Description
Die Erfindung betrifft ein Verfahren zur Herstellung von neuen 5-(Subst. phenyl)-oxazolidinonen und deren Schwefelanaloga der allgemeinen Formel I

R3 Wasserstoff, niederes Alkyl, Alkenyl, Alkinyl, Aryl, Aralkyl oder Acyl,
R4 Wasserstoff oder niederes Alkyl und R5 niederes Alkyl, Niederalkoxycarbonyl und Wasserstoff und
X! und X2 unabhängig voneinander Sauerstoff oder Schwefel bedeuten.
Die Verbindungen der allgemeinen Formel I besitzen ein asymmetrisches Kohlenstoffatom und können daher sowohl als Racemate als auch als optische Antipoden vorliegen.
Unter niederem Alkyl werden Alkylgruppen mit bis zu 6 C-Atomen verstanden. Genannt sei zum Beispiel Methyl, Äthyl, Propyl, Isopropyl, Butyl, Isobutyl, tert.-Butyl, Pentyl, 2-Methylbutyl, 2,2-Dimethylpropyl und Hexyl. Alkenyl-und Alkinylgruppen sind z.B. 1-Propenyl, 2-Propenyl, 3-Methyl-2-propenyl, Vinyl und Propargyl. Die niederen Alkylgruppen können auch ein- oder mehrfach substituiert sein, beispielsweise durch Halogen, insbesondere Fluor, Chlor und Brom. Beispiele für halogensubstituierte Alkylgruppen sind 2-Chloräthyl, 3-Chlorpropyl, 4-Brombutyl, Difluormethyl, Trifluormethyl, l,l,2-Trifluor-2-chloräthyl, 3,3,3-Trifluorpropyl, 2,2,3,3,3-Pentafluorpropyl, 1,1,1,3,3,3-Hexafluor-2-propyl. Als Substituenten der Alkylgruppen kommen ferner in Frage Hydroxygruppen, zum Beispiel als 2-Hydroxyäthyl oder 3-Hydroxypropyl, Carboxygruppen zum Beispiel als Carboxymethyl oder Carboxyäthyl, Alk-oxygruppen, wobei jede Alkoxygruppe 1 bis 5 C-Atome enthalten kann, zum Beispiel als Äthoxymethyl, Isopropoxyme-thyl, 2-Methoxyäthyl, 2-Isopropoxyäthyl, 2-Butoxyäthyl, 2-Isobutoxyäthyl, 3-Pentoxypropyl. Die Alkoxygruppen können aber auch ringgeschlossen sein (cyclische Ätherreste), wie z. B. der 3-Tetrahydropyranylrest.
Die niederen Alkylgruppen können auch endständig substituiert sein mit Aminogruppen, bei denen der Stickstoff gegebenenfalls durch Alkylgruppen mit vorzugsweise 1 bis 5 C-Atomen mono- oder disubstituiert sein kann oder Bestandteil eines 4- bis 7gliedrigen Ringes ist. Beispiele für N-sub-stituierte Alkylgruppen sind Aminomethyl, 2-Methylamino-äthyl, 2-Dimethylaminoäthyl, 2-Diäthylaminoäthyl, 3-Di-methylaminopropyl, 3-Äthylmethylaminopropyl, Pyrrolidi-no, Piperidino, N-Methylpiperazino, Hexamethylenimino usw.
Cycloalkyl- bzw. Cycloalkyl-alkylgruppen enthalten vorzugsweise 3 bis 7 C-Atome. Bevorzugte sind die Cyclopro-pyl-, Cyclopropylmethyl-, Cyclopentyl- und Cyclohexyl-gruppen.
Als Aryl- oder Aralkylgruppen kommen insbesondere die Phenyl- und die Benzylgruppe in Frage, die gegebenenfalls in an sich bekannter Weise substituiert sein können.
Als Acyl kommen an sich alle Reste von physiologisch verträglichen Carbonsäuren in Frage. Bevorzugt sind solche, die sich von Alkanoylsäuren mit 1 bis 18 Kohlenstoffatomen, vorzugsweise 2 bis 8 Kohlenstoffatomen, ableiten, wie z.B. monobasische Alkanoylsäuren wie die Ameisensäure, Essig-, Propion-, Butter-, Isobutter-, a-Äthylbutter-, Pivalin-, Valerian-, Isovalerian-, a-Äthylvalerian-, Trime-thylessig-, 2-Methylbutter-, 3-Äthylbutter-, Capron-, Tri-äthylessig-, Önanth- oder Caprylsäure, oder cyclische Säuren, vorzugsweise cycloaliphatische Säuren, wie die Cyclo-propylidenessig-, Cyclobutylcarbon-, Cyclopentylcarbon-, Cyclopentylessig-, ß-Cyclopentylpropion-, Cyclohexyl-carbon- oder Cyclohexylessigsäure oder auch carbocyclische Aryl- oder Aralkylsäuren wie die Benzoe, 2-, 3- oder 4-Me-thylbenzoesäure.
Da der chemische Charakter der Acylgruppe für die Eigenschaften der erfindungsgemäss herstellbaren neuen Verbindungen nicht kritisch ist, solange die Acylgruppe nicht toxisch wirkt, sind auch andere aliphatische und aromatische unsubstituierte und substituierte, mono-, di- und polybasische Carbonsäuren, gesättigte und ungesättigte aliphatische, araliphatische und aromatische Carbonsäuren mit bis zu 18 Kohlenstoffatomen, vorzugsweise bis zu 8 Kohlenstoffatomen, geeignet.
Genannt seien beispielsweise die Undecyl-, Dodecan-, Tetradecan-, Hexadecan-, Octadecan-, Palmitin-, Stearin-und ß-Cyclohexylpropionsäure, 2,3-, 2,4-, 2,6-, 3,4- und 3,5-Dimethylbenzoe-, Äthylbenzoe-, Naphthoe-, 3-Methyl-a-naphthoe-, ß-Phenylpropion-, Diphenylessig- und a-Naph-thylessigsäure oder Carbaminsäuren wie die Carbamin-, Phenylcarbamin-, n-Butylcarbamin-, Dimethylcarbamin-, Diäthylcarbamin- und Allophansäure oder heterocyclische Säuren wie die ß-Furylcarbon-, Pyrrolcarbon-, ß-Pyrrolidi-nopropion-, N-Methylpyrrolidino-2-carbon-, 6-Hydroxy-in-dolyl-3-essig-, N-Methylmorpholino-2-carbon- und Pyrrol-2-carbonsäure. Die Acylreste können auch ein- oder mehrfach substituiert sein. Als Substituenten seien beispielsweise folgende Reste genannt: Hydroxy, Halo, Alkoxy, Aralkoxy, Acyloxy, Sulfonyloxy, Amido, Sulfato, Nitro, Mercapto und Cyano, wie z.B. in den Acylresten der Glykol-, Milch-, Zitronen-, Wein-, Malein-, Glycerin-, Mannon-, Glucon- und Salicylsäure oder Acylreste von Aminosäuren wie Glycin, Aminopropion-, Diglykolamino- und Triglykolaminosäure, Methylglycin, Dimethylglycin, Diäthylglycin. Genannt seien weiter beispielsweise die Acylreste der p-Aminosalicyl-, p-Aminobenzoe-, Äthylmercaptoessig-, Benzylmercaptoessig-, Chloressig-, Fluoressig-, Trichloroessig-, Trifluoroessig-, Thioglycol-, m-Nitrobenzoe-, 2,3,4-Trimethoxybenzoe-, Phenoxyessig- und a-Naphthyloxyessigsäure. Weiterhin seien beispielsweise genannt die alkoxylierten und aralkoxylier-ten Acylreste der Ameisensäure wie z.B. Carbäthoxy und Carbobenzyloxy.
Die erfindungsgemäss herstellbaren neuen Verbindungen der allgemeinen Formel I besitzen wertvolle pharmakologische Eigenschaften. Sie zeigen zentral-depressive, antidopa-minerge, antinoziceptive und antikonvulsive Wirkungen und haben damit eine gewisse Ähnlichkeit zu Neuroleptika wie Chlorpromazin oder Haloperidol. Die erfindungsgemäss herstellbaren neuen Verbindungen unterscheiden sich jedoch von den klassischen Neuroleptika durch eine andersartige Beeinflussung rezeptorabhängiger, monoaminerger Rück4

kopplungsmechanismen (Verringerung der extrapyramidalen Nebenwirkungen).
Ausserdem besitzen die erfindungsgemäss herstellbaren Verbindungen starke phosphodiesterasehemmende Eigenschaften und beeinflussen damit den Stoffwechsel cyclischer Nukleotide. Da kontrolliertes Zellwachstum entscheidend von cyclischen Nukleotiden reguliert wird (Pastan et al„ 1975) und in unkontrolliert proliferierendem Gewebe cyclische Nukleotide z.T. vermindert sind (Ryan und Heidrick, 1974, Voorhees et al., 1974), eignen sich die Verbindungen zur Behandlung hyperproliferativer und mit unkontrolliertem Zellwachstum einhergehende Erkrankungen wie der Psoriasis, der Polycythämia vera, des Neuroblastoms, immunologischen Erkrankungen, wie des Asthma bronchiale und thromboembolischer Erkrankungen, bei denen eine Störung im Stoffwechsel cyclischer Nukleotide vorliegt (Tate-son, J.E.; Trist, D.G., Inhibition of adenosine-3', 5'-cyclic monophosphate Phosphodiesterase by potential antiallergic Compounds; Life Sei. 18, 153-162; De Gaetano, G., Phar-macology of platelet aggregation; Pharmacol. Res. Commun. 7, 301-309,1975).
Aufgrund der beschriebenen Wirkungen können die erfindungsgemäss herstellbaren neuen Verbindungen in Form pharmazeutischer Präparate zur Behandlung der vorgenannten Erkrankungen angewendet werden. Die Herstellung der Präparate erfolgt mit den für die enterale oder parenterale Applikation üblichen Trägerstoffen, wie zum Beispiel Wasser, Alkohol, Gelatine, Gummi arabicum, Milchzucker, Stärke, Magnesiumstearat, Talkum, pflanzliche Öle, Polyal-kylenglykol usw. Die Präparate können in fester Form als Tabletten, Kapseln, Dragées, Suppositorien oder in flüssiger Form als Lösungen, Suspensionen oder Emulsionen vorliegen.
Das erfindungsgemässe Verfahren zur Herstellung von Verbindungen der Formel I ist dadurch gekennzeichnet, dass man 2-Amino-l (3,4-disubst. phenyl)-äthanol der allgemeinen Formel II

Ringsauerstoff und/oder Sauerstoff der 2-Carbonylgruppe gegen Schwefel austauscht, oder, falls R3 Wasserstoff bedeutet, N-acyliert oder N-alkyliert.
Die Ringschlussreaktion bei Verbindungen der Formel II kann an sich mit allen Kohlensäure- bzw. Thiokohlensäure-derivaten der allgemeinen Formel III ausgeführt werden. R kann gleich oder verschieden sein. So bedeutet es vorzugsweise niederes Alkyl oder Aralkyl wie z. B. Methyl, Äthyl und Benzyl. R kann aber auch einen Alkylenrest, wie z.B. Propylen- oder iso-Butylen, bedeuten.
Die verwendeten Lösungsmittel sind zweckmässigerweise gegenüber den Reaktanten inert. Genannt seien beispielsweise Äther wie Diäthyläther, Glykoldimethyläther oder Di-äthylenglykoldimethyläther, aliphatische und aromatische Kohlenwasserstoffe wie Hexan, Benzol, Toluol,Xylol und Mesitylen, aber auch Alkohole wie Methanol, Äthanol, Pro-panol, Butanol sowie Dimethylsulfoxid, Glykolmonome-thyläther und Diäthylenglykolmonomethyläther.
Die Reaktion wird in der Wärme ausgeführt. Die Temperatur sollte oberhalb Raumtemperatur liegen und reicht im allgemeinen bis zum Siedepunkt des Reaktionsgemisches, wobei der Temperaturbereich 60 bis 200°, vorzugsweise 100 bis 160 °C, angewendet wird.
Die Cyclisierungsreaktion wird in Gegenwart eines basischen Katalysators ausgeführt. Geeignete basische Katalysatoren sind Alkalimetall- und Erdalkalimetallcarbonate und besonders -alkoholate wie Natriummethylat und Kaliumcar-bonat. Geeignet sind aber auch organische Basen wie Pyridin, Triäthylamin, Alkalimetall- und Erdalkalimetallhydroxide wie Natriumhydroxid, insbesondere für den Fall, dass X oder Y Halogen ist. Für den Fall, dass X und Y Halogen sind, wie z.B. Chlor, kann der Zusatz eines basischen Katalysators auch entfallen. Wenn X und Y Imidazolreste sind, wird vorzugsweise bei Raumtemperatur gearbeitet, wobei als Lösungsmittel Dimethylformamid, Tetrahydrofuran, Methylenchlorid geeignet sind. Die sich gegebenenfalls anschliessende reduktive Abspaltung einer araliphatischen Äthergruppe, wie der Benzylgruppe, erfolgt an sich gewöhnlich mit allen gängigen Metallkatalysatoren, wie z.B. Platin, Palladium, Rhodium oder Nickel, sowohl in reiner Form als auch auf Trägermaterialien wie Kohle, Calciumcarbonat und Bariumsulfat. Als Lösungsmittel lassen sich alle solche verwenden, die gegenüber den Reduktionsmitteln unter den erfindungsgemässen Bedingungen inert sind. Genannt seien beispielsweise organische Säuren wie Essigsäure und Propionsäure, niedere Alkohole wie Methanol und Äthanol, Ester wie Essigester, aliphatische, cycloaliphatische und aromatische Kohlenwasserstoffe wie Hexan, Cyclohexan und Benzol.
Die sich gegebenenfalls anschliessende O-Alkylierung (R2 bedeutet Wasserstoff) erfolgt normalerweise gleichfalls nach an sich bekannten Methoden. Die Alkylierung wird vorzugsweise mit dem entsprechenden R2-halogenid, -mesy-lat oder -tosylat durchgeführt. Als Halogenide sind die Chloride, Bromide und Jodide geeignet. Zur Alkylierung wird die Hydroxyverbindung der Formel II beispielsweise in einem polaren Lösungsmittel gelöst und in Gegenwart einer Base mit dem Alkylierungsmittel auf Temperaturen zwischen 30 und 150 °C erhitzt. Als Basen sind beispielsweise Natriumhydrid, Kaliumcarbonat, Alkalialkoholate, wie Natrium-äthylat, Kaliumbutylat und Kalium-tert.-butylat, geeignet. Als Lösungsmittel kommen beispielsweise Dimethylformamid, Dimethylacetamid, Hexamethylphosphorsäuretri-amid, Acetonitril, Dimethylsulfoxid, Tetrahydrofuran, Dio-xan, Ketone, wie Aceton und Methylisobutylketon, sowie Alkohole, wie Äthanol, Butanol und tert.-Butanol, in Frage.

kann nach an sich bekannten Methoden wie z. B. nach Scheeren et al. Synthesis 1973,149 ausgeführt werden.
Für diesen Zweck ist beispielsweise ein Polysulfid wie Phosphorpentasulfid in einem Lösungsmittel oder Lösungsmittelgemisch in Gegenwart einer Base geeignet. Die Umsetzung kann auch in einer Suspension vorgenommen werden. Geeignete Lösungs- bzw. Suspensionsmittel sind zum Beispiel Acetonitril, Tetrahydrofuran, Diäthyläther, Glykol-dimethyläther und Pyridin. Als Basen sind Natriumhydro-gencarbonat, Kaliumcarbonat usw. geeignet. Die Umsetzung ist bei 30 bis 120 °C nach etwa 3 bis 24 Stunden beendet.
Die sich gegebenenfalls anschliessende N-Acylierung bzw. N-Alkylierung (R3 = H) erfolgt in der Regel ebenfalls nach an sich bekannten Methoden. So wird die Aminoverbindung vorzugsweise in einem polaren Lösungsmittel gelöst und in Gegenwart eines Salzbildners mit einem Alkyl-oder Aryl- bzw. Acylhalogenid oder Acylanhydrid auf 40 bis 150 °C erhitzt. Als polare Lösungsmittel können Dimethyl-formamid, Dimethylacetamid, Tetrahydrofuran, Dioxan, sowie Alkohole, wie Äthanol und Butanol, verwendet werden. Geeignete Salzbildner sind zum Beispiel Natriumhydrid, Kaliumcarbonat, Alkalialkoholate, wie Natrium-äthylat, Kalium-tert.-butylat usw. Die Umsetzung mit Halo-genaryl, beispielsweise Jodbenzol, kann auch ohne Lösungsmittel, vorzugsweise in Gegenwart von Kupferpulver, durchgeführt werden.
Die nachfolgenden Beispiele sollen die Erfindung erläutern.
Beispiel 1
5-(3-Benzyloxy-4-methoxy-phenyl)-2-oxazolidinon
318,6 mMol 3-Benzyloxy-4-methoxybenzaldehyd werden in 1200 ml Äther gelöst. Dazu wird eine Lösung von 558 mMol Kaliumcyanid in 300 ml Wasser bei Raumtemperatur gegeben. Nach Abkühlen auf 0 °C werden zu der gut gerührten Mischung 237 mMol 2n-Schwefelsäure getropft und das Reaktionsgemisch 12 Stunden bei Raumtemperatur gerührt. Nach Abtrennen der wässrigen Phase wird der Äther mehrmals mit 50 ml halbgesättigter Kochsalzlösung gewaschen und über Calciumchlorid scharf getrocknet. Nach Abfiltrieren vom Trockenmittel und Nachwaschen mit zweimal 100 ml Äther werden die vereinten organischen Phasen, die das rohe 2-(3-Benzyloxy-4-methoxyphenyl)-2-hydroxyaceto-nitril enthalten, zur nächsten Stufe eingesetzt.
In analoger Weise wurden aus 3,4-Dimethoxybenzal-dehyd das 2-(3,4-Dimethoxyphenyl)-2-hydroxyacetonitril und aus 3-Cyclopentyloxy-4-methoxybenzaldehyd das 2-(3-Cyclopentyloxy-4-methoxyphenyl)-2-hydroxyacetonitril hergestellt.
1,3 Mol Lithiumaluminiumhydrid werden portionsweise in 1 Liter absolutem Äther suspendiert und unter Kühlung und gutem Rühren wird die Ätherlösung von 2-(3-Benzyl-oxy-4-methoxyphenyl)-2-hydroxyacetonitril so eingetropft, dass nur ein schwacher Rückfluss auftritt. Nach Rühren bei
Raumtemperatur über Nacht wird unter Kühlung erst 400 ml Essigester und später 600 ml Wasser zugetropft. Darauf wird von dem entstehenden Niederschlag abgesaugt und der Rückstand zweimal mit je 200 ml ÄthanokWasser (1:1) 5 nachgewaschen. Die gesammelten Filtrate werden stark eingeengt und in 300 ml halbkonzentrierter Salzsäure aufgenommen. Diese Lösung wird zweimal mit je 200 ml Essigester extrahiert. Die Essigesterphase wird verworfen, und die salzsaure Lösung wird gekühlt und mit Kalilauge alkalisch io gemacht - eventuell ausfallende Aluminiumsalze werden abgesaugt - und dreimal mit je 400 ml Äther ausgeschüttelt. Die vereinigten organischen Phasen werden getrocknet, nach Filtrieren eingeengt und aus Essigester umkristallisiert.
Das in 20%iger Ausbeute erhaltene 2-Amino-l-(3-i5 benzyloxy-4-methoxyphenyl)-äthanol schmilzt bei 101 bis 102 °C.
In analoger Weise werden aus 2-(3,4-Dimethoxyphenyl)-2-hydroxyacetonitril in 49%iger Ausbeute 2-Amino-l-(3,4-dimethoxyphenyl)-äthanol (Fp. 80 bis 81 °C, Benzol) und 20 aus 2-(3-Cyclopentyloxy-4-methoxyphenyl)-2-hydroxyaceto-nitril in 70%iger Ausbeute 2-Amino-l-(3-Cyclopentyloxy-4-methoxyphenyl)-äthanol (Öl) hergestellt.
2-Amino-l-(3-benzyloxy-4-methoxyphenyl)-äthanol kann auch wie folgt hergestellt werden:
25 50 mMol 3-Benzyloxy-4-methoxybenzaldehyd und 55 mMol Trimethylalylcyanid werden zusammen mit 0,567 mMol wasserfreiem Zinkjodid 4 Stunden unter Stickstoff und Feuchtigkeitsausschluss auf 90 °C erhitzt. Es wird anschliessend in 15 ml absolutem Tetrahydrofuran aufgenom-30 men und diese Lösung zu einer Suspension von 60 mMol Lithiumaluminiumhydrid in 35 ml Tetrahydrofuran getropft. Nach einstündigem Erhitzen auf 60 °C wird, wie im Beispiel 1 beschrieben, aufgearbeitet. Nach Umkristallisa-tion aus Essigester wird 2-Amino-l-(3-benzyloxy-4-35 methoxyphenyl)-äthanol vom Schmelzpunkt 100 bis 102 °C in 30% Ausbeute isoliert.
36 mMol 2-Amino-l-(3-Benzyloxy-4-methoxyphenyl)-äthanol werden mit 50 mMol Natriummethylat und 91,4 mMol Diäthylcarbonatin 120 ml absolutem Toluol suspen-40 diert und 2 Stunden auf 110 °C Badtemperatur unter Feuchtigkeitsausschluss erhitzt, wobei Methanol und Äthanol überdestillieren. Anschliessend wird das Toluol abdestilliert, zuletzt im Ölpumpenvakuum. Der Rückstand wird in 150 ml Chloroform aufgenommen und gegen 100 ml Wasser 45 verteilt. Die Wasserphase wird zweimal mit je 150 ml Chloroform extrahiert, die vereinigten Chloroformextrakte mit 100 ml Wasser gewaschen, getrocknet, filtriert, eingeengt und aus Essigester umkristallisiert. Es wird 5-(3-Benzyloxy-4-methoxyphenyl)-2-oxazolidinon vom Schmelzpunkt 132 so bis 133 °C (91% Ausbeute) erhalten.
Beispiel 2
Analog Beispiel 1 werden aus den entsprechenden Aminoalkoholen die in der folgenden Tabelle aufgeführten 55 Oxazolidinone hergestellt, wobei R5 Wasserstoff bedeutet.

Beispiel 3
88,3 mMol 2-Amino-l-(3-benzyloxy-4-methoxyphenyl)-äthanol werden in 200 ml absolutem Tetrahydrofuran gelöst und unter Feuchtigkeitsausschluss mit 92,6 mMol Carbonyl-diimidazol versetzt. Die klare Lösung wird über Nacht bei Raumtemperatur gerührt. Nach Abdestillieren des Tetra-hydrofurans wird in 300 ml Essigester aufgenommen, 2mal mit In Salzsäure ausgeschüttelt und dann mit gesättigter Natriumchlorid-Lösung neutral gewaschen, getrocknet, filtriert und eingeengt.
Nach Umkristallisieren aus Essigester werden in 66%iger Ausbeute 5-(3-Benzyloxy-4-methoxyphenyl)-2-oxazolidinon vom Schmelzpunkt 83 bis 91 °C erhalten.
Beispiel 4
5-(3-Hydroxy-4-methoxyphenyl)-2-oxazolidinon 40,6 mMol 5-(3-Benzyloxy-4-methoxyphenyl)-2-oxazoli-dinon werden in 700 ml Äthanol gelöst und in Gegenwart von 8 g Raney-Nickel (B 113, Fa. Degussa, Frankfurt) bei Raumtemperatur unter einem Wasserstoffdruck von 100 Atm. 2,2 Stunden hydriert. Nach Abfiltrieren vom Katalysator wird eingedampft und der Rückstand aus Methanol umkristallisiert. Es werden 6,74 g 5-(3-Hydroxy-4-methoxy-phenyl)-2-oxazolidinon vom Schmelzpunkt 157° bis 160°C (Methanol) erhalten.
Beispiel 5
5-(3-Cyclopentyloxy-4-methoxyphenyl)-2-oxazolidinon 7,2 mMol 5-(3-Hydroxy-4-methoxyphenyl)-2-oxazolidi-non werden in 10 ml absolutem Dimethylformamid gelöst 30
und mit 7,9 mMol Natriumhydrid 1 Stunde bei 50 °C gerührt. Nach dem Abkühlen wird mit 8,6 mMol Cyclopentyl-bromid versetzt und anschliessend 2 Stunden bei 80 °C gerührt. Nach beendeter Reaktion wird das Dimethylform-5 amid im Vakuum bei 40 °C abgezogen. Der Rückstand wird in 100 ml 2n Natronlauge aufgenommen und dreimal mit 150 ml Chloroform extrahiert.
Die vereinigten Chloroformphasen werden mit Wasser 10 gewaschen, getrocknet, filtriert und eingeengt. Der Rückstand wird aus Essigester/Äther umkristallisiert. Man erhält in 70%iger Ausbeute 5-(3-Cyclopentyloxy-4-methoxy-phenyl)-2-oxazolidinon vom Schmelzpunkt 111 bis 112 °C.

Beispiel 6
Nach dem in Beispiel 5 angegebenen Verfahren werden aus 5-(3-Hydroxy-4-methoxyphenyl)-2-oxazolidinon und dem Halogenid oder Tosylat R2X" die in der folgenden Ta-20 belle angegebenen Verbindungen hergestellt (R3 = R4 = R5 — H).

5-(3,4-Dimethoxyphenyl)-2-oxazolidinthion und 5-(3,4-Dimethoxyphenyl)-2-thiazolidinon 45 mMol 2-Amino-l-(3,4-dimethoxyphenyl)-äthanol werden in 65 ml absolutem Dimethylsulfoxid gelöst und nacheinander mit 1,8 g pulverisiertem Kaliumhydroxid und
1,4 ml Schwefelkohlenstoff bei 10 °C versetzt. Anschliessend wird das Reaktionsgemisch 2,5 Stunden unter Feuchtigkeits-65 ausschluss gerührt. Nach Abziehen des Dimethylsulfoxids unter Vakuum wird der Rückstand mit 100 ml Wasser versetzt und dreimal mit 100 ml Chloroform extrahiert. Die vereinigten Chloroformphasen werden über wasserfreiem

Magnesiumsulfat getrocknet, filtriert und eingeengt. Der Rückstand wird über 125 g Kieselgel mit ChlorofornrMe-thanol (96:4) chromatographiert. Nach Umkristallisation der entsprechenden Fraktionen aus Methanol werden in 9% Ausbeute 5-(3,4-Dimethoxyphenyl)-2-oxazolidinthion vom Schmelzpunkt 177 bis 178 °C neben 6% 5-(3,4-Dimethoxy-phenyl)-2-thiazolidinon vom Schmelzpunkt 167 bis 169 °C erhalten.
Beispiel 8
5-(3,4-Dimethoxyphenyl)-2-thiazolidinon
10 mMol 2-Amino-l-(3,4-dimethoxyphenyl)-äthanol werden in 6 ml Pyridin gelöst und unter Kühlung auf 0 °C mit 11 mMol Schwefelkohlenstoff in 10 mMol Triäthylamin tropfenweise versetzt, wobei die Temperatur stark ansteigt. Es wird 1 Stunde bei 0 °C gerührt und anschliessend bei der gleichen Temperatur 10 mMol Benzylchlorid zugetropft. Nach Rühren über Nacht bei 0 °C wird der Ansatz in 40 ml 3n Schwefelsäure aufgenommen und dreimal mit je 50 ml Chloroform extrahiert. Die vereinigten Chloroformphasen werden nacheinander mit Natriumhydrogencarbonat und Wasser gewaschen, getrocknet, filtriert und eingeengt. Der spontan kristallisierende Rückstand wird aus Benzol und Essigester umkristallisiert. Es wird in quantitaver Ausbeute [2-(3,4-DimethoxyphenyI)-2-hydroxyäthyl]-dithiocarbamin-säure-benzylester vom Schmelzpunkt 130 bis 132 °C erhalten.
Zu 6,07 mMol [2-(3,4-Dimethoxyphenyl)-2-hydroxy-äthyl]-dithiocarbaminsäure-benzylester in 50 ml absolutem Äther werden bei +5 °C 2,14 mMol Phosphortribromid zugetropft. Nach 20stündigem Rühren des Ansatzes bei 5 °C wird unter Kühlung 10 ml Methanol/Wasser so zugetropft, dass die Temperatur nicht über 20 °C steigt. Anschliessend wird das Reaktionsgemisch vorsichtig in 20 ml Natrium-hydrogencarbonatlösung eingerührt. Es wird dreimal mit je 50 ml Chloroform extrahiert und die vereinigten organischen Phasen nacheinander mit 50 ml Natriumhydrogencarbonat-lösung und 50 ml Wasser gewaschen, getrocknet, filtriert und eingeengt.
Es wird 2-Benzylthio-5-(3,4-dimethoxyphenyl-l,3-thi-azol-2-in als Öl in 80% Ausbeute erhalten.
Dieses Öl wird in 20 ml Äthanol gelöst, mit 20 ml 6n Salzsäure versetzt und 4 Stunden am Rückfluss gekocht. Nach Einengen wird der Rückstand in 25 ml Äthanol aufgenommen, mit 19 ml einer In Natronlauge versetzt und 3 Stunden bei Raumtemperatur gerührt. Darauf wird das Reaktionsgemisch mit 2n-Salzsäure neutralisiert, das Äthanol abgezogen und dreimal mit je 100 ml Chloroform ausgeschüttelt. Die Chloroformphase wird mit Wasser gewaschen, getrocknet und eingeengt. Der Rückstand wird über 50 g Kieselgel mit dem Laufmittel Chloroform/Methanol (96:4) chromatographiert. Die entsprechenden Fraktionen werden aus Methanol umkristallisiert, wobei 50% 5-(3,4-Di-methoxyphenyl)-2-thiazolidinon vom Schmelzpunkt 167 bis 169 °C erhalten werden.
Beispiel 9
5-(3,4-Dimethoxyphenyl)-2-oxazolidinon-3-carbonsäureani-
lid
0,5 mMol 5-(3,4-Dimethoxyphenyl)-2-oxazolidinon werden in 3 ml Phenylisocyanat 3 Stunden auf 160 °C erhitzt. Nach dem Abkühlen wird das Reaktionsgemisch im Vakuum eingeengt und über 50 g Kieselgel mit Cyclohexan/ Essigester (1:1) chromatographiert. Nach Umkristallisation der entsprechenden Fraktionen aus Essigester erhält man in 60%iger Ausbeute 5-(3,4-Dimethoxyphenyl)-2-oxazolidi-non-3-carbonsäureanilid vom Schmelzpunkt 175 bis 182 °C.
Beispiel 10
100 mMol 2-Amino-l-(3,4-dimethoxyphenyl)-äthanol werden in 100 ml Chloroform gelöst. Nach Kühlen auf 0 °C wird eine Lösung von 100 mMol Phosgen in 100 ml Chloro-5 form langsam so zugetropft, dass die Innentemperatur nicht über 5 °C steigt. Nach 1 Stunde Rühren bei 5 bis 10 °C werden 200 mMol Pyridin in 100 ml Chloroform zugetropft und 3 Stunden gerührt. Nach Waschen der organischen Phase mit halbgesättigter Kochsalzlösung wird eingedampft und io aus Methanol umkristallisiert. Man erhält in 75%iger Ausbeute 5-(3,4-Dimethoxyphenyl)-2-oxazolidinon vom Schmelzpunkt 114 bis 117 °C.
Beispiel 11
15 5-(3,4-Dimethoxyphenyl)-2-oxazolidinon
5 mMol 2-Amino-l-(3,4-Dimethoxyphenyl)-äthanol werden in 5 ml 2n-Natronlauge gelöst. Die auf 0 °C gekühlte Lösung wird tropfenweise mit 10 mMol Chlorameisensäureäthylester versetzt. Nach beendeter Zugabe wird 30 Minuten 20 bei 0 °C gerührt. Die gebildeten Kristalle werden abgesaugt mit Wasser gewaschen und getrocknet. Es werden 86% Ausbeute 2-(3,4-Dimethoxyphenyl)-2-hydroxyäthyl-carbamin-säureäthylester vom Schmelzpunkt 90 bis 92 °C erhalten. 3 mMol 2-(3,4-Dimethoxyphenyl)-2-hydroxyäthyl-25 carbaminsäureäthylester werden mit 4 mMol Natriumme-thylat in 12 ml Toluol suspendiert und 2 Stunden auf 110 °C Badtemperatur unter Feuchtigkeitsausschluss erhitzt. Dabei destillieren Methanol und Äthanol ab. Anschliessend wird das Toluol im Vakuum abgezogen, der Rückstand in 50 ml 30 Chloroform aufgenommen und gegen 10 ml Wasser verteilt. Nach Trocknen, Filtrieren und Einengen der organischen Phase wird aus Methanol umkristallisiert. Es wird in 80%iger Ausbeute 5-(3,4-Dimethoxyphenyl)-2-oxazolidinon vom Schmelzpunkt 114 bis 117 °C erhalten.

Beispiel 12
3-Acetyl-5-(3,4-dimethoxyphenyl)-2-oxazolidinon 8,9 mMol 5-(3,4-Dimethoxyphenyl)-2-oxazolidinon werden mit 10 ml Essigsäureanhydrid und 5 ml Pyridin versetzt 40 und 5 Stunden auf 100 °C erhitzt. Nach Einengen wird aus Essigester umkristallisiert. Man erhält 3-Acetyl-5-(3,4-di-methoxyphenyl)-2-oxazolidinon in 73%iger Ausbeute vom Schmelzpunkt 175 bis 182 °C.
45 Beispiel 13
5-(3,4-Dimethoxyphenyl)-5-propyl-2-oxazolidinon 56 mMol 3,4-Dimethoxybutyrophenon und 61,4 mMol Trimethylsilylcyanid unter Zusatz von 20Ô mg (627 mMol) wasserfreiem Zinkjodid werden 4 Stunden unter Stickstoff so und Feuchtigkeitsausschluss auf 90 °C erhitzt. Das Reaktionsgemisch wird in 15 ml absolutem Tetrahydrofuran aufgenommen und zu einer Suspension von 60 mMol Lithiumaluminiumhydrid in 35 ml absolutem Tetrahydrofuran zugetropft, eine Stunde unter Stickstoff auf 60 °C er-55 wärmt und der Überschuss an Lithiumaluminiumhydrid durch Zutropfen von Essigester und Wasser zerstört. Der Niederschlag wird über Kieselgel abgesaugt, gut mit Äthanol nachgewaschen und die Mutterlauge eingeengt. Der Rückstand wird in Essigester aufgenommen, mit 4n 6o Salzsäure angesäuert und ausgeschüttelt. Die wässrige Phase wird mit Natriumcarbonat auf pH 9 eingestellt, mit Kochsalz gesättigt und mit Chloroform extrahiert. Die Chloroform-Phase wird mit gesättigter Kochsalzlösung neutral gewaschen und das Lösungsmittel nach Trocknen über 65 Natriumsulfat abdestilliért. Man erhält 5-Amino-4-(3,4-di-methoxyphenyl)-4-pentanol in 75,5%iger Ausbeute als Öl. Eine Analysenprobe als Hydrochlorid schmilzt bei 175 bis 176 °C.
17,35 mMol 5-Amino-4-(3,4-dimethoxyphenyl)-4-pen-tanol werden in 10 ml absolutem Dimethylformamid gelöst und mit 18,3 mMol 98%igem Carbonyldiimidazol in 100 ml absolutem Tetrahydrofuran 4 Tage bei Raumtemperatur unter Feuchtigkeitsausschluss umgesetzt. Nach Abdestillieren des Lösungsmittels wird der Rückstand in Essigester aufgenommen, zweimal mit In Salzsäure ausgeschüttelt und mit gesättigter Kochsalzlösung neutral gewaschen. Das erhaltene öl wird über eine Kieselgelsäule (350 g) im System Chloroform/Methanol 30:1 gereinigt. Es wird in 81,7%iger Ausbeute 5-(3,4-Dimethoxyphenyl)-5-propyl-2-oxazolidinon als öl erhalten.
Beispiel 14
5-(3,4-Dimethoxyphenyl)-5-methyl-2-oxazolidinon Analog Beispiel 13 wird aus 3,4-Dimethoxyacetophenon das 5-(3,4-Dimethoxyphenyl)-5-methyl-2-oxazolidinon in 30%iger Gesamtausbeute mit dem Schmelzpunkt 98 bis 101 °C erhalten.
Beispiel 15
10 mMol 5-(3,4-Dimethoxyphenyl)-2-oxazoIidinon werden in 20 ml absolutem Dimethylformamid gelöst und mit 11 mMol Natriumhydrid versetzt. Anschliessend wird 40 Minuten bei 40 °C gerührt. Nach Abkühlen werden 20 mMol Methyljodid in 5 ml Dimethylformamid zugetropft und nach beendeter Zugabe 6 Stunden bei 50 °C gerührt. Nach Abziehen des Dimethylformamids wird in Chloroform aufgenommen, erst mit wenig Wasser und dann mit gesättigter Kochsalzlösung gewaschen, getrocknet, filtriert und eingeengt. Der Rückstand wird über 60 g Kieselgel mit Chloroform/Methanol (96:4) als Laufmittel chromatographiert. Nach Umkristallisation aus Äthanol erhält man in 60%iger Ausbeute 5-(3,4-Dimethoxyphenyl)-3-methyl-2-oxazolidi-non vom Schmelzpunkt 132 bis 133 °C.

Beispiel 16
5-(3,4-Dimethoxyphenyl)-2-oxazolidinon-3-carbonsäure-benzylester 6,25 mMol 5-(3,4-Dimethoxyphenyl)-2-oxazolidinon s werden in 30 ml Dioxan gelöst und mit 6,25 ml (12,50 mM) einer 2 N-Natronlauge versetzt. Bei +4 °C wird der Ansatz mit 12,5 ml Carbobenzoxychlorid versetzt und 4 Stunden bei dieser Temperatur gerührt.
Nach Versetzen mit 50 ml 2 N-Natronlauge wird 3mal
10 mit je 50 ml Essigester extrahiert. Die Essigesterphase wird lmal mit 50 ml 2 N-Natronlauge und 2mal mit je 50 ml Wasser gewaschen, über Sikkon® getrocknet, filtriert und eingeengt. Nach Abziehen des noch enthaltenen Carbobenzoxy-chlorids bei 100 °C und 1 Torr wird der Rückstand aus
15 Essigester/Petroläther umkristallisiert und ergibt in 64%iger Ausbeute 5-(3,4-Dimethoxyphenyl)-2-oxazolidinon-l-carbonsäurebenzylester vom Schmelzpunkt 124 bis 126 °C.
Beispiel 17
20 5-(3,4-Dimethoxyphenyl)-2-oxazolidinon-3-
carbonsäureäthylester 5 mMol 5-(3,4-Dimethoxyphenyl)-oxazolidinon werden in 20 ml Toluol (abs.) mit 10 mMol Triäthylamin bei 0 °C unter Feuchtigkeitsausschluss vorgelegt. Der Ansatz wird 10 25 mMol Chlorameisensäureäthylester versetzt, 4 Stunden am Rückfluss gekocht und über Nacht stehengelassen. Das auskristallisierte Produkt wird nach Absaugen nochmals mit 7,6 mMol Triäthylamin und 7,6 mMol Chlorameisensäureäthylester gekocht. Das auskristallisierte Produkt wird wieder ab-30 gesaugt; die vereinigten Filtrate werden eingedampft. Die Chromatographie des Rückstandes über 80 g Kieselgel mit Cyclohexan-Essigester 1:1 als Elutionsmittel ergibt in 43%iger Ausbeute 5-(3,4-Dimethoxyphenyl)-2-oxazolidi-non-1-carbonsäureäthylester vom Schmelzpunkt 181 bis 35 185 °C.

5-(3,4-Dimethoxyphenyl)-4-methyl-2-oxazolidinon 5,5 mMol 2-Amino-l-(3,4-dimethoxyphenyl)-propanol werden in 50 ml Chloroform gelöst und mit 1,05 g (6,5 mMol) Carbonyldiimidazol 2 Stunden unter Feuchtigkeitsausschluss gerührt. Nach Stehenlassen über Nacht wird gegen 50 ml destilliertes Wasser extrahiert, getrocknet, filtriert und eingeengt. Nach Chromatographie des Rückstandes über 50 g Kieselgel mit Chloroform/Methanol (95:5) als Laufmittel und Umkristallisation aus Essigester/Petroläther erhält man in 24%iger Ausbeute 5-(3,4-Dimethoxyphenyl)-4-methyl-2-oxazolidinon vom Schmelzpunkt 98 bis 99 °C.
Das Ausgangsmaterial 2-Amino-l-(3,4-dimethoxy)-pro-panol wurde wie folgt hergestellt:
Zu 100 mM 3,4-Dimethoxypropiophenon in 160 ml 60 Methylenchlorid werden 10 ml Sulfurylchlorid in 80 ml Methylenchlorid tropfenweise bei Raumtemperatur hinzugefügt. Anschliessend wird bei Raumtemperatur 3,5 Stunden weitergerührt. Nach Abziehen des Lösungsmittels und Umkristallisation des Rückstandes aus Cyclohexan/Petroläther 65 gewinnt man in 89%iger Ausbeute 2'-Chlor-3,4-dimethoxy-propiophenon vom Schmelzpunkt 56 bis 57 °C.
Je 43,86 mM 2'-Chlor-3,4-dimethoxypropiophenon in 230 ml Aceton gelöst, mit 437 mg Kaliumjodid und 87,72

mM Dibenzylamin werden unter Feuchtigkeitsausschluss 7 Tage gerührt. Der Ansatz wird auf 11 Lösung mit Diäthyl-äther verdünnt, filtriert und eingeengt. Der Rückstand wird auf Kieselgel aufgezogen und das Kieselgel zunächst mit Chloroform und anschliessend mit Äthanol ausgerührt und jeweils abgesaugt. Die Chloroformphase wird über 500 g Kieselgel mit Chloroform als Laufmittel chromatographiert und ergibt in 49%iger Ausbeute 2'-N,N-Dibenzylamino-3,4-dimethoxypropiophenon als Öl.
12,39 g (31,8 mMol) 2'-N,N-Dibenzylamino-3,4-dimethoxypropiophenon werden in 75 ml Isopropanol gelöst und mit 1,33 g (35,05 mMol) Natriumboranat versetzt. Nach 1 Stunde Rühren bei Raumtemperatur wird 2 Stunden am Rückfluss erhitzt. Nach Erkalten wird der Ansatz abgesaugt und der Rückstand mit Essigester ausgekocht. Nach Filtrieren kristallisiert aus dem Filtrat in 59%iger Ausbeute 2'-(N,N-Dibenzylamino)-1 -(3,4-dimethoxyphenyl)-propanol vom Schmelzpunkt 151 bis 152 °C aus.
13,2 mMol der Dibenzylverbindung werden in 50 ml Äthanol p. a. mit 2,64 g Palladium/Kohle (10%) 3 Stunden bei 90 °C unter 10 atü Wasserstoffatmosphäre hydriert.
Nach Absaugen wird eingeengt und aus Äthanol umkristallisiert. Das ergibt in 64%iger Ausbeute 2-Amino-l-(3,4-di-methoxyphenyl)-propanol vom Schmelzpunkt 131 bis 132 °C.
Beispiel 20
5-(3,4-Dimethoxyphenyl)-2-oxo-oxazolidin-4-carbonsäureäthylester 6 mMol 2-Amino-4-(3,4-dimethoxyphenyl)-3-hydroxy-propionsäureäthylester werden mit 12 mM Carbonyldiimidazol in 60 ml Chloroform für 4 Stunden bei Raumtempera-

tur unter Feuchtigkeitsausschluss gerührt. Anschliessend wird lmal gegen 50 ml Wasser extrahiert, über Nacht über Silicagel getrocknet, filtriert und eingeengt.
Der Rückstand wird über 130 g Kieselgel mit Chloro-s form/Methanol (90:10) als Elutionsmittel chromatographiert. Die Umkristallisation aus Essigester/Äthanol liefert in 34%iger Ausbeute 5-(3,4-Dimethoxyphenyl)-2-oxooxazo-lidin-4-carbonsäureäthylester vom Schmelzpunkt 188 bis 189 °C.
io Das Ausgangsmaterial 2-Amino-4-(3,4-dimethoxy-phenyl)-3-hydroxy-propionsäure-äthylester wurde wie folgt hergestellt:
44,37 mMol 3,4-Dimethoxyglycidsäureäthylester (hergestellt nach W. Schneider et al., Arch. Pharm. 299, 817 ls (1966)) werden mit 97,61 mM Dibenzylamin in 100 ml Äthanol für 4,5 Stunden unter Feuchtigkeitsausschluss am Rückfluss gekocht. Nach Stehenlassen über 3 Tage bei Raumtemperatur wird der Ansatz eingeengt und der Rückstand über 400 g Kieselgel mit Chloroform/Methanol (98:2) als 20 Elutionsmittel chromatographiert. Nochmalige Chromatographie der entsprechend zusammengefassten Fraktionen ergibt 2-N,N-Dibenzyl-4-(3,4-dimethoxyphenyl)-3-hydroxy-propionsäure-äthylester als Öl in 89%iger Ausbeute.
22,2 mMol 2-N,N-Dibenzylamino-4-(3,4-dimethoxy-25 phenyl)-3-hydroxypropionsäureäthylester werden mit 4,44 g Palladium/Kohle 10%ig in 100 ml unvergälltem Äthanol 3 Stunden bei 90 °C unter 10 atü Wasserstoffdruck hydriert. Nach Absaugen vom Katalysator wird eingeengt. Umkristallisation aus unvergälltem Äthanol ergibt in 80%iger Aus-30 beute 2-Amino-4-(3,4-dimethoxyphenyl)-3-hydroxypropion-säureäthylester vom Schmelzpunkt 211 bis 212°C (Zersetzung).

- 635 578PATENTANSPRÜCHE 1. Verfahren zur Herstellung von neuen 5-(subst. Phe-nyl)-oxyzolidinonen und deren Schwefelanaloga der allgemeinen Formel I
- X.worinR-! niederes Alkyl oder Cycloalkyl,R2 Wasserstoff, gegebenenfalls substituiertes niederes Alkyl, Cycloalkyl, Aryl, Aralkyl, Alkenyl, heterocyclische Reste mit einem Heteroatom, wobei dieses Heteroatom Sauerstoff oder Stickstoff ist, Alkinyl und Cycloalkyl-alkyl,R3 Wasserstoff, niederes Alkyl, Alkenyl, Alkinyl, Aryl, Aralkyl oder Acyl,R4 Wasserstoff oder niederes Alkyl und Rs niederes Alkyl, Niederalkoxycarbonyl und Wasserstoff undXj und X2 unabhängig voneinander Sauerstoff oder Schwefel bedeuten, dadurch gekennzeichnet, dass man 2-Amino-l-(3.4-disubst. phenyl)-äthanole der allgemeinenFormel IIORworinRj_5 die oben angegebene Bedeutung haben, mit einem Kohlensäure- oder Thiokohlensäurederivat der allgemeinen Formel IIIX=C (III),XYworinX für Sauerstoff oder Schwefel,X' und Y für OR, Chlor, Brom oder Imidazol, mit R in der Bedeutung von Alkyl, Aryl, Aralkyl und gemeinsam als Alkylen, stehen oder falls X Schwefel bedeutet, X' und Y auch gemeinsam Schwefel bedeuten können, in einem inerten Lösungsmittel in der Wärme in Gegenwart eines basischen Katalysators umsetzt.
- 2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man eine anwesende araliphatische Äthergruppe reduk-tiv mit Raney-Nickel oder mit einem Edelmetallkatalysator abspaltet.
- 3. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man eine freie Hydroxygruppe alkyliert.
- 4. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man den Ringsauerstoff und/oder den Sauerstoff der 2-Carbonylgruppe gegen Schwefel austauscht.
- 5. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man, falls R3 Wasserstoff bedeutet, N-acyliert.
- 6. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man, falls R3 Wasserstoff bedeutet, N-alkyliert.
- 7. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass man 5-(3-Benzyloxy-4-methoxy-phe-nyl)-2-oxazilidinon herstellt.
- 8. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass man 5-(3,4-Dimethoxyphenyl)-2-ox-azolidinon herstellt.
- 9. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass man 3-Methyl-5-(3,4-dimethoxyphe-nyl)-2-oxazolidinon herstellt.
- 10. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass man 5-(3-Methoxy-4-cyclo-pentyl-oxyphenyl)-2-oxazolidinon herstellt.
- 11. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass man 5-(3-Cyclopentyloxy-4-methoxyphenyl)-2-oxazolidinon herstellt.
- 12. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass man (3-Propyloxy-4-methoxy-phenyl)-2-oxazolidinon herstellt.
- 13. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass man 5-(3-Isobutyloxy-4-methoxyphenyl)-2-oxazolidinon herstellt.
- 14. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass man 5-(3-Cyclobutyloxy-4-methoxyphenyl)-2-oxazolidinon herstellt.
- 15. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass man 5-(3-n-Butoxy-4-methoxy-phenyl)-2-oxazolidinon herstellt.
- 16. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass man 5-(3-Allyloxy-4-methoxy-phenyl)-2-oxazolidinon herstellt.
- 17. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass man 5-(3-Propargyloxy-4-methoxyphenyl)-2-oxazolidinon herstellt.
- 18. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass man 5-(3-Methylallyloxy-4-methoxyphenyl)-2-oxazolidinon herstellt.
- 19. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass man 5-(3-Phenyloxy-4-methoxy-phenyl)-2-oxazolidinon herstellt.
- 20. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass man 5-(3,4-Dimethoxyphenyl)-2-oxazolidinthion herstellt.
- 21. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass man 5-(3,4-Dimethoxyphenyl)-2-thiazolidinon herstellt.
- 22. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass man 5-(3,4-Dimethoxyphenyl)-5-methyl-2-oxazolidinon herstellt.
- 23. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass man 5-(3,4-Dimethoxyphenyl)-5-propyl-2-oxazolidinon herstellt.
- 24. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass man 5-(3,4-Dimethoxyphenyl)-2-oxazolidinon-3-carbonsäure-benzylester herstellt.
- 25. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass man 5-(3,4-Dimethoxyphenyl)-3-isopropyl-2-oxazolidinon herstellt.

- X.worinR] niederes Alkyl oder Cycloalkyl,R2 niederes Alkyl, Cycloalkyl, Aryl, Aralkyl, Alkenyl oder einen heterocyclischen Rest mit einem Heteroatom, wobei dieses Heteroatom Sauerstoff oder Stickstoff ist,R3 Wasserstoff,R4 Wasserstoff oder niederes Alkyl,R5 Wasserstoff oder niederes Alkyl,Xj Schwefel undX2 Sauerstoff bedeuten, dadurch gekennzeichnet, dass man eine Verbindung der Formel II, worin R,_5 die oben angegebene Bedeutung haben, erst mit Schwefelkohlenstoff in Gegenwart einer organischen Base unter Kühlung und dann unter weiterer Kühlung mit Benzylchlorid umsetzt zu einer Verbindung der FormelOR,R1C »NOHworin R1; R2, R4 und Rs die oben angegebene Bedeutung haben, diese Verbindung anschliessend in einem inerten Lösungsmittel unter Kühlung mit Phosphortribromid zu einem 2-Benzylthio-l,3-thiazol-2 in der FormelR1s-bz.umsetzt und die so erhaltene Verbindung in der Wärme sauer hydrolysiert.
- 30. Verfahren zur Herstellung von neuen 5-(subst. Phe-nyl)-oxazolidinonen der allgemeinen Formel I,OR:1°worinRj niederes Alkyl oder Cycloalkyl,R2 niederes Alkyl, Cycloalkyl, Aryl, Aralkyl, Alkenyl oder einen heterocyclischen Rest mit einem Heteroatom, wobei dieses Heteroatom Sauerstoff oder Stickstoff ist, R3 Wasserstoff,R4 Wasserstoff oder niederes Alkyl,R5 Wasserstoff oder niederes Alkyl,X! und X2 Sauerstoff bedeuten, dadurch gekennzeichnet, dass man eine Verbindung der Formel II, worin R,_5 die oben angegebene Bedeutung haben, in einer organischen Base löst und mit Chlorameisensäureethylester unter Kühlung zu einem Carbaminsäureethylester der FormelOR,R1OHNH-COOEt worin R1; R2, R4 und R5 die oben angegebene Bedeutung haben, umsetzt und anschliessend diese Verbindung mit einem Alkalimetallalkohol in einem inerten Lösungsmittel in der Wärme behandelt.51015202530354045505560635 578
