REVENDICATIONS
1. Procédé de préparation d'un mélange des alcaloïdes difeuine 1. difeuine Il et difeuine III présente dans la racine d'une pentadiplandracée. plante du genre pentadipandra brazzeana appelée Difeu, caractérisé en ce que l'on évapore un extrait alcoolique concentré de la plante et que l'on soumet le résidu à l'action d'un acide, de manière à précipiter des sels d'acide des trois alcaloïdes, en ce qu'après séparation du préci pitié. on ajoute aux phases liquides un réactif constitué par l'iodomercurate de potassium en milieu neutre, en ce qu'après lavage du précipité obtenu.
on met celui-ci en suspension dans l'eau et on le soumet à l'action de l'acide sulfhydrique, en ce que l'on alcalinise la phase liquide obtenue après centrifugation et en ce que l'on extrait les trois alcaloïdes par un éther, les phases éthérées étant ultérieurement deshydratées et l'éther évaporé.
7. Procédé selon la revendication 1. caractérisé en ce que l'on effectue la préparation de l'extrait alcoolique en mettant en contact des racines de Difeu avec un alcool dans un récipient fermé hermétiquement et à l'abri de la lumière. et en ce que l'on filtre l'extrait obtenu et on concentre par distillation sous vide.
3. Procédé selon la revendication 1. caractérisé en ce que l'alcool est l'éthanol.
4. Procédé selon la revendication 1. caractérisé en ce que l'agent d'alcalinisation est le carbonate de sodium.
5. Procédé selon la revendication 1, caractérisé en ce que l'éther servant à l'extraction est l'éther éthylique.
La présente invention se rapporte à un procédé de préparation d'un mélange des alcaloïdes difeuine I. Il et III.
Ces alcaloïdes se présentent ensemble dans les racines d'une plante de la famille des pentadiplandracées, genre pentadiplandra brazeana, connue sous le nom de difeu dans le département du Nde (Cameroun).
Cette plante possède diverses propriétés thérapeutiques.
notamment pour le traitement de i'anorexie. des hémorroïdes, des états diarrhétiques et des oedèmes de carence.
Les éléments actifs de la plante sont trois alcaloïdes nommés difeuine 1. difeuine Il et difeuine III. Les propriétés et l'obtention de la difeuine III sont décrites dans le brevet suisse no. 566 139.
L'invention vise à préparer de façon particulièrement avantageuse un mélange des alcaloïdes difeuine 1.11 et III.
A cet effet. le procédé selon l'invention est caractérisé en ce que l'on évapore un extrait alcoolique concentré de la plante et que l'on soumet le résidu à l'action d'un acide, de manière à précipiter des sels d'acide des trois alcaloïdes. en ce qu'après séparation du précipité. on ajoute aux phases liquides du réactif de Mayer (iodomercurate de potassium en milieu neutre),en ce qu'après lavage du précipité obtenu. on met celui-ci en suspension dans l'eau et on le soumet à l'action de l'acide sulfhydrique. en ce que l'on alcalinise la phase liquide obtenue après centrifugation et en ce que l'on extrait les trois alcaloïdes par un éther, les phases éthérées etant ultérieurement déshydratées et l'éther évaporé.
Comme alcool servant de solvant pour l'extrait de départ.
on utilise généralement l'éthanol à 95" GL mais d'autres alcools tels que le méthanol et le butanol peuvent être aussi employés.
Cet extrait est préparé en plaçant des racines de difeu en contact avec un alcool (le même que celui du paragraphe précédent) dans un récipient fermé hermétiquement et à l'abri de la lumière. et l'extrait obtenu est filtré et concentré par distillation sous vide. Pour la distillation. on utilise de préférence un évaporateur sous vide et on arrête la distillation dès que l'extrait, à l'origine brun-jaunâtre. devient de plus en plus brun et laisse des traînées jaunâtres sur la paroi du ballon de l'évaporateur rotatif.
L'acide servant à provoquer une précipitation des sels des alcaloïdes peut être tout acide conduisant à une telle précipitation. par exemple l'acide chlorvdrique (donnant des chlorhydrates). L'acide bromhydrique. L'acide sulfurique. L'acide oxalique...
L'éther servant à extraire les trois alcaloïdes est. de préférence, L'éther éthylique mais d'autres éthers réalisant une telle extraction peuvent être employés. De même, tout agent alcalin (par exemple n'importe quelle hase ou n'importe quel sel à propriété alcaline) peut être employé pour l'alcalînisation de la phase liquide obtenue après l'action de H2S et la séparation du precipité.
On présentera dans ce qui va suivre un exemple de réalisation de la présente invention. exemple qui n'est donné qu'à titre d'illustration et non de limitation.
Des racines de Difeu, séchées et découpées en petits morceaux, sont mises dans un vase contenant de l'éthanol à 95t) GL.
en quantité suffisante. Le vase est fermé hermétiquement et mis à l'obscurité pendant 48 heures. On obtient alors un extrait brun-jaunâtre qui est filtré et concentré en chassant l'alcool par distillation sous vide à environ 15 mn Hg, dans un évaporateur à bain-marie thermostat régié à 40t8C.
On prend 160 ml d'extrait alcoolique concentré de la plante et on les évapore au bain-marie. Le résidu est repris par 80 ml d'acide chlorhydrique N110. ce qui donne les chlorhydrates des trois alcaloïdes. On centrifuge et on lave 2 fois le précipité à l'eau. Dans les phases liquides recueillies, on ajoute 200 ml du réactif de Mayer (iodomercurate de potassium en milieu neu tre). Le précipité blanc-jaunâtre obtenu est isolé par centrifugation, lavé deux fois à l'eau et puis. après élimination des eaux de lavage, on le met en suspension dans 300 ml d'eau et on fait passer H2S pendant 30 minutes.
La solution est laissée au repos pendant 12 heures; on centrifuge, on recueille la phase liquide qu'on alcalinise avec 7O ml de Na C03.
Les 3 alcaloïdes ainsi libérés sont extraits par 300 ml d'éther éthvlique.
On déshydrate les phases éthérées par Na2SO4 anhydre et on évaporte l'éther sous vide. Le résidu est repris par de l'alcool à 9: GL.

An exemplary embodiment of the present invention will be presented in what follows. example which is given only by way of illustration and not of limitation.
Difeu roots, dried and cut into small pieces, are placed in a vase containing ethanol at 95t) GL.
in sufficient quantity. The vase is sealed and put in the dark for 48 hours. A yellowish-brown extract is then obtained which is filtered and concentrated by removing the alcohol by distillation under vacuum at about 15 min Hg, in a thermostatically controlled water bath evaporator at 40 ° C.
160 ml of concentrated alcoholic extract of the plant are taken and evaporated in a water bath. The residue is taken up in 80 ml of N110 hydrochloric acid. which gives the hydrochlorides of the three alkaloids. The precipitate is centrifuged and washed twice with water. 200 ml of Mayer's reagent (potassium iodomercurate in neutral medium) are added to the liquid phases collected. The 3 alkaloids thus released are extracted with 300 ml of ethyl ether.
