Description
Gegenstand der Erfindung sind neue substituierte 4-Aryl--2,5-dihydrofuran-2-one und 4-Aryl-l,5-dihydro-2H-pyrrol--2-one der Formel (I)
worin R1, R2, R3, R4, R5 und A die im Anspruch 1 angegebene Bedeutung haben.

Ferner betrifft die Erfindung neue substituierte 4-Aryl-2,5-dihydro-furan-2-one und 4-Aryl-l,5--dihydTO-2H-pyrrol-2-one der Formel IV und V

0-ch--ch-ch oh worin R1; R2 und A die vorgenannte Bedeutung haben und worin X für ein Chlor- oder Bromatom steht, als Mittel zur Durchführung des erfindungsgemässen Verfahrens für die Herstellung der 4-Aryl-2,5-dihydro-furan-2-one und 4-Aryl--l,5-dihydro-2H-pyrrol-2-one der Formel (I).
Die erfindungsgemässen Verbindungen der allgemeinen Formel (I) und ihre Säureadditionssalze besitzen wertvolle therapeutische Eigenschaften. Sie zeichnen sich durch ß-adre-nolytische und blutdrucksenkende Wirkung aus und können daher zur Behandlung von Angina pectoris, Hypertonie und Arrhytmien eingesetzt werden. Die Dosierung beträgt beim Menschen etwa 15-200 mg pro Tag.
Die Wirksamkeit der neuen Verbindungen ist u.a. von der Stellung der 3-Alkylamino-2-hydroxy-l-propoxy-Seiten-kette abhängig. Besonders günstig haben sich solche Verbindungen erwiesen, in denen diese Seitenkette des Phenyl-rests in ortho-Stellung zum 2,5-Dihydrofuran-2-on Ring steht.
Die Verbindungen können als solche oder in Form ihrer Säureadditionssalze, gegebenenfalls mit geeigneten festen oder flüssigen pharmakologisch verträglichen Trägerstoffen und/oder Verdünnungsmitteln gebräuchlicher Art vermischt, zur Herstellung von Lösungen für Injektionszwek-ke und insbesondere von peroral zu verabreichenden pharmazeutischen Präparaten, wie Dragées, Tabletten oder Liquida, verwendet werden. Geeignete Trägerstoffe sind beispielsweise Milchzucker (Lactose), Gelatine, Maisstärke, Stearinsäure, Äthanol, Propylenglycol, Äther des Tetrahy-drofurfurylalkohols und Wasser.
Das erfindüngsgemässe Verfahren zur Herstellung der Verbindungen der Formel (I) ist dadurch gekennzeichnet,
dass man die neuen Verbindungen der Formeln (IV) oder

(V), worin R1, R2, A und X die obengenannte Bedeutung haben, mit einem Amin der Formel (VI)

in der R3, R4 und R6 die obengenannte Bedeutung haben, umsetzt, indem man zweckmässig eine Lösung der Verbindungen (IV) bzw. (V) in einem Alkohol, bevorzugt t-Bu-tanol, mit einem Überschuss des Amins bei erhöhter Temperatur behandelt und gegebenenfalls die freie Basen mit Säuren in die entsprechenden Säureadditionssalze umwandelt. Geeignete Säuren sind beispielsweise Chlor- oder Brom-wasserstoff-, Schwefel-, Oxal-, Fumar- oder Maleinsäure.
Als Amin der Formel VI wird bevorzugt Isopropylamin oder tert.-Butylamin verwendet.
Als Ausgangsverbindungen zur Herstellung der Verbindungen (IV) und (V) sind insbesondere die neuen Verbin dungen der Formel (III)

Ein bevorzugtes Verfahren zur Herstellung von Verbindungen der Formel (III), in der A für ein Sauerstoffatom steht und R1 und R2 die angegebene Bedeutung haben und R7 Wasserstoff, Methyl oder Benzyl bedeutet, wird durchgeführt, indem man ein 2-Halogen-acetophenon der Formel (VII), mit der angegebenen Bedeutung von R1, R2 und R7,
worin X ein Chlor- oder Bromatom ist, mit dem Alkali-Salz einer 2-Dialkylphosphono-essigsäure zu a-Ketoacylestern der allgemeinen Formel (VIII) umsetzt,

und dieses zunächst einer Ätherspaltung und Hydrolyse des Esters zur Abspaltung der Reste R7 und R10 unterworfen und anschliessend gegebenenfalls bei höherer Temperatur decarboxyliert, soweit nicht bereits eine spontane Decar-boxylierung im letzten Reaktionsschritt erfolgte.
Die Umsetzung zu den Malonsäurediestern (X) erfolgt vorzugsweise mit dem Kaliumsalz des Malonsäuremono-äthylesters bei Raumtemperatur. Sie lassen sich mit wässri-ger Natronlauge bei niedriger Temperatur zu den Buteno-liden XI cyclisieren. Anschliessend werden die Äther- und die Estergruppe durch Behandlung mit Jodwasserstoffsäure unter Erhitzen abgespalten. Soweit die gebildete freie Carbonsäure nicht bereits in diesem Reaktionsschritt decarboxyliert, ist eine nachfolgende thermische Decarboxylierung, beispielsweise in Diphenyläther bei 160°C erforderlich, um zu den genannten Verbindungen der Formel III zu gelangen.
Bevorzugte Ausgangsverbindungen sind solche 2-Halo-genacetophenone der allgemeinen Formel (VII), bei denen RT eine Methyl- oder Benzylgruppe bedeutet. Die Ausgangsverbindungen der allgemeinen Formel (VII) sind nach bekannten, in der Literatur beschriebenen Verfahren zugänglich.
Vorzugsweise verwendet man das Kaliumsalz der 2-Di-äthylphosphono-essigsäure, mit dem sich die Umsetzung bei Raumtemperatur durchführen lässt. Die a-Ketoacylester (VIII) lassen sich entweder mit Kalium-tert. butylat in Di-methoxyäthan bei niedrigen Temperaturen oder vorteilhafter durch Phasentransferkatalyse beispielsweise mit Benzyl-tributylammoniumbromid oder Tetrabutylammoniumbroihid zum Butenolid cyclisieren.
Verbindungen der Formel (III), in der A eine Imino-oder Alkyliminogruppe bedeutet, können durch Umsetzung von Verbindungen der Formel (III), in denen A für ein Sauerstoffatom steht, mit Ammoniak oder Aminen der allgemeinen Formel R9-NH2, wobei R9 eine Alkylgruppe mit 1 bis 4 Kohlenstoffatomen bedeutet, gewonnen werden. Dabei entstehen zunächst Verbindungen der allgemeinen Formel (IX)

die durch Erhitzen in einem inerten Lösungsmittel wie beispielsweise Toluol oder Chlorbenzol, gegebenenfalls in Anwesenheit eines Katalysators, beispielsweise p-Toluolsulfon-säure, durch Dehydratisierung in die 3-Pyrrolin-2-one der Formel (III) übergeführt werden.
Die Methyläther gemäss der allgemeinen Formel (III) werden vorzugsweise durch Erhitzen mit Bromwasserstoff/ Eisessig gespalten, während sich die Benzyläther schon bei Raumtemperatur mit Bromwasserstoff/Eisessig oder auch durch Hydrierung mit Wasserstoff in Gegenwart von Edelmetallkatalysatoren wie Palladium/Aktivkohle in die Phe-noleumwandeln lassen. Mit der säurekatalysierten Ätherspaltung erhält man aus Verbindungen mit Cyanomethyl-Substituenten der Phenylgruppe erfindüngsgemässe Verbindungen der allgemeinen Formel(I), in denen R1 bzw. R2 die Carbamoylmethylgruppe bedeuten.
Die Phenole der Formel (III) (R7 = H) lassen sich mit Epichlor- oder Epibromhydrin zu den Verbindungen der allgemeinen Formeln (IV) und (V) umsetzen, die als Gemische anfallen. Eine Trennung der Gemische ist durch Säu-lenchromatografie möglich, aber meist nicht erforderlich. Die Reaktion erfolgt in einem Überschuss des Epihalogen-hydrins in Gegenwart einer katalytischen Menge einer organischen Base bei Temperaturen von etwa 100°C.
Die zur Umsetzung mit den Verbindungen der allgemeinen Formeln (IV) und (V) geeigneten Amine der allgemeinen Formel (VI) sind in der Literatur beschrieben oder nach bekannten Methoden zu erhalten.
Die folgenden Beispiele dienen zur Erläuterung der Erfindung:
Beispiel 1
4-[2-(2-Hydroxy-3-tert.butylamino-propoxy)-4-chlor-phenyl]--2.5-dihydro-furan-2-on
6,8 g 4-(2-Hydroxy-4-chlor-phenyl)-2,5-dihydro-furan-2--on werden mit 70 ml Epibromhydrin und 2 Tropfen Pipe-ridin 2 Stunden auf 100°C erwärmt. Nach Abdampfen des überschüssigen Bromhydrins wird das Gemisch aus Epoxi-und Bromhydrin-Verbindung durch Hochdruckflüssigchro-matografie getrennt. Die Epoxi-Verbindung kristallisiert nach Anreiben mit Diisopropyläther.
Ausbeute: 3,0 g 4-[2-(2,3-Epoxi-propoxy)-4-chlor-phe-nyl]-2,5-dihydro-furan-2-on. Fp. 155-57°C.
Eine Lösung von 3,0 g 4-f2-(2,3-Epoxi-propoxy)-4-chIor-phenyl]-2,5-dihydro-furan-2-on und 20 ml tert. Butylamin

in 100 ml tert. Butanol und 50 ml Methylenchlorid wird 5 Tage bei Raumtemperatur stehengelassen, dann eingeengt. Der Rückstand wird zwischen Methylenchlorid und wässri-ger, verdünnter Salzsäure verteilt, die wässrige Schicht mit Natriumhydrogencarbonat alkalisch gestellt und mit CH2C12 extrahiert. Die CH2C12-Phase wird abgetrennt, eingedampft, der Rückstand in Essigester aufgenommen und mit äthanol. HCl versetzt. Nach Umkristallisation aus Äthanol/tert. Butanol erhält man 2,4 g 4-[2-(2-Hydroxy-3-tert.-butylamino--propoxy)-4-chlor-phenyl]-2,5-dihydro-furan-2-on als Hydro-chlorid.

Beispiel 2
4-[2-(2-Hydroxy-3-isopropylamino-propoxy}-phenyl]--1,5-dihydro-2H-pyrrol-2-on s 21,5 g 4-(2-Hydroxy-phenyl)-l,5-dihydro-2H-pyrrol-2-on werden mit 150 ml Epibromhydrin und einigen Tropfen Piperidin 3,5 Stunden auf 75°C erwärmt. Nach Säulenchro-matografie an Kieselgel erhält man 4,5 g Epoxi-Verbindung und 13 g Bromhydrin-Verbindung als Öle.
io 2,5 g des Bromhydrins werden in 30 ml tert. Butanol mit 30 ml Isopropylamin 15 Stunden bei Raumtemperatur gerührt. Danach wird im Vakuum eingedampft und der Rückstand nach Ansäuern mit 2n HCl mit Methylenchlorid extrahiert. Nach Alkalisieren der wässrigen Phase wird er-i5 neut mit Methylenchlorid ausgeschüttelt und die CH2C12-Phase nach Ansäuern mit äthanol. HCl eingeengt. Nach Zugabe von wenig Äthanol kristallisieren 1,76 g des Hydro-chlorids von 4-[2-(2-Hydroxy-3-isopropylamino-propoxy)--phenyl] -1,5-dihydro-2H-pyrrol-2-on aus. 20 Fp. 224-27°C (Z.).

Die als Ausgangsverbindungen verwendeten Phenole können wie in Beispiel 25 bis 28 beschrieben, hergestellt werden:
Beispiel 25
4-(2-Hydroxy-4-chlor-phenyl)-2,5-dihydro-furan-2-on a) 76,7 g (0,45 mol) 2-Hydroxy-4-chlor-acetophenon und 88,9 g (0,52 mol) Benzylbromid werden in 500 ml Äthanol gelöst und bei Siedetemperatur 29,6 g (0,45 mol) KOH (85%ig) in ca. 11 Äthanol zugetropft. Nach 2 Stunden Riick-fluss wird eingedämpft, der Rückstand mit Wasser versetzt und mit Methylenchlorid extrahiert. Die organische Phase wird mehrfach mit 2n NaOH und dann mit Wasser ausgeschüttelt. Nach Abdampfen des Lösungsmittels erhält man 100 g des Benzyläthers als Öl, aus dem nach Verreiben mit n-Hexan 75 g 2-Benzyloxy-4-chlor-acetophenon vom Fp. 64°C auskristallisieren.
b) Zu 68 g (0,26 mol) 2-Benzyloxy-4-chlor-acetophenon in 700 ml Methanol werden bei 0-10°C langsam 41,7 g (0,26 mol) Brom zugetropft und anschliessend wird noch 15 Stunden bei Raumtemperatur gerührt. Dabei kristallisiert das w-Bromacetophenon zusammen mit dem Ketal dieser Verbindung aus. Zur Spaltung des Ketals wird die Substanz in 300 ml Dioxan gelöst und eine Stunde bei 40°C mit 300 ml 6n HCl behandelt. Nach Filtration erhält man 77,8 g 2-Benzyloxy-4-chIor-phenacylbromid.
Fp. 75-78°C.
c) Eine Lösung von 40,5 g (0,12 mol) 2-Benzyloxy-4--chlorphenacylbromid in 400 ml Acetonitril werden mit 33,7 g (0,145 mol) des Kaliumsalzes der 2-Diäthylphospho-no-essigsäure 15 Stunden bei Raumtemperatur gerührt. Anschliessend saugt man das Kaliumbromid ab und entfernt das Lösungsmittel. Dann nimmt man den Rückstand in Methylenchlorid auf, schüttelt mit Wasser aus und engt im Vakuum ein. Man erhält 52,5 g rohen a-Ketol-phosphono-essig-ester, der nach Anreiben mit Diisopropyläther kristallisiert.
Ausbeute: 45,4 g
Fp. 84-85°C.
d) 45,4 g (0,1 mol) des obigen Phosphonesters werden zusammen mit 3,25 g (0,01 mol) Tetrabutylammonium--bromid in 350 ml Methylenchlorid gelöst und in einer Zweiphasenreaktion mit 7,9 g (0,12 mol) KOH (85% ig) in 250 ml Wasser eine Stunde bei 20°C kräftig gerührt. Die organische Phase wird mit Wasser neutral gewaschen und eingedampft. Durch Verreiben mit Diisopropyläther erhält man 28 g 4-(2-Benzyloxy-4-chlor-phenyl)-2,5-dihydro-fu-ran-2-on.

e) 17 g des obigen Benzyläthers werden in 80 ml 40%-iger Bromwasserstoffsäure in Eisessig eine Stunde bei Raumtemperatur gerührt. Nach Eindampfen wird der Rückstand mit Äther/Diisopropyläther verrieben und abgesaugt. 30 Ausbeute: 7,5 g 4-(2-Hydroxy-4-chlor-phenyl)-2,5-dihy-dro-furan-2-on.

8,5 g 4-(2-Benzyloxy-5-acetamido-phenyl)-2,5-dihydro--furan-2-on werden in 300 ml Äthanol/Essigester (1:1) mit Wasserstoff unter leichtem Überdruck bei Raumtemperatur 40 hydriert (Katalysator 10% Pd-C).
Nach üblicher Aufarbeitung erhält man 5,3 g 4-(2-Hy-droxy-5-acetamido-phenyl)-2,5-dihydro-furan-2-on.

4-(2-Hydroxy-5-carbamoylmethyl-phenyl)-2,5-dihydro--furan-2-on
4,4 g 4-(2-Benzyloxy-5-cyanomethyl-phenyl)-2,5-dihy-50 dro-furan-2-on werden in 30 ml HBr-Eisessig (40%) bis zur vollständigen Lösung der Substanz gerührt. Nach Abdampfen des Eisessigs wird der Rückstand mit Essigester/Äther verrieben und abgesaugt. Ausbeute: 3,2 g. Zur Entfernung von Bromwasserstoffsäure wird die Substanz in gesättigter ss Natriumhydrogencarbonat-Lösung suspendiert und nach Filtration mit Äthanol gewaschen.
Ausbeute: 1,6 g 4-(2-Hydroxy-5-carbamoylmethyl-phe-nyl)-2,5-dihydro-furan-2-on.

4-(2-Hydroxy-phenyl)-l,5-dihydro-2H-pyrrol-2-on
22,0 g 4-(2-Benzyloxy-phenyl)-2,5-dihydro-furan-2-on 65 werden in einem Autoklaven mit 300 ml flüssigem NH3 3 Stunden auf 100°C erhitzt. Nach Abdampfen des NH3 nimmt man den öligen Rückstand in Isopropanol auf und saugt 15 g eines kristallinen. Niederschlags ab. Fp. 163-66°C.

Aufgrund der NMR-spektroskopischen Daten handelt es sich um 3-Hydroxy-4-(2-benzyloxyphen.yl)-pyrrolidin-2-on (Formel IX}.
14 g dieser Verbindung werden durch halbstündiges Erhitzen in 50 ml Toluol unter Zusatz katalytischer Mengen p-Toluolsulfonsäure dehydratisiert. Nach Erkalten der Lösung kristallisieren 8,5 g 4-(2-BenzyIoxy-phenyI)-1,5-dihy-dro-2H-pyrrol-2-on aus.
Fp. 153-54°C.
8,0 g des Benzyläthers werden in 40 ml HBr (40%ig) in Eisessig suspendiert und bei Raumtemperatur gerührt. Nach ca. 15 Minuten ist eine klare Lösung entstanden, aus der sich das Phenol langsam abscheidet. Das Lösungsmittel wird abgedampft, der Rückstand mit Toluol versetzt und abgesaugt. Das Produkt wird anschliessend 5 Minuten in 50 ml Eiswasser gerührt, abgesaugt und getrocknet.
Ausbeute: 4,7 g 4-(2-Hydroxy-phenyl)-l,5-dihydro-2H--pyrrol-2-on.

Man erhält 220 grohen a-Keton-phosphono-essigester, der 5 ohne weitere Reinigung in 1 1 1,2-Dimethoxyäthan mit 65 g Kalium-tert.-butylat bei 0-10°C versetzt wird. Nach etwa einer Stunde ist die Reaktion beendet. Man säuert mit verdünnter Salzsäure an, giesst auf Wasser und trennt die organische Phase ab; nach Abdampfen des Lösungsmittels 10 bleiben 200 g des Butenolids als Öl zurück, das nach Verreiben mit Äther kristallin wird. Man erhält 4-(2-Benzyl-oxy-phenyl)-2,5-dihydrofuran-2-on (81,4 g).

b) 58,0 g 4-(2-Benzyloxy-phenyl)-2,5-dihydrofuran-2-on I5 (a) werden in 200 ml Methanol mit Wasserstoff hydriert
(Katalysator 10% Pd-C). Nach üblicher Aufarbeitung erhält man 30 g 4-(2-Hydroxyphenyl)-2,5-dihydrofuran-2-on.

c) 8,0 g 4-(2-Hydroxyphenyl)-2,5-dihydrofuran-2-on
2o werden in 80 ml Epibromhydrin mit einigen Tropfen Pipe-ridin 3 Stunden auf 70°C erwärmt. Nach Abdampfen des überschüssigen Bromhydrins wird der Rückstand in Chloroform aufgenommen und mit Wasser gewaschen. Durch Einengen erhält man ein dunkles Öl, bei dem es sich um ein 25 Gemisch von Epoxid und Bromhydrin-Verbindung handelt. Durch Säulenchromatografie an Kieselgel lassen sich daraus 4,5 g des Bromhydrins als Öl abtrennen, das in Methanol gelöst und nach Zusatz von 30 ml tert. Butylamin 19 Stunden bei Raumtemperatur gerührt wird. Danach wird im 30 Vakuum eingedampft und der Rückstand nach Ansäuren mit verdünnter Salzsäure mit Methylenchlorid extrahiert. Die salzsaure Phase wird dann alkalisiert und mit Methylenchlorid ausgeschüttelt. Man erhält nach Abdampfen des Lösungsmittels und Umkristallisation aus Benzol/Hexan 35 3,1 g 4-[2-(2-Hydroxy-3-tert. butylamino-propoxy)-phenyl]--2,5-dihydrofuran-2-on.

55 a) 40,0 g (0,18 M) m-Methoxyphenacylbromid und 61,2 g (0,36 M) Kalium-monoäthylmalonat werden in 500 ml Acetonitril 6 Stunden bei Raumtemperatur gerührt. Nach Abdampfen des Lösungsmittels erhält man ein bräunliches Öl, das in 300 ml Methylenchlorid aufgenommen wird und so nach Filtration des Kaliumbromids mit 90 ml 2n NaOH 15 Minuten lang bei + 10°C behandelt wird. Nach vorsichtigem Ansäuern mit 2n HCl wird die organische Phase abgetrennt, mit Wasser gewaschen und eingedampft. Als Rückstand verbleiben etwa 50 g 3-Carbäthoxy-4-(3-methoxyphe-65 nyl)-2,5-dihydrofuran-2-on die mit 300 ml Jodwasserstoffsäure (57% ig) 4 Stunden unter Rückfluss erhitzt werden. Anschliessend wurde die Lösung eingedampft, der gelbe Rückstand mit Wasser gewaschen und getrocknet. Rohaus

beute: 31,1 g, Fp. 161-2° (Z.). Nach spektroskopischen Befunden handelt es sich um 3-Carboxy-4-(3-hydroxyphenyl)--2,5-dihydrofuran-2-on. Zur Decarboxylierung werden 20 g der Carbonsäure kurze Zeit in Diphenyläther auf 160°C erhitzt. Nach Abkühlen erhält man 14,5 g 4-(3-Hydroxyphe-nyl)-2,5-dihydrofuran-2-on.

b) 14,0 g 4-(3-Hydroxyphenyi)-2,5-dihydrofuran-2-on werden in 150 ml Epichlorhydrin mit kataly tischen Mengen Piperidin 9 Stunden auf 100°C erwärmt. Nach üblicher Aufarbeitung erhält man ein Gemisch aus Chlorhydrin- und Epoxi-Verbindung. Durch Säulenchromatografie an Kieselgel erhält man 10 g kristallines Epoxid.

c) 4,0 g der Epoxid-Verbindung (b) werden in 50 ml Methylenchlorid und 50 ml Methanol gelöst und mit 17 ml tert. Butylamin (lOfacher Überschuss) 5 Stunden auf 40°C erwärmt. Danach wird eingedampft und der Rückstand in verdünnter Salzsäure aufgenommen. Durch Extraktion mit Methylchlorid werden nichtbasische Bestandteile entfernt. Nach Alkalisieren der wässrigen Phase wird das Amin in Methylenchlorid aufgenommen. Man erhält 5,4 g Rohbase 4-[3-(2-Hydroxy-3-tert.butylaminopropoxy)-phenyl]-2,5-di-
hydrofuran-2-on, die durch Säulenchromatografie an Kieselgel gereinigt wird.
Zur Herstellung des Fumarats löst man 1,1 g der Base in Äthanol und versetzt mit 0,42 g Fumarsäure. Durch Einen-
5 gen der äthanolischen Lösung erhält man 1,2 g des des Fumarats von 4-[3-(2-Hydroxy-3-tert.butylamino-propoxy)--phenyl]-2,5-dihydrofuran-2-on.

4-[4-(2-Hydroxy-3-tert. butylamino-prapoxy)-phenyl]-2,5--dihydrofuran-2-on i5 a) 7,9 g 4-(4-Hydroxyphenyl)-2,5-dihydrofuran-2-on werden in 15 ml Epichlorhydrin mit kataly tischen Mengen Piperidin 6 Stunden auf 100°C erhitzt. Nach üblicher Aufarbeitung erhält man 4,0 g der Epoxiverbindung.

20 b) Analog Beispiel 42 c) lässt sich das Fumarat des 4-[4--(2-Hydroxy-3-tert. butylamino-propoxy)-phenyl]-2,5-dihy-drofuran-2-on erhalten.

- 6414512PATENTANSPRÜCHE 1. Substituierte 4-Aryl-2,5-dihydro-furan-2-one und 4-Aryl-l,5-dihydro-2H-pyrrol-2-one der allgemeinen Formel (I)(I)(V)ö-ch0-ch-ch52 i i 2 Oi ih x0-cho-CH-cho-nh-c-ch5r' 2 i 2 i 2k worin R1, R2 und A die im Anspruch 1 angegebene Bedeu-i5 tung haben und X Chlor oder Brom darstellt, oder ein Gemisch dieser Verbindungen mit einem Amin der Formel (VI)in derR1 und R2, die gleich oder verschieden sein können, ein Wasserstoff- oder Halogenatom, eine Allcyl- oder Acylaminogruppe mit 1 bis 6 Kohlenstoffatomen oder eine Carba-moylmethylgruppe,R3 und R4, die gleich oder verschieden sein können, ein Wasserstoffatom oder eine Methylgruppe,R5 ein Wasserstoffatom, eine Dimethoxyphenylgruppe oder eine Acylaminogruppe der allgemeinen Formel (II)R6-CONH20 g |R -CH_-C-NH0z I /'4(VI)(II)worin R6 für eine Alkylgruppe mit 1 bis 4 Kohlenstoffatomen steht, undA ein Sauerstoffatom, eine Imino- oder Alkyliminogrup-pe mit 1 bis 4 Kohlenstoffatomen bedeuten, sowie deren physiologisch verträgliche Säureadditionssalze.
- 2. Verbindungen nach Anspruch 1 der Formel IaR5I0-CH~-CH-CH--NH-C-CH5R21 2 Ï4OH E25 in der R3, R4 und R5 die im Anspruch 1 angegebene Bedeu-einer Säure in das entsprechende Säureadditionssalz um-tung haben, umsetzt, und gegebenenfalls die freie Base mit wandelt.
- 4. Verfahren nach Anspruch 3, dadurch gekennzeich-30 net, dass man als Amin der Formel VI mit Isopropylamin oder tert.-Butylamin umsetzt.
- 5. Verbindungen der Formeln IV und V, worin R1, R2 und A die im Anspruch 1 angegebene Bedeutung haben und X Chlor oder Brom darstellt, als Mittel zur Durchführung35 des Verfahrens gemäss Anspruch 3.
- 6. Pharmazeutisches Präparat, dadurch gekennzeichnet, dass es als Wirkstoff eine oder mehrere der Verbindungen gemäss Anspruch 1 enthält.5(I)
