Abstract
Plynná fáze s obsahem.ethanolu projde membránou a oběma vrstvami do prostoru detektoru kyslíku, kde v důsledku oxidace v předchozích vrstvách dojde k, poklesu vodivosti elektrolytu. Množství ethanolu v plynné fázi lze charakterizovaz stechiometrickou spotřebou kyslíku.The sensor is based on the enzymatic oxidation of ethanol and is used to measure the amount of alcohol in the breath of intoxicated persons. It consists of an oxygen detector on which is applied a layer permeable to oxygen, or another layer with an immobilized enzyme alcohol oxidase, and both layers are covered with a membrane permeable to oxygen and ethanol. The gas phase containing ethanol passes through the membrane and both layers into the oxygen detector space, where, as a result of oxidation in the previous layers, the conductivity of the electrolyte decreases. The amount of ethanol in the gas phase can be characterized by the stoichiometric oxygen consumption.
Description
The invention relates to an electrochemical sensor of ethanol in the gas phase ; based on the enzymatic oxidation of ethanol.
Analytické stanovení ethanolu v plynné směsi lze uskutečnit řadou způsobů. Jsou známy a ověřeny především spektroskopické a chromatografické metody jeho detekce v širokém koncentračním rozmezí, od několika setin až po desítky objemových procent·Analytical determination of ethanol in a gas mixture can be accomplished in a number of ways. Especially spectroscopic and chromatographic methods of its detection in a wide concentration range from several hundredths to tens of volume percent are known and verified.
The disadvantages of these methods are their complexity and the fact that they are not suitable for field use as portable devices. Known foreign products use an inorganic semiconductor element detector as a detector sensor. A disadvantage, however, is the sensitivity not only to ethanol but also to acetone and to other organic vapors and gases such as ethylene or methane.
Enzyme analysis appears to be a relatively new way of indicating and, above all, a new way of quantifying many organic substances. The essence of these, in analytical perspective processes, is the fact that many interesting substances can be oxidized or possibly subject to other substance changes under normal conditions only in the presence of the corresponding enzyme. In fact, it is a catalytic conversion that the enzyme merely mediates without changing itself; moreover, it is a case of a highly specific reaction, that is, one type of enzyme affects only one type of substance of interest.
♦♦
V případě enzymové oxidace^ katalyzované oxidázami obsahujícími flavinový kofaktor, lze sledovat látkovou bilanci spotřebovaného kyslíku. Při aplikaci vhodného enzymu, kdy dochází k enzymové oxidaci etahnolu, lze množství etahnolu v plynné fázi charakterizovat steehiometrickou spotřebou kyslíku.In the case of enzymatic oxidation catalysed by flavin cofactor-containing oxidases, the mass balance of oxygen consumed can be monitored. When a suitable enzyme is applied, in which the enzymatic oxidation of ethanol is occurring, the amount of ethanol in the gas phase can be characterized by steehiometric oxygen consumption.

This principle is based on the electrochemical ethanol sensor formed by the oxygen detector according to the invention, which consists in that the oxygen detector is coated with an oxygen and ethanol permeable layer and immobilized in it by the enzyme alcohol oxidase. its surface is immobilized by the enzyme alcohol oxidase and the treated layer is covered with a membrane permeable to ethanol and oxygen. According to another variant of embodiment, another layer with the immobilized enzyme alcohol oxidase is placed between the layer and the membrane.
Elektrochemické čidlo podle vynálezu umožňuje rychle a spolehli vě měřit například množství alkoholu v dechu Intoxikováného probanta v rozsahu jednotek promile, případně až desetin promile. The electrochemical sensor according to the invention makes it possible to measure quickly and reliably, for example, the amount of alcohol in the breath of an Intoxicated Probant in the range of units per mille or up to tenths per mille.
An exemplary arrangement of an electrochemical ethanol sensor according to the invention is schematically shown in the accompanying drawing. The electrochemical ethanol sensor consists of an oxygen detector on which oxygen permeable layer 2 is applied. Layer 2 is deposited another layer of 4 8 immobilized enzyme alcohol oxidase. The layer 4 is covered by a membrane 2 permeable to ethanol and oxygen.
Plynná fáze s obsahem ethanolu přijde do styku s membránou prostoupí do další vrstvy 4 obsahující imobilizovaxý enzym alkoholoxidázu, kde se následkem oxidace sníží tok kyslíku vrstvou 2 do prostoru detektoru χ kyslíku, čímž poklesne vodivost elektrolytu, kterým je detektor χ kyslíku naplněn. Tímto způsobem je možno měřit množství alkoholu v dechu intoxikovaných osob. The ethanol-containing gas phase comes into contact with the membrane and passes to another layer 4 containing the immobilized enzyme alcohol oxidase, where, as a result of oxidation, the oxygen flow through the layer 2 to the oxygen detector space decreases, thereby reducing the electrolyte conductivity through which the oxygen detector is filled. In this way it is possible to measure the amount of alcohol in the breath of intoxicated persons.
