Process for the preparation of 5-amino thiazolidine compounds
The subject of the present invention is a methodological process for the preparation of S-amino-thiazolidine compounds, which was used for the preparation of valuable intermediates and especially in the first synthetic preparation of 7-amino-cephalosporanic acid and its derivatives and is particularly suitable for this peculiar synthesis .
7-Amino-cephalosporanic acid has the following formula
EMI1.1
Derivatives are primarily N-acyl compounds, in which acyl radicals in particular those of effective N-acyl derivatives of 7-amino-cephalosporanic acid, such as thienylacetyl, cyanoacetyl, chloroethylcarbamyl or phenylacetyl radical, or easily cleavable acyl radicals, such as the radical of a half-ester of carbonic acid , e.g. B. the tert-Butyloxycarbonylrest mean.
The synthesis of these compounds and their derivatives, which are important for the production of valuable drugs, is based on the idea of a 3,5-unsubstituted 2,2-disubstituted thiazolidine-4-carboxylic acid, e.g. B. a compound of formula I.

and carry out the novel synthesis according to the following formula:

The compound of formula X used as an intermediate is prepared as follows:

The 5-aminothiazolidine compounds of the formula which are valuable as intermediates

EMI4.3
treated with a phosphine and subsequently hydrolyzed. If desired, one substituent in a compound obtained can be converted into another and / or, if desired, a mixture of isomers obtained can be separated into the individual isomers.
The conversion of the azido group into a phosphinimino group by treatment with a suitable phosphine can e.g. By treating with a trialkylphosphine, e.g. B. tri-n-butylphosphine, and a triarylphosphine, e.g. B. triphenylphosphine happen; by subsequent hydrolysis, e.g. B. by treating with water, the desired amino compound is obtained.
The above reaction is carried out in the presence or absence of diluents, especially those which, if so desired, are inert towards the reactants and / or are able to dissolve them, and / or of catalytically active substances, with cooling, at room temperature or with heating, if necessary, in a closed vessel, under pressure and / or in an inert gas atmosphere.
In the compounds obtained, substituents can be converted into others by methods known per se. Acid derivatives obtained, such as esters, can be converted into the free acid without removing the acyl group, in particular an easily cleavable acyl group such as the tert-butyloxycarbonyl group, in the 3-position. So can a carbodiphenylmethoxy group under acidic conditions, e.g. B. in the presence of catalytic amounts of an acid such as trifluoroacetic acid, convert into a free carboxyl group. In a peculiar way, z. B.
a carboxy group esterified with a 2,2,2-trihaloethanol, especially 2,2,2-trichloroethanol, is converted into the free carboxyl group by means of reducing agents. Suitable reducing agents are chemical reducing agents such as the above-mentioned metals, metal alloys or metal amalgams in the presence of hydrogen-donating agents, and the like. a. also alkali metals, e.g. b. Calcium, in liquid ammonia, optionally with the addition of alcohols such as a lower alkanol. Furthermore, an ester with a 2,2,2-trihaloethanol, in particular 2,2,2-trichloroethanol, can also be treated with strongly reducing metal salts, such as chromium-II compounds, e.g. B.
Chromium (II) chloride or chromium (II) acetate preferably in the presence of aqueous media containing water-miscible solvents, such as lower alkanols, lower alkanecarboxylic acids or ethers, e.g. B.
Methanol, ethanol, acetic acid, tetrahydrofuran, dioxane, ethylene glycol dimethyl ether or diethylene glycol dimethyl ether can be converted into the free acid. The reductive cleavage of a 2,2,2-trihaloethyl ester, which is peculiar per se, can take place simultaneously with the reductive conversion of the azido group into the desired amino group.
In a compound obtained with a free carboxyl group, this can be converted into its functional derivatives, such as its esters, amides, hydrazides or azides, by methods known per se. So she can z. B. by treating with a diazo compound such as a diazo lower alkane, e.g. B.
Diazomethane or diazoethane, or with a phenyldiazoalkane, e.g. B. phenyldiazomethane or diphenyldiazomethane, or by reaction with a hydroxy compound suitable for esterification, such as e.g.
an alkanol, as well as a phenolic compound, or an N-hydroxy nitrogen compound, e.g. B. a hydroxamic acid, in the presence of an esterifying agent such as a carbodiimide, e.g. B. dicyclohexylcarbodiimide, as well as carbonyldiimidazole, or by any other known and suitable esterification process, such as reaction with a reactive ester of the hydroxy compound, especially an alcohol, and a strong inorganic acid or a strong organic sulfonic acid, if desired, in the presence of a salt-forming base, be esterified.
A functionally modified carboxy group in a compound obtained can also be converted into another functionally modified carboxy group by methods known per se, e.g.
esterified groups by transesterification, such as treatment with a hydroxy compound in the presence of a transesterification catalyst. Furthermore, esters and especially activated esters, such as. B. esters with N-hydroxy nitrogen compounds, or anhydrides, e.g. B. with chloroformic acid ester, such as methyl ester, by reaction with other hydroxy compounds such as alcohols or phenols or with ammonia, with at most secondary amines or hydrazines, can be converted into other esters or into amides or hydrazides. In an amide or hydrazide compound obtained with a hydrogen-containing nitrogen atom, this can be subsequently substituted, e.g. B. by treatment with a carboxylic acid or sulfonic acid derivative, such as with an acid halide, and / or a reactive esterified alcohol or by means of other suitable reagents; an N-unsubstituted amide can e.g.
B. can be converted into a nitrile by dehydration.
If necessary, the free amino group can temporarily, e.g. B. protected by suitable acylation.
Mixtures of isomers obtained can be prepared by methods known per se, e.g. B. by fractional crystallization or distillation, by adsorption chromatography, column or thin layer chromatography) or other methods can be separated into the individual isomers; resulting racemates can be separated by forming a mixture of diastereoisomeric salts with optically active, salt-forming agents, separating the mixture into the diastereoisomeric salts and converting the separated salts into the free compounds or into the antipodes.
The method also includes those embodiments according to which starting materials in the form of derivatives, eg. B. of salts, used or formed during the reaction.
Such starting materials are preferably used and the reaction conditions are selected such that the compounds listed at the beginning as being particularly preferred are obtained.
The starting materials used in the above process are determined in accordance with the application no.
16973/65 described method produced.
In the compounds obtained according to the invention, acyl radicals Ac are primarily those which occur in pharmacologically active N-acyl derivatives of 7-aminocephalosporanic acid, such as thienylacetyl, e.g. B. 2-Thienylacetyl-, Chloräthylcarbamyl- or Phenylacetylrest, or easily split off acyl residues, such as the remainder of a half ester of carbonic acid, z. B. the tert-butyloxycarbonyl radical, or any other suitable acyl radicals of organic acids.
The radical -X- stands in particular for the group of the formula
EMI5.1
wherein R2 and R3 represent hydrocarbon, especially aliphatic hydrocarbon radicals, such as lower alkyl, e.g. B. ethyl, n-propyl, isopropyl or preferably methyl groups, as well as aromatic, especially phenyl groups or araliphatic hydrocarbon radicals, especially phenylalkyl, z. B. benzyl or phenylethyl groups, and for functionally modified, especially esterified carboxyl groups, such as carbon-lower alkoxy, z. B. carbethoxy or carbomethoxy groups, or, when taken together, for a divalent hydrocarbon, especially aliphatic hydrocarbon radical, such as a lower alkylene, e.g. B. 1,4-butylene or 1,5-pentylene group, and a phthaloyl group, or an oxo or thiono group.
The above hydrocarbon radicals are unsubstituted or can, for. B. by lower alkyl, such as methyl or ethyl groups, lower alkoxy, such as methoxy or ethoxy groups, halogen, such as fluorine, chlorine or bromine atoms, haloalkyl, such as trifluoromethyl groups, or other suitable groups.
The group R1 stands for a free or preferably functionally modified carboxyl group, in particular an esterified carboxyl group. The latter is suitable for the esterification of carboxylic acids with hydroxy compounds of any kind, especially with aliphatic alcohols such as alkanols, especially lower alkanols, e.g. B. methanol, methanol, n-propanol or tert-butanol, cycloaliphatic alcohols such as cycloalkanols, z. B. cyclohexanol, or araliphatic alcohols such as phenylalkanols, e.g. B.
Benzyl alcohol or diphenylmethanol, as well as esterified with phenol compounds, where the above-mentioned hydroxy compounds are unsubstituted or may contain lower alkyl, lower alkoxy or trifluoromethyl groups or, in particular, halogen atoms and other groups as substituents; Halogenated lower alkanols, such as 2,2,2-trichloroethanol, are particularly suitable as substituted hydroxy compounds which esterify the carboxylic acid.
Other functionally modified carboxyl groups Rt are e.g. B. nitrogen-containing, functionally modified carboxyl groups, such as carbamyl groups, which are unsubstituted on the nitrogen atom or optionally containing lower alkyl, free esterified or etherified hydroxy, such as lower alkoxy, aralkoxy, lower alkanoyloxy or aroyloxy groups or halogen atoms, nitro or trifluoromethyl groups or containing aliphatic, aromatic, alicyclic groups araliphatic hydrocarbon radicals or heterocyclic radicals of aromatic character, such as lower alkyl, cycloalkyl, phenyl, phenyl-lower alkyl, phenyl-lower alkylidene or pyridyl radicals, as well as by free, etherified or esterified hydroxyl groups, such as the groups of this type mentioned above, by phosphorus-containing radicals, or by acyl residues such as residues of carboxylic acids, e.g. B.
Residues of half-esters or half-amides of carbonic acid or Niederalkanoyireste, or of sulfonic acids, such as arylsulfonic acids, for example phenylsulfonyl, can be mono or disubstituted, and nitrile or azidocarbonyl groups, and optionally on the nitrogen z. B.
hydrazinocarbonyl or azocarbonyl groups mono- or polysubstituted by the above-mentioned substituents of the carbamyl group.
The compounds obtained according to the invention can, as shown in the equation, be converted into 7-amino-cephalosporanic acid and its derivatives; this conversion can e.g. B. according to the registration G.Nr. 497 456, G.Nr. 497 371, G.Nr. 497 369, G.Nr. 497 457, G.Nr. 497 460, G.Nr.

After 2 hours at 0 the supernatant solution is poured off and the crystalline material is washed with 5 ml of pentane and dried; the product obtained melts at 180-181 and is identical to that obtained by the above procedure.
L-2,2-dimethyl-3-tert-butyloxycarbonyl5a-amino-thiazolidine-4-carboxylic acid 2,2,2-trichloroethyl ester, characteristic bands in the infrared absorption spectrum (in methylene chloride) at 2.85 u and 2.92.
The above-mentioned esters can be converted into the free acids as described; the L-2,2-dimethyl-3-tert-butyloxycarbonyl Sa-amino-thiazolidine-4-carboxylic acid shows in the infrared absorption spectrum (in methylene chloride) bands at 2.84u, 2.93 in, 5.76, 5, 90, 7, 7 u and 8.63 u.
