Die Erfindung bezieht sich auf ein Verfahren zur Herstellung von Verbindungen der Formel II

oder einem Salz dieser Verbindung umgesetzt wird, in welchen Formeln M ein Wasserstoff- oder Alkalimetallatom bedeutet, X ein Halogenatom oder eine Tosyloxygruppe, Y eine Acylgruppe und R'5 die gleiche Bedeutung hat wie R5, aber statt der Amino-, Methylamino- oder Athylaminogruppe eine mit einer oder zwei Acylgruppen geschützte derartige Gruppe darstellt,
und die erhaltene Zwischenverbindung durch Hydrolyse in die Verbindung der Formel II überführt wird.
Die Gruppe X in Verbindung IV ist vorzugsweise Chlor oder Brom.
Die Reaktion kann in einem geeigneten Lösungsmittel durchgeführt werden. Solche Lösungsmittel sind unter anderem: Alkohole, wie Methanol, Ketone, wie Aceton und Methyläthylketon, und Äther, wie Dioxan, Dimethyl- und Glykoläther. Die Abspaltung einer Trifluoracetylgruppe Y kann mit z. B. KOH in Alkohol, z. B. äthanol, erfolgen.
Wenn M in der Formel III ein Wasserstoffatom darstellt, ist es zu empfehlen, dem Reaktionsgemisch einen Säurebinder zuzusetzen. Als solche seien unter anderem erwähnt: Alkoholate, Kalium- und Natriumcarbonat, tertiäre Amine, Pyridin und dergleichen.
Die Temperatur des Reaktionsgemisches kann in ziemlich weiten Grenzen schwanken. In der Regel liegt sie jedoch zwischen 0 und 500 C.
Die Oxime der Formel III können auf die übliche Weise mit Hilfe von Hydroxylamin aus den entsprechenden Aldehyden oder Ketonen hergestellt werden.
Die Oximate lassen sich aus den Oximen dadurch herstellen, dass diese, gegebenenfalls in Alkohol gelöst, einer Lösung von Natrium- oder Kaliumalkoholat oder Natrium- oder Kaliumhydroxyd in Alkoholen zugesetzt werden.
Die acylierten Amine der Formel IV sind dadurch herstellbar, dass vom entsprechenden primären oder sekundären Amin und einem in z. B. Benzol gelöstem Säurechlorid ausgegangen wird.
Diese Verbindungen sind neu, ausgenommen O-(2-Aminoäthyl)-fluorenonoxim und -indanon-1-oxim.
Die erfindungsgemäss erhaltenen Verbindungen haben interessante pharmakolegische Eigenschaften. Sie haben insbesondere eine sehr starke zentrale Wirkung, die sich sowohl als eine antidepressive Wirkung, gegebenenfalls infolge einer Monoamino-oxydasenhemmung, als auch als eine sedative oder antikonvulsive Wirkung äussern kann.
Eine starke Wirksamkeit haben insbesondere die Verbindungen, bei denen sowohl R1 als R je ein Wasserstoffatom darstellen.
Die Monoaminooxydasenhemmungswirkung von derartigen Verbindungen wurde bei Versuchen gefunden, bei denen fünf männlichen Albinomäusen eine gewisse Menge der Versuchsverbindung intraperitoneal oder oral verabreicht wurde. Eine Stunde später erhielten die Tiere eine subkutane Einspritzung von Tryptaminhydrochlorid in einer Menge von 250 mg/kg. Diese Menge hat bei Tieren, welche die Versuchsverbindung nicht empfangen hatten, keine, bei den behandelten jedoch eine Sterblichkeit zur Folge. Achtzehn Stunden nach der Verabreichung des Tryptaminhydrochlorides wurde festgestellt, wie viele behandelte Tiere gestorben waren. Aus den Ergebnissen wurde die ED;o bestimmt.
Die antidepressive Wirkung von erfindungsgemäss erhaltenen Verbindungen wurde im Tetrabenazinversuch ermittelt. Bei diesem Versuch wurde 5 männlichen Albinomäusen eine Menge der Versuchsverbindung intraperitoneal oder oral verabreicht. Nach 45 Minuten erhielten die Tiere eine subkutane Einspritzung von 80 mg'log Tetrabenazin. Nach abermals 45 Minuten wurde der Grad der Ptosis bestimmt und mit der Ptosis von Tieren verglichen. die nur Tetrabenazin empfangen hatten. Aus den Ergebnissen wurde die ED50 bestimmt.
Die sedative Wirkung von erfindungsgemäss erhaltenen Verbindungen wurde im Hexobarbitalschlafversuch gefunden. Bei diesem Versuch wurde eine Versuchsverbindung 30 bzw. 60 Minuten vor der Verabreichung einer gerade nicht narkotischen Hexobarbitaldosis (30 rr.g/kg) intravenös bzw. oral verabreicht. Das Kriterium für die Wirksamkeit der Substanz war die Induktion der Narkose. Aus einer Reihe von Versuchen mit wechselnden Dosierungen wurde die ED50 errechnet.
Die antikonvulsive Wirkung von erfindungsgemäss hergestellten Verbindungen gegen maximalen Elektroschock wurde an weiblichen Mäusen 30 Minuten nach intraperilonealer oder 60 Minuten nach oraler Verabreichung der Versuchsverbindung bestimmt.
Die Einwirkung von Verbindungen auf die konvulsive Wirkung einer supramaximalen intravenös verabreichten Dosis Pentamethylentetrazol (50 mg/kg) wurde ebenfalls an weiblichen Mäusen 30 Minuten nach intraperitonealer oder 60 Minuten nach oraler Verabreichung einer Versuchsverbindung bestimmt.
Die antidepressiven Verbindungen gemäss der Erfindung sind besonders geeignet zur Verwendung bei der Therapie neurotischer und psychotischer Störungen, insbesonderc in der Therapie des depressiven Syndroms, und auch für die Behandlung psychosomatischer Störungen. Die Substanzen können denn auch depressiven Patienten als Psychostimulans verabreicht werden.
Die sedativen Verbindungen eignen sich ausgezeichnet zur Anwendung als Ataraktikum. Sie können mit Erfolg bei der Behandlung gelinder psychoneurotiseher Erscheinungen benutzt werden.
Die antikonvulsiven Verbindungen können bei der Behandlung epileptischer Patienten Anwendung finden.
Die Verbindungen können, nachdem sie in eine ge eignet Verabreichungsform gebracht worden sind, auf die übliche Weise verabreicht werden. Sie können eingespritzt, aber auch oral oder rektal verabreicht werden. Es kommen somit unter anderem die folgenden Verabreichungsformen in Betracht: Injektionsflüssigkei- ten, Pillen, Tabletten, Dragees. Kapseln, Pulver und dergleichen.
Die Weise, auf die die Menge in der und die Frequenz mit der die Stoffe dem Patienten verabreicht werden müssen, können bei jedem Patienten verschieden sein, unter anderem in Abhängigkeit vom Ernst der Störungen. Es dürfe den behandelnden Arzt im allgemeinen keine Mühe kosten, für einen bestimmten Patienten die richtige Therapie zu wählen.
Die Dosierung sedativer und antidepressiver Ver bindungen beträgt für Erwachsene im allgemeinen I 10 bis 500 m täglich. In der Regel diirfte eine Menge von 10 bis 150 mg genügen.
Erfindungsgemäss hergestellte Antikonvulsiva werden im allgemeinen in Dosierungen von 100 bis 500 mg täglich verabreicht.
Die erfindungsgemäss erhaltenen Verbindungen können gemäss in der Pharmazie üblichen Verfahren zu Präparaten verarbeitet werden. z. B. dadurch, dass ein Wirkstoff mit festen oder flüssigen Trägermaterialien gemischt oder in ihnen gelöst wird.
Als solche eignen sich die üblichen Träger. z. B.
Wasser, das gegebenenfalls. z. B. durch Kochsalz, isotonisch mit Blut gemacht worden ist, Glyzerin, Kreide, Kalziumphosphat, Milchzucker, Puderzucker und Kalziumkarbonat. Als Quellmittel können in Tabletten und Dragees z. B. Kartoffelstärke, Maisstärke, Pfeilwurzstärke (Amylum marantae), Karboxymethylzellulose, Gelatine und Akaziengummi benutzt werden.
Als Gleitmittel können Talk, Magnesium- und Kalziumstearat sowie Stearinsäure Verwendung finden.
Oral zu verabreichende Präparate können weiter Geschmackstoffe, wie Zucker oder Vanillenextrakt, enthalten.
Als KonservierungsmitteI können z. B. Propyl-phydroxybenzoat und Benzylalkohol benutzt werden. Die Präparate können weiter oberflächenaktive Stoffe enthalten, wie z. B. Mono-, Di- und Triester höherer Fettsäuren.
Als Beispiele pharmazeutisch akzeptabler Säuren, mit denen die Verbindungen Salze bilden können, seien erwähnt: Halogenwasserstoffsäuren, wie Salzsäure, Bromwasserstoffsäure, ferner andere anorganische Säu- ren, wie Schwefelsäure, Salpetersäure und Phosphorsäure und organische Säuren, wie Zitronensäure, Essigsäure, Oxalsäure, Fumarsäure, Milchsäure, Bernsteinsäure, Sulfaminsäure, Benzoesäure, Weinsteinsäure und Gallussäure.
Beispiel 1 O-(-Aminoäthyl)-3-n.propylindanon- l -oxim.HCI
0,030 Mol 3-n-Propylindanon-1-oxim (5,64 g) wurde in eine Lösung von 0,120 Mol Natrium (2,76 g) in 75 cm wasserfreiem Äthanol gegeben. Danach wurde 0,060 Mol 2-Bromäthylaminhydrobromid (12,3 g) zugesetzt. Nachdem dem Gemisch 25 cm3 wasserfreies Äthanol zugesetzt worden war, wurde es 2¸ Stunden bei Zimmertemperatur gerührt. Danach wurde vom gebildeten NaBr-Niederschlag abgesaugt. Das Filtrat wurde im Vakuum eingedampft, bis es lösemittelfrei war, wonach der Rückstand in einem Gemisch aus 50 cm3 Wasser und 50 cm Ather gelöst wurde. Die Schichten wurden getrennt und die Ätherschicht noch einmal mit 25 cm3 Wasser gewaschen.
Dann wurde die Ätherschicht mit 60 cm n-Salzsäure angesäuert. Die saure Wasserschicht wurde abgetrennt, mit 50 cm:3 2n NaOH basisch gemacht und dann 3mal mit je 75 cm Äther extrahiert. Die gesammelten Ätherextrakte wurden nach Trocknen über Na,CO im Vakuum eingedampft. Das erhaltene Öl wurde in 10 ml Äthanol gelöst und mit etwa 3,2n alkoholischer Salzsäure auf pH 4 angesäuert. Nach Zusatz von Diäthyläther kristallisierte ein Stoff mit Schmelzpunkt 117-118 C aus.
Beispiel 2 O-(2-Methylaminoäthyl)-fluorenonoximhydrochlorid
0,300 Mol Natrium (6,90 g) wurde in 200 cm3 wasserfreiem Äthanol gelöst. Der zweiten Lösung wurde 0,075 Mol Fluorenonoxim (14,65 g) zugesetzt. Danach wurde 0.150 Mol 2-Methylaminoäthylchloridhydrochlorid (19.35 g) zugesetzt. Das Gemisch wurde bei Zimmertemperatur 25 Stunden lang gerührt. Dann wurde vom gebildeten NaCI-Niederschlag abgesaugt. Das Filtrat wurde im Vakuum eingedampft, bis das Lösungsmittel entfernt war. Das erhaltene Ö1 wurde in 150 cm3 Wasser und 150 cm3 Äther gelöst und die Wasserschicht wurde abgetrennt. Danach wurde die Atherlösung noch dreimal mit je 50 cm3 Wasser gewaschen.
Die Ätherschicht wurde mit 150 cm 2n HC1-Lösung geschüttelt und die saure Wasserschicht wurde abgetrennt.
Diese Wasserschicht wurde dreimal mit je 30 cm3 Äther gewaschen und dann mit 200 cm3 2n NaOH basisch gemacht. Das dadurch freiwerdende basische Öl wurde dreimal mit je 50 cm3,Ather extrahiert. Die gesammelten Ätherextrakte wurden schliesslich dreimal mit je 25 cm3 Wasser gewaschen; nach Trocknen über Na2CO@ wurde der Ather im Vakuum entfernt. Das erhaltene Öl wurde in 4 cm3 absolutem Äthanol gelöst und mit etwa 4n alkoholischer Salzsäure auf den pH 6 bis 7 angesäuert. Der alkoholischen Lösung wurde Äther zugesetzt, wodurch sich ein kristallinischer Stoff niederschlug.
Dieser wurde eine Stunde stehengelassen, bei + 30 C abgesaugt, mit etwas Äther gewaschen und im Vakuum über festem NaOH getrocknet. Nach zweimaliger Umkristallisierung aus einem Gemisch aus absolutem Alkohol und Äther ergab sich eine Substanz, die unter Zersetzung bei 183,5-184,5 C schmolz.
Beispiel 3 a) 3-Fluor-O-{2-(äthoxycarbonylamin)äthyl}- fluorenonoxim
Eine Lösung von 2,06 g 2-Brom-N-(äthoxycarbonyl)-äthylamin in 6 ml absolutem Äthanol wurde unter Rühren einer Lösung von 1,95 g 3-Fluorfluorenonoxim und 0,24 g Natrium in 18 ml absolutem Äthanol zugesetzt. Das Reaktionsgemisch wurde einige Stunden gerührt und dann 24 Stunden bei Zimmertemperatur stehengelassen. Danach wurde das Gemisch mit Wasser verdünnt und dreimal mit Diäthyläther extrahiert.
Die Ätherschicht wurde über Natriumsulfat getrocknet und das Lösemittel im Vakuum entfernt. Durch fraktionierte Kristallisation des Rückstandes aus Äthanol wurde ein Stoff mit Schmelzpunkt 93,5-94 C erhalten. b) 3-Fluor-O-(2-aminoäthyl)-fluorenonoxim HCI
0,3 g der nach Beispiel 6a erhaltenen Verbindung wurde durch Erhitzung in 5 ml Äthanol gelöst. Dieser Lösung wurde 1,9 ml 3n Natronlauge zugesetzt und das Ganze gut eine Stunde gekocht. Das Reaktionsgemisch wurde dann mit 10 ml Wasser verdünnt und dreimal mit Diäthyläther extrahiert. Der Ätherextrakt wurde danach mit 2,5 ml 1,2n Salzsäure und dann mit etwas Wasser extrahiert. Die erhaltene saure Lösung wurde mit 1,8 ml 3n Natronlauge basisch gemacht und wiederum mit Äther extrahiert.
Der Ätherextrakt wurde über Natriumsulfat getrocknet und dann im Vakuum eingedämpft. Der Rückstand wurde in etwas Äthanol gelöst und die Lösung mit alkoholischer Salzsäure neutralisiert. Hierdurch kristallisierte ein Stoff aus, der abgesaugt, mit einem Gemisch aus Diäthyläther und Äthanol gewaschen und im Vakuum getrocknet wurde.
Schmelzpunkt 216-217,50 C (unter Zersetzung).

One hour later, the animals received a subcutaneous injection of tryptamine hydrochloride in the amount of 250 mg / kg. This amount does not result in any death in animals which did not receive the test compound, but causes death in the treated. The antidepressant effect of compounds obtained according to the invention was determined in the tetrabenazine test. The layers were separated and the ether layer was washed once more with 25 cm3 of water.
Then the ether layer was acidified with 60 cm N hydrochloric acid. The acidic water layer was separated, made basic with 50 cm: 3 2N NaOH and then extracted 3 times with 75 cm ether each time. After drying over Na, CO, the collected ether extracts were evaporated in vacuo. The oil obtained was dissolved in 10 ml of ethanol and acidified to pH 4 with about 3.2N alcoholic hydrochloric acid.
