Abstract

(A) Substituted phenylacetic acids and -esters (I): R1 = H, alkyl, alkenyl or alkinyl; R2 = H, alkyl, alkoxy or halogen; R3, R4 and R5 are H or alkyl. (B) Salts of (I) with inorganic and organic bases. Compounds (I) have analgesic-, antiphlogistic- and antipyretic activity with very favourable therapeutic index. They may be used to alleviate or remove pain as well as in the treatment of rheumatic- and other inflammatory processes. 0.20 mol of (p-aminophenyl) acetic acid (Radiszewski, Ber. 2, 209; Bedson, J.Chem. Soc 37, 92) and 0.20 mol of 2.5- dimethoxytetrahydrofuran are refluxed in 40 ml glacial acetic acid for 30 minutes. After cooling the mixture is poured into 160 ml. water, the crystals filtered off, washed with water and dried for 15 hours at 70 deg.C. The brown powder obtained is extracted with benzene. Evaporation of the extract gives the (p-(1-pyrryl) phenyl) acetic acid.

   Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen, substituierten Phenylessigsäuren und deren Estern, sowie ihrer Salze, mit wertvollen pharmakologischen Eigenschaften.



   Substituierte Phenylessigsäuren und deren Ester entsprechend der Formel I,

 in welcher R2 Wasserstoff, eine niedere Alkyl- oder Alkoxygruppe oder ein Halogenatom und R3, R4 und   Rs    unabhängig voneinander Wasserstoff oder niedere Alkylgruppen bedeuten, sowie Salze der unter die Formel I fallenden Carbonsäuren mit anorganischen und organischen Basen sind bisher nicht beschrieben worden.



   Wie nun gefunden wurde, besitzen diese neuen Stoffe wertvolle pharmakologische Eigenschaften, insbesondere analgetische, antipyretische und antiphlogistische Wirksamkeit mit sehr günstigem therapeutischem Index. Diese Eigenschaften charakterisieren sie als Wirkstoffe zur Linderung und Behebung von Schmerzen verschiedener Genese und zur Behandlung von rheumatischen und andern entzündlichen Krankheitsprozessen. Die Verabreichung kann oral, rektal oder parenteral erfolgen.



   In den Verbindungen der Formel I und den zugehörigen, weiter unten genannten Ausgangsstoffen ist R2 als niedere Alkylgruppe z.B. die Methyl-, Äthyl-, n-Propyl-, Isopropyl-, n-Butyl-, Isobutyl- oder tert.Butylgruppe und als Halogenatom z.B. Chlor, Brom oder Fluor. Niedere Alkylgruppen R3 und R4 sind z.B. Methyl-, Äthyl-, n-Propyl- oder Isopropylgruppen. Eine niedere Alkylgruppe   Rs    ist z.B. die Methyl-,   Athyl-,    n-Propyl-, Isopropyl-, n-Butyl-, Isobutyl-, tert.Butyl-, n-Pentyl-, Isopentyl- oder n-Hexylgruppe.



   Zur Herstellung der Verbindungen der Formel I und der Salze der unter diese Formel fallenden Carbonsäuren mit anorganischen und organischen Basen setzt man eine Verbindung der Formel II

 in welcher R2, R3 und R4 die unter Formel I angegebene Bedeutung haben, in Gegenwart eines Silbersalzes, von Silberoxid, Kupfer oder Platin als Katalysator mit Wasser oder einem niederen Alkanol um, und führt gewünschtenfalls eine erhaltene Carbonsäure in ein Salz mit einer anorganischen oder organischen Base über. Beispielsweise fügt man eine Lösung des Diazoketons der Formel II langsam zu einer wässrigen Lösung von Silbernitrat und Natriumthiosulfat oder zu einer Suspension von Silberoxid in einer wässrigen Lösung von   Natriumthiosulfat,    wobei die Temperatur der wässrigen Lösung bzw.

  Suspension ca.   60-700beträgt.    Gemäss einer vorteilhaften Verfahrensmodifikation versetzt man eine Lösung des Diazoketons in einem niederen Alkanol bei dessen Siedetemperatur portionenweise mit frisch bereitetem Silberoxid, bis keine Stickstoffentwicklung mehr festzustellen ist.



  Ausgangsstoffe der Formel II sind durch Umsetzung von entsprechend der Definition für R2, R3 und R4 substituiertem p (I-Pyrryl)-benzoylchlorid mit Diazomethan in an sich bekannter Weise herstellbar. Die benötigten Säurechloride sind ihrerseits durch Kondensation von gegebenenfalls entsprechend der Definition für R2 substituierter p-Aminobenzoesäure mit einer Verbindung der Formel III

 in welcher höchstens zwei R jeder Einheit niedere Alkylgruppen und die übrigen Wasserstoffatome bedeuten, X und X unabhängig voneinander Reste der Formeln R6-Ound R6-CO-O-, in welchen R6 für einen gegebenenfalls halogen-substituierten Kohlenwasserstoffrest steht, ferner Chloroder Bromatome, oder X zusammen mit Y auch den Oxorest = 0, Y und Y unabhängig voneinander Reste der vorstehend definierten Formeln R6-O- oder R6-CO-O- oder beide zusammen den Epoxyrest -0-, oder jedes zusammen mit Z bzw.

  Z je eine zusätzliche Bindung entsprechend den punktierten Linien, und Z und Z Wasserstoffatome bedeuten, sofern sie nicht die vorgenannte Bedeutung haben, und n Null oder eine niedere ganze Zahl bedeutet, und anschliessende Behandlung der erhaltenen, gegebenenfalls substituierten p-(1-Pyrryl)-benzoesäure mit Thionylchlorid in Gegenwart eines säurebindenden Mittels, wie z.B. Pyridin, oder mit Oxalylchlorid erhältlich.



   Als gewünschtenfalls herstellbare Salze von unter die Formel I fallenden Carbonsäuren seien z.B. die Natrium-, Kalium-, Lithium-, Magnesium-, Calcium- und Ammoniumsalze, sowie Salze mit Äthylamin, Triäthylamin, 2-Amino-äthanol,   2,2'-    Imino-diäthanol, 2-Dimethylamino-äthanol, 2-Diäthylamino äthanol, Äthylen-diamin, Benzylamin, Procain, Pyrrolidin, Piperidin, Morpholin, 1-Äthyl-piperidin oder 2-Piperidino äthanol oder mit basischen Ionenaustauschern genannt.



   Die neuen Verbindungen der Formel I sowie die Salze der unter diese Formel fallenden freien Säuren werden, wie weiter vorne erwähnt, peroral, rektal oder parenteral verabreicht. Die täglichen Dosen bewegen sich zwischen 50 und 3000 mg für erwachsene Patienten. Geeignete Doseneinheitsformen, wie Dragees, Tabletten, Suppositorien oder Ampullen, enthalten als Wirkstoff vorzugsweise 10-500 mg einer Verbindung der Formel I oder eines Salzes einer unter diese Formel fallenden freien Säure mit einer pharmazeutisch annehmbaren anorganischen oder organischen Base.



   In Doseneinheitsformen für die perorale Anwendung liegt der Gehalt an Wirkstoff vorzugsweise zwischen 10% und 90%.



  Zur Herstellung solcher Doseneinheitsformen kombiniert man den Wirkstoff z.B. mit festen, pulverförmigen Trägerstoffen, wie Lactose, Saccharose, Sorbit, Mannit; Stärken, wie Kartoffelstärke, Maisstärke oder Amylopektin, ferner Laminariapulver oder Citruspulpenpulver; Cellulosederivaten oder Gelatine, gegebenenfalls unter Zusatz von Gleitmitteln, wie Magnesium- oder Calciumstearat oder Polyäthylenglykolen, zu Tabletten oder zu Dragee-Kernen. Letztere überzieht man beispielsweise mit konzentrierten Zuckerlösungen, welche z.B.



  noch arabischen Gummi, Talk und/oder Titandioxid enthalten können, oder mit einem in leichtflüchtigen organischen Lösungsmitteln oder Lösungsmittelgemischen gelösten Lack.



  Diesen Überzügen können Farbstoffe zugefügt werden, z.B.



  zur Kennzeichnung verschiedener Wirkstoffdosen. Als weitere orale Doseneinheitsformen eignen sich Steckkapseln aus Gelatine sowie weiche, geschlossene Kapseln aus Gelatine und einem Weichmacher, wie Glycerin. Die erstern enthalten den Wirkstoff vorzugsweise als Granulat in Mischung mit Gleitmitteln, wie Talk oder Magnesiumstearat, und gegebenenfalls Stabilisatoren, wie Natriummetabisulfit   (Na2S2Os)    oder Ascorbinsäure. In weichen Kapseln ist der Wirkstoff vorzugsweise in geeigneten Flüssigkeiten, wie flüssigen Polyäthylenglykolen, gelöst oder suspendiert, wobei ebenfalls Stabilisatoren zugefügt sein können.



   Als Doseneinheitsformen für die rektale Anwendung kommen z.B. Suppositorien in Betracht, welche aus einer Kombination eines Wirkstoffes mit einer Suppositorien-Grundmasse auf der Basis von natürlichen oder synthetischen Triglyceriden (z.B. Kakaobutter), Polyäthylenglykolen oder geeigneten höheren Fettalkoholen bestehen, und Gelatine-Rektalkapseln, welche eine Kombination des Wirkstoffs mit Polyäthylenglykolen enthalten.



   Ampullenlösungen zur parenteralen, insbesondere intramuskulären oder intravenösen Verabreichung enthalten z.B. eine Verbindung der Formel I in einer Konzentration von vorzugsweise   0,5-5 %    als wässrige, mit Hilfe von üblichen Lösungsvermittlern und/oder Emulgiermitteln sowie gegebenenfalls von Stabilisierungsmitteln bereitete Dispersion, oder eine wässrige Lösung eines pharmazeutisch annehmbaren, wasserlöslichen Salzes einer unter die Formel I fallenden freien Säure.



   Als weitere parenterale Applikationsformen kommen beispielsweise mit den üblichen Hilfsstoffen bereitete Lotions, Tinkturen und Salben für die perkutane Anwendung in Betracht.



   Die folgenden Vorschriften sollen die Herstellung von Tabletten, Dragees usw. näher erläutern: a) 1000 g Wirkstoff, z.B.   [p-(1 -Pyrryl)-phenyl]-essigsäure    werden mit 550 g Lactose und 292 g Kartoffelstärke vermischt, die Mischung mit einer alkoholischen Lösung von 8 g
Gelatine befeuchtet und durch ein Sieb granuliert. Nach dem Trocknen mischt man 60 g Kartoffelstärke, 60 g Talk, 10 g Magnesiumstearat und 20 g hochdisperses Siliciumdioxid zu und presst die Mischung zu 10 000 Tabletten von je 200 mg
Gewicht und 100 mg Wirkstoffgehalt, die gewünschtenfalls mit Teilkerben zur feineren Anpassung der Dosierung versehen sein können.

 

   b) 200 g Wirkstoff, z.B.   [p-(1-Pyrryl)-phenyl}-essigsäure    werden mit 16 g Maisstärke und 6 g hochdispersem Silicium dioxid gut vermischt. Die Mischung wird mit einer Lösung von
2 g Stearin-Säure, 6 g Äthylcellulose und 6 g Stearin in ca.



   70 ml Isopropylalkohol befeuchtet und durch ein Sieb III (Ph.



   Helv. V) granuliert. Das Granulat wird ca. 14 Stunden getrocknet und dann durch Sieb-III-IIIa geschlagen. Hierauf wird es mit 16 g Maisstärke, 16 g Talk und 2 g Magnesium stearat vermischt und zu 1000 Dragee-Kernen gepresst. Diese werden mit einem konzentrierten Sirup von 2 g Lacca, 7,5 g arabischem Gummi, 0,15 g Farbstoff, 2 g hochdispersem Siliciumdioxid, 25 g Talk und 53,35 g Zucker überzogen und getrocknet. Die erhaltenen Dragees wiegen 360 mg und enthalten je 200 mg Wirkstoff.



   c) 50,0 g [p-(1-Pyrryl)-phenyl]-essigsäure werden in einem   Gemisch von 232 ml   1n    Natronlauge und 500 ml ausgekochtem, pyrogenfreiem Wasser gelöst und die Lösung mit ebensolchem Wasser auf 2000 ml ergänzt. Die Lösung wird filtriert, in 1000 Ampullen à 2 ml abgefüllt und sterilisiert. Eine Ampulle à 2 ml enthält 50 mg 2-   [p-(1-Pyrryl)-phenylj-essigsäure    als Wirkstoff in Form des Natriumsalzes.



   d) 50 g   [p-(1-Pyrryl)-phenyl]-essigsäure    und 1950 g fein geriebene Suppositoriengrundmasse (z.B. Kakaobutter) werden gründlich gemischt und dann geschmolzen. Aus der durch Rühren homogen gehaltenen Schmelze werden 1000 Suppositorien von 2,0 g gegossen. Sie enthalten je 50 mg Wirkstoff.



   e) 60,0 g Polyoxyäthylensorbitan-monostearat, 30,0 g Sorbitan-monostearat, 150,0 g Paraffinöl und 120,0 g Stearylalkohol werden miteinander   geschmolzen, 50,0    g   [p-(1-Pyrryl)-    phenyl]-essigsäure (fein pulverisiert) werden zugegeben und 590 ml auf   40D    vorgewärmtes Wasser einemulgiert. Die Emulsion wird bis zum Erkalten auf Raumtemperatur gerührt und in Tuben abgefüllt.



   Die nachfolgenden Beispiele erläutern die Herstellung der neuen Verbindungen der Formel I näher. Die Temperaturen sind in Celsiusgraden angegeben.



   Beispiel 1
7,4 g p-(1-Pyrryl)-benzoesäure werden mit 3,3 ml Thionylchlorid und 4,8 g Pyridin in 100 ml Methylenchlorid 3 Stunden in einer Stickstoffatmosphäre unter Rückfluss zum Sieden erhitzt. Dann wird das Reaktionsgemisch unter Vakuum eingedampft und der Rückstand mit 100 ml Tetrahydrofuran versetzt, wobei ein Teil ungelöst bleibt. Die erhaltene Suspension von rohem Säurechlorid wird unter Eiskühlung innerhalb einer Stunde zu einer Mischung von   200    ml 0.6n ätherischer Diazomethanlösung und 50 ml Dioxan getropft und das Ganze anschliessend 18 Stunden bei Raumtemperatur gerührt. So entsteht eine Lösung, die durch Filtration durch Hyflo (Diatomeenerde) von einer geringen Trübung befreit und bei   30O    unter reduziertem Druck eingedampft wird.



   Das zurückbleibende, rohe   4-(1-Pyrryl)-2-diazoacetophe-    non wird in 180 ml abs. Methanol gelöst, die Lösung unter Rückfluss zum Sieden erhitzt und portionenweise mit Silberoxid (aus 2 g Silbernitrat hergestellt und mit Methanol gewaschen) versetzt, bis keine Gasentwicklung mehr festzustellen ist, was ca. 4 Stunden dauert. Der Silberniederschlag wird abfiltriert, das Filtrat unter Vakuum eingedampft. Zurück bleibt der teilweise kristalline rohe   [p-(1-pyrryl)-phenyl]-essigsäure-methylester.   



   Die als Ausgangsstoff verwendete   p-(1-Pyrryl)-benzoesäure    wird wie folgt erhalten: a) Der   p-( 1 -Pyrryl)-benzoesäure-äthylester    wird aus 2,5 Dimethoxytetrahydrofuran und 4-Amino-benzoesäureäthylester hergestellt, Smp.   75-760    (aus Methanol), Kp.   140-145o1      0.01    Torr.



   b) Eine Lösung von 32 g   p-(1-Pyrryl)-benzoesäure-äthyl-    ester und 12 g Natriumhydroxid in 600 ml Äthanol wird 2 Stunden unter Rückfluss zum Sieden erhitzt, wobei allmählich ein kristalliner Niederschlag entsteht. Anschliessend wird das Reaktionsgemisch unter vermindertem Druck auf ca. 200 ml eingeengt und das ausgefallene Natriumsalz durch Zugabe von 200 ml Wasser in Lösung gebracht. Unter Rühren wird diese Lösung mit Eisessig langsam auf pH 5 gebracht, wobei die gewünschte   p-(1-Pyrryl)-benzoesäure    als feiner, dicker Brei ausfällt. Der Niederschlag wird abgenutscht, mit 100 ml Wasser portionenweise gewaschen und im Vakuumtrockenschrank 7 Stunden bei 70 getrocknet. Die so erhaltene p-(1-Pyrryl)benzoesäure schmilzt bei   265-270O    unter Zersetzung.

 

   Beispiel 2
5,7 g   [p-(1-Pyrryl)-phenyl]-essigsäure    werden in 40 ml Isopropanol suspendiert. Durch Zugabe von 8 ml Triäthylamin erhält man eine homogene Lösung. Diese wird mit 20 ml Äther versetzt und filtriert. Nach Zugabe von soviel Petrol äther (Kp.   40-600),    dass die entstehende Trübung sich noch löst, kristallisiert das Salz beim Abkühlen allmählich. Nach Trocknen bei 200 Torr während 12 Stunden schmilzt das Triäthylammonium-Salz der   [p-( 1 -Pyrryl)-phenylj-essigsäure    bei   67-73.

   In unit dose forms for oral use, the content of active ingredient is preferably between 10% and 90%.



  To produce such unit dosage forms, the active ingredient is combined e.g. with solid, powdery carriers such as lactose, sucrose, sorbitol, mannitol; Starches, such as potato starch, corn starch or amylopectin, also laminaria powder or citrus pulp powder; Cellulose derivatives or gelatin, optionally with the addition of lubricants, such as magnesium or calcium stearate or polyethylene glycols, to tablets or to dragee cores. The latter is coated, for example, with concentrated sugar solutions, which e.g.



  Arab gum, talc and / or titanium dioxide, or with a paint dissolved in volatile organic solvents or solvent mixtures.



  Dyes can be added to these coatings, e.g.



  for labeling different doses of active ingredients. Push-fit capsules made of gelatin and soft, closed capsules made of gelatin and a plasticizer, such as glycerine, are suitable as further oral unit forms. The former contain the active ingredient preferably as granules mixed with lubricants such as talc or magnesium stearate, and optionally stabilizers such as sodium metabisulphite (Na2S2Os) or ascorbic acid. In soft capsules, the active ingredient is preferably dissolved or suspended in suitable liquids, such as liquid polyethylene glycols, it also being possible for stabilizers to be added.



   Unit forms for rectal use are e.g. Consider suppositories, which consist of a combination of an active ingredient with a suppository base based on natural or synthetic triglycerides (e.g. cocoa butter), polyethylene glycols or suitable higher fatty alcohols, and gelatin rectal capsules which contain a combination of the active ingredient with polyethylene glycols.



   Ampoule solutions for parenteral, in particular intramuscular or intravenous administration contain e.g. a compound of the formula I in a concentration of preferably 0.5-5% as an aqueous dispersion prepared with the aid of customary solubilizers and / or emulsifiers and optionally stabilizers, or an aqueous solution of a pharmaceutically acceptable, water-soluble salt of one of the formula I falling free acid.



   Other parenteral forms of administration that can be used are, for example, lotions, tinctures and ointments for percutaneous use prepared with the usual auxiliaries.



   The following instructions are intended to explain the production of tablets, coated tablets, etc. in more detail: a) 1000 g of active ingredient, e.g. [p- (1 -Pyrryl) -phenyl] -acetic acid are mixed with 550 g of lactose and 292 g of potato starch, the mixture with an alcoholic solution of 8 g
Gelatin moistened and granulated through a sieve. After drying, 60 g of potato starch, 60 g of talc, 10 g of magnesium stearate and 20 g of highly dispersed silicon dioxide are mixed in and the mixture is compressed to form 10,000 tablets of 200 mg each

   b) 200 g of active ingredient, e.g. [p- (1-Pyrryl) -phenyl} -acetic acid are mixed well with 16 g of corn starch and 6 g of highly dispersed silicon dioxide. The mixture is made with a solution of
2 g stearic acid, 6 g ethyl cellulose and 6 g stearin in approx.



   70 ml of isopropyl alcohol moistened and filtered through a sieve III (Ph.



   Helv. V) granulated. The granulate is dried for about 14 hours and then passed through Sieve III-IIIa. It is then mixed with 16 g of corn starch, 16 g of talc and 2 g of magnesium stearate and pressed into 1000 coated tablets. These are coated with a concentrated syrup of 2 g Lacca, 7.5 g gum arabic, 0.15 g color, 2 g highly dispersed silicon dioxide, 25 g talc and 53.35 g sugar and dried. The dragees obtained weigh 360 mg and each contain 200 mg of active ingredient.



   c) 50.0 g of [p- (1-pyrryl) phenyl] acetic acid are dissolved in a mixture of 232 ml of 1N sodium hydroxide solution and 500 ml of boiled, pyrogen-free water and the solution is made up to 2000 ml with the same water. The solution is filtered, filled into 1000 ampoules of 2 ml each and sterilized. A 2 ml ampoule contains 50 mg 2- [p- (1-pyrryl) -phenylj-acetic acid as the active ingredient in the form of the sodium salt.



   d) 50 g of [p- (1-pyrryl) -phenyl] -acetic acid and 1950 g of finely grated suppository base (e.g. cocoa butter) are mixed thoroughly and then melted. 1000 suppositories of 2.0 g are poured from the melt, which is kept homogeneous by stirring. They each contain 50 mg of active ingredient.



   e) 60.0 g of polyoxyethylene sorbitan monostearate, 30.0 g of sorbitan monostearate, 150.0 g of paraffin oil and 120.0 g of stearyl alcohol are melted together, 50.0 g of [p- (1-pyrryl) phenyl] acetic acid (finely powdered) are added and 590 ml of water preheated to 40D are emulsified. The emulsion is stirred until it cools to room temperature and filled into tubes.



   The following examples explain the preparation of the new compounds of the formula I in more detail. The temperatures are given in degrees Celsius.



   example 1
7.4 g of p- (1-pyrryl) -benzoic acid are refluxed with 3.3 ml of thionyl chloride and 4.8 g of pyridine in 100 ml of methylene chloride for 3 hours in a nitrogen atmosphere. The reaction mixture is then evaporated in vacuo and 100 ml of tetrahydrofuran are added to the residue, some of which remains undissolved. The resulting suspension of crude acid chloride is added dropwise to a mixture of 200 ml of 0.6N ethereal diazomethane solution and 50 ml of dioxane, while cooling with ice, and the whole is then stirred for 18 hours at room temperature. The result is a solution that is freed from a slight cloudiness by filtration through Hyflo (diatomaceous earth) and evaporated at 30 ° under reduced pressure.



   The remaining, crude 4- (1-pyrryl) -2-diazoacetophenon is abs in 180 ml. Dissolved methanol, the solution heated to boiling under reflux and added in portions with silver oxide (made from 2 g of silver nitrate and washed with methanol) until no more gas evolution can be detected, which takes about 4 hours. The silver precipitate is filtered off and the filtrate is evaporated in vacuo. The partially crystalline crude [p- (1-pyrryl) -phenyl] -acetic acid methyl ester remains.



   The p- (1-pyrryl) -benzoic acid used as starting material is obtained as follows: a) The p- (1-pyrryl) -benzoic acid ethyl ester is prepared from 2,5 dimethoxytetrahydrofuran and 4-amino-benzoic acid ethyl ester, melting point 75- 760 (from methanol), b.p. 140-145o1 0.01 torr.



   b) A solution of 32 g of p- (1-pyrryl) -benzoic acid ethyl ester and 12 g of sodium hydroxide in 600 ml of ethanol is heated to boiling under reflux for 2 hours, a crystalline precipitate gradually forming. The reaction mixture is then concentrated to approx. 200 ml under reduced pressure and the precipitated sodium salt is dissolved by adding 200 ml of water. While stirring, this solution is slowly brought to pH 5 with glacial acetic acid, the desired p- (1-pyrryl) -benzoic acid precipitating as a fine, thick paste. The precipitate is filtered off with suction, washed in portions with 100 ml of water and dried at 70 for 7 hours in a vacuum drying cabinet. The p- (1-pyrryl) benzoic acid obtained in this way melts at 265-270 ° with decomposition.

 

   Example 2
5.7 g of [p- (1-pyrryl) phenyl] acetic acid are suspended in 40 ml of isopropanol. A homogeneous solution is obtained by adding 8 ml of triethylamine. This is mixed with 20 ml of ether and filtered. After adding so much petroleum ether (bp 40-600) that the resulting cloudiness still dissolves, the salt gradually crystallizes on cooling. After drying at 200 torr for 12 hours, the triethylammonium salt of [p- (1-pyrryl) -phenylj-acetic acid melts at 67-73.

PATENTANSPRUCH PATENT CLAIM Verfahren zur Herstellung von substituierten Phenylessigsäuren und deren Estern entsprechend der Formel I EMI3.1 in welcher R2 Wasserstoff, eine niedere Alkyl- oder Alkoxygruppe oder ein Halogenatom und R3, R4 und Rs unabhängig voneinander Wasserstoff oder niedere Alkylgruppen bedeuten, sowie von Salzen der unter die Formel I fallenden Carbonsäuren mit anorganischen und organischen Basen, dadurch gekennzeichnet, dass man eine Verbindung der Formel II EMI4.1 in welcher R2, R3 und R4 die oben angegebene Bedeutung haben, in Gegenwart eines Silbersalzes, von Silberoxid, Kupfer oder Platin als Katalysator mit Wasser oder einem niederen Alkanol umsetzt, und gewünschtenfalls eine erhaltene Carbonsäure in ein Salz mit einer anorganischen oder organischen Base überführt.
