Description
The invention relates to a film-forming dispersion for forming a protective layer on shaped articles, such as tablets, pellets, granules or their constituents in crystalline form, in particular β containing one or more active substances. The invention also relates to a process for the production of this film-forming dispersion.
As is well known, various medicaments formulated for oral administration are often sensitive to physical and / or chemical environmental influences such as oxygen, light, moisture, or tastiness. It also often happens that in one dosage form chemical reactions occur between two or more active substances or between an active substance and excipients. Therefore, it is necessary to protect either the finished molded article or some or all of its active ingredients by suitable deposition, i.e. by forming protective coatings (layers) with various film-forming substances, most often applied in the form of a solution.
Historically, the oldest type of induction is the auction. In this process, a thick layer of sucrose or other excipients including dyes is formed on the pressed cores.
The sugar syrup is used for the dragee coating, which is gradually poured over the cores placed in the draining cauldron with constant rotation; by introducing warm air, the water from the syrup evaporates and a compact layer is formed on the surface of the cores whose weight reaches or exceeds 10055 the weight of the core. Auctioning is a long-term process that lasts 24 to 32 hours or more. However, sugar syrup is not a film-forming solution in the strict sense.
Further encapsulated tablets are so-called coated tablets, which are basically cores coated with a thin layer of cellulose derivative or other suitable polymeric material. The most commonly used cellulose derivatives are hydroxypropylmethylcellulose or ethylcellulose, 229045 from other polymers, acrylates and methacrylates. A film-forming solution based on organic solvents or mixtures thereof is used for coating. The lining by the described technology is very well elaborated and there are available very powerful devices using fluid technology.
A common disadvantage of the preparation of the encapsulated tablets by the prior art methods is, in particular, that the film-forming solutions contain organic solvents or mixtures thereof. Usually a mixture of ethanol, isopropanol or acetone with chlorinated alkane (dichloromethane, chloroform) is used. The precondition for the coating is the evaporation of the solvent whose vapors escape into the atmosphere. Working with organic solvents is unpleasant both in terms of the protection of workers' health and in terms of fire protection. In addition to air pollution, the economic aspect of operation is not negligible: solvent vapors are diluted with large amounts of air, so they cannot be economically regenerated by available technical means. A further disadvantage is that it is not possible to use water-soluble dyes in these film-forming solutions and therefore only pigments need to be used.
Is therefore. The general aim is to eliminate film-forming solutions based on organic solvents from the technology of the impregnated products and replace them with aqueous solutions or dispersions. To this end, some water-soluble cellulose derivatives have been tested; however, they provide very viscous solutions which are difficult to apply. Commercially available for the purpose of induction are some special types of hydroxyethyl cellulose which provide aqueous solutions of low viscosity and which are useful for forming protective layers. In addition, aqueous dispersions of acrylate polymers, known under various commercial names, or ethyl cellulose dispersions are available.
However, even these aqueous film-forming solutions or dispersions are not entirely without disadvantages. Water is difficult to escape from the forming film, the cores have been in contact with water for quite a long time, and at high temperatures and non-continuous aeration, especially when working in the buoyancy, degradation of the active substances can occur.
In a systematic study of the various available water-soluble substances to be considered, in the formation of a protective film, in particular cellulose derivatives, it has been found that the hydroxyethylcellulose sodium provides a high-quality film from an aqueous solution which excels in low water retention and thus dries very quickly. However, difficulty in coring the cores with an aqueous solution of the substance, both when working on conventional boiling boilers and in fluid power: the cores ee strongly bonded and the coating formed was very irregular. Thus, it was necessary to look for various additives that would reduce the sticking of the cores, ensure their sliding and improve the uniformity of the film, the desired goal was achieved by adding polyethylene glycol and calcium or magnesium stearate to the film-forming solution.
It has been found that polyethylene glycol functions as a plasticizer and polish, and calcium and magnesium stearate as a smoothing agent. Furthermore, it has been found that in the preparation of the film-forming solution, it is appropriate to disperse the base film-forming agent, i.e., the hydroxyethylcellulose sodium salt, first in a small amount of a lower alcohol and then to dissolve it by adding water. This speeds up dissolution and a small amount of alcohol ensures more uniform film formation on the cores.
It is preferred that the polyethylene glycol be dissolved in the water beforehand. Additional excipients such as fillers, dispersion stabilizers, dyes or pigments, flavoring agents, preservatives, etc. may be added to the film-forming solution as desired.
The foregoing non-film dispersions do not have a film-forming dispersion for forming a protective layer on molded articles such as tablets, pellets, granules or their constituents in crystalline form, in particular and containing one or more active ingredients according to the invention, which comprises 10% by weight, preferably 2 to 4% by weight, of a water-soluble cellulose derivative, in particular an alkali metal salt of a hydroxyalkyl cellulose derivative, preferably a hydroxyethylcellulose sodium salt, further a polyethylene glycol having a molecular weight of 1,000 to 20,000, preferably 4,000 to 6,000 From 5 to 20% by weight, preferably from 8 to 12% by weight of cellulose, based on the weight of the cellulose derivative used, an alkali metal or aluminum alkanoic acid salt of 14 to 18 carbon atoms, preferably magnesium or calcium stearate, in an amount of 1 to 4 % by weight, based on the weight of the cellulose derivative used, and 4 to 16% by weight, preferably From 6 to 10% by weight of a C 1 -C 3 alkanol, preferably ethanol, the balance being up to 100% by weight of water, optionally with the addition of a filler and / or diluent and / or stabilizer of the dispersion and / or colorant C1 of pigment and / or flavorings and / or preservatives.
The invention also relates to a process for the production of a film-forming dispersion for the formation of a protective layer on shaped articles, which comprises the first step of dispersing a water-soluble cellulose derivative, in particular an alkali metal hydroxyalkyl cellulose derivative, and preferably hydroxyethylcellulose sodium. A solution of polyethylene glycol in water is added in the form of a C 1 to C 3 atom, whereupon an alkali earth metal or aluminum salt of C 14 to C 18 alkanoic acid is dispersed in the resulting solution, optionally with filler and / or dispersion stabilizer and / or colorant or pigment. or flavorings and / or preservatives.
In the production of the film-forming dispersion according to the invention, the most common procedure is to first disperse the sodium salt of hydroxyethylcellulose (or another useful derivative) in a small amount of ethanol or isopropanol and add a solution of polyethylene glycol in water to the resulting dispersion. After complete dissolution, magnesium or calcium stearate is added to the solution, optionally and further with additives such as talc, colloidal silica, soluble or pigmented colorant, flavor, preservative, and the like, and processed to a homogeneous dispersion which is ready for further processing. use, i.e.
This operation is carried out either on conventional rotary ellipsoid-shaped panning boilers, on large-capacity boilers or in fluidized bed equipment. The film-forming dispersion according to the invention is applied in an amount such that about 3% by weight of the salt of the hydroxyalkylcaluloay derivative is on the total weight of the feed of the induced material. By means of the film-forming dispersion according to the invention, it is possible to form a protective layer on the surface of cores, granules, crystals or pellets, which protects the material from external influences and from mutual interactions of individual components. The properties of this layer, which can also be suitably colored, can be varied, if necessary, by suitable auxiliaries.
The film-forming dispersion according to the invention eliminates the disadvantages of the compositions used so far: it is prepared at normal temperature, contains only a small amount of organic solvents, the active substances are not exposed to moisture for a long time because the film is very fast. The amount of alcohol used is insignificant from the fire, health and economic point of view. The film-forming dispersion according to the invention can be prepared either freshly, in a product-optimized composition or in advance, with the addition of the desired additive, for example a colorant, pigment or aroma, added as necessary to the base dispersion with thorough homogenization.
It goes without saying that the use of the film-forming dispersion according to the invention is not limited to drugs. It can also be used in food, chemical and other production, generally wherever it is advisable to protect molded products or their parts from environmental influences.
