Verfahren zur Herstellung von 1-Aminoadamantan-40n
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von l-Aminoadamantan-4-on sowie auch der entsprechenden pharmakologisch verträglichen Säureadditionssalze.
Die erfindungsgemäss erhältlichen Verbindungen werden durch folgende Formel wiedergegeben:

Die erfindungsgemäss erhältlichen Verbindungen können auch in die pharmakologisch verträglichen Säureadditionssalze überführt werden. Man stellt insbesondere Salze mit folgenden Säuren her: Schwefelsäure, Salzsäure, Salpetersäure, Phosphorsäure, Milchsäure, Benzoesäure, Methansulfonsäure, p-Toluolsulfonsäure, Salicylsäure, Essigsäure, Propionsäure, Maleinsäure, Apfelsäure, Weinsäure, Zitronensäure, Cyclohexansulfamsäure, Bernsteinsäure, Nikotinsäure, Ascorbinsäure und dergleichen.
Die erfindungsgemäss erhältlichen Verbindungen stimulieren das Zentralnervensystem. Sie beleben die Stimmung und stärken die psychischen Kräfte und eignen sich deshalb zur Behandlung von Geistes- und Gemütskrankheiten.
Die zur Behandlung von Menschen und Tieren geeigneten Verbindungen können als aktive Bestandteile von konventionellen pharmazeutischen Dosierungsformen wie Tabletten, Kapseln, Elixieren, Injektionslösungen und -suspensionen oder dergleichen verabreicht werden.
Die freien Basen der Verbindungen der Formel VA können auch Salze mit Fluorkieselsäure bilden, die als Mottenschutzmittel gemäss den US-Patenten Nummern 1 915 334 und 2 075 359 dienen. Die freien Basen bilden in der Regel auch Salze mit Thiocyansäure, die mit Formaldehyd unter Bildung harzartiger Materialien kondensierbar sind, welche man als Beizinhibitoren gemäss den US-Patenten Nrn. 2 425 320 und 2 606 155 verwenden kann.
Die Ausgangsprodukte der Formel IIA für das erfindungsgemässe Verfahren können auf mikrobiologischem Wege nach folgendem Reaktionsschema hergestellt werden:
In diesem Verfahren zur Herstellung der Ausgangsprodukte kann man ein l-Amidoadamantan (I) der oxydierenden Wirkung eines Mikroorganismsu der Gattung Sporotrichum sulfurescens, Curvularia lunata oder Rhizopus arrhizus unterwerfen, wobei man in der Regel die entsprechenden l-Amidoadamantan-4-ole und l-Amidoadamantan-3-ole der Formeln IIA und II13 erhält. Die Gattungen Sporotrichum und Curvularia gehören zur Familie der Moniliaceen der Gruppen Moniliales aus der Klasse der Deuteromyceten. Der Stamm Rhizopus gehört zur Familie der Mucoracen der Gruppe Mocorales aus der Klasse der Phycomyceten.
Spezielle Stämme, die zur Durchführung des erwähnten Oxydationsverfalrens bevorzugt werden, sind Sporotrichum sulfurescens, ATCC Nr. 7159; Curvularia lunata, ATCC Nr. 12017; und Rhizopus arrhizus ATCC Nr. 11145, die von der American Type Cultur Collection in Washington erhältlich sind. Selbstverständlich sind auch andere Stämme dieser Mikroorganismen zur Durchführung des Verfahrens geeignet.

Die Bedingungen des vorliegenden Oxydationsverfahrens entsprechen im allgemeinen den zur Durchführung von biochemischen Prozessen bekannten Verfahren, wie sie beispielsweise von Murray und Mitarbeitern in den US-Patenten Nrn. 2 602 769 und 2 735 800 beschrieben sind, wobei man hier die oxydierende Wirkung der Mikroorganismen Sporotrichum sulfurescens, Curvularia lunata oder Rhizopus arrhizus ausnutzt.
Die biologische Umwandlung kann mit einer wachsenden oder ruhenden Kultur des Mikroorganismus, mit Sporen, gewaschenen Zellen oder Enzymen des Mikroorganismus durchgeführt werden.
Die Züchtung des Mikroorganismus zur Durchführung der Oxydation erfolgt in der Regel in oder auf einem seiner Entwicklung günstigen Medium. Quellen für Stickstoff und Kohlenstoff müssen vor allem in dem Nährmedium vorhanden sein, ferner soll insbesondere eine entsprechende Zufuhr an steriler Luft während der Umwandlung aufrechterhalten werden, dies kann beispielsweise in konventioneller Weise vorgesehen werden, indem man eine grosse Oberfläche des Mediums dem Luftzutritt überlässt, oder indem man Luft durch eine submerse Kultur bläst.
Assimilierbarer Stickstoff kann in üblicher Weise zugeführt werden, beispielsweise durch Zugabe von Mais Weichwasser, Soyabohnenmehl, Hefeextrakten, Pepton, löslichem oder unlöslichem, pflanzlichem oder tierischem Protein, Lactalbumin, Casein, Molke, Rückständen der Alkoholdestillation, Aminosäuren, Nitraten und Ammoniumverbindungen, wie Ammoniumtartrat, -nitrat, -sulfat und dergleichen.
Auch die Versorgung mit Kohlenstoff kann in üblicher Weise erfolgen, beispielsweise durch Zugabe von Kohlehydraten wie Glukose, Fructose, Sucrose, Lactose, Maltose, Dextrinen, Stärken, Fleischextrakten, Peptonen, Aminosäuren, Proteinen, Fettsäuren, Glycerin, Molke und dergleichen. Diese Produkte können entweder in gereinigtem Zustand oder in Form von Konzentraten wie Molkekonzentraten, Corn steep liquor, Maische und dergleichen verwendet werden; auch Gemische dieser Produkte sind verwendbar.
Verschiedene der vorstehend genannten Kohlenstoffquellen können auch als Stickstoffquelle dienen.
Das Medium hat vorzugsweise vor der Inoculierung einen pH-Wert zwischen etwa 4 und 7, obgleich auch höhere oder niedrigere pH-Werte verwendet werden können. Zur Züchtung des Mikroorganismus wird im allgemeinen eine Temperatur zwischen etwa 25 und 320 C bevorzugt, jedoch kann auch bei höheren oder niedrigeren Temperaturen innerhalb eines relativ weiten Temperaturbereichs gearbeitet werden.
Das Substrat der Formel I kann dem Medium während der Züchtungsperiode des Mikroorganismus auf einmal oder portionsweise zugegeben werden. Ferner kann der Zusatz vor oder nach der Sterilisierung oder Inoculierung erfolgen, wobei durch Steuerung von pH Wert und/oder Temperatur für die Stabilität des Substrats gesorgt werden muss. Der bevorzugte, jedoch nicht ausschliessliche Konzentrationsbereich des Substrats im Medium liegt bei etwa 0,1 bis 10 g pro Liter. Das Substrat kann dem Medium in beliebiger, geeigneter Weise zugesetzt werden, bevorzugt derart, dass eine grosse Oberfläche des Substrats der oxydierenden Wirkung des Mikroorganismus ausgesetzt wird, was beispielsweise erfolgen kann durch Lösen des Substrats in einem organischen Lösungsmittel und Zumischen der Lösung zu dem Kulturmedium, oder durch Zugabe von feinen Teilchen des Substrats, z.
B. mikronisierten Teilchen, von denen vorzugsweise 90 Gew.% kleiner als 20 Mikron sind, in Form eines trockenen Pulvers oder in Form einer wässrigen Suspension. Bei Verwendung einer wässrigen Suspension empfiehlt sich der Zusatz von Dispergier- oder Suspendiermitteln.
Die Temperatur während der Fermentation ist zweckmässig der bei der Züchtung des Mikroorganismus verwendeten Temperatur gleich oder ähnlich. Sie muss vor allem innerhalb eines solchen Bereichs gehalten werden, dass Lebensfähigkeit, aktives Wachstum oder Enzymen aktivität des Mikroorganismus aufrecherhalten werden; der Temperaturbereich von 20 bis 350 C wird hier speziell bevorzugt. Der pH-Wert liegt im allgemeinen vorzugsweise bei etwa 4 bis 8 während des Wachstums der Mikroorganismen und während der biochemischen Umwandlung. Bei säureempfindlichen Substraten sollte der pH-Wert während der Fermentation vorzugsweise jedoch oberhalb 7 liegen. Die Belüftung kann durch Oberflächenkultur oder vorzugsweise durch submerse Fermentationsbedingungen in an sich bekannter Weise erfolgen.
Die zur Oxydation des Substrats durch das Enzymsystem der Mikroorganismen benötigte Zeit kann beträchtlich schwanken. Gewöhnlich liegt die Verfahrensdauer bei etwa 2 bis 120 Stunden, wobei hiermit jedoch keine Grenzwerte angegeben werden. Gewöhnlich werden nach 72 Stunden befriedigende Ergebnisse erzielt. Der Verlauf der biologischen Umwandlung und die Beendigung des Verfahrens werden zweckmässig durch Papierchromatographie, Dampfphasenchromatographie oder Dünnschichtchromatographie (Heftman, Chromatography (1961) Reinhold Publishing Co., New York, New York) verfolgt.
Die Oxydation des Substrats kann ferner auch unter aeroben Bedingungen erfolgen, indem man das Substrat der oxydierenden Wirkung der aus dem Mikroorganismen isolierten oxydierenden Enzyme, der Wirkung von Sporen des Mikroorganismus oder der Wirkung isolierter Zellen des Mikroorganismus unterwirft. Isolierte Enzympräparate können nach dem Verfahren von Zuidweg und Mitarbeitern, Biochim. Biophy. Acta, 58, 131-133 (1962) hergestellt werden. Mit Sporen kann die Oxydation nach dem Verfahren der US-Patente Nrn. 3 031 379 und 3 031 382 erfolgen. Die Abtrennung gewaschener Zellen aus dem Fermentationsmedium ist ebenfalls bekannt, vergleiche z. B. US-Patent Num mer 2831 789.
Unter oxydierender Wirkung oder Oxydationswirkung wird in vorliegender Beschreibung die enzymatische Wirkung einer wachsenden oder ruhenden Kultur des Mikroorganismus oder von Sporen, gewaschenen Zellen oder isolierten Enzymen des Mikroorganismus verstanden, welche die Einführung von Sauerstoff in das Molekül des Substrats unter aeroben Fermentationsbedingungen bewirkt.
Nach beendeter Fermentation können die oxydierten bzw. mit einer neuen Sauerstoff-Funktion versehenen Produkte IIA und IIB in konventioneller Weise aus der Fermentationsbrühe isoliert werden. Hierzu kann man beispielsweise das gesamte Medium mit einem mit Wasser nicht mischbaren organischen Lösungsmittel wie Methylenchlorid, Chloroform, Tetrachlorkohlenstoff, Äthylenchlorid, Trichloräthylen, Äther, Amylacetat, Benzol oder dergleichen extrahieren, oder man kann Brühe und Mycel trennen, z. B. durch Zentrifugieren oder Filtrieren, und dann getrennt mit geeigneten Lösungsmitteln extrahieren. Das Mycel kann sowohl mit wassermischbaren wie auch mit Wasser nichtmischbaren Lösungsmitteln extrahiert werden.
In Fällen, in welchen das Mycel wenig oder kein Produkt enthält, genügt ge wöhnlich blosses Waschen mit Wasser, und das Waschwasser wird dann der Fermentationsbrühe zugesetzt. Die vom Mycel befreite Brühe kann mit mit Wasser nichtmischbaren Lösungsmitteln, beispielsweise den vorstehend genannten, extrahiert werden. Die Extrakte werden vorzugsweise vereinigt, getrocknet, beispielsweise mit wasserfreiem Natriumsulfat, und das Lösungsmittel wird in konventioneller Weise entfernt, beispielsweise durch Abdampfen oder durch Destillation bei Normal- oder Unterdruck.
Die Produkte können ferner aus der Brühe durch Adsorption an Holzkohle, die nachher mit einem polaren organischen Lösungsmittel wie Methanol, Äthanol, Aceton, Äthylacetat oder dergleichen eluiert wird, gewonnen werden.
Die Produkte IIA und IIB, die entweder durch Extraktion oder Eluieren erhalten wurden, können in konventioneller Weise gereinigt werden, beispielsweise durch Chromatographieren und/oder Kristallisieren und dergleichen.
Die so erhaltenen 1-Amidoadamantan-4-ole werden erfindungsgemäss gemäss folgendem Reaktionsschema in die neuen Verbindungen überführt:

Die 1-Amidoadamantan-4-ole der Formel IIA werden erfindungsgemäss unter Bildung der entsprechenden 1 -Amidoadamantan-4-one der Formel IVA oxydiert; hierbei kann in zur Oxydation sekundärer Hydroylgruppen zu Ketonen in an sich bekannter Weise vorge gangen werden, vergleiche z. B. Fieser und Fieser, Natural Products Related to Phenathrene, 3. Auflage, S. 127-129, 193 und 194, Reinhold Publishing Corporation, New York, New York. Dabei wird z.
B. die Verbindung IIA in einem inerten organischen Lösungsmittel wie Aceton, Benzol, Methylenchlorid, t-Butanol oder dergleichen gelöst, dann mit wässriger Chromsäure, Kaliumpermanganat, t-Butylhypochlorit oder ähnlichen Oxydationsmitteln oxydiert, wobei die Hydroxylgruppe in eine Ketogruppe überführt wird. Das so erhaltene Keton IVA kann in konventioneller Weise aus dem Reaktionsgemisch isoliert und gereinigt werden. Beispielsweise kann man mit einem organischen Lösungsmittel, wie Methylenchlorid extrahieren, worauf aus einem geeigneten Lösungsmittel, wie z. B. Aceton, Ben zol, Methylchlorid oder dergleichen, umkristallisiert wird.
Die erhaltenen Verbindungen der Formel IVA werden anschliessend, z. B. nach an sich bekannter Weise, siehe z. B. Chem. Ber. 93, 229 (1960) in 1-Amino- adamantan-4-on (VA) überführt. Die Verbindungen der Formel IVA werden dazu vorzugsweise mit einer wässrigen starken Base in Natrium- oder Kaliumhydroxyd bei Rückflusstemperatur hydrolysiert. Die so erhaltenen freien Amine werden gewöhnlich durch Extraktion mit einem geeigneten organischen Lösungsmittel wie Me thylenchlorid, Äther, Benzol, Hexan oder Gemischen dieser Lösungsmittel aufgearbeitet. Das freie Amin kann ferner durch Kristallisation aus einem Lösungsmittel, wie Äther, Äther-Hexan, Benzol oder dergleichen, gereinigt werden. Die Hydrolyse kann jedoch auch mit einer starken Säure erfolgen. In diesen Fällen erhält man das Säureadditionssalz des Amins.
Die freien Amine der Formel VA können in an sich bekannter Weise in ihre pharmakologisch verträglichen Säureadditionssalze überführt werden, indem man das Amin mit einer geeigneten Säure, beispielsweise einer der vorstehend aufgeführten Säuren, in Gegenwart eines inerten Lösungsmittels, wie Methanol, Äthanol, Diäthyl äther, Athylacetat oder dergleichen, umsetzt.
Die Verbindungen der Formel VA können ausserdem in übliche Carbonylderivate wie Oxime, Hydrazone, Semicarbazone, cyclische Alkylenketale und dergleichen in an sich bekannter Weise überführt werden. Beispielsweise kann man die Carbonylgruppe durch Umsetzung mit einem Alkan-1,2-diol oder Alkan-1,3-diol mit bis zu 8 Kohlenstoffatomen ketalysieren, wie Äthylen-, Propylen-, Trimethylen-, 2,3-Butylen-, 2,4-Pentylen-, 4-Methyl-1,2 pentylen, 1,3-Hexylen-, 1,2-Heptylen-, 3,4 Heptylen-, 1,3-Octylenglycol oder dergleichen, vorzugsweise in einem organischen Lösungsmittel, wie Benzol, Toluol, Xylol, Methylenchlorid oder dergleichen, in Gegenwart eines sauren Katalysators wie p-Toluolsulfon- säure. Die Umsetzung erfolgt im allgemeinen bei Temperaturen zwischen etwa 20 und 2000 C, vorzugsweise zwischen etwa 40 und 1500 C.
Die Reaktionszeit ist gewöhnlich nicht kritisch und kann im allgemeinen zwischen etwa 1 und 48 Stunden liegen, je nach der angewandten Temperatur. Die Ketonderivate, wie Oxime und dergleichen, können nach bekannten Methoden erhalten werden, wie sie z. B. in Identification of Organic Compounds, Shriner und Fusion,, John Wiley and Sons, Inc., New York, New York, beschrieben sind.
Die cyclischen Alkylenketale, Oxime und anderen Ketonderivate der erfindungsgemäss erhältlichen Verbindung können gegebenenfalls wieder hydrolysiert werden, beispielsweise unter Verwendung wässriger Säuren oder Basen, wobei man die freien Ketoverbindungen erhält.
Präparat 1
Oxydation von l-Acetamidoadamantan
Ein Medium wurde hergestellt aus 40 g Mais Weichwasser (60S Feststoffgehalt) und 20 g handels üblicher Dextrose, worauf mit Leitungswasser auf ein Liter aufgefüllt und der pH-Wert auf 4,8 bis 5,0 eingestellt wurde. Ein ml Specköl wurde als Antischaummittel zugesetzt. 100 Liter dieses Mediums wurden sterilisiert und mit einer 72stündigen vegetativen Kultur von Sporotrichum sulfurescens, ATCC Nr. 7159 inoculiert und bei etwa 280 C unter einer Belüftung mit etwa 5 Liter pro Minute und Rühren mit etwa 300 Umdrehungen pro Minute bebrütet. Nach etwa 43 Stunden, oder sobald ein mässiges bis starkes Wachstum des Mycels beobachtet wurde, wurde eine Lösung von 55 g l-Acetamidoadamantan in etwa 500 ml N,N-Dimethylformamid zugesetzt.
Nach weiteren 72 Stunden Bebrütung wurden 1500 g Diatomeenerde (Celite) zugesetzt, und Gärbrühe und Mycel wurden durch Filtration getrennt.
Die so erhaltene Brühe wurde durch eine Kolonne mit 3 kg granulierter Kohle (Pittsburg Chemical Co.) geführt. Die Herstellung der Säule erfolgte, indem man die granulierte Kohle in eDtionisit rtem Wasser auf 80-900C erwärmte, abkühlte und feucht die Säule damit packte. Die Eluierung erfolgte dann mit 50 1 Methanol. Das die gewünschten Produkte enthaltende Eluat wurde bei vermindertem Druck zu einem kleinen Volumen eingeengt, in ein offenes Gefäss überführt und auf dem Dampfbad zur Trockne eingedampft. Der Rückstand wurde sorgfältig mit etwa 500 g Silikagel versetzt und auf einem Tablett an der Luft getrocknet. Die so erhaltene Masse wurde dann auf eine feucht gepackte (Athylacetat) Säule mit Silikagel aufgebracht.
Die Säule wurde zunächst mit 6 Litern Äthylacetat und dann mit von 2 bis 18X steigende Mengen Methanol enthaltendem Äthylacetat eluiert. Man sammelte Fraktionen von je 1 Liter.
Die durch Dünnschichtchromatographie ermittelten Fraktionen, die das l-Acetamidoadamantan-4-ol enthielten, wurden vereinigt und das Lösungsmittel wurde bei vermindertem Druck abgedampft. Der Rückstand wurde aus Aceton umkristallisert, wobei man 19,19 g l-Acetamidoadamantan-4-ol vom Schmelzpunkt 173 bis 1750 C erhielt. IR- und kernmagnetisches Resonanzspektrum bestätigten die zugeordnete Struktur.

C 68,86; H 9,15; N 6,69%
Gefunden: C 68,87; H 9,22; N 6,83%
Die ebenfalls durch Dünnschichtchromatographie bestimmten Fraktionen, die 1 -Acetamidoadamantan- 3-ol enthielten, wurden ebenfalls vereinigt, das Lösungsmittel wurde entfernt und der Rückstand wurde aus Aceton umkristallisiert, wobei man 5,08 g dieser Verbindung vom Schmelzpunkt 223-2250C erhielt. IR Spektrum und kernmagnetisches Resonanzspektrum bestätigten die zugeordnete Struktur.
Analyse: Berechnet für C1 2H19 NO:
C 68,86; R 9,15; N 6,69%
Gefunden: C 69,00; H 8,11; N 6,82%
Die andern Fraktionen, die Gemische der gewünschten Produkte enthielten, und die beim Umkristallisieren erhaltenen Mutterlaugen wurden nochmals an Silikagel chromatographiert, wobei man weitere 4,23 g l-Acetamidoadamantan-4-ol und 2,30 g l-Acetamidoadamantan-3-ol erhielt.
In gleicher Weise können Rhizopus arrhizus, ATCC Nr. 11145 oder Curvularia lunata, ATCC Nummer 12017 anstelle von Sporotrichum sulfurescens verwendet werden, wobei die gleichen Produkte erhalten werden.
In gleicher Weise können nach der Arbeitsweise von Präparat 1 andere l-Amidoadamantane der Formel I zu den entsprechenden 4-Olen und 3-Olen der Formel IIA und IIB umgesetzt werden, beispielsweise: 1 -Formamidoadamantan zu l-Formamido- adamantan-4-ol und 1 -Formamidoadamantan-3 -ol, l-Benzamidoadamantan zu l-Benzamido- adamantan-4-ol und 1-Benzamidoadamantan-3-ol, l-Propionamidoadamantan zu l-Propionamida- adamantan-4-ol und 1-Propionamidoadamantan-3-ol, l-Butyramidoadamantan zu l-Butyramido- adamantan-4-ol und 1 -Butyramidoadamantan-3-ol, l-Valeramidoadamantan zu l-Valeramido- adamantan-4-ol und <RTI

Präparat 2
Oxydation von l-Benzamidoadamantan
10 Liter des in Präparat 1 benutzten Mediums wurden sterilisiert und mit einer 72stündigen vegetativen Kultur von Sporotrichum sulfurescens, ATCC Nummer 7159 beimpft. Dann wurde bei etwa 280 C unter einer Belüftung von 0,5 1 pro Minute und Rühren mit 300 Umdrehungen pro Minute bebrütet. Nach 24 bis 48 Stunden, oder sobald ein mässiges bis starkes Mycelwachstum sichtbar war, wurde eine Lösung von 2 g l-Benzamidoadamantan in 20 ml N,N-Dimethylformamid der Fermentation zugesetzt. Nach weiteren 72 Stunden wurden Gärbrühe und Mycel mit 75 ml Methylenchlorid extrahiert. Die Extrakte wurden fil triert, über Natriumsulfat getrocknet und an synthetischem Magnesiumsilikat ( Florisil ), chromatographiert.
Beim Eluieren mit Skellysolve B , dem steigende Mengen Aceton zugesetzt wurden, erhielt man 0,51 g 1-Benzamidoadamantan-4-ol. Das so erhaltene Rohprodukt wurde aus Aceton- Skellysolve B umkristallisiert, wobei man 0,42 g der obigen Verbindung vom Schmelzpunkt 178-181 C erhielt.
Analyse: Berechnet für C17H21NOe:
C 75,24; H 7,80; N 5,16%
Gefunden: C 75,15; H 7,71; N 5,30S
Durch Dünnschichtchromatographie der Mutterlaugen wurde die Anwesenheit von l-Benzamidoadamantan-3-ol festgestellt.
Präparat 3
Oxydation von 1 -Acetamidoadamantan
Das Verfahren von Präparat 1 wurde in kleinerem Massstab durchgeführt unter Verwendung von 10 Litern des sterilisierten Mediums der gleichen Zusammensetzung, 4,0 g l-Acetamidoadamantan als Substrat und dem Mikroorganismus Rhizopus arrhizus ATCC Nummer 11145. Durch Papier- und Dünnschichtchromatographie der Eluate wurde die Anwesenheit von l-Acet amidoadamantan-bol und 1 -Acetamidoadamantan-3 -ol nachgewiesen.
Präparat 4
Oxydation von l-Acetamidoadamantan
Das Verfahren von Präparat 3 wurde wiederholt mit Curvularia lunata ATCC Nr. 12017 als Mikroorganismus. Auch hier konnte analytisch die Anwesenheit von l-Acetamidoadamantan-4-ol und l-Acetamidoadamantan-3-ol in den Eluaten nachgewiesen werden.
Beispiel 1
1 -Acetamidoadamantan-4-on
In ein Gemisch aus 2,45 g 1-Acetamidoadamantan- 4-ol und etwa 150 ml Aceton wurde eine Chromsäurelösung (hergestellt aus 267 g Chromsäureanhydrid, 230 ml konzentrierter Schwefelsäure und Wasser zum Auffüllen auf 1 Liter) zugetropft, bis ein geringer Über- schuss erreicht war, wobei die Temperatur unterhalb 400 C gehalten wurde. Das Gemisch wurde 5 Minuten lang gerührt, dann wurden 5,0 ml Isopropanol zugesetzt und es wurde bei vermindertem Druck auf etwa des Volums eingeengt, mit 100 ml Wasser verdünnt und mehrmals mit Methylenchlorid extrahiert.
Die vereinigten Extrakte wurden mit Wasser gewaschen, über Natriumsulfat getrocknet und zur Trockne eingedampft, wobei man 2,13 g 1-Acetamidoadamantan-4on erhielt. Eine durch Umkristallisieren aus Aceton erhaltene analysenreine Probe zeigte einen Schmelzpunkt von 176-1780 C.
Analyse: Berechnet für C12Hr7NO2:
C 69,53; H 8,27; N 6,76%
Gefunden: C 69,88; H 8,51; N 6,79%
Arbeitet man nach dem Verfahren von Beispiel 1, jedoch unter Verwendung anderer l-Amidoadamantan4-ole der Formel IIA als Ausgangsmaterial, so erhält man die entsprechenden Ketone der Formel IVA.
Beispielsweise werden unter Verwendung der im Anschluss an Präparat 1 aufgeführten Hydroxylverbindungen die folgenden Ketone erhalten:
1 -Formamidoadamantan-4-on,
1 -Benzamidoadamantan-4-on,
1 -Propionamido adamantan-4-on,
1 -Butyramidoadamantan-4-on,
1 -Valeramidoadamantan-4-on,
1 -Hexanamidoadamantan-4-on,
1 -Decanamidoadamantan-4-on,
1 -Dodecanamidoadamantan-4-on,
1 -Cyclopropancarboxamidoadamantan-4-on,
1 -Cyclohexancarboxamidoadamantan-4-on,
1 -Toluamidoadamantan-4-on,
1 -Naphthamidoadamantan-4-on,
1 -(p-Äthylbenzamido)-adamantan-4-on, l-Phenylacetamidoadamantan-4-on und
1 -Stearamidoadamantan-4-on.
Beispiel 2
1 -Aminoadamantan-4-on-hydrochlorid
Ein Gemisch aus 4,0 g 1-Acetamidoadamantan-4-on, 160 ml Äthylenglycol und 16 g Natriumhydroxyd wurde 4 Stunden lang am Rückfluss gekocht. Nach dem Abkühlen und Verdünnen mit 200 ml Wasser wurde die Lösung kontinuierlich mit Äther 8 Stunden lang extrahiert. Der Extrakt wurde über Natriumsulfat und Kaliumhydroxyd getrocknet.
Der getrocknete ätherische Extrakt wurde mit ätherischer Chlorwasserstofflösung behandelt, wobei das Aminsalz in einer Ausbeute von 2,60 g, Schmelzpunkt > 3000 C (Zersetzung) ausfiel.
Der Äther kann aus dem Extrakt auch abgedampft werden, wobei man l-Aminoadamantan-4-on erhält, das nach dem Umkristallisieren aus Äther einen Schmelzpunkt von 248-250 C (im verschlossenen Rohr, Ölbad) zeigt.
Analyse: Berechnet für C10H17NO HCl:
C 58,96; H 8,90; N 6,88; Cl 17,41 % Gefunden: C 58,79; H 8,82; N 6,73; C1 17,32%
Beispiel 3
1 -Aminoadamantan-4-on und l-Aminoadamantan-4-on-hydrochlorid
Ein Gemisch aus 300 mg l-Acetamidoadamantan- 4-on und 20 ml 0 %iger wässriger Natronlauge wurde 22 Stunden lang am Rückfluss gekocht. Das Gemisch wurde mit 10 ml Wasser verdünnt und mehrmals mit Äther extrahiert. Der ätherische Extrakt wurde über Kaliumhydroxyd getrocknet und das Lösungsmittel wurde abgedampft, wobei man 220 mg kristallines 1-Aminoadamantan-4-on erhielt. Eine aus Äther-Hexan umkristallisierte Probe schmolz im verschlossenen Rohr (Ölbad) bei 2670 C.
100 mg des so erhaltenen l-Aminoadamantan-4-ons wurden in 50 ml Äther gelöst und mit ätherischer Chlorwasserstofflösung behandelt, wobei das Hydrochlorid ausfiel, das abfiltriert und aus Methanol Methyläthylketon umkristallisiert wurde. Das Hydrochlorid besass einen Schmelzpunkt oberhalb 3000 C.
Analyse: Berechnet für CloHtgNOCl:
C 58,96; H 8,90; N 6,80%
Gefunden: C 59,07; H 9,05; N 7,21%
In analoger Weise können nach den Verfahren der Beispiele 2 und 3 andere 1-Amidoadamantane der Formel IVA in die entsprechenden 1-Aminoadamantane der Formel VA und deren Hydrochloride überführt werden.
Ferner können nach den Verfahren der Beispiele 2 und 3 andere pharmakologisch annehmbare Salze, beispielsweise die vorstehend aufgeführten, anstelle der Hydrochloride hergestellt werden.
Claims (1)
- PATENTANSPRUCHVerfahren zur Herstellung von l-Aminoadamantan 4on der Formel EMI7.1 und der entsprechenden pharmakologisch verträglichen Säureadditionssalze, dadurch gekennzeichnet, dass man in Verbindungen der Formel EMI7.2 <tb> H <tb> N-Acyl <tb> <SEP> (IIA) <tb> <SEP> OH <SEP> worin Acyl den Acylrest einer einbasischen Kohlenwasserstoffcarbonsäure mit 1 bis 18 C-Atomen bedeutet, die Hydroxylgruppe zur Ketogruppe oxydiert, wobei Verbindungen der Formel EMI7.3 <tb> N-Acyl <tb> <SEP> (IvA) <tb> <SEP> 0 <tb> entstehen, und anschliessend den Acylrest durch Hydrolyse entfernt.UNTERANSPRÜCHE 1. Verfahren gemäss Patentanspruch, dadurch gekennzeichnet, dass man erhaltene Verbindungen der Formel VA in die entsprechenden pharmakologisch verträglichen Säureadditionssalze überführt.2. Verfahren gemäss Patentanspruch, dadurch gekennzeichnet, dass man die Hydrolyse in Gegenwart wässriger starker Basen, vorzugsweise Natrium- oder Kaliumhydroxyd, ausführt.3. Verfahren gemäss Patentanspruch, dadurch gekennzeichnet, dass man die Hydrolyse in Gegenwart starker Säuren ausführt, wobei sich die entsprechenden Säureadditionssalze bilden.4. Verfahren gemäss Patentanspruch, dadurch gekennzeichnet, dass man eine erhaltene Verbindung der Formel VA durch Umsetzung mit Hydroxylamin, Hydrazinen oder Semicarbaziden in das entsprechende Oxim, die entsprechenden Hydrazone oder Semicarbazone überführt.5. Verfahren gemäss Patentanspruch, dadurch gekennzeichnet, dass man eine erhaltene Verbindung der Formel VA durch Umsetzung mit Alkan-1,2-diolen oder Alkan-1,3-diolen mit je bis zu 8 C-Atomen in die entsprechenden cyclischen Alkylenketale überführt.
