技术领域
本发明涉及生物材料及蜘蛛丝蛋白领域,特别涉及一种含苦参的蜘蛛丝蛋白组织修复材料。
背景技术
组织工程技术是一项用于治疗组织和器官缺损的新手段,由生物支架、细胞、生长因子等元素组成。其中,生物支架作为细胞生长、增殖以及新组织形成的物理支撑,在组织工程中起着重要作用。组织工程旨在恢复重建受损组织或器官的结构和功能完整性,而现今还不能解决复杂组织器官的修复。大组织、复杂器官不像简单的薄层组织可以通过小分子扩散获得营养物质和氧气,它们需要通过庞大的血管系统才能获得营养物质和氧气。修复损伤的大组织和复杂器官的生物材料必须具有促血管形成的能力才能使大组织和复杂器官具有生物功能。
丝素蛋白具有优良的生物相容性、可降解性、可改良性和机械拉伸性能等。丝素蛋白中含有羟基、氨基、羧基等活性基团,可以与其他高分子聚合物通过氢键、静电作用、螯合效应等相结合,形成性能优越的复合生物支架材料。但现有技术中多为单纯的丝素蛋白材料,很难达到理想促血管形成及组织修复功能。
发明内容
针对现有技术中存在的问题,本发明提供了一种含苦参的蜘蛛丝蛋白组织修复材料,相对现有技术中单纯的丝素蛋白材料具有更好的组织修复功能。
为了解决上述问题本发明采用以下技术方案:
一种含苦参的蜘蛛丝蛋白组织修复材料,为包含蜘蛛丝蛋白的膜状物,内部平均孔密度为1-2000个/mm2,平均孔径为1-300μm,所述的膜状物包含以下重量份数的原料:蜘蛛丝蛋白溶液70-90份、蜘蛛丝蛋白纳米纤维溶液5-6份、苦参提取液1-2份、塞来昔布1-2份、精油1-2份。
优选的,所述的苦参提取液采用以下方法制备:称取苦参25g,粉碎成粗粉,加入其重量10倍的水,在90-100℃条件下煎煮提取1-1.5h,抽滤得滤液1;再向滤渣中加入6倍滤渣重量的蒸馏水,在70-80℃条件下煎煮提取1-1.5h,抽滤得滤液2;将滤液1、滤液2合并,在温度为40-50℃、真空度为0.05-0.08Pa条件下减压浓缩至苦参重量的4倍,过滤即得苦参提取液。
优选的,所述的蜘蛛丝蛋白纳米纤维溶液采用以下方法制备:取蜘蛛丝蛋白溶液置于60℃恒温箱内浓缩至原体积的一半,再转入通风橱内浓缩至质量分数为20-30%得浓缩液,将浓缩液放入4℃冰箱储存2d后将浓缩液加水稀释到质量分数为0.5%,充分均匀搅拌后放入恒温箱内孵育至形成半透明凝胶状的蜘蛛丝蛋白纳米纤维溶液。
优选的,所述的蜘蛛丝蛋白纳米纤维溶液中蜘蛛丝蛋白纳米纤维的长度为1μm,直径为10-20nm。
优选的,所述的蜘蛛丝蛋白溶液采用以下方法制备:将2L去离子水烧热至沸腾,加入4.24g碳酸钠及5g蜘蛛丝,煮沸30min,用去离子水清洗干净、烘干后,用9.3mol/L的LiBr溶液溶解;待完全溶解后将溶液装入透析袋,去离子水透析72h,9000r/min离心20min,重复两次,得蜘蛛丝蛋白溶液。
优选的,所述的蜘蛛丝蛋白溶液为蜘蛛丝丝素蛋白溶液。
优选的,所述的膜状物采用以下方法制备:将蜘蛛丝蛋白溶液与蜘蛛丝蛋白纳米纤维溶液混合,并加入苦参提取液、塞来昔布、精油混合均匀制得纺丝液,纺丝液采用静电纺丝工艺制得三维无纺网状结构膜。
进一步优选的,所述的静电纺丝工艺为:电压12-18KV,纺丝高度5-10cm,卷绕速度100-300rpm,纺丝液流量2-5ml/h,加热温度50-150℃。
优选的,所述的三维无纺网状结构膜中纤维直径为50nm-20um,纤维间孔隙为l-500um,膜厚度为50nm-20mm。
本发明的有益效果在于:
1)本发明的组织修复材料相对于单纯的蚕丝丝素蛋白材料,为低模量的软组织修复材料,具有更好的促血管形成及组织修复功能。
2)用于组织工程的三维多孔材料必须具有较高的孔隙率,具有相互连通的三维结构,以便于细胞的增殖、迁移,为细胞的生长提供足够的营养物质。加入纳米纤维后,提高了支架的成孔性支架孔壁上具有纳米纤维结构,大大增加了支架的比表面积。
3)添加中草药成分苦参提取液,及塞来昔布、精油,进一步提高了材料的组织修复的能力。
4)苦参可以治疗阴肿阴痒,湿疹,湿疮,皮肤瘙痒,疥癣麻风,外治滴虫性阴道炎。
具体实施方式
下面结合实施例对本发明做进一步的描述。
实施例1
一种含苦参的蜘蛛丝蛋白组织修复材料,为包含蜘蛛丝蛋白的膜状物,内部平均孔密度为1-2000个/mm2,平均孔径为1-300μm,其特征在于,所述的膜状物包含以下重量份数的原料:蜘蛛丝蛋白溶液70份、蜘蛛丝蛋白纳米纤维溶液5份、苦参提取液1份、塞来昔布1份、精油1份。

所述的蜘蛛丝蛋白纳米纤维溶液采用以下方法制备:取蜘蛛丝蛋白溶液置于60℃恒温箱内浓缩至原体积的一半,再转入通风橱内浓缩至质量分数为20%得浓缩液,将浓缩液放入4℃冰箱储存2d后将浓缩液加水稀释到质量分数为0.5%,充分均匀搅拌后放入恒温箱内孵育至形成半透明凝胶状的蜘蛛丝蛋白纳米纤维溶液。
所述的蜘蛛丝蛋白纳米纤维溶液中蜘蛛丝蛋白纳米纤维的长度为1μm,直径为10-20nm。
所述的蜘蛛丝蛋白溶液采用以下方法制备:将2L去离子水烧热至沸腾,加入4.24g碳酸钠及5g蜘蛛丝,煮沸30min,用去离子水清洗干净、烘干后,用9.3mol/L的LiBr溶液溶解;待完全溶解后将溶液装入透析袋,去离子水透析72h,9000r/min离心20min,重复两次,得蜘蛛丝蛋白溶液。
所述的蜘蛛丝蛋白溶液为蜘蛛丝丝素蛋白溶液。
所述的膜状物采用以下方法制备:将蜘蛛丝蛋白溶液与蜘蛛丝蛋白纳米纤维溶液混合,并加入苦参提取液、塞来昔布、精油混合均匀制得纺丝液,纺丝液采用静电纺丝工艺制得三维无纺网状结构膜。
所述的静电纺丝工艺为:电压12-18KV,纺丝高度5-10cm,卷绕速度100-300rpm,纺丝液流量2-5ml/h,加热温度50-150℃。
所述的三维无纺网状结构膜中纤维直径为50nm-20um,纤维间孔隙为l-500um,膜厚度为50nm-20mm。
实施例2
一种含苦参的蜘蛛丝蛋白组织修复材料,为包含蜘蛛丝蛋白的膜状物,内部平均孔密度为1-2000个/mm2,平均孔径为1-300μm,其特征在于,所述的膜状物包含以下重量份数的原料:蜘蛛丝蛋白溶液90份、蜘蛛丝蛋白纳米纤维溶液6份、苦参提取液2份、塞来昔布2份、精油2份。
所述的苦参提取液采用以下方法制备:称取苦参25g,粉碎成粗粉,加入其重量10倍的水,在90-100℃条件下煎煮提取1.5h,抽滤得滤液1;再向滤渣中加入6倍滤渣重量的蒸馏水,在70-80℃条件下煎煮提取1.5h,抽滤得滤液2;将滤液1、滤液2合并,在温度为40-50℃、真空度为0.05-0.08Pa条件下减压浓缩至苦参重量的4倍,过滤即得苦参提取液。
所述的蜘蛛丝蛋白纳米纤维溶液采用以下方法制备:取蜘蛛丝蛋白溶液置于60℃恒温箱内浓缩至原体积的一半,再转入通风橱内浓缩至质量分数为30%得浓缩液,将浓缩液放入4℃冰箱储存2d后将浓缩液加水稀释到质量分数为0.5%,充分均匀搅拌后放入恒温箱内孵育至形成半透明凝胶状的蜘蛛丝蛋白纳米纤维溶液。
所述的蜘蛛丝蛋白纳米纤维溶液中蜘蛛丝蛋白纳米纤维的长度为1μm,直径为10-20nm。

所述的膜状物采用以下方法制备:将蜘蛛丝蛋白溶液与蜘蛛丝蛋白纳米纤维溶液混合,并加入苦参提取液、塞来昔布、精油混合均匀制得纺丝液,纺丝液采用静电纺丝工艺制得三维无纺网状结构膜。
所述的静电纺丝工艺为:电压12-18KV,纺丝高度5-10cm,卷绕速度100-300rpm,纺丝液流量2-5ml/h,加热温度50-150℃。
所述的三维无纺网状结构膜中纤维直径为50nm-20um,纤维间孔隙为l-500um,膜厚度为50nm-20mm。
将实施例1制得的组织修复材料(S)、实施例2制得的组织修复材料(SS),与由纯蚕丝丝丝素蛋白制成的组织修复材料(SSS)的性能进行对比,采用具有相同的组织损伤程度的小白鼠进行实验,将材料S、SS及SSS分别敷于小白鼠的受伤位置,每天更换两次,连续使用10天,采用观察评分法,分别按1-5分对小白鼠受伤组织恢复程度进行打分,各项使用效果最优为5分,得出的实验数据见表1。

由实验结果可以看出:采用蜘蛛丝蛋白替代蚕丝蛋白,并向蜘蛛丝丝素蛋白中添加蜘蛛丝蛋白纳米纤维、苦参、塞来昔布及精油,可以有效提升丝素蛋白的组织修复能力及抗炎效果。
Claims (8)
1.一种含苦参的蜘蛛丝蛋白组织修复材料,为包含蜘蛛丝蛋白的膜状物,内部平均孔密度为1-2000个/mm2,平均孔径为1-300μm,其特征在于,所述的膜状物包含以下重量份数的原料:蜘蛛丝蛋白溶液70-90份、蜘蛛丝蛋白纳米纤维溶液5-6份、苦参提取液1-2份、塞来昔布1-2份、精油1-2份;
所述的苦参提取液采用以下方法制备:称取苦参25g,粉碎成粗粉,加入其重量10倍的水,在90-100℃条件下煎煮提取1-1.5h,抽滤得滤液1;再向滤渣中加入6倍滤渣重量的蒸馏水,在70-80℃条件下煎煮提取1-1.5h,抽滤得滤液2;将滤液1、滤液2合并,在温度为40-50℃、真空度为0.05-0.08Pa条件下减压浓缩至苦参重量的4倍,过滤即得苦参提取液。
2.根据权利要求1所述的一种含苦参的蜘蛛丝蛋白组织修复材料,其特征在于,所述的蜘蛛丝蛋白纳米纤维溶液采用以下方法制备:取蜘蛛丝蛋白溶液置于60℃恒温箱内浓缩至原体积的一半,再转入通风橱内浓缩至质量分数为20-30%得浓缩液,将浓缩液放入4℃冰箱储存2d后将浓缩液加水稀释到质量分数为0.5%,充分均匀搅拌后放入恒温箱内孵育至形成半透明凝胶状的蜘蛛丝蛋白纳米纤维溶液。
3.根据权利要求2所述的一种含苦参的蜘蛛丝蛋白组织修复材料,其特征在于:所述的蜘蛛丝蛋白纳米纤维溶液中蜘蛛丝蛋白纳米纤维的长度为1μm,直径为10-20nm。

6.根据权利要求1所述的一种含苦参的蜘蛛丝蛋白组织修复材料,其特征在于,所述的膜状物采用以下方法制备:将蜘蛛丝蛋白溶液与蜘蛛丝蛋白纳米纤维溶液混合,并加入苦参提取液、塞来昔布、精油混合均匀制得纺丝液,纺丝液采用静电纺丝工艺制得三维无纺网状结构膜。
7.根据权利要求6所述的一种含苦参的蜘蛛丝蛋白组织修复材料,其特征在于,所述的静电纺丝工艺为:电压12-18KV,纺丝高度5-10cm,卷绕速度100-300rpm,纺丝液流量2-5ml/h,加热温度50-150℃。
