Unter «niederer Alkyl-», «niederer Alkoxy-» und «niederer Alkylmercapto-Rest» der Substituenten Ri, R2, R3, R4 25 und Rs sind Reste mit 1-6, vorzugsweise 1-3 Kohlenstoffatome zu verstehen. Insbesondere findet bei dem niederen Alkyl-Rest die Methyl-Gruppe Verwendung.
Unter Halogen versteht man Fluor, Chlor und Brom, vorzugsweise Chlor.
30 Unter einem «Dioxan-Ring» ist vorzugsweise der «1,3-Dioxan-Ring» zu verstehen.
Die niederen, geradkettigen oder verzweigten Alkylreste der Gruppe A können 1-4 Kohlenstoffatome aufweisen und bedeuten vorzugsweise die Methylen-, Äthylen-, n-Propylen-, 35 n-Butylen-, Methylmethylen-, Methyläthylen-, Methyl-pro-pylen-, Dimethylmethylen- oder Dimethyläthylen-Gruppe.
Die neuartigen Benzopyrone der Formel I haben eine antiallergische Wirksamkeit, die an der passiven cutanen Anaphylaxie bei Ratten nachgewiesen wurde. Sie besitzen 40 auch einen hemmenden Einfluss auf intradermal injiziertes Histamin.
Weiterhin wurde gefunden, dass Verbindungen der Formel I wertvolle Zwischenprodukte bei der Synthese pharmazeutisch verwertbarer Substanzen darstellen. 45 Das erfindungsgemässe Verfahren zur Herstellung der Verbindungen der Formel I ist dadurch gekennzeichnet, das man 2-Hydroxyacetophenone der Formel II

auch nachträglich ineinander überführt werden können, und anschliessend gewünschtenfalls entweder die unter den Reaktionsbedingungen anfallenden Salze in die freien Basen der Formel I oder die freien Bahnen in die pharmakologisch verträglichen Säureadditionssalze überführt.
Die «niedere Alkyl-Gruppe» des Substituenten R der Formel III kann 1-6, vorzugsweise 1-4 Kohlenstoffatome enthalten. Insbesondere kommen die Methyl- und die Äthyl-Gruppe in Frage. Reaktive Derivate der Formel IV sind die
Halogenide, Mesyloxy- und Tosyloxy-Verbindungen.
Das erfindungsgemässe Verfahren zur Herstellung der Verbindungen der Formel I wird vorteilhaft gemäss der bekannten «Claisen-Kondensation» durchgeführt, wobei zunächst 2-Hydroxy-acetophenone der Formel II mit Pipera-zinyl-fettsäureestern der Formel III in Gegenwart alkalischer Kondensationsmittel zu 1,3-Diketonen der Formel VI bzw. ihrer tautomeren Formel VII umgesetzt werden,

wobei in den Formeln VI und VII n, A, Ri, R2, R3, R4 und R5 die oben genannte Bedeutung haben.
Diese 1,3-Diketone werden anschliessend unter sauren Reaktionsbedingungen durch Wasserabspaltung zu den gewünschten Benzopyron-Derivaten cyclisiert.
Die 1,3-Diketone der Formel VI bzw. deren tautomere Formen der Formel VII sind neu und können als Natrium-Salze isoliert werden.
Als alkalische Kondensationsmittel kommen die in der Literatur für die Claisen-Kondensation bekannt gewordenen Metalle, Metallhydroxyde, Metallhydride, Alkoholate usw. bei Verwendung entsprechender Lösungsmittel wie Alkohol, nicht reaktive Aromaten, Dioxan, stark polare aprotische Lösungsmittel wie Hexametapol u.a. in Frage, wobei je nach Löslichkeit auch unpolare Lösungsmittel, wie Ligroin und andere Petrolfraktionen Anwendung finden können.
Die Ringschlussreaktion erfolgt vorteilhaft im sauren Milieu, vorzugsweise mit wässriger Salzsäure, alkoholischer Salzsäure, Dioxan, Polyphosphorsäure, Eisessig, konzen-triertre Schwefelsäure oder unter milden Bedingungen, wie beispielsweise mit Polyphosphorsäureester.
Für die Claisen-Kondensation kommen verschiedene spezielle Verfahrensvarianten in Frage. So kann z. B. die phenolische Hydroxyl-Gruppe der 2-Hydroxyacetophenone der Formel II vorübergehend durch eine leicht abspaltbare Schutzgruppe blockiert werden; desgleichen die Substituenten der R2, R3, R4 oder Rs, wenn diese eine Hydroxyl-Gruppe bedeuten. Als Schutzgruppe kommen vorzugsweise ein Formyl- oderTetrahydropyranyl-Rest in Betracht.
Für den Fall, dass in diesen Verbindungen R2, R3, R4 oder Rs einen durch Hydroxyl substituierten Alkoxy-Rest bedeuten, kann zusätzlich auch die alkoholische Hydroxyl-Gruppe durch die oben angeführte Gruppe geschützt werden.
Ferner kann auch, wenn R2, R3, R4 oder Rs in der Formel II einen mehrfach hydroxylierten Alkoxy-Rest bedeuten, dieser Rest zusätzlich zu der bereits geschützten phenolischen Hydroxy-Gruppe mit Aldehyden oder Ketonen unter Bildung eines Dioxan- oder Dioxolan-Ringes geschützt werden.
Die Abspaltung der oben aufgeführten Schutzgruppen erfolgt meist bereits in dem sauren Milieu der Ringschluss-reakion. Sie kann aber auch speziell durch eine wässrige saure Hydrolyse mit z. B. Salzsäure oder Schwefelsäure durchgeführt werden.
30 Weitere spezielle Verfahrensvarianten für Verbindungen der Formel I, in der R2-R5 eine gegebenenfalls durch Hydroxyl, Halogen oder durch den Oxiran-Ring substituierte Alkoxygruppe bedeuten, bestehen darin, dass man Verbindungen der Formel I, in der R2-R5 eine Hydroxylgruppe 35 bedeuten, nachträglich mit einer entsprechend substituierten Halogenalkyl-Verbindung alkyliert, wobei die Halogengruppe, beispielsweise durch den Mesyloxy- oder Tosyloxy-Rest ersetzt werden kann.
Als weitere Verfahrens variante zur Herstellung von Ver-40 bindungen der Formel I, in der R2-R5 eine Hydroxylgruppe bedeuten können, besteht darin, dass man Substanzen der Formel I, der der R2-R5 einen niederen Alkoxy-Rest bedeutet, synthetisiert und anschliessend auf bekannte Weise, z. B. unter schonenden Bedingungen mit Bortri-4s bromid entalkyliert
Die eingesetzten Piperazinyl-fettsäureester sowie die gegebenenfalls mit Schutzgruppen versehenen 2-Hydroxy-acetophenone sind zum Teil neue Verbindungen. Ihre Herstellung wird in den jeweiligen Beispielen beschrieben. 50 Die pharmakologisch verträglichen Salze erhält man in üblicher Weise z. B. durch Neutralisation der Verbindungen der Formel I mit nicht-toxischen anorganischen oder organischen Säuren wie z. B. Salzsäure, Schwefelsäure, Phosphorsäure, Bromwasserstoffsäure, Essigsäure, Milchsäure, Zitro-55 nensäure, Äpfelsäure, Salicylsäure, Malonsäure, Maleinsäure oder Bernsteinsäure.
Die erfindungsgemäss hergestellten neuen Substanzen der Formel I und ihre Salze können in flüssiger oder fester Form enterai oder parenteral appliziert werden. Hierbei kommen 60 alle üblichen Applikationsformen in Frage, beispielsweise Tabletten, Kapseln, Dragées, Sirupe, Lösungen, Suspensionen etc. Als Injektionsmedium kommt vorzugsweise Wasser zur Anwendung, welches die bei Injektionslösungen üblichen Zusätze wie Stabilisierungsmittel, Lösungsver-65 mittler und Puffer enthält. Derartige Zusätze sind z. B. Tar-trat- und Citrat-Puffer, Äthanol, Komplexbildner (wie Äthy-lendiamin-tetraessigsäure und deren nicht-toxische Salze), hochmolekulare Polymere (wie flüssiges Polyäthylenoxid)

zur Viskositätsregulierung. Flüssige Trägerstoffe für Injektionslösungen müssen steril sein und werden vorzugsweise in Ampullen abgefüllt. Feste Trägerstoffe sind z. B. Stärke, Lactose, Mannit, Methylcellulose, Talkum, hochdisperse Kieselsäuren, höher-molekulare Fettsäuren (wie Stearinsäure), Gelatine, Agar-Agar, Calciumphosphat, Magnesiumstearat, tierische und pflanzliche Fette, feste hochmolekulare Polymere (wie Plyäthylenglykole); für orale Applikation geeignete Zubereitungen können gewünschtenfalls Geschmacksund Süsstoffe enthalten.
Die folgenden Beispiele erläutern das erfindungsgemässe Verfahren.
Beispiel 1
2-{2-[4-(2-Methcxyphenyl)-piperazinyl-1 ]äthyl}-5-me-thoxy-4-oxo-4H-1 -benzopyran a) 2-Hydroxy-6-methoxy-acetophenon
126,3 g 2,6-Dihydroxyacetophenon (0.83 mol) werden in 900 ml Aceton gelöst, mit 229,4 g K2CO3 (1.66 mol) versetzt und nach Zugabe von 141,9 g Methyljodid (1 mol) 3 Stunden zum Sieden erhitzt. Anschliessend saugt man ab, dampft die Mutterlauge im Vakuum ein, nimmt den Rückstand in Äther auf und schüttelt die phenolischen Anteile mit 2 N Natronlauge aus. Die wässrig alkalische Schicht wird dann mit 2 N Salzsäure angesäuert und ausgeäthert. Aus der ätherischen Lösung gewinnt man durch Eindampfen 131g (95% der Theorie) praktisch reines 2-Hydroxy-6-methoxy-aceto-phenon vom Schmelzpunkt 53-54°C.
b) 3-[4-(2-Methoxyphenyl)-piperazinyl- l]-propionsäure-äthylester
Zu einer Lösung aus 30,0 g Acrylsäureäthylster (0,3 mol) in 100 ml Alkohol tropft man eine Lösung von 57,7 g 1-Metho-xyphenyl)-piperazin (0,3 mol) in 100 ml Alkohol, rührt 3 Stunden bei Raumtemperatur, dampft anschliessend ein und destilliert den Rückstand im Hochvakuum. Man erhält 75,9 g (86,5% der Theorie) (3-[4-(2-Methoxyphenyl)-piperazinyl-l]-propionsäureäthylester mit einem Siedepunkt von 163°C bei 0,01 mm.
c) 1 --(2-[4-(2-Methoxyphenyl)-piperazinyl-l]-äthyl}-3-(2-hydroxy-6-methoxy-phenyl)-propan-l,3-dion
7,2 g Natriumhydrid (0,3 mol) in Form von 50proz., handelsüblicher Dispersion werden in 100 ml trockenem Dioxan suspendiert und unter Rühren auf 80° erwärmt. Anschliessend tropft man eine Lösung von 16,6 g (0,1 mol) nach Beispiel la hergestelltem 2-Hydroxy-6-methoxy-acetophenon und 34,2 g (0,1 mol) gemäss Beispiel lb hergestelltem 3-[4-(2-Methoxyphenyl)-piperazinyl-l]-propionsäureäthylester in 75 ml Dioxan bei 80-90° zu der Natriumhydrid-Suspension zu, erhitzt 1 Stunde unter Rückfiuss und lässt dann abkühlen. Der Kolbeninhalt wird danach in 11 Ligroin eingerührt. Es fällt als Zwischenprodukt der allgemeinen Formel VI das gelblich gefärbte, amorphe Na-Salz von l-{2-[4-(2-Methoxy-phenyl )-piperazinyl-1 ]äthyl}-3-(2-hydroxy-6-methoxy-phenyI)-propan-l,3-dion aus, das mit Ligroin und Äther gewaschen wird. Die Ausbeute beträgt 36,9 g (85% der Theorie).
d) 2-{2-[4-(2-Methoxyphenyl)-piperazinyl-l]-äthyl}-5-methoxy-4-oxo-4H-1 -benzopyran
36,5 g des nach Beispiel lc hergestellten Propandions-1,3 werden in 250 ml Alkohol gelöst, man kühlt dann auf 0° ab, sättigt die Lösung mit HCl-Gas, rührt 0,5 Stunden nach und giesst die breiartige Masse auf Eis. Neutrale Verunreinigungen werden durch Ausschütteln mit Methylenchlorid entfernt. Die wässrige Phase neutralisiert man durch Zugabe von festem Natriumkarbonat und extrahiert erneut mit Methylenchlorid. Die organische Phase wird danach getrocknet, eingedampft, der Rückstand in trockenem Tetrahydrofuran aufgenommen und mit ätherischer Salzsäure versetzt. Es fällt das Hydrochlorid des 2-{2-[4-(2-Methoxyphenyl)-pipera-zinyl- l]-äthyl}-5-methoxy-4-oxo-4H-1 -benzopyrans aus, das abgesaugt und im Exsikkator getrocknet wird. Das Produkt ist chromatographisch rein, es besteht in der vorliegenden Form aus einem Gemisch des Mono- und Dihydrochlorids, Fp. 129-134°C (Zersetzung). Die Ausbeute ist 36,6 g, (ca. 75% der Theorie).
Die Struktur wird spektralanalytisch durch IR-, UV-, NMR und Massenspektren eindeutig bewiesen. UV: 316mii(MeOH), 321 mp.(pH 1), 320mp.(pH 13) NMR (DDMSO): 3-H; 6.16 ppm; Singulett
Ein Teil des vorliegenden Produkts wird mit wenig Methanol verrieben, nach anfänglicher Auflösung kristallisiert ein reines Dihydrochlorid vom Schmelzpunkt 192°C aus (1% Wassergehalt).
Aus einer Probe des Hydrochlorid-Gemisches wird mit Natriumkarbonat die Base in Freiheit gesetzt, die nach Zugabe von Äther oder Ligroin in kristallisierter Form anfällt. Schmelzpunkt 120-121 °C.
Bei verschiedenen Ansätzen werden Hydrochloride mit sehr unterschiedlichem Wassergehalt isoliert, der bis zu 12% erreichte.
Das Dihydrochlorid des nach Beispiel ld erhaltenen Produkts verliert je nach der Temperatur beim Trockenvorgang relativ leicht ein Mol Salzsäure.
Beispiel 2
2-[2-(4-Phenylpiperazinyl-l)-äthyl]-5-methoxy-4-oxo-4H-1-benzopyran a) Gemäss Beispiel lb werden 63 g Phenylpiperazin (0,39 mol) und 41,8 g Acrylsäureäthylester (0,39 mol) in 270 ml Alkohol umgesetzt. Man erhält 87,5 g (85,9%) 3-(4-Phenyl-piperazinyl-l)-propionsäureäthylester. Kp.0,01:140-145°C.
b) In Analogie zu Beispiel lc werden 3,6 g Natriumhydrid (0,15 mol), 8,3 g 2-Hydroxy-6-methoxy-acetophenon (0,05 mol/Beispiel la)und 14,4g(0,055 mol) 3-(4-Phenylpipera-zinyl-l)-propionsäure-äthylester (Beispiel 2a) in 100 ml Dioxan umgesetzt. Man isoliert l-[2-(4-Phenyl-piperazinyl-l)-äthyl]-3-(2-hydroxy-6-methoxy-phenyl)-propan-1,3-dion als Na-Salz durch Verdünnung mit Ligroin.
c) Das unter 2b genannte, noch Iigroinfeuchte Na-Salz wird in Analogie zu Beispiel ld in 70 ml Alkohol gelöst und mit 40 ml alkoholischer Salzsäure angesäuert. Man rührt 1 Stunde bei Raumtemperatur, sättigt bei 0°C mit Salzsäuregas und rührt bei 50°C nach. Der Kolbeninhalt wird auf Eis gegossen, und das Reaktionsgemisch mit Methylenchlorid extrahiert. Das gewünschte Produkt wird aus der organischen Phase in Form des Mono-hydrochlorids isoliert, eine Restmenge erhält man durch Neutralisation der wässrigen Phase. Die Produkte werden vereinigt und über eine sauer/schwach alkalische Aufarbeitung in ein Dihydrochlorid übergeführt. Man erhält 2-[2-(4-Phenylpiperazinyl-l)-äthyl]-5-methoxy-4-oxo-4H-l-benzopyran/Dihydrochlorid in einer Ausbeute von 14,6 g (61% der Theorie, 10% Wassergehalt) Schmelzpunkt 135-137°C.
Eine Probe der Substanz wird mit wässriger Natriumbikarbonatlösung in die freie Base übergeführt. Schmelzpunkt 122,5-123°C(Essigester/Ligroin).
Wie in Beispiel 1 wird der Strukturbeweis durch Spektralanalyse erbracht.
UV: 316 mn (MeOH), 321 mp. (pH 7), 322 mp. (pH 1), 321 mjj,(pH 13).

Nach der Ringschlussreaktion mit alkoholischer Salzsäure wird mit Chloroform extrahiert. Die gewünschte Verbindung wird als Hauptmenge in Form des Monohydrochlorids aus der organischen Phase isoliert. Die wässrige Phase wird anschliessend mit festem Natriumbikarbonat versetzt, die ausgefallene Rohbase mit dem Monohydrochlorid vereinigt und gemeinsam über eine sauer/schwach-basische Aufarbeitung in das Dihydrochlorid übergeführt. Man erhält 2-{2-[4-(2-Chlorphenyl)-piperazinyl-l]-äthyl}-5-methoxy-4-oxo-4H-
1-benzopyran-Dihydrochlorid mit einem Schmelzpunkt von 182°C (8% Wassergehalt). Ausbeute: 13,6 g (ca. 55% der Theorie).

Unter Verwendung von 3-[4-(2-Äthoxyphenyl)-pipera-zinyl-l]propion-säureäthylester (analog Beispiel lb, Kp.0,01: 162-164°C, Ausbeute: 94,4% der Theorie) wird wie in Beispiel 1 -3 mit 2-Hydroxy-6-methoxy-acetophenon die Verbindung
2-(2-[4-(2-Äthoxyphenyl)-6-methoxy-acetophenon die Verbindung 2-{2-[4-(2-Äthoxyphenyl)-piperazinyl- l]-äthyl}-5-methoxy-4-oxo-4H-l-benzopyran in Form eines Monohydrochlorids vom Fp. 118-120°C (9,5% Wassergehalt) in 53proz. Ausbeute erhalten.
Beispiel 5
2-{2-[4-(4-Methoxyphenyl)-piperazinyl-l]-äthyl}-5-me-thoxy-4-oxo-4H-1 -benzopyran
Unter Verwendung von 3-[4-(4-Methoxyphenyl)-pipera-zinyl-l]propion-säureäthylester (analog Beispiel lb, Kp.0,01: 170-175°C, Fp. 53°C, 93% der Theorie) wird in Beispiel 1-3 mit 2-Hydroxy-6-methoxy-acetophenon die Verbindung 2-{2-[4-(4-Methoxyphenyl)-piperazinyl-l]-äthyl}-5-methoxy-4-oxo-4H-l-benzopyran in Form eines Di-Hydrochlorids ( 13% Wassergehalt nach Gefriertrocknung der wässrigen Lösung) mit einem Schmelzpunkt von 140-143°C in 50proz. Ausbeute erhalten.
Der Wassergehalt entspricht 4 mol Kristallwasser. Die mit wässriger Bikarbonatlösung ausgefällte Base schmilzt analysenrein bei 123-124°C.
Aus einem Gemisch Tetrahydrofuran/Äther kann ein Dihydrochlorid vom Schmelzpunkt 211 °C isoliert werden.
Der spektralanalytische Strukturbeweis wird analog Beispiel 1 durchgeführt.
Beispiel 6
2-{2-[4-(3-Methoxyphenyl)-piperazinyl-l]-äthyl}-5-me-thoxy-4-oxo-4H-1 -benzopyran a) In Analogie zu Beispiel lb erhält man bei Verwendung von 50 g l-(3-Methoxyphenyl)-piperazin mit Acrylester 57,5 g (75,64% der Theorie) 3-[4-(3-Methoxyphenyl)-piperazin-l-ylj-propionsäure-äthylester, Kp.0,1-0,2:211-212°C (leicht überhitzt).
b) Chromon-Ringschluss: gemäss Beispiel 1 werden 8,3 g (0,05 mol) 2-Hydroxy-6-methoxy-acetophenon mit 16,08 g (0,055 mol) des nach Beispiel 6a hergestellten Propionsäure-esters mit 3,6 g Natriumhydrid in Dioxan umgesetzt. Die Extraktion des Reaktionsgemisches erfolgt mit Methylenchlorid. Man erhält auf diese Weise ein Hydrochlorid vom Schmelzpunkt 126°C (17,9 g Rohausbeute = 76,7% der Theorie). Wegen des hohen Wassergehaltes des Salzes kann mit ca. 75 ml Isopropanol ausgerührt werden. Die Substanz geht dabei grösstenteils in Lösung und kristallisiert nach

kurzer Zeit wieder aus. Es verbleiben 13,8 g 2-{2-[4-(3-Metho-xyphenyl)-piperazinyl-1 ]-äthyl}-5-methoxy-4-oxo-4H-1 -ben-zopyran Di-Hydrochlorid (59% der Theorie) vom Schmelzpunkt 146-148°C.
Beispiel 7
2-{2-[4-(2-Methylphenyl)-piperazinyl-l]-äthyl}-5-methoxy-4-OXO-4H-1 -benzopyran
Unter Verwendung von 3-[4-(2-Methylphenyl)-pipera-zinyl-l]-propionsäureäthylester (analog Beispiel lb, Kp.0,01: 147°C, Ausbeute: 69% der Theorie) wird nach der in den Beispielen 1-3 genannten Methode mit 2-Hydroxy-6-methoxy-acetophenon die Verbindung 2-{2-[4-(2-Methylphenyl)-pipe-razinyl- l]-äthyl)-5-methoxy-4-oxo-4H-1 -benzopyran in Form des analysenreinen Dihydrochlorids vom Fp.
161-162°C (Isopropanol) in 56proz. Ausbeute erhalten.
Die Struktur ist spektralanalytisch wie in Beispiel 1 gesichert (z. B. Massenspektrum).
Beispiel 8
2-{2-[4-(2-Methoxyphenyl)-piperazinyl-l]-äthyl}-4-oxo-4H-1-benzopyran
Nach der in den Beispielen 1-3 beschriebenen Methode werden 13,5 g (0,1 mol) handelsübliches 2-Hydroxy-aceto-phenon und 34,2 (0,11 mol) 3-[4-2-(Methoxyphenyl)-pipera-zinyl-l]-propionsäureäthylester (Beispiel lb) mit 4,8 g NaH (0,2 mol) in 100 ml Dioxan umgesetzt. Man erhält 2-{2-[4-(2-Methoxy phenyl)-piperazinyl-1 ]-äthyl}-4-oxo-4H-1 -benzopyran in Form des analysenreinen Dihydrochlorids vom Schmelzpunkt 197°C in 60,9proz. Ausbeute (26,63 g).
Die Struktur ist spektralanalytisch wie in Beispiel 1 gesichert z. B. UV (MeOH): 291 m^; 301 m|i (pH 1); 299 m|i (pH 13).
Beispiel 9
2-{2-[-(2-Methoxyphenyl)-piperazinyl-l]-äthyl}-5-me-thoxy-4-oxo-4H-1 -benzopyran a) In Analogie zu Beispiel la werden 152,15 g (1 mol) 2,4-Dihydroxy-acetophenon, 246 g (2 mol) Kaliumkarbonat und 170 g (1,2 mol) Methyljodid in 900 ml Aceton 3 h zum Sieden erhitzt. Nach gleicher Aufarbeitung erhält man 141,3 g (85% der Theorie) 2-Hydroxy-4-methoxy-acetophenon vom Schmelzpunkt 52°C.
b) Nach der in Beispiel 1 angegebenen Methode erhält man durch Umsetzung von 2-Hydroxy-4-methoxy-aceto-phenon (Beispiel 9a) mit 3-[4-(2-Methoxyphenyl)-pipera-zinyl-l]-propionsäureäthylester (Beispiel lb) die Verbindung 2-{2-[4-(2-Methoxyphenyl)-piperazinyl-1 ]-äthyl}-7-methoxy-4-oxo-4H-l-benzopyran in Form des Dihydrochlorids vom Fp. 205°C in 58proz. Ausbeute. Die freie Base schmilzt bei 154°C.
Beispiel 10
2-{2-[4-(2-Methoxyphenyl)-piperazinyl-l]-äthyl}-6-chlor-4-0X0-4H-1 -benzopyran
Nach der in Beispiel 1 beschriebenen Methode erhält man aus 17 g (0,1 mol) 2-Hydroxy-5-chlor-acetophenon (Lit. K.V. Auwers, G. Wittig, Berichte der Deutschen Chem. Gesellschaft, B 57, [1924], 1270 bis 1274) und 34,2 g (0,11 mol) 3-[4-(2-Methoxyphenyl)-piperazinyl-1 ]-propionsäureäthylester 26,9 g (62% der Theorie) der Verbindung 2-{2-[4-(2-Methoxy-phenyl)-piperazinyl-1 ]-äthyl}-6-chlor-4-oxo-4H-1 -benzopyran in Form des Monohydrochlorids vom Schmelzpunkt 205-207°C.
Wie in Beispiel 1 wird die Struktur spektralanalytisch bewiesen, z. B. UV: 311 mu (MeOH), 310 mu(pH 1), 308 mu. (pH 13).

Beispiel 11
2-{2-[4-(2-Methoxyphenyl)-piperazinyl-l]-äthyl}-6-methyl-4-oxo-4H-1 -benzopyran
Nach der in Beispiel 1 beschriebenen Methode, jedoch unter Verwendung eines Gemisches von Tetrahydrofuran/ Hexamethyl-phosphorsäuretriamid (8:2) werden bei einer Reaktionstemperatur von 60-65°C 15 g (0,1 mol) 2-Hydroxy-5-methylacetophenon und 34,2 g (0,11 mol) 3-[4-(2-Methoxy-phenyl)-piperazinyl-1 ]-propionsäureäthylester umgesetzt. Man erhält in 61proz. Ausbeute die Verbindung 2--(2-[4-(2-Methoxyphenyl)-piperazinyl-l]-äthyl)-6-methyl-4-oxo-4H-
1-benzopyran in Form des Monohydrochlorids vom Schmelzpunkt 173-175°C. Ein entsprechendes Dihydrochlorid schmilzt bei 199-201°C.
Wie üblich wird die Struktur spektralanalytisch bewiesen und die Reinheit dünnschicht-chromatographisch kontrolliert (Lösungsmittelgemisch Isopropanol/Essigsäurebutyl-ester/Wasser/konz. Ammoniak: 50/30/15/5).
Beispiel 12
2-[4-(2-Methoxyphenyl)-piperazin-1 -yl-methyl]-5-methoxy 4-OXO-4H-1 -benzopyran a) 38,4 g (0,2 mol) 2-Methoxyphenylpiperazin werden in 250 ml Tetrahydrofuran und 60,7 g (0,6 mol) Triäthylamin gelöst. Zu dieser Lösung tropft man bei Raumtemperatur 33,4 g (0,2 mol) Bromessigsäureäthylester. Unter stark exothermer Reaktion ist die Umsetzung nach 15 Minuten beendet. Man saugt danach ab, dampft das Lösungsmittel ab, destilliert den Rückstand im Hochvakuum und erhält 4-(2-Methoxyphenyl)-piperazinyl-1 -essigsäureäthylester in 81proz. Ausbeute (45,3 g), Kp.0,1:149-158°C.
b) Das nach Beispiel 12 a dargestellte Essigsäure-Derivat wird gemäss den Beispielen 1-3 umgesetzt. Man erhält aus 12,46 g 2-Hydroxy-6-methoxy-acetophenon (0,075 mol) und 22,26 g (0,08 mol) des unter 12a erhaltenen Esters 23,7 g (69,7% der Theorie) 2-[4-(2-Methoxyphenyl)-piperazin-l-yl]-methyl)-5-methoxy-4-oxo-4H-l-benzopyran in Form des Dihydrochlorids vom Schmelzpunkt 209-211 °C.
Beispiel 13
2-{2-[4-(4-Chlorbenzyl)-piperazinyl-l]-äthyl}-5-methoxy-4-0X0-4H-1 -benzopyran a) In Analogie zu Beispiel 1 a werden 30 g (0,3 mol) Acryl-säureäthylester und 63,2 g l-(4-Chlorbenzyl)-piperazin in 200 ml Alkohol umgesetzt. Man erhält 86,5 g (92,8% der Theorie) 3-[4-(4-Chlorbenzyl)-piperazin-1 -yl]-propionsäure-äthylester, Kp.0,1:150-160°C.
b) Aus 24,9 g (0,15 mol) 2-Hydroxy-6-methoxy-aceto-phenon und 51,3 g (0,16 mol) des Propionsäure-Derivats des Beispiels 13a erhält man mit 10,8 g Natriumhydrid (0,45 mol) in 260 ml Dioxan gemäss Beispiel 1 54 g (74,1% der Theorie)
2-{2-[4-(4-Chlorbenzyl)-piperazinyl-1 ]-äthyl}-5-methoxy-4-oxo-4H-l-benzopyran in Form des Dihydrochlorids vom Schmelzpunkt 247°C (Zersetzung).
Die Struktur wird durch Spektren bewiesen, unter anderem durch das Massenspektrum (gefunden 412).
Beispiel 14
2-{2-[4-(4-Chlorbenzy l)-piperazinyl-1 ]-äthyl}-5-hydroxy-4-OXO-4H-1 -benzopyran
9,8 g (0,02 mol) des in Beispiel 13 beschriebenen 5-Me-thoxy-benzopyrans werden in 200 ml Chloroform suspendiert und bei — 20°C mit einer Lösung von 10 g Bortribromid (0,04 mol) in 100 ml Chloroform versetzt. Man lässt anschliessend auf Raumtemperatur erwärmen, versetzt erneut mit 5 g Bortribromid in 25 ml Chloroform und rührt 2 Stunden nach. Überschüssiges Bortribromid wird danach durch Zugabe von Methanol zersetzt. Das Festprodukt isoliert man durch Absaugen, versetzt es mit wässriger Bikarbonatlösung und isoliert das Benzopyran durch Cloroformex-traktion. Man erhält nach Zugabe von ätherischer Salzsäure 6,2 g (65,8% der Theorie) 2-{2-[4-(4-Chlorbenzyl)-pipera-s zinyl-1 ]-äthyl}-5-hydroxy-4-oxo-4H-1 -benzopyran vom Schmelzpunkt >260 (Zersetzung).
Aus der Mutterlauge des Festprodukts werden weitere 0,8 g (8,5% der Theorie) des gewünschten Produkts erhalten.
Die Struktur wird spektralanalytisch bewiesen, die Masse 10 398 wird bestätigt, die Phenolgruppe u. a. durch das UV-Spektrum nachgewiesen. UV 330 mp. (MeOH), 368 mu (pH 13).
Beispiel 15
15 2-{2-[4-(2-Methoxyphenyl)-piperazin-1 -yl]-äthyl)-5-hydroxy-4-oxo-4H-1 -benzopyran a) 90,6 g (0,6 mol) 2,6-Dihydroxy-acetophenon werden in 200 ml 2,3-Dihydro-4-H-pyran suspendiert und mit einem Tropfen Polyphosphorsäure versetzt. Nach wenigen Minuten
20 tritt Erwärmung unter Auflösung des Ausgangsstoffes ein. Man lässt über Nacht stehen, versetzt mit Äther und schüttelt das Gemisch mit 2 N Natronlauge aus. Die ätherische Schicht wird getrocknet und eingedampft. Man erhält als Rohprodukt 72 g 2,6-Bis-(Tetrahydropyran-2-yloxy)-acetophenon 25 als zähflüssiges Öl.
Die wässrig alkalische Schicht versetzt man mit festem Kohlendioxid und äthert den gelblichen Niederschlag aus. Der Ätherextrakt wird getrocknet und eingedampft. Man erhält 110 g2-Hydroxy-6-(tetrahydropyran-2-yloxy)-aceto-30 phenon.
Sowohl das Mono- als auch das Di-(tetrahydropyran-2-yloxy)-Derivat können in der vorliegenden Form für die weitere Umsetzung vewendet werden.
b) 23,6 g (ca. 0,1 mol) des in Beispiel 15a genannten 3s2-Hydroxy-6-(tetrahydropyran-2-yloxy)-acetophenons werden mit 3-[4-(2-Methoxyphenyl)-piperazinyl-l]-propion-säureäthylester(34,16g/0,ll mol/Beispiel lb)mit7,2g(0,3 mol) Natriumhydrid gemäss Beispiel 2 in 170 ml Dioxan umgesetzt. Nach üblicher Aufarbeitung werden die ver-40 einigten Chloroform-Extrakte mit Bikarbonatlösung behandelt. Man erhält in über 90proz. Ausbeute die gewünschte Verbindung 2--(2-[4-(2-Methoxyphenyl)-piperazin-1 -yl]-äthyl}-5-hydroxy-4-oxo-4H-l-benzopyran vom Schmelzpunkt 113-115°C. Eine Probe wird in das entsprechende 45 Dihydrochlorid vom Schmelzpunkt 188-190°C (Zersetzung) übergeführt (3% Wassergehalt).
Die Struktur wird durch Spektralanalyse bewiesen, z. B. UV: 329 mu (MeOH), 329 mu (pH 1), 368 m^ (pH 13). Die Masse 380 wird durch Massenspektrum bewiesen.

Beispiel 16
2-{2-[4-(2-Methoxyphenyl)-piperazin-1 -yl]-äthyl}-5-(2,3-dihydroxy-propoxy)-4-oxo-4H-1 -benzopyran ss a) 2,2-Dimethyl-4-(2-acetyl-3-hydroxy-phenoxy-methyl)-dioxolan-1,3
11,3 g (0,05 mol) des durch Umsetzung von 2,6-Dihydroxy-acetophenon mit 1-Chlor-bzw. l-Brom-propandiol-(2,3)in Analogie zu Beispiel la hergestellten 2-Hydroxy-6-(2,3-dihy-60 droxy-propoxy)-acetophenons (Fp. 88-92°C) werden mit 20,8 g (0,2 mol) Acetondimethylacetal in 150 ml Tetrahydrofuran unter Zusatz von 20 ml dioxanischer Salzsäure gelöst. Danach dampft man partiell ein, rührt den Rückstand in wässrige Ammoniumkarbonatlösung ein, dekantiert vom 65 ausgefallenen Öl ab und nimmt das Produkt in Essigester und Äther auf. Über eine 2 N Natronlaugelösung fällt man durch Zugabe von festem Kohlendioxid 10,9 g (82% der Theorie) chromatographisch nahezu reines 2,2-Dimethyl-4-[(2'-acetyl-

3'-hydroxy-phenoxy)-methyI]-dioxolan-l,3 vom Schmelzpunkt 56-58°C.
b) Chromon-Ringschluss:
Wie in den Beispielen 1-3 beschrieben, wird das unter Beispiel 16a beschriebene Dioxolan-l,3-Derivat (10,9 g) dem Chromonringschluss mit 3-[4-(2-Methoxyhenyl)-piperazinyl-l]-propionsäureäthylester unter gleichzeitiger Abspaltung von Aceton unterworfen. Man erhält das Hydrochlorid von
2-{2-[4-(2-Methoxyhenyl)-piperazin-1 -yl]-äthyl}-5-(2,3-dihy-droxy-propoxy)-4-oxo-4H-l-benzopyran (14 g = 64,9% der Theorie).
Die Thermoanalyse zeigt einen Schmelzpunkt von 137°C bei einem Wassergehalt von 9,5%.
Die Struktur wird u. a. durch das Massenspektrum bewiesen, das nach Acetylierung den Molpeak des Diacetyl-Derivats von 538 zeigt (MG + 84).
UV: 315 mu (MeOH); 319 m\i (pH 1); 319 mp. (pH 13).
Beispiel 17
2-{2-[4-(2-Methoxybenzyl)-piperazinyl-l]-äthyl}-5-me-thoxy-4-oxo-4H-1 -benzopyran a) 1 -(2-Methoxybenzyl)-piperaziny 1-4-carbonsäureäthyl-ester
94,8 g Piperazin-N-carbonsäureäthylester werden mit 243 g 2-Methoxybenzaldehyd in 300 ml Alkohol in Gegenwart von Raney-Nickel 5 Stunden bei 50°C und 50 atm Wasserstoff-Druck katalytisch hydriert. Man reinigt über das Hydrochlorid und erhält das gewünschte Produkt als Öl, das als Rohprodukt (chromatogaphisch nahezu rein) weiterverarbeitet wird. Ausbeute: 110,2 g (66% der Theorie).
b) l-(2-Methoxybenzyl)-piperazin
Der nach Beispiel 17a erhaltene N-Carbonsäureäthylester wird mit 105 g Kaliumhydroxid in 1000 ml Alkohol 70 Stunden unter Rückfluss erhitzt. Der noch warme Kolbeninhalt wird mit konz. Salzsäure angesäuert und 30 Minuten nachgerührt. Anschliessend dampft man den Alkohol weitgehend ab, versetzt mit 10 N Natronlauge und destilliert das isolierte Öl im Pumpenvakuum. Man erhält 62,5 g (72,56% der Theorie) an reinem l-(2-Methoxybenzyl)-piperazin, Kp.o.oi: 162-168°C.
c) 3-[4-(2-Methoxybenzyl)-piperazin-1 -yl]-propionsäure-äthylester
Gemäss Beispiel lb wird das nach 17b hergestellte Pipe-razin (58 g) mit gleichmolaren Mengen Acrylester in 200 ml Alkohol umgesetzt. Man erhält den gewünschten basischen Propionsäureester in 72,6proz. Ausbeute (62,5 g), Kp.0,01: 162-168°C.
d) Chromonringschluss:
Man setzt 8,3 g (0,05 mol) 2-Hydroxy-6-methoxy-aceto-phenon (hergestellt nach Beispiel la) mit 16,85 g (0,055 mol)
3-[4-(2-Methoxybenzyl)-piperazin-l-yl]-propionsäureäthyl-ester (hergestellt nach Beispiel 17c) in insgesamt 90 ml Dioxan um. Nach der in Beispiel 1-3 erläuterten Aufarbeitung (in diesem Falle mit Methylenchlorid als Extraktionsmittel) erhält man 21 g (80%) 2-{2-[4-(2-Methoxybenzyl)-piperazinyl-l]-äthyl}-5-methoxy-4-oxo-4H-l-benzopyran in Form eines Dihydrochlorids (Wassergehalt 12,5%) vom Schmelzpunkt 134°C (Zersetzung, starkes Sintern ab 120°C).
Eine Probe der Substanz wird über die Base erneut aus Chloroform in das Dihydrochlorid übergeführt, Schmelzpunkt 21 1-213 °C (0,6% Wassergehalt).
Neben der Elementaranalyse werden wie in Beispiel 1 spektralanalytische Daten zum Strukturbeweis herangezogen, z. B.
UV: 314 m[i (MeOH), 320 mjj. (pH 7), 321 mp. (pH 1), 319 m|i (pH 13).
Beispiel 18
2-{2-[4-(2-Methoxyphenyl)-piperazinyl-l]-äthyl}-5,7-dimethyl-4-oxo-4H-1 -benzopyran
Aus 8,21 g (0,05 mol) 4,6-Dimethyl-2-hydroxy-aceto-5 phenon erhält man nach Beispiel 1-3 mit 16,08 g (0,055 mol) 3-[4-(2-Methoxyphenyl)-piperazinyl-1 -propionsäureäthyl-ester (hergestellt nach Beispiel lb) nach der Ringschlussreaktion in alkoholischer Salzsäure ein Reaktionsgemisch, das in Eiswasser eingerührt wird. Man schüttelt mit Methylen-10 chlorid aus (eine Probe zeigt, dass das gewünschte Produkt als Monohydrochlorid extrahiert wurde), säuert die organische Phase nach dem Trocknen mit Magnesiumsulfat mit ätherischer HCl an und erhält das Dihydrochlorid der Verbindung 2-{2-[4-(2-Methoxyphenyl)-piperaziny 1-1 ]-äthyl>-15 5,7-dimethyl-4-oxo-4H-l -benzopyran mit einem Schmelzpunkt von 183°C. Ausbeute der ersten Charge 13,3 g.
Die wässrig saure Phase der ersten Extraktion wird mit Bikarbonat neutralisiert, mit Methylenchlorid extrahiert und der organische Anteil nach dem Trocknen mit Natriumsulfat 20 mit ätherischer Salzsäure angesäuert. Auf diese Weise erhält man eine zweite Charge von 7,9 g mit einem Schmelzpunkt von 182-183°C, die auch analytisch mit dem 1. Anteil identisch ist.
Die Gesamtausbeute beträgt damit 21,2 g Dihydrochlorid 25 (89,5% der Theorie).
Beispiel 19
2-(2-[4-(2-Methoxyphenyl)-piperazinyl-l]-äthyl}-5-(2-hydroxy-propoxy)-4-oxo-4H-l-benzopyran a) 2-Hydroxy-6-(2-hydroxy-propoxy)-acetophenon 30 30 g 2,6-Dihydroxyacetophenon werden in 60 ml Dioxan mit 12 g 1,2-Epoxypropan nach Zugabe einiger Tropfen Triton B 48 Stunden im Glasautoklaven erhitzt (120°C Ölbadtemperatur). Anschliessend dampft man ein, nimmt den Rückstand in 2 N Natronlauge auf (durch Ätherextrak-35 tion wird disubstituiertes Produkt entfernt) und extrahiert nach Zugabe von 5 N Salzsäure mit Chloroform. Der Eindampfrückstand der organischen Phase wird anschliessend im Vakuum destilliert. Man erhält 21,4 g (51,7%) 2-Hydroxy-6-(2-hydroxy-propoxy)-acetophenon. Kp.0,01:151-155°C. 40 b) 2-Hydroxy-6-[2-(tetrahydropyran-2-yloxy)-propoxy]-acetophenon
8,4 g (0,04 mol) des nach Beispiel 19a hergestellten 2-Hydroxy-propoxy-Derivats werden mit 25 ml 2,3-Dihydro-pyran versetzt. Nach Zugabe von einem Tropfen Polyphos-45 phorsäure tritt unter Erwärmung Auflösung des Ausgangsstoffes ein. Man rührt 1 Stunde bei Raumtemperatur nach, nimmt in Äther auf und schüttelt mit wässriger Natriumkarbonatlösung aus. Der Eindampfrückstand der äherischen Schicht besteht neben wenig Ausgangsphenol zum grössten 50 Teil aus der gewünschten Mono-Tetrahydropyran-2-yloxy-Verbindung (Dünnschicht-Chromatogramm, FeCb-Detek-tion/Lösungsmittel Isopropanol/Essigsäurebutylester/ Wasser/konz. wässrigem Ammoniak = 50:30:15:5) und der Bis-substituierten Verbindung 2-(Tetrahydropyran-2-yloxy)-55 6-[2-(tetrahydropyran-2-yloxy)-propoxy]-acetophenon, die als viskoses Öl anfallen.
c) Chromonringschluss:
Eine Probe des nach Beispiel 19b mit Tetrahydropyran geschützten 2-Hydroxy-6-(2-hydroxypropoxy)-acetophenons 60 wird wie in Beispiel 1 mit3-[4-(2-Methoxyphenyl)-piperazin-1 -yl]-propionsäureäthylester mit Natriumhydrid kondensiert und anschliessend in isopropanolischer Salzsäure cyclisiert. Man erhält nach üblicher Aufarbeitung das Dihydrochlorid der gewünschten Verbindung 2-{2-[4-(2-Methoxyphenyl)-65 piperazinyl-l]-äthyl}-5-(2-hydroxy-propoxy)-4-oxo-4H-l-benzopyran vom Schmelzpunkt 166-168°C.
Die Struktur wird spektralanalytisch wie unter Beispiel 1 bestätigt.

Beispiel 20
2-{2-[4-(2-Methoxyphenyl)-piperazinyl-l]-äthyl)~5-me-thoxy-4-oxo-4H-1 -benzopyran
0,24 g (0,01 mol) Natriumhydrid werden in 25 ml Hexa-methyl-phosphorsäuretriamid (HMPT) suspendiert und s langsam mit einer Lösung von 3,8 g (0,01 mol) 2-{2-[4-(2-Me-thoxyphenyl)-piperazin-l-yl]-äthyl}-5-hydroxy-4-oxo-4H-l-benzopyran (Herstellung vgl. Beispiel 15) in 25 ml HMPT versetzt. Nach einstündigem Rühren bei Raumtemperatur lässt das Schäumen der Reaktion nach. Anschliessend ver- io setzt man tropfenweise mit 4,11 g (0,03 mol) Epibromhydrin, rührt 1 Stunde bei Raumtemperatur und 2 Stunden bei 40-50°C. Das Reaktionsgemisch wird danach in Eiswasser eingerührt und der Niederschlag abgesaugt. Man nimmt die Festsubstanz in Chloroform auf, schüttelt mit Wasser aus, is trocknet unter Zusatz von Kohle und dampft ein. Man erhält 2,4 g (55% der Theorie) 2-{2-[4-(2-Methoxyphenyl)-pipera-zinyl-1 ]-äthyl)-5-(oxiranyl-methoxy)-4-oxo-4H-1 -benzo-pyran als gelbliches Pulver. Der Schmelzpunkt ist unscharf (um 130°), da noch Spuren von HMPT vorhanden sind 20 (Nachweis im Massenspektrum).
Beispiel 21
2-{2-[4-(3-T rifluormethyl-phenyl)-piperazin-1 -yl]-äthyl}-5-methoxy-4-oxo-4H-1 -benzopyran 25
a) In Analogie zu Beispiel lb setzt man 25 g l-(3-Trifluor-methylphenyl)-piperazin (Handelsprodukt EMKA-Chemie) mit äquimolaren Mengen Acrylsäureäthylester in 80 ml Alkohol um und destilliert das Reaktionsprodukt im Pumpenvakuum. Man erhält 30,3 g (84,53% der Theorie) 30 3-[4-(3-Trifluormethyl-phenyl)-piperazinyl-l]-propionsäure-äthylester als farbloses Öl. Kp.0,1:158-159°C.
b) Chromonringschluss:
Wie in den Beispielen 1-3 beschrieben, werden 3,6 g (0,15 mol) Natriumhydrid, 8,3 g (0,05 mol) 2-Methoxy-6-hydroxy- 35 acetophenon und 18,16 g (0,055 mol) des nach Beispiel 21a erhaltenen basischen Esters in 100 ml Tetrahydrofuran umgesetzt. Nach erfolgtem Ringschluss in alkoholischer Salzsäure wird der Kolbeninhalt in Eiswasser eingerührt und mit Methylenchlorid ausgeschüttelt. Die Gesamtmenge der basischen 40 Anteile bleibt in diesem Falle in der wässrig sauren Lösung. Nach partieller Neutralisation mit Bikarbonat fällt ein gallertartiger Niederschlag aus, der in Methylenchlorid unter Zugabe von wenig Methanol gelöst werden kann. Aus der organischen Schicht erhält man durch Eindampfen ein Roh- 45 hydrochlorid, das mit wenig Isopropanol verrieben wird. Es kristallisieren 16,2 g (69,1% der Theorie) 2~(2-[4-(3-Trifluor-methyl-phenyl)-piperazin-1 -yl]-äthyl}-5-methoxy-4-oxo-4H-1-benzopyran als Monohydrochlorid vom Schmelzpunkt 206-207 °C. so
Die Struktur wird neben IR, UV und NMR-Spektren durch das Massenspektrum M+432) bestätigt.
Beispiel 22
2-{2-[4-(2-Trifluormethyl-phenyl)-piperazin-l-yl]-äthyl)- ss 5-methoxy-4-oxo-4H-1 -benzopyran
Wie in Beispiel 21 für das 3-Trifluormethyl-Derivat beschrieben, wird unter Verwendung des l-(2-Trifluor-
methyl-phenyl)-piperazins mit Acrylsäureäthylester zum basischen Propionsäure-Derivat umgesetzt (Ausbeute 76% der Theorie, Kp.0,01:160-168°C) und anschliessend bei gleicher Ansatzgrösse in 80 ml Dioxan mit 2-Methoxy-6-hydroxy-acetophenon und nachfolgendem Ringschluss in alkoholischer Salzsäure in 61proz. Ausbeute zu 2-(2-[4-(2-Trifluormethyl-phenyl)-piperazin-l-yl]-äthyl}-5-methoxy-4-oxo-4H-l-benzopyran umgesetzt. Das isolierte Hydrochlorid hat einen Schmelzpunkt von 143-146°C und besteht aus einem Gemisch aus Mono- und Dihydrochlorid der reinen Base.
Nach einmaligem Umkristallisieren aus Isopropanol erhält man das Monohydrochlorid vom Fp. 198-199°C (Zers.).
Beispiel 23
2-{2-[4-(4-Hydroxyphenyl)-piperazinyl-l]-äthyl}-5-me-thoxy-4-oxo-4H-1 -benzopyran a) Unter Stickstoffatmosphäre werden analog Beispiel lb 25 g (0,14 mol) l-(4-Hydroxyphenyl)-piperazin (Handelsprodukt EMA-Chemie) mit 15,2 ml Acrylsäureäthylester in 200 ml Alkohol umgesetzt. Nach einstündiger Reaktionszeit ist im Dünnschicht-Chromatogramm nur teilweise Umsetzung in der Suspension zu erkennen; man rührt über Nacht nach und saugt den neuen Niederschlag ab. Man erhält 34,7 g (88,93% der Theorie) 3-[4-(4-Hydroxyphenyl)-piperazinyl-l]-propion-säureäthylester vom Schmelzpunkt 118-120°C.
b) 15,3 g (0,055 mol) des Esters aus Beispiel 23a werden mit 20 ml Tetrahydrofuran nach Zugabe von einem Tropfen Polyphosphorsäure 3 Stunden auf dem Wasserbad auf 60° erwärmt. Man dampft dann teilweise ein, nimmt den Kolbeninhalt in Chloroform auf, schüttelt mit Bikarbonatlösung aus, trocknet die organische Schicht und dampft ein. Es verbleiben 17,2 g eines Produkts, das neben phenolischem Ausgangsstoff den gewünschten 3-[4-(4-Tetrahydropyran-2-yloxy-phenyl)-piperazin-1 -yl]-propionsäureäthylester enthält (DC, Chloroform: Methanol = 9:1), der in dieser Form weiter umgesetzt werden kann.
c) Chromon-Synthese:
Analog den Beispielen 1-3 setzt man den nach Beispiel 23b partiell durch den Tetrahydropyranyl-Rest geschützten Pro-pionsäureester mit 8,3 g 2-Methoxy-6-hydroxy-acetophenon (0,05 mol) in 120 ml Dioxan mit Natriumhydrid (0,15 mol) als Kondensationsmittel um und schliesst anschliessend den Chromonring mit alkoholischer Salzsäure. Das Reaktionsgemisch wird auf Eis gegossen und mit Methylenchlorid extrahiert. Die stark saure wässrige Schicht wird dann mit Natriumbikarbonat neutralisiert und erneut mit Methylenchlorid ausgeschüttet. Man erhält 12,2 g (62,17% der Theorie) 2-{2-[4-(4-Hydroxy-phenyl)-piperazinyl-l]-äthyl}-5-methoxy-4-oxo-4H-1 -benzopyran in Form der reinen Base vom Schmelzpunkt 202-204°C. Ein entsprechendes Hydrochlorid hat einen Schmelzpunkt von 196°C (ausgefällt aus Chloroform/ Isopropanol).
Die gleiche Verbindung erhält man durch Umsetzung des ungeschützten 3-[4-(4-Hydroxyphenyl)-piperazinyl-l]-pro-pionsäureäthylesters mit 2-Methoxy-6-hydroxy-acetophenon in Dioxan unter Zusatz von ca. 10% Hexamethyl-phosphor-säuretriamid bei 80° Reaktionstemperatur.
