Description
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung des neuen 3-Cyan-N-(N,N-dimethylaminopro-pyO-iminodibenzyls der Formel

Uh3
sowie von Säureadditionssalzen dieser Verbindung.
Es wurde gefunden, dass das erfindungsgemässe 3-Cyan--N-(N,N-dimethylaminopropyl)-iminodibenzyl und dessen Säureadditionssalze, welche neue Verbindungen darstellen, sich durch starke antidepressive Wirkung auszeichnen, welche sich tierexperimentell vor allem in einer ausserordentlich starken Hemmung der neuronalen Serotoninaufnahme äussert. Sie können somit beispielsweise zur Behandlung von Depressionen endogener oder exogener Genese eingesetzt werden. Von besonderem Vorteil ist, dass anticholinerge Nebenwirkungen, die sich beispielsweise in Mundtrockenheit, Obstipation, Tachykardie und/oder Accomodationsstörun-gen äussern können, auf ein Minimum beschränkt sind.
Das 3-Cyan-N-(N,N-dimethylaminopropyI)-iminodiben-zyl und dessen Säureadditionssalze werden erfindungsge-
15 mit Kupfer-I-cyanid umsetzt, wonach man erwünschtenfalls eine erhaltene Base der Formel I in ein Säureadditionssalz überführt.
Die erfindungsgemässe Umsetzung des Bromderivats der Formel VII mit Kupfer-I-cyanid wird vorzugsweise in einem 20 aprotischen, polaren organischen Lösungsmittel, wie Dime-thylformamid, bei erhöhter Temperatur, vorzugsweise in einem Temperaturbereich zwischen etwa 100°C und dem Siedepunkt des Reaktionsgemisches, durchgeführt. Das ge-(VII) wünschte Endprodukt der Formel I fällt zusammen mit 25 Ausgangsmaterial und geringeren Anteilen 3-Cyan- und 3-Bromiminodibenzyl an. Das gewünschte Produkt kann aus diesem Gemisch beispielsweise durch Extraktion in einem organischen Lösungsmittel mit einer wässerigen Säure und, nach Alkalisierung der wässerigen Phase, Extrak-30 tion mit einem organischen Lösungsmittel, Entfernung des Lösungsmittels und Destillation des Rückstandes, von den Nebenprodukten abgetrennt werden.
Das erfindungsgemässe 3-Cyan-N-(N,N-dimethylamino-propyl)-iminodibenzyI der Formel I bildet Salze sowohl mit 35 anorganischen als auch mit organischen Säuren, z.B. mit Halogenwasserstoffsäuren, wie Chlorwasserstoffsäure oder Bromwasserstoffsäure, mit anderen Mineralsäuren, wie Schwefelsäure, Phosphorsäure oder Salpetersäure, sowie mit organischen Säuren, wie Weinsäure, Citronensäure, Cam-40 phersulfonsäure, Methansulfonsäure, Toluolsulfonsäure, Salicylsäure, Ascorbinsäure, Maleinsäure, Fumarsäure oder Mandelsäure usw. Bevorzugte Salze sind die Hydrohaloge-nide, insbesondere das Hydrochlorid. Die Säureadditionssalze werden vorzugsweise in einem geeigneten Lösungsmittel, 45 wie Äthanol, Aceton oder Acetonitril, durch Behandeln der freien Base mit der entsprechenden, nicht wässerigen Säure hergestellt.
(I) Das 3-Cyan-N-(N,N-dimethylaminopropyl)-iminodiben-zyl der Formel I ist eine kristalline, feste Substanz, die in so Dimethylsulfoxid, Dimethylformamid oder in chlorierten Kohlenwasserstoffen, wie z.B. Chloroform, Methylenchlorid, in Alkanolen, wie Methanol oder Äthanol, oder auch in Äther oder Benzol relativ gut löslich und in Wasser relativ schwer löslich ist.
55 Die Säureadditionssalze entsprechend der Base der Formel I sind kristalline, feste Substanzen. Sie sind in Dimethylsulfoxid und Dimethylformamid, in Alkanolen, wie Methanol oder Äthanol, und in Chloroform, Methylenchlorid und Wasser gut löslich. Sie sind in Benzol, Äther und n-He-6o xan relativ schwer löslich.
Wie oben gesagt, zeichnen sich die erfindungsgemässen Endprodukte durch starke antidepressive Wirkung aus, welche beispielsweise am Modell der Hemmung der Serotoninaufnahme in den Neuronen des Hirns der Ratte sichtbar wird. 65 Zum Nachweis dieser antidepressiven Wirkung wurde das erfindungsgemässe 3-Cyan-N-(N,N-dimethylaminopropyl)--iminodibenzyl (Produkt A) mit bekannten Analoga gemäss den nachstehenden Versuchsanordnungen geprüft:

Produkt A: 3-Cyan-N-(N,N-dimethylaminopropyl)-imi-nodibenzyl-hydrochlorid (Produkt der Erfindung)
Produkt B: N-(N,N-Dimethylaminopropyl)-iminodiben-zyl-hydrochlorid (bekanntes Produkt)
Produkt C: 3-Chlor-N-(N,N-dimethylaminopropyl)-imi-nodibenzyl-hydrochlorid (bekanntes Produkt).
1. Hemmung der Serotoninaufnahme in vitro
Die Aufnahme von Serotonin in Synaptosomen des Vorderhirns von Ratten wurde nach J. Pharmacol. exp. Ther 181, 36, 1972, geprüft. Eine Hemmung von 50% (ED50) konnte bei folgenden Konzentrationen gemessen werden:

Das erfindungsgemässe Produkt A ist in diesem Test somit 52 bzw. 4,2 mal stärker wirksam als die beiden bekannten Vergleichspräparate.
2. Hemmung der Serotoninaufnahme ex vivo
Eine ähnliche Versuchsanordnung wie unter 1. wurde verwendet. Die Prüfsubstanzen wurden jedoch i.p. gespritzt; nach 1 h wurde die Serotoninaufnahme gemessen. Unbehandelte Ratten dienten als Kontrolle. Es konnten folgende ED50-Werte ermittelt werden:

Das erfindungsgemässe Produkt A ist in diesem Test in 21 bzw. 2,4 mal kleineren Dosen wirksam als die beiden bekannten Vergleichspräparate.
3. Hemmung der Serotoninwiederaufnahme in vivo
Die Wirkung auf die sogenannte «Membranpumpe»
wurde nach der in Biochem. Pharmacol. 20, 707, 1971, angegebenen Methode geprüft. Es konnten folgende ED50-Werte ermittelt werden:

Das erfindungsgemässe Produkt A ist in diesem Test in 18 bzw. 9 mal kleineren Dosen wirksam als die beiden bekannten Vergleichspräparate.
Als weitere Bestätigung der antidepressiven Wirkung der erfindungsgemässen Endprodukte dienen die nachstehenden Ergebnisse für die Hemmung der Serotoninaufnahme in Blutplättchen an der Ratte.
4. Hemmung der Serotoninaufnahme in vitro
0,5 ml Blutplasma wurde mit verschiedenen Konzentra tionen an Prüfsubstanz in 50 [i,I physiologischer Kochsalz lösung versetzt. Nach 5minutiger Inkubation bei 37°C
wird Serotonin (C14; 0,1 |iM) hinzugefügt und das Ganze noch 5 min bei 37°C inkubiert. Die Serotoninkonzentration wird an einem Cu-Messgerät ermittelt. Es wird ebenfalls ein Kontrollversuch ohne Prüfsubstanz durchgeführt. Durch
Variieren der Konzentration an Prüfsubstanz (drei verschiedene Konzentrationen) ermittelt man diejenige Konzentration, die zu 50% Hemmung der Serotoninaufnahme führt (ED50).

Das erfindungsgemässe Produkt ist in diesem Test somit 130 bzw. 20 mal stärker wirksam als die beiden bekannten Vergleichspräparate.
5. Hemmung der Serotoninaufnahme in vivo
Ratten erhielten zweimal täglich während 4 Tagen 5,0 mg Prüfsubstanz pro kg intraperitoneal injiziert. 12 h nach der letzten Administrierung wurden die Tiere geopfert; Serotonin wurde spektrofluorimetrisch gemäss J. Pharmacol. exp. Ther. 117, Seiten 82 ff, 1956, und der Proteingehalt kolorimetrisch gemäss J. Biol. Chem. 193, Seiten 265 ff, 1951, bestimmt.

Die erfindungsgemässen Produkte können als Heilmittel, z.B. in Form pharmazeutischer Präparate, Verwendung finden, welche sie in Mischung mit einem für die enterale, z.B. orale oder parenterale Applikation geeigneten pharmazeutischen, organischen oder anorganischen inerten Trägermaterial, wie z.B. Wasser, Gelatine, Milchzucker, Stärke, Ma-gnesiumstearat, Talk, pflanzliche Öle, Gummi arabicum, Polyalkylenglykole, Vaseline, usw., enthalten. Die pharmazeutischen Präparate können in fester Form, z.B. als Tabletten, Dragées, Suppositorien, Kapseln oder in flüssiger Form, z.B. als Lösungen, Suspensionen oder Emulsionen, vorliegen. Gegebenenfalls sind sie sterilisiert und bzw. oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten.
Zweckmässige pharmazeutische Dosierungsformen enthalten etwa 1-200 mg der Verbindung der Formel I bzw. eines ihrer Salze. Zweckmässige organische Dosierungsbereiche liegen bei etwa 1 mg/kg pro Tag bis etwa 10 mg/kg pro Tag. Zweckmässige parenterale Dosierungsbereiche liegen bei etwa 0,1 mg/kg pro Tag bis etwa 1 mg/kg pro Tag. Es können indessen die erwähnten Bereiche nach oben oder nach unten ausgedehnt werden, je nach individuellem Bedarf und Vorschrift des Fachmannes.
Das folgende Beispiel erläutert die Erfindung. Alle Temperaturen sind in °C angegeben.
Beispiel: 1,30 g (0,0036 Mol) 3-Brom-N-(N,N-dimethyl-aminopropyl)-iminodibenzyl und 0,41 g (0,0046 Mol) Kupfer-I-cyanid werden mit 5 ml Dimethylformamid 6 h unter Rückfluss gekocht. Man kühlt auf Raumtemperatur ab,
gibt 20 ml Chloroform zu und filtriert. Dann wird die Chloroformphase mit Wasser gewaschen, getrocknet und einge5

dampft. Als Rückstand verbleibt ein braunes Öl mit der folgenden Zusammensetzung (gemäss Gaschromatogramm): 72 % 3-Cyan-N-(N,N-dimethylaminopropyl)-iminodibenzyl, 4 % 3-Brom-N-(N,N-dimethylaminopropyl)-iminodibenzyl, 16% 3-Cyan-iminodibenzyl und 1% 3-Bromiminodiben-zyl.
Die Reinigung des Rohproduktes kann wie folgt erfolgen: Das braune Öl wird in 20 ml Chloroform gelöst und dreimal mit 10 ml 5%iger Methansulfonsäure extrahiert. Die sauren Teile wäscht man mit 20 ml Essigester und stellt mit konzentrierter wässeriger Natronlauge alkalisch. Die erhaltene Emulsion wird zweimal mit je 20 ml Chloroform ausgeschüttelt. Man wäscht dann die Chloroformphase mit 50 ml Wasser, trocknet sie über Magnesiumsulfat und dampft sie unter vermindertem Druck ein. Der ölige Rückstand wird unter stark vermindertem Druck destilliert. Man erhält 3-Cyan-N-(N,N-dimethylaminopropyl)-iminodibenzyl als ein gelbes Öl vom Siedepunkt 240° (0,12 mm), das über Nacht kristallisiert; Fp. 53-55°.
Das als Ausgangsverbindung verwendete 3-Brom-N--(N,N-dimethyIaminopropyl)-iminodibenzyl kann wie folgt hergestellt werden:
1,50 g Natriumhydrid-Dispersion (50%ig in Mineralöl) werden mit 10 ml n-Hexan gewaschen und in 5 ml N,N-Di-methylformamid suspendiert. Dieser Suspension tropft man bei Raumtemperatur und innert 10 min eine Lösung von 5 2,2 g 3-Brom-iminodibenzyl in 5 ml Dimethylformamid zu. Man rührt die erhaltene Lösung V4 h bei 70°, kühlt sie dann auf Raumtemperatur ab und versetzt sie tropfenweise mit einer Lösung von 3,67 g N,N-Dimethylaminopropyl-chlorid in 4 ml Dimethylformamid. Das Reaktionsgemisch io wird 16 h bei 60° gerührt und anschliessend mit 40 ml Wasser versetzt. Die erhaltene Emulsion extrahiert man zweimal mit 20 ml Essigester; die Essigester-Extrakte werden vereinigt, achtmal mit 50 ml Wasser gewaschen und dreimal mit 20 ml 10%iger Methansulfonsäure extrahiert. Man 15 wäscht die sauren Teile zweimal mit 20 ml Essigester und stellt sie mit konzentrierter Natronlauge wieder alkalisch. Das basische Produkt wird zweimal mit 30 ml Chloroform extrahiert, über Magnesiumsulfat getrocknet und unter vermindertem Druck eingeengt. Der braune, ölige Rückstand 20 wird unter stark vermindertem Druck destilliert. Man erhält 3-Brom-N-(N,N-dimethylaminopropyl)-iminodibenzyl, das bei 230° (0,08 mmHg) siedet.
Claims (2)
- 625227PATENTANSPRÜCHE 1. Verfahren zur Herstellung einer tricyclischen Verbindung der Formel mäss dadurch hergestellt, dass man eine Verbindung der Formel(I) 10(VII)ch2ch2ch2n yC H3ch2ch2ch2nXch-/CH3 ch3und von Säureadditionssalzen dieser Verbindung, dadurch gekennzeichnet, dass man eine Verbindung der Formelch2ch2ch2i<ch3 ch3mit Kupfer-I-cyanid umsetzt, wonach man erwünschten-falls eine erhaltene Base der Formel I in ein Säureadditionssalz überführt.
- 2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man eine erhaltene Base der Formel I in das Hy-drochlorid überführt.
