Description
Die Erfindung betrifft ein Verfahren zur Herstellung ei- In der DE-OS 2 439 458 sind Verbindungen beschrieben,
ner neuen Klasse von Aralkylanilinen, die den Fettgehalt im die den Fettgehalt im Blut zu senken vermögen und welche Blut zu senken vermögen. die nachstehende allgemeine Formel I besitzen

Es ist jetzt eine neue Klasse von substituierten Anilinen 35 gefunden worden, von denen bestimmte Verbindungen innerhalb dieser Klasse die Wirkung besitzen, den Fettgehalt im Blut zu senken.

(b) ein Wasserstoffatom bzw. die 4-Hydroxygruppe.
(c) die 3-Methoxygruppe bzw. die 4-Hydroxygruppe oder
(d) Wasserstoffatome sind,
oder deren pharmakologisch verträgliche Säureadditionssalze.
Beispiele geeigneter Esterreste für den Rest R1 sind Alkylester, insbesondere die niederen Alkylester. Bevorzugte Esterreste R1 sind die Methyl- und Äthylester.
Bedeutet R1 einen Alkylrest, so kann er zweckmässigerweise 1 bis 10 Kohlenstoffatome aufweisen. Beispiele derartiger Alkylreste sind die Methyl-, Äthyl-, gerad- oder verzweigtkettigen Propyl-, Butyl-, Pentyl- und Hexylgruppen, welche auch an beliebiger Stelle durch eine oder mehrere Hydroxylgruppen substituiert sein können.
Zweckmässige Acylreste für den Rest R1 sind Alkanoyl-reste, insbesondere die Acetyl-, Propionyl- und Butyryl-gruppen.
Zweckmässigerweise hat q den Wert 0 oder besitzt den Zahlenwert von 1 bis 6, insbesondere von 1 bis 2.
Der Rest Alk kann zweckmässigerweise ein Alkylenrest mit 1 bis 10 Kohlenstoffatomen, insbesondere 1 bis 6 Kohlenstoffatomen, sein, beispielsweise die Methylen-, Äthylen-, Propylen- oder Butylengruppe. Vorzugsweise steht Alk für die Methylengruppe.
Beispiele zweckmässiger Reste für R2 und R3 sind Wasserstoff-, Chlor-, Brom- oder Fluoratome, ferner die Methyl-, Äthyl-, n- und Isopropyl-, n-, Iso-, sek.- und tert.-Bu-tyl-, Pentyl-, Hexyl-, Methoxy-, Äthoxy-, n- und Isoprop-oxy-, n-, Iso-, sek.- und tert.-Butoxy-, Pentyloxy- und die Hexyloxygruppe.
Vorzugsweise ist einer der Reste R2 oder R3 ein Wasserstoffatom.
Eine Verbindung, die eine möglichst grosse Wirkungskraft als Mittel zur Senkung des Fettgehalts im Blut aufweisen soll, muss den Lipoidspiegel im Serum in bedeutsamer Weise herabsetzen und keine oder nur eine geringe Wirkung auf das Wachstum, das Lebergewicht und die Le-berlipoide besitzen. Eine Klasse von Verbindungen innerhalb der allgemeinen Formel II mit einer vorteilhaften Kombination dieser Faktoren weist die allgemeine Formel III auf

in der R1 und q die bei der allgemeinen Formel II angegebenen Bedeutungen besitzen, Rs ein Wasserstoff- oder Halogenatom oder ein Alkoxyrest ist und R6 ein Wasserstoffatom oder die Hydroxygruppe bedeutet. Beispiele von speziellen Verbindungen, die unter die allgemeine Formel III fallen und bei denen q den Wert 0 besitzt, sind: 4-(4'-Chlor-benzylamino)-benzoesäure-äthylester, 4-(4'-Hexyloxy-benzylamino)-benzoesäure-äthylester, 4-(4'-Fluor-benzylamino)-benzoesäure, 4-(4'-Chlor-benzylamino)-benzoesäure, 4-(3'-Chlor-benzylamino)-benzoesäure-äthylester, 4-(2'-Chlor-benzylamino)-benzoesäure-äthylester, 4-f4'-Chlor-benzylamino)-benzoesäure-methylester, 4-(3'-Brom-benzylamino)-benzoesäure-äthylester, 4-(2'-Brom-benzylamino)-benzoesäure-äthylester, 4-(4'-Methoxy-benzylamino)-benzoesäure-äthylester, 4-(2'-Hydroxy-5'-brom-benzylamino)-benzoesäure-äthyl-ester,
4-(4'-Brom-benzylamino)-benzoesäure-äthylester, 4-(4'-Fluor-benzylamino)-benzoesäure-äthylester und 4-(4'-Fluor-benzylamino)-benzoesäure-methylester.
Beispiele von Verbindungen der allgemeinen Formel III, bei denen q einen Zahlenwert von 1 bis 6 aufweist, sind: 4-(Benzylamino)-phenylessigsäure, 4-[4'-(Benzylamino)-phenyl]-buttersäure, 4-(4'-Chlor-benzylamino)-phenylessigsäure, 3-[(4'-Chlor-benzylamino)-phenyl]-propionsäure, 3-[(4'-Chlor-benzylamino)-phenyl]-propionsäure-äthylester und
3-[(4'-Chlor-benzylamino)-phenyl]-propionsäure-methyl-ester.
Beispiele anderer Verbindungen der allgemeinen Formel II, bei denen q Zahlenwerte von 1 bis 6 aufweist, sind: 3-[(4'-Fluor-benzylamino)-phenyl]-propionsäure-äthylester, 3-[(4'-Fluor-benzylamino)-phenyl]-propionsäure-methyl-ester,
3-[(3'-Trifluormethyl-benzylamino)-phenyl]-propionsäure-äthylester,
3-[(3'-Trifluormethyl-benzylamino)-phenyl]-propionsäure-methylester,
4-(3'-Trifluormethyl-benzylamino)-phenylessigsäure-äthyl-ester und
4-(3'-Trifluormethyl-benzylamino)-phenylessigsäure-me-thylester.
Eine andere Gruppe von Verbindungen der Formel II, die in Arzneimitteln zur Senkung des Fettgehalts im Blut wertvoll sind, werden durch die allgemeine Formel IV veranschaulicht

Spezielle Verbindungen der allgemeinen Formel IV, bei denen p den Wert 1 hat, sind: 4-(4'-Methyl-benzylamino)-benzoesäure-äthylester, 4-(4'-Dimethylamino-benzylamino)-benzoesäure-äthylester, 4-(4'-Carboxy-benzylamino)-benzoesäure-äthylester, 4-(l'-Naphthylmethylamino)-benzoesäure-äthylester, 4-(3',5'-Di-tert.-butyl-4-hydroxy-benzylamino)-benzoesäure-äthylester,
4-(3',4'-Dichlor-benzylamino)-benzoesäure-äthylester, 4-(3',4'~Methylendioxy-benzylamino)-bepzoesäure-äthyl-ester,
4-(3',4,-Dimethoxy-benzylamino)-benzoesäure-äthylester, 4-(3'-Nitro-benzylamino)-benzoesäure-äthylester, 4-(3'-Nitro-4'-chlor-benzyIamino)-benzoesäure-äthylester, 4-(3'-Äthoxy-4'-methoxy-benzylamino)-benzoesäure-äthyl-ester und
4-(3'-Trifluormethyl-benzylamino)-benzoesäure-äthylester.
Eine besondere Verbindung der allgemeinen Formel IV, bei der p einen Zahlenwert von 2 bis 6 besitzt, ist 4-[2'-(4"-Chlor-phenyl)-äthylamino]-benzoesäure-äthylester.
Eine weitere Klasse von Verbindungen der Formel II, die in Arzneimitteln zur Senkung des Fettgehalts im Blut wertvoll sind, sind solche, bei denen der Rest R1 ein Alkylrest, der durch eine oder mehrere Hydroxylgruppen substituiert sein kann, ein Acylrest oder die Cyanogruppe ist. Besonders brauchbar sind solche Verbindungen, bei denen q einen Zahlenwert von 1 bis 12, insbesondere 1 bis 6, aufweist.
Spezielle Verbindungen innerhalb dieser Unterklasse, bei denen q den Wert 0 hat, sind: N-4-Acetylphenyl-N-4-chlor-benzylamin, N-4-Methylphenyl-N-4-chlor-benzylamin und N-4-Hydroxymethylphenyl-N-4-chlor-benzylamin.
Des weiteren sind vorteilhafte Verbindungen, bei denen q einen Zahlenwert von 1 bis 6 besitzt, wie: N-4-Acetylmethylphenyl-N-4-chlor-benzylamin, N-4-Acetylmethylphenyl-N-4-fluor-benzylamin, N-4-Acetylmethylphenyl-N-3-trifluormethyl-benzylamin, N-4-(2'-Acetyläthyl)-phenyl-N-4-chlor-benzylamin, N-4-(2'-Acetylathyl)-phenyl-N-4-fluor-benzylamin und N-4-(2'-Acetyläthyl)-phenyl-N-3-trifluormethyl-benzylamin.
Die obengenannten Arzneimittel können zur Verabreichung auf jedem beliebigen Weg formuliert werden, obgleich die orale Verabreichung bevorzugt wird. Die Arzneimittel können in Form von Tabletten, Kapseln, Pulvern, Granulaten, Pastillen oder flüssigen Zubereitungen, wie oralen oder sterilen parenteralen Lösungen oder Suspensionen, vorliegen.
Tabletten oder Kapseln für eine orale Verabreichung können in Form von Einzeldosierungen vorliegen und übliche Exzipientien enthalten, wie Bindemittel, beispielsweise Sirup, Akaziengummi, Gelatine, Sorbit, Traganth oder Poly-vinyl-pyrrolidon, ferner Füllstoffe, wie Lactose, Zucker, Maisstärke, Calciumphosphat, Sorbit oder Glycin, des weiteren Gleitmittel, wie Magnesiumstearat, Talcum, Poly-äthylenglykol oder Siliciumdioxid, weiterhin Zerfallhilfsmittel, wie Kartoffelstärke, oder schliesslich pharmakologisch unbedenkliche Netzmittel, wie Natrium-laurylsulfat.
Die Tabletten können nach Methoden, die in der üblichen pharmazeutischen Praxis an sich bekannt sind, überzogen sein. Orale flüssige Zubereitungen können in Form von wässrigen oder öligen Suspensionen, Lösungen, Emulsionen, Sirupen oder Elixieren vorliegen oder können als Trockenprodukte zur Rekonstituierung mit Wasser oder anderen geeigneten Vehikeln vor Gebrauch angeboten werden. Derartige flüssige Zubereitungen können übliche Zusatzstoffe, wie Suspensionsmittel, beisbielsweise Sorbit, Methylcel-lulose, Glukosesirup, Gelatine, Hydroxyäthylcellulose, Carboxymethylcellulose, Aluminiumstearatgel oder gehär4

tete Speisefette, ferner Emulgiermittel, beispielsweise Lecithin, Sorbitan-monooleat oder Akaziengummi, des weiteren nicht-wässrige Vehikel, die Speiseöle umfassen können, beispielsweise Mandelöl, fraktioniertes Kokosnussöl, ölige Ester, wie von Glycerin, Propylenglykol oder Äthanol, fer- 5 ner Konservierungsmittel, beispielsweise p-Hydroxy-benzoe-säure-methylester oder -propylester oder Sorbinsäure, sowie gegebenenfalls übliche Geschmackskorrigenzien oder Farbstoffe, enthalten.
Suppositorien enthalten übliche Suppositorien-Grund- i0 massen, wie Kakaobutter oder andere Glyceride.
Für eine parenterale Verabreichung werden flüssige Einzeldosen unter Verwendung der Verbindungen und eines sterilen Vehikels hergestellt, wobei Wasser bevorzugt wird. Je nach dem verwendeeten Vehikel oder der angewendeten Kon- 15 zentration kann die Verbindung entweder suspendiert oder dispergiert in dem Vehikel vorliegen. Bei der Herstellung von Lösungen kann die Verbindung in Wasser für Injektionszwecke gelöst und durch Filtrieren sterilisiert werden,
bevor ein Abfüllen in geeignete Fläschchen oder Ampullen 20 und ein Verschliessen erfolgt. Vorteilhafterweise können in dem Vehikel Adjuvantien, wie Lokalanästhetika, Konservierungsstoffe und/oder Puffersubstanzen, gelöst sein. Um die Stabilität zu erhöhen, können die Arzneimittel nach dem Abfüllen in Fläschchen oder Ampullen gefroren werden, 2s und das Wasser kann unter vermindertem Druck entfernt werden. Das trockene, lyophilisierte Pulver wird dann in einem Fläschchen oder einer Ampulle eingesiegelt. Parenterale Suspensionen werden praktisch in der gleichen Weise hergestellt, mit der Massgabe, dass die Verbindung in einem Vehi- 30 kel suspendiert, anstatt gelöst wird und dass die Sterilisierung nicht mit einer Filtration verbunden wird. Die Sterilisierung der Verbindung kann durch Einwirkung von Äthylenoxid vor einem Suspendieren in dem sterilen Vehikel erfolgen. Vorzugsweise wird bei den Mitteln zur Erleich- 35 terung einer gleichmässigen Verteilung der Verbindung eine grenzflächenaktive Verbindung oder ein Netzmittel mitverwendet.
Die Arzneimittel können 0,1 bis 99 Gewichtsprozent, vorzugsweise 10 bis 60 Gewichtsprozent, Aktivsubstanz je 40 nach der Verabreichungsmethode enthalten. Wenn die Arzneimittel aus Einzeldosierungen bestehen, enthält jede Dosis vorzugsweise 250 mg bis 3 g Aktivsubstanz. Bei einer Behandlung von erwachsenen Menschen liegt die angewendete Tagesdosis bei 1 bis 10 g, beispielsweise 3 g, je nach dem 45 Verabreichungsweg und der Häufigkeit der Verabreichung.
Eine bevorzugte neue Verbindung ist der 4-(4'-Chlor-benzylamino)-benzoesäure-äthylester der nachstehenden Formel

reduziert. Ein geeignetes Reagens zur Reduzierung einer Verbindung der allgemeinen Formel IX ist Borwasserstoff in Dimethylsulfid.
Weitere Verfahren zur Herstellung von Verbindungen, in denen R1 eine veresterte Carboxylgruppe ist, sind dadurch gekennzeichnet, dass man eine erhaltene Verbindung der allgemeinen Formel II, in der R1 die Carboxylgruppe oder eine in Salzform vorliegende Carboxylgruppe ist, verestert oder eine Verbindung der allgemeinen Formel II, in der R1 eine andere veresterte Carboxylgruppe ist, umestert. Die Veresterung kann nach üblichen Verfahren erfolgen, beispielsweise durch Umsetzen der freien Säure mit einem geeigneten Alkohol in Gegenwart eines Katalysators, wie einer starken Säure, wasserfreiem Chlorwasserstoff oder mit p-Toluol-sulfonsäure.
Die Bildung von Verbindungen der allgemeinen Formel II, bei denenR1 eine veresterte Carboxylgruppe ist, kann auch mittels üblicher Umesterungsverfahren durchgeführt werden, beispielsweise durch die Umsetzung eines Esters mit einem geeigneten zweiten Alkohol in Gegenwart eines Katalysators, wie dem Natriumsalz eines Alkohols oder mit wasserfreiem Chlorwasserstoff, mit p-Toluolsulfonsäure oder mit Kaliumcyanid.
Verbindungen, bei denen R1 die Carboxylgruppe ist, können auch durch eine mittels Säure oder Base katalysierte Hydrolyse einer entsprechenden Verbindung der allgemeinen Formel II hergestellt werden, in der R1 einer der nachstehenden Reste ist:
(a) die Cyanogruppe (—C = N) oder
(b) eine veresterte Carbonsäuregruppe.
Bei der Hydrolyse einer Verbindung, bei der R1 die Cyanogruppe ist, wird Ammoniak freigesetzt, und demzufolge ist ein bevorzugter Katalysator eine Säure, die Ammoniak bindet, beispielsweise eine Halogenwasserstoffsäure, wie Chlor- oder Bromwasserstoffsäure. Wenn eine basisch katalysierte Hydrolyse angewendet wird, wird ebenfalls Ammoniak freigesetzt, und die Säure wird als Alkalisalz oder nach einer Neutralisierung als freie Säure erhalten.
Bei der Hydrolyse einer veresterten Carboxylgruppe erfolgt das Verfahren vorzugsweise mittels einer Hydrolyse mittels einer starken Base, wie Natriumhydroxid. Die veresterte Carboxylgruppe R1 kann beispielsweise eine niedere Alkoxycarbonylgruppe, wie die Methoxycarbonyl- oder die tert.-Butoxycarbonylgruppe, sein. Die früheren Ausführungen hinsichtlich der Salze der erhaltenen freien Säure sind auch in diesem Falle anwendbar.
Die Verbindungen der Formel II können allein oder in Kombination mit einem oder mehreren pharmakologisch unbedenklichen Trägermaterialien und/oder Verdünnungsmitteln oder als Teil einer Gesamtnahrung verabreicht werden. In letzterem Falle kann die Menge der verwendeten

Verbindung unter 1 Gewichtsprozent und vorzugsweise nicht über 0,5 Gewichtsprozent der Nahrung ausmachen. Die Nahrung für Menschen kann aus normalen Lebensmitteln bestehen, denen ein Ester zugesetzt wird, und in gleicher Weise kann die Nahrung für Tiere aus Futtermitteln bestehen, wobei die Verbindung allein oder mit einer Vormischung zugegeben werden kann.
Die Beispiele erläutern die Erfindung.
Beispiel 1
4-[2'-(4"-Chlor-phenyl)-äthylamino]-benzoesäure-äthylester 8,91 g (0,03 Mol) N-(4-Äthoxycarbonylphenyl)-(4-chlor-phenyl)-acetamid werden unter Stickstoff in 70 ml wasserfreiem Tetrahydrofuran gelöst und mit 3,8 ml (0,03 Mol) eines Borwasserstoff-dimethylsulfid-Komplexes bei Raumtemperatur versetzt. Das Gemisch wird 30 Minuten bei Raumtemperatur gerührt, dann 4 Stunden unter Rühren unter Rückfluss zum Sieden erhitzt, gekühlt und mit 70 ml 1-n Salzsäure versetzt. Die organische Phase wird abgetrennt, und die wässrige Phase wird zweimal mit je 80 ml Dichlor-methan extrahiert. Die vereinigten organischen Phasen werden mit 100 ml Wasser gewaschen und über wasserfreiem Magnesiumsulfat getrocknet. Nach dem Abdampfen des Lösungsmittels erhält man ein Öl, das an 120 g Silikagel mit Di-chlormethan als Eluierungsmittel chromatographiert wird.
Man erhält in einer Ausbeute von 4,02 g (= 44% der Theorie) 4-[2'-(4"-Chlor-phenyl)-äthylamino]-benzoesäure-äthyl-ester vom Fp. 75 bis 77 "C
5 Beispiel 2
4-(4'-Chlor-benzylamino)-benzoesäure-isopropylester 7,0 g (0,024 Mol) 4-(4'-Chlor-benzylamino)-benzoesäure-äthylester werden zu einer Lösung von 0,5 g (0,022 Mol) Natrium in 70 ml Isopropanol gegeben. Das Gemisch wird 16 io Stunden unter Rückfluss erhitzt, dann auf Raumtemperatur abgekühlt und mit eiskaltem Wasser versetzt. Der Feststoff wird abfiltriert, getrocknet und aus Isopropanol umkristallisiert. Man erhält in einer Ausbeute von 5,38 g (= 74% der Theorie) den 4-(4'-Chlor-benzylamino)-benzoesäure-iso-15 propylester vom ungefähren Schmelzpunkt 115 °C.

Claims (3)
- 631 432PATENTANSPRÜCHE 1. Verfahren zur Herstellung von Verbindungen der FormelNH.CH -(Alk)z.in derR1 die Carboxylgruppe, die verestert oder in Form eines pharmakologisch unbedenklichen Salzes vorliegen kann, ein Alkylrest, der durch mindestens eine Hydroxylgruppe substituiert sein kann, ein Acylrest oder die Cyanogruppe ist, q den Wert 0 hat oder eine Zahl von 1 bis 12 bedeutet, Alk einen gerad- oder verzweigtkettigen Alkylenrest darstellt,x den Wert 0 oder 1 hat,R2 und R3 gleich oder verschieden sind und Wasserstoff-oder Halogenatome oder Alkyl- oder Alkoxyreste mit jeweils 1 bis 8 Kohlenstoffatomen bedeuten undR4 für ein Wasserstoff- oder Halogenatom, die Nitro-, Hydroxy-, Amino-, Phenyl- oder die Carboxylgruppe, die verestert oder in Salzform vorliegen kann, für einen Alkoxy-202530oder Halogenalkylrest mit jeweils 1 bis 8 Kohlenstoffatomen oder für einen Alkylamino- oder Acylaminorest steht, wobei auch zwei der Reste R2, R3 und R4 an benachbarten Kohlenstoffatomen den Rest eines Benzolringes oder eine Methylendioxygruppe bilden können, oder deren pharmakologisch verträgliche Säureadditionssalze, mit der Massgabe, dass, wenn q den Wert 0 hat, R1 die Carboxylgruppe oder eine veresterte Carboxylgruppe ist und R2 ein Wasserstoffatom bedeutet, dann R3 und R4 nicht(a) ein Wasserstoffatom bzw. die 2-Methoxygruppe,(b) ein Wasserstoffatom bzw. die 4-Hydroxygruppe,(c) die 3-Methoxy- bzw. die 4-Hydroxygruppe oder(d) Wasserstoffatome sind, dadurch gekennzeichnet, dass man eine Verbindung der FormelR^CH2]q(IX)c i!o- (Alk)reduziert.
- 2. Verfahren gemäss Patentanspruch 1 zur Herstellung von Verbindungen der Formel II, in denen R1 eine veresterte Carboxylgruppe ist, dadurch gekennzeichnet, dass man eine erhaltene Verbindung der Formel II, in der R1 die Carboxylgruppe oder eine in Salzform vorliegende Carboxylgruppe ist, verestert.55
- 3. Verfahren gemäss Patentanspruch 1 zur Herstellung von Verbindungen der Formel II, in denen R1 eine veresterte Carboxylgruppe ist, dadurch gekennzeichnet, dass man eine erhaltene Verbindung der Formel II, in der R1 eine andere veresterte Carboxylgruppe ist, umestert.
