Látky vzorce I jsou meziprodukty přípravy farmakodynamicky účinných látek, jejich hydrochloridy při nízké toxicitě mají inhibiční účinnost vůči celé řadě pathogenních i nepathogenních mikroorganismů a methojodidy mají myorelaxační účinnost. The compounds of formula (I) are intermediates for the preparation of pharmacodynamically active substances, their hydrochlorides at low toxicity have inhibitory activity against a variety of pathogenic and non-pathogenic microorganisms, and methoiodides have myorelaxant activity.

Vynález se týká způsobu přípravy nových 2-(4-alkylsulfonylfenyl)-2-hydroxy-4-(2-hydroxyethyljmorfolinů obecného vzorce IThe present invention relates to a process for the preparation of novel 2- (4-alkylsulfonylphenyl) -2-hydroxy-4- (2-hydroxyethyl) morpholines of the formula I

ve kterém R značí alkyl s 1 až 4 uhlíkovými atomy a s rovně probíhajícím nebo rozvětveným řetězcem jejich hydrochloridů a methojodidů.wherein R is C 1 -C 4 alkyl and straight or branched chain of their hydrochlorides and methoiodides.

Látky obecného vzorce I jsou meziprodukty přípravy farmakodynamicky účinných látek, tj. potenciálních léčiv. Jejich hydrochloridy jsou jen málo jedovaté (LDso 400 až 800 mg/kg při intravenosním podání J a samy o sobě mají v koncentracích 50 až 100 ,ug/ml inhibiční účinnost vůči celé řadě pathogenních i nepathogenních mikroorganismů, zejména vůči Streptococcus jS-haemolyticus, Staphylococcus pyogenes aureus, Pseudomonas aeruginosa, Próteus vulgaris, Mycobacterium tuberculosis, Saccharomyses pasterianus, Trichophyton mantagrophytes. Relativně toxický methojodid látky I, R = CH2CH(CH3)2 (LDso = 12,5 mg/kg i. v.) vykazuje myorelaxační účinnost na krysím musc. gastrocnemius. The compounds of formula (I) are intermediates in the preparation of pharmacodynamically active substances, i.e. potential drugs. Their hydrochlorides are of low toxicity (LD 50 of 400 to 800 mg / kg by intravenous administration of J) and in themselves have concentrations of 50 to 100 µg / ml inhibiting activity against a variety of pathogenic and non-pathogenic microorganisms, in particular Streptococcus jS-haemolyticus, Staphylococcus pyogenes aureus, Pseudomonas aeruginosa, Proteus vulgaris, Mycobacterium tuberculosis, Saccharomyses pasterianus, Trichophyton mantagrophytes, Relatively toxic methoxide of compound I, R = CH 2 CH (CH 3) 2 (LD 50 = 12.5 mg / kg iv) exhibits myrocnemius activity in rats. .

The process for the preparation of the compounds of the formula I according to the invention consists in reacting 4- (alkylsulfonyl) phenacylbromides of the formula II (ti) in which R is the same as in formula I with diethanolamine in boiling chloroform. they are formed instead of the expected N-substituted diethanolamines.

Chem. Soc. 71, 993, 1949; Lutz R. E., Jordán R. H.: J. Amer. Chem. Soc. 71, 996, 1949).However, a reaction of this type has already been observed in the reactions of N-substituted ethanolamines with phenacyl bromide (Cromwell NH, Tsou K.-C .: J. Amer. Chem. Soc. 71, 993, 1949; Lutz RE, Jordan RH: J. Amer. Chem. Soc., 71, 996 (1949).

V IČ spektrech lze rozlišit pásy příslušející primární a terciární alkoholické skupině a dále pás etherického kyslíku, vázaného na dva uhlíkové atomy. The compounds of the formula I according to the invention have been found to lack the conjugated keto group of the starting compounds of the formula II by means of ultraviolet and infrared spectra. In the IR spectra, bands belonging to the primary and tertiary alcohol groups can be distinguished, as well as a band of etheric oxygen bonded to two carbon atoms.

Accordingly, the structure of the hemiacetals of formula I could be attributed to the reaction products. Conversion to the hydrochlorides by treatment with hydrochloric acid or hydrochloric acid in the presence of water as part of the process of the invention does not lead to hydrolysis of the hemiacetal ring. .

I.Thus, the hemiacetals of the formula I are highly stable to acid hydrolysis and their hydrochlorides have the structure of the appropriately protonated derivatives of the formula I. Also the reaction of the bases of the formula I with the methyl iodide prepared by the methoiodides which are part of the invention are derived from the structure I.

The following examples are intended to illustrate some of the possibilities for preparing the compounds of formula (I) of the present invention, but are not intended to exhaustively describe all of them. It is by no means intended to limit the invention to the conditions described in the examples.
