Description
The invention relates to a process for the production of proteins from whey and protein and starch-containing natural products, in particular from cheese whey and cereals.
Whey from cheese factories poses a worldwide problem. On the one hand, it is a poorly usable by-product in cheese production in large quantities, but on the other hand it contains such valuable ingredients that destruction, which still occurs in some cases, is incompatible with the problems of world hunger and environmental pollution.
Methods for utilizing whey are known. Depending on its origin, whey consists of approx. 4-5% lactose and approx. 0.7% protein, the rest are salts and water. Dry whey as an additive for animal feed is obtained by evaporation and drying. The end product consists of about 11-12% protein, about 60-65% lactose with salts. However, the production is very energy-intensive and therefore expensive. 5 The different ways of fermenting the milk sugar in the whey are described in French Pat. No. 2,049,425, British Pat. No. 1,383,703, US Pat. No. 3,818,109 and DE-OS 2 403 306 and DE-OS 2 410 349.
However, none of these processes lead to a product that is a balanced diet for humans and animals. Not even the proteins contained in the products have a balanced amino acid spectrum. With regard to the amino acid composition, dry whey appears to be the most suitable, as it contains sufficient amounts of methionine and lysine, but due to the excess of carbohydrates in the form of milk sugar it cannot be regarded as a wholesome food.
Yeasts grown on whey that comply with the F.D.A. regulations have a deficit in the amino acids mentioned and cannot be used as sole foods because of their high hydrochloric and nucleic acid content. The usual proportion in food and feed is between 5 and 30%.
An improvement in the amino acid composition can be achieved by adding, for example, artificially produced methionine. These compositions are also unsatisfactory, since even then only a mixed component for animal feed can be obtained.
Mixtures of single-cell proteins, cereals and whey powder are known. For this purpose, however, the individual components are previously produced in the desired purity. However, the production of the individual components is associated with high costs, the main costs being the evaporation and drying of the whey, the processing of the substrate solution, which is only thinned in relatively diluted form, for example on molasses or sulfite waste liquor, the drying of the yeast and the processing of the cereals .
The object of the invention is to provide a method for producing a balanced food and feed, preferably for the rearing of calves.
Another task is to save on drying costs in the manufacture of the end product.
The solution according to the invention lies in a process for the production of proteins from whey and natural products containing protein and 45 starch, which is characterized in that whey is combined with natural products containing protein and starch and the mixture obtained in a first stage of acidic and / or enzymatic starch hydrolysis is subjected, whereupon the unchanged protein is separated and isolated as the first protein fraction, and the sugars formed are reacted with microorganisms in single-cell protein in a second stage and the second protein fraction thus obtained is then evaporated and dried.
A process was thus found in which the whey obtained from the 55 cheese factories also serves as a swelling agent for cereals, for example, and also as a diluent in their hydrolysis. The diluted whey, as it comes from the cheese dairy, is mixed with cereals, which are broken down into sugar in the whey itself. The sugar-containing 60 whey is then subjected to fermentation with yeast. After the fermentation, the mixture is evaporated and dried.
Durch die Vermeidung von zu/ The fermentation can be carried out with Torula yeast, which does not attack the milk sugar, so that a dry product containing protein and lactose is obtained. However, it is also possible to work with a yeast that can be used to both sugar from cereal hydrolysis and lactose, such as Saccharomyces fragilis. This way, all sugar is bound. By avoiding too /

The diagram shows a storage container 1 for whey. Between the lines 1 and 4 there is a pump 3. The two containers 5 and 5 'are of identical construction and are equipped with an agitator 6 and 6', a heating water inlet 7 and 7 ', a heating water outlet 8 and 8', respectively Connection pieces are connected with a double jacket (not shown) or with heating coils. Both containers 5 and 5 'are connected to each other by a line 9. A line 10 leads from container 5 'via a pump 11 and a connecting line 12 to the fermenter 13. The fermenter 13 consists of a boiler 14 which has a ventilation device 15 in the lower part of the boiler 14 and a foam separator 16 in the upper part of the boiler 1 . Baffles 17 and 17 'are attached to the boiler walls. The inlet connector 18 and the outlet connector 19 are provided for cooling water. Between the reservoir 1 there is a further connecting line 20, pump 21 and line 22 directly to the fermenter 13. In the lower part of the fermenter 13 is the harvesting line 23, in which the control valve 24 is installed. The fermenter boiler stands on a pressure cell 25, which connects a cable 26 to the optical weight display 27 and a further cable 28 to the control valve 24. Line 29 leads via a throttle valve 30 and line 31 to a multi-stage evaporator system 32. The evaporator shown consists of three stages. The way of working is known. The vapor from the first stage 321 goes via line 33 and heats up the concentrate, which goes via line 34 to the second stage 322. The same is done with the vapors via line 35, which is the concentrate which is passed through line 36 to the 3rd stage 323 of the evaporator. The pipeline 37 leads to the condenser (not shown). The pump 39 is located in the concentrate line 38; the connecting line 40 leads to the spray dryer 41. A container 42 is equipped with a stirrer 43, a feed line 44 for the substrate from the fermenter and a discharge line 45 for the premixing to the homogenizer 46 and its discharge line 47. 48 represents the packaging, 49 a Mixing plant.
In operation, a partial stream of the whey is conveyed from the container 1 via the line section 2 by means of the pump 3 and the further line section 4 into the stirred tank 5. The high-speed comminution stirrer 6 mixes the added grain 50, flour or corn. After uniform heating of the contents by introducing the heating medium into connection 7 and discharging through 8, the first stage of the enzymation takes place, which leads to a liquefaction of the starch of the grain. The liquefied starch is conveyed via line 9 into the second enzymatic tank 5 ', where the liquefied starch is saccharified. After the remaining solids have been separated off by means of separators or filters (not shown), the liquid is pumped via line 10 by means of pump 11 via line 12 into a fermenter 13. Here the conversion into yeast protein takes place in a known manner with vigorous aeration and the participation of special yeasts, such as, for example, Candida utilis, Candida tropicalis, Saccharomy-ces fragilis. In order to dilute the enzymed substrate, whey is conveyed from the storage container 1 via the line pieces 20 and 22 and pump 21 into the fermenter. The fermentation takes place automatically and is controlled via line 28 via a weighing system, consisting of pressure cell 25, connecting line 26, display 27 and control valve 24. The yeast-containing substrate is harvested via a line 23, the valve 24, and can be fed directly to a multi-stage evaporator 23 in the case of continuous operation.
However, some or all of the crop with oil or liquid fat can also be emulsified in a container 43 via line 44 via a homogenizer 46 and then passed through the evaporator 2.
The degree of evaporation can be selected, but the product must still be pumpable. Via a pump 39 and a line 40, the concentrate is fed, for example, to a spray dryer 41 and dried there, packaged in bags 48, possibly mixed with further ingredients in a mixing system 49 and stored as finished product 51.
In the process, cereal products such as raw cereals, flours, etc. are mixed with the whey, subjected to acidic or enzymatic degradation and then the cereal sugar (together with the lactose of the whey) is combined after the protein components, whether by centrifugation, filtration or Ultrafiltration have been separated.
Since it is known that the fiber content of the grain is of great importance in the nutrition of humans and animals and also contains important vitamins, these cereals can be used either in unpeeled or peeled form. In the case of unhulled cereals, such as maize, the fiber components are in the precipitated protein production and form an important component there.
On the other hand, if you want to work on the highest possible protein concentration, the cereals are used in the form of flours. The cereals can also be degreased and sterilized before they are added to the whey and before hydrolysis. Corn can be subjected to the known swelling with water. The corn steep liquor is an excellent accelerating agent for the subsequent fermentation.
Depending on the content of fiber components, these can be separated by separation before the actual protein extraction and processed for themselves. Such products are then preferably mixed into the finished products. However, the separation can also take place between the possible 2-stage amylase degradation.
For faster solution resp. Grain degradation during hydrolysis can also be subjected to a previous temperature treatment.
The whey proteins can be precipitated at different pH, taking into account the isoelectric point, for the purpose of separation into different protein fractions. In general, work is carried out at pH 3–5, when using ultrafiltration also at higher pH (6-8).
If the process described above is carried out with a higher grain content, a protein fraction with a smaller albumin content and thus also with less lysine in the total protein is obtained: when using less grain, relatively more albumin is obtained in the protein fraction and consequently a higher lysine content.
Since such protein fractions can be used as a mixture directly or after the addition of yeast as food or feed, the desired protein composition and also the type of cereals are precisely defined beforehand.
The protein fractions can be used as the basis for a low-carbohydrate diet, such as in the baked state as protein biscuits or as a beverage basis. By adding and emulsifying fats and oils, these basic products can be used to produce nutrients for humans and animal feed. The latter in particular for rearing calves and other young cattle.
The sugar-liquor solutions obtained after protein separation can either be wholly or partially stitched and dried, depending on the desired residual sugar content.
In It is also possible to separate the proteins together with the yeast, be it by separation or ultrafiltration. In

Bei Verwendung von Bakterien ist dieser Eiweissgehalt noch beträchtlich höher. this results in a protein-containing dry yeast, which can contain up to 65% protein. When using bacteria, this protein content is considerably higher.
Beispiel example
1250 liters of cheese whey with approx. 4.2% lactose and 0.7% albumin content are poured into a 2000-liter stirred kettle equipped with a high-quality comminution stirrer and heated to 50-60 ° C. Now 40 kg of broken, unhulled corn and 50 kg of wheat flour with about 12% protein content are stirred in. After adding a little calcium chloride, about 0.15% (calculated on starch) of a high-temperature alpha-amylase is added and the temperature is increased to 75-80 ° C. After 1-2 hours the grain portion is practically in solution, i.e. H. with the exception of the grain fibers and a certain amount of protein.
After this time, a little calcium chloride is added and the pH is adjusted to 3.8 with hydrochloric acid. The temperature is lowered to 50-55 ° C. Now 0.15% (based on starch) amidoglucosidase is added, the mixture is stirred a little further and the whole is left to stand for 24 hours. During this time, the corn starch has been degraded to such an extent that it can then be fermented to yeast.
After 24 hours, the mixture is heated to 80-90 ° C. The egg whites are centrifuged off together with the fiber components and the whey albumin which has also been precipitated. The result is 17-25 kg, depending on the fiber content and the degree of drying, 5 protein fractions with 60-65% protein. The amino acid composition shows an excellent spectrum, especially with regard to essential amino acids.
The centrifuged liquid, which represents the saccharified starch of the cereals and the milk sugar of the whey, is mixed with a yeast, Candida utilis or Saccharomyces fragi-lis, in a fermentation plant.
A yeast strain must be used, which can supply glucose as well as maltose and lactose. The breeding is carried out after addition of the known amounts of ammonium sulfate, ammonium phosphate and trace elements.
Depending on the starch content of the grain, this results in 50-60 kg of dry yeast. Part of this dry yeast can be used to improve the protein fraction, although for certain uses this addition may only be so high that the nucleic acids present in the yeast do not interfere.
Of course, the yeast can be freed of nucleic acids in a known manner before being added to the protein fraction.
