Die Cyanhydrinspaltung kann thermisch, säuren- oder basenkatalysiert durchgeführt werden.
Im Falle der thermischen Durchführung erhitzt man die Verbindung der Formel II zweckmässigerweise auf Temperaturen von ca. 50 bis 400 "C, insbesondere auf ca.
80-250''C
Im Falle der säurenkatalysierten Cyanhydrinspaltung versetzt man die Verbindung der Formel II, zweckmässigerweise mit katalytischen Mengen, z.B. mit '/l000-/loÄqui- valenten, oder aber auch mit grösseren, z. B. molaren Mengen einer Säure. Wesentlich ist bloss, dass der pH-Wert des Reaktionsgemisches unter 7 liegt.
Die Natur der Säure ist nicht kritisch. Als Säuren kommen anorganische Säuren, z.B. 112SO4, HCI, H3PO4, oder organische Säuren, z. B. Ameisensäure, Essigsäure, Zitronensäure, Oxalsäure, oder saure Ionentauscher, z. B. Amberlite JRC 50 usw., in Frage
Im Falle der basenkatalytischen Cyanhydrinspaltung versetzt man die Verbindung der Formel II zweckmässigerweise mit katalytischen Mengen, z.B. mit l/lOOO-I/IoÄqui- valenten, oder aber auch mit grösseren, z. B. molaren Mengen einer Base. Wesentlich ist bloss, dass der pH-Wert des Reaktionsgemisches über 7 liegt.
Die Natur der Base ist nicht kritisch. Als Basen kommen z. B. in Frage: anorganische Basen wie Alkalimetallhydroxyde, z. B. NaOH, Erdalkalimetallhydroxyde, z. B. Ca(OH)2, Mg(OH)2, Alkalimetallcarbonate, z.B. Na2 CO3, K2CO3, Alkalimetallbicarbonate wie NaHCO3, Ammoniak, andere basische Salze wie Na3PO4, K2HPO4, Borax, basische Puffersysteme wie z. B. NaHCO3/Na2CO3, K2HPO4/K3PO4 usw., organische Basen wie Amine, z. B. Triäthylamin, Pyridin, Morpholin usw., Salze organischer Säuren mit starken Basen, wie Natriumacetat, -formiat, -oxalat, -citrat-, 4acetat, oder basische Ionentauscher, z. B. Amberlite JRA 400, Dowex 2 usw.
Die Cyanhydrinspaltung kann in der Gasphase oder in flüssiger Phase durchgeführt werden. Anwesenheit eines Lösungsmittels ist nicht erforderlich, jedoch zweckmässig.
Als Reaktionstemperaturen kommen insbesondere 50-200 "C, bevorzugt ca. 100 -C in Frage.
Die Natur des Lösungsmittels ist nicht kritisch, es können polare Lösungsmittel - wie Wasser, Ammoniak, Alkohole, oder apolare Lösungsmittel, wie Toluol, Benzol, Toluol, Äther, Petroläther usw., zur Verwendung gelangen.
Bevorzugte Systeme sind basische Ionentauscher in der OH--Form/H2O oder organische Säuren bzw. deren Salze, wie CH3COOH/H2O, Natriumoxalat/H2O, oder Pyridin/ Toluol bei Reaktionstemperaturen von rund 100 C.
Die Verbindungen der Formel II sind neu. Sie bilden als Mittel zur Ausführung des erfindungsgemässen Verfahrens ebenfalls Gegenstand der vorliegenden Erfindung.
Die Verbindungen der Formel II können vorteilhaft durch Oxydation von Verbindungen der Formel

Als Oxydationsmittel sind insbesondere Alkalimetallcaroate, z. B. KH SOs, geeignet. Bevorzugt ist Caroat @ (KHSO5, welches KHSO4 und K2SO4 enthält).
Das Caroat wird zweckmässigerweise in einer Menge von 1-2,5 Äquivalenten, insbesondere 1,1-1,5 Aequivalenten, verwendet.
Die Oxydation wird vorzugsweise in polaren Lösungsmitteln, wie Wasser, Alkoholen, Aceton, Acetonitril, oder Gemischen solcher Lösungsmittel durchgeführt.
Der pH-Wert des Mediums beträgt zweckmässigerweise ca. 3-11, wie dies durch entsprechende Puffersysteme vom Carbonat-, Phosphat-, Citrat-, Borat-, NH3/NH4+- oder Oxalattyp in an sich bekannter Weise erzeugt werden kann.
Die Reaktionstemperatur kann beispielsweise zwischen - 10 bis 60 "C, bevorzugt zwischen 0 bis 20 C liegen.
Die Verbindungen der Formel III, worin X + 0, sind ebenfalls neu. Diese Verbindungen der Formel III' können insbesondere nach der Thorpe-Ziegler-Methode (siehe z. B.
The Merck Index, Encyclopedia of Chemicals and Drugs, 9th ed., 1976, O.N.R. 87, Merck and Co., Rahway, N.J.) aus einem Dinitril IV oder durch eine Dieckmann-Zyklisierung (The Merck Index, siehe oben, O. N. R. 23) eines entsprechenden Cyanoesters V gemäss:

Die Verbindungen der Formel I sind generell bekannt; sie stellen Geschmackstoffe und/oder Zuckerabbauprodukte dar.
Beispiel 1
Herstellung von 3,4-Dimethylcyclopent-2-en-2-olon a) In einen Sulfierkolben werden 12 g Magnesiumspäne vorgelegt. Unter Stickstoffatmosphäre, Rühren und Eiskühlung wird bei 0 bis 5 C langsam ein Gemisch von 53,7 g Crotonsäurenitril, 64 g Methanol und 40 ml Toluol zugetropft. Darauf wird 30 Minuten bei 5 C und 60 Minuten bei Raumtemperatur weitergerührt. Der dicke graue Brei wird mit 2-n Schwefelsäure neutralisiert, die organische Phase abgetrennt und dreimal mit je 150 ml Toluol nachextrahiert.
Die eingeengten, vereinigten organischen Phasen ergeben 20 g (38% der Theorie) 3,4-Dimethyladiponitril, Sdp.
107-109 C/0,15 Torr.
a') In ein Reaktionsgefäss legt man 67,1 g (1 Mol) Crotonsäurenitril und 76 g (0,65 Mol) Ammoniumperchlorat vor und versetzt das Gemisch unter Kühlung auf ca. - 70 "C mit ca. 260 g wasserfreiem Ammoniak. Mittels einer am Gefässboden liegenden Quecksilberkathode und einer hängenden Graphitanode wird während 15 Stunden bei - 75 "C eine Gleichstromspannung von 15 Volt (4-5 Ampere) angelegt. Man lässt das Ammoniak verdampfen. Die dunkelbraune Lösung wird mit 700 ml Ather aufgenommen und mit 250 ml Wasser gewaschen. Die eingeengte Ätherlösung ergibt 31 g (46%) 3,4-Dimethyladiponitril, welches bei 107-109 "C/0,15 Torr fraktioniert destilliert wird.
b) 5,6 g Kalium-tert.butylat werden mit 6,8 g 3,4-Dimethyladiponitril in 50 ml Toluol während 60 Minuten rückflussiert. Die erkaltete Lösung wird mit 50 ml Wasser gewaschen und eingeengt. Man erhält 6,4 g (94% der Theorie) 2 Cyano-3,4-dimethyl-cyclopent- 1 -enylamin, Schmp.

IR (CHCl3) 3500 und 3400 (NH), 2180 (CN), 1638 und 1596 (C=C); MS: 136(M+), 121 (100%), 94.
c) 2 g des obigen Produktes werden 10 Minuten mit 50 ml 2-n H2SO4 gerührt und anschliessend dreimal mit je 30 ml Äther extrahiert. Die getrockneten und eingeengten Ätherphasen enthalten 1,8 g (89%) 2-Cyano-3,4-dimethylcyclopentanon, Sdp. 75-76 C/0,05 Torr.
IR: 2250 (CN), 1750 (stark, C=O); MS: 137 (M+, 100%) 122, 108,94, 69, 68.
d) 2 g des erhaltenen Produktes werden zusammen mit 1,4 g Borax und 1,17 g NaOH in 20 ml Wasser gelöst und mit 6,4 g Caroat (Degussa) versetzt. Nach 30 Minuten wird filtriert, mit H2SO4 auf pH 1 angesäuert und viermal mitje 30 ml CH2CI2 extrahiert. Nach dem Einengen erhält man 1,57 g (71 %) eines Enantiomerengemisches von 2-Cyano-2 hydroxy-3,4-dimethylcyclopentanon, nD20 = 1,5490.
IR: 3150 (OH), 2250 (sehr schwach, CN), 1710 (C = O); MS: 139 (M+), 121, 112, 95, 83 (100%).
e) 1 g des oben erhaltenen Produktes wird mit 0,8 g Natriumacetat während 6 Stunden in 15 ml Wasser rückflussiert und darauf viermal mit Essigester extrahiert. Durch Einengen des Essigesters erhält man 0,578 g (70%) braune Kristalle von 3,4-Dimethyl-cyclopent-2-en-2-ol- 1 -on. Smp.
69-71 C (Wasser).
NMR (CDCl3): 6 = 5,8 ppm Singulett, breit/l Pr (OH); 2,8-2,5 Multiplett/2Pr (H5); 2,2-1,8 Multiplett/lPr (H-4); 1,98 Singulett/3 Pr (CH3 an C-4).
Dasselbe Produkt kann aus dem gemäss Beispiel Id) erhaltenen 2-Cyano-2-hydroxy-3,4-dimethylcyclopentanon hergestellt werden, indem 2 g des letzteren in 60 ml gesättigter Natriumbicarbonatlösung während 4 Stunden rückflussiert werden und die erkaltete Lösung viermal mit je 30 ml Methylenchlorid extrahiert wird. Aus der eingeengten organischen Phase erhält man 0,98 g (60% Ausbeute) des reinen (GC und DC) kristallinen Produktes. Smp. 66-68 "C.
Die Umkristallisation erfolgt aus Wasser.
Beispiel 2
Herstellung von 3-Hydroxy-4,5-dimethyl-2(5H)-furanon a) 10 g 2-Cyano-3-methyl-valero-Y-lacton (hergestellt gemäss Chem. Abstr. 58, 5508 b (1962)) werden zusammen mit 14 g Borax in 150 ml Wasser mit 24,5 g Caroat versetzt, gleichzeitig werden 65 ml 2-n Natronlauge zugetropft. Nach 30 Minuten wird mit 100 ml Essigester gewaschen, mit 80 ml 2-n H2SO4 auf pH 1 angesäuert und viermal mit 100 ml Essigester extrahiert. Der getrocknete und eingeengte Extrakt ergibt 6,7 g (60%) eines gelben Ols von 2-Cyano-2-hydroxy 3-methyl-valero-y-lacton (Epimerengemisch). nD20 = 1,4509.
IR: 3370 (OH); 2250 (schwach, CN); 1782 (C=O) MS: 155 (2%, M+), 128 (M-HCN), 133,96,83 (100%).
b) 3,5 g des Lactons werden mit 2,75 g Natriumacetat während 3 Stunden in 50 ml Wasser rückflussiert. Die erkaltete Lösung wird viermal mit je 50 ml Essigsäure extrahiert und der Extrakt mit Na2 SO4 getrocknet und eingeengt. Man erhält 1,43 g (50%) gaschromatographisch reines 3-Hy droxy-4,5-dimethyl-2(SH)-furanon.
NMR (CDCl3) 8 = 7,2 ppm Singulett breitlI Pr (OH); 4,91 Quartett x Quartett (Jl = 6,5 Hz, J2= l,SHz)/lPr(HS); 1,95 Dublett (J= 1,5 Hz)/3Pr (CH5 an C-4); 1,44 Dublett (J=6,5 Hz)/3Pr (CH3 an C-5); 128 (M+), 113, 85, 83 (100%), 72, 57, 55.
Beispiel 3
Herstellung von 3-Methylcyclopent-2-en-2-olon a) Eine Lösung von 30,6 g Natriumäthylat in 400 ml Äthanol wird mit 80 g Diäthylphosphono-acetnitril versetzt.
Nach 2 Stunden tropft man bei 5 C 71,5 g Lävulinsäure äthylester zu, was 60 Minuten erfordert, und rückflussiert hierauf die trübe Emulsion während 30 Minuten. Nun wer den ca. 300 ml Äthanol abdestilliert, und es werden 400 ml
In-HCI zugegeben. Die organische Phase wird abgetrennt, die wässrige Phase dreimal mit je 200 ml Äther extrahiert und die vereinigten organischen Phasen eingeengt. Man er hält 50,5 g (67%) einer klaren, gelblichen Flüssigkeit des cis trans-Gemisches von 5-Cyano-4-methyl-4-pentensäure äthylester, Sdp. 85-86 C/0,05 Torr.
b) 25 g des obigen Esters werden mit 1 g Palladium auf
Aktivkohle in 150 ml Methanol während 2 Stunden hydriert, darauf wird filtriert und eingeengt. Man erhält 24,9 g (99%)
2-Methyl-adipomononitril-monoäthylester. Sdp. 76-81"C/
0,04 Torr.

MS: 169(M+), 154, 142, 129, 124 (M+-OAe, 100%), 101,
96, 88.
c) 3,3 g des Esters werden zusammen mit 3,3 g Kalium tert.butylat in 50 ml Toluol während 30 Minuten rückflus siert. Darauf wird mit 50 ml In-Salzsäure versetzt, die Was serphase abgetrennt und dreimal mit je 40 ml CH2CIz ex trahiert. Die vereinigten organischen Phasen werden über Na2 SO4 getrocknet, eingeengt und am Hochvakuum destil liert und ergeben so 2,22 g (92%) 2-Cyano-3-methyl-cy clopentanon, Sdp. 90 C/0,04 Torr.

MS: 123 (M+, 100%), 108,94, 80,68, 55.
d) 1,45 g des erhaltenen Nitrils werden zusammen mit 2,3 g Borax und 0,96 g NaOH in 15 ml Wasser gegeben und bei 15 "C mit 5,4 g Caroat (Degussa), gelöst in 18 ml Wasser versetzt. Nach 45minütigem Rühren wird mit 25 ml Essigester gewaschen, mit 50 ml 4n-H2SO4 angesäuert und dreimal mit je 25 ml Essigester extrahiert. Die Essigesterphasen werden über Na2 SO4 getrocknet und eingeengt und ergeben 1,35 g (82% Ausbeute) eines braunen Öls von 2-Cyano-2-hydroxy-3-methylcyclopentanon. nD20 = 1,4708, Sdp. 120-130 "C/0,04 Torr.
IR: 3380 (OH), 2250 (schwach, C=N), 1760 und 1712 (C=O); MS: 153 (M+), 135, 126, 83, 69 e) 1,5 g des oben erhaltenen Cyanhydrins werden zusammen mit 1,35 g Natriumacetat in 25 ml Wasser während 5 Stunden rückflussiert. Die erkaltete Reaktionslösung wird sechsmal mit je 25 ml Essigester extrahiert. Die organischen Phasen werden vereinigt, über Na2 SO4 getrocknet und eingeengt und ergeben so 554 mg (45% Ausbeute) eines gelben Pulvers von 3-Methylcyclopent-2-en-2-olon. Smp.

Beispiel 4
Herstellung von 3,5-Dimethylcyclopent-2-en-2-olon a) 2,9 g (23,5 mMol) 2-Cyano-3-methyl-cyclopentanon (hergestellt gemäss Beispiel 3c) werden in 60 ml trockenem Tetrahydrofuran und 2,8 ml Hexamethylphosphortriamid gelöst und mit 0,62 g (25,9 mMol) Natriumhydrid versetzt.
Darauf werden unter Argonatmosphäre bei - 8 "C 25,9 mMol einer 1,6 molaren Hexanlösung von Butyllithium und anschliessend 3,67 g (25,9 mMol) Methyljodid zugegeben.
Das Gemisch wird weitere 20 Minuten bei ca. 0 C gehalten (exotherme Reaktion) und darauf auf 75 ml In-Salzsäure und 100 g Eis geworfen. Die wässrige Phase wird dreimal mit je 50 ml Ather extrahiert, mit Na2 SO4 getrocknet und eingeengt. Das erhaltene Ol wird an 140 g Kieselgel mit zwei Teilen Hexan und einem Teil Äther fraktioniert chromatographiert. Man erhält 1,03 g (32% der Theorie) eines gelben Öls eines Diastereomerengemisches von 2-Cyano-3,5-dimethylcyclopentanon.
IR: 2250 (CN), 1755 (C=0); NMR (CDCl3) : 6 = 1,6-3,5 ppm komplexes Multiplett /SPr (H-2, H-3, H-4, H-5); 1,29 und 1,14 je ein Dublett/6Pr (J = 6 Hz, CH3-3 und CH3-5); MS: 137 (M+), 122, 108, 94, 81, 68 (100%).
b) 24 g (175 mMol) der obigen Cyanoverbindung werden zusammen mit 33,3 g (87,5 mMol) Borax und 14 g (350 mMol) Natriumhydroxid in 300 ml Wasser gelöst und unter Kühlung mit 87 g (257 mMol) Caroat in266 ml Wasser versetzt. Nach weiteren 30 Minuten Rühren beträgt der pH der Lösung 5,6. Mittels 2n-Schwefelsäure wird auf pH 1 angesäuert, und es wird dreimal mit je 150 ml Essigester extrahiert. Die organischen Phasen werden über Natriumsulfat getrocknet und eingeengt und ergeben so 26,2 g (98% der Theorie) eines farblosen dickflüssigen Öls von 2-Cyano-2hydroxy-3,5-dimethylcyclopentanon. Dieses Öl lässt sich bei 50-60 C/0,035 Torr destillieren. nD20 = 1,4577.
IR: 3450 (stark, OH), 2250 (schwach, CN), 1760 (stark, C=O); NMR (CDCl3): 6 = 4,4 ppm, Singulett, breit/l Pr (OH); 3,1-1,6 Multiplett/4 Pr (H-3, H-4, H-5,); 1,5-1,0 Multiplett/ 6 Pr (CH3-3 und CH3-5); MS: 153 (M+), 135, 126, 120, 109, 96, 83 (100%), 74.
c) 23,2 g (152 mMol) des obigen Cyanhydrins werden mit 18,6 g (227 mMol) Natriumacetat in 380 ml Wasser unter Einleiten eines kräftigen Stickstoffstroms 40 Minuten rückflussiert. Die erkaltete Reaktionslösung wird dreimal mit je
150 ml Methylenchlorid extrahiert und die vereinigten Extrakte über Natriumsulfat getrocknet und eingeengt. Man erhält 12,0 g (63%) der Theorie) 3,5-Dimethylcyclopent-2en-2-olon. Smp: 92-95 "C (Äther/Hexan).
