10-Methoxy-vincamin bzw. 10-Hydroxy-vincamin in seinen.

The subsequent acid cleavage is preferably carried out in a hydrogen halide solution, e.g. B. hydrochloric acid in methanol, aqueous hydrogen iodide or hydrogen bromide in glacial acetic acid at temperatures between 0 and 80O C, preferably at temperatures between 10 and 60 C performed. The resulting 10-methoxy-vincamine, 10-methoxy-apovincamine, 10-hydroxy-vincamine and 10-hydroxyapovincamine are then isolated from the reaction mixture in a manner known per se and purified.
The 10-methoxy-apovincamine or 10-hydroxy-apovincamine obtained is then allowed to react with absolutely dry and halogen-free hydrogen halide at temperatures below 0 ° C., preferably at temperatures between -20 and -150 ° C. Hydrogen bromide is preferably used as the hydrogen halide However, hydrogen chloride or hydrogen iodide can also be used. The solution is then evaporated to dryness, the residue is then at temperatures below 0 C, preferably at temperatures between -20 and -150 C, in a suitable one that does not freeze at temperatures below 0 C. Solvents such as
Suspended acetone or tetrahydrofuran, then with a water-containing mixture that does not freeze at temperatures below 0 C, consisting of a solvent such as. B. acetone or tetrahydrofuran, an organic or inorganic base such as. B. pyridine, sodium hydrogen carbonate or alkali hydroxide, and water, preferably added in a ratio of 70:20:10 and warmed to room temperature.
The resulting 10-methoxyvincamine or 10-hydroxyvincamine can then be isolated from the reaction mixture and purified in a manner known per se.
The process according to the invention can be used both for the preparation of the optically active forms and for the preparation of the racemate of 10-methoxyvincamine or 10-hydroxyvincamine. The application of the method according to the invention to optically active compounds makes it possible, in particular, to obtain (3S, 14S, 16S) -10-methoxy-vincamine or (3S, 14S, 16S) -10-hydroxy-vincamine, which has the same stereochemistry as that natural vincamine, the z. B. from Vinca minor L. is isolated. The process also enables (3R, 14R, 16R) -10-methoxy-vincamine or
(3R, 14R, 16R) -10-hydroxy-vincamine, the optical antipodes of the above-mentioned compounds.
This is preferably based on an optically active compound of the formula II. The compound of the formula II is new; its preparation is described in the example below. To prepare an optically active compound of the formula II, a racemate is separated into its optical isomers in one of the precursors which lead to this compound; z. B. by converting it into the mixture of the diastereoisomeric salts by means of optically active bases or acids and isolating the optical antipodes therefrom. But it is also possible to start from an optically active compound of the formula IV, V or VI which can be obtained by resolving the corresponding racemate.
10-methoxy-vincamine or 10-hydroxy-vincamine in his.
Optically active forms as well as in the form of its racemate are characterized by interesting pharmacological properties and can be used as medicinal products.
In the observation test on the mouse, an increase in excitability could be seen at doses of 10 to 100 mg / kg p.o. to be observed. The sleep phases in the electroencephalogram of the rat showed a decrease in sleep and an increase in wakefulness at doses of 10 mg / kg i.p. Such changes are an expression of an increase in vigilance and psychostimulation. The compounds are therefore indicated for cerebral vascular damage, cerebral sclerosis, cerebral insufficiency or loss of consciousness due to head trauma.
The doses to be used naturally vary depending on the substance used, the type of administration and the condition to be treated. In general, however, satisfactory results are achieved in animal experiments with a dose of 10 to 100 mg / kg body weight. For larger mammals, the daily dose is around 1 to 500 mg. If necessary, these doses can be administered in 2 to 3 parts of 0.5-30 mg 10-methoxy-vincamine or 10-hydroxy-vincamine in addition to solid or liquid carriers or diluents or as a sustained-release form.
In the following examples, which explain the invention in more detail but are not intended to restrict its scope in any way, all temperatures are given in degrees Celsius and are uncorrected.
The signs of the rotation values for all optically active compounds relate to measurements with chloroform as solvent.
