Abstract

Crystalline cefazolin sodium, 7-(1-(1H)-tetrazolylacetamido)-3-[2-methyl-1,3,4-thiadiazolyl)-thiomet hyl]-3-cephem-4-carboxylic acid sodium salt, a standard antibiotic for parenteral administration, is prepared by a freeze-drying process in which a solution of cephazolin sodium in ethanol/water is slowly cooled within the space of 3 to 12 hours from room temperature down to -10 DEG C and is then cooled rapidly, until the mixture has completely solidified, prior to this frozen solution being subjected to a high vacuum and to moderate heating in order to sublime the frozen solvent. The powder thus obtained is notable for dissolving rapidly in pharmaceutically acceptable diluents and as a powder is capable of being stored for a very long time in a stable condition.

   PATENTANSPRÜCHE
1. Verfahren zur Herstellung von kristallinem, leicht auflösbarem Natriumsalz der   7-[1-(lH)-Tetrazolylacetamido]-       -3- [2-(5-methyl-1,3,4thiadiazolyl)-thiomethylj-3-cephem-4-      -carbonsäure,    gekennzeichnet durch die folgenden Stufen:

   a) langsames Abkühlen einer Lösung des genannten Na triumsalzes in einem Lösungsmittel aus 2 bis 25   Vol.-%    eines Alkanols mit 1 bis 3 Kohlenstoffatomen und 75 bis 98   Vol.-%    Wasser während 3 bis 12 Stunden auf    -100C;    b) weiteres Abkühlen des nach a) erhaltenen Präparats bis zur vollständigen Verfestigung der Mischung;

   c) Verminderung des Drucks der Umgebung, in der sich das nach b) erhaltene Präparat befindet, auf ein Maxi mum von 1 mm Hg, d) Erhöhen der Temperatur der Umgebung, in der sich das nach c) erhaltene Präparat befindet, auf ein Maximum von   50"C    unter Vermeidung des Schmelzens dieses Prä parats, und e) Sublimieren des Lösungsmittels des nach d) erhaltenen
Präparats bis zu einem Feuchtigkeitsgehalt der gebilde ten Kristalle des genannten Natriumsalzes von höchstens
6,0 Gew.-% und einem Alkanolgehalt von höchstens    1,0 Gew.-%.   



   2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass ein Lösungsmittel aus 10 bis 25   Vol.-%    eines Alkanols mit 1 bis 3 Kohlenstoffatomen und 75 bis 90 Vol.-% Wasser verwendet wird.



   3. Verfahren nach Anspruch 2, dadurch gekennzeichnet, dass eine Lösung verwendet wird, die 20 bis 40 g des genannten Natriumsalzes in 100 ml Lösung enthält.



   4. Verfahren nach Anspruch 3, dadurch gekennzeichnet, dass eine Lösung verwendet wird, die 23 bis 28 g des genannten Natriumsalzes in 100 ml Lösung enthält.



   5. Verfahren nach einem der Ansprüche 1 bis 4, dadurch gekennzeichnet, dass in Stufe c) der Druck der Umgebung auf 0,05 bis 0,20 mm Hg vermindert und dabei gehalten wird.



   6. Verfahren nach einem der Ansprüche 1 bis 5, dadurch gekennzeichnet, dass in Stufe d) die Temperatur der Umgebung langsam auf 10 bis   40"C    unter Vermeidung des Schmelzens des Präparats erhöht wird.



   7. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass als Alkanol mit 1 bis 3 Kohlenstoffatomen Ethanol oder vergälltes Ethanol verwendet wird.



   8. Verfahren nach Anspruch 1, gekennzeichnet durch folgende Stufen: a) Auflösen des genannten Natriumsalzes in einem Lö sungsmittel aus 2 bis 15% Ethanol und 98 bis 85%
Wasser; b) Filtrieren der nach a) erhaltenen Lösung durch ein steri lisierendes Filter; c) Abkühlen des nach b) erhaltenen sterilen Filtrats von
Zimmertemperatur bis zu einer Temperatur   von - 100C    in einer Zeit von 3 bis 12 Stunden; d) rasches Abkühlen des nach c) erhaltenen Präparats auf eine Temperatur von -30   bis 40CC;    e) Aufrechterhalten der Temperatur des nach d) erhaltenen
Präparats bei   unter -30"C    während 1 bis 2 Stunden; f) Verminderung des Drucks der Umgebung, in der das nach e) erhaltene Präparat gehalten wird, auf ein Maxi mum von 1 mm Hg;

   g) Erhöhen der Temperatur der Umgebung, in der das nach f) erhaltene Präparat gehalten wird, auf ein Maximum von   50"C    unter Vermeidung des Schmelzens des Präpa rats, und h) Absublimieren des Lösungsmittels aus dem nach g) er haltenen Präparat, bis die gebildeten Kristalle des Na triumsalzes einen Feuchtigkeitsgehalt von nicht mehr als
6,0   Gew.-%    und einen Ethanolgehalt von nicht mehr als
1,0 Gew.-% haben.



   Die Erfindung betrifft ein Verfahren zur Herstellung des Natriumsalzes der   7- [1-( 1H)-Tetrazolylacetamido] -3 -    -methyl- 1,3,4-thiadiazolyl)-thiomethyl] -3-cephem-4-carbonsäure, welches nach der freien internationalen Kurzbezeichnung, von der Weltgesundheitsorganisation registriert, Cefa   zolin-Natrium    heisst.    Übliche Zubereitungen von Cefazolin-Natrium sind Trokkenpulver, die unmittelbar vor ihrer Verwendung in einem wässrigen Verdünnungsmittel gelöst werden. Das   grocken-    pulver soll in praktisch kristalliner Form vorliegen, weil amorphes Cefalozolin-Natrium bei der Lagerung nachteilige Veränderungen erleidet und sich nur langsam lösen lässt.



  Aufgabe der Erfindung ist ein verbessertes Verfahren zur Herstellung von kristallinem Cefazolin-Natrium, das sich sehr schnell auflöst.



   Die Gefriertrocknung ist eine seit langem bekannte   Me    thode zur Entfernung eines Lösungsmittels von einem gelösten Stoff. Das   Verfahren    ist zwar mühselig, aufwendig und zeitraubend, führt jedoch zu einer Entfernung von Lösungsmitteln, ohne wärmeempfindliche gelöste Stoffe zu schädigen. Antibiotika und andere pharmazeutische Wirkstoffe sowie Nahrungsmittel, insbesondere Sofort-Kaffee, werden seit vielen Jahren auf diese Weise hergestellt. Eine Lösung, aus der der getrocknete gelöste Stoff gewonnen werden soll, wird bis zu einem Feststoff gefroren und einem Hochvakuum unterworfen, worauf die Temperatur erhöht wird, um die bei der Sublimation des gefroreren Lösungsmittels verbrauchte Wärme zu liefern. Die Temperatur wird unter derjenigen gehalten, bei der ein Schmelzen der gefroreren Lösung eintreten würde.

  Sie wird zusammen mit dem Vakuum so eingestellt, dass eine möglichst hohe Sublimationsgeschwindigkeit erzielt und gleichzeitig ein Schmelzen der gefrorenen Masse vermieden wird.



   Im allgemeinen ist Wasser das Lösungsmittel, das bei einem Gefriertrocknungsverfahren verwendet wird. Es   kapn    sich dabei aber auch um andere Lösungsmittel oder Lösungsmittelmischungen handeln, doch kommen nur solche in Betracht, die in dem praktisch anwendbaren Temperaturbereich fest werden und im Vakuum sublimieren.



   Bei der Gefriertrocknung von Antibiotika und anderen Arzneistoffen wird üblicherweise wie oben beschrieben verfahren, d.h. es wird eine Lösung hergestellt, die bis zur Ausbildung einer festen Masse gefroren und einem Hochvakuum unterworfen wird, worauf Wärme zugeführt und das Lösungsmittel sublimiert wird. Wenn jedoch diese herkömmliche Arbeitsweise auf Cefazolin-Natrium angewandt wird, dann wird ein langsam lösliches Pulver erhalten.

 

   Cefazolin-Natrium kann aus organischen Lösungsmitteln in ausreichend kristalliner Form gewonnen werden.



   Solch kristallines Cefazolin-Natrium bietet jedoch wieder andere Schwierigkeiten. Beispielsweise gibt es keinen wirklich wirksamen Weg zur Sterilisierung von aus organischen Lösungsmitteln gewonnenem Cefazolin-Natrium, weshalb das gesamte Kristallisationsverfahren unter aseptischen Be dingungen durchgeführt werden muss. Bei dem komplizier ten Verfahren, das zur Kristallisation von Cefazolin-Natrium angewandt werden muss, gibt es viele Möglichkeiten, wie verunreinigende Fremdstoffe in das Produkt gelangen können. Darüber hinaus ist noch keine Vorrichtung entwickelt worden, mit der es möglich wäre, trockenes Material mit der Genauigkeit und Gleichmässigkeit in Ampullen abzufüllen,  



  die mit Flüssigkeitsabfüllvorrichtungen ohne weiteres erreicht wird.



   In der US-PS 4 029 655 ist ein Gefriertrocknungsverfahren zur Herstellung von kristallinem Cefazolin-Natrium angegeben. Hierbei wird eine wässrige Lösung sehr rasch abgekühlt, um während des Kühlvorgangs Kerne von Cefazolin-Natrium auszubilden. Diese Kernbildung führt zur Kristallisation des Cefazolin-Natriums vor der Verfestigung des Lösungswassers. Deshalb liegt das Cefazolin-Natrium, wenn mit der Sublimation begonnen wird, bereits in Form von Kristallen vor und ist nicht von der Kristallisation abhängig, die bei der Entfernung des Lösungsmittels erfolgt.



  Das Lösungsmittel wird absublimiert, und das Cefazolin Natrium bleibt zurück.



   Dieses Verfahren führt zwar zu für die parenterale Verabreichung geeignetem stabilem sterilem kristallinem Cefazolin-Natrium, aber die gebildeten Kristalle lösen sich in annehmbaren pharmazeutischen Verdünnungsmitteln nicht rasch auf. Im allgemeinen ist zwei Minuten langes oder längeres kräftiges Schütteln erforderlich, um solches Cefazolin-Natrium wieder in Lösung zu bringen. Diese verhältnismässig lange Zeit ist eine Belastung für das Personal, das die injizierbare Lösung für die Verabreichung herstellen muss.



   Gegenstand der Erfindung ist ein Verfahren zur Herstellung von kristallinem leicht auflösbarem Cefazolin-Natrium, das durch folgende Stufen gekennzeichnet ist: a) langsames Abkühlen einer Lösung von Cefazolin-Na trium in einem Lösungsmittel aus 2 bis 25% eines Alka nols mit 1 bis 3 Kohlenstoffatomen und 75 bis   98%     (Volumen/Volumen) Wasser während 3 bis 12 Stunden    auf -100C;    b) weiteres Abkühlen des nach a) erhaltenen Präparats bis zur vollständigen Verfestigung der Mischung; c) Verminderung des Drucks der Umgebung, in der sich das nach b) erhaltene Präparat befindet, auf ein Maximum von 1 mm Hg;

   d) Erhöhen der Temperatur der Umgebung, in der sich das nach c) erhaltene Präparat befindet, auf ein Maximum von   50"C    unter Vermeidung des Schmelzens dieses Prä parats, und e) Sublimieren des Lösungsmittels des nach d) erhaltenen
Präparats bis zu einem Feuchtigkeitsgehalt der gebilde ten Kristalle von Cefazolin-Natrium von höchstens 6,0% und einem Alkanolgehalt von höchstens 1,0% (Gewicht/
Gewicht).



   Eine bevorzugte Ausführungsform des erfindungsgemässen Verfahrens ist durch folgende Stufen gekennzeichnet: (a) Cefazolin-Natrium wird in einem Lösungsmittel aus 2 bis 15% Ethanol und 98 bis 85% Wasser gelöst, (b) die nach (a) erhaltene Lösung wird in einen vorher sterilisierten Behälter steril filtriert, (c das nach (b) erhaltene Cefazolin-Natriumpräparat wird langsam innerhalb von 3 bis 12 Stunden   auf - 100C    abgekühlt, (d) die Temperatur des nach (c) erhaltenen Cefazolin-Natriumpräparats wird dann rasch auf -30   bis -40"C    gesenkt, (e) das nach (d) erhaltene Cefazolin-Natriumpräparat wird 1 bis 2 Stunden   bei -30"C    gehalten,   (f)    das nach (e) erhaltene Cefazolin-Natriumpräparat wird einer Umgebung ausgesetzt,

   worin der Druck bei höchstens 1 mm Hg absolut gehalten wird und (g) die Temperatur der Umgebung, in der sich das nach (f) erhaltene Cefazolin-Natriumpräparat bei einem Maximum von 1 mm Hg absolut befindet, wird auf bis zu   50"C    erhöht, wodurch das Lösungsmittel aus dem Cefazolin-Natriumpräparat sublimiert und ein kristallines Cefazolin-Natrium mit einem Feuchtigkeitsgehalt von nicht über 6,0% und einem Ethanolgehalt von nicht über 1,0% (Gewicht/Gewicht) erhalten wird.



   Der Ausdruck  Prozent  bezieht sich entweder auf Gewicht/Volumen-(G/V)-Prozent (g gelöster Stoff je 100 ml
Lösung) oder auf Volumen/Volumen-(V/V)-Prozent (ml einer reinen Flüssigkeit je 100 ml des Flüssigkeitsgemischs) oder auf Gewicht/Gewicht-(G/G)-Prozent (g eines Bestandteils je 100 g Gesamtgewicht).



   Das bei dem erfindungsgemässen Verfahren verwendete Alkanol mit 1 bis 3 Kohlenstoffatomen kann Methanol, Ethanol, Propanol oder Isopropanol sein. Alkanolgemische können ebenso wie vergällter Alkohol verwendet werden, unter der Voraussetzung, dass das Vergällungsmittel pharmazeutisch annehmbar oder flüchtig ist. Mit Methanol vergälltes Ethanol ist ein besonders zweckmässiges Alkanol.



  Ethanol und vergällte Ethanole sind die für das erfindungsgemässe Verfahren bevorzugten Alkanole.



   Das erfindungsgemässe Verfahren umfasst eine bevorzugte Arbeitsweise unter Anwendung einer Gefriertrocknung, wobei Cefalozolin-Natrium in einem Lösungsmittelsystem aus 2 bis 25% Alkanol und 75 bis 98% (V/V) Wasser gelöst wird. Diese Lösung wird dann langsam von Zimmertemperatur   auf - 100C    abgekühlt. Die Abkühlungsdauer soll 3 bis 12 Stunden betragen, in denen 50% oder mehr Cefazolin-Natrium aus der Lösung auskristallisieren, ehe ein Gefrieren einsetzt.



   Nach dem langsamen Abkühlen   auf - 100C    in 3 bis 12 Stunden wird die abgekühlte Lösung weiter stark gekühlt, bis die Mischung vollständig fest geworden ist. Nach den oben beschriebenen Stufen von ausschlaggebender Bedeutung wird eine bei der Gefriertrocknung herkömmliche Massnahme angewandt, um das Eis zu sublimieren, wodurch Cefazolin-Natriumkristalle mit einem Wassergehalt von höchstens   6%    und einem Alkoholgehalt von höchstens   1 %    anfallen.



   Diese Kristalle haben eine Lagerbeständigkeit von 3 Jahren oder darüber bei Zimmertemperatur und lösen sich in einem pharmazeutisch annehmbaren Verdünnungsmittel in für die parenterale Verabreichung geeigneten Konzentrationen in einer Minute oder weniger auf. Die Verbesserung der Löslichkeitseigenschaften des nach dem   erfindungsge-    mässen Verfahren hergestellten Cefazolin-Natriums gegen über in herkömmlicher Weise erhaltenen Cefazolin-Natriumkristallen besteht in einer etwa 50prozentigen Verminderung der Zeit, die zur Lösung des Cefazolin-Natriums in einem pharmazeutisch annehmbaren Verdünnungsmittel erforderlich ist.



   Das Wesen der Erfindung liegt in der Kombination der Verwendung des Lösungsmittels aus Alkanol und Wasser und in der Anwendung des langsamen Abkühlens der Lösung des Cefazolin-Natriums in diesem Lösungsmittel auf   -10"C.    Das Alkanol in dem Lösungsmittel ermöglicht das Abkühlen der Lösung   auf - 100C,    ohne dass sich darin Eis bildet. Es hat sich gezeigt, dass Cefazolin-Natrium in einem Lösungsmittel aus Alkanol und Wasser wesentlich weniger löslich ist als in Wasser allein. Deshalb kristalisieren bei der Temperatur   von - 100C    in Abhängigkeit von der Konzentration des gelösten Stoffs 50% oder mehr des Cefazolin Natriums aus der Lösung aus, ehe sich Eis zu bilden beginnt. 

  Darüber hinaus sind die durch die langsame Verminderung der Temperatur der Lösung von Zimmertemperatur bis auf - 100C gebildeten Kristalle gross und haben eine kleine Oberfläche im Vergleich zu Kristallen, wie sie bei der herkömmlichen Gefriertrocknung von Cefazolin-Natrium erhalten werden.



   Sobald sich Eis zu bilden beginnt, wird die Temperatur der Lösung vorzugsweise rasch vermindert, um die Entwicklung von Dendrit-Eis zu begünstigen, das die Kernbildung des in der Lösung verbliebenen Cefazolin-Natriums fördert.



  Bei der bevorzugten Durchführung des Verfahrens wird die Temperatur rasch auf -30   bis -40"C    gesenkt und 1 bis 2   Stunden aufrechterhalten, um vollständiges Gefrieren zu gewährleisten.



   Das erfindungsgemäss verwendete Lösungsmittel aus Alkanol und Wasser kann 2 bis 25% Alkanol und 75 bis 98% (V/V) Wasser enthalten. Das bevorzugte Lösungsmittelsystem besteht aus 10 Volumenprozent Alkanol und 90 Volumenprozent Wasser.



   Eine Konzentration von Cefazolin-Natrium von 20 bis 40% (G/V) in dem Lösungsmittel ist ausreichend. Der bevorzugte Bereich liegt zwischen 23 und 28% (G/V).



   Das kristalline Produkt wird vorzugsweise durch Filtrieren der Alkanol-Wasser-Lösung durch ein auf diesem Gebiet allgemein bekanntes steriles Filtermittel und Auffangen des Filtrats in einem vorher sterilisierten Behälter sterilisiert, ehe mit der Durchführung des erfindungsgemässen Gefriertrocknungsverfahrens begonnen wird. Die Sterilfiltration kann beispielsweise unter   Verwendung    einer hitzesterilisierten Platten-Rahmen-Filterpresse durchgeführt werden, die mit einem Asbestbausch oder einer Filtermembran aus Celluloseacetat oder -nitrat oder einer Wachsschicht mit einer Porosität von unter 0,22   tjm    ausgerüstet ist. Auf diese Weise wird die gesamte Lösung sterilisiert, und die Sterilität des kristallinen Produkts ist gesichert.



   Das langsame Abkühlen der Lösung von Cefazolin-Natrium kann am besten dadurch erzielt werden, dass die Lösung zuerst einer Umgebung von etwa   15"C    ausgesetzt und dann diese Temperatur langsam alle 30 Minuten um etwa   4"C    gesenkt wird, bis sich die Temperatur der Lösung auf - 100C eingestellt hat. Die Temperatur der Lösung kann durch ein Thermoelement bestimmt werden, das sich etwa in dem Zentrum der Lösung befindet und damit die Temperatur an diesem Punkt anzeigt.



   Wenn die Temperatur   von - 100C    erreicht worden ist, kann die Lösung rasch abgekühlt werden, bis sich die Mischung vollständig verfestigt hat. Die Geschwindigkeit der Abkühlung ist nicht von kritischer Bedeutung, und mit der Erniedrigung der Temperatur unter -400C ist kein Vorteil verbunden.



   Wird die Temperatur der gefrorenen Lösung auf etwa   -300C    oder tiefer erniedrigt, genügt eine Dauer von 1 bis 2 Stunden zur vollständigen Verfestigung der Masse. Bei diesem Punkt liegt praktisch das gesamte Cefalozin-Natrium als freie Kristalle in der gefrorenen Masse vor.



   Nach praktisch vollständiger   Kernbildung    oder Nukleation der Cefazolin-Natriumkristalle wird eine herkömmliche Gefriertrocknungsmassnahme angewandt, um das Lösungsmittel von der gefrorenen Masse abzusublimieren, wodurch ein Klumpen aus praktisch kristallinem Cefazolin-Natrium hinterbleibt. Das gefrorene Cefazolin-Natriumpräparat wird einer Umgebung ausgesetzt, wo der Druck auf einen Maximalwert von nicht mehr als 1 mm Hg absolut reduziert werden kann. Es ist bevorzugt, den Druck auf viel weniger als 1 mm Hg absolut zu erniedrigen. Die besten Ergebnisse werden mit einem absoluten Druck von 0,05 bis 0,2 mm erzielt.



  Dieser Druckbereich lässt sich ohne weiteres in Laboratoriums- und technischen Gefriertrocknungsvorrichtungen erreichen, deren Konstruktion und Betrieb allgemein bekannt sind.



   Nach der Senkung des Drucks der Umgebung auf einen wie oben angegebenen Wert wird der Umgebung Wärme zugeführt. Die Temperatur der Umgebung wird auf einen Punkt erhöht, wo die maximale Sublimationsgeschwindigkeit ohne Schmelzen der gefroreren Masse erreicht werden kann. Als allgemeine Regel stehen Temperatur und Druck in umgekehrtem Verhältnis zueinander, und die in der Sublimationsstufe angewandte Temperatur kann um so höher sein, je wirksamer die Druckerniedrigung ist. Als allgemeine Richtlinie kann angegeben werden, dass eine Höchsttemperatur der Umgebung von 500C mit einem hochwirksamen Vakuumsystem erreicht werden kann, mit dem der absolute Druck bei etwa 0,05 mm Hg absolut (50   m)    gehalten wird.



  Auf jeden Fall soll die Temperatur langsam erhöht werden, um eine Überlastung des Druckverminderungssystems zu vermeiden, was zu einem unerwünschten Schmelzen der gefrorenen Masse führen kann. Vorzugsweise wird die Temperatur der Umgebung in der Sublimationsstufe zwischen 10 und   40 C    bei einem Druck von 0,2 mm Hg absolut oder darunter gehalten.



   Das Absublimieren des Eises von der gefroreren Masse wird fortgesetzt, bis der Feuchtigkeitsgehalt der Cefalozolin Natriumkristalle unter 6% und der Alkanolgehalt unter   1 %    (G/G) liegt. Diese Werte sichern die physikalische Beständigkeit der schliesslich erhaltenen Kristalle. Cefalozolin Natrium kristallisiert als das Pentahydrat. Werden die Kristalle nach der vollständigen Absublimierung des Eises bei 25 bis   30"C    gehalten, dann wird das Pentahydrat in das Hemihydrat übergeführt, das bis zu etwa 6% (G/G) Feuchtigkeit enthalten kann.



   Das wie vorstehend beschrieben hergestellte Cefazolin Natrium ist weitgehend kristallin. Die physikalische Analyse von Cefazolin-Natrium-hemihydrat ergibt eine Kristallinität von über 90%. Auf jeden Fall wird ein Grad der   Kri-    stallinität erreicht, der genügt, um Lagerbeständigkeit zu verleihen. Diese Cefazolin-Natriumkristalle können in den richtigen Mengen für die Herstellung von Lösungen für die parenterale Verabreichung steril in vorher sterilisierte Ampullen abgefüllt werden.



   Bei einer bevorzugten Ausführungsform des erfindungsgemässen Verfahrens wird noch eine weitere Stufe durchgeführt, die darin besteht, dass ein abgemessenes Volumen steriler Alkanol-Wasser-Lösung in eine vorher sterilisierte Ampulle abgefüllt wird, wobei dieses abgemessene Volumen die Menge an Cefazolin-Natrium enthält, die nach der Gefriertrocknung in dieser Ampulle erwünscht ist. Die die Lösung von Cefazolin-Natrium enthaltenden Ampullen werden dann in der gleichen Weise behandelt, wie dies oben beschrieben wurde. Die erhaltenen gefriergetrockneten Cefazolin-Natriumampullen brauchen dann nur noch steril verschlossen werden.



   Die Gefriertrocknung in der Ampulle bringt wenigstens zwei Vorteile mit sich. Der erste Vorteil besteht darin, dass eine genauere und gleichbleibende Menge Cefazolin-Natrium in eine Ampulle in Form einer Flüssigkeit statt in fester Form (Kristalle) abgefüllt werden kann. Zweitens lassen sich sterile Betriebsbedingungen bei der Abfüllung von Flüssigkeit leichter erreichen und aufrechterhalten als bei der Abfüllung von trockenem Material. Darüber hinaus ist Luftverschmutzung bei der Hantierung von Flüssigkeiten ein geringeres Problem als bei der von trockenen Materialien.

 

   Durch die folgende Beispiele wird die Erfindung weiter erläutert.



   Beispiel 1
348 g Cefazolin-Natrium mit einem Feuchtigkeitsgehalt von etwa 3,5% werden in Wasser für Injektionen, U.S.P., gelöst, und die erhaltene Lösung wird mit weiterem Wasser für Injektionen auf 1200 ml eingestellt.



   Zu diesen 1200   ml    wässriger Lösung von Cefazolin-Natrium werden unter kräftigem Rühren 120 ml absolutes Ethanol gegeben.



   Die so erhaltene   ethanolisch-wäsmige    Lösung von Cefazolin-Natrium wird durch eine Membran mit Porenweiten von 0,45   m    filtriert und in vorher sterilisierte Phiolen in einer Menge von 4,35 ml/Phiole abgefüllt. Die Lösungsmenge ist so berechnet, dass jede Ampulle 1 g Cefazolin Natrium mit einem ungefähren   Überschuss    von 7% enthält.  



   Die gefüllten Phiolen werden in einen üblichen Gefriertrockner eingebracht, und die Temperatur der Lösung wird langsam innerhalb von 3 bis 4 Stunden   auf - 100C    gesenkt, worauf sie so schnell wie möglich auf   unter -30"C    erniedrigt wird. Die Phiolen werden 1 bis 2 Stunden nach dem Erreichen einer Temperatur   unter -30DC    bei dieser Temperatur belassen. Der Druck in dem Gefriertrockner wird auf unter 0,2 mm Hg absolut gesenkt, und die Temperatur wird für die Sublimation des Ethanol-Wasser-Lösungsmittels auf   lO C    erhöht. Schliesslich wird die Temperatur auf   25"C    erhöht, wobei dafür Sorge getragen wird, dass die gefrorene Masse in den Phiolen nicht schmilzt.

  Nach dem Ende der Sublimation wird das Vakuum aufgehoben, und die Phioleninhalte werden auf Feuchtigkeitsgehalt, Ethanolgehalt und Auflösungsgeschwindigkeit geprüft.



   Der Feuchtigkeitsgehalt einzelner Phiolen beträgt 2,16, 2,11, 2,05 und 2,24 Prozent (G/G).



   Zwei auf ihren Ethanolgehalt geprüfte Phiolen ergeben 0,42 und 0,45% (G/G).



   Fünf Phiolen werden hinsichtlich der Auflösungszeit geprüft; dabei werden Werte zwischen 30 und 60 Sekunden für die Auflösung ber Cefazolin-Natriums in 2,5 ml Wasser zur Injektion, U.S.P., erhalten.



   Beispiel 2
1 g Cefazolin-Natrium wird in 3 ml Wasser gelöst und mit 1 ml 95prozentigem Methanol versetzt. Die Lösung wird in einer Ampulle nach der in Beispiel 1 beschriebenen Temperatur- und Druckfolge gefriergetrocknet. Das nach dem Ende des Verfahrens erhaltene getrocknete Produkt wird m kroskopisch untersucht, wobei festgestellt wird, dass es zu   100%    aus kristallinem Cefazolin-Natrium besteht.



   Beispiel 3
1 g Cefazolin-Natrium wird in 3 ml Wasser gelöst und mit 1 ml 95prozentigem Ethanol versetzt. Es wird wiederum das Gefriertrocknungsverfahren nach Beispiel 1 angewandt, und das getrocknete Produkt wird mikroskopisch analysiert. Dabei zeigt sich, dass das Produkt zu etwa   100N    aus kristallinem Cefazolin-Natrium besteht.

 

   Beispiel 4
Bei Zimmertemperatur wird eine Lösung von 1 g Cefazolin-Natrium in 3 ml Wasser hergestellt. Nach Zugabe von 1 ml Isopropanol wird die Lösung wie in Beispiel 1 beschrie ben gefriergetrocknet. Die mikroskopische Prüfung des getrockneten Produkts ergibt, dass es praktisch zu 100% aus kristallinem Cefazolin-Natrium besteht.



   Beispiel 5
1 g Cefazolin-Natrium wird in 3 ml Wasser gelöst und mit 1 ml n-Propanol versetzt. Die wässrig-alkoholische Lösung wird nach dem in Beispiel 1 beschriebenen Verfahren gefriergetrocknet, und das Produkt wird mikroskopisch analysiert. Dabei zeigt sich, dass es aus kristallinem Cefazolin-Natrium besteht, worin praktisch keine amorphe Verbindung enthalten ist.

   The invention relates to a process for the preparation of the sodium salt of 7- [1- (1H) -tetrazolylacetamido] -3 - -methyl-1,3,4-thiadiazolyl) -thiomethyl] -3-cephem-4-carboxylic acid, which according to Free international short name, registered by the World Health Organization, is called Cefa zolin sodium.    Common preparations of cefazolin sodium are dry powders which are dissolved in an aqueous diluent immediately before use. The grocken powder is said to be in practically crystalline form because amorphous cefalozoline sodium undergoes adverse changes during storage and can only be dissolved slowly.



  The object of the invention is an improved process for the production of crystalline sodium cefazolin which dissolves very quickly.



   Freeze-drying is a long-known method for removing a solvent from a solute. While the process is tedious, time-consuming and time-consuming, it does remove solvents without damaging heat-sensitive solutes. Antibiotics and other active pharmaceutical ingredients as well as foods, especially instant coffee, have been produced in this way for many years. A solution from which the dried solute is to be recovered is frozen to a solid and subjected to a high vacuum, whereupon the temperature is raised to provide the heat consumed in the sublimation of the frozen solvent. The temperature is kept below that at which the frozen solution would melt.

  It is adjusted together with the vacuum so that the highest possible sublimation speed is achieved and at the same time melting of the frozen mass is avoided.



   In general, water is the solvent used in a freeze-drying process. However, these can also be other solvents or solvent mixtures, but only those that come into consideration are those which become solid in the practically applicable temperature range and sublime in a vacuum.



   Freeze-drying of antibiotics and other drugs is usually carried out as described above, i.e. a solution is prepared which is frozen and subjected to a high vacuum until a solid mass is formed, whereupon heat is applied and the solvent is sublimed. However, when this conventional procedure is applied to cefazolin sodium, a slowly soluble powder is obtained.

 

   Cefazolin sodium can be obtained in sufficient crystalline form from organic solvents.



   However, such crystalline cefazolin sodium presents other difficulties. For example, there is no really effective way to sterilize cefazolin sodium obtained from organic solvents, which is why the entire crystallization process must be carried out under aseptic conditions. In the complicated process that must be used to crystallize sodium cefazoline, there are many ways in which contaminants can enter the product. In addition, no device has been developed with which it would be possible to fill dry material into ampoules with the accuracy and uniformity,



  which is easily achieved with liquid filling devices.



   US Pat. No. 4,029,655 discloses a freeze-drying process for the production of crystalline sodium cefazoline. Here, an aqueous solution is cooled very quickly in order to form nuclei of cefazolin sodium during the cooling process. This nucleation leads to the crystallization of the cefazolin sodium before the solution water solidifies. Therefore, when sublimation is started, the cefazolin sodium is already in the form of crystals and is not dependent on the crystallization that occurs when the solvent is removed.



  The solvent is sublimed and the cefazolin sodium remains.



   While this method results in stable sterile crystalline cefazolin sodium suitable for parenteral administration, the crystals formed do not dissolve quickly in acceptable pharmaceutical diluents. In general, vigorous shaking for two minutes or longer is required to redissolve such cefazolin sodium. This relatively long time is a burden on the personnel who have to prepare the injectable solution for administration.



   The invention relates to a process for the preparation of crystalline, easily dissolvable cefazolin sodium, which is characterized by the following steps: a) slow cooling of a solution of cefazolin sodium in a solvent of 2 to 25% of an alkanol with 1 to 3 carbon atoms and 75 to 98% (volume / volume) water for 3 to 12 hours at -100C; b) further cooling of the preparation obtained according to a) until the mixture has solidified completely; c) reducing the pressure of the environment in which the preparation obtained according to b) is located to a maximum of 1 mm Hg;

   The alkanol having 1 to 3 carbon atoms used in the process according to the invention can be methanol, ethanol, propanol or isopropanol. Alcohol mixtures can be used as well as denatured alcohol, provided that the denaturant is pharmaceutically acceptable or volatile. Ethanol denatured with methanol is a particularly useful alkanol.



  Ethanol and denatured ethanols are the preferred alkanols for the process according to the invention.



   The process according to the invention comprises a preferred procedure using freeze drying, in which cefalozoline sodium is dissolved in a solvent system consisting of 2 to 25% alkanol and 75 to 98% (v / v) water. This solution is then slowly cooled from room temperature to -100C. The cooling time should be 3 to 12 hours, in which 50% or more of cefazolin sodium crystallize from the solution before freezing begins.



   After slowly cooling to -100C in 3 to 12 hours, the cooled solution is further cooled strongly until the mixture has completely solidified. According to the stages of crucial importance described above, a conventional freeze-drying measure is used to sublimate the ice, thereby producing cefazolin sodium crystals with a water content of at most 6% and an alcohol content of at most 1%.



   These crystals have a shelf life of 3 years or more at room temperature and dissolve in a pharmaceutically acceptable diluent in concentrations suitable for parenteral administration in one minute or less. The improvement in the solubility properties of the cefazolin sodium produced by the process according to the invention compared to cefazolin sodium crystals obtained in a conventional manner consists in an approximately 50 percent reduction in the time required for dissolving the cefazolin sodium in a pharmaceutically acceptable diluent.



   The essence of the invention lies in the combination of the use of the solvent from alkanol and water and in the application of slowly cooling the solution of the cefazoline sodium in this solvent to -10 ° C. The alkanol in the solvent enables the solution to be cooled to 100C without the formation of ice in it. It has been shown that cefazolin sodium is much less soluble in a solvent of alkanol and water than in water alone, which is why it crystallizes at a temperature of - 100C depending on the concentration of the solute 50% or more of the cefazolin sodium out of solution before ice begins to form.

  In addition, the crystals formed by slowly lowering the temperature of the solution from room temperature to -100C have a large surface area and have a small surface area compared to crystals obtained in the conventional freeze drying of cefazolin sodium.



   Once ice begins to form, the temperature of the solution is preferably rapidly reduced to favor the development of dendrite ice, which promotes the nucleation of the cefazolin sodium remaining in the solution.



  In the preferred practice of the process, the temperature is rapidly lowered to -30 to -40 "C and maintained for 1 to 2 hours to ensure complete freezing.



   The solvent of alkanol and water used according to the invention can contain 2 to 25% alkanol and 75 to 98% (v / v) water. The preferred solvent system consists of 10 volume percent alkanol and 90 volume percent water.



   A concentration of cefazolin sodium of 20 to 40% (w / v) in the solvent is sufficient. The preferred range is 23-28% (w / v).



   The crystalline product is preferably sterilized by filtering the alkanol-water solution through a sterile filter medium well known in the art and collecting the filtrate in a previously sterilized container before beginning the freeze-drying process of the invention. The sterile filtration can be carried out, for example, using a heat-sterilized plate-frame filter press equipped with an asbestos pad or a filter membrane made of cellulose acetate or nitrate or a wax layer with a porosity of less than 0.22 μm. In this way, the entire solution is sterilized and the sterility of the crystalline product is ensured.



   The slow cooling of the solution of cefazolin sodium can best be accomplished by first exposing the solution to an environment of about 15 "C and then slowly lowering that temperature by about 4" C every 30 minutes until the temperature of the solution increases is set to - 100C. The temperature of the solution can be determined by a thermocouple, which is located approximately in the center of the solution and thus indicates the temperature at this point.



   When the temperature of -100C has been reached, the solution can be cooled rapidly until the mixture has solidified completely. The rate of cooling is not critical and there is no advantage in lowering the temperature below -400C.



   If the temperature of the frozen solution is lowered to about -300C or lower, a period of 1 to 2 hours is sufficient for the mass to solidify completely. At this point practically all of the cefalozin sodium is present as free crystals in the frozen mass.



   After virtually complete nucleation or nucleation of the cefazolin sodium crystals, a conventional freeze-drying measure is used to sublime the solvent from the frozen mass, leaving a lump of practically crystalline cefazolin sodium. The frozen cefazolin sodium preparation is exposed to an environment where the pressure can be absolutely reduced to a maximum value of no more than 1 mm Hg. It is preferred to lower the pressure to much less than 1 mm Hg absolute. The best results are achieved with an absolute pressure of 0.05 to 0.2 mm.



  This pressure range can easily be achieved in laboratory and technical freeze-drying devices, the construction and operation of which are generally known.



   After the pressure of the environment is reduced to a value as stated above, heat is supplied to the environment. The temperature of the environment is raised to a point where the maximum sublimation rate can be achieved without melting the frozen mass. As a general rule, temperature and pressure are inversely related, and the more effective the pressure reduction is, the higher the temperature used in the sublimation step. As a general guideline it can be stated that a maximum ambient temperature of 500C can be achieved with a highly effective vacuum system, with which the absolute pressure is kept at about 0.05 mm Hg absolute (50 m).



  In any case, the temperature should be slowly increased in order to avoid overloading the pressure reduction system, which can lead to an undesired melting of the frozen mass. The temperature of the environment in the sublimation stage is preferably kept between 10 and 40 ° C. at a pressure of 0.2 mm Hg absolute or below.



   The sublimation of the ice from the frozen mass continues until the moisture content of the cefalozolin sodium crystals is below 6% and the alkanol content is below 1% (w / w). These values ensure the physical stability of the crystals finally obtained. Cefalozolin sodium crystallizes as the pentahydrate. If the crystals are kept at 25 to 30 ° C. after the ice has been completely sublimed, the pentahydrate is converted into the hemihydrate, which can contain up to about 6% (w / w) moisture.



   The cefazolin sodium produced as described above is largely crystalline. The physical analysis of cefazolin sodium hemihydrate shows a crystallinity of over 90%. In any case, a level of crystallinity is achieved which is sufficient to give shelf life. These cefazolin sodium crystals can be filled into the sterilized ampoules in the correct amounts for the preparation of solutions for parenteral administration.



   In a preferred embodiment of the method according to the invention, a further step is carried out, which consists in filling a measured volume of sterile alkanol-water solution into a previously sterilized ampoule, this measured volume containing the amount of cefazolin sodium which after freeze-drying in this ampoule is desired. The ampoules containing the solution of cefazolin sodium are then treated in the same manner as described above. The freeze-dried cefazolin sodium ampules obtained then only need to be sealed under sterile conditions.



   Freeze-drying in the ampoule has at least two advantages. The first advantage is that a more precise and constant amount of cefazolin sodium can be filled into an ampoule in the form of a liquid instead of a solid (crystals). Second, sterile operating conditions are easier to achieve and maintain when filling liquid than when filling dry material. In addition, air pollution is less of a problem when handling liquids than when handling dry materials.

 

   The invention is further illustrated by the following examples.



   example 1
348 g of cefazolin sodium with a moisture content of about 3.5% are dissolved in water for injections, U.S.P. and the resulting solution is adjusted to 1200 ml with further water for injections.



   120 ml of absolute ethanol are added to these 1200 ml of aqueous solution of cefazolin sodium with vigorous stirring.



   The ethanolic aqueous solution of cefazolin sodium thus obtained is filtered through a membrane with a pore size of 0.45 m and filled into previously sterilized vials in an amount of 4.35 ml / vial. The amount of solution is calculated so that each ampoule contains 1 g of cefazolin sodium with an approximate excess of 7%.



   The filled vials are placed in a conventional freeze dryer, and the temperature of the solution is slowly lowered to -100 ° C. in the course of 3 to 4 hours, after which it is reduced to below -30 ° C. as quickly as possible. The vials become 1 to 2 hours after reaching a temperature below -30DC, remain at this temperature, the pressure in the freeze dryer is reduced to below 0.2 mm Hg absolute, and the temperature is raised to 10 ° C. for the sublimation of the ethanol-water solvent Temperature increased to 25 "C, taking care that the frozen mass in the vials does not melt.

  After sublimation has ended, the vacuum is released and the vial contents are checked for moisture content, ethanol content and dissolution rate.



   The moisture content of individual vials is 2.16, 2.11, 2.05 and 2.24 percent (w / w).



   Two vials tested for their ethanol content give 0.42 and 0.45% (w / w).

   Example 2
1 g of cefazolin sodium is dissolved in 3 ml of water and 1 ml of 95 percent methanol is added. The solution is freeze-dried in an ampoule according to the temperature and pressure sequence described in Example 1. The dried product obtained after the end of the process is examined microscopically, whereby it is found that it consists of 100% crystalline sodium cefazolin.

1 g of cefazolin sodium is dissolved in 3 ml of water and 1 ml of 95 percent ethanol is added. The freeze-drying method according to Example 1 is again used and the dried product is analyzed microscopically. It turns out that the product consists of about 100N crystalline cefazolin sodium.

 

   Example 4
A solution of 1 g of cefazolin sodium in 3 ml of water is prepared at room temperature. After adding 1 ml of isopropanol, the solution is freeze-dried as described in Example 1. Microscopic examination of the dried product shows that it is practically 100% crystalline cefazolin sodium.



   Example 5
1 g of cefazolin sodium is dissolved in 3 ml of water and 1 ml of n-propanol is added. The aqueous-alcoholic solution is freeze-dried according to the procedure described in Example 1 and the product is analyzed microscopically. It shows that it consists of crystalline sodium cefazolin, which contains practically no amorphous compound.

PATENTANSPRÜCHE 1. Verfahren zur Herstellung von kristallinem, leicht auflösbarem Natriumsalz der 7-[1-(lH)-Tetrazolylacetamido]- -3- [2-(5-methyl-1,3,4thiadiazolyl)-thiomethylj-3-cephem-4- -carbonsäure, gekennzeichnet durch die folgenden Stufen:  PATENT CLAIMS 1.
