PATENTANSPRÜCHE
1. Verfahren zur Trennung von racemischen Alkalime tallsalzen und vom Lacton der cis-2-Hydroxy-cyclopent-4en-l-yl-essigsäure, dadurch gekennzeichnet, dass man ein Alkalimetallsalz oder das Lacton der racemischen cis-2 Hydroxy-cyclopent-4-en-l -yl-essigsäure im wässrigen Medium mit optisch aktivem a-Phenyl-äthylamin und/oder mit dessen Salz mit einer starken Säure umsetzt; das eine, aus dem Reaktionsgemisch ausgeschiedene Teil des entstandenen diastereomeren Salzpaares von der Mutterlauge trennt, dann mit einer Säure in das entsprechende optisch aktive Lacton, beziehungsweise mit Alkalimetallhydroxyd in das entsprechende optisch aktive Salz der cis-2-Hydroxy-cyclopent-4-en-1-yl-essigsäure überführt;
die, nach der Trennung des einen, während der Abtrennung aus dem Reaktionsgemisch ausgeschiedenen diastereomeren Salzes erhaltene Mutterlauge mit starker anorganischer Säure ansäuert und das entstandene cis-2-Hydroxy-cyclopent-4-en- 1 -yl-essigsäu- re-Lacton mit einem nicht mit Wasser vermischbaren Lösungsmittel extrahiert und in einer Lösung von einem niederen Alkanol, mit einer, mit dem Lacton-Gehalt des Racemats äquivalenten Menge von Alkalimetallhydroxyd hydrolysiert; das ausgeschiedene Alkalimetallsalz der racemischen cis-2-Hydroxy-cyclopent-4-en- 1 -yl-essigsäure von der Mutterlauge trennt;
die erhaltene Mutterlauge mit einer, mit ihrem Lacton-Gehalt äquivalenten Menge von Alkalimetallhydroxyd hydrolisiert und das ausgeschiedene Alkalimetallsalz des Antipoden der cis-2-Hydroxy-cyclopent-4-en-l-yl- essigsäure, das gegensätzlich vom diastereomeren Salz ist, das durch das Trennmittel ausgeschieden wurde, trennt.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man das Lacton der racemischen cis-2-Hydroxy-cyclopent-4-en-l-yl-essigsäure als Ausgangsstoff nimmt.
3. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man das Natriumsalz der racemischen cis-2-Hydroxy cyclopent-4-en-1-yl-essigsäure als Ausgangsstoff nimmt.
4. Verfahren nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass man die Trennung durch Verwendung von 0,3-1,5 Mol optisch aktives a-Phenyl-äthylamin auf 1 Mol cis-2-Hydroxy-cyclopent-4-en- 1 -yl-essigsäure-salz be ziehungsweise Lacton gerechnet durchführt.
5. Verfahren nach einem der Ansprüche 1-4, dadurch gekennzeichnet, dass man bei der Trennung das Natriumsalz der racemischen cis-2-Hydroxy-cyclopent-4-en-1-yl-essigsäu- re als Ausgangsstoff nimmt, vom optisch aktiven a-Phenyl äthylamin weniger als die stöchiometrische Menge verwendet und die Trennung in Gegenwart eines, mit Wasser unvermischbaren organischen Lösungsmittels durchführt.
6. Verfahren nach einem der Ansprüche 1 bis 5, dadurch gekennzeichnet, dass man bei der Trennung die Hydrolyse des Lactons, das durch Behandlung mit einer Säure der Mutterlauge, die nach der Trennung des einen diastereomeren Salzes erhalten wurde, in zwei Schritten durch Verwendung von Natriumhydroxyd durchführt.
7. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man das getrennte Alkalimetallsalz der racemischen cis 2-Hydroxy-cyclopent-4-en-1-yl-essigsäure in den Trennungsprozess zurückführt.
Gegenstand der Erfindung ist ein neues Verfahren zur
Trennung von racemischen Alkalimetallsalzen und/oder vom Lacton der cis-2-Hydroxy-cyclopent-4-en- 1 -yl-essigsäu- re.
Die cis-2-Hydroxy-cyclopent-4-en- l-yl-essigsäure besitzt zwei optische Antipoden, die im Weiteren (+), beziehungs weise ()-cis-2-Hydroxy-cyclopent-4-en-1-yl-essigsäure genannt werden. Diese Säuren und die racemische Säure, die von deren äquimolaren Gemisch gebildet wird, haben die gleiche Eigenschaft und zwar die, dass sie sich im sauremwässrigen Medium in ein bicyclisches Lacton der allgemeinen Formel

umwandeln.
Da die Hydroxyl- und Carboxymethyl-gruppe in der cis-2-Hydroxy-cyclopent-4-en- 1 -yl-essigsäure der allgemeinen Formel

die gleiche Seite des Cyclopenten-Ringes einnehmen, das heisst in dem einer Antipode beide Gruppen a, während in der anderen Antipode beide Gruppen ss sterische Stellung haben, steht der Lactonring in dem durch Ringschluss entstandenen Lacton der allgemeinen Formel I bei der einen Antipode hinter dem Cyclopenten-Ring und bei der anderen Antipode davor. Bei der Darstellung der Formeln haben wir darauf keine Rücksicht genommen, deshalb werden die sterischen Stellungen der einzelnen Substituten beziehungsweise die anellierenden Richtungen der Ringe nicht gezeichnet.
Die optisch aktiven und racemischen cis-2-Hydroxy cyclopent-4-en-1-yl-essigsäure-lactone der allgemeinen Formel I werden zur Herstellung von Prostaglandinen und anderen Verbindungen mit Cyclopenten-Gerüst, wie zum Beispiel cis-Jasmon, verwendet (J.A.C.S., 95, 6382. [1973], beziehungsweise J. Org. Chem., 37, 2363. [1972]). Die Verbindungen der allgemeinen Formel I werden auf folgender Weise hergestellt: Cyclopentadien und Dichloracetylchlorid reagieren, in Gegenwart von Triäthylamin miteinander, wodurch das entstehende Dichlorketon auf das Cyclopentadien additioniert wird und ein bicyclisches Dihalogen-Keton entsteht.
Diese Verbindung wird unter Reduktion dehalogeniert, dann wird die racemische Verbindung der allgemeinen Formel I durch Behandlung des entstandenen bicyclischen Ketons unter gleichen Umständen wie bei der Baeyer-Villiger-Oxidation hergestellt (Tetrahedron Letters 307. [19703).
Zur Herstellung von Prostaglandinderivaten von natürlicher Konfiguration ist es aber nicht vorteilhaft die racemische Verbindung der allgemeinen Formel I als Ausgangsstoff zu nehmen, weil dadurch auch die enantiomere Verbindung als Ballaststoff in der Synthese durchgeführt werden muss und das heisst ausser der Verminderung der Ausbeute auch, dass die Trennung bei einem späteren Schritt der Synthese mit empfindlicheren Stoffen durchgeführt werden muss.
Die Trennung der Verbindung der allgemeinen Formel I durch Verwendung vom optisch aktiven a-Phenyl-äthylamin wird in der Literatur beschrieben (J.A.C.S. 95, 6832. [1973]).
Gemäss der, in der Literatur beschriebenen Methode wird folgendermassen vorgegangen: die racemische Verbindung der allgemeinen Formel I wird mit äquimolarer Menge einer
wässrigen Base hydrolisiert, die erhaltene Lösung vom Salz der racemischen Säure der allgemeinen Formel II wird bei 0 0C in Gegenwart von Äthylacetat auf den pH-Wert 3,5-4 gesäuert und die frei gewordene racemische Säure der allgemeinen Formel II wird mit Äthylacetat extrahiert. Die erhaltenen Äthylacetat-Lösung wird bei 0 "C entwässert. Dazu wird die optisch aktive Base ((+)-a-Phenyl-äthylamin) im ca. 5% Überschuss zugegeben. Das ausgeschiedene kristalline Salz wird durch Umkristallisieren gereinigt.
Das erhaltene Salz wird in der äquimolaren Menge einer wässrigen Base gelöst, das auf dieser Weise freigewordene (+)-a-Phenyl äthylamin wird durch Extrahieren mit Äther entfernt, und dann wird die erhaltenen wässrige Lösung beim pH-Wert und bei 20 "C mit Jod reagiert und dadurch in ein sogenanntes Jodlacton überführt.
Es gibt noch ein bekanntes Verfahren (ungarische Patentanmeldung Nr. CI-1678) zur Reinigung der Verbindungen der allgemeinen Formel I beziehungsweise II durch Salzbildung. Dieses Verfahren ermöglicht auch die Herstellung von Alkalimetall- und Erdalkalimetallsalzen der Säure der allgemeinen Formel II.
Der gemeinsame Nachteil all dieser Verfahren ist, dass die Trennung und die Salzbildung mit deren Hilfe nicht in den für die industrielle Produktion notwendigen Mengen gelöst werden können. Zwar sichern die bekanntgemachten Verfahren die Herstellung reiner Produkte, aber sie würden bei der Steigerung der Mengen wegen der relativ kleinen Lösungskonzentrationen die Aufarbeitung eines grossen Lösungsvolumens benötigen, was ungerechtfertigt wäre. Überraschenderweise wurde nun gefunden, dass die Alkalimetallsalze und das Lacton der racemischen cis-2-Hydroxy-cyclo pent-4-en-1-yl-essigsäure auch, in für industrielle Mengen lösbarer Weise durch Anwendung vom optisch aktiven -Phenyl-äthylamin getrennt werden können, wenn das Verfahren nach Anspruch 1 durchgeführt wird.
Das während des Verfahrens entstehende racemische, beziehungsweise in kleinem Masse optisch aktive cis-2-Hydro xy-cyclopent-4-en-l-yl-essigsäure-salz kann in den Trennungsprozess zurückgeführt werden.
Bei der Durchführung der Trennung kann man das racemische Lacton als Ausgangsstoff wählen, aber in einem noch vorteilhafteren Verfahren wird das racemische Lacton in wässriger Alkalimetallhydroxyd-Lösung gelöst und die erhaltene Lösung des Alkalimetallsalzes getrennt. Beim erfindungsgemässen Verfahren können Natrium- oder Kaliumhydroxyd als Alkalimetallhydroxyd gleicherweise verwendet werden. Vorteilhaft ist Natriumhydroxyd zu verwenden.
Was für die Alkalimetallhydroxyde angegeben wurde, bezieht sich sinngemäss auch auf die Alkalimetallsalze.
Beim erfindungsgemässen Verfahren kommen die aromatischen und aliphatischen Kohlenwasserstoffe und deren verschiedene halogenierte Derivate als mit Wasser unvermischbare Lösungsmittel in erster Linie in Betracht. Jedes organisches Lösungsmittel, das mit Wasser nicht vermischbar und den Reaktionspartnern gegenüber inaktiv ist und das cis-2-Hydroxy-cyclopent-4-en- 1 -yl-essigsäure-lacton löst, kann aber verwendet werden. Dichloräthan und Dichlormethan wurden äusserst vorteilhaft gefunden.
Beim erfindungsgemässen Verfahren ist es vorteilhaft 0,3-1,5 Mol vom optisch aktiven a-Phenyl-äthylamin für 1 Mol racemisches Lacton zu verwenden. Wenn die Menge des Trennungsmittels nicht die stöchiometrische Menge [aufs Lacton gerechnet] erreicht, ist es vorteilhaft, wenn die Trennung in Gegenwart der oben angeführten organischen Lösungsmittels durchgeführt wird und so die andere Säure Antipode, die in Form eines, als diastereomeres Salz nicht ausgeschiedenen Lactons anwesend ist, in Lösung gehalten wird.
Als, zum Verfahren verwendetes, mit Wasser vermischbares organisches Lösungsmittel wird ein niederer Alkanol genommen. Solche Alkanole sind die, gegebenenfalls geraden oder verzweigten aliphatischen Alkanole mit 14 Kohlenstoffatomen, wie zum Beispiel Methanol, Äthanol, n- und i-Propanol, sowie n-, sek.- und tert.-Butanol. Sehr vorteilhaft ist die Verwendung von Äthanol.
Nähere Einzelheiten des erfindungsgemässen Verfahrens werden an Hand der folgenden Beispiele erläutert.
Beispiel 1
32,8 g Natriumsalz der racemischen cis-2-Hydroxycyclopent-4-en-l-yl-essigsäure (0,200 Mol) werden in 35 ml Wasser gelöst. Zur Lösung werden 54 ml Dichloräthan und 14,0 g (0,116 Mol) (+)-a-Phenyl-äthylamin zugegeben. Das erhaltene Gemisch wird unter 5 C gekühlt und 14 ml (0,168 Mol) wässrige Salzsäure-Lösung (37,0 Gew.-%, spezifisches Gewicht: 1,19 g/ml) werden bei dieser Temperatur unter Rühren dazu gegeben. Das Reaktionsgemisch wird eine weitere Stunde lang gerührt, dann filtriert und der ausgeschiedene Stoff wird mit 2 x 5 ml Dichloräthan gewaschen und getrocknet. Man erhält 19,7 g (75,0%) (-)-cis-2-Hydroxy- cyclopent-4en-1-yl-essigsäure-(+)-a-phenyl-äthylamin Salz.

Beispiel 2
Die, im Beispiel 1 erhaltene zweiphasige Mutterlauge wird mit 20 ml cc. Salzsäure unter Rückfluss gekocht. Nach Abkühlung und Trennung der organischen Phase wird die wässrige Phase mit 2 x 100 ml Chloroform ausgeschüttelt.
Die vereinigte organische Phase wird mit wasserfreiem Natriumsulfat getrocknet und eingeengt. Als Rückstand erhält man 15,5 g (100%) Lacton der Formel I (Gemisch von Racemat und (+)-Antipode).

Beispiel 3
Das, im Beispiel 2 erhaltene Lacton der Formel I wird in 45 ml absoluten Äthylalkohol gelöst und 2,2 g (0,055 Mol) festes, granuliertes Natriumhydroxyd werden dazu gegeben, und dann wird das Gemisch eine Nacht (12 Stunden lang) bei Zimmertemperatur stehen gelassen.
Nachher wird das Gemisch unter 5 "C gekühlt und filtriert. Der erhaltene feste Stoff wird mit 2 x 1 ml Äthylalkohol gewaschen und getrocknet. Man erhält 9,1 g (27,6%, gerechnet auf die Messung von Beispiel 1) cis-2-Hydroxy cyclopent-4-en-1 -yl-essigsäure-natriumsalz (Gemisch vom Racemat und (+)-Antipode).

Beispiel 4
Zur im Beispiel 3 erhaltenen Mutterlauge werden 2,8 g (0,07 Mol) weiteres Natriumhydroxyd zugegeben. Die Lösung wird wieder eine Nacht stehen gelassen und schliesslich eingeengt. Der Rückstand beträgt 11,4 g (69,6%) (+)-cis-2 Hydroxy-cyclopent-4-en- 1 -yl-essigsäure-natriumsalz.

Beispiel 5
19,7 g gemäss Beispiel 1 erhaltenes (-)-cis-2-Hydroxy- cyclopent-4-en- 1 -yl-essigsäure-( + )-a-phenyl-äthylamin Salz werden in 40 ml warmen Wasser gelöst. Die Lösung wird 3 Stunden lang stehen gelassen, dann eine Stunde lang unter 5 "C gekühlt und schliesslich filtriert. Das erhaltene Salz wird mit 2 > < x 2 ml eisigen Wasser gewaschen und getrocknet.
Man erhält 17,1 g (86,8%) reines (-)-cis-2-Hydroxy-cyclo- pent-4-en- 1 -yl-essigsäure-( + )-a-phenyl-äthylamin Salz.

Beispiel 6
Zur im Beispiel 5 erhaltenen Mutterlauge werden 30 ml Chloroform und 5 ml cc. Salzsäure zugegeben und nachher wird sie eine Stunde lang unter Rückfluss gekocht. Nach Kühlung wird die organische Phase getrennt und die wässrige Phase mit 2 x 20 ml Chloroform extrahiert. Die vereinigte organische Phase wird mit wasserfreiem Natriumsulfat getrocknet und eingeengt. Zum Rückstand werden 5 ml absoluter Äthylalkohol und 0,38 g Natriumhydroxyd zugegeben.
Nachdem die Lösung 45 Stunden lang stehen gelassen wurde, wird sie eingeengt. Man erhält 1,5 g (4,6%, gerechnet auf die Messung von Beispiel 1) cis-2-Hydroxy-cyklopent-4-enl-yl-essigsäure-natriumsalz. (Ein racemisches Produkt mit kleinem Gehalt vom (-)-Antipode.) [a]20540 = -2"0"(c = 2, Salzsäure).
Beispiel 7
22,8 g racemisches cis-2-Hydroxy-cyclopent-4-en- 1 -yl- essigsäure-natriumsalz und'l0,0 g gemäss Beispiel 3 beziehungsweise 6 regeneriertes racemisches und optisch aktives cis-2-Hydroxy-cyclopent-4-en- 1 -yl-essigsäure-natriumsalz, wobei beachtet wird, dass der [a]20 54-Wert des Gemisches kleiner als + 10 sein soll (c = 2, Wasser). Im Weiteren wird gemäss Beispiel 1 gehandelt. Als Produkt erhält man 19,3 g (73,8 (- )-cis-2-Hydroxy-cyclopent-4-en-1 -yl-essigsäure (+ )-a-phenyl-äthylamin-salz.

Beispiel 8
Zu 5,7 g (0,047 Mol) (+ )-a-Phenyl-äthylamin werden 25 ml Wasser zugegeben. Der pH-Wert wird mit cc. Salzsäure-Lösung auf 8 eingestellt und dann wird cis-2-Hydroxy cyclopent-4-en-1-yl-essigsäure-natriumsalz zur erhaltenen Lösung zugegeben. Vorübergehend entsteht eine Lösung, aus der bald Kristalle ausgeschieden werden. Das Reaktionsgemisch wird bei Zimmertemperatur 3 Stunden lang stehen gelassen und dann unter 5 "C gekühlt, filtriert, das ausgeschiedene Salz wird mit 2 > < x 0,5 0,5 ml eisigen Wasser ge- waschen und getrocknet. Man erhält 3,3 g (67,3%) (-)-cis2-Hydroxy-cyclopent-4-en- 1 -yl-essigsäure- (+ )-a-phenyl äthylaminsalz.

Das erhaltene Salz wird gemäss Beispiele 5 und 6 gereinigt und die Mutterlauge wird gemäss Beispiele 2, 3 und 4 aufgearbeitet.
Beispiel 9
Zum Gemisch von 11,4 g (0,092 Mol) (+ )-a-Phenyl- äthylamin und von 80 ml Wasser wird bis zum pH-Wert 7 cc. Salzsäure-Lösung zugegeben. Danach werden 24,4 g (0,148 Mol) racemisches cis-2-Hydroxy-cyclopent-4-en-1-yl- essigsäure-natriumsalz dazu gemessen. Im Weiteren wird gemäss Beispiel 8 gehandelt. Als Produkt erhält man 12,0 g (61,7%) (- )-cis-2-Hydroxy-cyclopent-4-en-1-yl-essigsäure- (+ )-a-phenyl-äthylaminsalz.

Die weitere Reinigung des Produkts und die Aufarbeitung der Mutterlauge wird gemäss Beispiel 8 vorgenommen.
Beispiel 10
Zu 4,52 g (0,036 Mol) racemischen cis-2-Hydroxy-cyclo pent-4-en-1-yl-essigsäure-lacton (racemische Verbindung der Formel I) werden 4,8 g (0,039 Mol) (+)-a-Phenyl-äthylamin und 30 ml Wasser zugegeben. Das Gemisch wird gerührt und nach dem Anfang der Kristallisierung wird es 3 Stunden lang bei Zimmertemperatur und 2 Stunden lang unter 5 "C gehalten. Das ausgeschiedene (- )-cis-2-Hydroxy-cyclopent- 4-en- 1 -yl-essigsäure-( + )-a-phenyl-äthylaminsalz wird filtriert, mit 2 x 0,5 ml Eis-Wasser gewaschen und getrocknet.
Man erhält 1,7 g (35,9%) trockenes Produkt.

Beispiel 11
Zu 24,8 g (0,2 Mol) recemischen cis-2-Hydroxy-cyclo pent-4-en-1-yl-essigsäure-lacton werden 35 ml Wasser, 54 ml Dichloräthan und 8,0 g (0,2 Mol) Natriumhydroxyd zugegeben. Im Weiteren wird gemäss Beispiel 1 gehandelt und das dort angegebene Produkt von gleicher Qualität und in gleicher Menge erhalten.
Beispiel 12
Es wird gemäss Beispiel 1 gehandelt, bloss die Trennung wird mit (-)-a-Phenyl-äthylamin ausgeführt. Man erhält 19,7 g (75%) (+ )-cis-2-Hydroxy-cyclopent-4-en- 1 -yl-essig- säure-( - )-a-phenyl-äthylaminsalz.

Beispiel 13
26,3 g (0,100 Mol) (-)-cis-2-Hydroxy-cyclopent-4-en-1- yl-essigsäure-(+)-a-phenyl-äthylaminsalz werden in 80 ml Wasser gelöst. Der pH-Wert der Lösung wird mit cc. Salzsäure-Lösung auf 1 eingestellt, dann wird die Lösung eine Stunde lang bei Zimmertemperatur gerührt und schliesslich mit 3 x 40 ml Dichlormethan extrahiert. Die vereinigte organische Lösung wird mit wasserfreiem Natriumsulfat getrocknet und dann eingeengt. Als Rückstand erhält man 11,9 g (- )-cis-2-Hydroxy-cyclopent-4-en- 1 -yl-essigsäure-lacton.

Beispiel 14
Die, gemäss Beispiel 13 erhaltene lactonfreie wässrige Phase wird mit 20% Natriumhydroxyd-Lösung gelaugt und mit 3 x 25 ml Dichlormethan extrahiert. Die vereinigte organische Phase wird getrocknet und eingeengt. Als Rückstand erhält man 13,8 g (+)-Phenyl-äthylamin.

1. The optically active and racemic cis-2-hydroxycyclopent-4-en-1-yl-acetic acid lactones of the general formula I are used for the preparation of prostaglandins and other compounds with a cyclopentene skeleton, such as, for example, cis-jasmon (JACS , 95, 6382. [1973], or J. Org. Chem., 37, 2363. [1972]). The compounds of the general formula I are prepared in the following way: cyclopentadiene and dichloroacetyl chloride react with one another in the presence of triethylamine, as a result of which the dichloro ketone formed is added to the cyclopentadiene and a bicyclic dihalogen ketone is formed.
This compound is dehalogenated under reduction, then the racemic compound of the general formula I is prepared by treating the resulting bicyclic ketone under the same circumstances as in the Baeyer-Villiger oxidation (Tetrahedron Letters 307. [19703).
For the production of prostaglandin derivatives of natural configuration, however, it is not advantageous to use the racemic compound of the general formula I as the starting material, because it also means that the enantiomeric compound must be carried out as fiber in the synthesis, and that means besides the reduction in the yield that the Separation at a later step of the synthesis must be done with more sensitive substances.
The separation of the compound of general formula I by using the optically active a-phenyl-ethylamine is described in the literature (J.A.C.S. 95, 6832. [1973]).
According to the method described in the literature, the following procedure is followed: the racemic compound of the general formula I is mixed with an equimolar amount
hydrolysed aqueous base, the solution obtained from the salt of racemic acid of the general formula II is acidified at 0 ° C. in the presence of ethyl acetate to a pH of 3.5-4 and the racemic acid of the general formula II which has been liberated is extracted with ethyl acetate. The ethyl acetate solution obtained is dewatered at 0 ° C. To this end, the optically active base ((+) - a-phenyl-ethylamine) is added in a 5% excess. The crystalline salt which has separated out is purified by recrystallization.
The salt obtained is dissolved in the equimolar amount of an aqueous base, the (+) - a-phenylethylamine released in this way is removed by extraction with ether, and then the aqueous solution obtained is at pH and at 20 ° C. with Iodine reacts and thereby converted into a so-called iodine lactone.
There is also a known method (Hungarian patent application No. CI-1678) for purifying the compounds of general formula I or II by salt formation. This process also enables the production of alkali metal and alkaline earth metal salts of the acid of the general formula II.
The common disadvantage of all these processes is that the separation and salt formation cannot be solved with the help of them in the amounts necessary for industrial production. Although the processes disclosed secure the production of pure products, they would require a large volume of solution to be worked up if the quantities were to increase because of the relatively small solution concentrations, which would be unjustified. Surprisingly, it has now been found that the alkali metal salts and the lactone of the racemic cis-2-hydroxy-cyclopent-4-en-1-yl-acetic acid are also separated from the optically active -phenyl-ethylamine in a manner which is detachable for industrial quantities can, if the method according to claim 1 is carried out.
The racemic or, to a small extent, optically active cis-2-hydroxy-cyclopent-4-en-1-yl-acetic acid salt formed during the process can be recycled into the separation process.
When carrying out the separation, the racemic lactone can be selected as the starting material, but in an even more advantageous process, the racemic lactone is dissolved in aqueous alkali metal hydroxide solution and the resulting solution of the alkali metal salt is separated. In the process according to the invention, sodium or potassium hydroxide can equally be used as the alkali metal hydroxide. It is advantageous to use sodium hydroxide.
What has been stated for the alkali metal hydroxides also applies analogously to the alkali metal salts.
In the process according to the invention, the aromatic and aliphatic hydrocarbons and their various halogenated derivatives are primarily considered as water-immiscible solvents. However, any organic solvent which is immiscible with water and is inactive towards the reactants and which dissolves the cis-2-hydroxy-cyclopent-4-en-1-yl acetic acid lactone can be used. Dichloroethane and dichloromethane have been found to be extremely advantageous.
In the process according to the invention, it is advantageous to use 0.3-1.5 mol of the optically active a-phenylethylamine for 1 mol of racemic lactone. If the amount of the separating agent does not reach the stoichiometric amount [based on the lactone], it is advantageous if the separation is carried out in the presence of the organic solvent mentioned above and so the other acid antipode, which is in the form of a lactone not excreted as a diastereomeric salt is present is kept in solution.
A lower alkanol is used as the water-miscible organic solvent used in the process. Such alkanols are the optionally straight or branched aliphatic alkanols with 14 carbon atoms, such as, for example, methanol, ethanol, n- and i-propanol, and also n-, sec- and tert-butanol. The use of ethanol is very advantageous.

Example 2
The two-phase mother liquor obtained in Example 1 is cc with 20 ml. Hydrochloric acid boiled under reflux. After cooling and separation of the organic phase, the aqueous phase is extracted with 2 x 100 ml of chloroform.

Example 3
The lactone of formula I obtained in Example 2 is dissolved in 45 ml of absolute ethyl alcohol and 2.2 g (0.055 mol) of solid, granulated sodium hydroxide are added, and then the mixture is left to stand at room temperature for one night (12 hours) .

To the mother liquor obtained in Example 5, 30 ml of chloroform and 5 ml of cc. Hydrochloric acid added and afterwards it is refluxed for one hour. A mixture of 11.4 g (0.092 mol) of (+) -a-phenyl-ethylamine and 80 ml of water is added to a pH of 7 cc. Hydrochloric acid solution added. Example 10
To 4.52 g (0.036 mol) of racemic cis-2-hydroxy-cyclopent-4-en-1-yl-acetic acid lactone (racemic compound of the formula I) are added 4.8 g (0.039 mol) (+) - a-Phenyl-ethylamine and 30 ml of water added.
