Abstract
本发明公开了一种漱口水及其制备方法,该漱口水以总质量满足100%计,由下述质量百分比的各原料组成:壳聚糖0.1‑0.5%;低聚糖2‑8%;三聚磷酸盐0.1‑0.4%;复合氨基酸0.05‑0.2%;茶多酚0.1‑0.3%;木瓜蛋白酶0.05‑0.15%;木糖醇1‑10%;表面活性剂4‑12%;余量为纯化水。按质量百分比称取各原料,经溶解、超声处理、混合、湿热灭菌等步骤,即得。具有能有效抑菌、无毒副作用、于口腔内正常菌群无害、生物安全性高的特点。
Description
技术领域
本发明属于医疗卫生技术领域,具体来说涉及一种漱口水,同时还涉及该漱口水的制备方法。
背景技术
就口腔卫生新理念而言,仅早晚刷牙已经无法达到口腔卫生标准,但随时随地拿出牙刷、牙膏进行口腔清洁又极不现实。
漱口水,能清洁口腔、预防龋齿和牙周炎等;使用十分方便,可有效拒绝食物等残渣对牙齿的侵蚀;是牙齿脱落的老人、不会刷牙的小孩以及不能忍受刷牙的牙病患者口腔卫生保障的最佳选择。
目前,漱口水大致可以分为含氟化物漱口水、防牙菌膜漱口水、抑制牙菌膜漱口水、防敏感漱口水、草本漱口水以及传统中草药类漱口水。这些已有的漱口水,均属于广谱高效抑菌抗菌类漱口水。而越来越多的研究表明,长期使用广谱抑菌抗菌类漱口水会造成口腔内正常菌群失调,从而出现黏膜溃烂、脱膜甚至坏死的症状,严重时更会导致多处溃疡。这是本领域呈待解决的技术问题。
发明内容
本发明的目的在于克服前述困难而提供一种能有效抑菌、无毒副作用、于口腔内正常菌群无害、生物安全性高的漱口水。
本发明的另一目的在于提供该漱口水的制备方法。
本发明的一种漱口水,以总质量满足100%计,由下述质量百分比的各原料组成:
上述的一种漱口水,其中:所述低聚糖为低聚果糖、低聚木糖、水苏糖、低聚异麦芽糖中的一种或按任意质量比混合的两种以上混合物。
上述的一种漱口水,其中:所述三聚磷酸盐为三聚磷酸钠、三聚磷酸钾、三聚磷酸铵中的一种或按任意质量比混合的两种以上混合物。
上述的一种漱口水,其中:所述复合氨基酸为半胱氨酸、亮氨酸、色氨酸、苯丙氨酸、谷氨酸按任意质量比混合的两种以上混合物。
上述的一种漱口水,其中:所述表面活性剂为泊洛沙姆、蔗糖酯、亲水单甘脂中的一种或按质量比2:1:1的混合物。
本发明的一种漱口水的制备方法,包括以下步骤:
(1)按质量百分比称取下述原料:壳聚糖0.1-0.5%,低聚糖2-8%,三聚磷酸盐0.1-0.4%,复合氨基酸0.05-0.2%,茶多酚0.1-0.3%,木瓜蛋白酶0.05-0.15%,木糖醇1-10%,表面活性剂4-12%,余量为纯化水;
(2)将纯化水按体积平均分为4份,取1/4体积的纯化水溶解壳聚糖,搅拌至溶解完全后超声处理9-20min,得壳聚糖水溶液;
(3)将低聚糖加入壳聚糖水溶液中,搅拌至溶解完全后用孔径为0.45-1um的滤膜过滤,得滤液A;
(4)另取1/4体积的纯化水溶解复合氨基酸,搅拌至溶解完全后加入茶多酚及木瓜蛋白酶,继续搅拌至溶解完全,得混合溶液B,控制所述混合溶液B的温度为4-30℃;
(5)将混合溶液B加入滤液A中,得混合溶液C;
(6)取剩余1/2体积的纯化水溶解三聚磷酸盐,得三聚磷酸盐水溶液;
(7)将三聚磷酸盐水溶液加入混合溶液C中,一边加入一边搅拌,加入速度为45-50ml/min,搅拌速度为50-60r/min;再加入木糖醇及表面活性剂,得混合溶液D;
(8)将混合溶液D用孔径为0.45-5um的滤膜过滤后,得滤液E;将滤液E于115℃,8≤F0≤30条件下采常规方法进行湿热灭菌,即得。
本发明与现有技术相比,具有明显有益效果,从以上技术方案可知:本发明所提供的漱口水,采壳聚糖、低聚糖、三聚磷酸盐、复合氨基酸、茶多酚、木瓜蛋白酶及木糖醇为主要原料,通过各原料之间的协同作用,可有效清洁口腔、防治牙菌斑、促进口腔及肠道有益菌的繁殖生长且抑制有害菌的增殖。其中:壳聚糖是一种具有生物活性的非水溶性物质,在漱口水中主要发挥防治牙菌斑、修复口腔溃疡创面及抗渗出功能,配合多聚磷酸盐,可发生交联反应生成纳米粒,纳米粒作为药物载体,既有助于药物吸收,又具有靶向性作用;低聚糖具有选择性抑菌作用,特别是能被双歧杆菌利用,可促进肠内有益菌的增殖;木瓜蛋白酶可以水解多种食物蛋白,有助于人体对有效物质的吸收及肠道消化,通过茶多酚与它络合,可形成比较稳定的络合物,解决木瓜蛋白酶所存在的不稳定及对温度敏感的问题,利于原料的综合利用;木糖醇具有降低血糖、防治龋齿、改善肝功能、调节肠道功能等作用,不仅可改善漱口水的口感,还能作为营养补充剂使用。因不属于广谱高效抑菌抗菌类漱口水,在能有效抑菌及抗渗出的同时,还具有无毒副作用、于口腔内正常菌群无害、生物安全性高的特点。
所提供的制备方法工艺科学、操作简便且易于推广。
具体实施方式
下面通过实验例及实施例来进一步说明本发明的有益效果。
实验例:
采实施例1所得漱口水作为实验样品进行抑菌实验(以实施例2或实施例3作为实验样品进行抑菌实验,方法相同),抑菌实验中采用的菌株均为常规口腔致病菌,包括:金黄色葡萄球菌、变形链球菌、大肠杆菌及白色念珠菌。
A实验器材:
营养琼脂培养基、玫瑰红钠培养基、硫乙醇培养基、改良马丁培养基、培养皿、烧杯、三角瓶、接种环、酒精灯、微量移液枪、直角玻璃棒、金黄色葡萄球菌、变形链球菌、大肠杆菌、白色念珠菌、实验样品、对照品、高压蒸汽灭菌锅、100mL具塞试管。
B实验方法:
(1)培养基的制备:
固体培养基:
营养琼脂培养基(用于金黄色葡萄球菌、大肠杆菌、变形链球菌):称取33g营养琼脂培养基,用1000mL已加热至100℃的纯化水溶解完全,120℃高压(101kpa)蒸汽灭菌20min,冷却至50-60℃,恒温保存,备用;
玫瑰红钠琼脂培养基(用于白色念珠菌):称取30.5g玫瑰红钠琼脂培养基,用1000mL已加热至100℃的纯化水溶解完全,120℃高压(101kpa)蒸汽灭菌20min,冷却至50-60℃,恒温保存,备用。
液体培养基:
硫乙醇液体培养基(用于金黄色葡萄球菌、大肠杆菌、变形链球菌):称取硫乙醇培养基29.3g,用1000mL已加热至100℃的纯化水溶解完全;取6支100mL试管,分别倒15-20mL已溶解完全的硫乙醇培养基于试管中;将剩下的培养基与6支试管于121℃高压(101kpa)蒸汽灭菌20min,冷却,备用;
改良马丁液体培养基(用于白色念珠菌):称取改良马丁培养基28.5g,用1000mL已加热至100℃的纯化水溶解完全;取2支试管,分别倒入15-20mL已溶解完全的改良马丁培养基于试管中;将剩下的培养基与2支试管于121℃高压(101kpa)蒸汽灭菌20min,冷却,备用。
(2)液体培养:
取上述装有硫乙醇液体培养基的6支试管及装有改良马丁液体培养基的2支试管,在超净工作台中进行转接。
6支装有硫乙醇液体培养基的试管每两支分一组,共三组;第一组转接金黄色葡萄球菌,用接种环转接金黄色葡萄球菌,并标记金黄色葡萄球菌;第二组转接大肠杆菌,用接种环转接大肠杆菌,并标记大肠杆菌;第三组转接变形链球菌,用接种环转接变形链球菌,并标记变形链球菌。
2支装有改良马丁液体培养基的试管为一组,转接白色念珠菌,用接种环进行转接,并标记;
细菌35℃、真菌28℃培养24h后取出,选择每组中长势较好的试管中所培养得到的菌株,作为液体菌株,备用。
(3)菌株稀释:
分别取所得4种液体菌株0.1mL,用灭菌后的生理盐水进行10倍梯度稀释,稀释至10-5倍,得金黄色葡萄球菌待测培养物、大肠杆菌待测培养物、变形链球菌待测培养物及白色念珠菌待测培养物。
(4)实验样品:
采实施例1所得漱口水作为实验样品,每种菌株做两个平行,最后取平均值。
(5)实验步骤:
①样品组:
a、取8支15mL离心管,两支为一组,每组中标记第一支为样品组1号,第二支为样品组2号,共四组,前三组分别为大肠杆菌、金黄色葡萄球菌、变形链球菌,最后一组为白色念珠菌;
b、前三组每支离心管分别添加8mL硫乙醇液体培养基,第四组添加8mL改良马丁液体培养基;
c、第一组每支添加0.5mL大肠杆菌待测培养物,第二组每支添加0.5mL金黄色葡萄球菌待测培养物,第三组每支添加0.5mL变形链球菌待测培养物,第四组每支添加0.5mL白色念珠菌待测培养物;
d、最后于每支离心管中添加0.5mL样品液,即实施例1所得漱口水;
e、细菌35℃、真菌28℃培养1h后取出,离心,去除沉淀取滤液,得样品。
②对照组:
a、取4支15mL离心管,前三支离心管添加8.5mL硫乙醇液体培养基,第四支添加8.5mL改良马丁液体培养基;
b、第一支添加0.5mL大肠杆菌待测培养物,第二支添加0.5mL金黄色葡萄球菌待测培养物,第三支添加0.5mL变形链球菌待测培养物,第四支添加0.5mL白色念珠菌待测培养物;
c、细菌35℃、真菌28℃培养1h后取出,离心,去除沉淀取滤液,得对照品。
③取出3个培养皿为一组,即白色念珠菌组,在超净工作台内将已灭菌且冷却至50-60℃玫瑰红钠琼脂培养基分别倒入3个培养皿中,每个培养皿20-25mL,得3个平板;其中,第一个平板标记为A,第二个平板标记为B,第三个平板标记为C;待玫瑰红钠琼脂培养基冷却凝固后,用微量移液枪移取100μl白色念珠菌样品组1号于A号平板中,再用微量移液枪移取100μl白色念珠菌样品组2号于B号平板中,最后用微量移液枪移取100μl白色念珠菌对照品于C平板中,进行涂平板,即用直角玻璃棒分别将白色念珠菌均匀地涂布于平板内(在酒精灯灯芯10cm范围内进行);
再取出9个培养皿,在超净工作台内将已灭菌且冷却至50-60℃的营养琼脂培养基分别倒入9个培养皿中,每个培养皿20-25mL,得9个平板;其中,三个一组,第一组为大肠杆菌组,第二组为金黄色葡萄球菌组,第三组为变形链球菌组;按照白色念珠菌组进行实验;
标记日期、样品及菌株名称;
培养条件:细菌:35℃,48h;真菌:28℃,72h。
C实验结果:
根据样品组与对照组菌落数来确定抑菌率,抑菌率=对照组菌落数-样品组菌落数/对照组菌落数;
样品组和对照组菌落数如表1所示:

每个样品对应每种菌株的抑菌率如表2所示:

从以上结果可知:任一实施例对应的任一菌株的抑菌率均大于90%,故,无论是真菌还是细菌,使用本发明漱口水都能有效起到抑菌作用。
实施例1
一种漱口水,包括以下步骤:
(1)称取壳聚糖0.1g,低聚果糖2g,三聚磷酸钠0.1g,按质量比3:2混合的天冬氨酸及谷氨酸0.05g,茶多酚0.1g,木瓜蛋白酶0.05g,木糖醇1g,泊洛沙姆4g,纯化水92.6g;
(2)将纯化水按体积平均分为4份,取1/4体积的纯化水溶解壳聚糖,搅拌至溶解完全后超声处理9min,得壳聚糖水溶液;
(3)将低聚果糖加入壳聚糖水溶液中,搅拌至溶解完全后用孔径为0.45um的滤膜过滤,得滤液A;
(4)另取1/4体积的纯化水溶解天冬氨酸及谷氨酸,搅拌至溶解完全后加入茶多酚及木瓜蛋白酶,继续搅拌至溶解完全,得混合溶液B,控制所述混合溶液B的温度为4℃;
(5)将混合溶液B加入滤液A中,得混合溶液C;
(6)取剩余1/2体积的纯化水溶解三聚磷酸钠,得三聚磷酸盐水溶液;
(7)将三聚磷酸盐水溶液加入混合溶液C中,一边加入一边搅拌,加入速度为45ml/min,搅拌速度为50r/min;再加入木糖醇及泊洛沙姆,得混合溶液D;
(8)将混合溶液D用孔径为0.45um的滤膜过滤后,得滤液E;将滤液E于115℃,F0=8条件下采常规方法进行湿热灭菌,即得。
实施例2
一种漱口水,包括以下步骤:
(1)称取壳聚糖0.5g,低聚木糖8g,按质量比1:3混合的三聚磷酸钠及三聚磷酸钾0.4g,按质量比3:1谷氨酸及亮氨酸0.2g,茶多酚0.3g,木瓜蛋白酶0.15g,木糖醇10g,蔗糖酯12g,纯化水68.45g;
(2)将纯化水按体积平均分为4份,取1/4体积的纯化水溶解壳聚糖,搅拌至溶解完全后超声处理20mi n,得壳聚糖水溶液;
(3)将低聚木糖加入壳聚糖水溶液中,搅拌至溶解完全后用孔径为1um的滤膜过滤,得滤液A;
(4)另取1/4体积的纯化水溶解谷氨酸及亮氨酸,搅拌至溶解完全后加入茶多酚及木瓜蛋白酶,继续搅拌至溶解完全,得混合溶液B,控制所述混合溶液B的温度为30℃;
(5)将混合溶液B加入滤液A中,得混合溶液C;
(6)取剩余1/2体积的纯化水溶解三聚磷酸钠及三聚磷酸钾,得三聚磷酸盐水溶液;
(7)将三聚磷酸盐水溶液加入混合溶液C中,一边加入一边搅拌,加入速度为50ml/min,搅拌速度为60r/min;再加入木糖醇及蔗糖酯,得混合溶液D;
(8)将混合溶液D用孔径为5um的滤膜过滤后,得滤液E;将滤液E于115℃,F0=30条件下采常规方法进行湿热灭菌,即得。
实施例3
一种漱口水,包括以下步骤:
(1)称取壳聚糖0.3g,按质量比2:3混合的水苏糖、低聚异麦芽糖5g,按质量比1:1:2混合的三聚磷酸钠、三聚磷酸钾及三聚磷酸铵0.25g,按质量比2:2:1混合的天冬氨酸、谷氨酸及亮氨酸0.125g,茶多酚0.2g,木瓜蛋白酶0.1g,木糖醇5.5g,按质量比2:1:1混合的泊洛沙姆、蔗糖酯及亲水单甘脂8g,纯化水80.525g;
(2)将纯化水按体积平均分为4份,取1/4体积的纯化水溶解壳聚糖,搅拌至溶解完全后超声处理14.5min,得壳聚糖水溶液;
(3)将水苏糖、低聚麦芽糖加入壳聚糖水溶液中,搅拌至溶解完全后用孔径为0.7um的滤膜过滤,得滤液A;
(4)另取1/4体积的纯化水溶解天冬氨酸、谷氨酸及亮氨酸,搅拌至溶解完全后加入茶多酚及木瓜蛋白酶,继续搅拌至溶解完全,得混合溶液B,控制所述混合溶液B的温度为17℃;
(5)将混合溶液B加入滤液A中,得混合溶液C;
(6)取剩余1/2体积的纯化水溶解三聚磷酸钠、三聚磷酸钾及三聚磷酸铵,得三聚磷酸盐水溶液;
(7)将三聚磷酸盐水溶液加入混合溶液C中,一边加入一边搅拌,加入速度为47.5ml/min,搅拌速度为55r/min;再加入木糖醇及泊洛沙姆、蔗糖酯、亲水单甘脂,得混合溶液D;
(8)将混合溶液D用孔径为2.7um的滤膜过滤后,得滤液E;将滤液E于115℃,F0=19条件下采常规方法进行湿热灭菌,即得。

1.一种漱口水,其特征在于:以总质量满足100%计,由下述质量百分比的各原料组成:

3.如权利要求1或2所述的一种漱口水,其特征在于:所述三聚磷酸盐为三聚磷酸钠、三聚磷酸钾、三聚磷酸铵中的一种或按任意质量比混合的两种以上混合物。
4.如权利要求1所述的一种漱口水,其特征在于:所述复合氨基酸为半胱氨酸、亮氨酸、色氨酸、苯丙氨酸、谷氨酸按任意质量比混合的两种以上混合物。
5.如权利要求1或2或4所述的一种漱口水,其特征在于:所述表面活性剂为泊洛沙姆、蔗糖酯、亲水单甘脂中的一种或按质量比2:1:1的混合物。
6.一种漱口水的制备方法,包括以下步骤:
(1)按质量百分比称取下述原料:壳聚糖0.1-0.5%,低聚糖2-8%,三聚磷酸盐0.1-0.4%,复合氨基酸0.05-0.2%,茶多酚0.1-0.3%,木瓜蛋白酶0.05-0.15%,木糖醇1-10%,表面活性剂4-12%,余量为纯化水;
(2)将纯化水按体积平均分为4份,取1/4体积的纯化水溶解壳聚糖,搅拌至溶解完全后超声处理9-20min,得壳聚糖水溶液;
(3)将低聚糖加入壳聚糖水溶液中,搅拌至溶解完全后用孔径为0.45-1um的滤膜过滤,得滤液A;
(4)另取1/4体积的纯化水溶解复合氨基酸,搅拌至溶解完全后加入茶多酚及木瓜蛋白酶,继续搅拌至溶解完全,得混合溶液B,控制所述混合溶液B的温度为4-30℃;
(5)将混合溶液B加入滤液A中,得混合溶液C;
(6)取剩余1/2体积的纯化水溶解三聚磷酸盐,得三聚磷酸盐水溶液;
(7)将三聚磷酸盐水溶液加入混合溶液C中,一边加入一边搅拌,加入速度为45-50ml/min,搅拌速度为50-60r/min;再加入木糖醇及表面活性剂,得混合溶液D;
(8)将混合溶液D用孔径为0.45-5um的滤膜过滤后,得滤液E;将滤液E于115℃,8≤F0≤30条件下采常规方法进行湿热灭菌,即得。
