Abstract

   PATENTANSPRÜCHE
1. Einrichtung für einen Fermentationsapparat zur sterilen Probenahme von Kulturflüssigkeit aus dem Fermentationsapparat, die einen Probenehmer, der an einem   Formen    tationsapparat in der Zone der Vermischung der Kulturflüssigkeit anbringbar ist und einen sterilen Probeauffänger enthält, dadurch gekennzeichnet, dass der Probeauffänger (11) bei der Probenahme am Probenehmer (1) aufgestellt und mit diesem verbunden ist, wobei der Probenehmer (1) als im Ruhezustand geschlossenes Ventil ausgeführt ist,

   an dessen Gehäuse (4) an der Seite der Berührung mit dem Probeauffänger   (11)    ein Ringansatz (8) ausgeführt ist und der vakuumierbare Probeauffänger am Hals ein-im Ruhestand geschlossenes Ventil (12) aufweist und an dessen Gehäuse (13) an der Seite der Berührung mit dem Probenehmer (1) eine Ringnut (17) mit einer Dichtung (18) ausgeführt ist, welche Ringnut mit dem Ringansatz (8) des Probenehmers (1) zusammenfügbar ist, welche beide bei Deckung und Wechselwirkung einen hermetisch abgeschlossenen Kanal (19) für den Durchlauf der Kulturflüssigkeit aus dem Fermentationsapparat (3) in den Probeauffänger   (11) bilden.   



   2. Einrichtung nach Anspruch 1, dadurch gekennzeichnet, dass das in Schliessstellung sich befindliche Ventil (12) mit dem Hals des Probeauffängers (11) mittels eines elastischen Einsatzes (20) lösbar verbunden ist.



   3. Einrichtung nach Anspruch 2, dadurch gekennzeichnet, dass im elastischen Einsatz (20) zwischen dem im Ruhezustand geschlossenen Ventil (12) und dem Hals des Probeauffängers (11) ein Bakterienfilter (21) vorgesehen ist.



   Die vorliegende Erfindung bezieht sich auf eine Einrichtung zur sterilen Probenahme aus einem Fermentationsapparat gemäss Anspruch 1.



   Die vorliegende Erfindung kann in der mikrobiologischen, medizinischen, chemischen, Nahrungsmittelindustrie sowie in der Forschungspraxis zur sterilen Probenahme einer Kul   turflüssigkeit    verwendet werden.



   Eine sterile Entnahme der Kulturflüssigkeit aus einem Fermentationsapparat ist zur Durchführung der Kontrolle des Zustandes des zu untersuchenden Objektes und zur   Bestim-    mung der Bedingungen der Umgebung seiner Lebenstätigkeit notwendig. Sämtliche Labor- und   lndustriefermentationsap-    parate sind mit Einrichtungen zur sterilen Probenahme der Kulturflüssigkeit versehen. Ungeachtet eines bedeutenden Unterschieds in den Konstruktionen der Fermentationsapparate, unterscheiden sich die Einrichtungen zur sterilen Probenahme der Kulturflüssigkeit konstruktiv wenig voneinander.



  Die Probenahme der Kulturflüssigkeit aus einem Fermentationsapparat erfolgt manuell und umfasst solche Vorgänge, die gleichzeitig erfüllt werden müssen. In der Regel führen eine Probenahme zwei Forscher durch, dazu wird in der Flamme eines brennenden   Spiritusbrenners    der Stöpsel von einem sterilen Probeauffänger abgenommen und die Klemme eines Ablaufrohres der Kulturflüssigkeit geöffnet bzw. werden Ventile gesteuert. Die Dauer der Probenahme ermöglicht keine Analyse des Zustandes der   Übergangspro-    zesse der Entwicklung von Mikroorganismen, was in der Forschungspraxis besonders wichtig ist. Das letztere diente letzten Endes als Beginn der Suche nach neuen konstruktiven Lösungen der Einrichtungen zur sterilen Probenahme der Kulturflüssigkeit aus   einem-    Fermentationsapparat.



   Bekannt ist eine Einrichtung zur sterilen Probenahme aus einem Fermentationsapparat (siehe z.B. Katalog der Firma New Brunswick Scientific Co. Inc., Modell FM 250), die ein Ablaufrohr mit einem Ventil enthält, das am Fermentationsapparat angebracht ist, dem Ablaufstutzen dessen eine Rohrleitung zugeführt ist, durch welche Dampf eingeführt wird.



  Zur Durchführung der Probenahme der Kulturflüssigkeit aus dem Fermentationsapparat ist es notwendig, einen Probeauffänger, der mit einem Watte- bzw. leichtabnehmbaren Stöpsel geschlossen ist, im voraus zu sterilisieren. Danach wird der Probeauffänger dem Ablaufstutzen angeschlossen, zur Sterilisation mit Dampf behandelt und das Ventil wird zur Probenahme geöffnet.



   Bei Verwendung der genannten Einrichtung ist es notwendig, einen Teil der in der Rohrleitung befindlichen Kul   turflüssigkeit    abzugiessen, das führt zu unerwünschten Verlusten an Kulturflüssigkeit, insbesondere bei der Durchführung periodischer Prozesse der Züchtung von Mikroorganismen.



  Ausserdem ist es bei Benutzung der genannten Einrichtung notwendig, dass sich Kulturflüssigkeit im Fermentationsapparat unter einem Druck befindet, was bei der Züchtung von Mikroorganismen nicht immer günstig und möglich ist, bzw.



  ist es in der Ablaufleitung notwendig, eine Pumpe zum Umpumpen zu verwenden. Durch den Ablaufstutzen wird Dampf in den Raum abgelassen, was ebenfalls die Arbeit des Bedienungspersonals erschwert.



   Breit bekannt ist eine Einrichtung zur sterilen Probenahme aus einem Fermentationsapparat unter Verwendung von Antiseptika, z.B. von Chloramin, die eine Rohrleitung mit einem Ventil für den Durchlauf der Kulturflüssigkeit aus dem Fermentationsapparat und eine Rohrleitung mit einem Ventil für den Durchlauf des Antiseptikums enthält. Nach den Ventilen werden die Rohrleitungen in eine Rohrleitung vereint, die einen Ablaufstutzen besitzt. Als sterilisierte Probeauffänger verwendet man Reagenzgläser, die mit einem Wattestöpsel verschlossen sind. Zur Probenahme wird das Ventil zum Ablauf der Kulturflüssigkeit geöffnet und nachdem sie zum Teil abgegossen wird, öffnet man den Stöpsel des Probeauffängers, diesen unter den Strahl führend. Nachdem die erforderliche Menge der Kulturflüssigkeit entnommen ist, schliesst man den Probeauffänger mit dem Stöpsel und das Ablaufventil wird geschlossen.

  Danach wird das Ventil an der Rohrleitung mit Antiseptikum geöffnet und dieses wird zum Teil abgegossen, wobei das Antiseptikum im Ablaufstutzen bis zur nächsten Probenahme bleiben muss.



   Die genannte Einrichtung sieht einen zusätzlichen Ablauf der Stauungszone der Kulturflüssigkeit vor, was letzten Endes zu den Verlusten an der letzteren führt. Beim vorherigen Teilablauf der Kulturflüssigkeit ausserdem bleibt das Antiseptikum zum Teil an den Wandungen des Ablaufstutzens, das führt zum Gelangen des Antiseptikums in den Probeauffänger und zur Entstellung der Analyseergebnisse, insbesondere bei der Arbeit mit Fermenten.



   Bekannt ist eine Einrichtung zur sterilen Probenahme aus einem Fermentationsapparat, die einen Probenehmer, der am Fermentationsapparat in der Zone der Vermischung der Kulturflüssigkeit angebracht ist, sowie einen sterilen Probeauffänger enthält (siehe z.B. Katalog der Firma New Brunswick Scientific Co. Inc. BioFLO, Modell C 30).

 

   Die Einrichtung enthält weiter ein Ablaufrohr mit einer Schraubklemme, an dem eine Kammer mit einem Gummiball und einer Spritze befestigt ist, und eine Stossbaueinheit.



  Der sterilisierte Probeauffänger wird aufgestellt und an die Stossbaueinheit unter der Flamme eines Brenners angeschlossen. Die Probenahme geschieht wie folgt. Bei offener Schraubklemme drückt man an den Gummiball, dabei wird die Luft aus dem Probeauffänger in den Fermentationsapparat verdrängt. Beim Freilassen des Gummiballs wird seine Form wiederhergestellt und gleichzeitig wird die Kulturflüssigkeit aus dem Fermentationsapparat in den Probeauffänger eingesaugt. Danach wird die Schraubklemme geschlossen und unter der Flamme des Brenners wird der Probeauffänger  



  mit entnommener Probe gegen einen sterilen leeren Probeauffänger ausgetauscht.



   Die Verwendung der genannten Einrichtung fordert von einem Forscher bei Probenahme die Verwirklichung einer ganzen Reihe nacheinanderfolgend durchzuführender Vorgänge, was mit einem grossen Zeitaufwand verbunden ist.



  Das letztere gewährt keine Möglichkeit, eine grosse Zahl von Proben in einer Zeiteinheit zu entnehmen, was bei Untersuchung der Dynamik der Entwicklung von Mikroorganismen notwendig ist. Die Verwendung offener Flamme beim Anschliessen und bei der Abnahme des Probeauffängers erlaubt nicht, die genannte Einrichtung während der Arbeit mit explosionsgefährlichen Gasen zu benutzen, in deren Umgebung einige Arten von Mikroorganismen gezüchtet werden. Die Rückleitung der Luft eines Überdrucks in den Fermentationsapparat beim Druck auf dem Gummiball ist nicht immer erwünscht, während der Arbeit mit einem Objekt aber, das bei einem strengen Verhältnis der zuzuführenden Gase wächst, z.B. bei der Arbeit mit wasserstoffoxydierenden Bakterien, ist unzulässig.

  In der genannten Einrichtung ausserdem gibt es auch Verluste an Kulturflüssigkeit, welche vor der Probenahme aus der Stauungszone des Ablaufrohrs immer abgegossen werden muss.



   Der vorliegenden Erfindung liegt die Aufgabe der Entwicklung einer Einrichtung zur sterilen Probenahme der Kulturflüssigkeit aus einem Fermentationsapparat mit solcher Ausführung eines Probenehmers und seiner Verbindung mit einem Probeauffänger zugrunde, die die Möglichkeit gewähren, die Zeit der Probenahme wesentlich zu verkürzen, die Sterilität der Probe beizubehalten, die Verluste an Kulturflüssigkeit während der Probenahme auszuschliessen sowie während der Probenahme das Filtrat unmittelbar aus dem Fermentationsapparat zu erhalten und die Sterilität des Probeauffängers sichtbar zu machen.



   Die gestellte Aufgabe wird gelöst durch eine Einrichtung nach Anspruch 1.



   Es ist zweckmässig, das im Ruhezustand geschlossene Ventil am Hals des Probeauffängers mittels eines elastischen Einsatzes anzubringen.



   Es wird bevorzugt, im elastischen Einsatz zwischen dem normal geschlossenen Ventil und dem Hals des Probeauffängers ein Bakterienfilter anzubringen.



   Die erfindungsgemässe Einrichtung zur sterilen Probenahme aus dem Fermentationsapparat erlaubt, die Probenahme durch einen Forscher durchzuführen und dabei die Zeit, die zur Entnahme einer Probe in Anspruch genommen wird, wesentlich zu verkürzen. Der vakuumierte Probeauffänger ermöglicht die Beibehaltung der Sterilität der Probe und die Sicherung einer langen Lagerung der entnommenen sterilen Probe. Die Anbringung eines Bakterienfilters erlaubt, Filtrat bei Probenahme unmittelbar aus dem Fermentationsapparat zu erhalten. Das Fehlen irgendwelcher Stauungszonen gewährt die Möglichkeit, die Verluste an Kulturflüssigkeit bei Probenahme völlig auszuschliessen. Der am Hals des Probeauffängers aufgestellte elastische Einsatz macht die Dichtheit d.h. die Sterilität des Probeauffängers sichtbar, dabei wird die Probenahme in einen nicht sterilen Probeauffänger ausgeschlossen.



   Nachstehend wird die Erfindung durch ihre konkreten Ausführungsbeispiele mit Hinweisen auf beigefügte Zeichnungen erläutert, wobei es zeigen:
Fig. 1 erfindungsgemässe Einrichtung zur sterilen Probenahme aus einem Fermentationsapparat (Längsschnitt);
Fig. 2 dito wie in Fig. 1 mit einem elastischen Einsatz, in dem ein Bakterienfilter aufgestellt ist.



   Die Einrichtung zur sterilen Probenahme aus einem Fermentationsapparat enthält einen Probenehmer 1 (Fig. 1), der an einer Rohrleitung 2 angebracht ist, mittels dessen der Rückumlauf der Kulturflüssigkeit aus einem Fermentationsapparat 3 verwirklicht wird. Die Entnahme der Kulturflüssigkeit erfolgt aus der Zone einer aktiven Vermischung.



   Der Probenehmer   list    in Form eines im Ruhezustand geschlossenen Ventils ausgeführt, welches ein Gehäuse 4, eine Feder 5 und ein Absperrelement 6 enthält, das an einem Schaft 7 angebracht ist. Am Gehäuse 4 ist ein Ringansatz 8 ausgeführt. Das Gehäuse 4 des im Ruhezustand geschlossenen Ventils ist an der Rohrleitung 2 mittels einer Einlage 9 und einer Schraubenverbindung 10 angebracht (bedingt dargestellt).



   Die Einrichtung enthält weiter einen sterilen Probeauffänger 11, als solcher wurde in dem zu schildernden Beispiel ein vakuumiertes Reagenzglas verwendet. Am Hals des Probeauffängers 11 ist ein normal geschlossenes Ventil 12 aufgestellt, welches dem obengeschilderten ähnlich ist und ein Gehäuse 13, eine Feder 14 und ein Absperrelement 15 enthält, das an einem Schaft 16 angeordnet ist. Am Gehäuse 13 ist eine Ringnut 17 mit einer Dichtung 18 ausgeführt, welche mit dem Ringansatz 8 des Probenehmers 1 gekoppelt ist. Der Ansatz 8 und die Nut 17 bilden bei Deckung und Wechselwirkung einen hermetischen abgeschlossenen Kanal 19 für den Durchlauf der Kulturflüssigkeit aus dem Fermentationsapparat 3 in den Probeauffänger 11.



   Das im Ruhezustand geschlossene Ventil 12 ist am Hals des Probeauffängers 11 mittels eines elastischen Einsatzes 20 angebracht, der in dem zu schildernden Beispiel zusammen mit dem Ventil 12 innerhalb des Probeauffängers 11 untergebracht ist.



   Möglich ist ein Beispiel, wenn der elastische Einsatz 20 (Fig. 2) eine verlängerte Form hat und mit einer Seite den Hals des Probeauffängers 11, mit der anderen Seite aber das Gehäuse 13 des normal geschlossenen Ventils 12 umfasst. Es ist günstig, erstens, zur Anzeige des vorhandenen Vakuums im Probeauffänger 11, da in diesem Fall der elastische Einsatz 20 zusammengepresst oder verflacht ist, und zweitens, es ermöglicht die Verwendung der Behälter mit unterschiedlichem Fassungsvermögen als Probeauffänger 11, abhängig vom erforderlichen Volumen der zu entnehmenden Probe.



   Zur Adsorption der Mikroorganismen und zur Herstellung des Filtrats aus der Kulturflüssigkeit ohne Schleudern ist im elastischen Einsatz 20 zwischen dem normal geschlossenen Ventil 12 und dem Hals des Probeauffängers 11 ein Bakte   rienfilter    21 in einem Gehäuse 22 aufgestellt.



   Die Arbeitsweise der erfindungsgemässen Einrichtung zur sterilen Probenahme aus einem Fermentationsapparat ist die folgende:
Ein Forscher stellt den sterilen vakuumierten Probeauffänger 11 (Fig. 1) auf den Probenehmer 1 so   aul,    dass der Ringansatz des Probenehmers 1 in die Ringnut 17 des Probeauffängers 11 eingeht. Dabei wird der hermetisch abgeschlossene Kanal 19 gebildet, der mittels Dichtungsringes 18 abgedichtet wird. Dann beim Druck auf den Probeauffänger so, dass die Richtung der Beanspruchung mit der Längsachse des Ventils 12 zusammenfällt, erfolgt die Wechselwirkung der Schäfte 7 und 16. Infolgedessen drücken sich die Federn 5 und 14 zusammen, die Absperrelemente 6 und 15 öffnen sich und geben den Kanal 19 frei, durch den die Kulturflüssigkeit aus dem Fermentationsapparat 3 in den Probeauffänger 11 überströmt. 

  Nachdem die erforderliche Menge der Kulturflüssigkeit entnommen ist, entspannt der Forscher den Probeauffänger, dabei nehmen die Federn 5, 14 die normal geschlossene Ausgangslage der Absperrelemente 6 und 15 wieder ein.



   Eine sterile Probenahme aus dem Fermentationsapparat 3 mit nachfolgender Filtratherstellung geschieht wie folgt.  



   Aus einem Satz der sterilisierten und   vakuumierten Probe-    auffänger 11 wählt der Forscher die, in denen die elastischen Einsätze 20, welche als Indikator der Vakuumbeibehaltung im Probeauffänger 11 dienen, verflacht sind. Danach erfüllt der Forscher die Vorgänge zur Probenahme, die den obengeschilderten ähnlich sind. Die Kulturflüssigkeit strömt durch den hermetisch abgeschlossenen   Kanal 19    in die Zone A des elastischen Einsatzes 20 über. Nach der Probenahme entlastet der Forscher den Probeauffänger, die Absperrelemente 6, 15 kehren in die normal geschlossene Stellung zurück, dabei dehnt sich der elastische Einsatz 20 aus. Weiter wird der Probeauffänger 11 senkrecht mit dem Ventil 12 nach oben in einen Ständer aufgestellt. 

  Im Probeauffänger 11 und in der Zone A des elastischen Einsatzes 20 erfolgt der Druckausgleich auf Kosten des Durchlaufs der Kulturflüssigkeit aus der Zone A durch das Bakterienfilter 21 in den Probeauffänger 11. Der elastische Einsatz 20 verflacht sich dabei wieder.

   The present invention can be used in the microbiological, medical, chemical, food industry and in research practice for sterile sampling of a culture liquid.



   A sterile removal of the culture fluid from a fermentation apparatus is necessary to carry out the control of the state of the object to be examined and to determine the conditions in the surroundings of its life activity. All laboratory and industrial delivery devices are equipped with facilities for sterile sampling of the culture fluid. Regardless of a significant difference in the construction of the fermentation apparatus, the facilities for sterile sampling of the culture fluid differ little from one another in terms of design.



  Sampling of the culture fluid from a fermentation apparatus is done manually and includes processes that must be carried out at the same time. As a rule, two researchers take a sample. To do this, the plug is removed from a sterile sample collector in the flame of a burning spirit burner and the clamp of a drain pipe of the culture fluid is opened or valves are controlled. The duration of the sampling does not allow an analysis of the state of the transition processes in the development of microorganisms, which is particularly important in research practice. The latter ultimately served as the beginning of the search for new constructive solutions for the facilities for sterile sampling of the culture fluid from a fermentation apparatus.



   A device for sterile sampling from a fermentation apparatus is known (see, for example, the catalog of the New Brunswick Scientific Co. Inc., model FM 250), which contains a drain pipe with a valve which is attached to the fermentation apparatus and to the drain pipe of which a pipeline is fed through which steam is introduced.



  To carry out the sampling of the culture fluid from the fermentation apparatus, it is necessary to sterilize a sample catcher, which is closed with a cotton or easily removable stopper, in advance. Then the sample catcher is connected to the drain socket, treated with steam for sterilization and the valve is opened for sampling.



   When using the above-mentioned device, it is necessary to pour off part of the culture fluid in the pipeline, which leads to undesirable losses of culture fluid, in particular when carrying out periodic processes for growing microorganisms.



  In addition, when using the above-mentioned device, it is necessary for the culture fluid in the fermentation apparatus to be under a pressure, which is not always cheap and possible in the cultivation of microorganisms, or



  it is necessary in the drain line to use a pump for pumping around. Steam is released into the room through the outlet connection, which also complicates the work of the operating personnel.



   A device for sterile sampling from a fermentation apparatus using antiseptics, e.g. of chloramine, which contains a pipe with a valve for the passage of the culture liquid from the fermentation apparatus and a pipe with a valve for the passage of the antiseptic. After the valves, the pipelines are combined in a pipeline that has a drain socket. Test tubes sealed with a cotton plug are used as the sterilized sample collector. For sampling, the valve for the drainage of the culture fluid is opened and after it has been poured off in part, the stopper of the sample catcher is opened, guiding it under the jet. After the required amount of culture fluid has been removed, the sample collector is closed with the stopper and the drain valve is closed.

  Then the valve on the pipeline is opened with antiseptic and this is poured off in part, whereby the antiseptic must remain in the drain pipe until the next sampling.



   The said device provides an additional drainage of the stagnation zone of the culture fluid, which ultimately leads to the losses in the latter. In the previous partial drain of the culture fluid, the antiseptic also remains partly on the walls of the drain socket, which leads to the antiseptic getting into the sample catcher and to the distortion of the analysis results, especially when working with ferments.



   A device for sterile sampling from a fermentation apparatus is known, which contains a sampler, which is attached to the fermentation apparatus in the zone of mixing of the culture liquid, and a sterile sample collector (see, for example, the catalog of the New Brunswick Scientific Co. Inc. BioFLO, Model C 30).

 

   The device further includes a drain pipe with a screw clamp, to which a chamber with a rubber ball and a syringe is attached, and a butt assembly.



  The sterilized sample collector is set up and connected to the butt assembly under the flame of a burner. Sampling is done as follows. With the screw clamp open, press on the rubber ball, the air is forced out of the sample collector into the fermentation apparatus. When the rubber ball is released, its shape is restored and at the same time the culture liquid is sucked out of the fermentation apparatus into the sample collector. Then the screw clamp is closed and the sample catcher is placed under the flame of the burner



  with the sample removed, exchanged for a sterile empty sample collector.



   The use of the above-mentioned device requires a researcher to carry out a whole series of processes to be carried out in succession, which is time-consuming.



  The latter does not allow a large number of samples to be taken in one unit of time, which is necessary when examining the dynamics of the development of microorganisms. The use of an open flame when connecting and removing the sample collector does not permit the use of the device mentioned when working with explosive gases, in the vicinity of which some types of microorganisms are grown. Returning the air of an overpressure to the fermentation apparatus when pressure is on the rubber ball is not always desirable, but when working with an object that grows with a strict ratio of the gases to be supplied, e.g. when working with hydrogen-oxidizing bacteria, is not permitted.

  In the facility mentioned, there are also losses of culture fluid, which must always be poured out of the stowage zone of the drain pipe before sampling.



   The present invention is based on the object of developing a device for sterile sampling of the culture liquid from a fermentation apparatus with such a design of a sampler and its connection to a sample collector, which allow the possibility of significantly reducing the time of sampling, while maintaining the sterility of the sample, to exclude the loss of culture fluid during sampling and to receive the filtrate directly from the fermentation apparatus during sampling and to make the sterility of the sample collector visible.



   The object is achieved by a device according to claim 1.



   It is advisable to attach the valve, which is closed at rest, to the neck of the sample catcher by means of an elastic insert.



   It is preferred to attach a bacterial filter in the elastic insert between the normally closed valve and the neck of the sample collector.



   The device according to the invention for sterile sampling from the fermentation apparatus allows the sampling to be carried out by a researcher and thereby significantly reduces the time it takes to take a sample. The vacuum-sealed sample collector enables the sterility of the sample to be maintained and the sterile sample to be stored for a long time. The attachment of a bacterial filter allows filtrate to be obtained directly from the fermentation apparatus when sampling. The absence of any stagnation zones enables the loss of culture fluid during sampling to be completely ruled out. The elastic insert placed on the neck of the sample catcher makes the tightness, i.e. the sterility of the sample catcher is visible; sampling in a non-sterile sample catcher is excluded.



   The invention is explained below by its concrete exemplary embodiments with references to the attached drawings, in which:

   The device for sterile sampling from a fermentation apparatus contains a sampler 1 (FIG. 1), which is attached to a pipeline 2, by means of which the recirculation of the culture fluid from a fermentation apparatus 3 is realized. The culture fluid is removed from the zone of active mixing.



   The sampler is designed in the form of a valve which is closed in the idle state and which contains a housing 4, a spring 5 and a shut-off element 6 which is attached to a shaft 7. A ring extension 8 is implemented on the housing 4. The housing 4 of the valve which is closed in the idle state is attached to the pipeline 2 by means of an insert 9 and a screw connection 10 (shown conditionally).



   The device also contains a sterile sample collector 11, as such a vacuum test tube was used in the example to be described. A normally closed valve 12, which is similar to the one described above and contains a housing 13, a spring 14 and a shut-off element 15, which is arranged on a shaft 16, is set up on the neck of the sample catcher 11. On the housing 13 there is an annular groove 17 with a seal 18 which is coupled to the ring extension 8 of the sampler 1. The neck 8 and the groove 17 form a hermetically sealed channel 19 for the passage of the culture fluid from the fermentation apparatus 3 into the sample collector 11 when the cover and interaction are present.



   The valve 12, which is closed in the idle state, is attached to the neck of the sample catcher 11 by means of an elastic insert 20 which, in the example to be described, is accommodated together with the valve 12 within the sample catcher 11.



   An example is possible if the elastic insert 20 (FIG. 2) has an elongated shape and comprises the neck of the sample catcher 11 on one side, but the housing 13 of the normally closed valve 12 on the other side. It is favorable, firstly, to display the vacuum in the sample collector 11, since in this case the elastic insert 20 is compressed or flattened, and secondly, it enables the use of the containers with different capacities as the sample collector 11, depending on the volume required taking sample.



   To adsorb the microorganisms and to produce the filtrate from the culture fluid without spinning, a bacterial filter 21 is placed in a housing 22 in the elastic insert 20 between the normally closed valve 12 and the neck of the sample collector 11.



   The method of operation of the device according to the invention for sterile sampling from a fermentation apparatus is as follows:
A researcher places the sterile, vacuumed sample catcher 11 (FIG. 1) on the sampler 1 in such a way that the ring attachment of the sampler 1 enters the annular groove 17 of the sample catcher 11. The hermetically sealed channel 19 is formed, which is sealed by means of a sealing ring 18. Then when the sample catcher is pressed so that the direction of the stress coincides with the longitudinal axis of the valve 12, the shafts 7 and 16 interact. As a result, the springs 5 and 14 compress, the shut-off elements 6 and 15 open and give the Channel 19 free, through which the culture liquid flows from the fermentation apparatus 3 into the sample collector 11.

  After the required amount of the culture fluid has been removed, the researcher relaxes the sample catcher, the springs 5, 14 taking up the normally closed starting position of the shut-off elements 6 and 15 again.



   Sterile sampling from the fermentation apparatus 3 with subsequent production of the filtrate takes place as follows.



   The researcher chooses from a set of the sterilized and vacuumed sample catchers 11 those in which the elastic inserts 20, which serve as indicators of the vacuum maintenance in the sample catcher 11, are flattened. The researcher then performs the sampling procedures that are similar to those described above. The culture fluid flows through the hermetically sealed channel 19 into zone A of the elastic insert 20. After sampling, the researcher relieves the load on the sample catcher, the shut-off elements 6, 15 return to the normally closed position, the elastic insert 20 thereby expanding. Furthermore, the sample catcher 11 is placed vertically with the valve 12 upwards in a stand.

  In the sample catcher 11 and in zone A of the elastic insert 20, the pressure equalization takes place at the expense of the passage of the culture fluid from zone A through the bacterial filter 21 into the sample catcher 11. The elastic insert 20 flattens out again.
