Im Falle, dass R" Wasserstoff bedeutet, so wird" die Acylierung mit dem Säureadditionssalz der genannten Verbindung der Formel II durchgeführt.
Das bevorzugte Derivat ist dasjenige, das durch Umsetzung mit Essigsäureanhydrid erhalten wird. Diese Verbindungen weisen antidepressive Wirkung auf und man kann sie in täglichen Dosen von 1 bis 300 mg einnehmen, vorzugsweise in Form ihrer nichttoxischen Säureadditionssalze.
Den Ausgangsstoff der Formel II, in der R" Nieder alkyl ist, stellt man z. B. zweckmässig durch Umsetzung des entsprechenden 10,11-Dihydro-5-(3-dimethylamino propyl)-5,10-epoxy-11-keto-5H- dibenzo[a,d]cycloheptens mit einem Alkylmagnesiumhalogenid her. Mit Äthyl- magnesiumjodid z.
B. erhält man das 10,11-Dihydro-5-(3-dimethylaminopropyl)- 5,10-epoxy-11-äthyl-11-hydroxy 5H- dibenzo[a,d]cyclohepten. Die 11-Ketoverbindung kann man ihrerseits bequem durch Oxydation der entsprechenden 11-Hydroxyver- bindung nach der Methode Oppenauer erhalten, worauf die Grignardverbindung hydrolysiert wird.
Den Ausgangsstoff der Formel II, in der R" Was serstoff ist, kann man aus dem, wie eben beschrie ben, erhaltenen 11-Alkyl-11-hydroxy-5-(3-tertiäramino- propyl)-Derivat durch Desalkylierung bzw. durch Be handeln. mit einem Hlalogenformiat und darauffolgender Hydrolyse hergestellt werden, oder man desalkyüert direkt das durch Oxydation erhaltene 11-Ketoderivat mittels Grignardierung und anschliessender Hydrolyse. Die nach dem erfindungsgemässen Verfahren erhaltenen Dibenzocyeloheptene haben in 11-Stellung zwei ver schiedene Substituenten, weshalb diese Verbindungen im allgemeinen als Razemate der entsprechenden geo metrischen Isomere anfallen. Eine Aufspaltung in die Enantiomorphen nach bekannten Verfahren ist leicht möglich; z.
B. durch fraktionierte Krisfallisation der Salze mit optisch aktiven Säuren. Beide Isomere weisen die angegebene pharmakologische Wirkung auf, doch kann die Wirkung der einen Form wesentlich stärker sein als die der anderen.
Beispiel A. 10,11-Dihydro-5-(3-dimethylaminopropyl) 5,10-epoxy-11-hydroxy-11-methyl-5H-dibenzo[a,d]- cyclohepten Ein mit Rührwerk, Rückflusskühler und Gaszulei tungsrohr ausgerüsteter 50-ml-Kolben wird mit 145 mg (0,06 g -Atom) Magnesiumdrehspänen, einem Jodkristall und 25 ml Tetrahydrofuran beschickt. Die Vorrichtung wird mit trockenem Stickstoff ausgespült und mittels eines Trockenröhrchens vor atmosphärischer Feuchtig keit geschützt. Die Umsetzung wird mittels eines Trop- fens Methyljodid eingeleitet, worauf Methylchlorid unter die Oberfläche des gerührten Gemisches bei Raumtemperatur eingeleitet wird. Die Einleitung des Gases wird fortgesetzt, bis -alles Magnesium gelöst ist.
Die Lösung des Grignard-Reagens wird dann tropfen weise in eine gerührte Lösung von 900 mg (0,0029 Mol) 10,11-Dihydro-5-(3-dimethylaminopropyl) 5,10-epoxy-11-keto-5H-dibenzo[a,d]- cyclohepten in 25 ml Tetrahydrofuran gegeben, die m einer Stick stoffatmosphäre gehalten und in einem Eisbad gekühlt wird. Das Gemisch wird 45 Minuten bei Raumtempera tur gerührt und der Hauptteil des Tetrahydrofurans bei einer Temperatur von unter 50 C unter vermindertem Druck verdampft. Der Rückstand wird in Benzol gelöst und durch die tropfenweise Beigabe von 10 ml Wasser hydrolysiert. Die Benzolschicht wird abgegossen und der gelatinöse Niederschlag mehrmals mit Benzol ge waschen. Nach Waschen mit Wasser werden die ver einigten Benzollösungen dreimal mit je 25 ml 0,5 m Zitronensäure extrahiert.
Der Säureextrakt wird alka lisch gemacht und die ölige Base mit Benzol extrahiert. Der gewaschene und getrocknete Benzolextrakt wird unter vermindertem Druck verdampft, wobei das Pro dukt als öliger Rückstand zurückbleibt, der bei Verrei ben mit absolutem Äther fest wird. Man erhält 700 mg (75%) Produkt, F = 108-110 C. Die Base kann durch Behandlung einer äthanolischen Lösung mit einem 10 % igen überschuss an in absolutem Äthanol gelöster Maleinsäure in das saure Maleatsalz übergeführt werden. Bei Verdünnen mit absolutem Äther fällt das gebildete 10,11-Dihydro-5-(3-dimethylaminopropyl) 5,10-epoxy-11-hydroxy-11-methyl-5H- dibenzo[a,d]cyclohepten-wasserstoffmaleat aus, F = 182-184 C. Der Schmelzpunkt erhöht sich bei weiterem Umkristallisieren nicht mehr.
Analyse für C21H25N02.C4H404: Berechnet: C, 68,32; H, 6,65; N, 3,19; Gefunden: C, 67,89; H, 6,62; N, 3,30.
B. 11-Acetoxy-10,11,dihydro-5-(3-dimethylamino propyl)-5,10-epoxy-11-methyl-5H-dibenzo[a,d]- cyclohepten Eine Lösung von 650 mg (0,00204 Mol) 10,11-Dihydro-5-(3-dimethylaminopropyl) 5,10-epoxy-11-hydroxy-11-methyl- 5H-dibenzo[a,d]cyclohepten in 12 ml Essigsäureanhydrid wird 41/2 Stunden bei Rückfluss erhitzt. Die dunkelgelbe Lösung wird bei ver mindertem Druck zur Trockne verdampft und der ölige Rückstand in 20 ml 1 ml 6n Salzsäure enthaltendem Wasser gelöst. Nach einmaligem Extrahieren mit Benzol wird die wässrige Lösung mit Natriumhydroxyd alka lisch gemacht und die ölige Base mit Benzol extrahiert. Der gewaschene Benzolextrakt wird unter vermindertem Druck verdampft.
Man erhält das Produkt in Form weisser Kristalle in 49 % iger Ausbeute. F =l08 bis 122 C. Nach Umkristallisieren aus einem Gemisch von Äther und Petroläther und Sublimation bei 115 C und 0,05 mm Hg schmilzt eine analytische Probe bei 122-126 C.

The washed benzene extract is evaporated under reduced pressure.
The product is obtained in the form of white crystals in a 49% yield. F = l08 to 122 C. After recrystallization from a mixture of ether and petroleum ether and sublimation at 115 C and 0.05 mm Hg, an analytical sample melts at 122-126 C.
