Description
Vynález se týká způsobu výroby nitrilu kyseliny cyklohexylfenyloctové vzorce I C6H5 ^CHCN (I) který představuje významný meziprodukt při výrobě biologicky účinných látek (Kleemann A.,The invention relates to a process for the preparation of cyclohexylphenylacetic acid nitrile of the formula I C 6 H 5 · CHCN (I) which represents an important intermediate in the production of biologically active substances (Kleemann A.,
Engel J. v knize Pharmazeutische Wirkstoffe, Synthesen, Patente, Anwendungen, Thieme Verlag Stuttgart, 1978) např. spasmolytika methojodidu methylthioethylcyklohexylfenylacetátu (M. Perles, skriptum Výroba léčiv, V3CHT - SNTL Praha, 1981) . Nitril vzorce I se podle známého stavu techniky vyrábí ze sodné soli nitrilu kyseliny fenyloctové vzorce IIEngel J. in Pharmazeutische Wirkstoffe, Synthesen, Patente, Anwendungen, Thieme Verlag Stuttgart, 1978), e.g. The nitrile of the formula I is prepared according to the prior art from the sodium salt of the phenylacetic acid nitrile of the formula II
CgH5CH2CH (II) získané působením silných bázických činidel, reakcí s cyklohexylbromidem. Ve většině případů se jako bázlcké činidlo používá natrium amid v různých prostředích jako kapalný amoniak-toluen, ether-benzen, toluen, xylen (J. Amer, Chem. Soc. 65, 1999 (1943), Z. obšč. chim. 7, 2823 (1937); Cg H 5 CH 2 CH (II) obtained by treatment with strong basic agents, by reaction with cyclohexyl bromide . In most cases, sodium amide in various media such as liquid ammonia-toluene, ether-benzene, toluene, xylene (J. Amer, Chem. Soc. 65, 1999 (1943), Z. ob. Chim. 2823 (1937);
J. Chem. Soc. 1949, 3156, Org. Synth. Coll. Vol III, 219 (1955), US pat. spisy 2 265 284).J. Chem. Soc. 1949, 3156, Org. Synth. Coll. Vol III, 219 (1955), US Pat. documents 2 265 284).
Tyto způsoby nenalezly významného technického využití z hlediska nákladnosti a bezpečnosti a hygieny práce. Další způsob AO 200 438 chrání přípravu sloučeniny vzorce I ze sloučeniny vzorce II, cyklohexylbromidu, hydroxidu alkalického kovu a katalyzátoru fázového přenosu na bázi kvartérní amoniové soli v prostředí aprotického dipolárního rozpouštědla, přičemž podle AO 202 448 je možné uvedenou reakci provést se stejným výsledkem bez přítomnosti katalyzátoru.These methods have not found significant technical applications in terms of cost and safety and occupational hygiene. Another method of AO 200 438 protects the preparation of a compound of formula I from a compound of formula II, cyclohexyl bromide, alkali metal hydroxide and a quaternary ammonium salt phase transfer catalyst in an aprotic dipolar solvent environment, according to AO 202 448 of the catalyst.
Metoda (Bull. Chem. Soc. Jan 56, 3306 (1983)), přípravy sloučeniny I, která provádí uvedenou alkylaci v benzenu za přítomnosti oxidu hlinitého a hydroxidu sodného nebo draselného má spíše teoretický význam'.According to the aforementioned AO 200 438 and 202 448, an oily product with a boiling point in a wide range is obtained, which can reasonably be assumed to be not a pure compound of formula I. In addition, the use of relatively expensive solvents associated with difficulties in waste water treatment economically disadvantageous. The process of classical two-phase catalysis carried out in a water-aromatic hydrocarbon such as toluene, xylene and tetra-n-butylammonium hydrogen sulphate or methyltrioctylammonium chloride as a phase transfer catalyst has resulted in considerable simplification, higher yields and purity of the product. One of its disadvantages is the cost of the phase transfer catalyst and its regeneration from wastewater. The method (Bull. Chem. Soc. Jan 56, 3306 (1983)) for the preparation of compound I which performs said alkylation in benzene in the presence of alumina and sodium or potassium hydroxide is of theoretical importance.
I.These known processes are followed in a positive manner by the process according to the invention. The process according to the invention consists in treating the nitrilophenylootic acid of the formula II with a mixture of sodium hydride and hydroxide and cyclohexyl bromide or cyclohexyl chloride in a hydrocarbon medium at a temperature of 100 ° C to the boiling point of the solvent. 1.5: 1.05-1.2, after which the insoluble material is suctioned off after completion of the reaction and, after evaporation of the solvents under reduced pressure, the product of formula I is obtained.
As the solvent, aromatic hydrocarbons such as benzene, toluene, xylene are preferred. The use of a hydride-hydroxide mixture containing 18 to 25% sodium hydride has the advantage that it is not self-igniting and can be handled under conventional safety conditions such as sodium hydroxide.
According to the process of the invention, a mixture of hydride and sodium hydroxide is added in portions to the mixture of nitrile of formula II, cyclohexyl bromide or chloride in a preferably aromatic hydrocarbon solution at a reaction temperature of 100 ° C to the boiling point of the solvent. Alternatively, the sodium nitrile II is first prepared by treatment with the above mixture and then the cyclohexyl halide is added. The progress of the reaction can be monitored by chromatographic methods, with the gas chromatography method being particularly preferred.
V případě vyreagováni cyklohexylhalogenidu se přidá do reakční směsi další množství, až se v reakční směsi spotřebuje veškerý nitril vzorce II, přičemž konečný molární poměr reakčnich složek nitril vzorce II : cyklohexylhalogenid : směs hydrid-hydroxid sodný je 1:1,1 až 1,5:1,05 až 1,2.If the cyclohexyl halide is reacted, an additional amount is added to the reaction mixture until all of the nitrile of formula II has been consumed, the final molar ratio of the reactants of the nitrile of formula II: cyclohexyl halide: sodium hydride-hydroxide being 1: 1.1-1.5 : 1.05 to 1.2.
Po ukončení reakce většinou za 1,5 až 4 hodiny se nerozpustné podíly odfiltrují, roztok odbarví aktivním uhlím, rozpouštědla oddestilují za sníženého tlaku a získá se produkt ve výtěžku 60 až 70 % hmot., který podle plynově chromatografické analýzy obsahuje nad 95 i hmot. sloučeniny vzorce I.After completion of the reaction for about 1.5 to 4 hours, insoluble materials are filtered off, the solution is decolorized with activated carbon, the solvents are distilled off under reduced pressure, and the product is obtained in a yield of 60 to 70% by weight. compounds of formula I.
The advantage of the process according to the invention is the simple carrying out of the reaction, the use of inexpensive reagents and solvents, the minimum requirements for waste water treatment, while the regenerated solvents can be used in further repeated batches.
The process according to the invention is illustrated by several special examples which are merely illustrative and do not limit the scope of the invention in any way.
