Description
Verfahren zur Herstellung von neuen Piperidinderivaten Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Piperidinderivate mit wertvollen pharmakologischen Eigenschaften. Es wurde überraschenderweise gefunden, dass Piperidinderivate der allgemeinen Formel I, EMI1.1 in welcher R1 einen aliphatischen Kohlenwasserstoffrest mit höch stens 10 Kohlenstoffatomen, die Cinnamylgruppe, eine unsubstituierte oder am Benzolkern durch Ha logen bis Atomnummer 35 oder niedere Alkyl- oder Alkoxygruppen substituierte Phenyl- (niederalkyl) -gruppe mit höchstens 12 Kohlenstoffatomen, in der die Phenylgruppe mit der niederen Alkylgruppe statt direkt auch über Sauerstoff, die Carbonylgruppe, Al kyliminogruppe oder Alkenyliminogruppe verbunden sein kann, bedeutet und R2 und R3 eine Alkylgruppe mit 2-4 Kohlenstoffatomen bedeuten, und ihre Additionssalze mit anorganischen und organischen Säuren wertvolle pharmakologische Eigenschaften, insbesondere analgetische sowie antitussive und teilweise auch zentral dämpfende Wirksamkeit bei günstigem therapeutischem Index besitzen. Die analgetische Wirksamkeit der Verbindungen der allgemeinen Formel I und ihrer Salze, z.B. der Fumarate des 1-[l-(3-Phenylpropyl)-4-propyl- 4 -piperidyl]-1 - butanon und des 1 -[-Cinnamyl-4-propyl-4-piperidyl]- 1 -buta- non, lässt sich z.B. durch Messung der durch ihre intraperitoneale oder orale Verabreichung an Mäuse bewirkten Verlängerung der Reaktionszeit bei der Reizung des Schwanzes durch Wärmestrahlung gemäss der Versuchsanordnung von H. Friebel und Cl. Reichle, Arch. exp. Path. und Pharmakol. 226, 551 (19i5) nachweisen. Gemäss den Versuchsresultaten sind die Verbindungen der allgemeinen Formel I mit der n-Propylgruppe als R5 und ihre Salze starke Analgetica, solche mit andern Gruppen R3, wie der Äthyl- und n-Butylgruppe, milde bis mittlere Analgetica. Die antitussive Wirksamkeit der obengenannten und weiterer Verbindungen der allgemeinen Formel I zeigt sich beispielsweise bei intravenöser Applikation an Katzen nach der Methode von R. Domenjoz. Arch. exp. Path. und Pharmakol. 215, 19-24 (1952). Eine weitere Methode zum Nachweis der antitussiven Wirksamkeit ist die Bestimmung der durch subcutane oder orale Verabreichung der Prüfsubstanzen bewirkten Hemmung des durch Schwefeldioxid ausgelösten Reizhustens von Meerschweinchen: in einem Vorversuch zur Auswahl der Versuchstiere werden in einer Plexiglaskammer männliche Meerschweinchen einem bei Atmosphärendruck durchströmenden SO2-CO2-Luft-Gemisch vom konstanten Mischungsverhältnis von 20 ml: 1,5 Li ter:10,5 Liter pro Minute bis zum Eintritt des Hustens bzw. maximal 120 sec. ausgesetzt. Die Beurteilung des Husteneintritts erfolgt durch Inspektion. Die mit Husten reagierenden Meerschweinchen (ca. 2/3 aller Tiere) werden in Gruppen von je 6 Tieren zusammengefasst. Solche Versuchstiergruppen erhalten ca. 24 Stunden nach dem Vorversuch die Prüfsubstanz in verschiedenen, zur Ermittlung der EDso in mg/kg geeigneten Dosen subcutan oder peroral verabreicht. Die Reizgasexposition erfolgt in gleicher Weise wie beim Vorversuch nach 30 u. nach 90 Minuten nach Applikation der Prüfsubstanzen. Die Beurteilung des Husteneintritts erfolgt wiederum durch Inspektion. Aus den bei verschiedenen Dosen ermittelten Prozentzahlen von auf SO3 nicht mehr reagierenden Tieren wird die bei 50% der Tiere den Husteneintritt verhindernde Dosis = ED30 durch graphische Interpolation mit dem Wahrscheinlichkeitsnetz Schleicher und Schüll 29872 ermittelt. Die neuen Piperidinderivate der allgemeinen Formel I und ihre pharmazeutisch annehmbaren Säureadditionssalze eignen sich als Wirkstoffe für pharmazeutische Präparate zur Linderung und Behebung von Schmerzen verschiedener Genese sowie des Hustenreizes. Die Verabreichung kann oral, rektal oder parenteral erfolgen. In den Verbindungen der allgemeinen Formel I und den zugehörigen, weiter unten genannten Ausgangsstoffen ist R1 beispielsweise durch Alkylgruppen, wie die Methyl-, Äthyl-, n-Propyl-, Isopropyl-, n-Butyl-, Isobutyl-, sek. Butyl-, n-Pentyl-, Isopentyl-, n-Hexyl-, n-Heptyl-, n-Octyl-, n-Nonyl-, oder n-Decylgruppe; durch Alkenylgruppen, wie die Allyl-, Crotyl-, l-Methylallyl- oder 2 -Methylallylgruppe; durch die Benzylgruppe, die p-Fluor-, o-, m- oder p-Chlor-, p-Brom-, 3,4-Dichlor-, p-Methyl-, p-Isopropyl-, p-Methoxy-, p-Äthoxy-, p-Isopropoxy-, 3,4 -Dimethoxy-, 3,4,5-Trimethoxy-, Benzylgruppe, die im Benzolkern unsubstituierte 2-Phenyläthyl-, 3-Phenylpropyl-, 2-Phenylpropyl-, 4-Phenylbutyl-, 2-Phenoxyäthyl-, 3-Phenoxypropyl-, 4-Phenoxybutyl-, 2-Benzoyläthyl-, 3 -Benzolpropyl-Gruppe oder durch die entsprechenden, im Benzolkern analog den vorgenannten Benzylgruppen substituierten Gruppen; durch Methyl-, Äthyl-, n-Propyl-, die in ihrer -Stellung durch die Anilino-, o-Toluidino-, m-Toluidino-, p-Toluidino-, 3,4-Xylidino- oder 2,6-Xylidino-Gruppe substituiert sind. Neue Piperidinderivate der allgemeinen Formel I und ihre Säureadditionssalze werden hergestellt, indem man auf eine Verbindung der allgemeinen Formel II, EMI2.1 in welcher R1 und R die unter Formel I angegebene Bedeutung haben, R3 die Methyl-, Äthyl- oder Propylgruppe, Y eine niedere Alkylgruppe und Z die Gruppe -COO-Y oder die Cyanogruppe bedeuten, eine Säure einwirken lässt und gewünschtenfalls aus einem erhaltenen Säureadditionssalz eine Verbindung der allgemeinen Formel I freisetzt und diese oder das zunächst erhaltene Salz in ein anderes Additionssalz mit einer anorganischen oder organischen Säure überführt. Beispielsweise lässt man auf eine Verbindung der allgemeinen Formel II konz. Mineralsäuren, wie z.B. konz. Salzsäure oder konz. Bromwasserstoffsäure oder Schwefelsäure bei Temperaturen von 600 bis 1500C einwirken, wobei die nötige Reaktionstemperatur und -dauer anhand der Kohlendioxidentwicklung leicht feststellbar ist. Für die Herstellung der Ausgangsstoffe der allgemeinen Formel II setzt man beispielsweise ein Säurehalogenid der allgemeinen Formel III, EMI2.2 in welcher Hal Chlor oder Brom bedeutet und R1 und R2 die unter Formel I angegebene Bedeutung haben, mit einer Alkalimetall- oder einer Alkoxymagnesiumverbindung eines Malonsäurediesters oder Cyanessigsäureesters der allgemeinen Formel Formel IV EMI2.3 in welcher Y, Z und R5, die unter Formel II angegebene Bedeutung haben, um, wobei man als Lösungsmittel z.B. Äther, Äthanol, Methanol oder Benzol verwendet. Gewünschtenfalls werden die nach den erfindungsgemässen Verfahren erhaltenen Piperidinderivate der allgemeinen Formel I anschliessend in üblicher Weise in ihre Additionssalze mit anorganischen und organischen Säuren übergeführt. Beispielsweise versetzt man eine Lösung eines Piperidinderivates der allgemeinen Formel 1 in einem organischen Lösungsmittel, wie Diäthyläther, Methanol oder Äthanol, mit der als Salzkomponente gewünschten Säure oder einer Lösung derselben und trennt das unmittelbar oder nach Zufügen einer zweiten organischen Flüssigkeit, wie z.B. Diäthyläther zu Methanol, ausgefallene Salz ab. Zur Verwendung als Wirkstoffe für Arzneimittel können anstelle freier Basen pharmazeutisch annehmbare Säureadditionssalze eingesetzt werden, d.h. Salze mit solchen Säuren, deren Anionen bei den in Frage komrnenden Dosierungen entweder keine oder erwünschte eigene pharmakologische Wirkung zeigen. Ferner ist es von Vorteil, wenn die als Wirkstoffe zu verwendenden Salze gut kristallisierbar und nicht oder wenig hygroskopisch sind. Zur Salzbildung mit Piperidinderivaten der allgemeinen Formel I kann z.B. Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Methansulfonsäure, Äthansulfonsäure, p-Hydroxyäthansulfonsäure, Essigsäure, Apfelsäure, Weinsäure, Citronensäure, Milchsäure, Bernsteinsäure, Fumarsäure, Maleinsäure, Benzoesäure, Salicylsäure, Phenylessigsäure, Mandelsäure, Embonsäure, 1,5-Naphthalindisulfonsäure oder Cyclohexylaminsulfonsäure verwendet werden. Das nachfolgende Beispiel erläutert die Herstellung der neuen Verbindungen der allgemeinen Formel I. Die Temperaturen sind in Celsiusgraden angegeben. Beispiel In einem Vierhalskolben werden 28,2 g Äthyl-Malonsäurediäthylester mit 3,45 g fein veteiltem Natrium 15 Stunden in 900 ml abs. Äther gerührt. Zu der dicken Suspension von Natrium-äthyl-malonsäurediäthylester werden 17,3 g 1 -(2-Phenyläthyl-4-propyl-isonipecotinoyl- chlorid-hydrochlorid zugegeben und zuerst bei Zimmertemperatur und anschliessend am Rückfluss noch eine Stunde erhitzt. Anschliessend wird mit Eis und 5N Salzsäure zersetzt, die Ätherphase abgetrennt, noch einmal mit Äther ausgezogen und dann die wässrige Phase mit konz. Ammoniak alkalisch gestellt. Es wird mit Äther ausgezogen, wobei nach dem Verdampfen das (1-Phenyl äthyl-4-propyl-4-piperidyl)-dicarbäthoxy-butanon zurückbleibt. 8,9 g des rohen oben erwähnten Produktes werden mit 20 ml konz. Salzsäure versetzt und 2 Stunden am Rückfluss erhitzt. Dann wird im Vakuum verdampft, der Rückstand in wenig Wasser aufgenommen, mit konz. Ammoniak alkalisch gestellt und mit Äther ausgezogen. Es wird getrocknet, verdampft und im Luftbad bei 0,02 Torr und 135-145 destilliert. Eventuell wird ein Silicagel in Chloroform-Methanol 97: 3 chromatographiert. Anschliessend wird die Base in üblicher Weise in das Fumarat übergeführt. Das 1-[1-(2-Phenyläthyl)-4-propyl-4 -piperidyl]-1-butanon-fumarat schmilzt bei 1760. In gleicher Weise wurden hergestellt: 1 -[1 -(3 -Phenyl-propyl)-4-propyl-4-piperidyl]- 1 -butanon -fumarat, Smp. 172-173 , 1 -(1 -Cinnamyl-4-propyl-4-piperidyl]-1 -butanon-fumarat, Smp. 178-179 und 1-[1-(2-Phenoxy-äthyl)-4-propyl- 1-piperidyl]- 1 -butanon- -fumarat, Smp. 159-160 .

- PATENTANSPRUCHVerfahren zur Herstellung von neuen Piperidinderivaten der allgemeinen Formel I, EMI3.1 in welcher R1 einen aliphatischen Kohlenwasserstoffrest mit höch stens 10 Kohlenstoffatomen, die Cinnamylgruppe, eine unsubstituierte oder am Benzolkern durch Ha logen bis Atomnummer 35 oder niedere Alkyl- oder Alkoxygruppen substituierte Phenyl- (niederalkyl) -gruppe mit höchstens 12 Kohlenstoffatomen, in der die Phenylgruppe mit der niederen Alkylgruppe statt direkt auch über Sauerstoff, die Carbonylgruppe, Al kyliminogruppe oder Alkenyliminogruppe verbunden sein kann, bedeutet und R2 und Rg eine Alkylgruppe mit 2-4 Kohlenstoffatomen bedeuten, dadurch gekennzeichnet, dass man auf eine Verbindung der allgemeinen Formel II, EMI3.2 in welcher R1 und R die unter Formel I angegebene Bedeutung haben, R3, die Methyl-,Äthyl- oder Propylgruppe, Y eine niedere Alkylgruppe und Z die Gruppe -COO-Y oder die Cyanogruppe bedeuten, eine Säure einwirken lässt und gewünschtenfalls aus dem erhaltenen Säureadditionssalz eine Verbindung der allgemeinen Formel I freisetzt und sie oder das zunächst erhaltene Salz in ein anderes Additionssalz mit einer anorganischen oder organischen Säure überführt.
