背景技术
裂环环烯醚萜甙类化合物是一类具有抗菌、消炎、抗病毒、抗氧化、抗癌、降血糖等作用的物质,且毒性很低,可用于医疗和保健功能方面。但是,这类化合物在天然植物中含量很低,且化学结构复杂,利用化学方法进行全合成非常困难,且不经济,严重地限制了对其进一步的开发和利用。
发明内容
本发明的目的在于提供一种适于工业化生产的合成裂环环烯醚萜甙类化合物的生物转化合成方法即组织(细胞)培养方法。
为了达到上述目的,本发明采用如下技术方案:一种合成裂环环烯醚萜甙类化合物的方法,包括培养基的制备、外植体的诱导、组织(细胞)培养和目标分子的提取四个步骤:在外植体诱导步骤,取木犀科丁香属、女贞属、木犀属、木犀榄属、茉莉属、雪柳属和梣属的植物叶片、叶芽或根茎消毒后接种于固体培养基上,于15-32℃培养15-108天;在目标分子提取步骤,将收获的愈伤组织置于提取器中用有机溶剂回流提取2-6小时或冷浸8-48小时,除去有机溶剂得目标分子裂环环烯醚萜甙类化合物。

固体培养基中还含有吡多醇0.125-1.25mg/l、烟酸0.025-0.25mg/l。
有机溶剂包括醇类、酮类和醚类有机溶剂。
有机溶剂为醇类中的甲醇或者乙醇,用量为愈伤组织重量的10-20倍。
一种合成裂环环烯醚萜甙类化合物的方法,包括固体培养基和液体培养基的制备、外植体的诱导、组织(细胞)培养和目标产物的提取四个步骤:在外植体诱导步骤,取木犀科丁香属、女贞属、木犀榄属、茉莉属和梣属的植物叶片、叶芽或根茎消毒后接种于固体培养基上,于15-32℃培养14-49天;再取出愈伤组织转移到液体培养基中于震荡培养箱或摇床或生物反应器或反应釜中在18-32℃培养10-60天;在目标产物提取步骤,将培养物和培养基先过滤分开再分别置于提取器中用有机溶剂提取,除去有机溶剂得目标产物裂环环烯醚萜甙类化合物。
液体培养基中含有硫酸镁160-689mg/l,磷酸二氢钾或磷酸二氢钠或二者之和70-350mg/l,硝酸钾860-4200mg/l,硝酸铵725-4600mg/l,氯化钙180-980mg/l,硫胺0.05-1.25mg/l,肌醇25-250mg/l,硼酸1.6-26.8mg/l,硫酸锰3.9-62.5mg/l,硫酸锌2.1-32.2mg/l,钼酸钠0.0125-0.0725mg/l,硫酸铜0.0125-0.0725mg/l,氯化钴0.0125-0.0725mg/l,碘化钾0.0215-2.65mg/l,硫酸亚铁6.5-65.5mg/l,EDTA钠盐9.8-76.4mg/l或EDTA铁盐21.5-86mg/l,2,4-D、N6-呋喃甲基腺嘌呤、6-苄基腺嘌呤、6-糠氨基腺嘌呤之一或二种以上的和0.05-25mg/l,萘乙酸、萘甲酸、吲哚-3-乙酸、4-氨基-3,5,6-三氯吡啶甲酸之一或二种以上的和0.1-50mg/l,蔗糖12000-60000mg/l,逐一称取除蔗糖外的培养基成分,用水溶解、混合并加水稀释到所需浓度,然后称取配方量蔗糖,加入上述溶液中溶化,调PH=4.5-7.0,经灭菌后得液体培养基;固体培养基比液体培养基配方中增加琼脂成分,含量为4500-19500mg/l,其制法为:称取除蔗糖、琼脂外的培养基成分,用水溶解、混合并加水稀释到所需浓度,然后称取配方量蔗糖和琼脂,加入上述溶液中溶化,调PH=4.5-7.0,经灭菌后得固体培养基。
固体培养基和液体培养基中各含有吡多醇0.125-1.25mg/l、烟酸0.025-0.25mg/l。
有机溶剂包括醇类、酮类和醚类有机溶剂。
有机溶剂为醇类中的甲醇或者乙醇,用量为愈伤组织的10-20倍。
本发明采用生物学和细胞工程中的组织(细胞)培养方法合成裂环环烯醚萜甙类化合物,不受自然资源限制,并且有利于保护生态环境,方法中培养基为组织(细胞)培养中广泛采用的植物细胞培养基,成分易得,培养基中含有氮、磷、钾等大量元素,硼、锌、锰、钼、铜、钴、钠、碘、铁等微量元素,硫胺、吡多醇、烟酸、肌醇等维生素和2,4-D、N6-呋喃甲基腺嘌呤、6-苄基腺嘌呤、6-糠氨基腺嘌呤、萘乙酸、萘甲酸、吲哚-3-乙酸、4-氨基-3,5,6-三氯吡啶甲酸等植物生长调节物质,能够提供组织(细胞)培养所需的各种营养成分,培养条件温和,提取方法简便易行。整个方法易于工业化生产。本发明两种方法中,第一种方法仅采用固体培养基进行培养;第二种方法先采用固体培养基培养然后转入液体培养基进行培养,更易于工业化大规模生产,且缩短培养时间,产物收率大大提高,生产成本得以大大降低。

具体实施方式
实施例1,一种合成裂环环烯醚萜甙类化合物的方法,取硫酸镁370mg,磷酸二氢钾170mg,硝酸钾1900mg,硝酸铵1650mg,氯化钙440mg,硫胺1.0mg,肌醇100mg,硼酸6.2mg,硫酸锰15.6mg,硫酸锌8.6mg,钼酸钠0.025mg,硫酸铜0.025mg,氯化钴0.025mg,碘化钾0.085mg,硫酸亚铁27.8mg,EDTA钠盐37.3mg,2,4-D1.0mg,6-糠氨基腺嘌呤2.0mg,萘乙酸3.0mg,蔗糖30000mg,琼脂12000mg,逐一称取除蔗糖、琼脂外的培养基成分,用去离子水溶解、混合,然后称取配方量蔗糖和琼脂,加入上述溶液中溶化并加去离子水稀释到1000ml,调PH=5.8,经121℃灭菌25分钟后得固体培养基。
将小叶丁香叶片或幼芽或嫩茎消毒后接种于灭菌后的固体培养基上,于25℃培养箱内诱导产生愈伤组织并且培养49天,取出产物,置于索氏提取器中每次加10倍重量的无水乙醇提取2次,然后蒸出乙醇,得目标分子。其代表物橄榄苦甙结构式见图1,其NMR数据列于表1中。
实施例2,本实施例中将消毒后的女贞叶片置于灭菌后的固体培养基上,于28℃培养箱内诱导并培养49天,取出产物,置于索氏提取器中每次加15倍重量的无水乙醇提取2次,然后蒸出乙醇,得目标分子。其它同实施例1。
实施例3,一种合成裂环环烯醚萜甙类化合物的方法,本实施例中,固体培养基及制法同实施例1。液体培养基配方为硫酸镁370mg,磷酸二氢钾170mg,硝酸钾1900mg,硝酸铵1650mg,氯化钙440mg,硫胺1.0mg,肌醇100mg,硼酸6.2mg,硫酸锰15.6mg,硫酸锌8.6mg,钼酸钠0.025mg,硫酸铜0.025mg,氯化钴0.025mg,碘化钾0.085mg,硫酸亚铁27.8mg,EDTA钠盐37.3mg,2,4-D 1.0mg,6-糠氨基腺嘌呤2.0mg,萘乙酸3.0mg,蔗糖30000mg,逐一称取上述培养基成分,用去离子水溶解、混合,并加去离子水稀释到1000ml,调PH=5.6,经121℃灭菌分钟后得液体培养基。
取小叶丁香叶片消毒后置入固体培养基上,28℃培养42天,将得到的愈伤组织转入液体培养基中,于振荡培养箱内25℃培养21天,过滤分开培养物和培养液。培养物用甲醇提取2次,每次加10倍重量的甲醇;培养液用甲醇提取2次,每次加3倍重量的甲醇。合并提取物即目标产物。其代表物橄榄苦甙结构式见图1,其NMR数据列于表1中。
实施例4,本实施例中,将固体培养基培养所得愈伤组织转入盛有液体培养基的50升反应釜中,25℃培养18天,过滤分开培养物和培养液,分别用甲醇提取。提取物即目标产物。其他同实施例3。

Claims (10)
1.一种合成裂环环烯醚萜甙类化合物的方法,其特征在于,包括培养基的制备、外植体的诱导、组织培养和目标分子的提取四个步骤;在外植体诱导步骤,取木犀科丁香属、女贞属的植物叶片、叶芽或根茎消毒后接种于固体培养基上,于15-32℃培养15-108天;在目标分子提取步骤,将收获的愈伤组织置于提取器中用有机溶剂回流提取2-6小时或冷浸8--48小时,除去有机溶剂得目标分子裂环环烯醚萜甙类化合物。
2.如权利要求1所述的方法,其特征在于,固体培养基中含有硫酸镁160-689mg/l,磷酸二氢钾或磷酸二氢钠或二者之和70-350mg/l,硝酸钾860-4200mg/l,硝酸铵725-4600mg/l,氯化钙180-980mg/l,硫胺0.05-1.25mg/l,肌醇25-250mg/l,硼酸1.6-26.8mg/l,硫酸锰3.9-62.5mg/l,硫酸锌2.1-32.2mg/l,钼酸钠0.0125-0.0725mg/l,硫酸铜0.0125-0.0725mg/l,氯化钴0.0125-0.0725mg/l,碘化钾0.0215-2.65mg/l,硫酸亚铁6.5-65.5mg/l,EDTA钠盐9.8-76.4mg/l或EDTA铁盐21.5-86mg/l,2,4-D、N6-呋喃甲基腺嘌呤、6-苄基腺嘌呤、6-糠氨基腺嘌呤之一或二种以上的和0.05-25mg/l,萘乙酸、萘甲酸、吲哚-3-乙酸、4-氨基-3,5,6-三氯吡啶甲酸之一或二种以上的和0.1-50mg/l,蔗糖12000-60000mg/l,琼脂4500-32000mg/l,逐一称取除蔗糖、琼脂外的培养基成分,用水溶解、混合并加水稀释到所需浓度,然后称取配方量蔗糖和琼脂,加入上述溶液中溶化,调PH=4.5-7.0,经灭菌后得固体培养基。
3.如权利要求2所述的方法,其特征在于,固体培养基中还含有吡多醇0.125-1.25mg/l、烟酸0.025-0.25mg/l。
4.如权利要求1、2或3所述的方法,其特征在于,有机溶剂包括醇类、酮类和醚类有机溶剂。
5.如权利要求4所述的方法,其特征在于,有机溶剂为醇类中的甲醇或者乙醇,用量为愈伤组织重量的10-20倍。
6.一种合成裂环环烯醚萜甙类化合物的方法,其特征在于,包括固体培养基和液体培养基的制备、外植体的诱导、组织培养和目标产物的提取四个步骤;在外植体诱导步骤,取木犀科丁香属、女贞属的植物叶片、叶芽或根茎消毒后接种于固体培养基上,于15-32℃培养14-49天;再取出愈伤组织转移到液体培养基中于震荡培养箱或摇床或生物反应器或反应釜中在18-32℃培养10-60天;在目标产物提取步骤,将培养物和培养基先过滤分开再分别置于提取器中用有机溶剂提取,除去有机溶剂得目标产物裂环环烯醚萜甙类化合物。
7.如权利要求6所述的方法,其特征在于,液体培养基中含有硫酸镁160-689mg/l,磷酸二氢钾或磷酸二氢钠或二者之和70-350mg/l,硝酸钾860-4200mg/l,硝酸铵725-4600mg/l,氯化钙180-980mg/l,硫胺0.05-1.25mg/l,肌醇25-250mg/l,硼酸1.6-26.8mg/l,硫酸锰3.9-62.5mg/l,硫酸锌2.1-32.2mg/l,钼酸钠0.0125-0.0725mg/l,硫酸铜0.0125-0.0725mg/l,氯化钴0.0125-0.0725mg/l,碘化钾0.0215-2.65mg/l,硫酸亚铁6.5-65.5mg/l,EDTA钠盐9.8-76.4mg/l或EDTA铁盐21.5-86mg/l,2,4-D、N6-呋喃甲基腺嘌呤、6-苄基腺嘌呤、6-糠氨基腺嘌呤之一或二种以上的和0.05-25mg/l,萘乙酸、萘甲酸、吲哚-3-乙酸、4-氨基-3,5,6-三氯吡啶甲酸之一或二种以上的和0.1-50mg/l,蔗糖12000-60000mg/l,逐一称取除蔗糖外的培养基成分,用水溶解、混合并加水稀释到所需浓度,然后称取配方量蔗糖,加入上述溶液中溶化,调PH=4.5-7.0,经灭菌后得液体培养基;固体培养基比液体培养基配方中增加琼脂成分,含量为4500-19500mg/l,其制法为:称取除蔗糖、琼脂外的培养基成分,用水溶解、混合并加水稀释到所需浓度,然后称取配方量蔗糖和琼脂,加入上述溶液中溶化,调PH=4.5-7.0,经灭菌后得固体培养基。
8.如权利要求7所述的方法,其特征在于,固体培养基和液体培养基中各含有吡多醇0.125-1.25mg/l、烟酸0.025-0.25mg/l。
9.如权利要求6、7或8所述的方法,其特征在于,有机溶剂包括醇类、酮类和醚类有机溶剂。
10.如权利要求9所述的方法,其特征在于,有机溶剂为醇类中的甲醇或者乙醇,用量为愈伤组织的10-20倍。
