植物病虫害往往使农业生产蒙受严重损失,农作物产量损失的三分之一可归因于病害。培育和栽培抗病品种,使用农药,高水平的生产管理技术等是防治作物病害的重要措施。但通过常规育种手段获得抗病害作物品种选育历程较长,存在抗性丧失问题,缺乏抗病害的亲本资源;大量使用农药会产生农药残留、病原物产生抗药性等问题。植物基因工程的发展为培育抗病害的作物品种提供了新的手段。
植物的抗病方式有固有抗性(被动抗性)和病原诱导抗性(主动抗性)。植物的固有抗性又分固有的结构抗性和固有的化学抗性。固有的结构抗性如植物的蜡质角质层、周皮、表皮毛和根毛、表皮细胞、自然孔口、伤口等;固有的化学抗性如不饱和内酯、含硫化合物、酚类化合物如皂苷(saponin)等植物次生代谢产物。植物的主动抗性也分诱导的结构抗性和诱导的化学抗性。诱导的结构抗性如乳突的形成、维管束内胼胝质的形成、晕斑形成、细胞壁的木质化、细胞壁富含羟脯氨酸糖蛋白积累及细胞壁的交联强化等。诱导的化学抗性如酚类物质含量的增加、植保素的产生和积累、病程相关蛋白(pathogenesis-related protein)如几丁质酶、葡聚糖酶等的产生、糖苷氧化产生有毒物质如HCN、蛋白酶抑制剂的生成、各种活性氧(O2-,H2O2)以及发生过敏性细胞死亡反应等(Dixon RA,Harrison MJ,Lamb CJ.1994.Earlyevents in the activation of plant defense response.Annu.Rev Phytopathol.32:479~501)。Flor(Flor.1971.Current status of the gene for gene concept.Annu.Rev.Phytopathol.9:275-296.)根据他对亚麻和亚麻锈病菌(Melampsoralini)间小种特异性抗性的研究,提出了基因对基因(gene-for-gene)假说。该假说认为寄主分别含有感病基因(r)和抗病基因(R)、病原物分别含有毒性基因(vir)和无毒基因(avr),只有当具有相应抗病基因的植物与具有相应无毒基因的病原相遇时,才会激发植物的抗病反应,植物表现为抗病,否则,寄主植物表现为感病。现在多个植物抗病基因和病原物无毒基因的克隆证实了该假说的正确性。
过敏性反应(Hypersensitive Response,HR)是植物最常见的抗病表现形式,它几乎总被看作为抗病的标志,但并不是所有的抗病反应均表现HR(KeenNT.1992.The molecular biology of disease resistance.Plant MolBiol.19:109-114)。在病原菌侵入点及周围组织的细胞突然死亡,限制了病原菌在体内的定殖和扩展。这种局部的过敏性反应有些情况也可激发整个植株的非专化性的抗病性即系统获得抗性(systemic acquired resistance,以下简称SAR)的表达,此时整个植株对广泛的病原物具有抗性。SAR是植物受生物或化学因子在某部位作用后整株表现出的对较广泛的病原微生物(病毒、细菌、真菌)的抗性,且这种抗性往往能持续较长时间。SAR是植物防卫反应基因特别是PR蛋白类基因被系统诱导表达的结果。
水稻(Oryza sativa L.)是我国重要的粮食作物。病害是水稻生产的一个重要制约因素,每年需投入大量的人力、物力和财力用于水稻病害的防治。稻瘟病和白叶枯病是水稻二大主要病害。水稻桂99为广西主栽杂交稻常用恢复系,它为籼稻,对稻瘟病、白叶枯病均表现为中抗。
发明创造内容
本发明的一个目的是提供一种植物抗病相关蛋白及其编码基因。
本发明所提供的植物抗病相关蛋白,名称为SDKB,来源于水稻籼稻品种桂99,具有序列表中SEQ ID №:4的氨基酸残基序列或具有将SEQ ID №:4的氨基酸残基序列经过一个或几个氨基酸残基的取代、缺失或添加且与SEQ ID №:4限定的蛋白质具有相同活性的由SEQ ID №:4衍生的氨基酸残基序列。
序列表中的SEQ ID №:4由582个氨基酸残基组成,自N端的第55-194位氨基酸残基为BTB功能域(Broad-complex,Tramtrack and Bric a brac),自N端的第269-297、298-328和332-361位氨基酸残基为三个连续的锚蛋白重复序列(ankyrin repeats)功能域,该两个功能域可能都与蛋白质相互作用有关。
上述植物抗病相关蛋白的编码基因,它的cDNA序列具有下述核苷酸序列之一:
1)序列表中序列3的多核苷酸;
2)编码序列表中SEQ ID №:4蛋白质序列的多核苷酸;
3)与序列表中序列3限定的核苷酸序列具有90%以上同源性,且编码相同功能蛋白质的核苷酸序列;
4)在高严谨条件下可与序列表中的SEQ ID №:3限定的DNA序列杂交的核苷酸序列。
所述高严谨条件为杂交后用含0.1×SSPE(或0.1×SSC)、0.1%SDS的溶液在65℃下洗膜。
序列表中的序列3由1826位碱基组成,该cDNA序列的编码序列为序列表中序列3的自5′端第16-1764位碱基。
与所述cDNA序列对应的所述植物抗病相关蛋白的基因组基因,具有下述核苷酸序列之一:
1)序列表中序列5的多核苷酸;
2)编码序列表中SEQ ID №:4蛋白质序列的DNA;
3)与序列表中序列5限定的核苷酸序列具有90%以上同源性,且编码相同功能蛋白质的核苷酸序列;
4)在高严谨条件下可与序列表中的SEQ ID №:5限定的DNA序列杂交的核苷酸序列。
所述高严谨条件为杂交后用含0.1×SSPE(或0.1×SSC)、0.1%SDS的溶液在65℃下洗膜。
序列表中的序列5由4241位碱基组成,自5′端的第1-561位碱基为该基因组基因的第一个外显子,自5′端的第2284-3031位碱基为该基因组基因的第二个外显子,自5′端的第3240-3440位碱基为该基因组基因的第三个外显子,自5′端的第4003-4241位碱基为该基因组基因的第四个外显子,自5′端的第1-3位碱基为该基因组基因的起始密码子ATG,自5′端的第4239-4241位碱基为该基因组基因的终止密码子TGA;自5′端的第562-2283位碱基为该基因组基因的第一个内含子,自5′端的第3032-3239位碱基为该基因组基因的第二个内含子,自5′端的第3441-4002位碱基为该基因组基因的第三个内含子。
所述植物抗病相关蛋白的基因组基因的5′端和3′端连接有非编码区,为具有序列表中的序列1的核苷酸序列的DNA。
序列表中的序列1由7417个碱基组成,自5′端的第2365-2925位碱基为植物抗病相关蛋白的基因组基因的第一个外显子,自5′端的第4648-5395位碱基为该基因组基因的第二个外显子,自5′端的第5604-5804位核苷酸为该基因组基因的第三个外显子,自5′端的第6367-6605位核苷酸为该基因组基因的第四个外显子,自5′端的第2365-2367位核苷酸为该基因组基因的起始密码子ATG,自5′端的第6603-6605位核苷酸为该基因组基因的终止密码子TGA;自5′端的第2926-4647位碱基为该基因组基因的第一个内含子,自5′端的第5396-5603位碱基为该基因组基因的第二个内含子,自5′端的第5805-6366位碱基为该基因组基因的第三个内含子。
含有本发明的植物抗病相关蛋白编码基因的载体、细胞系及宿主菌,如质粒pGXN7410,均属于本发明的保护范围。
本发明的第二个目的是提供一种提高水稻抗病性的方法。
本发明所提供的提高水稻抗病性的方法,是将上述植物抗病相关蛋白编码基因导入水稻中。
所述植物抗病相关蛋白编码基因可按照植物基因工程领域中的常规方法,如农杆菌介导转化法、基因枪介导转化法或花粉管通道法导入水稻中。
实验证明转正义的SDKB的基因组基因的水稻TP309对水稻白叶枯病和稻瘟病的抗性显著增强,转反义的SDKB的基因组基因的水稻桂99植株对水稻白叶枯病的抗性显著降低,说明该基因与水稻广谱抗病有关。本发明的植物抗病相关蛋白编码基因可广泛用于增强水稻等植物的抗病性。

具体实施方式
在本发明的实施例中所用的材料包括:大肠杆菌(Escherichia coli)株系JM109,购自Invitrogen公司的3’RACE试剂盒(3’RACE System for RapidAmplification of cDNA Ends,目录号18373-027)和Clontech公司的5’RACE试剂盒(SMARTTM RACE eDNA Amplification kit,目录号K1811-1),购自Promega、Stratagene、TaKaRa、Sigma、Qiagen的限制性内切酶、修饰酶等试剂。
实施例1、植物抗病相关蛋白SDKB的基因组基因的克隆
设计合成一对引物(引物序列RiF2:5’CTTGAAAACCGAGTTGCT 3’;RiR2:5’TTTCCCGAGCTCCACTGT 3’),用该对引物,按照常规方法对经苯(1,2,3)噻重氮-7-硫代羟酸S-甲酯(benzo(1,2,3)thiadiazole-7-carbothioc acid S-methyl ester,简称BTH)处理(处理方法为:向生长了3周的盆栽桂99的浇灌水中加入BTH至终浓度约0.3mM,同时用0.3mM的BTH喷雾处理水稻桂99的叶片,处理后48小时采集水稻叶片)的水稻桂99的总RNA进行RT-PCR,结果得到一个约200bp的PCR产物(图1),将该PCR产物克隆到经SmaI酶切的pGEM-3Zf(+)(购自Promega公司)中,双向双链测定了两个转化子的插入片段的DNA序列,结果表明该两个克隆的外源DNA的序列相同,大小均为228bp,和GenBank索引号AP002537、AP002746、NM_189701、NM_188219、AK120715、AY323485序列中的对应序列100%相同。图1中,1为100bp ladder(片段大小从大到小依次为:3kb,2kb,1.5kb,1.2kb,1kb,900bp,800bp,700bp,600bp,500bp,400bp,300bp,200bp,100bp);2为PCR产物。
用该PCR产物作探针和分别经EcoRI、BamHI或BamHI/EcoRI酶切的桂99总DNA杂交,结果如图2所示,表明分别得到一个约25kb、11kb和7.4kb的杂交带。图2中,1为λ/EcoRI(片段大小从大到小依次为:21.2kb,7.4kb,5.8kb,5.6kb,4.9kb,3.5kb);2-4为桂99总DNA分别经EcoRI、BamHI、EcoRI/BamHI酶切的电泳图;5-7为桂99总DNA分别经EcoRI、BamHI、EcoRI/BamHI酶切及电泳后与探针的杂交带型。用BamHI/EcoRI双重酶切桂99总DNA,回收7.4kb左右的DNA片段,用经BamHI/EcoRI酶切的pGEM-3Zf(+)来克隆这些片段构建得到部分基因文库,用菌落原位杂交筛选约12000个转化子,结果如图3所示,得到1个阳性克隆pGXN7410。双向双链测定了该阳性克隆的DNA序列,结果表明该BamHI/EcoRI片段大小为7417bp(序列表中的序列1,图4),上面含有一个完整的基因,即本发明的植物抗病相关蛋白SDKB的基因组基因,大小为4241bp(序列表中的序列5),推测其含有四个外显子、三个内含子,自5′端的第1-561位碱基为该基因组基因的第一个外显子,自5′端的第2284-3031位碱基为该基因组基因的第二个外显子,自5′端的第3240-3440位碱基为该基因组基因的第三个外显子,自5′端的第4003-4241位碱基为该基因组基因的第四个外显子,自5′端的第1-3位碱基为该基因组基因的起始密码子ATG,自5′端的第4239-4241位碱基为该基因组基因的终止密码子TGA;自5′端的第562-2283位碱基为该基因组基因的第一个内含子,自5′端的第3032-3239位碱基为该基因组基因的第二个内含子,自5′端的第3441-4002位碱基为该基因组基因的第三个内含子。
所克隆得到的7417bp BamHI/EcoRI片段的自5’端的第2365-2925位碱基为SDKB的基因组基因的第一个外显子,自5’端的第4648-5395位碱基为SDKB的基因组基因的第二个外显子,自5’端的第5604-5804位碱基为SDKB的基因组基因的第三个外显子,自5’端的第6367-6605位碱基为SDKB的基因组基因的第四个外显子,自5’端的第2365-2367位碱基为SDKB的基因组基因的起始密码子ATG,自5’端的第6603-6605位碱基为SDKB的基因组基因的终止密码子TGA。
上述基因组基因编码一个含582个氨基酸的蛋白质(序列表中的序列4),推测的该基因的mRNA和拟南芥NPR1基因的mRNA同源性为57.2%,编码的蛋白质产物和拟南芥NPR1蛋白质的相似性为66%,其中48%氨基酸是相同的。图4中,1为1kb ladder(片段大小从大到小依次为:10.0kb,8.0kb,6.0kb,5.0kb,4.0kb,3.5kb,3.0kb,2.5kb,2.0kb,1.5kb,1kb);2为pGXN7410/EcoRI+BamHI。
实施例2、编码植物抗病相关蛋白SDKB的全长cDNA的克隆和测序
根据实施例1中得到的植物抗病相关蛋白SDKB的基因组基因的第2个外显子序列设计两个嵌套引物(引物RACE3-1:5’GAGCCCTTGACTCTGACG 3’;RACE3-2:5’GATGCTGCTCACTGAAGG 3’),应用3’RACE的方法(3’RACE System for RapidAmplification of cDNA Ends),利用购自Invitrogen公司的3’RACE试剂盒(3’RACE System for Rapid Amplification of cDNA Ends,目录号18373-027)克隆得到了引物RACE3-2下游的cDNA序列并连接在pBluescript M13 KS(+)(其物理图谱见《分子克隆实验指南》(第二版)(J.萨母布鲁克,E.F.弗里奇,T.曼尼阿蒂斯著,金冬雁,黎孟枫等译,1992年,科学出版社,北京)第16页)的EcoRV位点,用HindIII/EcoRI酶切获得的重组质粒,结果如图5所示,重组质粒给出和预期大小(约1.2kb)一致的外源片段,表明重组质粒结构正确。图5中,1为1kb ladder(片段大小从大到小依次为:10.0kb,8.0kb,6.0kb,5.0kb,4.0kb,3.5kb,3.0kb,2.5kb,2.0kb,1.5kb,1kb);2为用HindIII/EcoRI酶切的重组质粒。对该克隆的cDNA片段的序列测定分析表明该片段长度为1182bp(序列表序列2),3’端有17个碱基的poly(A),其余1165个碱基组成的核苷酸序列和SDKB基因的对应序列100%一致,含有外显子2下游(序列表中的序列1自5′端第4947-5395位碱基)449个碱基组成的核苷酸序列,全长的外显子3和外显子4以及276个碱基的3’端非编码序列。
根据3’RACE产物中的终止密码子下游的序列设计了2个嵌套引物(引物序列RiFuR1:5’GGGGGGCAATTGACAGAACGCAACCATCAGGTG 3’;3’END:5’GGTGAGTAGCTCCAGGAGGACAG TTAGCTATCAA 3’),应用SMART(Switch Mechanism Atthe 5’end of RNA Transcript)5’RACE的方法,利用Clontech公司的5’RACE试剂盒(SMARTTMRACE cDNA Amplification kit,目录号K1811-1),用下游引物RiFuR1特异性地合成该基因的全长cDNA,再用试剂盒提供的上游嵌套引物与3’END引物进行PCR扩增,把扩增产物克隆到pUC19的SmaI位点,用BamHI/EcoRI酶切得到的重组质粒,结果如图6所示,重组质粒给出和预期大小(约1.8kb)一致的外源片段,表明重组质粒结构正确,得到该基因的全长cDNA。图6中,1为1kb ladder(片段大小从大到小依次为:10.0kb,8.0kb,6.0kb,5.0kb,4.0kb,3.5kb,3.0kb,2.5kb,2.0kb,1.5kb,1kb);2为用BamHI/EcoRI酶切的重组质粒。挑取3个克隆进行测序并将得到的序列和克隆的7417bp EcoRI/BamHI序列比较,结果表明有1个克隆在PCR过程中没有出错,编码SDKB的全长cDNA序列(序列表中的序列3)和编码SDKB的基因组基因的对应序列100%一致,结果证明该基因是在水稻桂99中真实表达的基因,且预测的四个外显子、三个内含子的位置都是正确的。序列表中的序列3总长度为1826bp,含有一个长度为1749个碱基的编码序列,该编码序列为序列3的自5′端第16-1764位碱基,第1-15位碱基属于该基因的5’非编码区,第1765-1826位碱基属于该基因的3’非编码区。
实施例3、SDKB基因正义及反义水稻表达载体的构建
水稻表达载体的构建
以玉米的总DNA为模板,PCR扩增玉米Ubiquitin(泛素)启动子,所用引物为UbiF(5’ACTAAGCTTCTGCAGTGCAGCGTGACCCG 3’,在其5’端含HindIII酶切位点)和UbiR(5’GACGGATCCCTGCAGAAGTAACACCAAACAA 3’,在其5’端含BamHI酶切位点)。将玉米Ubiquitin启动子克隆到植物表达载体pCambia-1301(其详细情况及物理图谱请见http://www.cambia.org.au/main/r et 1300 1301.htm)的多克隆位点HindIII和BamHI上。再以pCambia-1301为模板,PCR扩增NOS(胭脂碱合成酶基因的3’-端转录终止序列)终止子,所用引物为nosF(5’AGCGGTACCCGTTCAAACATTTGGCAATAAAGTT 3’,在其5’端含KpnI酶切位点)和nosR(5’GTCGAGCTCCCCGATCTAGTAACATAGATGACACC 3’,在其5’端含SacI酶切位点),将NOS终止子克隆到已连上玉米Ubiquitin启动子的pCambia-1301的KpnI和SacI上,得到水稻表达载体pCambia1301-UbiN,如图7所示,pCambia1301-UbiN载体的T-DNA结构包括:T Border(L)(T-DNA左边界);T Border(R)(T-DNA的右边界);Ubiquitin启动子;NOS终止子;HYG(R)(潮霉素B磷酸转移酶(hygromycin Bphosphotransferase)基因);GUS(葡萄糖醛酸糖苷酶基因,GUS基因内有一内含子序列,使GUS基因只在转基因植物细胞中表达)。
1、SDKB的基因组基因正义水稻表达载体的构建
用XbaI/XhoI酶切pGXN7410,电泳后回收4842bp XbaI/XhoI片段(序列表中的序列1自5′端第1956-6797位碱基),把回收得到的含全长SDKB基因组基因的XbaI/XhoI片段亚克隆到pBluescript M13 KS(+)(其物理图谱见《分子克隆实验指南》(第二版)(J.萨母布鲁克,E.F.弗里奇,T.曼尼阿蒂斯著,金冬雁,黎孟枫等译,1992年,科学出版社,北京)第16页)的XbaI和XhoI位点上,得到重组质粒pGXN4834,再PCR扩增SDKB的基因组基因起始密码子ATG到第一个外显子的SmaI位点188bp的序列(模板为pGXN7410,引物为NPR1-S:5’AATCTAGAATGGAGCCGCCGACCAGCCA 3’;NPR1-A:5’CCCGGGACGGCGATGCGCGC 3’),在扩增产物的5′端加上了XbaI位点,用回收的经XbaI/SmaI酶切后的扩增产物取代pGXN4834上的597bp的XbaI/SmaI片段,得到重组质粒pGXN4821,这样该重组质粒的XbaI位点的下游紧接着就是用作全长SDKB的基因组基因的起始密码子的ATG。用XbaI和XhoI双酶切pGXN4821后,回收4439bp的外源片段,用大连(宝)生物工程公司的T4 DNA聚合酶补平两端后回收,再将该片段克隆到水稻表达载体pCambia1301-UbiN的SmaI位点,用限制性内切酶HindIII酶切重组质粒以筛选SDKB的基因组基因以正义方向插入到Ubiquitin启动子下游的克隆(SDKB的基因组基因如果是以正义方向克隆在Ubiquitin启动子下游则HindIII酶切后应该释放出一条约3kb的片段,如果是以反义方向克隆在启动子的下游酶切后应释放出一条约5.5kb的片段),得到桂99 SDKB的基因组基因全长正义水稻表达载体pCambia1301-UbiN-4408(图8)。应用三亲本接合的方法(Rogers SG,Horsch RB,Fraley RT.1986.Gene transfer in plants:production of transformed plantsusing Ti-plasmid vectors.Methods Enzymol.118:627-640)将重组质粒pCambia1301-UbiN-4408导入到根癌农杆菌EHA105中。
2、SDKB的基因组基因反义水稻表达载体的构建
用XbaI和SalI酶切pGXN4821,回收456bp小片段,该片段含有SDKB的基因组基因第一个外显子从起始密码子ATG起始的56bp序列。用大连(宝)生物工程公司的T4DNA聚合酶补平两端后再连接到水稻表达载体pCambia1301-UbiN的SmaI位点,筛选以反义方向插入到Ubiquitin启动子下游的重组质粒(首先用EcoRI酶切重组质粒鉴定是否连上456bp外源片段,如酶切后释放出约1.4kb的BNA片段则说明已克隆上456bp DNA片段,再用EcoRI/SacI双酶切已克隆上456bp DNA片段的重组质粒,如酶切后释放出两条大小分别为约1kb和400bp的DNA片段,则说明外源片段是以正义方向克隆到Ubiquitin启动子下游的;如酶切后释放出两条大小分别为约680bp和720bp的DNA片段,则说明外源片段是以反义方向克隆到Ubiquitin启动子下游的)。构建得到反义水稻表达载体pCambia1301-UbiN-400,它含有水稻SDKB的基因组基因从起始密码子ATG起始的456bp的序列,连接方向是反义的(图9)。应用三亲本接合的方法将重组质粒pCambia1301-UbiN-400导入到根癌农杆菌EHA105中。
实施例4、水稻的遗传转化及抗性鉴定
组织培养和转化的培养基列于表1。

1、水稻的遗传转化
(1)水稻种子的灭菌
将去好壳的水稻TP309(水稻台北309)和桂99的种子先用70%乙醇表面消毒2min,再用0.1%升汞溶液灭菌15-20min,最后用无菌水冲洗6-7次,再用无菌水浸泡3-4小时,然后将其接种到NB培养基(pH 5.8)上,于26±1℃暗培养8天左右。
(2)愈伤组织的继代
将长好的愈伤组织去掉芽和种子后,转到NB培养基(pH 5.8)进行继代培养14天,以备根癌农杆菌侵染。
(3)根癌农杆菌的培养
在愈伤组织转化前两天就开始用YEB培养基(每升含蛋白胨5g,牛肉膏5g,MgSO41g,酵母膏1g,蔗糖5g,pH 7.0)培养菌株EHA105/pCambia-UbiN-4408(含正义水稻表达载体)或EHA105/pCambia1301-UbiN-400(含反义水稻表达载体),同时加入利福平(Rif)、卡那霉素(Km)(终浓度均为25μg/ml),于28℃摇床培养过夜,第二天转接大瓶YEB培养基,大瓶培养基中含同样浓度的抗生素和100μmol/L的乙酰丁香酮(AS),培养到OD600=0.5时收集菌体,再用等体积的含100μmol/L乙酰丁香酮的AMM培养基(AA大量,AA氨基酸,MS有机,水解酪蛋白(Casamino acid),500mg/L;蔗糖,68.5g/L;pH5.2)悬浮菌体以备转化。
(4)共培养
取待转化的愈伤组织于灭菌三角瓶中,加入以上备好的含正义水稻表达载体pCambia1301-UbiN-4408的根癌农杆菌菌液转化水稻TP309的愈伤组织,含反义水稻表达载体pCambia1301-UbiN-400的根癌农杆菌菌液转化水稻桂99的愈伤组织,浸泡20分钟后倒去菌液,把愈伤组织倒入无菌培养皿中,再用无菌滤纸吸干多余的菌液,转接到置于NB培养基(含100μmol/L乙酰丁香酮,pH5.2)上的一张无菌滤纸上,28℃暗培养3天。
(5)筛选培养
将共培养3天后的愈伤组织转接于含头孢霉素(cefotaxime,300μg/ml)和潮霉素的NB培养基(TP309和桂99筛选的潮霉素终浓度分别为30μg/ml和25μg/ml,pH5.8)上筛选培养4周(每两周更换一次培养基),再将抗性愈伤组织转接于含头孢霉素(300μg/ml)和高浓度潮霉素的NB培养基(TP309和桂99筛选的潮霉素终浓度分别为50μg/ml和40μg/ml)上筛选培养4周(每两周更换一次培养基)。
(6)抗性愈伤组织的分化
取胚性抗性愈伤组织转接于MSF培养基(pH5.8)上,26℃光照(每天光照14小时,光照强度为3000Lux)培养2-3周长成小苗。
(7)抗性小苗的生根、炼苗及移栽
将再生出的小苗转接于1/2MS培养基(pH5.8)上,继续26℃光照培养以生根。将根长好的抗性小苗移于含1/2MS培养基(pH5.8)的玻璃管或瓶中继续26℃光照开放培养3天以炼苗,然后移栽到温室中(土壤盆栽)。
(8)植株鉴定
用重组质粒pCambia1301-UbiN-4408转化水稻TP309得到了潮霉素抗性分化小苗113株,全部移栽成活,目前已收获得到62个转基因株系的T1代种子。对成活的植株进行PCR和DNA测序及Southern杂交鉴定。
①以按照常规方法提取的未转基因TP309植株(阴性对照)和转基因的TP309植株的总DNA以及质粒pCambia 1301-UbiN-4408 DNA(阳性对照)为模板,用引物FSDKB1:5’TGAATAGGTAGCCTAGTTAATCTTTCTTGGGGTGC 3’,和RNOS:5’GCGGGACTCTAATCATAAAAACCCATCTCATAAAT 3’进行PCR扩增,结果如图10a所示,表明待检测的转基因植株都给出了与阳性对照同样大小的PCR片段。以已测序的经分析是正确的阳性对照的PCR产物作探针对转SDKB基因组基因的TP309 T1代植株的PCR产物做Southern杂交,结果如图10b所示,表明所有PCR产物都能和探针杂交,证实得到的PCR产物是特异性正确产物,从而证明它们都是转基因植株。图10a和图10b中,M为1kb ladder(片段大小从大到小依次为:10.0kb,8.0kb,6.0kb,5.0kb,4.0kb,3.5kb,3.0kb,2.5kb,2.0kb,1.5kb,1kb,750bp,500bp,250bp);1为阳性对照(以pCambia 1301-UbiN-4408为模板的PCR产物);2为阴性对照(未转基因TP309植株);3-28为26个转正义SDKB的基因组基因的TP309株系的T1代植株。
②随机取步骤①中的3株转正义SDKB的基因组基因的TP309的T1代植株的PCR产物进行双向双链测序,结果如图11所示,表明每个PCR产物都测得738bp的核苷酸序列且序列均相同,其中第1-602位碱基为SDKB的基因组基因3’端序列(带下划线-----的核苷酸序列);第603-612位碱基为载体序列(带下划线-的核苷酸序列);第613-738位碱基(带下划线-的核苷酸序列)为NOS终止子的上游序列。测得的PCR产物序列都和用同样引物测得的pCambia1301-UbiN-4408上对应序列完全一致,由此进一步证明PCR产物的序列都是正确的,PCR分析为阳性植株的皆为转SDKB的基因组基因的转基因植株。
③按照常规方法提取未转基因TP309植株(阴性对照)和转基因的TP309植株的总DNA,利用限制性内切酶HindIII酶切上述总DNA,以质粒pGX4821的3.1kbEcoRV/XhoI片段(序列表中序列1的自5’端第3606-6797位碱基)为探针,按照常规方法进行Southern杂交,结果如图12所示,表明所有植株都有一条3.9kb的内源杂交带,而转基因植株除了有内源杂交带外,还多出至少一条大小不同的杂交带,说明桂99的SDKB的基因组基因确实已被导入到TP309中并已整合到TP309基因组的不同位置上。图12中,1为未转基因的TP309;2-9为转基因植株。
用重组质粒pCambia1301-UbiN-400转化水稻桂99得到了潮霉素抗性分化小苗50株,移栽成活32株,目前已收获得到29个转基因株系的T1代种子。PCR检测表明这32个株系均为转基因植株,所有转基因植株都给出了与阳性对照同样大小的PCR片段,说明反义SDKB基因已导入到水稻桂99(图13)。其中,PCR鉴定所用引物为UBRNF(5’TGCATATACATGATGGCATATGCAGCATCTATTC 3’)和RNOS(5’GCGGGACTCTAATCATAAAAACCCATCTCATAAA 3’)。图13中,M为1kb ladder(片段大小从大到小依次为:10.0kb,8.0kb,6.0kb,5.0kb,4.0kb,3.5kb,3.0kb,2.5kb,2.0kb,1.5kb,1kb,750bp,500bp,250bp);1为阳性对照(以质粒pCambia1301-Ubi-400为模板);2为未转基因的水稻桂99;3-14为转基因植株。
2、转基因植株对水稻白叶枯病、稻瘟病的抗性检测
(1)应用剪叶接种法检测了21个株系的转正义的SDKB的基因组基因的TP309T1代植株对水稻白叶枯病的抗性,接种的水稻白叶枯病菌为水稻黄单胞菌水稻致病变种(Xanthomonas oryzae pv.oryzae)中国菌株13751,接种浓度为OD600=0.001,接种后14天量取病斑长度,结果是对照未转基因的TP309的平均病斑长度为9.21cm,而转正义的SDKB的基因组基因的植株的平均病斑长度均短于2.5cm(表2,图14),表明TP309获得了正义的SDKB基因组基因后对水稻白叶枯病的抗性显著增强。图14中,1为未转基因的TP309;2-9为转SDKB基因组基因的TP309的T1代植株。

(2)应用喷雾孢子接种法检测了20个株系的转正义的SDKB的基因组基因的TP309T2代植株对稻瘟病的抗性,接种的稻瘟病菌为CHL0742中国菌株,接种浓度为每个视野30-50个孢子,接种后8天调查发病情况,结果表明未转基因的TP309(对照)的平均病情指数为52.63,而转SDKB的基因组基因植株有12个株系的平均病情指数均在30以下,有7个株系的平均病情指数在30-40之间,只有1个株系的平均病情指数在50以上,为51.85(表3,图15)。说明感病的TP309获得了正义的SDKB基因组基因后对稻瘟病的抗性显著增强。图15中,1为未转基因的TP309;2为转SDKB基因组基因的TP309。

用实施例1中RT-PCR得到的228bp的PCR产物作探针和经EcoRI酶切的7个水稻品种(838,K17B,ZSP选1,P702B,博B,ZSP517,IR36)的总DNA杂交。用经EcoRI酶切的桂99的总DNA作对照。IR36抗稻瘟病、白叶枯病,ZSP517抗稻瘟病但感白叶枯病,其它5个水稻品种(838,K17B,ZSP选1,P702B,博B)均感这两种病害。结果该7个水稻品种和桂99均给出相同的一个约25kb的杂交带(图17)。说明SDKB基因在水稻中普遍存在。M为λDNA/HindIII;1为IR36/EcoRI,2为D702B/EcoRI;3为K17B/EcoRI;4为ZSP选1/EcoRI;5为ZSP517/EcoRI;6为博IIR/EcoRI;7为838/EcoRI;8为桂99/EcoRI。
