Description
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen substituierten o-Phenylendiamin-derivaten.
Es ist bereits bekannt geworden, dass Phenylguanidine der allgemeinen Formel

R' sowohl niederes Alkyl als auch Wasserstoff bedeuten kann, anthelmintische Wirkungen aufweisen (s. dazu deut-5 sehe Offenlegungsschrift 2 117 293).
Sie zeigen jedoch den Nachteil, dass ihre anthelmintische Wirkung wesentlich schwächer ausgeprägt ist als bei den er-findungsgemässen substituierten o-Phenylendiamin-derivaten.
io Es wurde nun gefunden, dass die neuen erfindungsge-mäss herstellbaren substituierten o-Phenylendiaminderivate der Formel I

Als gegebenenfalls substituiertes Alkyl R und R5 steht geradkettiges oder verzweigtes Alkyl mit vorzugsweise 1 bis 6, insbesondere 1 bis 4 Kohlenstoffatomen. Beispielhaft seien gegebenenfalls substituiertes Methyl, Äthyl, n- und i.-Propyl, n.-, i.- unt t.-Butyl, genannt.
Als gegebenenfalls substituiertes Alkenyl R steht geradkettiges oder verzweigtes Alkenyl mit vorzugsweise 2 bis 6, insbesondere 2 bis 4 Kohlenstoffatomen. Beispielhaft seien gegebenenfalls substituiertes Äthenyl, Propenyl-(l), Pro-penyl-(3) und Butenyl-(3) genannt.
Als gegebenenfalls substituiertes Alkinyl R steht geradkettiges oder verzweigtes Alkinyl mit vorzugsweise 2 bis 6, insbesondere 2 bis 4 Kohlenstoffatomen. Beispielhaft seien gegebenenfalls substituiertes Äthinyl, Propinyl-(l), Propinyl-(3) und Butinyl-(3) genannt.
Als Alkyl R3 und R4 steht geradkettiges oder verzweigtes Alkyl mit vorzugsweise 1 bis 6, insbesondere 1 bis 4 Kohlenstoffatomen. Beispielhaft seien gegebenenfalls substituiertes Methyl, Äthyl, n.- und i.-Propyl, n.-, i. und t.-Bu-tyl genannt.
Als Alkoxy R5 steht geradkettiges oder verzweigtes Alkoxy mit vorzugsweise 1 bis 6, insbesondere 1 bis 4 Kohlenstoffatomen. Beispielhaft seien gegebenenfalls substituiertes Methoxy, Äthoxy, n.- und i.-Propoxy und n.-, i.- und t.-But-oxy genannt.
Als Substituenten für den gegebenenfalls substituierten Alkyl-, Alkenyl- oder Alkinylrest R seien Halogen, insbesondere Fluor, Chlor, Brom genannt, weiterhin Cyano, Alkoxy (Q-C4) und Alkylthio (Q-Q).

Als Verdünnungsmittel für die Umsetzung von Verbindungen der Formel (II) mit Isothioharnstoffen der Formel (III) kommen beispielsweise alle polaren organischen Lösungsmittel in Frage. Hierzu gehören vorzugsweise Alkohole, wie Methanol, Äthanol, iso-Propanol, sowie deren Gemische mit Wasser, Ketone, wie Aceton (auch mit Wasser gemischt), aber auch Äther wie Dioxan oder Tetrahydrofuran.
Die als reaktionsfördernde Katalysatoren zugesetzten Säuren können im Prinzip aus der Reihe der bekannten organischen oder anorganischen Säuren beliebig ausgewählt werden. Vorteilhaft verwendet man jedoch die leicht zugänglichen, technisch bedeutenden Vertreter dieser Klassen. Als Beispiel seien genannt: Salzsäure, Schwefelsäure, Salpetersäure, Ameisensäure, Essigsäure, p-Toluolsulfonsäure.
Die Reaktionstemperaturen können in einem grösseren Bereich variiert werden. Im allgemeinen arbeitet man zwischen 0 und 120 °C, vorzugsweise zwischen 0 und 80 °C. Die Umsetzung wird im allgemeinen bei Normaldruck durchgeführt.
Bei der Durchführung der Umsetzung von Verbindungen der Formel (II) mit solchen der Formel (III) werden im allgemeinen molare Mengen eingesetzt.
Die neuen Verbindungen sind oral und soweit sie ein basisches Zentrum tragen, überraschenderweise auch parenteral applizierbar, wobei die letzteren auch in Form ihrer physiologisch verträglichen Salze, wie beispielsweise Hy-drohalogenide - vorzugsweise Hydrochloride - Sulfate, Phosphate, Nitrate, Maleinate, Fumarate, Acetate, Methan-sulfonate, Naphthalindisulfonate u.a.m. angewendet werden. Das erfindungsgemässe Verfahren stellt somit eine Bereicherung der Pharmazie dar.
Die erfindungsgemäss dargestellten neuen Verbindungen zeigen eine überraschend gute und breite Wirkung gegen fol-

Die Wirkung wurde im Tierversuch nach oraler und parenteraler Applikation bei stark mit Parasiten befallenen Ver-
60 suchstieren geprüft. Die angewendeten Dosierungen wurden sehr gut von den Versuchstieren vertragen.
Die neuen erfindungsgemäss herstellbaren Wirkstoffe können als Anthelmintika sowohl in der Human- als auch in der Veterinärmedizin verwendet werden.
Die neuen Wirkstoffe können in bekannter Weise in die üblichen Formulierungen übergeführt werden.
Die neuen Verbindungen können entweder als solche oder aber in Kombination mit pharmazeutisch annehm65

baren Trägern zur Anwendung gelangen. Als Darreichungsformen in Kombination mit verschiedenen inerten Trägern kommen beispielsweise Tabletten, Kapseln, Granulate, wässrige Suspensionen, injizierbare Lösungen, Emulsionen und Suspensionen, Elixiere, Sirup, Pasten und dergleichen in Betracht. Derartige Träger umfassen feste Verdünnungsmittel oder Füllstoffe, ein steriles, wässriges Medium sowie verschiedene nicht toxische organische Lösungsmittel und dergleichen. Selbstverständlich können die für eine orale Verabreichung in Betracht kommenden Tabletten und dergleichen mit Süssstoffzusatz und ähnlichem versehen werden. Die therapeutsich wirksame Verbindung soll im vorgenannten Fall in einer Konzentration von etwa 0,5 bis 90 Gew.-% der Gesamtmischung vorhanden sein, d.h. in Mengen, die ausreichend sind, um den obengegannten Dosierungsspielraum zu erreichen.
Die Formulierungen werden in bekannter Weise hergestellt, z.B. durch Verstrecken der Wirkstoffe mit Lösungsmitteln und/oder Trägerstoffen, gegebenenfalls unter Verwendung von Emulgiermitteln und/oder Dispergiermitteln, wobei z.B. im Fall der Benutzung von Wasser als Verdünnungsmittel gegebenenfalls organische Lösungsmittel als Hilfslösungsmittel verwendet werden können.
Als Hilfsstoffe seien beispielhaft aufgeführt: Wasser,
nicht toxische organische Lösungsmittel, wie Paraffine (z.B. Erdölfraktionen), pflanzliche Öle (z.B. Erdnuss-/Sesamöl), Alkohole (z.B. Äthylalkohol, Glycerin), Glykole (z.B. Propylenglykol, Polyäthylenglykol) und Wasser; feste Trägerstoffe, wie z.B. natürliche Gesteinsmehle (z.B. Kaoline, Tonerden, Talkum, Kreide), synthetische Gesteinsmehle (z.B. hochdisperse Kieselsäure, Silikate), Zucker (z.B. Roh-, Milch- und Traubenzucker); Emulgiermittel, wie nicht iono-gene und anionische Emulgatoren (z.B. Polyoxyäthylen-Fettsäure-Ester, Polyoxyäthylen-Fettalkohol-Äther, Alkyl-sulfonate und Arylsulfonate), Dispergiermittel (z.B. Lignin, Sulfitablaugen, Methylcellulose, Stärke und Polyvinylpyrr-olidon) und Gleitmittel (z.B. Magnesiumstearat, Talkum, Stearinsäure und Natriumlaurylsulfat).
Im Falle der oralen Anwendung können Tabletten selbstverständlich ausser den genannten Trägerstoffen auch Zusätze, wie Natriumeitrat, Calciumcarbonat und Dicalcium-phosphat, zusammen mit verschiedenen Zuschlagstoffen, wie Stärke, vorzugsweise Kartoffelstärke, Gelatine und dergleichen, enthalten. Weiterhin können Gleitmittel, wie Magnesiumstearat, Natriumlaurylsulfat und Talkum, zum Tablettieren mitverwendet werden.
Im Falle wässriger Suspensionen und/oder Elixieren, die für orale Anwendungen gedacht sind, können die Wirkstoffe ausser mit den obengenannten Hilfsstoffen mit verschiedenen Geschmacksaufbesserern oder Farbstoffen versetzt werden.
Für den Fall der parenteralen Anwendung können Lösungen der Wirkstoffe unter Verwendung geeigneter flüssiger Trägermaterialien eingesetzt werden.
Die Wirkstoffe können in Kapseln, Tabletten, Pastillen, Dragees, Ampullen usw. auch in Form von Dosierungseinheiten enthalten sein, wobei jede Dosierungseinheit so ange-passt ist, dass sie eine einzelne Dosis des aktiven Bestandteils 5 liefert.
Die neuen Verbindungen können in den Formulierungen auch in Mischungen mit anderen bekannten Wirkstoffen vorliegen.
Die neuen Wirkstoffe können in üblicher Weise angelo wendet werden. Die Applikation erfolgt vorzugsweise oral, eine parenterale, insbesondere subkutane, aber auch eine dermale Applikation sind jedoch ebenfalls möglich.
Im allgemeinen hat es sich als vorteilhaft erwiesen, Mengen von etwa 0,1 bis etwa 50 mg der neuen Verbindungen je is kg Körpergewicht pro Tag zur Erzielung wirksamer Ergebnisse zu verabreichen.
Trotzdem kann es gegebenenfalls erforderlich sein, von den genannten Mengen abzuweichen, und zwar in Abhängigkeit vom Körpergewicht des Versuchstieres bzw. der Art 20 des Applikationsweges, aber auch aufgrund der Tierart und deren individuellem Verhalten gegenüber dem Medikament bzw. der Art von dessen Formulierung und dem Zeitpunkt bzw. Intervall, zu welchem die Verabreichtung erfolgt. So kann es in einigen Fällen ausreichend sein, mit weniger als 25 der vorgenannten Mindestmenge auszukommen, während in anderen Fällen die genannte obere Grenze überschritten werden muss. Im Fall der Applikation grösserer Mengen kann es empfehlenswert sein, diese in mehrere Einzelgaben über den Tag zu verteilen. Für die Applikation in der Hu-30 man- und Veterinärmedizin ist der gleiche Dosierungsspielraum vorgesehen. Sinngemäss gelten auch die weiteren obigen Ausführungen.
Die anthelmintische Wirkung der erfindungsgemäss herstellbaren neuen Wirkstoffe sei anhand der folgenden An-35 Wendungsbeispiele näher erläutert.
Beispiel A
Magen- und Darmwurm-Test / Schaf
40 Experimentell mit Haemonchus contortus oder Tricho-strongylus colubriformis infizierte Schafe wurden nach Ablauf der Präpatenzzeit der Parasiten behandelt. Die Wirkstoffmenge wurde als reiner Wirkstoff in Geltatinekapseln oral appliziert.
45 Der Wirkungsgrad wird dadurch bestimmt, dass man die mit dem Kot ausgeschiedenen Wurmeier vor und nach der Behandlung quantitativ auszählt.
Ein völliges Sistieren der Eiausscheidung nach der Behandlung bedeutet, dass die Würmer abgetrieben wurden 5o oder so geschädigt sind, dass sie keine Eier mehr produzieren können (Dosis effectiva).
Geprüfte Wirkstoffe und wirksame Dosierungen (Dosis effectiva minima) sind aus der nachfolgenden Tabelle ersichtlich.

Durch Aufarbeitung der Mutterlauge lässt sich die Aus-45 beute erhöhen.

18,3 g (0,0815 Mol) 2-Amino-4-propylthio-acetanilid werden in 350 ml Methanol und 9,5 ml Eisessig mit 18,1 g
(0,088 Mol) N,N'-bis-Methoxycarbonyl-isothioharnstoff-S-60 methyläther 15 Stunden lang bei Raumtemperatur gerührt. Nach dem Erkalten saugt man den Niederschlag ab, wäscht mit Äther und kristallisiert aus Essigester um. Man erhält 17 g N-(2-Acetylamino-5-propylthiophenyl)-N',N"-bis-methoxycarbonyl-guanidin vom Fp. 138 °c.
«s Das als Ausgangsmaterial verwendete 2-Amino-4-pro-pylthioacetanilid (Fp. 105 °c) wird durch katalytische Hydrierung von 2-Nitro-4-propylthio-acetanilid, Fp. 76 °c, erhalten. Das 2-Nitro-4-propylthio-acetanilid wird durch Um

setzung von 2-Nitro-4-thiocyanato-acetanilid mit Propyl-bromid in Gegenwart von NaBH4 in Dimethylformamid als Lösungsmittel oder durch Acetylierung von 2-Nitro-4-pro-pylthio-anilin, Fp. 41-42 °C mit Acetanhydrid oder Acetyl-chlorid hergestellt.

19,8 g N-(2-Propionylamino-5-propylthio-phenyl)-N',N"-bis-methoxycarbonyl-guanidin (Beispiel 5) werden in 650 ml Acetanhydrid gelöst. Man fügt 42 g Wasserstoffperoxid (30%ig) zu, rührt über Nacht und dampft im Vakuum ein.
Der Rückstand kristallisiert (nach Verreiben mit einem Gemisch aus Essigester und Äther). Man saugt ab und kristallisiert aus Essigester um.
Ausbeute: 22,2 g.
In entsprechender Arbeitsweise erhält man durch Oxida-tion der entsprechenden Thioäther folgende Sulfone:

30 Ausbeute: 7,37 g.
In entsprechender Arbeitsweise erhält man durch Oxi-dation der entsprechenden Thioäther folgende Sulfoxide:

- 2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass die Umsetzung in Gegenwart einer Säure erfolgt.2PATENTANSPRÜCHE 1. Verfahren zur Herstellung von neuen substituierten o-Phenylendiaminderivaten der Formel I(I)NH-Rin welcherR für gegebenenfalls substituiertes Alkyl, gegebenenfalls substituiertes Alkenyl oder gegebenenfalls substituiertes AlkinylX für O, S, SO oder S02 steht, und R1 und R2 voneinander verschieden sind und wechselseitig für einen der ResteN-COOR*bzw. -CO-R*NH-COORstehen, in welchenR3 und R4 gleich oder verschieden sind und für Alkyl stehen undR5 für Wasserstoff, gegebenenfalls substituiertes Alkyl oder Alkoxy steht,und deren physiologisch verträgliche Salze, dadurch gekennzeichnet, dass man Phenylaminoverbindungen der Formel IIR-X(II)"NH-R"'in welcherX, R und R2 die oben angegebene Bedeutung haben und in welcher X mit Stellung 4 oder Stellung 5 der substituierten 1-Amino-phenylgruppe der Formel (II) verknüpft ist mit Isothioharnstoffen der Formel III7 / r-s-c;N-COOR"(III). ^NH-COORin welcherR3 und R4 die oben angegebene Bedeutung haben undR7 für Alkyl (Ci-C^ steht, in Gegenwart eines Verdünnungsmittels umsetzt.
- 3. Verfahren nach einem der Ansprüche 1 bis 2, dadurch gekennzeichnet, dass man die entstehenden Verbindungen der Formel (I) isoliert und diese gegebenenfalls mit organischen oder anorganischen Säuren zu physiologisch verträglichen Salzen umsetzt.
