Description
Es ist bereits bekannt, das 7-Acetonyl-1,3- bzw. das 1 Acetonyl-3,7-dimethylxanthin durch Umsetzung des 1,3- bzw.
des 3,7-Dimethylxanthins mit Halogenaceton in Gegenwart von Alkalihydroxyd herzustellen. Auch das 7-(3'-Oxobutyl) 1,3-dimethylxanthin, das aus 1,3-Dimethylxanthin und Methyl-ss-diäthylaminoäthylketon erhalten wird, ist bereits beschrieben. Die Carbonylgruppe ist in diesen bekannten Verbindungen über ein bzw. zwei Kohlenstoffatome mit dem Xanthingerüst verbunden.
Gegenstand der Erfindung ist demgegenüber ein Verfahren zur Herstellung von 1-[((ss-1)-Oxoalkyl]-dimethylxanthinen der Formel

worin X, falls die -X-CO-CH3-Gruppe in 7-Stellung sitzt, eine Alkylengruppe mit 3 bis 6 C-Atomen, oder, falls die -X-CO CH3-Gruppe in 1-Stellung sitzt, eine Alkylengruppe mit 2 bis 5 C-Atomen bedeutet, wobei die Ketogruppe mindestens 2 C Atome vom Xanthingerüst entfernt ist.
Die vorstehend ausgeschlossenen Verbindungen (X = Alkylen mit 1 oder 2 C-Atomen bzw. X = Alkylen mit 1 C Atom) weisen, wie überraschenderweise gefunden wurde, eine sehr viel geringere Wasserlöslichkeit auf als die übrigen Verbindungen, so dass sie therapeutisch nicht universell anwendbar sind. Demgegenüber sind die nach dem erfindungsgemässen Verfahren hergestellten Verbindungen sowohl in Lipoiden als auch im Wasser gut löslich und ausserdem ist ihre Gefässwirkung gegenüber den bekannten mit einer kürzeren Alkylengruppe besser.
Das erfindungsgemässe Verfahren lässt sich in einfacher Weise durchführen, indem bei erhöhter Temperatur ein Dimethylxanthin der Formel

oder dessen Alkalimetallsalze mit einer Verbindung der Formel III

umgesetzt wird, wobei Y eine a,ss-ungesättigte Gruppe H2C=CR-, eine a,l3-Dialkylaminoäthylgruppe mit 1 oder 2 Kohlenstoffatomen in der Alkylgruppe und X = 0 ist oder wobei Y Brom oder Chlor bedeutet und X die für Formel I angegebene Bedeutung hat, und R in der ungesättigten Gruppe bei Umsetzung mit dem 1,3-Dimethylxanthin ein n Alkylrest mit 1 bis 4 Kohlenstoffatomen und bei Umsetzung mit dem 3,7-Dimethylxanthin Wasserstoff oder ein n-Alkylrest mit 1 bis 3 Kohlenstoffatomen ist.
In der oben genannten Formel I kann X vorzugsweise in 7 Stellung angeordnet und eine Alkylgruppe mit 3 bis 5 C Atomen oder in 1-Stellung und eine Alkylengruppe mit 2 bis 4 C-Atomen sein. Ferner kann der Rest X unverzweigt oder auch eine Äthylengruppe darstellen, wobei im letztgenannten Falle das der Carbonylgruppe benachbarte C-Atom eine Alkylseitengruppe trägt. Ferner ist es ausserdem möglich, dass der Rest R in der ungesättigten Gruppe der Verbindung gemäss Formel III und die Alkylseitengruppe in dem Rest X Methyl oder Äthyl sind.
Das erfindungsgemässe Verfahren wird zweckmässig mit einem a,ss-ungesättigten Methylketon in alkalischem Medium durchgeführt. Möglicherweise lässt sich die Umsetzung auch in wässriger-organischer Lösung durchführen. Dies ist vor allem bei der Umsetzung mit dem Methylketon zweckmässig. Wenn ein ungesättigtes Methylketon in wässrig-organischer Lösung umgesetzt wird, arbeitet man mit Vorteil mit starken Alkalien.
Als geeignete Lösungsmittel kommen mit Wasser mischbare Verbindungen in Frage, z.B. Methanol, Äthanol, Propanol, Isopropanol oder die verschiedenen Butanole, Aceton, Pyridin, mehrwertige Alkohole, wie Äthylenglykol, ferner Äthy- lenglykolmonomethyl- oder -äthyläther.
Wenn bei dem erfindungsgemässen Verfahren die Umsetzung mit dem Oxoalkylbromid oder -chlorid erfolgt, verwendet man als Reaktionspartner vorteilhaft ein Alkalisalz des Dime thylxanthins. Die Alkalisalze können als solche eingesetzt werden. Vorteilhaft erzeugt man sie aber im Reaktionsge misch.
Als Verbindungen gemäss Formel III werden insbesondere a,ss-ungesättigte Methylketone, a,B-Dialkylaminoäthyl- methylketone und (o - 1)-Oxoalkylhalogenide genannt.
Die genannten Umsetzungen werden in an sich bekannter
Weise im allgemeinen bei einer Temperatur von 5O15OOC, vorzugsweise von 60-1200C, gegebenenfalls bei erhöhtem oder vermindertem Druck, aber gewöhnlich bei Atmosphärendruck durchgeführt. Die einzelnen Komponenten können in stöchio metrischen oder - aus wirtschaftlichen Gründen - auch in nichtstöchiometrischen Mengen verwendet werden.
Soweit man nach der vorgenannten Verfahrensweise die
Oxohexylverbindung herstellt, kann man das für die Reaktion benötigte (co - 1)-Oxohexylhalogenid durch Kondensation des
1,3-Dihalogenpropans, insbesondere des 1,3-Dibrompropans mit einem Mol Acetessigester und zwei Mol metallischem
Natrium in Gegenwart von absolutem Alkohol erhalten. Das dabei erhaltene 2-Methyl-3-carbäthoxydihydropyran der

wurde dann durch Halogenwasserstoff, meistens Bromwasserstoff, zum 1-Bromhexanon-5 gespalten, das seinerseits mit einem Alkalimetallsalz des Theobromins bzw. Theophyllins in der angegebenen Weise umgesetzt wurde.
Wie weiter gefunden wurde, kann man das 1-(5!-Oxohexyl)-3,7- bzw.
7-(5'-Oxohexyl)-1 ,3 -dimethylxanthin in wesentlich einfacherer Weise wie folgt erhalten: Man kondensiert zunächst ein 1,3-Dihalogenpropan mit einem Mol Acetessigester statt mittels metallischem Natrium mittels mindestens zwei Mol, vorzugsweise wasserfreiem, Kaliumcarbonat in mindestens 90%igem, vorzugsweise dem üblichen 96%gen Alkohol bei 60 bis llO"C, vorzugsweise unter Rückfluss. Als Dihalogenpropane kommen solche in Frage, in denen das Halogen ein Atomgewicht von mindestens 35 hat, z.B. das 1 Brom-3-chlor-propan und vorzugsweise das 1,3-Dibrompropan. Die Anwendung eines Überschusses an Kaliumcarbonat oder Dihalogenpropan ist zwar möglich, bringt jedoch im allgemeinen keine besonderen Vorteile mit sich.
Das anfallende 2-Methyl-3-carbäthoxy-5,6-dihydropyran wird dann in üblicher Weise, vorzugsweise durch Destillation, gereinigt und in an sich bekannter Weise mit mindestens zwei Mol Halogenwasserstoff, vorzugsweise Bromwasserstoff, z.B. unter Rückfluss in das 1-Halogenhexanon-5 übergeführt. Diese Reaktion lässt sich jedoch auch bei gewöhnlicher Temperatur, z.B. von 030OC, durchführen, wenn man statt der üblichen etwa 50%igen wässrigen Bromwasserstofflösung eine konzentrierte, z.B. mindestens 60%ige Lösung verwendet. Das Halogenhexanon wird dann mit einem Alkalimetallsalz des Theobromins bzw. Theophyllins in der angegebenen Weise zum 1-(5/-Oxohexyl)-3,7- bzw.
7-(5'-Oxohexyl)- 1 ,3-dimethylxanthin umgesetzt.
Es ist ein Vorteil der vorliegenden Ausführungsform, dass das Arbeiten mit metallischem Natrium vermieden wird. Ausserdem kann statt des besonders herzustellenden absoluten Alkohols der gewöhnliche Alkohol verwendet werden. Dabei kann die Reaktion in einer wesentlich geringeren Menge Alkohol ausgeführt werden als die bekannte Kondensation. Ferner wird die Aufarbeitung des 2-Methyl-3-carbäthoxy-5,6-dihy- dropyrans dadurch erleichtert, dass die im Reaktionsgemisch vorhandenen anorganischen Salze eine aussalzende Wirkung auf das gewünschte Reaktionsprodukt ausüben. Schliesslich kann der wiedergewonnene Alkohol ohne weitere Reinigung oder Entwässerung für die vorliegende Reaktion wiederverwendet werden.
Die erfindungsgemäss hergestellten Verbindungen zeichnen sich durch eine ausgeprägte gefässerweiternde Wirkung bei geringer Toxizität aus. Sie haben die überraschende Eigenschaft, gut in Lipoiden und Wasser löslich zu sein. Es waren bisher keine leicht wasserlöslichen Theophyllin- bzw. Theobromin-Derivate bekannt, die gleichzeitig eine gute Lipoidlöslichkeit haben. Aufgrund der günstigen Löslichkeitsverhältnisse sind die Substanzen in der therapeutischen Anwendung universell verwendbar.
Beispiel 1
A) Eine Mischung von 560 g Kaliumcarbonat, 700 ml Äthanol (96 %mg), 404 g 1,3-Dibrompropan und 260 g Acetessigsäureäthylester wird unter Rühren bis 60OC aufgeheizt. Es setzt eine Reaktion ein, nach deren Abklingen das Reaktionsgemisch 5 Stunden unter Rückfluss erhitzt wird. Anschliessend wird die Hauptmenge des Alkohols unter gewöhnlichem Druck abdestilliert und der Rückstand mit 1,5 Liter Wasser versetzt; dabei scheidet sich ein Öl ab, welches abgetrennt wird. Die wässrige Phase wird mit Benzol extrahiert und die Benzolschicht mit dem Öl vereinigt. Nach dem Trocknen mit Natriumsulfat wird das Benzol abdestilliert und der Rückstand fraktioniert. Man erhält 250 g 2-Methyl-3-carbäthoxy-5,6dihydropyran vom Kp14 105-1080C (73 % der Theorie).
In 128 g 2-Methyl-3-carbäthoxy-5,6-dihydropyran werden bei Zimmertemperatur langsam 140 ml 63 %ige Bromwasserstoffsäure eingetragen. Es setzt zunächst eine starke Kohlensäureentwicklung ein. Nach 1-2-tägigem Stehen bei Raumtemperatur wird mit dem gleichen Volumen Eiswasser verdünnt; das sich abscheidende dunkel gefärbte Öl wird abge trennt, die wässrige Phase mit Chloroform extrahiert, der Extrakt mit dem t)l vereinigt und mit gesättigter Natriumbicarbonat-Lösung gewaschen. Die Lösung wird mit Natriumsulfat getrocknet, Chloroform unter Normaldruck abdestilliert und der Rückstand im Vakuum fraktioniert. Man erhält 109 g 1-Bromhexanon-5 vom Kpl2 94-98 C (81% der Theorie).
Die Lösung von 10,0 g 1-Bromhexanon-5 in 100 ml Äthanol wird in der Siedehitze unter kräftigem Rühren allmählich mit 11,3 g Theophyllin-Natrium in 100 ml Wasser versetzt.
Nach dreistündigem Erhitzen unter Rückfluss wird der Alkohol abdestilliert, die zurückbleibende wässrige Phase nach Abkühlen alkalisch gemacht und mit Chloroform extrahiert.
Nach Umkristallisation des Rückstandes der Chloroformlösung aus wenig Isopropanol wird 7-(5i-Oxohexyl)-1,3-dimethylxan- thin vom Schmelzpunkt 75-760C in etwa 80 %iger Ausbeute (bezogen auf umgesetztes Theophyllin) erhalten.
B) Geht man von 26,0 g 1-Brompentanon-4, gelöst in 100 ml Äthanol, 31,0 g Theophyllin-Natrium in 175 ml Wasser aus und arbeitet sonst in gleicher Weise, so erhält man nach Chromatographie über Kieselgel mit einem Gemisch aus Chloroform und Äthanol (Raumverhältnis 9:1) und Umkristallisation der Hauptfraktion aus Cyclohexan-Essigsäureäthylester (Raumverhältnis etwa 4:1) ganz analog das 7-(4/-Oxopentyl)-1,3-dimethylxanthin vom Schmelzpunkt 86-880C.
Beispiel 2
A) Die Lösung von 25,0 g 2-Äthylbutenon in 50 ml absolutem Äthanol wird in der Siedehitze unter Rühren allmählich mit einer Lösung von 25,0 g Theophyllin und 13 ml n-Natronlauge in 150 ml eines Gemisches von Äthanol und Wasser (Raumverhältnis 1:1) versetzt. Nach dreistündigem Erhitzen unter Rückfluss wird der Alkohol abdestilliert, die zurückbleibende wässrige Phase alkalisch gemacht und mit Chloroform extrahiert. Nach Umkristallisation des Rückstandes der Chloroformlösung aus Isopropanol wird 7- (2'-Äthyl-3'-oxobutyl)- 1,3-dimethylxanthin vom Schmelzpunkt l071080C in etwa 60!7ciger Ausbeute erhalten.
B) Geht man von 33,0 g 2-Methylbutenon, gelöst in 50 ml Äthanol und einer Lösung von 38,5 g Theophyllin und 20 ml n-Natronlauge in 200 ml eines Gemisches aus Äthanol und Wasser (Raumverhältnis 1:1) aus und arbeitet sonst in gleicher Weise, so erhält man analog das 7-(2 -Methyl-3'-oxobutyl)-
1,3-dimethylxanthin vom Schmelzpunkt 133-1350C.
Beispiel 3
1,8 g Theobromin, 0,8 g Methylvinylketon und 15 ml trokkenes Pyridin werden 3 Stunden unter Rückfluss erhitzt. Das Lösungsmittel wird in Vakuum entfernt und der Rückstand aus Isopropanol umkristallisiert. Man erhält 1 -(3'-Oxobutyl)-3,7- dimethylxanthin vom Schmelzpunkt 144-1460C in etwa 70zeiger Ausbeute.
Beispiel 4
1,8 g Theobromin, 5,0 g 2-Methylbutenon, 1,0 ml n Natronlauge und 50 ml eines Gemisches von Äthanol und Wasser (Raumverhältnis 1:1) werden 3 Stunden unter Rückfluss erhitzt. Die erhaltene klare Lösung wird anschliessend mit 1,0 ml n-Salzsäure neutralisiert und in Vakuum bis zur Trockne eingeengt. Der Rückstand wird mit Wasser aufgenommen und mit Chloroform extrahiert. Die Chloroform Lösung wird nach Trocknen über Natriumsulfat eingedampft.
Der farblose, sirupöse Rückstand wird über Kieselgel mit einem Gemisch aus Chloroform und Äthanol (Raumverhältnis 9:1) chromatographiert. Man erhält als reine Hauptfraktion in etwa 75 %iger Ausbeute das 1-(2-Methyl-3'-oxobutyl)-3,7- dimethylxanthin in Form eines farblosen Sirups. Die Verbindung bildet ein 2,4-Dinitrophenylhydrazon vom Schmelzpunkt 203-2050C.
Beispiel 5
A) Das Beispiel 1 A wird mit der Abweichung wiederholt, dass die zweite Arbeitsstufe wie folgt ausgeführt wird: Ein Gemisch von 66 g 2-Methyl-3-carbäthoxy-5,6-dihydropyran und 150 ml 48%iger Bromwasserstoffsäure wird bei Zimmertemperatur stehen gelassen. Dabei findet eine starke Kohlensäureentwicklung statt. Anschliessend wird das Reaktionsgemisch 1 Stunde unter Rückfluss gekocht. Nach dem Erkalten wird mit Chloroform extrahiert, die Chloroformlösung mit gesättigter Natriumbicarbonatlösung gewaschen und mit Natriumsulfat getrocknet. Nach dem Abdestillieren des Chloroforms unter Normaldruck wird das Keton im Vakuum fraktioniert; man erhält 49 g 1-Bromhexanon-5, entsprechend 71,5% der Theorie, vom Kp12 94-98OC.
B) Die Lösung von 35,4 g 1-Bromhexanon-5 in 200 ml Äthanol wird in der Siedehitze unter kräftigem Rühren all mählich mit 39,7 g Theobromin-Natrium in 100 ml Wasser versetzt. Nach dreistündigem Erhitzen unter Rückfluss wird nicht umgesetztes Theobromin abgesaugt, das Filtrat zur Trockne eingedampft, der Rückstand in Wasser aufgenommen und mit Chloroform extrahiert. Aus der Chloroformlösung wird als Rückstand das 1 -(5'-Oxohexyl)-3 ,7-dimethylxanthin erhalten, das nach dem Umkristallisieren aus Isopropanol in etwa 25 %Der Ausbeute (bezogen auf umgesetztes Theobromin) erhalten wird und bei 102-103oC schmilzt.
C) Das vorliegende Beispiel wird mit der Abweichung wiederholt, dass die zweite Arbeitsstufe wie folgt ausgeführt wird: Eine Mischung von 34 g 2-Methyl-3-carbäthoxy-5,6-dihydropyran und 50 ml 38teige Salzsäure wird eine Stunde bei Zimmertemperatur gehalten. Während dieser Zeit findet eine lebhafte Kohlensäureentwicklung statt. Anschliessend sättigt man das Reaktionsgemisch bei OoC mit gasförmigem Chlorwasserstoff und hält 3 Stunden bei dieser Temperatur; danach lässt man das Reaktionsgemisch sich langsam auf Zimmertemperatur erwärmen. Nach 12-stündigem Stehen wird mit Chloroform extrahiert, die Chloroformschicht mit gesättigter Natriumbicarbonatlösung entsäuert und mit Natriumsulfat getrocknet. Nach Abdestillieren des Chloroforms bei Normaldruck wird der Rückstand im Vakuum fraktioniert.
Man erhält 15 g 1-Chlorhexanon-5 vom Kpl2 83850C, entsprechend 55% der Theorie, das sich analog dem 1-Bromhexanon-5 umsetzen lässt.
Beispiel 6
10,7 g Methyl-Bchloräthylketon, gelöst in 150 ml Äthanol, und 20,2 g Theobromin-Natrium, gelöst in 100 ml Wasser, werden nach dem Verfahren des Beispiels 8 umgesetzt. Nach Umkristallisation aus Isopropanol wird 1-(3'-Oxobutyl)-3,7- dimethylxanthin vom Schmelzpunkt 144146OC in etwa 70Ssiger Ausbeute erhalten.
Beispiel 7
25,0 g Theobromin, 27,0 g 2-Äthylbutenon, 14,0 ml n Natronlauge und 150 ml eines Gemisches von Äthanol und Wasser (Raumverhältnis 1:1) werden 12 Stunden unter Rückfluss erhitzt. Die erhaltene Lösung wird anschliessend neutralisiert und das nicht umgesetzte Theobromin abfiltriert. Nach Entfernen des Alkohols im Vakuum wird die zurückbleibende wässrige Lösung alkalisch gemacht, einmal mit wenig Petrol äther und dann mit Chloroform extrahiert. Der erhaltene Rückstand der Chloroformlösung, der als farbloser Sirup in einer Ausbeute von etwa 70% (bezogen auf umgesetztes Theobromin) anfällt, entspricht dem 1-(2/-Äthyl-3'-oxobutyl)- 3,7-dimethylxanthin, das ein 2,4-Dinitrophenylhydrazon vom Schmelzpunkt 200-2050C bildet.
Beispiel 8
15,6 g Theobromin, 100 ml Wasser und 10,0 g ss-Dimethyl- aminoäthyl-methyl-keton werden 3 Stunden unter Rückfluss erhitzt. Anschliessend wird das Reaktionsgemisch mit 10% iger Salzsäure schwach angesäuert und vom nicht umgesetzten Theobromin abfiltriert. Das Filtrat wird dann alkalisch gemacht und mit Chloroform extrahiert. Nach Umkristallisation des Rückstandes der Chloroformlösung aus Isopropanol wird 1-(3'-Oxobutyl)-3,7-dimethylxanthin vom Schmelzpunkt 143-1440C in etwa 40 %iger Ausbeute (bezogen auf umgesetztes Theobromin) erhalten.
Die Eigenschaften einiger erfindungsgemäss hergestellter Verbindungen, und zwar des 7-(6-Oxoheptyl)-1,3-dimethylxanthins (I), des 7-(5 -Oxohexyl)-1,3-dimethylxanthins (II) (Beispiel 1), des 7-(4 -Oxopentyl)-1,3-dimethylxanthins (III) (Beispiel 1 B), des 7-(2-Methyl-3-oxobutyl)-1,3-dimethylxanthins (1V) (Beispiel 2 B), des 7-(2 -Äthyl-3 -oxobutyl)- 1 ,3-dimethylxanthins (V) (Beispiel 2A) werden in der nachstehenden Tabelle mit denen ihres Grundkörpers 1,3-Dimethylxanthin (Theophyllin) (VI), des im Handel befindlichen 7-(ss,-Dihydroxypropyl)-1,3-dimethylxanthins (VII), des bekannten 7-Acetonyl-1,3-dimethylxanthins (VIII) und 7-(3!-Oxobutyl)-1,3-dimethylxanthins (IX) verglichen,
ferner die des 1-(5/-Oxohexyl)-3,7-dimethylxanthins (X) (Beispiel 5), des 1-(4'-Oxopentyl)-3,7-dimethylxanthins (XI), des 1-(3'-Oxobutyl)-3,7-dimethylxanthins (XII) (Beispiel 3, 6 und 8), des 1-(2'-Methyl-3'-oxobutyl)-3 ,7-dimethylxanthins (XIII) (Beispiel 4) und des 1-(2'-Äthyl-3'-oxobutyl)-3 ,7-dimethylxanthins (XIV) (Beispiel 7) mit denen ihres Grundkörpers 3,7-Dimethylxanthin (Theobromin) (XV), des im Handel befindlichen 1-(2'-Hydroxypropyl)3,7-dimethylxanthins (XVI) und des bekannten, aber pharmakologisch bis jetzt nicht untersuchten 1-Acetonyl-3,7-dimethylxanthins (XVII) verglichen.

Die Gefässwirkung wurde nach Krawkow-Pissemski am isolierten Kaninchenohr mit einer Lösung von 0,1 mg Substanz/ml Ringer-Lösung bestimmt. Zur Gefässtonisierung wurde der Nährlösung 1-(m-Hydroxyphenyl)-2-amino-äthanol-hydrochlorid (0,5#/ml) zugesetzt.
Aus der Tabelle ist ersichtlich, dass die erfindungsgemäss erhältlichen Verbindungen im Gegensatz zu den bisher bekannten Derivaten des 1,3- bzw. 3,7-Dimethylxanthins bei guter Lipoidlöslichkeit eine überraschenderweise sehr gute Wasserlöslichkeit zeigen.
Ein weiterer pharmazeutischer Verwendungszweck der gut, z.B. zu mindestens 4% in Wasser löslichen erfindungsgemässen Oxoalkyldimethylxanthin-Derivate besteht überraschenderweise darin, dass diese die Wasserlöslichkeit von anderen therapeutisch nützlichen Substanzen erheblich erhöhen.
Bekanntlich können zahlreiche therapeutisch wirksame Substanzen am Menschen und am Tier nicht parenteral, also durch Injektionen, zugeführt werden, obwohl eine solche parenterale Anwendung gegenüber einer peroralen oder rektalen therapeutische Vorzüge hätte, weil damit wässrige Lösungen einer Konzentration, wie sie für Injektionen benötigt werden, wegen der geringen Löslichkeit solcher Substanzen in Wasser nicht hergestellt werden können. Es ist schon versucht worden, solche Substanzen durch geeignete Lösungsvermittler in Lösung zu bringen. So hat man modifizierte Polyoxyäthylene, sogenannte Tweene, verwendet. Diese sind aber pharmakologisch nicht indifferent und verursachen somit unerwünschte Nebenwirkungen.
Es ist beispielsweise bekannt, das therapeutisch viel verwendete Theophyllin, das in Form der Base, aber auch in Form bestimmter mässig wasserlöslicher Additionsverbindungen wegen zu geringer Wasserlöslichkeit für Injektionszwecke nicht geeignet ist, unter Verwendung von Äthylendiamin, das hier als Lösungsmittel fungiert, in wässrige Lösungen zu bringen, die für Injektionszwecke geeignet sind und vielfach angewandt werden.
Es ist daher ein erheblicher Fortschritt, dass die erfindungsgemässen Verbindungen beträchtliche löslichkeitsvermittelnde Eigenschaften für zahlreiche therapeutisch nützliche Substanzen aufweisen. So gelingt es z.B. von Khellin, von dem bei Zimmertemperatur nur 20 mg in 100 ml Wasser löslich sind, in 10 %aber wässriger Lösung von 1 -Oxohexyl-3 ,7-dimethylxan- thin 1,4%ige wässrige Lösungen herzustellen. Auch die Löslichkeit von Coffein lässt sich durch Zusatz von 1-Oxohexyl3,7-dimethylxanthin um mehr als das Zehnfache steigern.
Diese löslichkeitsvermittelnde Wirkung beschränkt sich nicht nur auf die genannten Substanzen, sondern gilt überraschenderweise auch für Alkaloide wie Yohimbin, Corynanthin und ihre Derivate, und andere Substanzen, wie Nikotinsäureester.
Entsprechende Angaben sind aus der nachfolgenden Tabelle ersichtlich.

Claims (1)
- PATENTANSPRUCH Verfahren zur Herstellung von l-[(w-l)-Oxoalkyl]- dimethylxanthinen der Formel I EMI6.1 worin X, falls die EMI6.2 Gruppe in 7-Stellung sitzt, eine Alkylengruppe mit 3 bis 6 C Atomen, oder, falls die EMI6.3 Gruppe in 1-Stellung sitzt, eine Alkylengruppe mit 2 bis 5 C Atomen bedeutet, wobei die Ketogruppe mindestens 2 C Atome vom Xanthingerüst entfernt ist, dadurch gekennzeichnet, dass bei erhöhter Temperatur ein Dimethyixanthin der Formel II EMI6.4 oder dessen Alkalimetallsalz mit einer Verbindung der Formel III EMI6.5 umgesetzt wird, wobei Y eine a,ss-ungesättigte Gruppe H2C= CR-,eine a,ss-Dialkylaminoäthylgruppe mit 1 oder 2 C Atomen in der Alkylgruppe und X = Null ist oder wobei Y Brom oder Chlor bedeutet und X die für Formel I angegebene Bedeutung hat, und R in der ungesättigten Gruppe bei Umsetzung mit dem 1,3-Dimethylxanthin ein n-Alkylrest mit 1 bis 4 C-Atomen und bei Umsetzung mit dem 3,7-Dimethylxanthin Wasserstoff oder ein n-Alkylrest mit 1 bis 3 C-Atomen ist.UNTERANSPRÜCHE 1. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man die Umsetzung mit einem a,ss-ungesättigten Methylketon in alkalischem Medium durchführt.2. Verfahren nach Patentanspruch oder Unteranspruch 1, dadurch gekennzeichnet, dass man die Umsetzung in wässriger organischer Lösung durchführt.3. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man die Umsetzung mit dem Oxoalkylbromid oder -chlorid mit dem Alkalisalz des Dimethylxanthins durchführt.4. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man die Umsetzung bei einer Temperatur zwischen 50oC und 1500C durchführt.5. Verfahren nach Patentanspruch oder einem der Unteransprüche 1 bis 4, dadurch gekennzeichnet, dass der Rest X unverzweigt ist.6. Verfahren nach Patentanspruch oder einem der Unteransprüche 1 bis 4, dadurch gekennzeichnet, dass der Rest X eine Äthylengruppe darstellt, der an dem der Carbonylgruppe benachbarten C-Atom durch eine Alkylseitengruppe substituiert ist.
