Description
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Cephalosporin- und Penicillinderivate, die als Antibiotika brauchbar sind.
Die erfindungsgemäss herstellbaren Verbindungen weisen die folgende Formel auf

Für geradkettige oder verzweigte Alkylreste mit 1 bis 4 Kohlenstoffatomen, aus denen Y, R1, R2, R3, R7, R8, R10, R13 und R14 bestehen können, sind der Methyl-, Äthyl-, n-Propyl-, Isopropyl-, n-Butyl- und tert-Butylrest zu nennen.
Beispiele für Alkylreste mit 1 bis 4 Kohlenstoffatomen, aus denen R9, R11 und R12 bestehen können, sind der Methyl-, Äthyl-, n-Propyl- und n-Butylrest.
Beispiele für Alkoxyreste, aus denen Y bestehen kann, sind der Methoxy-, Äthoxy-, n-Propoxy- und n-Butoxyrest.
Beispiele für geradkettige oder verzweigte Alkylreste, die den Substituenten R8 bilden können, sind der Methoxy-, Äth-oxy-, n-Propoxy-, Isopropoxy-, sek-Butoxy- und n-Butoxyrest.
Gemäss Formel I ist der Substituent X ausser Wasserstoff oder der Acetoxygruppe einer der folgenden heterozyklischen Mercaptoreste, nämlich ein l,3,4-Thiadiazol-5-ylthio-, 3-Methyl-l,2,4-thiadiazol-5-ylthio-, Tetrazol-5-ylthio-, 1-Methyltetrazol-5-ylthio-, 2-Methyl-l,3,4-oxydiazolrest-5-yl-thio- oder 2-Methyl-l,3,4-thiadiazol-5-ylthiorest folgender Formeln:

Bevorzugte Bindungsstellen für den Isothioharnstoffmethyl-rest in obiger Formel II sind die ortho- und para-Stellung des Phenylrings. In obiger Formel II besitzen die Substituenten Y, R1, R2, R3, Z, n, W, R5 und R6 die für Formel I geltende Bedeutung.
Ist der Arylrest in den Verbindungen der Formel I ein 2-Thienylrest, so bedeutet Y Wasserstoff und Z eine Bindung. Verbindungen dieser Art werden durch folgende Formel III wiedergegeben:

In den Verbindungen der Formel III kann der Isothioharn-stoffsubstituent in 4- oder 5-Stellung des Thienylrests gebunden sein. Die Substituenten R1, R2, R3, n, W, R5 und R6 besitzen die in Verbindung mit Formel I angegebene Bedeutung.

In den Verbindungen der Formeln IV und V kann der Substituent R5 eis- oder trans-ständig zum Wasserstoffatom in 6-Stellung der Cephalosporinderivate der Formel IV oder dem Wasserstoffatom in 5-Stellung der Penicillinderivate der Formel V vorliegen. Die Verbindungen der Formeln IV und V mit cis-ständigem Substituent R5 werden bevorzugt.
Weitere bevorzugte Ausführungsformen sind: (A) Verbindungen, bei welchen W Wasserstoff, die Hydroxyl-, Amino-oder S03H-Gruppe oder den Rest COOH darstellt. Bei derartiger Substitution erhält man üblicherweise Verbindungen mit breiterem Wirkungsspektrum und/oder verbesserter oraler Wirkung. So sind z. B.
(1) Verbindungen, worin W die Hydroxylgruppe ist, resistenter gegen ß-Lactamase-Organismen.
(2) Verbindungen, worin W SO3H oder COOH bedeutet, besitzen ein breiteres Wirkungsspektrum gegen gram-negative Bakterien.
(3) Verbindungen, worin W die Aminogruppe bedeutet, besitzen verbesserte orale Wirkung.
(B) Verbindungen, bei welchen R5 die Methoxygruppe ist, sind von spezieller Bedeutung, da sie eine antibakterielle Wirkung gegen Cephalosporinase produzierende gram-negative Organismen ausüben.
(C) Verbindungen, worin X den Acetoxy-, 2-Methyl-1,3,4-thiadiazol-5-ylthio- oder l-Methyltetrazol-5-ylthiorest bedeutet.
Unter den bevorzugten Ausführungsformen gemäss den Verbindungen (A), (B) und (C) sind solche besonders bevorzugt, bei denen Z eine Bindung darstellt.

Von den Verbindungen der Formel VI werden solche bevorzugt, bei denen die Wasserstoffatome in 6- und 7-Stel-lung cis-ständig zueinander sind.
In den Rahmen der Erfindung fallen üblicherweise auch die einzelnen optischen Isomeren solcher Verbindungen, bei denen W von Wasserstoff verschieden ist.
Ferner fallen in den Rahmen der Erfindung die nicht-toxischen Säureadditionssalze der Verbindungen, z.B. die Additionssalze mit Mineralsäuren wie das Hydrochlorid, Hydrobro-

55 mid, Hydrojodid, Sulfat, Sulfamat und Phosphat, oder mit organischen Säuren wie das Maleat, Acetat, Citrat, Oxalat, Succinat, Benzoat, Tartrat, Fumarat, Malat, Mandelat und Ascorbat.
In den Rahmen der Erfindung fallen im allgemeinen auch 60 die nicht-toxischen pharmazeutisch zulässigen Salze solcher Verbindungen, bei denen W die Carboxyl- oder Sulfonsäuregruppe bedeutet und M Wasserstoff darstellt. Beispiele für pharmazeutisch zulässige Salze dieser Säurederivate sind solche mit primären, sekundären oder tertiären Aminen, z.B. 65 Cyclohexylamin, Äthylamin und Pyridin.
Die pharmazeutisch zulässigen Kationen, die als Rest M in den Verbindungen der Formeln I bis IV vorliegen können, umfassen insbesondere die Alkalimetallionen wie das Natrium-,

Kalium- und Calciumion und das Ammoniumion, ferner organische Aminkationen wie z.B. niedrig-Alkylammoniumgrup-pen wie Triäthylammoniunrand N-Äthylpiperidin.
Die Salze der Verbindungen der Formeln I bis IV, bei denen M ein pharmazeutisch zulässiges Kation bedeutet, können in bekannter Weise hergestellt werden, und zwar in situ oder durch Umsetzung der entsprechenden Säure mit einer Base wie z.B. Natriumbicarbonat oder Triäthylamin.
Die erfindungsgemäss herstellbaren Verbindungen können analog wie viele bekannte Cephalosporinverbindungen, z.B. Cephalexin, Cephalothin oder Cephaloglycin, verabreicht werden. Man kann sie allein oder in Form pharmazeutischer Präparate oral, parenteral und topisch an Warmblütler, d. h. Vögel und Säugetiere, z.B. Katzen, Hunde, Rindvieh und Pferde sowie an Menschen verabreichen. Zur oralen Verabreichung können die Verbindungen in Form von Tabletten, Kapseln oder Pillen oder in Form von Elixieren oder Suspensionen gegeben werden. Zur parenteralen Verabreichung verwendet man am besten eine sterile wässrige Lösung, die andere gelöste

Stoffe enthalten kann, z.B. Kochsalz oder Glucose, womit die Lösung isotonisch gestellt wird. Zur topischen Verabreichung werden die Verbindungen in Cremes und Salben eingearbeitet.
Beispiele für Bakterien, gegen die die genannten Verbindungen wirksam sind, sind Staphylococcus aureus, Salmonella schottmuelleri, Klebsiella pneumoniae, Diplococcus pneumo-nia und Streptococcus pyogenes.
Ein Beispiel eines erfindungsgemäss herstellbaren Cephalo-sporinderivats ist die 3-[(Acetyloxy)methyl]-7-[[2-[4-(isothio-harnstoffmethyl)-phenyl]acetyl]amino]-8-oxo-5-thia-l-azabi-cyclo[4.2.0]oct-2-en-2-carbonsäure, und ein Beispiel eines weiteren neuen Penicillinderivats liegt in der 6-[[2-[4-(Isothio-harnstoffmethyl)phenyl]acetyl]amino]-3,3-dimethyl-7-oxo-4-thiazin-l-azabicyclo[3.2.0]heptan-2-carbonsäure vor. Weitere Beispiele dieser Verbindungen werden nachstehend gegeben.

Erfindungsgemäss erhalterreVerbindungen der Formel I, in welchen W den Rest COOH darstellt, können dazu verwendet werden, um Verbindungen der Formel I herzustellen, worin W COOR4 ist, in welcher Gruppe R4 Indanyl bedeutet. Dazu kann man die genannten Verbindungen mit 5-Indanol in einem inerten Lösungsmittel in Gegenwart von N,N'-Dicyclo-hexylcarbodiimid beim pH-Wert etwa 2,5 bei einer Temperatur von 20 bis 30°C umsetzen.

Die erfindungsgemässe Reaktion wird in einem alkoholischen Lösungsmittel bei Temperaturen von 0 bis 100°C, vorzugsweise von 25 bis 50°C, während V2 bis 6 Stunden durchgeführt, wobei man das entsprechende Isothiuronium-hydro-chlorid bzw. -hydrobromid gewöhnlich in Lösung erhält. Das organische Lösungsmittel wird üblicherweise abgedunstet und das zurückbleibende Öl wird mit entsprechenden organischen Lösungsmitteln wie Äther, Hexan, Äthylacetat oder Acetoni-tril verrieben, wobei man die durch den Isothioharnstoffme-thylrest substituierte Verbindung erhält.
Ist der Substituent W in Formel VII eine Aminogruppe, so können zum Schutz der Aminofunktion geeignete blockierende Gruppen, z.B. ein Säuresalz wie das Hydrochlorid, eine Acylgruppe oder der tert.-Butoxycarbonylrest verwendet werden. Die Schutzgruppen können nach der Kupplungsreaktionen in bekannter Weise entfernt werden, siehe z.B. die Vorschrift der US-PS 3 657 232.

Ist der Substituent W in Formel X eine Aminogruppe, so verwendet man vorzugsweise geeignete blockierende Gruppen, z.B. ein Säuresalz wie das Hydrochlorid, den tert.-Butoxycar-bonyl- oder Carbobenzyloxyrest, um die Aminofunlction analog wie vorstehend beschrieben zu schützen.
Funktionelle Äquivalente der Säuren der Formel X sind in der Regel die Säurehalogenide, z.B. das Säurechlorid, die Säureanhydride einschliesslich gemischte Anhydride mit beispielsweise Alkylphosphorsäuren, niederen aliphatischen Monoestern der Kohlensäure oder Alkyl- oder Arylsulfonsäu-ren. Ausserdem kann das Säureazid oder ein aktiver Ester jder Thioester, z. B. mit p-Nitrophenol, 2,4-Dinitrophenol oder Thioessigsäure verwendet werden, oder man kann die freie Säure der Formel X mit dem 7-Aminocephalosporansäu-rederivat oder einem 6-Aminopenicillansäurederivat der Formel IX kuppeln, nachdem man die Säure zunächst mit N,N'-Dimethylchloroformiumchlorid umgesetzt hat, oder unter Verwendung eines Carbodiimids wie N,N'-Diisopropylcarbodi-imid, N,N'-Dicyclohexylcarbodiimid oder N-Cyclohexyl-N'-(2-morpholinoäthyl)carbodiimid.
Die Kupplungsreaktion wird im allgemeinen in Gegenwart eines Lösungsmittels durchgeführt. Geeignete Lösungsmittel sind z.B. Äthylacetat, Aceton, Dioxan, Acetonitril, Chloroform, Methylenchlorid, Tetrahydrofuran und Dimethylform-amid. Werden hydrophile Lösungsmittel eingesetzt, so können diese auch im Gemisch mit Wasser verwendet werden. Die Kupplungsreaktion kann in Gegenwart einer Base, z.B. eines Alkalibicarbonats, durchgeführt werden. Die Reaktionstemperatur kann zwischen -10 und 100°C liegen, die Reaktionszeit kann etwa l/2 bis 10 Stunden betragen. Die Cephalosporin-Produkte werden üblicherweise nach konventionellen Methoden isoliert.
Die Verbindungen der Formel VII können auch hergestellt werden, indem man ein modifiziertes Polystyrol, welches Nitrophenol- oder Hydroxysuccinimidgruppen enthält, mit einer Säure der Formel X vereinigt und die so gebildete aktivierte Säure mit einer Verbindung der Formel IX nach dem Verfahren der CA-PS 892 580 umsetzt, wobei man das dort beschriebene Penicillansäurederivat durch eine Verbindung der Formel IX ersetzt.

Verbindungen der Formeln XI und XII, worin M einen Alkanoyloxymethylrest darstellt, können hergestellt werden, indem man die entsprechende Säure in Form eines Salzes, z.B. eines Alkalimetallsalzes oder des Triäthylammoniumsalzes, mit einer Verbindung der Formel

Verbindungen der Formeln XI und XII, worin M einen 45 Alkanoylaminomethyl- oder Alkoxycarbonylaminomethylrest bedeutet, können erhalten werden, indem man das Natriumsalz von Säurederivaten der Formeln XI oder XII in einem organischen Lösungsmittel wie Dimethylformamid oder Hexa-methylphosphoramid bei Raumtemperatur mit einer äquiva-so lenten Menge eines Alkanoylaminomethylhalogenids oder eines Alkoxycarbonylaminomethylhalogenids '/2 bis 3 Stunden behandelt, worauf das Gemisch in Eis/Wasser gegossen wird. Das dabei ausfallende Produkt wird nach Standardverfahren isoliert.
ss Verbindungen der Formeln XI und XII, worin M einen p-Alkanoyloxy-benzylrest bedeutet, kann man erhalten, indem man 2 Äquivalente des p-Alkanoyloxy-benzylalkohols zu einer Suspension des Natriumsalzes des Säurederivats der Formeln XI oder XII in Dimethylformamid oder Hexamethylphospho-60 ramid zusetzt, worauf das Gemisch auf 0°C gekühlt wird.
Vorzugsweise werden unter Rühren 1,2 Äquivalente Dicyclohe-xylcarbodiimid in Dimethylformamid zugetropft. Das Gemisch wird gewöhnlich bei 0°C 1h bis 3 Stunden und dann noch 2 bis 5 Stunden bei Raumtemperatur gerührt. Der gebildete Dicy-65 clohexylharnstoff kann abfiltriert und das Filtrat wird mit Chloroform, Methylenchlorid oder Äthylacetat verdünnt, mit Wasser gewaschen und getrocknet werden, wobei man das Produkt erhält.

Verbindungen der Formeln XI und XII, worin M einen Aminoalkanoyloxymethylrest bedeutet, werden üblicherweise hergestellt, indem man eine Suspension des Natriumsalzes einer Säure der Formel XI oder XII und einen Uberschuss eines entsprechenden Aminoalkanoyloxymethylhalogenids mit geschützter Aminogruppe in einem Lösungsmittel wie Dimethylformamid, Hexamethylphosphoramid oder Dimethylsulfo-xid 2 bis 96 Stunden vermischt. Das Gemisch wird gewöhnlich dann mit einem Lösungsmittel wie Methylacetat oder Methylenchlorid verdünnt und mit Wasser, wässriger Base und Wasser gewaschen. Die organische Phase kann abgetrennt und der Niederschlag in konventioneller Weise isoliert werden, dann wird die Aminogruppe freigesetzt, wobei man das Produkt erhält.
Verbindungen der Formel XII, worin X eine heterocyclische Mercaptogruppe bedeutet, wie bei Formel IV beschrieben, kann man herstellen, indem man 1 Äquivalent der Säure in Form eines Salzes, z.B. des Natriumsalzes, worin X den Acet-oxyrest bedeutet, in etwa 500 bis 2000 ml Wasser bei 50 bis 80°C in Stickstoffatmosphäre löst, worauf 1 Äquivalent einer Base wie Triäthylammonium oder Natriumbicarbonat und 1 bis 3 Äquivalente des heterozyklischen Mercaptans zugesetzt werden. Das Reaktionsgemisch wird in der Regel bei 50 bis 90°C etwa 2 bis 6 Stunden gerührt, dann kann das Wasser abgedunstet und der Rückstand in einem organischen Lösungsmittel wie Methanol, Äthanol oder Dimethylformamid aufgenommen und mit einem organischen Lösungsmittel wie Acetonitril, Aceton, Äthylacetat oder Chloroform ausgefällt werden.
Die Verbindungen der Formel X können durch direkte, nachstehend näher beschriebene Halomethylierung einer Säure der Formel

hergestellt werden, wobei in dieser Formel Aryl, Y, Z, n und W2 die in Verbindung mit Formel VII angegebene Bedeutung besitzen. Diese Säuren sind im Handel erhältlich oder nach bekannten Methoden herstellbar.
Ist der Substituent W in den Verbindungen der Formel XIII eine Aminogruppe, so wird diese durch eine geeignete Schutzgruppe blockiert, z.B. durch eine Acylgruppe wie den Acetyl- oder Trifluoracetylrest, ehe die Halomethylierungsre-aktion durchgeführt wird. Nach beendeter Halomethylierung können die Schutzgruppen durch saure Hydrolyse in an sich bekannterWeise abgespalten werden.
Die halomethylierten Derivate der Verbindungen der Formel XIII können nach verschiedenen Methoden erhalten werden. Z.B. erhält man aus einer Verbindung der Formel XIII mit einem Formaldehydspender wie Paraformaldehyd, CICH2OCH3 oder Formalinlösung in Gegenwart einer Lewis-Säure wie ZnCb, AlCh, SnCU oder CISO3H in einem Lösungsmittel wie Petroläther, Chloroform, Tetrachlorkohlenstoff oder Benzol bei einer Temperatur von -10 bis 100°C unter Einleiten von Chlorwasserstoff- oder Bromwasserstoffgas in das Reaktionsgemisch Verbindungen der allgemeinen Formel X.
Die Umsetzung einer Säure der Formel XIII mit 34 bis 38%igem Formalin in konzentrierter Salzsäure bei Temperaturen von -10 bis 100°C unter Hindurchleiten von Chlorwasserstoff- oder Bromwasserstoffgas durch das Reaktionsgemisch führt im allgemeinen ebenfalls zu Verbindungen der allgemeinen Formel X.
Auch durch Umsetzung einer Säure der Formel XIII mit
Trioxan in Essigsäure oder Phosphorsäure bei Temperaturen von —10 bis 100°C unter Hindurchleiten von Chlorwasserstoffoder Bromwasserstoffgas durch das Reaktionsgemisch kann man Verbindungen der Formel X erhalten. Ferner ergibt gewöhnlich die Umsetzung einer Säure der Formel XIII in Gegenwart einer Lewis-Säure (siehe oben) mit Chlormethyläther bei Temperaturen von -10 bis 100°C oder die Umsetzung der Säure in Essigsäure oder konzentrierter Schwefelsäure mit Dichlormethyläther in Gegenwart von Zinkchlorid Verbindungen der Formel X.
Die Verbindungen der Formel X, worin W die Carboxyl-gruppe und Aryl den Phenylrest bedeuten, werden vorzugsweise erhalten, indem man den entsprechenden Diäthylester der Formel XIII mit 40 %igem Formalin in Gegenwart von wasserfreiem Zinkchlorid in Benzol bei etwa 50°C umsetzt, während Chlorwasserstoff- oder Bromwasserstoffgas durch das Reaktionsgemisch geleitet wird, und anschliessend sauer hydrolysiert.
Verbindungen der Formel X, worin W die Sulfonsäuregruppe darstellt, können durch obige Halomethylierungsreak-tionen erhalten werden unter Verwendung einer Säure der Formel XIII, worin W die SCbH-Gruppe bedeutet, oder deren Carboxymethylester, wobei in letzterem Fall die resultierende halomethylierte Verbindung durch saure Hydrolyse in die freie COOH-Verbindung überführt werden kann.
Bei der Halomethylierung von Verbindungen der Formel XIII, bei denen W die Hydroxylgruppe bedeutet, kann sich der Schutz der Hydroxylgruppe vor der Halomethylierung empfehlen, wie von V. Reichert, et. al., Pharmazie 5, 10 (1950) beschrieben.
Verbindungen der Formel I mit einem 5-Indanylrest können erhalten werden, indem man die entsprechende Säure in einem inerten Lösungsmittel in Gegenwart von N,N'-Dicyclohexyl-carbodiimid bei einem pH-Wert von etwa 2,5 und einer Temperatur von 20 bis 30"C mit 5-Indanol umsetzt. Man arbeitet üblicherweise mit äquimolaren Mengen der Reaktionsteilnehmer oder mit einem geringen Uberschuss des 5-Indanols. Die molare Menge des verwendeten N,N -Dicyclohexylcarbodi-imids ist der molaren Menge des 5-Indanols äquivalent. Geeignete Lösungsmittel für diese Reaktion können Dioxan, Tetra-hydrofuran, Äthylacetat, Dimethylformamid und Methylenchlorid sein.
Im folgenden soll beschrieben werden, wie die Verbindungen a) bis h), die für die erfindungsgemässe Herstellung der neuen Derivate benötigt werden, erhalten werden können, bevor das erfindungsgemässe Verfahren selbst anhand von Beispielen illustriert wird.
a) p-Chlormethylphenyl-acetylchlorid
(A) Bei einer Temperatur von -10 bis 0°C wird gasförmiger Chlorwasserstoff unter Rühren in ein Gemisch aus 102 g Phe-nylessigsäure, 67,5 g Paraformaldehyd und 67,5 g Zinkchlorid in 1000 ml Petroläther 1 Stunde lang eingeleitet. Dann wird noch etwa 1 Stunde bei Raumtemperatur gerührt, danach wird das Gemisch etwa 2 Stunden am Rückfluss gekocht, wobei während dieser Zeit gasförmiger Chlorwasserstoff in das Gemisch geleitet wird. Dann werden dem Reaktionsgemisch je 1000 ml Methylenchlorid und Wasser zugesetzt. Die organische Phase wird abgetrennt und die wässrige Phase wird zweimal mit Methylenchlorid extrahiert. Die vereinigten organischen Phasen werden viermal mit gesättigter Nafriumbicarbo-natlösung extrahiert. Die organische neutrale Phase wird über wasserfreiem Natriumsulfat getrocknet und filtriert und das Lösungsmittel wird im Vakuum entfernt, wobei man ein neutrales Nebenprodukt erhält, das in Beispiel 5 identifiziert wird. Die basische wässrige Phase wird abgesondert und mit kalter konzentrierter Salzsäure auf pH 2 bis 3 angesäuert.

dann dreimal mit Methylenchlorid extrahiert. Der Methylen-:hloridextrakt wird über wasserfreiem Natriumsulfat getrocknet, filtriert und vom Lösungsmittel befreit. Das resultierende ölige Säureprodukt wird an Silikagel unter Verwendung von Benzol und Benzol/Aceton als Eluierungsmittel chromatogra-phiert, wobei man die p-Chlormethylphenylessigsäure erhält, die aus heissem Chloroform umkristallisiert wird. F. 147-149°C.
(B) Ein Gemisch aus 1 g p-Chlormethylphenylessigsäure und 6 ml Thionylchlorid wird bei Raumtemperatur 25 Stunden gerührt, dann wird überschüssiges Thionylchlorid im Vakuum entfernt, wobei man das p-Chlormethylphenylacetylchlorid erhält.
Ersetzt man in a) (A) die Phenylessigsäure durch eine Säure aus folgender Tabelle I, so erhält man die entsprechenden Chlormethylderivate, die gemäss ai (B) in das Säurechlorid überführt werden können:

p-Chlormethylphenylglycin-hydrochlorid Ein Gemisch aus 2,03 g trifluoracetyliertem Phenylglycin und 0,8 g Zinkchlorid in Chlormethyläther wird 12 Stunden auf 65°C erwärmt. Überschüssiges Reagens wird im Vakuum entfernt und der Rückstand wird in Methylenchlorid gelöst, mit gesättigter Natriumbicarbonatlösung und mit gesättigter
40 Natriumchloridlösung gewaschen. Die neutrale organische Phase wird über Natriumsulfat getrocknet und zu einem Öl eingeengt, das durch Säulenchromatographie gereinigt wird. Auf gleiche Weise können die in Tabelle II aufgeführten Chlormethylderivate aus den dort angegebenen Säuren herge-45 -stellt werden:

5-Chlormethyl-2-tbienylacetylchlorid 2-Thiophencarbonsäure wird in Chloroformlösung mit Chlormethyläther in Gegenwart von 0,9 bis 2,2 Äquivalenten Aluminiumchlordig umgesetzt, wobei man die 5-Chlormethyl-2-thienylcarbonsäure erhält. Behandelt man die Säure mit überschüssigem Thionylchlorid bei Raumtemperatur während ca. 16 Stunden, so erhält man das Säurechlorid, das mit Diazo-methan das entsprechende Diazoketon ergibt. Eine methanolische Lösung des Diazoketons wird unter Stickstoff etwa 1 Stunde mit einer Hochdruck-Quecksilberlampe unter Verwendung eines Quarzfilters bestrahlt. Nach Aufarbeitung und Säulenchromatographie an Silikagel erhält man den 5-Chlor-methyl-2-thienylessigsäuremethylester. Dieser wird mit einem l:l-Gemisch aus Essigsäure und konzentrierter Salzsäure bei Raumtemperatur über Nacht hydrolysiert, wobei man die 5-Chlormethyl-2-thienylessigsäure erhält.
Ersetzt man im a) (B) die p-Chlormethylphenylessigsäure durch 5-Chlormethyl-2-thienylessigsäure, so erhält man das 5-Chlormethyl-2-thienylacetylchlorid.
e)
o-Hydroxymethylphenylessigsäure-lacton Das in a) erhaltene neutrale Nebenprodukt wird durch Sublimation im Vakuum (0,05 mm Hg bei 80°C) gereinigt, wobei man das o-Hydroxymethylphenylessigsäure-lacton vom F. 82°C erhält.
f)
o-Brommethylphenylacetylchlorid Zu einer Lösung von 5 ml mit Bromwasserstoff gesättigtem Eisessig wird bei 0°C eine Lösung von 0,55 g o-Hydroxyme-thylphenylessigsäurelacton in 2 ml Eisessig zugegeben. Das Gemisch wird bei Raumtemperatur 2 Stunden gerührt und dann 1 Stunde am Rückfluss gekocht, während Bromwasserstoffgas eingeleitet wird. Überschüssiges Lacton und Lösungsmittel werden im Hochvakuum bei Raumtemperatur entfernt,
is der resultierende ölige Rückstand wird dreimal mit Hexan verrieben, wobei man die o-Brommethylphenylessigsäure vom F. 110°C erhält.
Eine Lösung von 0,18 g o-Brommethylphenylessigsäure in überschüssigem Thionylchlorid wird bei Raumtemperatur 18 20 Stunden gerührt, dann wird nicht umgesetztes Thionylchlorid im Hochvakuum entfernt, wobei man das o-Brommethylphe-
nylacetylchlorid als öligen Rückstand gewinnt.
g)
25 o-Chlormethyl-p-methoxymandelsäurechlorid
Eine Lösung von 1,1 g 2-Chlormethyl-4-methoxymandel-säure (Herstellung siehe B. Reichert et al., Pharmazie 5,10 (1950)) in 25 ml Thionylchlorid wird bei Raumtemperatur etwa 16 Stunden gerührt, dann wird überschüssiges Thionyl-30 chlorid im Vakuum entfernt und man erhält das o-Chlorme-thyl-p-methoxymandelsäurechlorid in Form eines Öls.

azabicyclo[4.2.0]oct-2-en-2-carbonsäure-hydrochlorid
Eine warme Lösung von 0,12 g 3-[(Acetyloxy)methyl]-7-[[2-[4-(chlormethyl)phenyl]acetyl]-8-oxo-5-thia-l-azabicy-clo[4.2.0]oct-2-en-2-carbonsäure und 0,12 g Isothioharnstoff 40 in 30 ml Äthanol wird 4 Stunden am Rückfluss gekocht, dann wird das Lösungsmittel im Hochvakuum bei Raumtemperatur entfernt, wobei man ein Öl erhält. Dieses wird mit 80 ml Benzol/Aceton (2:1) verrieben, wobei das 3-[(Acetyloxy)methyl]-7-[[2-[4-(isothioharnstoffmethyl)phenyl]acetyl]amino]-8-oxo-f 45 thia-l-azabicyclo[4.2.0]oct-2-en-2-carbonsäurehydrochlorid erhalten wird.
Auf analoge Weise erhält man bei Verwendung der aus folgender Tabelle IV ersichtlichen halomethylierten Derivate und Harnstoffderivate die ebenfalls aus Tabelle IV ersicht-50 liehen Produkte:

Ein Gemisch aus 1,2 g des Natriumsalzes der 3-[(Acetyl-oxy)-methyl]-7-[[2-[4-(isothioharnstoffmethyl)phenyl]acetyl]-amino]-8-oxo-5-thia-l-azabicyclo[4.2.0]oct-2-en-2-carbon-säure und 0,5 g N-Chlormethyl-N-methylurethan in 40 ml Dimethylformamid wird 2 Stunden bei Raumtemperatur gerührt 10 und dann in Eiswasser gegossen und abdekantiert. Der ölige Rückstand wird in 75 ml Äthylacetat aufgenommen, dann wird mit 5 ml verdünnter wässriger Natriumbicarbonatlösung und 15 ml Wasser gewaschen und über Magnesiumsulfat getrocknet, abfiltriert und eingeengt, wobei man den 3-[(Acetyloxy)me- 1S thyl]-7-[[2-[4-(isothioharnstoffmethyl)phenyl]acetyl]amino]-8-oxo-5-thia-l-azabicyclo[4.2.0]oct-2-en-2-carbonsäure-N-äth~ oxycarbonyl-N-methylaminomethylester erhält.

3-[(Acetyloxy)methyl]-7-[[2-[4-(isothioharnstoff-methyl)phenyl]2-(5-indanyloxycarbonyl)acetyl]amino]-8-oxo-5-thia-l-azabicyclo[4.2.0]oct-2-en-2-carbonsäure Zu 25,3 Millimol des Cephalosporins in 35 ml Dioxan wird 6 n-Salzsäure bis zum pH-2,5 zugesetzt. Dann werden 24,1 Millimol N^'-Dicyclohexylcarbodiimid in 35 ml Dioxan zugegeben und das Gemisch wird bei Raumtemperatur 15 bis 20 Minuten gerührt, worauf 24,1 Millimol 5-Indanol zugegeben werden. Das Gemisch wird dann 4 Stunden gerührt, der gebildete N,N'-Dicyclohexylharnstoff wird abfiltriert und das Filtrat wird dreimal mit Methylisobutylketon extrahiert. Der organische Extrakt wird mit Wasser gewaschen, über Magnesiumsulfat getrocknet und im Vakuum zur Trockene eingeengt, wobei man die 3-[(Acetyloxy)methyl]-7-[[2-[4-(isothioharnstoffme-thyl)phenyl]-2-(5-indanyloxycarbonyl)acetyl]amino]-8-oxo-5-thia-l-azabicyclo[4.2.0]-oct-2-en-2-carbonsäure erhält.
Beispiel 6
3-[(Acetyloxy)methyl]-7-[[2-[4-(isothioharnstoff-
methyl)-phenyl]acetyl]amino]-8-oxo-5-thia-l-azabicyclo[4.2.0]oct-2-en-2-carbonsäure Ein Gemisch aus 1,2 g 3-[(Acetyloxy)methyl]-7-amino-8-oxo-5-thia-l-azabicyclo[4.2.0]oct-2-en-2-carbonsäure und 1,2 g p-Isothioharnstoffmethylphenylessigsäurechlorid-HCl in 55 ml Äthylacetat wird 1,5 Stunden am Rückfluss gekocht, dann wird das Lösungsmittel entfernt, wobei man die 3-[(Ace-tyloxy)-methyl]-7-[[2-[4-(isothioharnstoffmethyl)phenyl]-acetyl]amino]-8-oxo-5-thia-l-azabicyclo[4.2.0]oct-2-en-2-carbonsäure erhält, die durch Säulenchromatographie weiter gereinigt werden kann.
Verwendet man eine entsprechende Menge der 6-Amino-3,3-dimethyl-7-oxo-4-thia-l-azabicyclo[3.2.0]heptan-2-car-bonsäure anstelle der 3-[(Acetyloxy)methyl]-7-amino-8-oxo-5-thia-l-azabicyclo[4.2.Öjoct-2-en-2-carbonsäure in obigem Verfahren, so wird die 6-[[2-[4-(Isothioharnstoffmethyl)phe-nyl]acetyl]amino]-3,3-dimethyl-7-oxo-4-thia-l-azabicy-clo[3.2.0]heptan-2-carbonsäure erhalten.
Verwendet man in obigem Verfahren entsprechende Mengen des Cephalosporinderivats und der Isothioharnstoffverbin-dung als Säurechlorid-Hydrochlorid gemäss folgender Tabelle VI anstelle von 3-[(Acetyloxy)methyl]-7-amino-8-oxo-5-thia-l-azabicyclo[4.2.0]oct-2-en-2-carbonsäure und p-Isothioharn-stoffmethylphenylessigsäure, so erhält man die ebenfalls aus Tabelle VI ersichtlichen Produkte:

Verwendung der nach dem Verfahren gemäss Anspruch 1 erhaltenen Verbindungen der Formel I zur Herstellung entsprechender Verbindungen, in welchen W den Rest-COOR.4, worin R4 Indanyl ist, bedeutet, dadurch gekennzeichnet, dass man die Verbindungen der Formel I, in denen W den 10 Rest —COOH bedeutet, mit 5-Indanol in einem inertenLösungsmittel in Gegenwart von N,N'-Dicyclohexylcarbodi-imid beim pH-Wert etwa 2,5 bei einer Temperatur von 20 bis 30°C umsetzt.R1^IIR- N — C - S - CR3Aryl iv-(che)n
