Abstract
本发明提供了一种青蒿口腔抑菌液。本发明的抑菌液以青蒿、生姜、柠檬、刺梨、鱼腥草、金银花等为主要原料进行仿生微生态发酵,熬合后有效地将发酵原料里面的活性成分萃取出来,经过发酵的原液具有良好的作用和效果。本发明的青蒿口腔抑菌液口感好、柔和、无激刺、无异味,有特定的发酵气味,有强的抑菌效果,适合作为口腔护理液特别是喷雾型护理液。

技术领域
本发明涉及口腔护理技术领域,具体来说,本发明涉及一种口腔抑菌液。
背景技术
口腔位于消化道的最前端,为病原微生物侵入的主要途径之一,与人的健康生存息息相关。细菌及病毒感染,空气污染,用声过度,烟酒及辛辣燥热刺激食物等因素易导致口腔问题和咽喉炎症,如果失治、误治、错治,易演变成牙周炎、慢性咽喉炎等。口腔护理不仅能预防口腔疾病,减少继发感染等并发症,而且还能增进食欲,促进患者舒适,为患者日常护理工作中重要的部分。
过去传统口腔护理常用的护理溶液有呋喃西林,过氧化氢,碳酸氢钠,生理盐水等,作用均不是很明显,长期应用含抗菌化学成分的漱口液可能引起口腔菌群失调,易产生耐药性,副反应也相对较多。我国是中草药的大国,利用各种植物提取物制作口腔护理溶液应用广泛,但所获得的口腔护理溶液大多存在抑菌效果不佳、刺激性强等问题,因此需要开发更多的抑菌效果强、刺激性小的口腔抑菌液。
发明内容
为了解决现有技术中存在的上述问题,本发明提供了一种青蒿口腔抑菌液,其对金黄色葡萄球菌、大肠埃希菌、白色念珠菌、牙龈卟啉单胞菌等多种病菌具有强的抑菌效果,并且无毒无刺激。
本发明的青蒿口腔抑菌液包括:
抑菌原液20~40份
乙醇5~8份
薄荷油0.03~0.05份
香兰素香精0.3~1份
甜菊糖苷0~1份
水30~65
所述抑菌原液由青蒿、艾叶、生姜、银杏叶、桂花、柠檬、刺梨、金银花、鱼腥草、蒲公英经酶解、发酵、熬合制得。
本发明另一方面提供所述青蒿口腔抑菌液的制备方法。
有益效果
本发明提供了一种青蒿口腔抑菌液。所述口腔抑菌液具有强的抑菌效果,主要是针对清除口腔异味,抑制口腔有害菌,同时对于一些皮肤细菌感染,部分真菌感染也有一定的抑制效果。本发明的抑菌液以青蒿、生姜、柠檬、刺梨、鱼腥草、金银花等为主要原料进行仿生微生态发酵,熬合后有效地将发酵原料里面的活性成分萃取出来,经过发酵的原液,可溶性固形物3~5%,pH3~4,总酸20~30g/kg,具有良好的作用和效果。本发明添加的薄荷油在作用于口腔后有清凉感,加入乙醇一是可以解决油水分离问题溶解性问题,二是喷雾形态更佳,加入甜菊糖苷可进一步提高抑菌液的口感。此外,本发明的抑菌液刺激性小,毒性低。总之,本发明的青蒿口腔抑菌液口感好、柔和、无激刺、无异味,有特定的发酵气味,有强的抑菌效果,适合作为口腔护理液特别是喷雾型护理液。

在一个实施方案中,酶解所用的酶为纤维素酶、脂肪酶、中性蛋白酶、淀粉酶的复合酶,重量比为1~2:1~2:1~2:1~2。
在一个实施方案中,发酵所用的菌为鼠李糖乳杆菌、枯草芽孢杆菌、乳酸杆菌的复合菌,重量比为1~2:1~2:1~2。
在一个实施方案中,青蒿、艾叶、生姜、银杏叶、桂花、柠檬、刺梨、金银花、鱼腥草、蒲公英的重量比为2~8:0~2:6~10:1~3:1~3:8~12:8~12:2~5:2~5:1~3。
优选的,青蒿、艾叶、生姜、银杏叶、桂花、柠檬、刺梨、金银花、鱼腥草、蒲公英的重量比为2~4:0.1~0.5:6~8:1~2:1~2:8~10:8~10:2~3:2~3:1~2。
在一个实施方案中,所述抑菌原液通过以下方法制备:
将青蒿、生姜、银杏叶、桂花、柠檬、刺梨、金银花、鱼腥草、蒲公英和任选的艾叶分别粉碎至12~65目并过筛,按比例称重并混匀,添加温水软化1~24小时;随后向其中添加复合酶,并补水至水的重量为原料总量的5~15倍,搅拌均匀,在37±1℃的条件下酶解5~10天得到酶解液;向该酶解液中加入复合菌,在37±1℃的条件下进行发酵,发酵1~3个月后,通过压滤设备将发酵基质分离出来,然后过滤收集滤液,滤液转移到熟化熬合罐进行熟化陈酿,得到发酵原液。
优选的,复合酶的用量为原料总量的10~20%,优选为13~18%。
优选的,复合菌的用量为原料总量的10~20%,优选为13~18%。
优选的,水的用量为原料总量的6~10倍。
优选的,熬合时间在半年以上。
在一个实施方案中,抑菌原液为30~40份。
在一个实施方案中,乙醇为5~6份。本发明中,添加的乙醇既可以解决油水分离问题溶解性问题,又使得抑菌液在喷雾使用时喷雾形态更佳。
在一个实施方案中,薄荷油为0.03~0.04份。本发明中,添加的薄荷油在作用于口腔后能够产生清凉感,提高使用口感。
在一个实施方案中,所述香精选自香兰素香精、葡萄香精、柠檬香精中的至少一种,优选为香兰素香精。优选的,其为0.5~0.8份。
在一个实施方案中,所述甜菊糖苷为0.01~0.3份。在本发明的青蒿口腔抑菌液中引入甜菊糖苷能够进一步提高抑菌液的口感。
在一个实施方案中,水为50~65份。
在一个实施方案中,所述青蒿口腔抑菌液还包括pH调节剂,其选自氢氧化钠、碳酸钠的至少一种。通过添加pH调节剂,将青蒿口腔抑菌液的pH调节至5.5~8.0,优选调节至6.0~7.0。
在一个实施方案中,所述青蒿口腔抑菌液为漱口水或口腔喷雾。
本发明的另一方面还提供了所述青蒿口腔抑菌液的制备方法,包括:
按比例称取抑菌原液、乙醇、薄荷油、香精和任选的甜菊糖苷,将薄荷油溶于乙醇中,将抑菌原液、香精、任选的甜菊糖苷与水于60~70℃下混匀,加入pH调节剂调节pH至5.5~8,转至灭菌罐升温70~95℃灭菌,然后转移到混料罐降温至40~50℃,加入薄荷油溶于乙醇的溶液,搅拌均匀得到青蒿口腔护理液。
本发明中,pH调节剂可以直接加入,也可以先溶于水中再加入。
在一个实施方案中,加入pH调节剂调节pH至6.0~7.0。
在一个实施方案中,所述方法还包括:
将青蒿、生姜、银杏叶、桂花、柠檬、刺梨、金银花、鱼腥草、蒲公英和任选的艾叶分别粉碎至12~65目并过筛,按比例称重并混匀,添加温水软化1~24小时;随后向其中添加复合酶,并补水至水的重量为原料总量的5~15倍,搅拌均匀,在37±1℃的条件下酶解5~10天得到酶解液;向该酶解液中加入复合菌,在37±1℃的条件下进行发酵,发酵1~3个月后,通过压滤设备将发酵基质分离出来,然后过滤收集滤液,滤液转移到熟化熬合罐进行熟化陈酿,熬合时间在半年以上,得到发酵原液。
下面将结合本发明实施例的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明的保护范围。
下面实施例中的实验方法,如无特殊说明,均为常规方法。实施例中未注明具体技术或条件者,按照本领域内的文献所描述的技术或条件,或者按照产品说明书进行。
制备例:发酵液的制备
步骤1)酶解
将青蒿、艾叶、生姜、银杏叶、桂花、柠檬、刺梨、金银花、鱼腥草、蒲公英分别粉碎至12~65目并过筛,取青蒿24.75份、艾叶3.50份、生姜60.00份、银杏叶10.50份、桂花17.50份、柠檬85.00份、刺梨87.50份、金银花27.50份、鱼腥草24.50份、蒲公英10.00份混匀,添加温水2000份软化20小时;随后向其中添加复合酶(见表1)50.00份,并补水至总体系重量为3350份,搅拌均匀,在37±1℃的条件下酶解7天得到酶解液。
步骤2)发酵
向步骤1)的酶解液中加入复合菌(见表1)50.00份,在37±1℃的条件下进行发酵,发酵2个月后,通过压滤设备将发酵基质分离出来,然后过滤收集滤液,滤液转移到熟化熬合罐进行熟化陈酿,熬合时间在半年以上,得到发酵原液。

对比制备例8:发酵液的制备
将青蒿、艾叶、生姜、银杏叶、桂花、柠檬、刺梨、金银花、鱼腥草、蒲公英分别粉碎至12~65目并过筛,取青蒿24.75份、艾叶3.50份、生姜60.00份、银杏叶10.50份、桂花17.50份、柠檬85.00份、刺梨87.50份、金银花27.50份、鱼腥草24.50份、蒲公英10.00份混匀,添加温水2000份软化20小时;随后向其中添加鼠李糖乳杆菌:枯草芽孢杆菌:乳酸杆菌重量比为1:1:1的复合酶50.00份,并补水至总体系重量为3350份,搅拌均匀,在37±1℃的条件下发酵3个月后,通过压滤设备将发酵基质分离出来,然后过滤收集滤液,滤液转移到熟化熬合罐进行熟化陈酿,熬合时间在半年以上,得到发酵原液。其中可溶性固形物含量2.4%,总酸含量13.6g/kg。
对比制备例9:提取液的制备
将青蒿、艾叶、生姜、银杏叶、桂花、柠檬、刺梨、金银花、鱼腥草、蒲公英分别粉碎至12~65目并过筛,取青蒿24.75份、艾叶3.50份、生姜60.00份、银杏叶10.50份、桂花17.50份、柠檬85.00份、刺梨87.50份、金银花27.50份、鱼腥草24.50份、蒲公英10.00份混匀,用1000份×3的水水煮提取,合并提取液,其中可溶性固形物含量4.9%。
实施例1-7和对比例1-9:青蒿口腔护理液的制备
将各制备例和对比制备例的发酵原液/提取液经硅藻土过滤,取350.75份,向其中加入香兰素香精5份、水573.75份,于60℃混合均匀,然后加入3.35份的氢氧化钠于16.75份水中的溶液,调节pH至6.0~6.5,转至灭菌罐升温90℃灭菌1h,然后转移到混料罐降温至45℃,加入0.4份的薄荷油预溶于50.00份乙醇的溶液,搅拌均匀得到青蒿口腔护理液,灌装。
试验例1:抑菌试验
1.1制备抑菌片取灭菌并干燥的直径为5mm的滤纸片。每片滴加实施例1制备的青蒿口腔抑菌液20μl,将滤纸片铺于无菌平皿内,半密封状态置干燥箱(37℃)中烘干,备用。
1.2制备阴性对照样片同1.1项,取灭菌处理并且干燥后的滤纸片,每张滤纸片上滴加无菌蒸馏水20μl,干燥后备用。
1.3接种试验菌用涂布工具,酒精灯火焰灭菌并冷却后,蘸取浓度为5×105~5×106cfu/ml试验菌悬液,在营养琼脂培养基平板表面均匀涂布。盖好平皿,室温下干燥5min。
1.4抑菌剂样片贴放用无菌镊子取样片贴放于平板表面。贴放好后,用无菌镊子轻压样片,使其紧贴于平板表面。每个平皿贴放5张样片,其中4张抑菌片、1张阴性对照样片,使各样片中心之间相距25mm以上,与平板的周缘相距15mm以上,盖好平皿,置37℃恒温培养箱中,培养16~18小时观察结果。用游标卡尺测量抑菌环的直径(包括贴片)并记录,每个平板上的4个抑菌圈大小取平均值作为该平板的读数;试验平行三次,取其平均值作为试验结果记录。
1.5评价规定抑菌环直径大于7mm者,判为有抑菌作用。抑菌环直径小于或等于7mm者,判为无抑菌作用。3次重复试验均有抑菌作用结果者,判为合格。阴性对照组应无抑菌环产生。否则试验无效。
试验结果如表2所示:

试验结果显示,本发明实施例1-7的青蒿口腔抑菌液具有十分优异的抑菌活性,其对包括金黄色葡萄球菌、大肠埃希菌、白色念珠菌、牙龈卟啉单胞菌在内的多种病菌均有很强的抑制活性;而对比例1-8由于酵解、发酵过程存在不足而导致抑菌液的抑菌活性下降较多;对比例9采用提取液而非发酵液,同样存在抑菌活性明显不足的问题。
试验例2:急性眼刺激试验
参照《消毒产品卫生安全评价规定》,用于黏膜的抗(抑)菌制剂应当进行眼刺激性试验。
2.1实验动物准备试验前24小时内检查每只新西兰兔的双眼是否有异常现象,发现有异常现象则淘汰。
2.2染毒方法采用同体左右侧自身对比法,取受试物原液0.1ml滴入左侧眼结膜囊内,每组3只,将眼睛被动闭合4s,30s后用氯化钠注射液冲洗。右侧眼睛同法给予氯化钠注射液0.1ml作为阴性对照组。
2.3结果记录滴受试物后,将眼被动闭合4s,30s后用生理盐水冲洗。分别在24h、48h、72小时肉眼观察新西兰兔眼结膜、虹膜和角膜的损伤与恢复情况并记录。如有刺激反应情况下,可继续观察记录7天、14天和21天情况;如果72小时内未出现刺激反应,或第7天或第14天,眼睛刺激反应完全恢复,即可提前终止试验。
2.4评价规定按《消毒技术规范》(2002年版)表2-13对家兔眼角膜、虹膜和结膜的急性刺激反应进行评分,并分别计算每只动物在3个不同观察时间(24,48和72h)角膜损害、虹膜损害、结膜充血和结膜水肿四方面的“平均评分”。分别以动物眼角膜、虹膜和结膜充血、水肿的平均评分和恢复时间进行,按《消毒技术规范》(2002年版)表2-14、表2-15眼刺激反应分级标准判定受试物对眼睛的刺激强度。各组均取平均值。
结果如表3所示:

试验结果显示,本发明实施例的青蒿口腔抑菌液对眼睛没有刺激作用,安全性高;而采用提取液而非发酵液的对比例9对眼睛存在一定程度的刺激。
以上描述了本发明优选实施方式,然其并非用以限定本发明。本领域技术人员对在此公开的实施方案可进行并不偏离本发明范畴和精神的改进和变化。
Claims (10)
1.一种青蒿口腔抑菌液包括:
抑菌原液20~40份
乙醇5~8份
薄荷油0.03~0.05份
香精0.3~1份
甜菊糖苷0~1份
水30~65
所述抑菌原液由青蒿、艾叶、生姜、银杏叶、桂花、柠檬、刺梨、金银花、鱼腥草、蒲公英经酶解、发酵、熬合制得。
2.根据权利要求1所述的抑菌液,其特征在于,酶解所用的酶为纤维素酶、脂肪酶、中性蛋白酶、淀粉酶的复合酶,重量比为1~2:1~2:1~2:1~2;发酵所用的菌为鼠李糖乳杆菌、枯草芽孢杆菌、乳酸杆菌的复合菌,重量比为1~2:1~2:1~2。
3.根据权利要求2或3所述的抑菌液,其特征在于,青蒿、艾叶、生姜、银杏叶、桂花、柠檬、刺梨、金银花、鱼腥草、蒲公英的重量比为2~8:0~2:6~10:1~3:1~3:8~12:8~12:2~5:2~5:1~3。
4.根据权利要求3所述的抑菌液,其特征在于,所述抑菌原液通过以下方法制备:
将青蒿、生姜、银杏叶、桂花、柠檬、刺梨、金银花、鱼腥草、蒲公英和任选的艾叶分别粉碎至12~65目并过筛,按比例称重并混匀,添加温水软化1~24小时;随后向其中添加复合酶,并补水至水的重量为原料总量的5~15倍,搅拌均匀,在37±1℃的条件下酶解5~10天得到酶解液;向该酶解液中加入复合菌,在37±1℃的条件下进行发酵,发酵1~3个月后,通过压滤设备将发酵基质分离出来,然后过滤收集滤液,滤液转移到熟化熬合罐进行熟化陈酿,熬合时间在半年以上,得到抑菌原液。
5.根据权利要求4所述的抑菌液,其特征在于,复合酶的用量为原料总量的10~20%,优选为13~18%;复合菌的用量为原料总量的10~20%,优选为13~18%。
6.根据权利要求1所述的抑菌液,其特征在于,抑菌原液为30~40份;乙醇为5~6份;薄荷油为0.03~0.04份;所述香精选自香兰素香精、葡萄香精、柠檬香精中的至少一种,其为0.5~0.8份;水为50~65份。
7.根据权利要求3所述的抑菌液,其特征在于,青蒿、艾叶、生姜、银杏叶、桂花、柠檬、刺梨、金银花、鱼腥草、蒲公英的重量比为2~4:0.1~0.5:6~8:1~2:1~2:8~10:8~10:2~3:2~3:1~2。
8.根据权利要求1-7任一项所述的抑菌液,其特征在于,所述青蒿口腔抑菌液还包括pH调节剂,其选自氢氧化钠、碳酸钠的至少一种。
9.根据权利要求8所述的青蒿口腔抑菌液的制备方法,包括:
按比例称取抑菌原液、乙醇、薄荷油、香精,将薄荷油溶于乙醇中,将抑菌原液、香精与水于60~70℃下混匀,加入pH调节剂调节pH至5.5~8,转至灭菌罐升温70~95℃灭菌,然后转移到混料罐降温至40~50℃,加入薄荷油溶于乙醇的溶液,搅拌均匀得到青蒿口腔护理液。
10.根据权利要求9所述的制备方法,其特征在于,所述方法还包括:
将青蒿、生姜、银杏叶、桂花、柠檬、刺梨、金银花、鱼腥草、蒲公英和任选的艾叶分别粉碎至12~65目并过筛,按比例称重并混匀,添加温水软化1~24小时;随后向其中添加复合酶,并补水至水的重量为原料总量的5~15倍,搅拌均匀,在37±1℃的条件下酶解5~10天得到酶解液;向该酶解液中加入复合菌,在37±1℃的条件下进行发酵,发酵1~3个月后,通过压滤设备将发酵基质分离出来,然后过滤收集滤液,滤液转移到熟化熬合罐进行熟化陈酿,得到抑菌原液。
