan verrieben, bis ein pulvriges Produkt erhalten wird. Zur weiteren Reinigung nimmt man das Benzylidenderivat in 10 ml Aceton auf und tropft die Acetonlösung unter Rühren zu vorgelegten 100 ml Pentan, wobei eine hellgelbe bis weiss gefärbte Fällung erhalten wird. Das Rohprodukt kann auch durch Chromatographie an Kieselgelsäulen gereinigt werden.
Dazu filtriert man eine möglichst konzentrierte Lösung von I g des rohen Benzylidenderivates in einem Gemisch von Chloroform + 2% Methanol auf eine Säule aus 200 g Kieselgel und eluiert mit dem gleichen Lösungsmit- telgemisch. Die dünnschichtchromatographisch einheitlichen Fraktionen werden vereinigt und aus Aceton-Pentan umgefällt. 4'-Demethyl-epipodophyllotoxin-e-D-ben- zyliden-glucosid wird als weisses Pulver vom Schmelz- punkt 182-185 erhalten. Die optischen Drehwerte betragen [x] = n=-99 in Methanol und-104 in Chloroform.
Beispiel 5
Epipodophyllotoxin-benzylident-ss-D-glucosid Durch MethS lierung von 4'-Demethyl-epipodopillotoxin -benzyliden-ss-D-glucosid mit Diazomethan
400 mg 4-Demethyl-epipodophyllotoxi n-benzyl iden-p- -D-glucosid werden in 8 ml Dioxan-Methanol (9. 1) gelöst und portionenweise eine ätherische Diazomethanlösung zugetropft. Der Verlauf der Methylierung (die im Dunkeln bei Raumtemperatur erfolgt) wird dünnschicht- chromatographisch auf Kieselgelplatten mit Cyclohexan Methyläthylketon (1 : 1) + 2% Methanol als Fliessmit- tel verfolgt. Die Sichtbarmachung der Flecken erfolgt durch Besprühen mit einer 0, 2%igen Cer (IV)-sulfat-Lö- sung in 50gciger Schwefelsäure und Erwärmen auf 130 .
Erst nach 12 Tagen ist im Dünnschichtchromatogramm nur noch 1 Hauptfleck zu erkennen. Zur Aufarbeitung entfernt man das Lösungsmittel im Vakuum. Das erhaltene Material wird durch Chromatographie an der 500fachen Menge Kieselgel ( < Merck gereinigt. Als Elutionsmittel wird durchgehend Cyclohexan-Methyläthylketon-(10 : 5) + 2% Methanol verwendet. Die dünn schichtchromatographisch einheitlichen Fraktionen werden in 2 ml Chloroform gelöst und mit 50 ml n-Hexan ausgefällt. Epipodophyllotoxin-benzyliden-jss-D-glucosid wird in Form eines hellgelben amorphen Pulvers vom Smp. 166-169 erhalten.
Process for the production of a new glucoside
The present invention relates to a process for the preparation of epipodophyllotoxin benzylidene; S-D-glucoside (Formula I).
According to the invention, epipodophyl lotoxin-benzylidene-iS-D-glucoside is obtained by methylating 4'-demethyl-epipodophyllotoxin-ss-D-benzylidene-glucoside (formula II) with diazomethane.
According to the invention, the cited method can be carried out so that 4'-demethyl-epipodophyl lotoxin - D-benzylidene-glucoside in an inert solvent, such as. B. dioxane, to which a little methanol is added, and the solution is added at 0 to 30, preferably at room temperature, in the dark with a solution of diazomethane in ether. The course of the methylation is followed by thin layer chromatography. For working up, the solvent is removed in vacuo and the material obtained is purified by means of chromatography.
The previously unknown 4'-demethyl-epipodophyllotoxin - D-benzylidene-glucoside used as the starting material can be obtained as follows: 4'-demethyl-epipodophyllotoxin (formula III) is converted to 4'-carbobenzoxy-4'-demethyl- epipodophyllotoxin (formula IV) implemented (see example 1), the latter reacted with x-acetobromoglucose in the presence of zinc oxide, from the resulting tetra-O-acetyl-4'-carbobenzoxy-4'-demethyl-epipodophyllotoxin - D-glucoside (Formula V) removed the carbobenzoxy group (see Example 2), the tetra-O-acetyl-4'-demethyl-epipodophyllotoxin- eD-glucoside obtained in this way (formula Vl)
deacetylated by methanolysis in the presence of zinc acetate (see Example 3) and the 4'-demethyl-epipodophyllotoxin-3-D- glucoside (formula VII) thus obtained is finally treated with benzaldehyde in the presence of a catalyst, e.g. B. anhydrous zinc chloride, implemented (see Example 4).
Epipodophyllotoxin benzylidene - D-glucoside has a high cytostatic effect on mastocytoma cells and fibroblast cultures in vitro. The compound mentioned is also distinguished by a strong activity against experimental tumors, in particular mouse leukemia L-1210.
The new compound can be used as a medicament alone or in appropriate dosage forms for oral, enteral or parenteral administration.
In order to produce suitable dosage forms, it is processed with organic or inorganic, pharmacologically indifferent auxiliary substances. As auxiliaries are used z. B. for tablets and dragees:
Lactose, starch, talc, stearic acid, etc. for syrups:
Cane sugar, invert sugar, glucose solutions and the like a. for injection preparations:
Water, alcohols, glycerine, vegetable oils and the like. For suppositories: natural or hydrogenated oils, waxes and the like. a. more.
In addition, the preparations can contain suitable preservatives, stabilizers, wetting agents, solubilizers, sweeteners, colorants, flavorings, etc.
In the following examples, which illustrate the implementation of the process but are not intended to restrict the scope of the invention in any way, all temperatures are given in degrees Celsius. The melting or Decomposition points are determined on the Kofler block.

4'-carbobenzoxy-4'-demethyl-epipodophyllotoxin
60 g of powdered 4'-demethyl-epipodophyllotoxin are suspended in 1000 ml of anhydrous ethylene chloride and after adding 19 ml of abs.
Pyridine cooled to -10. With stirring and exclusion of moisture, a solution of 34 g of benzyl chloroformate in 100 ml of ethylene chloride is added dropwise at -10 within 2.5 hours and then allowed to react for a further half an hour. The reaction solution is then washed with water, the organic phase is dried over sodium sulfate and evaporated in vacuo and the residue is dried in a high vacuum at 70-80. Crystallization of the crude product from acetone-ether and then twice from methanol gives 4'-carbobenzoxy -4'-demethyl-epipodophyllotoxin, v.
Double melting point 117-119 / 202-205. By drying in a high vacuum first at 95-110 and then at 130 or crystallization from acetone-ether, the solvent-free form of m.p. 201-204, [α] 21D = -43.9 (c = 0.535) in CHCl3, receive.
Example 2 Tetra-O-aceryS-4'-demetllyl-e pipodophyllotoxin-1- -D-glucoside
8.56 g of 4'-carbobenzoxy-4'-demethyl-epipodophyllo-toxin are stirred in 55 ml of acetonitrile at 65 to dissolve, 3.92 g of dry zinc oxide are added and, after the addition of 20.0 g of α-acetobromoglucose, with exclusion of moisture 65 stirred. After every 10 minutes a sample is taken and examined in a thin-layer chromatogram (silica gel / chloroform + 10% acetone).
After complete conversion of the acetobromoglucose (approx. 40-60 minutes after the start of the reaction), the mixture is cooled to room temperature, diluted with 50 ml of chloroform, unreacted zinc oxide is filtered off and the filtrate is evaporated in vacuo at 40 to a volume of approx. 20 ml one. This concentrate is dissolved in 250 ml of chloroform, the chloroform phase is washed twice with 300 ml of water-methanol (9: 1) each time and, after drying over sodium sulfate, the chloroform layer is evaporated in vacuo.
The dried residue is extracted four times with 70 ml of boiling-hot ethanol / water 1: 3 mixture each time. The ethanol-insoluble fraction is chromatographed on 50 times the amount of silica gel with chloroform-acetone (95: 5) as the eluent.
The glucoside fractions are combined and, after drying in a high vacuum at 80.7 g of tetra-O-acetyl-4'-carbobenzoxy-4'-demethyl-epipodophyllotoxin-, 3-D-glucoside. To split off the carbobenzoxy radical, 7 g of this compound are dissolved in 250 ml of ethanol-acetone -4: 1, 10 ml of glacial acetic acid and 2 g of palladium-carbon (with 10% Pd) are added and the mixture is hydrogenated at 20. The catalyst is then filtered off, washed with a warm chloroform-methanol mixture (1: 1) and the filtrate is evaporated in vacuo. The residue is crystallized from methanol and then several times from ethanol.
Tetra-O-acetyl-4'-demethyl-epipodophyllotoxin- --D-glucoside crystallizes in fine needles of m.p.
224-227, [a] 21 D = 61.0 (c = 0.356) in chloroform.
Example 3 4'-Demethyl-e pipodophylotoxin - D-glucoside
2.0 g of the tetra-0-acetyl--4'-demethyl-epipodophyllotoxin-0-D-glucoside obtained according to Example 2 and 1 g of anhydrous zinc acetate are heated under reflux in 30 ml of absolute methanol for 25 hours. The resulting white precipitate is then brought into solution by adding a few ml of glacial acetic acid and gentle heating, the solvent is removed in vacuo at 40 and the residue is taken up in 50 ml of chloroform-butanol (4: 1).
The organic phase is washed twice with 10 ml of water each time and, after drying over sodium sulphate, evaporated in vacuo and the residue is chromatographed on silica gel. Isopropyl acetate-methanol (9: 1), saturated with water, elutes first non-polar components, then pure 4'-demethyl-epipodophyllotoxin - D-glucoside.
The individual fractions are saturated by thin-layer chromatography on silica gel plates with the flow agent isopropyl acetate-methanol (8: 1). examined and the glucoside fractions combined and crystallized twice from methanol. 4'-Demethyl-epipodophyllotoxin-ss-D-glucoside melts at 222 to 230, another modification at 262-264, [ap'o = -88 (c = 0.507) in methanol.
Example 4 4'-Demethyl-e pipodophyllotoxin - D-ben ylidene-gf ocoside
I g of dried 4'-demethyl-epipodophyllotoxin-p-D- -glucoside is dissolved in 20 ml of pure benzaldehyde and, after adding 0.5 g of anhydrous zinc chloride, shaken for 5-6 hours at 20 on the machine with the exclusion of moisture. The progress of the reaction is continuously monitored by means of thin-layer chromatography. Silica gel plates and the like are suitable for this. Chloroform with 6% methanol as eluent.
To make the substances visible, the plates are sprayed with an ISO solution of cerium (IV) ammonium nitrate in 50% sulfuric acid and then heated to 100-120. To work up the condensation product, the clear, red-brown colored reaction solution is mixed with chloroform and extracted with water. The aqueous phase is extracted twice with chloroform.
All the chloroform phases are combined, washed twice with water, dried over sodium sulfate and the solvents evaporated in vacuo. The oily residue obtained is triturated with pentane to remove any benzaldehyde that is still adhering until a powdery product is obtained. For further purification, the benzylidene derivative is taken up in 10 ml of acetone and the acetone solution is added dropwise, with stirring, to 100 ml of pentane, a light yellow to white colored precipitate being obtained. The crude product can also be purified by chromatography on silica gel columns.
For this purpose, a solution, which is as concentrated as possible, of 1 g of the crude benzylidene derivative in a mixture of chloroform + 2% methanol is filtered onto a column of 200 g of silica gel and eluted with the same solvent mixture. The fractions which are uniform according to thin-layer chromatography are combined and reprecipitated from acetone-pentane. 4'-Demethyl-epipodophyllotoxin-e-D-benzylidene-glucoside is obtained as a white powder with a melting point of 182-185. The optical rotations are [x] = n = -99 in methanol and -104 in chloroform.
Example 5
Epipodophyllotoxin-benzylidene-ss-D-glucoside By methS lation of 4'-demethyl-epipodopillotoxin -benzylidene-ss-D-glucoside with diazomethane
400 mg of 4-demethyl-epipodophyllotoxi n-benzylidene-p- -D-glucoside are dissolved in 8 ml of dioxane-methanol (9.1) and an ethereal diazomethane solution is added dropwise in portions. The course of the methylation (which takes place in the dark at room temperature) is followed by thin-layer chromatography on silica gel plates using cyclohexane methyl ethyl ketone (1: 1) + 2% methanol as the eluent. The spots are made visible by spraying with a 0.2% cerium (IV) sulfate solution in 50% sulfuric acid and heating to 130.
Only after 12 days can only 1 main spot be seen in the thin-layer chromatogram. For working up, the solvent is removed in vacuo. The material obtained is purified by chromatography on 500 times the amount of silica gel (<Merck. Cyclohexane-methyl ethyl ketone (10: 5) + 2% methanol is used throughout as eluent. The fractions which are uniform by thin layer chromatography are dissolved in 2 ml of chloroform and mixed with 50 ml n-Hexane precipitated.Epipodophyllotoxin-benzylidene-jss-D-glucoside is obtained in the form of a light yellow amorphous powder with a melting point of 166-169.
