Abstract
N-benzoyl-N-phenyl-amino acid derivs. - with anti-inflammatory and analgesic activity. Title compounds of formula (I): (where R1 is H or opt. substd. alkyl, aryl, alkylaryl, cycloaliphaticyl or heterocyclyl; n is 0 and R2 is morpholino, NH2, NHR' or NHR'R" in which R' and R" are alkyl, or n is 1 and R2 is H, an alkali(ne earth) metal, or opt. substituted alkyl, aryl, arylalkyl, cycloaliphaticyl or heterocyclyl; and each X and Y is H, halogen, NO2, a sulphonic group, opt. substituted amono, opt. substituted alkyl, or a group OR1 in which R1 is as defined above), which have anti-inflammatory and analgesic properties, are prepd. by reacting an N-phenyl-amino acid of formula (II) with an agent capable of introducing the residue of formula (III), e.g. the corresponding benzoic acid chloride, anhydride, mxed anhydride or methyl or ethyl ester (the free acid can also be used).
Description
Procédé de préparation des dérivés des acides a-aniinés
N-disubstitués par benzoylation
Les dérivés de divers acides a-aminés N-disubstitués peuvent être obtenus par diverses méthodes classiques d'obtention des acides a-aminés.
Certains de ces dérivés, toutefois, qui ont montré des propriétés anti-inflammatoires et antalgiques, font intervenir des substituants sur le groupe amino (phényle substitué ou benzoyle substitué) provoquant un encombrement stérique de la molécule.
Aussi, selon les cas, devrait-on choisir une méthode de synthèse particulière pour obtenir les dérivés désirés avec un rendement suffisant et un produit brut d'une certaine pureté. Dans le groupe de molécules intéressant la titulaire, la présence du substituant R1 sur le carbone, directement voisin de l'azote, influence de façon très sensible le déroulement de la réaction.
La présente invention a pour objet la préparation par benzoylation des dérivés N-(X,Y-phényl)-N-(X,Ybenzoyl) de différents acides a-aminés ainsi que de leurs esters ou de leurs sels répondant à la formule générale:

Rt représente un atome H ou un reste alkyle, aryle, alkylaryle, cycloaliphatique ou hétérocyclique, ces restes étant substitués ou non;
n est égal à 0, lorsque R2 est un reste morpholino, NH2. ou NHR' ou NR'R" dans lesquels R' et R" représentent un groupe alkyle, et 1, lorsque R2 est un atome d'hydrogène ou bien un métal alcalin ou alcalino-terreux correspondant au sel de cet acide, ou un reste alkyle, aryle, arylalkyle, cycloaliphatique ou hétérocyclique, ces restes étant substitués ou non;

X et Y sont semblables ou différents et représentent un atome d'hydrogène ou d'halogène, un groupe nitro ou sulfonique, un reste amine substitué ou non, un groupe alkyle substitué ou non, ou le groupe -OR1, dans lequel R1 est défini comme indiqué ci-dessus, caractérisé en ce que l'on fait réagir dans un solvant un composant répondant à la formule générale:

Parmi les substituants précédemment définis les suivants sont préférentiels:
R1 = H, -CH3, -QH5, -C6H5, -CH2CH2OH
X et Y = H, -OH, OCH3, Cl, F, Br, -NO2, -NH2, -CH3, -CH3

L'agent apte à introduire le groupement répondant à la formule (3) peut être choisi parmi les chlorures, les anhydrides, les anhydrides mixtes, ou les esters méthyliques ou éthyliques d'un acide X, Y-benzoique agissant sur des dérivés N-(X, Y-phényl) d'acides -aminés, notamment glycine, alanine, phénylanine ou de leurs esters méthyliques ou éthyliques répondant à la formule (2).
La benzoylation peut également s'effectuer en présence de catalyseurs, par exemple BF3, acide p-toluène sulfonique, etc., par action d'un acide X, Y-ben zolque libre sur des N-(X, Y-phényl) ix-amino-acides, ou leurs esters.
D'une façon générale, la benzoylation des N-(palkoxyphényl)-glycines et -alanines est conduite en les dissolvant dans une solution aqueuse de NaOH, en ajoutant au mélange le chlorure de l'acide X, Y-benzoïque et en acidifiant pour obtenir la benzamide correspondante que l'on filtre, lave, sèche et délaye dans l'éther et que l'on recristallise dans l'éthanol absolu.
La benzoylation peut s'effectuer également dans le toluène, avec la formation du chlorhydrate de l'acide α-aminé que l'on sépare de la benzamide, et que l'on recristallise dans le toluène, ou dans un mélange de toluène-benzine.
Dans le cas où l'agent de la benzoylation est un halogénure d'acide, la réaction s'écrit:

Exemples
1) Préparation de la N-(p-méthoxyphényl)-N-(pchlorobenzoyl)-glycine.
Le composé de départ peut être obtenu de la manière suivante : 123,2 g p-anisidine, 108 g d'eau, 123 g d'acétate de sodium anhydre sont mélangés et chauffés sous agitation à 500. On ajoute par portions en 1/2 g 94,5 g d'acide chloroacétique, puis on garde 1 h à 95980. La masse de réaction est dissoute dans 1680g de
NaOH 5%, puis on extrait à l'éther 30g de p-anisidine non transformée. De la solution alcaline acidifiée avec 600g HCI 25 %, on extrait à l'éther 30 g d'acide
N-(p-méthoxyphényl)-imino-diacétique, F=122-3 .
Après traitement avec NaOH concentré 35% jusqu'à pH 3,5, 60g de N-(p-méthoxyphényl)-glycine cristallisent. F = 142-3 .
La benzoylation est ensuite réalisée de la manière suivante:
181,2 g de N-(p-méthoxyphényl)-glycine dans 600g d'eau sont dissous avec 1200g NaOH 10%. On ajoute à 200 192,5 g de chlorure de l'acide p-chlorobenzoique en 1/2 h, puis on agite encore 3 h. Après acidification à pH 2, on obtient 318,5 g de p-chlorobenzamide insoluble dans l'eau. Le produit est filtré, lavé et séché.
On le délaye ensuite dans l'éther, puis on le recristallise dans méthanol absolu. On obtient alors 196 g de
N-(p-méthoxyphényl)-N-(p - chlorobenzoyl)-glycine, F = 191-20.
Formule brute: CtaHt4ClNO4 M= 319,75 Analyse:

2) Préparation de la N-(p-oxyphényl) N-(p-chloro- benzoyl)glycine.
On peut également procéder de la manière suivante, pour obtenir le composé de départ:
109 g de p-aminophénol sont chauffés à 750 dans 350 ml d'éthanol. On introduit 61,3 g de chloroacétate d'éthyle, puis on porte à reflux 3 h. Après refroidissement, on filtre le chlorhydrate de p-aminophénol, puis on concentre sous vide à sec. Le résidu est dissous dans 1460g HCl 5%. On extrait à l'éther des impuretés, puis on neutralise à pH 5,5 avec NaOH conc. 35 % en présence d'éther. Après distillation on obtient 86,1 g de résidu cristallin. On recristallise dans 750 ml de toluène pour obtenir 76 g de N-(p-oxyphényl)-glycine éthylester. F. = 74-50.
La benzoylation s'effectue ainsi:
39 g de N-(p-oxyphényl) glycine éthylester et 200 ml de toluène sont chauffés à 800. On introduit 17,5 g de chlorure de l'acide p-chlorobenzoïque dissous dans 50 ml de toluène, puis on chauffe 1 h. Après refroidissement, la réaction est filtrée. Le précipité séché est délayé avec 73 g HCl 5% dans 100 ml d'eau. On filtre le précipité et on le sèche. Par recristallisation dans le toluène, on obtient 29,5 g de N-(p-oxyphényl) N-(pchlorobenzoyl) glycine éthylester. F = 148-1500.

3) Préparation de la N-(p-oxyphényl) N-(p-chloro- benzoyl)alanine éthylester.
De la même façon on a traité 109 g de p-aminophénol dans 350 ml d'éthanol avec 75,1 g d'α-chloro- propionate d'éthyle à reflux pendant 6 heures. On a obtenu 74,5 g de N-(p-oxyphényl) alanine éthylester,
F = 80-1 . 41,8 g de cet N-(p-oxyphényl)alanine éthylester sont benzoylés avec 17,5 g de chlorure de l'acide p-chlorobenzoique à 800 dans le toluène 6 heures. On filtre le chlorhydrate et concentre la solution toluénique à sec pour récupérer 23,2g de N-(p-oxyphényl) N-(pchlorobenzoyl) alanine éthylester, F = 106-7 .
Selon la nature des radioaux X et Y sur le groupement phényle ou benzoyle, il est possible de transformer ces radicaux par des procédés connus:
On pourra, par exemple, dans le cas où ces radicaux sont des groupements OH, procéder à une alkylation, au moyen d'un acide mono-méthyl- ou mono-éthylsulfonique, d'un ester méthylique ou éthylique de l'acide p-toluène-sulfonique, d'un halogène de méthyle ou d'éthyle, du diazométhane ou diazoéthane.
4) Préparation de la N-(p-méthoxyphényl)-N-(p- chlorobenzoyl) -glycine.
33,4 g de N-(p-oxyphényl)-N-(p-chlorobenzoyl) glycine éthylester, 264 g NaOH 5% sont mélangés à 20 .
On ajoute sous agitation en 1/2h 41,6g de sulfate de méthyle. Après quelques heures de réaction le pH tombe à 6. On ajoute alors 344 g NaOH 5 %, puis on chauffe 1/2 h à 60-650 jusqu'à disparition du précipité.
On traite au charbon, filtre, puis neutralise avec HCI 25 '0/o à pH 2. On obtient ainsi 24,1 g de N-(p-méthoxy phényl)-N- (p.chlorobenzoyl) glycine recristallisé dans l'éthanol absolu, F = 191.20. Le produit est identique à celui obtenu par l'exemple No 1.
Les dérivés p-éthoxy sont obtenus par aikylation
avec le sulfate d'éthyle.
5) Autres exemples
108,7 g de N-(p-méthoxyphényl)-glycine et 750 mi de toluène sont chauffés à 75-800. On introduit 55,7 g de chlorure de l'acide p-nitrobenzoïque dissous dans 150 ml de toluène en 1/2h sous agitation, puis on garde encore 2 h à 80 . Après la cristallisation, on filtre et sèche le précipité. On d6lave dans 440 g HCl 2,5 %, la p-nitrobenzamide insoluble est récupérée par filtration.
On obtient 97,8 g de N- (p-méthoxyphényl)-N-(p-nitro- benzoyl)glycine, F = 161.20.

Process for the preparation of α-anine acid derivatives
N-disubstituted by benzoylation
The derivatives of various N-disubstituted α-amino acids can be obtained by various conventional methods of obtaining α-amino acids.
Some of these derivatives, however, which have shown anti-inflammatory and analgesic properties, involve substituents on the amino group (substituted phenyl or substituted benzoyl) causing steric hindrance of the molecule.
Also, depending on the case, one should choose a particular synthesis method to obtain the desired derivatives with a sufficient yield and a crude product of a certain purity. In the group of molecules of interest to the holder, the presence of the substituent R1 on the carbon, directly adjacent to nitrogen, very significantly influences the course of the reaction.
A subject of the present invention is the preparation by benzoylation of N- (X, Y-phenyl) -N- (X, Ybenzoyl) derivatives of various α-amino acids as well as their esters or their salts corresponding to the general formula:

Benzoylation can also be carried out in the presence of catalysts, for example BF3, p-toluenesulfonic acid, etc., by the action of a free X, Y-ben zolque acid on N- (X, Y-phenyl) ix -amino acids, or their esters.
In general, the benzoylation of N- (palkoxyphenyl) -glycines and -alanines is carried out by dissolving them in an aqueous solution of NaOH, adding to the mixture the chloride of X, Y-benzoic acid and acidifying for obtain the corresponding benzamide which is filtered, washed, dried and diluted in ether and which is recrystallized from absolute ethanol.

Examples
1) Preparation of N- (p-methoxyphenyl) -N- (pchlorobenzoyl) -glycine.
The starting compound can be obtained as follows: 123.2 g p-anisidine, 108 g of water, 123 g of anhydrous sodium acetate are mixed and heated with stirring at 500. After treatment with 35% concentrated NaOH to pH 3.5, 60 g of N- (p-methoxyphenyl) -glycine crystallize. F = 142-3.
Benzoylation is then carried out as follows:
181.2 g of N- (p-methoxyphenyl) -glycine in 600 g of water are dissolved with 1200 g of 10% NaOH. To 200,192.5 g of chloride is added p-chlorobenzoic acid over 1/2 h, followed by stirring for a further 3 h. After acidification to pH 2, 318.5 g of p-chlorobenzamide insoluble in water are obtained. 2) Preparation of N- (p-oxyphenyl) N- (p-chlorobenzoyl) glycine.
It is also possible to proceed as follows, to obtain the starting compound:
109 g of p-aminophenol are heated to 750 in 350 ml of ethanol. 61.3 g of ethyl chloroacetate are introduced, then the mixture is refluxed for 3 h. After cooling, the p-aminophenol hydrochloride is filtered off and then concentrated under vacuum to dryness. The residue is dissolved in 1460g 5% HCl. The impurities are extracted with ether, then neutralized to pH 5.5 with conc. NaOH. 35% in the presence of ether. After distillation, 86.1 g of crystalline residue are obtained. Recrystallized from 750 ml of toluene to obtain 76 g of N- (p-oxyphenyl) -glycine ethyl ester. Mp = 74-50.
Benzoylation is carried out as follows:
39 g of N- (p-oxyphenyl) glycine ethyl ester and 200 ml of toluene are heated to 800. 17.5 g of p-chlorobenzoic acid chloride dissolved in 50 ml of toluene are introduced, then heated for 1 h. After cooling, the reaction is filtered. The dried precipitate is diluted with 73 g 5% HCl in 100 ml of water. The precipitate is filtered off and dried. By recrystallization from toluene, 29.5 g of N- (p-oxyphenyl) N- (pchlorobenzoyl) glycine ethyl ester are obtained. F = 148-1500.

3) Preparation of N- (p-oxyphenyl) N- (p-chlorobenzoyl) alanine ethyl ester.
