Description
Die Erfindung betrifft neue Riech- und/oder Geschmackstoffe. Es handelt sich dabei um Verbindungen der Formel worin R Butyl, But-2-en-yl oder 1,3-Butadienyl bedeutet.

Die Formeln sollen sämtliche im Hinblick auf die vorhandene cis/trans-Isomerie möglichen geometrischen Isomeren umfassen.
Die Erfindung betrifft ferner ein Verfahren zur Herstellung der Verbindungen der Formel I.
Dieses Verfahren ist dadurch gekennzeichnet, dass man entweder a) 4-(2,6,6-T rimethyl-3-oxo-1 -cyclohexen-1 -yl)-but-3-en-2-ol in saurem Medium erhitzt und die exocyclischen Doppelbindungen des erhaltenen Reaktionsproduktes der Formel Ia gegebenenfalls hydriert, um zu der Verbindung der Formel Ic zu gelangen, oder dass man b) 3-Oxodihydro-ß-jonylacetat der Pyrolyse unterwirft, um zu der Verbindung der Formel Ib zu gelangen.
Das saure Medium gemäss Verfahrensvariante a) kann unter Zuhilfenahme von Mineralsäuren, wie Schwefelsäure oder Salzsäure, von sauren Salzen, wie Bisulfaten, beispielsweise KHSO4, oder sauren Erden, wie Diatomeenerde, z.B. Filtrol, erzeugt werden. Aber auch geeignete organische Säuren, wie mittelstarke bis starke solcher Säuren kommen in Frage. Beispiele hierfür sind Alkansulfonsäuren, wie Me-thansulfonsäure, oder Pikrinsäure.
Die Reaktionstemperatur ist nicht kritisch. Sie beträgt zweckmässigerweise ca. 20-200 °C, vorzugsweise 60-130 °C. Sie liegt zweckmässigerweise höher bei Verwendung schwächerer Säuren und niedriger bei Verwendung stärkerer Säuren.
Die Umsetzung wird vorzugsweise unter Zuhilfenahme eines organischen Lösungsmittels, insbesondere eines aromatischen Lösungsmittels, durchgeführt. Beispiele sind Benzol, Toluol, Xylol, etc.
Die Hydrierung der exocyclischen Doppelbindungen der Verbindung der Formel Ia kann nach an sich bekannten Methoden, also zweckmässigerweise katalytisch durchgeführt werden. Geeignete Katalysatoren sind Edelmetalle, wie Pd, Pt, Rh, oder Ni, z.B. Raney-Nickel, etc. Man arbeitet vorzugsweise in Anwesenheit eines Lösungsmittels, wie Wasser, Äthanol, Methanol, Dioxan, etc.
Die Reaktion wird zweckmässigerweise bei Raumtemperatur durchgeführt, es kommen aber auch tiefere oder höhere Temperaturen in Frage - je nach eingesetztem Katalysator und Lösungsmittel, z.B. der Bereich von 5° bis 100 °C.
Die Pyrolyse des bekannten 3-Oxo-dihydro-ß-jonylace-tats [siehe z. B. H. Jde, S. Toki, Biochem. J. 119,281 (1970)] zwecks Herstellung der Verbindung Ib gemäss Verfahrensvariante b) wird zweckmässigerweise bei Temperaturen von 350-700 °C, insbesondere bei 400-500 °C durchgeführt.
Man arbeitet zweckmässigerweise in Inertgasatmosphäre, z.B. unter Stickstoff.
Bei der Herstellung der Verbindungen Ia und Ib fallen üblicherweise Gemische der cis/trans-Isomeren an. Im Falle Ia überwiegt das trans-Isomere (~ 93%) stark, im Falle von Ib ist das Verhältnis trans/cis = ~ 4 : 1. Eine Auftrennung der Gemische in die einzelnen Isomeren ist nicht nötig, jedoch möglich. Hierbei kann von den bekannten Techniken, wie Säulenchromatographie, präp. Gaschromatographie, etc. Gebrauch gemacht werden.

Die Verbindungen I weisen besondere organoleptische Eigenschaften auf, auf Grund derer sie sich in vorzüglicher Weise, insbesondere in reiner Form oder in Form von Gemischen, also mit Ausnahme von natürlichen, Verbindungen I enthaltenden Gemischen, als Riech- und/oder Geschmackstoffe verwenden lassen.
Unter reinen I sollen solche I verstanden werden, die frei von Begleitstoffen sind, wie sie neben I in natürlichen Extrakten vorliegen. Als reine I im Sinne der vorliegenden Erfindung sollen deshalb insbesondere synthetisch hergestellte I verstanden werden.
Es hat sich nämlich erwiesen, dass die Verbindungen Ia und Ib in äusserst geringen Mengen im Osmanthus absolue vorkommen. In Helv. Chim. Acta 61(7), (1978), Seiten 2328-2335 berichten Kaiser & Lamparsky über die Isolierung und Identifizierung der bisher unbekannten Stoffe Ia und Ib. Dieses natürliche Vorkommen von Verbindungen I [d.h. die natürlichen, Verbindungen der Formel I - in nichtisolierter Form - enthaltenden Gemische] bilden jedoch nicht Gegenstand der vorliegenden Erfindung.
Die als Riech- und/oder Geschmackstoffe verwendbaren Verbindungen I - insbesondere Ia - zeichnen sich durch feine, teeartige, würzige, an Trockenfrüchte erinnernde, leicht holzige Geruchsnuancen aus. Die Verbindungen können demgemäss beispielsweise zur Parfümierung bzw. Aromatisierung von Produkten, wie Kosmetika (seifen, Salben, Puffern, etc.), Detergentien, bzw. Nahrungsmitteln, Genussmitteln und Getränken dienen, wobei die Verbindungen vorzugsweise nicht allein, sondern in Form von Kompositionen mit anderen Riech- bzw. Geschmackstoffen eingesetzt werden. Solche Riech- bzw. Geschmackstoffkompositionen mit einem Gehalt an Verbindungen I bilden ebenfalls gegenständ der Erfindung.
Ihre Herstellung kann auf an sich bekannte Art erfolgen, also z.B. durch Zugabe von I zu bekannten Riech- bzw. Geschmackstoffkompositionen oder Vermischung von I mit als Bestandteil von Riech- bzw. Geschmackstoffkompositionen geeigneten natürlichen oder synthetischen Verbindungen oder Gemischen.
Als Riechstoffe eignen sich die Verbindungen I, insbesondere Ia, auf Grund ihrer sehr natürlichen Noten, insbesondere in Kombination mit einer Reihe von natürlichen und synthetischen Riechstoffen wie z.B. Galbanum, Mastix, Vetiveröl, Patchouliöl, Bergamotteöl, Petitgrainöl, Basili-kumöl, Baummoos absolue, Jasmin absolue, Aldehyden, Acetalen, Alkoholen, Estern, schwefelhaltigen Verbindungen, wie p-Menthan-8-thiol-3-on, etc., stickstoffhaltigen Verbindungen, wie Indol, etc., oder 5-Methylheptan-3-onoxim und Moschuskörpern.
Die unter Verwendung von Verbindungen I hergestellten Riechstoffkompositionen bestechen insbesondere durch ihre eindrückliche Frische. Diesbezüglich unterscheiden sich die neuen Verbindungen I in grundlegender Weise von den bekannten Megastigmatrienonen ähnlicher Struktur (Acta Chem. Scand. 26, 2573 (1972), USP 3268 589, USP 3 217 718). Diese bekannten Trienone der Formeln

führen in den Kompositionen zu schweren, süssen Geruchseindrücken.
Die Konzentration der Verbindungen I kann je nach dem Verwendungszweck innerhalb weiter Grenzen variieren, beispielsweise zwischen etwa 0,01 (Detergentien) und etwa 15 Gew.% (alkoholische Lösungen). In Parfumbasen bzw. Konzentraten können die Konzentrationen selbstverständlich auch höher liegen. Die Parfumbasen können in üblicher Weise zur Parfümierung von Eaux de Cologne, Eaux de toilette, Lotionen, Crèmes, Shampoos, Seifen und Detergentien, etc. verwendet werden.
Als Geschmackstoffe können die Verbindungen I beispielsweise zur Erzeugung bzw. Verbesserung, Verstärkung, Steigerung oder Modifizierung von Frucht- oder Beerenaromen in Nahrungsmitteln (Joghurt, Süsswaren, etc.); in Genussmitteln (Tee, etc.) und Getränken (Limonaden, etc.) verwendet werden.
Die ausgeprägten geschmacklichen Qualitäten von insbesondere praktisch reinen, insbesondere von synthetisch hergestellten Verbindungen I ermöglichen die Verwendung in geringen Konzentrationen. Eine geeignete Dosierung um-fasst beispielsweise den Bereich von 0,1-100 ppm, vorzugsweise von 1-20 ppm im Fertigprodukt, d.h. dem aromatisierten Nahrungsmittel, Genussmittel oder Getränk.
In der folgenden Tabelle sind einige Effekte zusammengestellt, wie sie sich mit den Ketonen I erzielen lassen.

Die Verbindungen können auf übliche Weise mit den für Geschmackstoffkompositionen verwendeten Bestandteilen vermischt bzw. solchen Aromen zugesetzt werden. Unter den erfindungsgemäss verwendeten Aromen werden Geschmackstoffkompositionen verstanden, die sich auf an sich bekannte Art verdünnen bzw. in essbaren Materialien verteilen lassen. Sie können nach an sich bekannten Methoden in die üblichen Gebrauchsformen, wie Lösungen, Pasten oder Pulver übergeführt werden. Die Produkte können sprühgetrocknet, vakuumgetrocknet oder lyophilisiert werden.
Für die Herstellung solcher üblicher Gebrauchsformen kommen beispielsweise folgende Trägermaterialien, Verdik-kungsmittel, Geschmackstoffverbesserer, Gewürze und Hilfsingredientien, etc. in Frage:
Gummi arabicum, Tragant, Salze oder Brauereihefe, Al-ginate, Carrageen oder ähnliche Absorbentien; Indole, Mal-tol, Gewürzoleoresine, Raucharomen; Gewürznelken, Na-triumcitrat; Mononatriumglutamat, Dinatrium-inosin-5'-monophosphat (IMP), Dinatriumguanosin-5-phosphat (GMP); oder spezielle Aromastoffe, Wasser, Äthanol, Pro-pylenglykol, Glycerin.
Beispiel 1
Eine Lösung von 70 g rohem 3-Oxo-ß-jonol und 1,5 g para-Toluolsulfonsäure in 300 ml Toluol wird während 40 Minuten unter gleichzeitiger Entfernung des Reaktionswassers mit Hilfe eines Wasserabscheiders bei Rückflusstemperatur gerührt. Man versetzt das abgekühlte Reaktionsgemisch mit 300 ml Äther, wäscht die organische Phase dreimal mit Sodalösung und dreimal mit Wasser, trocknet und engt ein. Es verbleiben 61g Rohprodukt, welche gemäss gaschromatographischer Analyse > 90% 2,4,4-Trimethyl-3-(buta-l,3-dienyl)cyclohex-2-en-l-on enthalten. Die Destillation des Rohproduktes über eine 15-cm-Kolonne ergibt 45 g olfaktisch gutes 2,4,4-Trimethyl-3-(buta-l,3-dienyl)-cyclo-hex-2-en-l-on vom Siedepunkt 74-75°/Ö,06 mmHg.

Das als Ausgangsmaterial verwendete 3-Oxo-ß-jonol kann wie folgt erhalten werden:
Zu einer auf 10° gekühlten und gut gerührten Suspension von 192 g (1 Mol) a-Jonon und 198 g wasserfreiem Natrium-acetat in 1 Liter Methylenchlorid lässt man im Verlaufe einer Stunde 200 g (~ 1,05 Mol) 40%ige Peressigsäure so zutrop-fen, dass die Reaktionstemperatur von 20° nicht überschritten wird. Anschliessend entfernt man die externe Kühlung und rührt das Reaktionsgemisch während 5 Stunden bei Raumtemperatur. Im Verlaufe der ersten Stunde steigt die Temperatur des Reaktionsgemisches auf 28° und fällt hierauf wieder langsam ab. Das Reaktionsgemisch wird durch Filtration von Natriumacetat befreit und die klare Lösung zweimal mit Wasser, viermal mit Sodalösung und nochmals mit Wasser bis zum Neutralpunkt gewaschen. Nach Trocknen mit Natriumsulfat und Einengen verbleiben 215 g Rohprodukt, die gemäss gaschromatographischer Analyse > 94% a-Jononepoxid enthält.
Zu einer Lösung von 215 g rohem a-Jononepoxid (~ 1 Mol) in 700 ml Methanol lässt man im Verlaufe von 5 Minuten eine frisch bereitete Natriummethylatlösung (durch Auflösen von 2,9 g Natrium in 100 ml Methanol erhalten) zu-tropfen. Anschliessend rührt man das Reaktionsgemisch während 3 Stunden bei der Rückflusstemperatur des Methanols, destilliert die Hälfte des Methanols unter dem Wasserstrahlvakuum ab, versetzt das Konzentrat mit der 4-fachen Menge Wasser und extrahiert das Reaktionsprodukt mit Äther. Nach Trocknen und Einengen der Ätherphase verbleiben 213 g Rohprodukt, welche gemäss gaschromatographischer Analyse > 85% 4-(2,6,6-Trimethyl-3-hydroxycyclo-hex-l-en-l-yl)-(but-3-en-2-on enthalten. Die Destillation des Rohproduktes über eine 15-cm-Widmer-Kolonne ergibt 155 g > 94%iges Produkt (Ausbeute: 75% bezogen auf eingesetztes a-Jonon) mit einem Siedepunkt von 131-132°/ 0,05 mmHg.

Zu einer auf 0° vorgekühlten Lösung von 140 g (0,068 Mol) 4-(2,6,6-T rimethyl-3-hydroxycyclohex-1 -en-1 -yl)-but-3-en-2-on in 500 ml Äther lässt man im Verlaufe von 10 Minuten 700 ml einer auf 0° gekühlten wässrigen Lösung von 202,6 g Natriumdichromat-dihydrat und 186 g konzentrierter Schwefelsäure unter intensiver Kühlung und gutem Rühren so zutropfen, dass eine Reaktionstemperatur von 5° nicht überschritten wird. Anschliessend rührt man während weiteren 5 Minuten bei 5°, versetzt mit 500 ml Äther, wäscht die Ätherphase dreimal mit Wasser, dreimal mit Sodalösung

und nochmals dreimal mit Wasser, trocknet mit Natriumsulfat und engt ein. Die Destillation der erhaltenen 125 g Rohprodukt über eine 10-cm-Widmer-Kolonne ergaben 103 g > 94%iges 4-(2,6,6-Trimethyl-3-oxo-cyclohex-l-en-l-yl)-but-3-en-2-on vom Siedepunkt 109-110°/0,05 mmHg, Smp. 50-51°.

Zu einer auf 10° gekühlten Lösung von 72 g (0,35 Mol) 4-(2,6,6-T rimethyl-3-oxo-cyclohex-1 -en-1 -yl)-but-3-en-2-on in 480 ml Methanol gibt man im Verlaufe von 10 Minuten unter gleichzeitigem Rühren und externer Kühlung portionenweise insgesamt 4,0 g (0,106 Mol) Natriumborhydrid so zu, dass eine Reaktionstemperatur von 13° nicht überschritten wird. Anschliessend rührt man das Reaktionsgemisch während weiteren 15 Minuten bei 10°, versetzt hierauf mit der 4-fachen Menge Wasser und extrahiert das Reaktionsprodukt mit Äther. Nach Trocknen und Einengen der Ätherlösung verbleiben 73 g Rohprodukt, die gemäss gaschromatographischer Analyse zu > 90% aus 4-(2,6,6-Trime-thyl-3-oxo-cyclohex-1 -en-1 -yl)-but-3-en-2-ol (3-Oxo-ß-Jonol) bestehen, enthält. 3 g des Rohproduktes werden auf säulenchromatographischem Wege gereinigt. Zur Charakterisierung dient das > 97%ige Produkt.

Beispiel 2
Als Pyrolyseapparatur dient ein Quarzrohr von 50 cm Länge und 5 cm Durchmesser, welches mit Glasringen von 2-4 mm Durchmesser gefüllt und mit einem Heizmantel auf eine Temperatur von 500° konditioniert wird.
8,0 g (0,034 Mol) 3-Oxodihydro-ß-jonylacetat werden in 30 ml Benzol gelöst und im Verlaufe von 20 Minuten unter gleichzeitigem Durchsatz eines Stickstoffstromes von 0,31/ Minute in das Pyrolyserohr getropft. Das Pyrolysegut wird in einem gekühlten Gefass kondensiert, mit 30 ml Hexan verdünnt, zweimal mit einer Natriumbicarbonat-Lösung und zweimal mit Wasser gewaschen, getrocknet und eingeengt. Die Destillation des so erhaltenen Rohproduktes (5,0 g) über eine 5-cm-Widmer-Kolonne ergibt 3,5 g Produkt vom Siedepunkt 69-70°/0,05 mmHg, das sich zu 80% aus 2,4,4-T rimethyl-3-(but-trans-2-en-1 -yl)-cyclohex-2-en-1 -on und zu 20% aus dem entsprechenden cis-Isomeren zusammensetzt. Zur Charakterisierung gelangt eine mittels präpa-rativer GC gereinigte Probe:

Die Verbindung weist einen balsamischen, camphrigen, fruchtigen, holzigen Geruch auf und erinnert an gewisse Aspekte von Myrrhe und Opoponax.
Beispiel 3
Eine Lösung von 3,80 g (0,02 Mol) des nach Beispiel 1 hergestellten 2,4,4-Trimethyl-3-(buta-l ,3-dienyl)-cyclohex-2-en-l-ons in 20 ml Äthanol wird mit 0,1 g Katalysator (Palladium, 10% auf Aktivkohle) versetzt und bis zur Aufnahme von 0,04 Mol Wasserstoff bei Normaldruck hydriert. Nach Filtration, Einengen und Kugelrohrdestillation resultieren 3,2 g Produkt, die gemäss gaschromatographischer Analyse zu über 85% aus 2,4,4-Trimethyl-3-butyl-cyclohex-2-en-l-on bestehen.

Beispiel 4
Analog Beispiel 3 lässt sich auch das nach Beispiel 2 hergestellte 2,4,4-Trimethyl-3-(but-trans-2-en-l-yl)-cyclohex-2-en-l-on katalytisch zu 2,4,4-Trimethyl-3-butyl-cyclohex-2-en-l-on hydrieren. Ausbeute: 92%.
In den folgenden Beispielen 5-17 wird gezeigt, wie eine breite Palette von Riechstoffbasen durch Zugabe der neuen Verbindungen der Formel I in wünschenswerter Weise veredelt bzw. verbessert werden können, oder auch Basen ganz neuer Geruchsrichtungen erzeugt werden können. In den Kompositionen der Beispiele 5-15 konnte andererseits der durch I erzielte Effekt durch Zugabe derselben Menge Me-gastigmatrienon (in Form seiner Gemische) nicht erhalten werden. Allen Kompositionen fehlte insbesondere die charakteristische Frische, die ihnen durch den Zusatz der neuen Verbindungen I vermittelt wird. Besonders auffallend ist ausserdem die Tatsache, dass die neuen Verbindungen I in Kombination mit bestimmten bekannten Parfümerierohstof-fen (vgl. Beispiele 8,12) zu einer vorher nicht beobachteten Tabak-Note führen, die sich durch Zugabe der als Tabakin-haltsstofife bekannten Megastigmatrienone erstaunlicherweise nicht realisieren lässt.

Gibt man zu dieser Grünbase 160 Gewichtsteile Ia, so wird das etwas harte Grün der Base sehr schön abgerundet. Wirkt in der Grünbase hauptsächlich das Galbanum geruchsbestimmend, so wird mit dem Zusatz von Ia das weiche Grün des Mastix absolut angenehm unterstrichen. Die Base wirkt nun viel blumiger.

Dominiert in der ursprünglichen Base das Citronellol, so ist in der neuen Base das Geraniol im Vordergrund. Die neue Base wirkt viel blumiger. Sie hat mehr Strahlungskraft und Diffusion.

Claims (8)
1. Verbindungen der Formel worin R Butyl, But-2-enyl oder 1,3-Butadienyl bedeutet.
2. 2,4,4-Trimethyl-3-(buta-l,3-dienyl)-cyclohex-2-en-l-on als Verbindung nach Anspruch 1.

PATENTANSPRÜCHE
3. Verbindungen nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass sie in reiner Form vorliegen.
4. Riech- und/oder Geschmackstoffkompositionen, gekennzeichnet durch einen Gehalt an einer Verbindung der Formel worin R Butyl, But-2-enyl oder 1,3-Butadienyl bedeutet, mit Ausnahme der natürlichen, Verbindungen der Formel I enthaltenden Gemische.
5. Riech- und/oder Geschmackstoffkompositionen nach Anspruch 4, gekennzeichnet durch einen Gehalt an 2,4,4-Trimethyl-3-(buta-l,3-dienyl)-cyclohex-2-en-l-on, mit Ausnahme der natürlichen 2,4,4-Trimethyl-3-(buta-l,3-dienyl)-cyclohex-2-en-l-on enthaltenden Gemische.
6. Verfahren zur Herstellung von 2,4,4-Trimethyl-3-(buta-l,3-dienyl)-cyclohex-2-en-l-on der Formel dadurch gekennzeichnet, dass man 4-(2,6,6-Trimethyl-3-oxo-l-cyclohexen-l-yl)-but-3-en-2-ol in saurem Medium erhitzt.
7. Verfahren zur Herstellung von 2,4,4-Trimethyl-3-(but-2-en-l-yl)-cyclohex-2-en-l-on der Formel dadurch gekennzeichnet, dass man 3-Oxodihydro-ß-jonyl-acetat der Pyrolyse unterwirft.
8. Verfahren zur Herstellung einer Verbindung der Formel
Ic dadurch gekennzeichnet, dass man nach dem Verfahren gemäss Anspruch 6 eine Verbindung der Formel Ia herstellt und anschliessend die exocyclischen Doppelbindungen hydriert.
