Beschreibung
Hintergrund der Erfindung Erfindungsgebiet
Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von Halogenphenoxyfettsäu-re-Derivaten der Formel (I):
worin bedeuten:
R - eine Cyanogruppe;
- eine Gruppe -CON(R3)R4, worin R3 und R4, die gleich oder unterschiedlich sind, jeweils für ein Wasserstoffatom; eine niedere Alkylgruppe; eine Phenylgruppe; eine Phenylgruppe mit 1 bis 5 gleichen oder unterschiedlichen Substituenten aus der Gruppe Halogenatom, Nitrogruppe, Cyanogruppe, niedere Alkylgruppe, niedere Halogenalkylgruppe, niedere Alkoxygruppe, niedere Halogenalkoxygruppe, niedere Alkylthiogruppe und niedere Halogenalkylthiogruppe; eine Benzylgruppe oder eine Benzylgruppe mit 1 bis 5 gleichen oder unterschiedlichen Substituenten aus der Gruppe niedere Alkylgruppe, niedere Halogenalkylgruppe, niedere Alkoxygruppe, niedere Halogenalkoxygruppe, niedere Alkylthiogruppe und niedere Halogenalkylthiogruppe stehen, oder R3 und R4 bedeuten zusammen eine Alkylengruppe, oder - eine Gruppe -COAR5, worin R5 für ein Wasserstoffatom; eine niedere Alkylgruppe; eine Phenylgruppe; eine Phenylgruppe mit 1 bis 5 gleichen oder unterschiedlichen Substituenten aus der Gruppe Halogenatom, Nitrogruppe, Cyanogruppe, niedere Alkylgruppe, niedere Halogenalkylgruppe, niedere Alkoxygruppe, niedere Halogenalkoxygruppe, niedere Alkylthiogruppe und niedere Halogenalkylthiogruppe; eine Benzylgruppe oder eine Benzylgruppe mit 1 bis 5 gleichen oder unterschiedlichen Substituenten aus der Gruppe niedere Alkylgruppe, niedere Halogenalkylgruppe, niedere Alkoxygruppe, niedere Halogenalkoxygruppe, niedere Alkylthiogruppe und niedere Halogenalkylthiogruppe steht und A ein Sauerstoff- oder Schwefelatom darstellt,
R1 und R2 jeweils ein Wasserstoffatom oder eine niedere Alkylgruppe, wobei R1 und R2 gleich oder unterschiedlich sind,
X ein Halogenatom oder eine niedere Alkylgruppe und Y ein Halogenatom,
und sie bezieht sich weiterhin auf einen Teil der nach dem Verfahren erhältlichen Halogenphenoxyfett-säure-Derivate.
Der Ausdruck «niedere» Alkylgruppe oder eine ähnliche im vorliegenden Dokument bezeichnet eine Gruppe mit 1 bis 6 Kohlenstoffatomen.
Im allgemeinen werden Halogenphenoxyfettsäure-Derivate durch Halogenierung von Monohalogen-phenolen hergestellt, woraus Dihalogenphenole entstehen, und dann stellt man die Halogenphenoxyfett-säure-Derivate aus den erhaltenen Dihalogenphenolen her. Diese Dihalogenphenole erhält man nach dem Verfahren der japanischen Offenlegungsschriften (Kokai) Nrn. 60-193939, 62-223140 und 3-99033 sowie der japanischen Auslegeschrift (Kokoku) Nr. 3-22377.
Bei diesen Verfahren besteht jedoch die Gefahr, toxische Dioxine als Nebenprodukte zu erhalten. Ausserdem ist es notwendig, im Verfahren der japanischen Offenlegungsschrift 62-223140 Wasserstoffperoxid zu verwenden, stickstoffhaltige Basen im Verfahren der japanischen Auslegeschrift Nr. 3-22377, und Wasser und ein Zweiphasensystem im Verfahren der japanischen Offenlegungsschrift Nr. 3-99033 zusätzlich zu den Chlorierungsmittein.
Weiterhin ist ein Verfahren zur Herstellung von Halogenphenoxyfettsäure-Derivaten durch Halogenierung von Monohalogenphenoxyfettsäure-Derivaten bekannt geworden, bei dem man die Halogenierung mit Halogenierungsmitteln sowie Katalysatoren wie lod und Eisen ausführt (J.O.C., 426 (1946)). Bei diesem Verfahren tritt das Problem einer Nachbehandlung der verwendeten Katalysatoren auf.
Zusammenfassung der Erfindung
Die Erfinder haben intensive Forschungen durchgeführt, um ein neues Verfahren zur Herstellung von Halogenphenoxyfettsäure-Derivaten in hohen Ausbeuten ohne Verwendung von Katalysatoren zu entwickeln. Die vorliegende Erfindung stellt ein Ergebnis der Forschungen dar.

findung erhalten werden, sind als Zwischenprodukte für Pharmazeutika, Agrikulturchemikalien und andere chemische Produkte wertvoll, und einige von ihnen, die durch die folgende Formel (I') dargestellt werden, sind neue Verbindungen, die bisher in der Literatur noch nicht erwähnt wurden:

R' eine Cyanogruppe oder -CON(R3')R4', in der R3' und R4', die gleich oder unterschiedlich sind, jeweils für ein Wasserstoffatom; eine niedere Alkylgruppe; eine Phenylgruppe; eine Phenylgruppe mit 1 bis 5 gleichen oder unterschiedlichen Substituenten aus der Gruppe Halogenatom, Nitrogruppe, Cyanogruppe, niedere Alkylgruppe, niedere Halogenalkylgruppe, niedere Alkoxygruppe, niedere Halogenalkoxygruppe, niedere Alkylthiogruppe und niedere Halogenalkylthiogruppe; eine Benzylgruppe oder eine Benzylgruppe mit 1 bis 5 gleichen oder unterschiedlichen Substituenten aus der Gruppe niedere Alkylgruppe, niedere Halogenalkylgruppe, niedere Alkoxygruppe, niedere Halogenalkoxygruppe, niedere Alkylthiogruppe und niedere Halogenalkylthiogruppe stehen,
R1' und R2' jeweils ein Wasserstoffatom oder eine niedere Alkylgruppe, wobei R1' und R2' gleich oder unterschiedlich sind,
X' ein Fluoratom, ein lodatom, ein Bromatom oder eine niedere Alkylgruppe, und Y' ein Halogenatom, wobei
R' keine Cyanogruppe ist, wenn R1' und R2' ein Wasserstoffatom darstellen, X' ein Fluoratom und Y' ein Chloratom bedeutet.
Eingehende Beschreibung bevorzugter Ausführungsformen
Die erfindungsgemässen Halogenphenoxyfettsäure-Derivate können beispielsweise nach dem unten erläuterten Verfahren hergestellt werden:
worin R, R1, R2, X und Y die obenstehenden Bedeutungen haben.
Die angestrebten Halogenphenoxyfettsäure-Derivate können durch Halogenierung der Phenoxyfett-säuren gemäss Formel II mit Halogenierungsmitteln in Gegenwart inerter Lösungsmittel selektiv erhalten werden.
Die in der vorliegenden Erfindung verwendeten inerten Lösungsmittel können aus allen denen ausgewählt werden, die das Verfahren der obigen Reaktion nicht wesentlich behindern. Beispiele solcher Lösungsmittel sind halogenierte Kohlenwasserstoffe wie Methylenchlorid, Chloroform, Tetrachlorkohlenstoff und Perchlorethylen, Carbonsäuren wie Ameisensäure, Essigsäure und Propionsäure, aromatische Kohlenwasserstoffe wie Nitrobenzol, Chlorbenzol, Dichlorbenzol und Trichlorbenzol, Ester wie Ethylacetat, Nitrile wie Acetonitril und BenzonitriI, Amide wie Dimethylformamid und Dimethylacetamid, Sulfolan, Di-methylsulfon, Dimethylsulfoxid, 1,3-Dimethyl-2-imidazolidinon sowie Wasser. Diese inerten Lösungsmittel können jeweils für sich oder in Kombinationen eingesetzt werden.
Als Halogenierungsmittel können beispielsweise Chlor, Brom, lod, Chlorwasserstoffsäure-Wasserstoff-peroxid, Sulfurylchlorid, Phosphorpentachlorid, N,N-Dichlorharnstoff, N-Chlorsuccinimid, N-Bromsucci-nimid, t-Butylester unterhalogeniger Säuren, Trichlorisocyanursäure und Trichlormethansulfonylhaloge-nide verwendet werden. Die Menge an Halogenierungsmittel kann im Bereich von 0,1 Mol bis ein überschüssiges Mol pro Mol der Phenoxyfettsäuren gemäss Formel (II) gewählt werden.
Wenn notwendig können organische Basen, anorganische Basen oder organische Salze im erfindungsgemässen Verfahren eingesetzt werden. Beispiele organischer Basen sind Amine wie Triethylamin

und Pyridin, Beispiele anorganischer Basen sind Hydroxide von Alkalimetallen oder Erdalkalimetallen wie Natriumhydroxid, Kaliumhydroxid, Calciumhydroxid und Bariumhydroxid sowie Carbonate von Alkalimetallen oder Erdalkalimetallen wie Natriumcarbonat, Kaliumcarbonat und Calciumcarbonat, und Beispiele organischer Salze sind Alkali- oder Erdalkalisalze organischer Säuren wie Natriumacetat, Kalium-acetat, Calciumacetat und Natriumbenzoat sowie Ammoniumacetat. Deren Menge kann nach Belieben im Bereich von 0,1 Mol bis ein überschüssiges Mol pro Mol der Phenoxyfettsäuren gemäss Formel (II) gewählt werden.
Die Reaktionstemperatur liegt im allgemeinen im Gebiet von -20°C bis zum Siedepunkt der verwendeten inerten Lösungsmittel.
Die Reaktionsdauer hängt von der Reaktionstemperatur und der Grösse des Reaktionsansatzes ab, beträgt aber im allgemeinen zwischen einigen Minuten und 48 Stunden.
Nach Vervollständigung der Reaktion wird das Reaktionsgemisch mit den Zielprodukten nach den üblichen Arbeitsweisen wie Extraktion mit Lösungsmitteln isoliert und, wenn nötig, durch Säulenchromatographie, Umkristallisieren, Destillation oder ähnlichen gereinigt, und man erhält die Halogenphenoxyfett-säure-Derivate gemäss Formel (I) oder (I')-
Repräsentative Beispiele der Halogenphenoxyfettsäure-Derivate gemäss Formel (I'), welche neue Verbindungen darstellen, sind in Tabelle 1 gezeigt.

Die vorliegende Erfindung wird durch die folgenden, nicht einschränkenden Beispiele weiter erläutert.
1,51 g (0,01 Mol) 4-Fluorphenoxyacetonitril und 0,82 g (0,01 Mol) Natriumacetat wurden in einem Lösungsmittelgemisch aus 7 ml Essigsäure und 3 ml Wasser gelöst, und in die Lösung wurde Chlorgas mit einer Mengengeschwindigkeit von 12,3 ml/Min 60 Minuten lang bei 50°C unter Rühren eingeleitet.
Nach Beendigung der Reaktion wurden dem Reaktionsgemisch 3 ml einer 10%igen wässerigen Natri-umthioisulfatlösung zugegeben und das Lösungsmittel unter vermindertem Druck entfernt. Zum erhaltenen Rückstand wurden 20 ml einer 10%igen wässrigen Natriumbicarbonatlösung gegeben und das Zielprodukt mit Ethylacetat extrahiert (30 ml x 2).
Die organische Schicht wurde über wasserfreiem Natriumsulfat getrocknet und unter vermindertem Druck konzentriert. Nach Destillation des Konzentrates erhielt man 1,54 g 2-Chlor-4-fluorphenoxyaceto-nitril.
Diese Verbindung ist eine bekannte Verbindung und hatte den gleichen Schmelzpunkt und das gleiche NMR-Spektrum als diejenige gemäss Literatur.
Ausbeute: 83,0%
1,86 g (0,01 Mol) 4-Chlorphenoxyacetamid wurden in 11 ml Essigsäure unter Erwärmen auf 50°C gelöst, und in diese Lösung wurde mit einer Mengengeschwindigkeit von 12,3 ml/Min 50 Minuten lang bei der genannten Temperatur Chlorgas eingeleitet. Nach Beendigung der Reaktion wurden dem Reaktionsgemisch 3 ml einer 10%igen wässerigen Natriumthiosulfatlösung beigegeben und das Lösungsmittel unter vermindertem Druck abdestilliert. Der Rückstand wurde mit 20 ml einer 10%igen wässerigen Natriumbicarbonatlösung versetzt, und das Zielprodukt wurde mit Ethylacetat extrahiert (30 ml x 2).
Die organische Schicht wurde über wasserfreiem Natriumsulfat getrocknet und unter vermindertem Druck konzentriert. Die erhaltenen Kristalle wurden mit n-Hexan gewaschen, und man erhielt 2,02 g des Zielproduktes 2,4-Dichlorphenoxyacetamid.
Ausbeute: 92,0%
Diese Verbindung ist eine bekannte Verbindung und hat den gleichen Schmelzpunkt und das gleiche NMR-Spektrum als diejenige gemäss Literatur.

1,87 g (0,01 Mol) 4-Chlorphenoxyessigsäure wurden zu 10 ml Dimethylformamid gegeben, und in die Mischung wurde bei 50°C 45 Minuten lang Chlorgas mit einer Mengengeschwindigkeit von 12,3 ml/Min unter Rühren eingeleitet.
Nach Beendigung der Reaktion wurden zur Reaktionsmischung 3 ml einer 10%igen wässerigen Natri-umthiosulfatlösung gegeben und das Lösungsmittel unter vermindertem Druck abdestilliert. 20 ml einer 10%igen wässerigen Natriumbicarbonatlösung wurden zum Rückstand gegeben, und das Zielprodukt wurde mittels Ethylacetat extrahiert (30 ml x 2).
Die organische Schicht wurde über wasserfreiem Natriumsulfat getrocknet und unter vermindertem Druck eingedampft. Die erhaltenen Kristalle wurden mit n-Hexan gewaschen, und man erhielt 2,07 g des Zielproduktes 2,4-Dichiorphenoxyessigsäure.
Ausbeute: 93,6%
Diese Verbindung ist eine bekannte Verbindung und besass den gleichen Schmelzpunkt und das gleiche NMR-Spektrum als diejenige gemäss Literatur.
Tabelle 3 zeigt typische Beispiele der Halogenphenoxyfettsäure-Derivate gemäss Formel I, die auf die gleiche Weise wie in den Beispielen 1 bis 3 hergestellt wurden.
Da die Verbindungen gemäss Tabelle 3 bekannte Verbindungen sind, wurden sie durch die Identität ihrer Schmelzpunkte und der NMR- und IR-Spektren im Vergleich zur Literatur identifiziert.

Die durch Formel (I) dargestellten Halogenphenoxyfettsäure-Derivate sind insbesondere als Zwischenprodukte zur Herstellung von Herbiziden wichtig, die in der japanischen Offenlegungsschrift (Kokai) Nr. 3-163063) offenbart sind. Die Endprodukte, nämlich die typischen Herbizide, können beispielsweise nach dem untenstehenden Verfahren hergestellt werden.

1. Verfahren zur Herstellung von Haiogenphenoxyfettsäure-Derivaten der Formel (I):

R - eine Cyanogruppe;
- eine Gruppe -CON(R3)R4, worin R3 und R4, die gleich oder unterschiedlich sind, jeweils für ein Wasserstoffatom, eine niedere Alkylgruppe, eine Phenylgruppe, eine Phenylgruppe mit 1 bis 5 gleichen oder unterschiedlichen Substituenten aus der Gruppe Halogenatom, Nitrogruppe, Cyanogruppe, niedere Alkylgruppe, niedere Halogenalkylgruppe, niedere Alkoxygruppe, niedere Halogenalkoxygruppe, niedere Alkylthiogruppe und niedere Halogenalkylthiogruppe, eine Benzylgruppe oder eine Benzylgruppe mit 1 bis 5 gleichen oder unterschiedlichen Substituenten aus der Gruppe niedere Alkylgruppe, niedere Halogenalkylgruppe, niedere Alkoxygruppe, niedere Halogenalkoxygruppe, niedere Alkylthiogruppe und niedere Halogenalkylthiogruppe stehen, oder R3 und R4 bedeuten zusammen eine Alkylengruppe, oder - eine Gruppe -COAR5, worin R5 für ein Wasserstoffatom, eine niedere Alkylgruppe, eine Phenylgruppe, eine Phenylgruppe mit 1 bis 5 gleichen oder unterschiedlichen Substituenten aus der Gruppe Halogenatom, Nitrogruppe, Cyanogruppe, niedere Alkylgruppe, niedere Halogenalkylgruppe, niedere Alkoxygruppe, niedere Halogenalkoxygruppe, niedere Alkylthiogruppe und niedere Halogenalkylthiogruppe, eine Benzylgruppe oder eine Benzylgruppe mit 1 bis 5 gleichen oder unterschiedlichen Substituenten aus der Gruppe niedere Alkylgruppe, niedere Halogenalkylgruppe, niedere Alkoxygruppe, niedere Halogenalkoxygruppe, niedere Alkylthiogruppe und niedere Halogenalkylthiogruppe steht und A ein Sauerstoff- oder Schwefelatom darstellt,
R1 und R2 jeweils ein Wasserstoffatom oder eine niedere Alkylgruppe, wobei R1 und R2 gleich oder unterschiedlich sind,
X ein Halogenatom oder eine niedere Alkylgruppe und
Y ein Halogenatom, durch selektive Halogenierung einer Phenoxyfettsäure der folgenden Formel (II) in Gegenwart eines Halogenierungsmittels:
Rl worin R, R1, R1 und X die bereits genannten Bedeutungen haben.
2. Verfahren nach Anspruch 1, worin das Halogenierungsmittel ein Chlorierungsmittel ist.
3. Halogenphenoxyfettsäure-Derivat gemäss Formel (!'):
worin bedeuten:
R' eine Cyanogruppe oder -CON(R3') R4', in der R3'und R4', die gleich oder unterschiedlich sind, jeweils für ein Wasserstoffatom, eine niedere Alkylgruppe, eine Phenylgruppe, eine Phenylgruppe mit 1 bis 5 gleichen oder unterschiedlichen Substituenten aus der Gruppe Halogenatom, Nitrogruppe, Cyanogruppe, niedere Alkylgruppe, niedere Halogenalkylgruppe, niedere Alkoxygruppe, niedere Halogenalkoxygruppe, niedere Alkylthiogruppe und niedere Halogenalkylthiogruppe, eine Benzylgruppe oder eine Benzylgruppe mit 1 bis 5 gleichen oder unterschiedlichen Substituenten aus der Gruppe niedere Alkylgruppe, niedere Halogenalkylgruppe, niedere Alkoxygruppe, niedere Halogenalkoxygruppe, niedere Alkylthiogruppe und niedere Halogenalkylthiogruppe stehen,
R1' und R2' jeweils ein Wasserstoffatom oder eine niedere Alkylgruppe, wobei R1' und R2' gleich oder unterschiedlich sind,
X' ein Fluoratom, ein iodatom, ein Bromatom oder eine niedere Alkylgruppe, und Y' ein Halogenatom,
wobei R' keine Cyanogruppe ist, wenn R1' und R2' ein Wasserstoffatom darstellen, X' ein Fluoratom und Y' ein Chloratom bedeutet.
