The subject of the solution is a device, especially for aerobic and anaerobic fermentations and bioconversions. The device comprises a reaction vessel with a hollow rotor rotating in the liquid phase and formed by a jacket, a bottom, or possibly provided with an extension, which has an inlet opening in the center of the bottom for the entry of the liquid phase into the rotor. A gas distributor is located inside the hollow rotor or in the inlet opening or in the extension of the hollow rotor, which may also have a separate axis of rotation. The apparatus comprises a reaction vessel with a hollow rotor rotating in the liquid phase and formed by a jacket, a bottom, optionally provided with an extension having an inlet opening in the center of the bottom for the inlet of the capillary phase into the rotor. Inside the hollow rotor or in the inlet or in the extension of the hollow rotor there is a gas distributor, which may also have a separate axis of rotation. If there is a solid phase cartridge or solid-state assembly inside the rotor, the gas distributor can also be located in these spaces inside the hollow rotor. A further advantage is that the hollow rotor can be provided with a feed or discharge tube for solid phase particles.
The invention relates in particular to apparatus for aerobic and anaerobic fermentations and bioconversions.
For the processing of flowing thin and very viscous media, particularly for the fermentation of microorganisms, but also for other chemical reactions in particular, for the purpose of mixing (homogenizing), spraying and pumping liquid upwards, a device consisting of a liquid medium container has been developed. a hollow rotor, for example a cylindrical or truncated cone with a driven shaft, is placed, either wholly or partially, in the liquid medium, the hollow rotor being formed by a jacket, a bottom or a lid.
(AO č. 228 221, AO č. 228 350 a další.]The bottom of the rotor is provided with at least one inlet opening for the liquid medium and there are outlet openings for the liquid to be processed, which exit from the rotor back into the vessel in the housing or cover of the hollow rotor. (AO No 228 221, AO No 228 350 and others.)
Toto zařízení bylo pak uzpůsobeno zejména pro biokatalytické reakce pomocí imobilizavných enzymů a buněk (AO č. 256 203).Later, the hollow rotor has been adapted to receive a solid phase, preferably a charge or built-in reagent particle, in the hollow rotor, which is filled with the particles and converted into a universal solid-liquid contact device. (AO No. 255 257.) This device was then adapted especially for biocatalytic reactions using immobilizing enzymes and cells (AO No. 256 203).
When rotating the rotor in a liquid medium (regardless of whether or not solid particulate solids are embedded in the rotor cavity), the liquid is sucked into the rotor through an inlet at the bottom of the rotor and forms a fluid spindle inside the rotor centering a surface liquid paraboloid. . Intense circulation of the liquid through the hollow rotor takes place, with the liquid exiting the rotor through the outlet openings at the top of the housing or the openings in the lid. If the rotor is not provided with these openings or a lid, it is sprayed freely over the upper edge of the rotor back into the liquids.
When the rotating rotor with the outlet openings or its free upper edge into the space above the surface of the liquid is sprayed, the exiting liquid is sprayed in this space, for example on the wall of the mixing vessel in the form of tiny droplets. liquid medium.
If a solid-phase cartridge or solid-phase installation is installed in the rotor, the liquid circulation in the rotor, in addition to the liquid mixing, also intensively contacts the solid phase with the liquid phase, which still maintains the shape of the liquid spindle inside the rotor.
Said known apparatus is not particularly suited for bioprocesses in which intensive contact of the liquid medium with an additional gaseous phase is required, e.g. air, oxygen, carbon dioxide, Nz, optionally other inert gases and the like.
Některé alternativy popsaného zařízení sice umožňují současně s mícháním i rozstřik kapalného, např. fermentačního média, do plynného, např. vzduchového prostoru nad hladinu média, ale např. pro růst některých prokaryontních a enkaryontních mikroorganismů nebo pro některé biokatalytické procesy se vyžaduje mnohem intenzivnější styk kapalné a plynné fáze.While some alternatives of the described apparatus allow for the spraying of a liquid, e.g. fermentation medium, into a gaseous, e.g. and gaseous phases.
This is the case, for example, in submerged ergot alkaloid fermentations, where it is necessary to simultaneously feed Ch or air into the bioprocess at a higher pressure or in a larger quantity than a mere spray of liquid medium above its surface could provide.
These disadvantages are overcome by the aerobic and anaerobic fermentation and bioconversion apparatus of the present invention comprising a vessel and a hollow rotor with a driven shaft inside the vessel, which has a gas distributor located inside the hollow rotor, in the inlet or in the hollow rotor extension.
The apparatus is adapted for both conventional fermentations where the presence of solid phase particles is not required, as well as bioreaction where, in addition to the liquid and gaseous phases, the presence of solid phase particles is also required in the form of a pack or embedded. In this case, the gas distributor may be placed directly in the solid phase charge or in a built-in solid or perforated lid with a solid phase charge which is housed inside the hollow rotor.
Vzhledem k otáčejícímu se rotoru může se pak distributoi plynu nebo' jeho hlavaA further feature of the device according to the invention is that the gas distributor can be provided with a separate axis of rotation. Due to the rotating rotor, the gas or its head can then be distributed
Rotate in the same direction as the rotor, but at a different speed (faster or slower, especially if it is freely rotated by the liquid spindle formed in the rotor).
2. otáčet v opačném směru než rotor libovolnou rychlostí nebo2. Rotate in the opposite direction to the rotor at any speed; or
3. být v relativním klidu vůči otáčejícímu se rotoru (je-li např. pevně spojen s reakční nádobou, např. fermentační).3. be relatively at rest with the rotating rotor (if, for example, it is firmly attached to a reaction vessel, eg, a fermentation vessel).
The advantage is that in this way it is possible to model a number of operating modes in contact of the liquid and solid phase or all three phases, ie liquid, gaseous and solid. At the same time, the gas distributor, located eg inside the solid phase charge, assumes the function of a mechanical fluid mixer in the space of the charge inside the hollow rotor.
Tím dochází k další intenzifikaci bioprocesu, při kterém plyn, např. vzdušný kyslík, rozpouští se dokonaleji v kapalné fázi a proniká do částic pevné fáze, např. tvaru globulí, obsahujících zakotvené mikrobiální buňky nebo jejich částice, čímž se vy5This further intensifies the bioprocess, in which the gas, e.g. air oxygen, dissolves more fully in the liquid phase and penetrates into solid particles, e.g. globules, containing embedded microbial cells or particles thereof, thereby eliminating
253735 tváří aerobní podmínky, nezbytné pr© řadu fermentací, např. pří výrobě alkaloidů.253735 faces aerobic conditions, necessary for a number of fermentations, eg in the production of alkaloids.
The gas distributor consists of a head and a gas supply tube which is seen by a control valve. Tube: for gas supply, it is usually routed through a central inlet opening in the middle of the bottom of the hollow rotor, possibly by a hollow rotor extension.
1).For example, the distributor head may be movable vertically, and may also have a control function for entering the liquid phase into the hollow rotor (see FIG. 1 below).
The shape of the gas distributor head having the outlet openings to distribute the gas into the liquid phase may be varied, e.g., hollow ring shape, straight tube, perforated circular plate, hemispherical sieves etc. Similarly, the shape of the hollow body forming the rotor itself, its bottom, shell, lid, shape of rotor inlet and outlet openings, the shape of the rotor cavity in which the solid-phase charge is located, or the shape of the solid-state embedded rotor assembly, is not limited.
Another feature of the invention is that the hollow rotor can be provided with a feed and discharge tube for the 'solid' phase particles. For example, the tube may be a hollow rotor axis through which the solid phase particles in the rotor cavity can be filled or emptied by pressure or vacuum. For this purpose, the hollow axis of the rotor may be perforated or provided with channels for introducing the solid phase into this space at the locations where the solid phase charge is to be stored. The solid phase particles can also be dosed by means of a tube while the rotor is rotating, so that the solid cavity of the rotor cavity is then filled by 1 centrifugal force and centrifugal pressure.

Between the bottom 4 of the rotor 3 and the lower lid 18, a specific vortex field is formed, containing the bubbles of the incoming gas, which is sucked in by the continuously forming liquid spindle inside the solid phase charge 14.
Na obr. 3 je zobrazeno další možné zařízení dle vynálezu, u kterého distributor 12 plynu (O2) je otočně uložen v prostoru náplně pevné fáze 14 a má tvar duté trubice rozšířené na obou koncích do tvaru lopatky, která je opatřena výstupními otvory pro přiváděný plyn do prostoru, ve kterém je náplň pevné fáze 14.Fig. 3 shows another possible device according to the invention, in which the gas distributor (O2) is rotatably mounted in the solid-phase filling space 14 and has the shape of a hollow tube extended at both ends to a vane shape having gas outlet ports into the space in which the solid phase is 14.
The rotor 3, which rotates below the surface of the liquid phase 15, is provided with a removable lid 6 with outlet openings 17 that are displaced towards the center of the lid 6. Inside the rotor 3 is a cavity with a lower perforated lid 18 filled with bulk solids 14 particles. so that they can whirl in the space of this cavity in the form of a suspension when the rotor 3 is rotated.
Přírodní trubice 19 plynu s regulačním ventilem 8, otáčející se pomocí elektromotoru 13, je vedena ucpávkou 9 skrze dno nádoby 1 a ložiskem 20 skrze perforované víčko 18. Osa 10 otáčení má zároveň funkci plnicí případně vyprazdňovací trubice 11 pro pevnou fázi 14, a to pomocí přetlaku případně vakua z vnější strany nebo pomoci odstředivé síly.The natural gas tube 19 with the control valve 8 rotating by the electric motor 13 is guided through the seal 9 through the bottom of the container 1 and through the bearing 20 through the perforated cap 18. The rotation axis 10 also functions as a filling or discharge tube 11 for the solid phase 14. overpressure or vacuum from outside or by centrifugal force.
Zde vstupuje do dutiny rotoru 3, ve které jsou volně loženy částice pevné fáze 14.When the hollow rotor 3 is rotating, the circulating liquid phase 15 is sucked into the interior of the rotor 3, where the solid phase cartridge 14 is located through the central opening 16 in the bottom 4 of the rotor 3 and fed upward through the perforated periphery of the lower cover 18. enters the cavity of the rotor 3 in which the solid phase particles 14 are loosely loaded.
Lopatky distributoru 12 se buď neotáčejí vůbec, otáčejí se opačným směrem než rotor 3 nebo libovolným směrem, ale odlišnou rychlostí než rotor 3.The blades of the distributor 12 either do not rotate at all, rotate in the opposite direction to the rotor 3 or in any direction, but at a different speed than the rotor 3.
Distributor 12 plynu zajišťuje jednak přívod plynu a jednak působí zde jeho mechanické vzduchové míchadlo kombinované s lopatkovým, které vytváří specifický homogenizační režim uvnitř rotoru a uvnitř náplně pevné fáze, vhodný pro styk všech tří fází, kapalné, pevné i plynné.The gas distributor 12 provides both a gas supply and a mechanical air mixer combined with a vane, which provides a specific homogenization mode inside the rotor and inside the solid phase charge, suitable for contacting all three phases, liquid, solid and gaseous.
Also, due to the location of the outlet openings 17 in the rotor cover 6, which form an annulus but moved towards the center of rotation, a pressure pocket is formed under the cover 6 so that the contact of all three phases in this space takes place under increased hydraulic pressure.
Thus, the gas distributor head 12 acts simultaneously as a mechanical stirrer which is hollow and is supplied with the necessary gas. Its shape can be any.
Osa 10 otáčení, kterou tvoří trubice s distribučními otvory 27, má zároveň funkci plnicí, případně vyprazdňovací trubice 11 pro částice pevné fáze 14.FIG. 4 shows schematically the device according to the invention also in front view, in which both the cylindrical hollow rotor 3 with the interchangeable insert 22 and the gas distributor 12 in the form of a perforated circular plate are provided with a separate pivot axis 10 each with its own adjustable drive by an electric motor 13 The axis of rotation 10, which consists of a tube with distribution openings 27, also functions as a filling or discharge tube 11 for solid phase particles 14.
Vestavba 22 s náplní pevné fáze má tvar válcovité kazety, s průlinčitými nebo perforovanými víky 24, která dosedá na kruhovou vložku 26, umístěnou na dnu 4 rotoruThe solid-phase cartridge 22 is in the form of a cylindrical cassette with punctured or perforated lids 24 that abuts a circular insert 26 located on the rotor bottom 4
3.3.
The installation 22 is divided internally by transverse baffles 23 in the form of an annular ring into horizontal channels through which the liquid phase 15 together with the gas phase bubbles is led from bottom to top.
Rotor 3 je ponořen pod hladinou kapalné fáze 15 a výstupní otvory 17 pro cirkulující kapalinu jsou provedeny ve víku 6 rotoru 3.The rotor 3 is submerged below the surface of the liquid phase 15 and the outlet openings 17 for the circulating liquid are provided in the cover 6 of the rotor 3.
Systém vytváří další možný režim pro zintenzivnění styku všech tří fází, kapalné, pevné a plynné.The system creates another possible mode for intensifying the contact of all three phases, liquid, solid and gaseous.
Obr. 1 až 4 sice znázorňují zařízení, ve kterém dochází ke styku všech tří fází, obdobně však bude pracovat i zařízení dle vynálezu, u kterého dutý rotor nebude obsahovat částice pevné fáze ve formě náplně nebo vestavby (pro styk jen kapalné a plynné fáze).Giant. 1 to 4 illustrate a device in which all three phases are in contact, the device according to the invention will work similarly, in which the hollow rotor will not contain solid phase particles in the form of a pack or a built-in (for liquid-gas contact only).
