Description
Die Erfindung betrifft therapeutisch wirksame Imidazo-und Pyrimido [2,l-b]chinazoline der allgemeinen Formel:

in der Ri, R2 und R3 unabhängig voneinander Wasserstoffoder Halogenatome, C 1-3-Alkyl- oder Ci-3-Alkoxyreste bedeuten, R4 einen Ci-s-Alkyl, Cs-s-Alkenyl, Aralkyl- oder Cyclo-C3-8-alkyl-Ci-3-alkylrest darstellt, Rs ein Wasserstoffatom oder eine Hydroxylgruppe ist und A einen gegebenenfalls durch ein oder zwei Ci-3-Àlkylreste substituierten C2-3-Alkylenrest bedeutet.
Unter « Alkylresten» werden geradkettige und verzweigte gesättigte aliphatische Kohlenwasserstoffreste verstanden, wobei die Ci-5-Alkylreste z.B. Methyl-, Äthyl-, n-Propyl-,'Iso-propyl-, n-Butyl-, Isobutyl-, tert.-Butyl- odern-Pentyl-gruppen sind. Ci-3-Alkoxyreste sind z.B. die Methoxy-, Äthoxy-, n-Propoxy- oder Isopropoxygruppe. Geeignete «CVj-Alkenylreste» sind z.B. die Allyl-, 2- und 3-Butenyl-gruppe. Unter « Aralkylresten» werden Arylreste verstanden,
die mit C 1-3-Alkylresten substituiert sind, wobei der Arylrest eine Phenyl- oder halogensubstituierte Phenylgruppe ist. «Cyclo-C3-8-alkyl» sind z.B. die Cyclopropyl-, Cyclobutyl-, Cyclopentyl-, Cyclohexyl-, Cycloheptyl-und die Cyclooctyl-5 gruppe. Der Ausdruck «Halogen» umfasst alle vier Halogene, vorzugsweise Chlor und Brom. Beispiele für den gegebenenfalls substituierten C2-3-Alkylenrest A sind die Äthylen-, Propylen-, 1,1-Dimethyläthylen-, 1,2-Dimethyl-äthylen-, 1-Äthyläthylen-, l-(n-Propyl)-äthylen-undTri-10 methylengruppe. Gegenstand der Erfindung sind auch die Additionssalze der Verbindungen der Formel I mit pharmazeutisch verträglichen Säuren. Derartige Säuren umfassen sowohl organische als auch anorganische Säuren, wie Salzsäure, Bromwasserstoffsäure, Jodwasserstoffsäure, Schwefel-15 säure, Phosphorsäure, Essigsäure, Oxalsäure, Maleinsäure, Fumarsäure, Bernsteinsäure, Weinsäure, Citronensäure, Ascorbinsäure, Glutaminsäure, Asparaginsäure, Stearinsäure und Palmitinsäure.
Die tricyclischen Chinazoline der Formel I haben wert-20 volle pharmakologische Eigenschaften, insbesondere besitzen sie diuretische und vasodilatatorische Wirkung und eignen sich zur Behandlung von kardiovaskulären Erkrankungen.

Bei der Bestimmung der diuretischen Aktivität an Ratten nach der Methode von W.L. Lipschits et al., J. Pharmacol. Exp. Ther., Bd. 79, S. 97 ( 1943) wurden die in Tabelle I genannten Ergebnisse erzielt, wobei zum Vergleich die Werte für eine Standarddosis von 10 mg/kg angegeben sind.

Die vasodilatatorische Aktivität wird nach der Methode gibt man Papaverin in einer Konzentration von 3 x 10-5 g/ml von N.Tada et al., J. Pharmacol. Exp. Ther., Bd. 191, S. 139 50 zu, wobei die dadurch induzierte Relaxation mit 100% ange-

Für die Verabreichung an Warmblüter können die erfin-dungsgemässen Verbindungen mit festen oder flüssigen pharmazeutischen Trägerstoffen kombiniert und z.B. zu Tabletten, Pulverpräparaten, Kapseln oder ähnlichen festen Dosierungsformen formuliert oder für die enterale oder parenterale Verabfolgung in geeigneten Lösungsmitteln oder Verdünnungsmitteln gelöst oder suspendiert werden. Die Arzneimittel der Erfindung können darüber hinaus auch andere bekannte Wirkstoffe enthalten.
Gegenstand der Erfindung sind ferner die folgenden Verfahren zur Herstellung der tricyclischen Chinazoline der Formel I.
Verbindungen der Formel:
in der Ri, R2, R3, R4 und A die vorstehende Bedeutung haben, werden dadurch hergestellt, dass man eine Verbindung der Formel:

Ein weiteres Verfahren zur Herstellung der Verbindungen der Formel Ia besteht darin, dass man eine Verbindung der Formel:

in der Ri, R2, R3, R4 und A die vorstehende Bedeutung haben, mit Phosphoroxychlorid und dann mit einer Alkalibase umsetzt.
Die Reaktion der Verbindung II mit Phosphoroxychlorid kann bei erhöhten Temperaturen in Gegenwart oder Abwesenheit eines inerten Lösungsmittels durchgeführt werden. Vorzugsweise erfolgt die Umsetzung unter Rückfluss in überschüssigem Phosphoroxychlorid, wobei gegebenenfalls ein inertes Lösungsmittel, wie Benzol, Toluol, Xylol, Chloroform oder Äthylendichlorid, eingesetzt wird. Nach dem Abdestillieren von nicht umgesetztem Phosphoroxychlorid wird der Rückstand mit einer Alkalibase, wie Natriumhydroxid, Kaliumhydroxid, Natriumcarbonat, Kaliumcar-bonat oder Ammoniakwasser basisch gemacht, wobei die gewünschte Verbindung der Formel Ia anfällt.
Die Ausgangsverbindungen der Formel II können dadurch hergestellt werden, dass man eine Verbindung der Formel:

in der Ri, Ra, R3, R4 und A die vorstehende Bedeutung haben und Y ein Halogenatom ist, mit einer Alkalibase in einem inerten Lösungsmittel umsetzt. Geeignete Alkalibasen sind z.B. Natriumhydroxid, Kaliumhydroxid, Natriumcarbonat, Kaliumcarbonat, und Ammoniakwasser. Als Lösungsmittel können z.B. Methanol, Äthanol, Isopropanol, Tetrahydro-furan, Aceton, Benzol oder Toluol verwendet werden.
Die Ausgangsverbindungen der Formel V lassen sich dadurch herstellen, dass man eine Verbindung der Formel II mit Thionylchlorid, Phosphortrichlorid, Phosphortribromid, Phosphoroxychlorid oder dergleichen umsetzt.
Ein weiteres Verfahren zur Herstellung der Verbindungen der Formel I besteht darin, dass man eine Verbindung der Formel:

in der R.3 die vorstehende Bedeutung hat und Z Lithium, oder MgX', wobei X' ein Halogenatom ist, darstellt, in einem inerten Lösungsmittel und anschliessend mit Wasser umsetzt, wobei eine Verbindung der Formel:

in der Ri, R2, R3, R4 und A die vorstehende Bedeutung haben, entsteht.
Die erhaltene Verbindung der Formel Ib wird dann mit einem Reduktionsmittel, das sich für die Hydrogenolyse von tertiären Alkoholen eignet, weiter umgesetzt.
Die Reaktion in der ersten Stufe kann bei einer Temperatur im Bereich von Raumtemperatur bis zum Siedepunkt des angewandten Lösungsmittels durchgeführt werden. Als Lösungsmittel eignen sich z.B. Diäthyläther, Dimethoxyä-than, Diäthylenglykoldimethyläther, Tetrahydrofuran und Dioxan.
Die Reduktion kann z.B. mit einem unedlen Metall in einem Alkohol oder einer Säure erfolgen, z.B. Zink und Salzsäure oder Natrium und Alkohol, durch katalytische Hydrierung über Raney-Nickel oder Palladium-auf-Aktivkohle oder mit Natriumborhydrid und Trifluoressigsäure nach dem Verfahren von G.W. Gribble et al., Synthesis (1977), S. 172.

Die Beispiele erläutern die Erfindung.
Beispiel 1
Eine Suspension von 7,6 g 6-Chlor-3,4-dihydro-l-(n-60 butyl)-4-phenyl-2-(l H)-chinazolinthion in 80 ml Methanol wird mit 4,9 g Methyljodid versetzt. Hierauf erhitzt man das Gemisch 2 Stunden unter Rückfluss und kühlt dann mit Eis ab. Der erhaltene Niederschlag wird abfiltriert, mit Methanol gewaschen und getrocknet, wobei 10 g 6-Chlor-l,4-dihydro-65 1 -(n-butyl)-2-methylthio-4-phenylchinazolin-hydrojodid erhalten werden, F. 203 °C (Zers.).
Ein Gemisch aus 6,15 g des erhaltenen Chinazolin-hydro-jodids und 10 ml Monoäthanolamin wird 5 Stunden bei 110

bis 120°C gerührt. Nach dem Abkühlen giesst man das Gemisch in Eiswasser und extrahiert mit Chloroform. Die Chloroformschicht wird mit Wasser gewaschen, über Natriumsulfat getrocknet und unter vermindertem Druck eingedampft. Durch Chromatographieren des Rückstands an Sili-cagel mit Chloroform/Methanol (Volumenverhältnis 15:1) als Eluiermittel erhält man 4,6 g 6-Chlor-1,4-dihydro-2-(ß-hydroxyäthylamino)-l-(n-butyl)-4-phenylchinazolin als hellbraunes Öl.
Ein Gemisch dieses Öls mit 15 ml Phosphoroxychlorid wird 1 Stunde unter Rückfluss erhitzt, dann abgekühlt und in Eiswasser gegossen. Das erhaltene Gemisch wird mit Ammoniakwasser basisch gemacht und mit Chloroform extrahiert. Die Chloroformschicht wird mit Wasser gewaschen, über Natriumsulfat getrocknet und unter vermindertem Druck zur Trockene eingeengt. Durch Chromatographieren des Rückstands an Silicagel mit Chloroform als Eluiermittel erhält man 3,2g7-Chlor-10-(n-butyl)-5-phenyl-2,3,5,10-tetrahy-droimidazo[2,1 -bjchinazolin als hellbraunes Öl, das aus n-Hexan in Form farbloser Prismen kristallisiert, F. 97 bis 97,5°C.
Beispiel 2
Nach dem Verfahren von Beispiel 1 werden aus den entsprechenden Ausgangsverbindungen die folgenden 2,3,5,10-Tetrahydroimidazo[2,1 -bjchinazolinderivate hergestellt:
7-Chlor-l 0-methyl-5-phenyl-2,3,5,10-tetrahydroimidazo[2,1 -b]-chinazolin, F. 129-129,5°C,
7-Chlor-10-isopropyl-5-phenyl-2,3,5,10-tetrahydroimi-dazo[2,l-bjchinazolin, hellgelbes Öl,
7-Chlor-10-cyclopropylmethyl-5-phenyl-2,3,5,10-tetrahy-droimidazo[2,1 -bjchinazolin-hydrochlorid,
1/2 Isopropanol, F. 291 bis 292°C (Zers.).
8-Chlor-10-cyclopropylmethyl-5-phenyl-2,3,5,10-tetrahy-droimidazo[2,l-bjchinazolin-hydrochlorid, F. 301 bis 302°C,
9-Chlor-10-cyclopropylmethyl-5-phenyl-2,3,5,10-tetrahy-droimidazo[2,l-b]chinazolin, hellbraunes Öl,
10-Cyclopropylmethyl-7-methoxy-5-phenyl-2,3,5,10-tetrahy-droimidazo[2,1 -bjchinazolin-hydrochlorid,
F. 265,5-266°C (Zers.).
10-(n-Butyl)-5-phenyl-2,3,5,10-tetrahydroimidazo[2,1 -bj-chinazolin-hydrochlorid, F. 297°C (Zers.).
Beispiel 3
Eine Suspension von 14,2 g 6-Chlor-3,4-dihydro-l-methyl-4-phenyl-2(l H)-chinazolinthion in 140 ml Methanol wird mit 7,68 g Methyljodid versetzt. Hierauf erhitzt man das Gemisch 3 Stunden unter Rückfluss und engt dann ein. Nach dem Abkühlen mit Eis wird der Niederschlag abfiltriert, mit Methanol gewaschen und getrocknet, wobei 20,5 g 6-Chlor-1,4-dihydro-1 -methyl-2-methylthio-4-phenylchinazolin-hydro-jodid, F. 196 bis 197°C (Zers.) erhalten werden.
Ein Gemisch aus 4,31 g des erhaltenen Chinazolin-hydro-jodids und 15 ml n-Propanolamin wird 2 Stunden auf 100°C erhitzt. Nach dem Abkühlen gibt man Isopropyläther zu und filtriert den erhaltenen Niederschlag ab. Die Kristalle werden dann in Chloroform gelöst, worauf man die Lösung nacheinander mit verdünntem Ammoniakwasser und Wasser wäscht, über Natriumsulfat trocknet und unter vermindertem Druck zur Trockene einengt. Durch Kristallisieren des Rückstands aus Chloroform/Isopropyläther erhält man 3,1 g 6-Chlor-1,4-dihydro-2-(y-hydroxypropylamino)-1 -methyl-4-phenyl-chinazolin, F. 165 bis 166°C.
Ein Gemisch aus 2,85 g des erhaltenen 2-(y-Hydroxypro-pylamino)-chinazolins und 10 ml Phosphoroxychlorid wird 3 Stunden unter Rückfluss erhitzt. Nach dem Abtrennen von nicht umgesetztem Phosphoroxychlorid unter vermindertem
Druck macht man den Rückstand mit Ammoniakwasser unter Eiskühlung basisch. Das Gemisch wird dann mit Chloroform extrahiert, worauf man die Chloroformschicht mit Wasser wäscht, über Natriumsulfat trocknet und unter ver-s mindertem Druck zur Trockene einengt. Durch Chromatographieren des zurückbleibenden braunen Öls an aktiviertem Aluminiumoxid mit Chloroform als Eluiermittel erhält man 2,7 g 8-Chlor-l l-methyl-6-phenyl-3,4,6,1 l-tetrahydro-2H-pyrimido[2,l-bjchinazolin. Das Produkt wird dann in einer 10 Lösung von Chlorwasserstoff in Methanol gelöst und unter vermindertem Druck wieder eingeengt. Durch Umkristallisieren des Rückstands aus Methanol/Isopropyläther erhält man das Hydrochlorid in Form von farblosen Prismen, F. 257,5 bis 258°C (Zers.).

Beispiel 4
Nach dem Verfahren von Beispiel 3 werden aus den entsprechenden 3,4-Dihydro-2( 1 H)-chinazolinthionderivaten die folgenden 3,4,6,1 l-Tetrahydro-2H-pyrimido[2,l-b]china-20 zolinderivate hergestellt:
8-Chlor-11-(n-butyl)-6-phenyl-3,4,6,11-tetrahydro-2H-pyri-mido[2,l-bjchinazolin-hydrochlorid, F. 273-274°C (Zers.),
9-Chlor-1 l-cyclopropylmethyl-6-phenyl-3,4,6,11-tetrahydro-25 2H-pyrimido[2,l-bjchinazolin-hydrochlorid, F. 276-277°C
(Zers.).
Beispiel 5
Ein Gemisch aus 3,3 g 6-Chlor-1,4-dihydro-2-(y-hydroxy-30 propylamino)-l-methyl-4-phenylchinazolin, 3,6 gThionyl-chlorid und 50 ml Chloroform wird bei Raumtemperatur über Nacht gerührt und dann weitere 2 Stunden unter Rückfluss erhitzt. Durch Einengen des Reaktionsgemisches zur Trockene unter vermindertem Druck erhält man 3,5 g 35 6-Chlor-1,4-dihydro-2-(y-chlorpropylamino)-1 -methyl-4-phenylchinazolin als Rückstand. Der Rückstand wird mit 100 ml wasserfreiem Äthanol und 2,8 g Kaliumcarbonat versetzt und 5 Stunden unter Rückfluss erhitzt. Nach dem Abkühlen filtriert man unlösliche Bestandteile ab und engt 40 das Filtrat zur Trockene ein. Durch Chromatographieren des Rückstands an aktiviertem Aluminiumoxid mit Chloroform als Eluiermittel erhält man 8-Chlor-11-methyl-6-phenyl-3,4,6,11 -tetrahydro-2H-pyrimido[2,1 -bjchinazolin.
45 Beispiel 6
Eine Phenylmagnesiumbromid-Lösung, die aus 6,9 g p-Chlorphenylbromid, 0,88 g Magnesium und 25 ml wasserfreiem Diäthyläther hergestellt worden ist, wird unter Rühren und Eiskühlung eine Lösung von 2,42 g 10-Methyl-2,3,5,10-50 tetrahydroimidazo[2,l-b]chinazolin-5-on in 30 ml wasserfreiem Tetrahydrofuran getropft. Das Gemisch wird 2 Stunden unter Rückfluss erhitzt und dann abgekühlt. Hierauf tropft man 200 ml Wasser zu und rührt noch eine Weile. Anschliessend extrahiert man das erhaltene Gemisch mit heissem Chloroform, filtriert die unlöslichen Bestandteile ab und wäscht sie mit heissem Chloroform. Die Chloroformschicht wird über Natriumsulfat getrocknet und unter vermindertem Druck zur Trockene eingeengt. Durch Kristallisieren des Rückstands aus Äthanol und Umkristallisieren des 60 erhaltenen Produkts aus Dimethylformamid erhält man 1,0 g 5-(p-Chlorphenyl)-5-hydroxy-10-methyl-2,3,5,10-tetrahy-droimidazo[2,l-bjchinazolin in Form von farblosen Nadeln, F. 207 bis 209°C (Zers.).

Beispiel 8
20 ml Trifluoressigsäure werden portionsweise unter Rühren unterhalb 0°C unter Stickstoff mit 0,48 g 5-Hydroxy-10-(n-butyl)-5-phenyl-2,3,5,10-tetrahydroimidazo[2,1-bjchinazolin und 0,57 g Natriumborhydrid versetzt. Das Reaktionsgemisch wird 1 Stunde bei 0 bis 5°C gerührt,
worauf man die Trifluoressigsäure unter vermindertem Druck abdampft. Der Rückstand wird mit 20 ml Eiswasser und dann mit 15 ml 40prozentiger Natronlauge versetzt. Nach kurzem Rühren extrahiert man das Gemisch mit Chloroform, wäscht die Chloroformschicht mit Wasser, trocknet über Natriumsulfat und engt unter vermindertem Druck ein. s Durch Chromatographieren des Rückstands an Silicagel mit Chloroform als Eluiermittel erhält man 0,3 g 10-(n-Butyl)-5-phenyl-2,3,5,10-tetrahydroimidazo[2,l-bjchinazolin als hellgelbes Öl. Eine Äthanollösung des Produkts wird mit einer Lösung von Chlorwasserstoff in Äthanol behandelt und 10 unter vermindertem Druck eingeengt. Durch Umkristallisieren des Rückstands aus Äthanol erhält man das Hydro-chlorid in Form von farblosen Prismen, F. 297°C (Zers.)
Beispiel 9
15 Nach dem Verfahren von Beispiel 8 werden die folgenden Verbindungen hergestellt:
8-Chlor-10-cyclopropylmethyl-5-(p-chlorphenyl)-2,3,5,10-tetrahydroimidazo[2,1 -bjchinazolin-hydrochlorid. Isopro-panol, F. 319 bis 320°C (Zers.). 20 10-(p-Fluorbenzyl)-5-phenyl-2,3,5,10-tetrahydroimi-dazo[2,l-bjchinazolin-hydrochlorid, F. 283,5 bis 284,5°C. 8-Chlor-l l-(n-butyl)-6-phenyl-3,4,6,l l-tetrahydro-2H-pyri-mido[2,l-bjchinazolin-hydrochlorid, F. 273 bis 274°C (Zers.).

Claims (10)
- 6368762PATENTANSPRÜCHE 1. Therapeutisch wirksame Imidazo- und Pyrimido-[2,l-b]chinazoline der allgemeinen Formel:(I)in der Ri, R2 und R3 unabhängig voneinander Wasserstoffoder Halogenatome, Ci-3-Alkyl- oder Ci-3-Alkoxyreste bedeuten, Ri einen Ci-s-Alkyl, C3-s-Alkenyl-, Aralkyl- oder Cyclo-C3-s-alkyl-Ci-3-alkylrest darstellt, Rs ein Wasserstoffatom oder eine Hydroxylgruppe ist und A einen gegebenenfalls durch ein oder zwei Ci-3-Âlkylreste substituierten C2-3-Alkylenrest bedeutet, und deren pharmazeutisch verträgliche Säureadditionssalze.
- 2. Verbindungen nach Anspruch 1, dadurch gekenn- . zeichnet, dass Rs ein Wasserstoffatom ist.
- 3. Verbindungen nach Anspruch 1, dadurch gekennzeichnet, dass Rs eine Hydroxylgruppe ist.
- 4. Verbindungen nach Anspruch 1, dadurch gekennzeichnet, dass entweder Ri oder R3 ein Halogenatom, einen Ci-3-Alkyl- oder Ci-3-Alkoxyrest bedeutet.
- 5. Verbindungen nach Anspruch 4, dadurch gekennzeichnet, dass A eine Äthylen- oder Trimethylengruppe ist.
- 6. Verfahren zur Herstellung der Verbindungen nach Anspruch 2, dadurch gekennzeichnet, dass man eine Verbindung der Formel:NH-A-A-OH(V)in der Ri, R2, R3, R4 und A die in Anspruch 1 genannte Bedeutung haben und Y ein Halogenatom ist, mit einer Alkalibase in einem inerten Lösungsmittel umsetzt.
- 8. Verfahren zur Herstellung der Verbindungen nach20 Anspruch 2, dadurch gekennzeichnet, dass man eine Verbindung der Formel (I), in der Rs eine Hydroxylgruppe ist, mit einem Reduktionsmittel, das sich zur Hydrogenolyse von tertiären Alkoholen eignet, umsetzt.
- 9. Verfahren zur Herstellung der Verbindungen nach25 Anspruch 3, dadurch gekennzeichnet, dass man eine Verbindung der Formel:(VI)40 in der Ri, R2, R4 und A die in Anspruch I genannte Bedeutung haben, mit einer Organometallverbindung der Formel:
- R.(VII)(IX)50in der R3 die in Anspruch 1 genannte Bedeutung hat und Z Lithium oder MgX', wobei X' ein Halogenatom bedeutet, darstellt, in einem inerten Lösungsmittel und anschliessend mit Wasser umsetzt.55 10. Arzneimittel, enthaltend mindestens eine Verbindung nach Anspruch 1 oder ein pharmazeutisch verträgliches Säureadditionssalz derselben.60in der Ri, R2, R3, R4 und A die in Anspruch 1 genannte Bedeutung haben, mit Phosphoroxychlorid umsetzt und dann mit einer Alkalibase behandelt.
- 7. Verfahren zur Herstellung der Verbindungen nach Anspruch 2, dadurch gekennzeichnet, dass man eine Verbindung der Formel:65
