Description
Verfahren zur Herstellung neuer Pyrido[1,2-a]-s.triazindione
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Pyrido[1,2-a]-s-triazin-dione
Die neuen Pyrido[1,2-al-s-triazin-dione entsprechen der Formel I:

In dieser Formel bedeuten: Rt Wasserstoff, Halogen,. Nitro, niederes Alkyl, niederes Alkoxy oder Halogenalkyl,
R2 einen aliphatischen Kohlenwasserstoffrest, einen Alkoxyalkyl- oder Halogenalkylrest und
R3 Wasserstoff, Halogen, Nitro, niederes Alkyl, niederes Alkoxy oder Halogenalkyl.
In der Formel I sind unter niederen Alkylresten Rl und R3 geradkettige oder verzweigte Reste mit 1 bis 5 Kohlenstoffatomen zu verstehen, zum Beispiel Methyl, Äthyl, n-Propyl, Isopropyl, n-Butyl, tert. Butyl, Isobutyl sowie Pentylreste. Niedere Alkoxyreste Rl und R3 weisen vorzugsweise 1 bis 4 Kohlenstoffatome auf, wie beispielsweise Methoxy, Isopropoxy, n-Butoxy, Isobutoxy. Als Halogenalkyl kommen für R, R2 und R3 in erster Linie niedere Alkylreste, die ein- oder mehrfach durch Fluor, Chlor und/oder Brom substituiert sein können, in Betracht. Ein aliphatischer Kohlenwasserstoffrest R2 kann z.
B. ein geradkettiger bzw. verzweigter Alkylrest mit 1 bis 8 bzw. 3 bis 8 Kohlenstoffatomen, ein niederer Alkenylrest mit 2 bis 4 Kohlenstoffatomen, insbesondere der Allyl- oder Methallylrest oder ein Älkinylrest mit 2 bis 4 Kohlenstoffatomen, wie der Propinyl- oder 1-Methyl-propinylrest, sowie ein monocyclischer oder polycyclischer aliphatischer Rest mit 3 bis 9 Kohlenstoffatomen sein, z. B. Cyclopropyl, 1-Methyl-cyclopropyl, Cyclobutyl, Cyclopentyl, Cyclohexyl, Cycloheptyl, Cyclooctyl, Bicyclohexyl, Bicycloheptyl, Bicyclooctyl, Tricyclononyl, Tetracyclononyl. Alkylreste als Teil von Substituenten, z. B. AlkaxyalkyI, weisen vorzugsweise je 1 bis 3 Kohlenstoffatome auf. Unter Halogen ist für Rt und R3 F]uor, Chlor oder Brom zu verstehen.
Das erfindungsgemässe Verfahren zur Herstellung von neuen Pyrido[1,2-a]-s-triazin-dionen der Formel I ist dadurch gekennzeichnet, dass man einen Pyridylharnstoff der Formel II

in der R1, R2 und R3 die unter Formel I angegebenen Bedeutungen haben, mit dem Chlorid oder einem Ester der Chlorameisensäure umsetzt. Für diese Umsetzung ist die Verwendung von gegenüber den Reaktionsteilnehmern inerten Lösungs- oder Verdünnungsmitteln wie zum Beispiel aromatischen und aliphatischen Kohlenwasserstoffen und Halogenkohlenwasserstoffen, N,Ndialkylierten Carbonsäureamiden, Äthern und ätherartigen Verbindungen ratsam. Als Ester der Chiorameisen- säure kommen niedermolekulare Alkylester, besonders der ethylester in Frage.
Die als Ausgangsstoffe dienenden Pyridylharnstoffe der Formel II können durch Umsetzung eines 2-Aminopyridins der Formel III

mit einem Isocyanat der Formel IV R2NCo ( in1 in welchen Formeln Rr, R2 und R3 die unter Formel I angegebenen Bedeutungen haben, in Anwesenheit von gegenüber den Reaktionsteilnehmern inerten Lösungsoder Verdünnungsmitteln wie Kohlenwasserstoffen, zum Beispiel Benzol, Toluol, Xylolen, Äthern und ätherartigen Verbindungen z. B. Dialkyläthern, Dioxan, Tetrahydrofuran, N,N-disubstituierten Carbonsäureamiden und -acetamiden, Nitrilen, zum Beispiel Acetonitril, chlorierten Kohlenwasserstoffen, z. B. Chlorbenzol, Chloroform und Methylenchlorid, und gegebenenfalls von basischen Kondensationsmitteln wie anorganischen Basen erhalten werden.
Die neuen Pyrido[1,2-a]-s-triazin-dione der Formel I besitzen ausgezeichnete herbizide Eigenschaften und können zur Bekämpfung mono- und dicotyler Unkräuter und Ungräser verwendet werden. In Konzentrationen über 6 kg/ha wirken diese Wirkstoffe als Totalherbizide, in Konzentrationen unter 6 kg/ha als selektive Herbizide. Von den neuen Wirkstoffen werden schwer bekämpfbare, sowie tiefwurzelnde Unkrautarten, zum Beispiel Gräser, Leguminosen und Doldengewächse, erfasst. Die Applikation der Wirkstoffe kann mit gleich gutem Erfolg sowohl vor dem Auflaufen (preemergence) als auch nach dem Auflaufen (postemergence) der Pflanzen erfolgen.
Zur Bekämpfung von Unkräutern und Ungräsern werden jene Pyrido[1,2-a]-s-triazin-dione der Formel I, in der Rt Wasserstoff oder einen Alkylrest mit 1 bis 5 Kohlenstoffatomen, R2 verzweigtes Alkyl mit 3 bis 8 Kohlenstoffatomen oder Cycloalkyl mit 3 bis 9 Kohlenstoffatomen, und R3 Wasserstoff bedeuten, besonders jene, bei denen R2 einen verzweigten, über den Verzweigungskohlenstoff an den Triazinring gebundenen Alkylrest, wie z. B. Isopropyl, sec. Butyl, tert. Butyl, sec. Pentyl, tert. Pentyl etc., sowie einen Cycloalkylrest darstellt, bevorzugt.
Solche Wirkstoffe sind selektiv und können zur Bekämpfung von Ackerunkräutern dienen, beispielsweise zur Bekämpfung von Hirsearten (Panicum sp.), Senfarten (Sinapis sp.), Gänsefussarten (Chenopodiaceaen), Rispengras (poa annua), Ackerfuchsschwanz (Alopecurus agrestis), Kamille (matricaria), Pastinak (pastinaca sativa), ohne Nutzpflanzen, wie Getreide, Mais, Hackfrüchte, Ölpflanzen, Gemüse, Baumwolle, Sorghum, Soyabohnen und Luzerne, vor oder nach dem Ablaufen zu schädigen.
Zur Herstellung von herbiziden Mitteln werden die Wirkstoffe mit geeigneten Trägerstoffen und/oder Verteilungsmitteln vermischt.
Die folgenden Beispiele veranschaulichen das erfindungsgemässe Verfahren. Sofern nichts anderes vermerkt ist, sind die Temperaturen in Celsiusgraden angegeben.
Beispiel 1
Eine Lösung von 5,8 g (0,025 Mol) N-3 -Methylpyridyl-(2)-N'-cyclohexyl-harnstoff in 50 ml absolutem Toluol wird unter Rühren und Kühlung mit Eiswasser der Lösung von 5,0g (0,05 Mol) Phosgen in 50 ml absolutem Toluol zugetropft. Die entstandene Suspension wird anschliessend unter gleichzeitiger Einleitung eines schwachen Phosgenstromes 2 Stunden lang am Rückfluss erhitzt. Nach dem Abkühlen auf Raumtemperatur wird der gebildete Niederschlag abfiltriert, in 200ml Wasser suspendiert und die Suspension mit konzentriertem Ammoniak neutralisiert. Der Niederschlag wird abfiltriert und getrocknet. Man erhält 5,85 g (90ozon 3 -Cyclohexyl-9-methyl-2H-pyrido [1,2-a] -s-triazin-2,4(3H)-dion vom Smp. 179-180 C.
Beispiel 2
5,5 g (0,023 Mol) N-3-Methylpyridyl-(2)-N'-cyclohexyl-hamstoff in 100 ml absolutem Toluol angeschlämmt, werden mit 23,3 g (0,23 Mol) Chlorameisensäureäthylester 15 Stunden zum Rückfluss erhitzt. Die Lösung wird am Rotationsverdampfer eingeengt und das zurückbleibende Öl kristallisieren gelassen. Der kristalline Rückstand wird aus Benzol-Petroläther umkristallisiert. Man erhält 3,1 g (52,30/o) 3 -Cyclohexyl-9-methyl-2H-pyrido [1,2-a] -s-triazin-2,4(3H)-dion vom Smp. 179-180 .
Der für obige Beispiele als Ausgangsmaterial verwendete N-3 -Methylpyridyl-(2)-N'-cyclohexyl-harnstoff wird wie folgt hergestellt:
Zur Lösung von 47 g (0,5 Mol) 2-Aminopyridin in 500 ml Benzol lässt man unter Kühlung mit Eiswasser und unter Rühren 62,5 g (0,5 Mol) Cyclohexylisocyanat zutropfen. Das Gemisch wird 3 Stunden lang bei 50 C weitergerührt, wobei farblose Kristalle ausfallen.
Der Niederschlag wird abgetrennt. Durch Umkristallisieren des Rohproduktes aus Isopropyl-äther erhält man den N-3-Methylpyridyl-(2)-N'-cyclohexyl-harnstoff vom Smp. 1040 C.
In analoger Weise wie in den Beispielen 1 und 2 beschrieben wurden noch folgende Wirkstoffe der allgemeinen Formel I hergestellt:

Der Wirkstoff wird als 100/oige wässrige Suspension in Gartenerde eingearbeitet, so dass die Wirkstoffkonzentration 0,5 g pro Liter Erde ist. In Saat schalen werden dann folgende Testpflanzen in die so vorbereitete Erde eingesät:
Hafer, Senf, Raygras, Zuckerrübe, Gurke, Wicke.
Die Schalen werden dann im Gewächshaus bei 20 bis 24O C und 70 /o relativer Luftfeuchtigkeit unter Tageslicht gehalten.
Die Auswertung des Versuches erfolgte nach 20 Tagen und die Beurteilung nach dem 10er Index:
10 = Pflanzen unbeschädigt = KontrolIe
0 = Pflanzen abgestorben
9- 1 = Zwischenstufen der Schädigung
Die 10%ige Suspension wurde aus einem 10%igen Spritzpulver folgender Zusammensetzung: 10 Teile Wirkstoff, 0,6 Teile Natriumdibutylnaphthylsulfonat, 10 Teile Natrium-Aluminiumsilikat, 78,4 Teile Kaolin und 1 Teil Naphthalinsulfonsäuren Phenolsulfonsäuren-Formaldehyd-Kondensat 3:2:1, durch Verdünnen mit Wasser erhalten.

Selektivversuche im Freiland a) Vorauflauf-Versuch
In ein frisch zubereitetes Saatbeet werden als Testpflanzen eingesät: Vicia faba, Erbse, Luzerne und Lein. Der Wirkstoff wird als wässrige Dispersion, hergestellt aus einem 250/oigen Spritzpulver, direkt nach der Saat auf die Bodenoberfläche appliziert.
Natürliche vorhandene Unkrautflora: Chenopodium album, Polygonum convolvulus, Polygonum persicaria, Capsella brusa pastoris, Sinapis arvensis.
Die Beurteilung der phytotoxischen Wirkung auf die Kulturpflanzen und die herbizide Wirkung auf die natürlichen Unkräuter werden nach 62 Tagen ermittelt.
Die Beurteilung wird bei Kulturpflanzen nach der folgenden Skala vorgenommen:
1 = keine Symptome oder Schädigung (wie unbehandelte Kulturpflanzen)
2-4 = logarithmische Zunahme der reversiblen phytotoxischen Symptome
5-8 = logarithmische Zunahme der irreversiblen phytotoxischen Symptome
9 = vollständiges Absterben
Für die Unkräuter wird die herbizide Wirkung in Prozent angegeben:
100 O/o = vollständige Abtötung 90-10 /o = Zwischenstufen der Abtötung 0 o/o = keine Schädigung (wie unbehandelte Kontrollpflanzen).

Die Kulturpflanzen Mais, Sonnenblume und Karotte und die schwerbekämpfbaren Unkräuter Galium spez. Raygras ital. und Amaranthus spez. werden als Testpflanzen in ein frisch zubereitetes Saatbeet eingesät. Direkt nach der Saat wird der Wirkstoff als wässrige Dispersion, hergestellt aus 250/oigem Spritzpulver, auf die Bodenfläche appliziert. Nach 54 Tagen wird die phytotoxische Wirkung auf die Kulturpflanzen und die herbizide Wirkung auf die eingesäten Unkräuter bewertet.
Die Beurteilung wird bei Kulturpflanzen nach der folgenden Skala vorgenommen:
1 = keine Symptome oder Schädigung (wie unbehandelte Kulturpflanzen)
2-4 = logarithmische Zunahme der reversiblen phytotoxischen Symptome
5-8 = logarithmische Zunahme der irreversiblen phytotoxischen Symptome
9 = vollständiges Absterben

Als Wirkstoff wurde bei diesem Versuch eingesetzt: 3-Isopropyl-9-methyl-2H-pyrido[1,2-a]- s-triazin-2,4(3H)dion (Wirkstoff 4)
Resultat:

Beide Versuche zeigen, dass die Wirkstoffe 4 und 13 gemäss vorliegender Anmeldung eine sehr gute- herbizide Wirkung sowohl gegen die natürlich vorkommende tinkrautilora als auch gegen die eingesäten schwer bekämpfbaren Unkräuter besitzen. Bei den Kulturpflanzen traten keine, wenn dann lediglich schwache, jedoch reversible phytotoxische Symptome auf.
Claims (1)
- PATENTANSPRUCHVerfahren zur Herstellung von neuen Pyrido[1,2a]-s-triazin-dionen der Formel I EMI6.1 in der Rt Wasserstoff, Halogen, Nitro, niederes Alkyl, niederes Alkoxy oder Halogenalkyl, R2 einen aliphatischen Kohlenwasserstoffrest, einen Alkoxyalkyl- oder Halogenalkylrest und R3 Wasserstoff, Halogen, Nitro, niederes Alkyl, niederes Alkoxy oder Halogenalkyl bedeuten, dadurch gekennzeichnet, dass man einen N-Pyridylharnstoff der Formel II EMI6.2 worin Rj, Rr und R3 die oben angegebenen Bedeutungen haben, mit dem Chlorid oder einem Ester der Chlorameisensäure umsetzt.UNTERANSPRÜCHE 1. Verfahren gemäss Patentanspruch, dadurch gekennzeichnet, dass man die Umsetzungen in Gegenwart von gegenüber den Reaktionsteilnehmern inerten Lösungs- und/oder Verdünnungsmitteln vornimmt.2. Verfahren gemäss Patentanspruch und Unteranspruch 1, dadurch gekennzeichnet, dass man Chlorameisensäureäthylester verwendet.
