Description
Die Erfindung betrifft ein Verfahren zur Herstellung von neuen kondensierten Pyrimidinderivaten. Zum Gegenstand der Erfindung gehören ferner die Herstellung der mit Säuren oder Basen gebildeten Salze dieser Verbindungen sowie, falls es sich um optisch aktive Verbindungen handelt, deren optisch aktive Isomere.
Die erfindungsgemäss hergestellten Verbindungen weisen wertvolle pharmakologische Wirkungen auf. Insbesondere ihre antiarterosklerotische Wirkung ist hervorzuheben, jedoch wirken einzelne Vertreter der erfindungsgemässen Verbindungen auch auf das Zentralnervensystem.
Die erfindungsgemäss hergestellten Verbindungen ent-

R6 für Wasserstoff, gegebenenfalls substituiertes Aryl, gegebenenfalls substituierten Heterocyclus, Trihalogenmethyl, Carboxyl oder funktionell abgewandeltes Carboxyl steht,
R9 Wasserstoff, gegebenenfalls substituiertes Hydroxyl, gegebenenfalls substituiertes Amino, gegebenenfalls substituiertes Alkyl oder Alkylthio, gegebenenfalls substituiertes Aryl, gegebenenfalls substituiertes Aralkyl, Carboxyl oder funktionell abgewandeltes Carboxyl oder einen über das Stickstoffatom an den Pyrimidinring gebundenen stickstoffhaltigen Heterocyclus bedeutet,

Die Verbindung der allgemeinen Formel (I), ihre gegebenenfalls optisch aktiven Isomere sowie ihre Salze werden J erfindungsgemäss erhalten, indem man a) gegebenenfalls optisch aktive kondensierte Pyrimidinderivate der allgemeinen Formel (II)

worin die Bedeutung von R13 und R14 unabhängig voneinander Hydroxy oder Alkoxy ist oder R13 und R14 zusammen für =S stehen, und gewünschtenfalls die erhaltenen kon-35 densierten Pyrimidinderivate der allgemeinen Formel (I) mit physiologisch verträglichen Säuren zu ihren Säureadditionssalzen oder mit organischen oder anorganischen Basen zu ihren Salzen umsetzt und/oder gewünschtenfalls aus ihren Salzen freisetzt.
40 Beide Verfahrensvarianten, sowohl a) wie auch b), werden gegebenenfalls in einem Lösungsmittelmedium vorzugsweise bei Temperaturen zwischen —20 und 250°C ausgeführt. Als Lösungsmittel kommen vor allem protische, apolare oder dipolar-aprotische Lösungsmittel sowie deren Ge-45 mische in Frage.
Als protische Lösungsmittel werden vorzugsweise C1-6-Alkanole, wie Methanol, Äthanol, Isopropanol, Glycerin usw., ferner aliphatische Carbonsäuren wie Ameisen- oder Essigsäure, oder Formamid usw. eingesetzt. Als apolare jo Lösungsmittel eignen sich Kohlenwasserstoffe, zum Beispiel Benzol, Toluol, Xylol, chlorierte Kohlenwasserstoffe wie Chloroform oder Tetrachlorkohlenstoff, auch Chlorbenzol, ferner Äther, wie Diäthyläther, Tetrahydrofuran, Dioxan, Pyridin usw., und als dipolare Lösungsmittel können zum 55 Beispiel Dimethylformamid, Dimethylacetamid, Dimethyl-sulfoxyd, ferner Ketone wie Aceton, Äthylmethylketon usw. ferner Nitromethyn, Nitrobenzol, Acetonitril, Hexamethyl-phosphortriamid oder entsprechend gewählte Gemische der aufgeführten Lösungsmittel verwendet werden. 60 Bei beiden Verfahrensvarianten (a) und (b) können gewünschtenfalls auch basische oder saure Katalysatoren verwendet werden. Die katalytische Wirkung kann auch durch ein entsprechend ausgewähltes Lösungsmittel erzielt werden. So kann bei Säurekatalyse in den Lösungsmitteln Ameisen-65 säure oder Essigsäure gearbeitet werden. Als saure Katalysatoren kommen ferner anorganische und organische Säuren, zum Beispiel Salzsäure, Schwefelsäure, Phosphorsäure, Essigsäure usw. in Frage.

Als basische Katalysatoren können zum Beispiel Piper-idin, Diäthylamin eingesetzt werden, aber auch die Verwendung von bifunktionellen, d.h. gleichzeitig sauer und basisch katalysierenden Verbindungen, wie zum Beispiel Piperidin-acetat oder Pyridin, ist möglich.
Bei beiden Verfahrens Varianten kann die Reaktion auch so geführt werden, dass man nicht das unmittelbare Additionsprodukt (d.h. Verbindungen der allgemeinen Formel (I), in denen R2 für Hydroxyl, Mercapto oder Alkoxy steht), sondern gleich das partiell dehydrierte Produkt (Verbindungen der allgemeinen Formel (I), in denen R2 durch eine a-Doppelbindung ersetzt ist, aus dem Reaktionsgemisch isoliert.
Gewünschtenfalls kann der Substituent R6 in an sich bekannter Weise zu einem anderen Substituenten R6 umgebildet werden. Steht R6 zum Beispiel für die Carboxylgruppe, so kann diese mit dem gewünschten Alkohol verestert werden. Bei der Veresterung werden die an sich bekannten Veresterungsmethoden angewendet.
Vorzugsweise leitet man dabei in die alkoholische Lösung trockenes Salzsäuregas ein. Es kann auch so vorgegangen werden, dass man das Gemisch aus Carbonsäure und Alkohol in Gegenwart von konzentrierter Schwefelsäure erwärmt und eventuell das gebildete Wasser durch mit Benzol oder Chloroform vorgenommene azeotrope Destillation entfernt. Der Substituent R6 = Carboxylgruppe kann ferner über die Methode des aktiven Esters bei Verwendung von Triäthylamin, Chlorameisensäure und Chloroform mit Ammoniak zum Säureamid oder mit anderen Aminen zu einer N-substituierten Säureamidgruppe umgebildet werden.
Falls R6 bereits für eine Estergruppe steht, so kann diese mit einem anderen Alkolol — zweckmässig in Gegenwart von Chlorwasserstoff — umgeestert werden. Sie kann ferner mit Ammoniak zum Beispiel in alkoholischer Lösung zur Säureamidgruppe, mit Hydrazinhydrat zur CarbohydraziS-gruppe oder mit einem Amin zu einer N-substituierten Säureamidgruppe umgebildet werden.
Zur Ausbildung einer gegebenenfalls substituierten Säureamidgruppe ist es auch möglich, die Gruppe R6 = Carboxyl zuerst zum Beispiel mit Thionylchlorid, Phosphoroxychlorid oder Phosphorpentachlorid zu einer Säurehalogenidgruppe umzusetzen und auf diese Ammoniak oder das in gewünschter Weise substituierte Amin einwirken zu lassen.
Der Substituent R6 = Säureamidgruppe kann durch Wasserentzug zur Nitrilgruppe umgebildet werden. Als wasserentziehende Mittel kommen zum Beispiel Phosphoroxychlorid, Thionylchlorid oder Phosphorpentoxyd in Frage.
Der Substituent R6 = Nitrilgruppe kann mit Ammoniak zur Amidingruppe, mit Wasser zur Säureamidgruppe und mit Schwefelwasserstoff zur Thiosäureamidgruppe umgesetzt werden.
Steht R6 für Ester-, Säureamid- oder Nitrilgruppe, so können diese Gruppen durch saure oder alkalische Hydrolyse zur Carboxylgruppe umgesetzt werden. Aus dem Substituenten R6 = Carboxylgruppe kann gewünschtenfalls durch Decarboxylierung ein Wasserstoffatom gebildet werden. Zur Decarboxylierung wird die Verbindung vorzugsweise in Chinolin, Polyphosphorsäure, usw. erwärmt.
Der Substituent R9 kann gewünschtenfalls in an sich bekannter Weise zu einem anderen Substituenten R9 umgebildet werden. Steht R9 zum Beispiel für Carbonsäureester-, Säureamid- oder Nitrilgruppe, so kann auf die für den Substituenten R6 bereits beschriebene Weise vorgegangen werden.
Auch der Substituent R10 kann gewünschtenfalls in einen anderen Substituenten R10 umgebildet werden, wobei ebenfalls an sich bekannte Methoden zur Anwendung gelangen; steht R10 zum Beispiel für Carbonsäureester-, Säureamid-
oder Nitrilgruppe, so kann auf die für den Substituenten R6 bereits beschriebene Weise vorgegangen werden.
Schliesslich kann auch der Substituent R11 mit an sich bekannten Methoden zu einem anderen Substituenten Ru 5 umgebildet werden. Steht Ru zum Beispiel für Sauerstoff, so kann dieser durch in Pyridin vorgenommene Behandlung mit Phosphorpentasulfid gegen Schwefel ausgetauscht werden. Steht R11 für die Gruppe, =NH, so kann diese durch Acylie-ren mit einem Säurehalogenid oder Säureanhydrid zu einer io =N-Acyl-Gruppe umgebildet werden. Steht R11 für die Gruppe =N-R12, so kann an deren Stelle durch saure oder alkalische Hydrolyse ein Sauerstoffatom gebunden werden.
Die erhaltenen Verbindungen der allgemeinen Formel (I) können gewünschtenfalls mit physiologisch verträglichen 15 Säuren zu ihren Salzen umgesetzt werden. Als Säuren kommen zum Beispiel Salzsäure, Bromwasserstoff, Schwefelsäure, Phosphorsäure, Milchsäure, Zitronensäure, Weinsäure, Maleinsäure, Nicotinsäure, usw. in Frage.
Steht wenigstens einer der Substituenten R6, R9 oder R10 20 für Carboxyl beziehungsweise ist in wenigstens einem dieser Substituenten die Carboxylgruppe vorhanden, so können aus den betreffenden Verbindungen der allgemeinen Formel (I) mit Basen gewünschtenfalls die Salze gebildet werden. Vorzugsweise werden die Alkalisalze, insbesondere die Natrium-25 und Kaliumsalze, die Erdalkalisalze, insbesondere die Calcium- und Magnesiumsalze, ferner die mit Aluminium, Vanadium, Äthylendiamin, 2-Aminoäthanol, 2-Dimethylamino-äthanol, Diisopropylamin usw. gebildeten Salze der Verbindungen der allgemeinen Formel (I) hergestellt.
30 Unter dem in der vorliegenden Beschreibung benutzten Ausdruck «gegebenenfalls substituierte Hydroxylgruppe»
sind folgende Gruppen zu verstehen: Hydroxyl, C^-Alkoxy, vorzugsweise Methoxy, Äthoxy, ferner C,-12-Aralkoxy, vorzugsweise Benzyl, sowie C6-10-Aryloxy, vorzugsweise Phen-35 oxy.
Unter dem Ausdruck «gegebenenfalls substituierte Ami-nogruppe» sind Aminogruppen zu verstehen, die gewünschtenfalls 1 oder zwei gleiche oder verschiedene Substituenten tragen. Solche Aminogruppen sind zum Beispiel Cj-g-Alkan-40 oylamino, vorzugsweise Acetylamino oder Propionylamino, ferner C6-10-Aroylamino, vorzugsweise Benzoylamino, sowie Ci-g-Alkylamino, vorzugsweise Methylamino oder Äthyl-amino, C1-6-Dialkylamino, vorzugsweise Dimethylamino, oder Diäthylamino, C7-12-Aralkylamino, vorzugsweise Ben-45 zylamino, sowie C6-10-Arylamino, vorzugsweise Phenylamino, schliesslich Piperidyl, Pyrrolidinyl und Piperazinyl.
Unter dem Ausdruck «gewünschtenfalls substituierte Ausdruck «gegebenenfalls substituierte Alkylgruppe» werden folgende Gruppen verstanden: Alkyl mit 1-6 Kohlenstoff-J0 atomen, vorzugsweise Methyl, Äthyl, n-Propyl, usw., welche Gruppen einen oder mehrere Substituenten tragen können. Als Substituenten kommen in Frage: Hydroxyl, vorzugsweise in 1-Stellung an Äthyl gebundenes Hydroxyl, Carboxyl oder funktionell abgewandeltes Carboxyl, z.B. Alkoxy-55 carbonyl, Carboxamido oder Nitrii.
Unter dem Ausdruck «gewünschtenfalls substituierte Arylgruppe» sind gewünschtenfalls einen oder mehrere Substituenten tragende Phenylgruppen zu verstehen. Als Substituenten kommen in Frage: Alkyl mit 1-6 Kohlenstoff atomen, 60 Dialkylamino und Alkylthio mit je 1-6 Kohlenstoffatomen im Alkylteil, ferner Nitro, Methylendioxyhalogen, Carboxyl, funktionell abgewandeltes Carboxyl oder Cj-6-AIkoxy.
Der Ausdruck «gegebenenfalls substituierte Aralkyl-grappe» steht hier für Aralkylgruppen mit 7-12 Kohlen-« stoffatomen, die durch Alkyl, Alkoxy, Halogen, Amino,
Nitro einfach oder mehrfach substituiert sein können.
Unter dem Ausdruck funktionell abgewandeltes Carboxyl sind folgende Gruppen zu verstehen: Alkoxycarbonyl

mit 1-6 Kohlenstoffatomen, Aralkoxycarbonyl mit 7-12 Kohlenstoffatomen, gegebenenfalls substituiertes Aryloxycarbo-nyl mit 6-10 Kohlenstoff atomen, Alkoxy thiocarbonyl mit 1-6 Kohlenstoffatomen, gegebenenfalls substituiertes Säureamid, Säurehydrazid, gegebenenfalls substituiertes Amidin oder Nitrii.
Der Ausdruck «gegebenenfalls substituiertes Säureamid» steht für Säuramid, N-Alkyl-, N,N-Dialkyl-, N-Phenylalkyl-und N-Acylsäureamid.
Unter dem Ausdruck «gegebenenfalls substituierter Heterocyclus» sind monocyclische Ringe mit 5. 6 oder 7 Gliedern, bicyclische Ringe mit 9, 10 oder 11 Gliedern sowie tri-cyclische Ringe mit 14 Gliedern zu verstehen, die als Fremdatome Sauerstoff, Schwefel oder Stickstoff enthalten und gegebenenfalls durch C^-Alkyl oder Alkoxy, ferner durch Nitro. Halogen, Methylendioxy, Dialkylamino einfach oder mehrfach substituiert sein können. Bevorzugte Heterocyclen sind zum Beispiel Furanyl, Pyrrolyl-, Pyridyl und Chinolyl.
Die als Ausgangsstoffe zu verwendenden, gegebenenfalls optisch aktiven Verbindungen der allgemeinen Formel (II) können auf die in den ungarischen Patentschriften 156 119, 158 085, 162 384, 162 373 und 166 577 und in der holländischen Patentschrift 72 12 286 beschriebene Weise oder analog dazu hergestellt werden.
Die als Ausgangsstoffe zu verwendenden Verbindungen der allgemeinen Formeln (III) und (IV) sind handelsübliche Produkte. Als Aldehyde der allgemeinen Formel (III) können bevorzugt Benzaldehyd, Isovanillin, Vanillin, Trimeth-oxybenzaldehyd, o-Chlorbenzaldehyd, p-Chlorbenzaldehyd, Glyoxylsäuremonohydrat, Glyoxylsäure, Methylendioxybenz-aldehyd, 5-Nitro-2-furanaldehyd, 2-Thiofenaldehyd-,-Pyridin--3-aldehyd, Pyridin-2-aldehyd, Pyridin-4-aldehyd, Zimtaldehyd, o-Nitrozimtaldehyd, Fluorbenzaldehyd, Trifluor-
methylaldehyd, Methylbenzaldehyd, Furfurol, 2-Pyrrolalde-hyd, l-Methyl-2-pyrrolaldehyd, Diäthoxybenzaldehyd, Brombenzaldehyd, Hydroxybenzaldehyd, Veratrumaldehyd, Anisaldehyd, Salicylaldehyd, Dimethylaminobenzaldehyd, Nitro-5 benzaldehyd, Alkoxycarbonylbenzaldehyd, Phthalaldehyd, Terephthalaldéhyd, Formaldehyd, Chloral, Bromal usw. eingesetzt werden.
Als Aldehydderivate der allgemeinen Formel (IV) kommen zum Beispiel die Halbacetale, Acetale, Acylale, geminalo len Diamine und Aminoalkohole, Schiff'sehen Basen, gemi-nalen Glycole, Aldehydnatriumbisulfite, Aldehydcyanhydride der entsprechenden Aldehyde in Frage.
Die Verbindungen der allgemeinen Formel (I) weisen wertvolle pharmakologische Eigenschaften, insbesondere eine 15 bedeutende antiarterosklerotische Wirkung auf, wobei nicht nur die Menge der Lipide im Serum, sondern auch die Menge der an den Blutgefässwänden abgelagerten Lipide, in erster Linie die des Cholesterins, vermindert wird.
Die Verbindungen der allgemeinen Formel (I) haben 20 günstige Toxizitätswerte. Die LD50-Werte, gemessen im Tierexperiment an Ratten bei per os Applikation, liegen im allgemeinen über 2000 mg/kg.
Die günstigen pharmakologischen Eigenschaften der er-findungsgemässen Verbindungen wurden in verschiedenen 25 Experimenten geprüft. In der Tabelle I sind die Ergebnisse zusammengefasst, die mit dem Cholesterinfütterungstest an Hasen (Beitr. path. Anat. 56, 379-403 [1913]) erhalten wurden.
Aus der Tabelle 1 geht hervor, dass die Verbindungen C 30 und D den Lipidspiegel des Serums sowie den Lipid- beziehungsweise Cholesteringehalt der Aorta wesentlich stärker senken als Clofibrat.
Die mathematisch-statistischen Daten sind aus Gründen der Übersichtlichkeit in der Tabelle nicht angegeben.

Die Kontrollgruppe 1 wurde normal gefüttert, während die Kontrollgruppe 2 neben dem normalen Futter noch 2 g Cholesterin erhielten.
Im Gegensatz zum Clofibrat riefen die Verbindungen bei dem Experiment keinen Gewichtszusatz der Leber hervor.
Die Ergebnisse der mit normolipaemischen Ratten durchgeführten Versuche sind in Tabelle 2 zusammengestellt.

Andere Vertreter der Verbindungen wirken auf das Zentralnervensystem. So zeigen zum Beispiel manche Verbindungsgruppen eine bedeutende analgetische, antipyretische, entzündungshemmende Wirkung, tranquillante Narkose-potenzierung, ferner leberprotektive, antidepressive, antibakterielle oder antituberkulotische Wirkung beziehungsweise Wirkung gegen Asthma.
Die erfindungsgemäss hergestellten Verbindungen können in erster Linie als Arzneimittel Verwendung finden, sie können jedoch auch als Ausgangsstoffe zur Herstellung anderer pharmazeutisch wirksamer Stoffe dienen.
Die Verbindungen der allgemeinen Formel (I) können an sich oder in Form von inerte, nichttoxische feste oder flüssige Streck- und Trägerstoffe enthaltenden Präparaten angewendet werden. Die Präparate können fest sein (zum Beispiel Tabletten, Kapseln, Dragees, Mikrokapseln) oder in flüssiger Form vorliegen (zum Beispiel Lösungen, Suspensionen, Emulsionen).
Die zu verabreichende Dosis hängt vom Anwendungsgebiet und der Art der Formulierung ab. Im allgemeinen werden Präparate mit 1-1000 mg Wirkstoff hergestellt.
Als Trägerstoffe können die für diesen Zweck üblichen Substanzen, zum Beispiel Talkum, Calciumcarbonat, Magne-siumstearat, Wasser, Polyäthylenglycolat usw. verwendet werden. Die Präparate können gewünschtenfalls auch die üblichen Hilfs- und Zusatzstoffe, zum Beispiel Emulgatoren, Sprengmittel usw., enthalten. Die Präparate können gewünschtenfalls auch als Retardpräparate hergestellt werden
Beispiel 1
Ein Gemisch aus 23,6 g 3-Äthoxycarbonyl-6-methyl-4--oxo-6,7,8,9-tetrahydro-4H-pyrido[l,2-a]pyrimidin und 10,6 g Benzaldehyd wird bei 40°C gerührt, dann mit 10 ml 5 Äthanol versetzt und über Nacht stehen gelassen. Die ausgefallenen Kristalle werden abfiltriert. Man erhält weisses 3-Äthoxycarbonyl-9-(l'-hydroxybenzyl)-6-methyl-4-oxo--6,7,8,9-tetrahydro-4H-pyrido[2,3-a]pyrimidin, das nach Umkristallisieren aus Äthanol bei 186-187°C schmilzt. 10 Analyse:

Beispiel 2
15 H8,0g 3-Äthoxycarbonyl-6-methyl-4-oxo-6,7,8,9-tetra-hydro-4H-pyrido[l,2-a]pyrimidin werden unter schwacher Aussenkühlung bei einer Temperatur unter 40°C mit 73,7 g Chloral (oder 82,7 g Chloralhydrat) umgesetzt. Das Gemisch wird einen Tag lang stehen gelassen und dann aus 20 Äthanol umkristallisiert. Man erhält 3-Äthoxycarbonyl-9-(l--hydroxyl-2,2,2-trichloräthyl)-6-methyl-4-oxo-6,7,8,9-tetra-hydro-4H-pyrido[l,2-a]pyrimidin, das bei 156°C schmilzt. Analyse:

Beispiel 3
34,2 g 3-Äthoxycarbonyl-9-(l'-hydroxybenzyl)-6-4-oxo--6,7,8,9-tetrahydro-4H-pyrido[l,2-a]pyrimidin werden in 30 einem Gemisch aus 100 ml Äthanol und 10 ml 15 Gew.-%-iger äthanolischer Salzsäure gekocht. Nach dem Abkühlen werden die ausgeschiedenen gelben Kristalle abfiltriert und aus Äthanol umkristallisiert. Das erhaltene 3-Äthoxycarbo-nyl-9-benzyliden-6-methyl-4-oxo-6,7,8,9-tetrahydro-4H-35 -pyrido[l,2-a]pyrimidin schmilzt bei 140-141°C und zeigt mit dem Produkt gemäss Beispiel 4 keine Schmelzpunktsdepression.
Beispiel 4
40 23,6 g 3-Äthoxycarbonyl-6-methyl-4-oxo-6,7,8,9-tetra-hydro-4H-pyrido[l,2-a]pyrimidin werden in einem Gemisch aus 10 ml Äthanol und 5 ml 15 Gew.-%iger äthanolischer Salzsäure mit 10,6 g Benzaldehyd umgesetzt. Das entstandene 3-Äthoxycarbonyl-9-(r-hydroxylbenzyl)-6-methyl-4-45 -oxo-6,7,8,9-tetrahydro-4H-pyrido[l,2-a]pyrimidin wird ohne Isolierung unter Rühren und Erwärmen durch Wasserentzug zu 3-Äthoxycarbonyl-9-benzyliden-6-methyl-4-oxo-6,7,8,9--tetrahydro-4H-pyrido[l,2-a]pyrimidin umgesetzt. Dieses bildet gelbe Kristalle, die nach Umkristallisieren aus Ätha-50 noi bei 140-141°C schmelzen. Ausbeute: 70%.

35,6 g 3,6-Dimethyl-4-oxo-6,7,8,9-tetrahydro-4H-pyrido-[l,2-a]pyrimidin und 28,2 g 5-Nitrofuran-2-aldehyd werden miteinander umgesetzt und das erhaltene 3,6-Dimethyl-9--(5'-nitro-r-hydroxyl-2-furfuryl)-4-oxo-6,7,8,9-tetrahydro--4H-pyrido[l,2-a]pyrimidin ohne -Isolierung durch Kochen in einem Gemisch aus 60 ml Benzol und 0,2 ml Essigsäure unter Wasseraustritt zu 3,6-Dimethyl-9-(5-nitro-2-furfuryl-iden)-4-oxo-6,7,8,9-tetrahydro-4H-pyrido [ 1,2-a] pyrimidin umgesetzt. Das Reaktionsgemisch wird auf 10°C gekühlt.
Die ausgeschiedenen Kristalle werden abfiltriert und aus Äthanol umkristallisiert. In 46,5 %iger Ausbeute wird 3,6--Dimethyl-9-(5-nitro-2-furfuryliden)-4-oxo-6,7,8,9-tetrahydro--4H-pyrido[l,2-a]pyrimidin erhalten, das bei 183-184°C 5 schmilzt.

55,6 g 3-Äthoxycarbonyl-9-carboxymethylen-4-oxo-6,7,-8,9-tetrahydro-4H-pyrido[l,2-a] pyrimidin werden in 500 ml Äthanol in Gegenwart von 20 g 9%iger Palladiumkohle hydriert. Nach Aufnahme der äquivalenten Menge Wasser-is stoff wird der Katalysator durch Filtrieren aus dem Reaktionsgemisch entfernt. Das Filtrat wird unter vermindertem Druck eingedampft und der Rückstand aus Äthanol umkristallisiert. Auf diese Weise wird in 57%iger Ausbeute 3-Äthoxycarbonyl-4-oxo-6,7,8,9-tetrahydro-4H-pyrido [ 1,2-a] -m pyrimidin-9-essigsäure erhalten, die bei 156-157°C schmilzt. Analyse:

Die erhaltene 3-Äthoxy-carbonyl-4-oxo-l,6,7,8,9,9a-hexahydro-4H-pyrido[l,2-a]py-rimidin-9-essigsäure schmilzt bei 205°C.

Beispiel 86
0,65 g Magnesiumspäne werden in Gegenwart von 0,1 g Jod als Katalysator mit 20 ml Äthanol umgesetzt. Zu dem erhaltenen Magnesiumäthylat werden 300 ml Äthanol und 14,7 g 3-Äthoxycarbonyl-6-methyl-4-oxo-6,7,8,9-tetrahydro--4H-pyrido[l,2-a]pyrimidin-9-essigsäure gegeben. Das Reaktionsgemisch wird zum Sieden erhitzt und dann filtriert. Auf diese Weise werden 13,0 g rohes, langsam kristallisierendes Magnesium-3-äthoxycarbonyl-6-methyl-4-oxo-6,7,8,9--tetrahydro-4H-pyrido[ 1,2-a] pyrimidin-9-acetat erhalten, welches nach Umkristallisieren aus einem Äthanol-Äthyl-acetat-Gemisch bei über 360°C unter Zersetzung schmilzt. 50 g 3-(Äthoxycarbonylmethyl)-6-methyl-4-oxo-6,7,8,9--tetrahydro-4H-pyrido[l,2-a]pyrimidin und 28,2 g 5-Nitro--2-furfurol werden miteinander umgesetzt, und das erhaltene 3-(Äthoxycarbonylmethyl)-6-methyl-9-[(5-nitro-2-furyl)-hy-droxymethyl]-4-oxo-6,7,8,9-tetrahydro-4H-pyrido[l,2-a]-pyrimidin wird ohne Isolieren in einem Gemisch aus 600 ml Benzol und 2 ml 16 Gew.-%iger äthanolischer Salzsäure unter einem Wasserabscheideaufsatz 3 Stunden lang gekocht. Anschliessend wird das Reaktionsgemisch unter vermindertem Druck eingedampft. Der erhaltene Rückstand wird aus äthanolischer Salzsäure zweimal umkristallisiert. In 73%iger Ausbeute wird 3-(Äthoxycarbonylmethyl)-6-methyI-9-(5-ni-tro-2-furfuryliden)4-oxo-6,7,8,9-tetrahydro-4H-pyrido[l,2-a]-pyrimidin-hydrochlorid erhalten, das bei 191-192°C schmilzt. Analyse:

Nach dem Beispiel 75 arbeitend wird aus 3-(Äthoxycar-bonyl-methyl)-6-methyl-9-(carboxy-methylen)-4-oxo-6,7,8,9--tetrahydro-4H-pyrido[l,2-a]pyrimidin mit einer Ausbeute 5 von 80% das 3-(Äthoxycarbonyl-methyl)-9-(äthoxycarbonyl--methyl)-4-oxo-6,7,8,9-4H-pyrido[l,2-a]pyrimidin, in Form eines nicht kristallisierenden blassgelben Öls erhalten. Beispiel 91
1,1 g 6-Methyl-3-(N-methyl-imino-methyl)-2-hydroxy-4--oxo-6,7,8,9-tetrahydro-4H-pyrido[l,2-a]pyrimidin und 1,06 g Benzaldehyd werden in 20 ml Benzol 90 Minuten 30 lang zum Sieden erhitzt. Nach Abkühlen werden die ausgeschiedenen Kristalle filtriert. Nach Einengen der Futterlauge werden weitere Kristalle erhalten. Insgesamt werden 1,15 g [74%] 9-Benzyliden-6-methyl-3-(N-methyl-imino--methyl)-2-hydroxy-4-oxo-6,7,8,9-tetrahydro-4H-pyrido-35 [l,2-a]pyrimidin erhalten, welches nach Umkristallisieren aus Äthanol bei 253-255°C schmilzt.

- 2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man die Umsetzung nach den Verfahrensvarianten a)20 und b) bei —20 bis +400°C vornimmt.3. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man die Umsetzung nach den Verfahrensvarianten a) und b) in Gegenwart eines Lösungsmittels, insbesondere eines aprotischen Lösungsmittels, eines protischen Lösungsmittels25 oder eines aprotischdipolaren Lösunsmittels, vornimmt.4. Verfahren nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass man als Aldehyd der allgemeinen Formel (III) Benzaldehyd, Zimtaldehyd, Formaldehyd, Chloral, Glyoxylsäure, 5-Nitrofuran-2-aldehyd, N-Methylpyrrol-2-al-30 dehyd, Pyridin-2-aldehyd, Pyrrol-2-aldehyd, Furfurol, 3,4,5-Trimethoxybenzaldehyd, Isovanillin, Vanillin, o-Nitrozimt-aldehyd, Pyridin-3-aldehyd, Piperonal, 4-Hydroxybenzalde-hyd, 4-Chlorbenzaldehyd, Veratrumaldehyd, Anisaldehyd, Salicylaldehyd, 2-Nitrobenzaldehyd, 3-Chlorbenzaldehyd, 35 3-Nitrobenzaldehyd, 2-Methoxybenzaldehyd, 4-Nitrobenzal-dehyd, 3-Hydroxybenzaldehyd, 3-Methoxybenzaldehyd, 2-Äthoxybenzaldehyd, 3-Fluorbenzaldehyd, 4-Methylbenzalde-hyd, 3-Cyanobenzaldehyd, 5-Bromsalicylaldehyd, 4-Benzyl-oxy-3-methoxybenzaldehyd, 4-Dimethylaminosalicylaldehyd, 40 3,4-Dichlorbenzaldehyd, Alkoxycarbonylbenzaldehyde, 4-Acetaminobenzaldehyd oder Phthalaldehyd verwendet5. Salzsäure, Schwefelsäure, Phosphorsäure oder Essig-50 säure, oder eines basischen Katalysators, wie z.B. Piperidin oder Diäthylamin, vorgenommen wird.7. Verfahren nach Anspruch 6, dadurch gekennzeichnet, dass als Katalysator Piperidin-Acetat oder Pyridin verwendet wird.55 8. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass als Ausgangsmaterial optisch aktives 3-Äthoxycarbonyl--6-methyl-4-oxo-6,7,8,9-tetrahydro-4H-pyrido-[l,2-a]pyrimi-din verwendet wird.9. Verwendung der Verbindungen der allgemeinen For-60 mei I, worin R2 eine Hydroxylgruppe bedeutet und die anderen Symbole im Anspruch 1 definiert sind, für die Herstellung der entsprechenden Verbindungen in der R2 für Wasserstoff steht, durch Abspaltung eines Moleküls Wasser und Reduktion der so gebildeten Äthylengruppe.10. Verwendung nach Ansprach 9, dadurch gekennzeichnet, dass die Reduktion durch Hydrierung in Gegenwart von Palladium Katalysator durchgeführt wird.11.
