Abstract
Fosfolipoproteiny jsou nové komplexy obsahující kyselé fosfolipidy a peptidy nebo · bílkoviny v hmotnostním poměru 1 až 50 : 99 až 50. Peptidy musejí obsahovat nejméně dva aminokyselinové zbytky, z nichž aspoň jeden je tvořen bazickou aminokyselinou. Získávají se smísením kyselých fosfolipidů s peptidy nebo bílkovinami za intenzivního ml- ■ cháni při 10 až 100 °C. Fosfolipoproteiny najdou uplatnění jako nové biologické fak.- tory v lékařství, kosmetice, v potravinářském průmyslu a v zemědělství.Phospholipoproteins are new complexes containing acidic phospholipids and peptides or proteins in a weight ratio of 1 to 50 : 99 to 50. Vynález se týká fosfolipoproteinů, obsahujících kyselé fosfolipidy komplexně vázané na peptidy nebo proteiny a způsobu jejich výroby.The invention relates to phospholipoproteins containing acidic phospholipids complexed with peptides or proteins and a method for their production.
Fosfolipoproteiny jsou nové biologické faktory, k jejichž přípravě lze použít fosfolipidy získané známými izolačními a syntetickými postupy. Pro průmyslovou přípravu jsou vhodné zejména směsi fosfatidových kyselin a bis/diacylglycerol/fosforečných kyselin a jejich lysoderivátů, které se připravují fosforylací směsi parciálních acylglycerolů, například postupem podle čs. autorského osvědčení č. 173 220, čs. autorského osvědčení č. 203 421, čs. autorského osvědčení č. 256 691 a čs. autorského osvědčení č. 259 440.Phospholipoproteins are new biological factors, for the preparation of which phospholipids obtained by known isolation and synthetic procedures can be used. • Komplexy, které vznikají interakcí bílkovin s fosfolipidy, se vyskytují ve všech živých objektech, kde plní řadu funkcí, z nichž nejdůležitější je funkce strukturní /biomembrány/ a transportní /přenášeče látek přes membrány nebo lipoproteiny/. V biologických membránách se komplexy tvoří na fázovém rozhraní lipidové dvojvrstvy s integrálními a periferními proteiny. Z hlediska struktury vzniklých komplexů s integrálními bílkovinami jde o útvary, kde vazba obou složek je zprostředkována především hydrofobními interakcemi mezi uhlovodíkovými řetězci acylových skupin fosfolipidů a lipofilními částmi aminokyselin v polypeptidových molekulách, jako jsou aromatická jádra v tryptofanu, tyrosinu a fenylalaninu a alifatické, popřípadě rozvětvené řetězce leucinu, isoleucinu a valinu. Periferní bílkoviny, které jsou lokalizovány na povrchu membrán jsou na něj vázány polárními vazbami a většinou také vodíkovými vazbami. I tyto vazby jsou relativně slabé, protože periferní bílkoviny lze z povrchu snadno uvolnit, například účinkem zředěných roztoků neutrálních solí. Počet molekul fosfolipidů vázaných na molekulu bílkoviny závisí jak na velikosti molekuly integrálního proteinu, tak i na složení a struktuře jeho povrchu, který je ve stykuslipidovou dvojvrstvou. Fosfolipidy vázané na bílkovinnou molekulu se neustále vyměňují rychlostí, srovnatelnou s rychlostí laterální difúze těchto molekul v prostoru membrány a jejich složení významně ovlivňuje funkci daného komplexu. Lipoproteiny, které jsou nejvýznamnější transportní formou lipidů krevní plazmy jsou, podobně jako biomembrány, tvořeny jádrem /s obsahem triacylglycerolů a esterů cholesterolu/ a dále pláštěm tvořeným fosfolipidovou vrstvou, na které jsou vázány speciální proteinové molekuly, tzv. apoproteiny. Také zde se vazba realizuje především hydrofobními interakcemi mezi hydrofobními řetězci fosfolipidů a hydrofobními postranními řetězci apolipoproteinů /o jejichž struktuře se říká, že je amfifilní/. Hydrofilní postranní řetězce apoproteinů vyčnívají do vodné fáze a umožňují transport lipidů v krevní plazmě. Vazba všech složek v lipoproteinových částicích je poměrně volná, protože při pohybu lipoproteinů v krevní plazmě se jednotlivé molekuly apoproteinů, ale i lipidů, snadno vyměňují za jiné. Existence shora popsaných komplexů fosfolipoproteinů v biomembránách i v lipoproteinech byla odvozena především z fyzikálních vlastností nativních systémů. Pokud je nám známo, takové komplexy nebyly, vzhledem k jejich malé stabilitě, zatím izolovány. Interakce lipidů s peptidy a bílkovinami se významně uplatňují i při některých jiných fyziologických dějích, například při myelinizaci nervových vláken, kdy se uskutečňuje mimo jiné vazba lysofosfatidylcholinu na basický protein myelinu, dále při tvorbě antiatelektaktického lipoproteinů, který vystýlá vnitřní povrch plicních alveolů a při vzniku ionoforů, komplexů lipidů s nižšími peptidy, které umožňují transport iontů přes membrány a podobně. V posledních letech je také věnována pozornost významu interakce lipidů a proteinů v potravinách, kde se vzniku komplexu přisuzuje významná úloha při trávení a při resorpci živin ze zařívacího traktu. V této souvislosti byly především studovány interakce neutrálních /amfoterních/ fosfolipidů /fosfatidylcholinu, lysofosfatidylcholinu/ s některými potravinářskými bílkovinami a jejich frakcemi, zejména s mléčnými a sójovými bílkovinami. Také z těchto prací vyplynulo, že při vzniku komplexů se uplatňují převážně hydrofobní interakce mezi acylovými řetězci fosfolipidů a hydrofobními skupinami bílkoviny a že polární část fosfolipidů zůstává relativně volná. Peripheral proteins that are localized on the surface of membranes are bound to it by polar bonds and mostly also by hydrogen bonds. These bonds are also relatively weak, because peripheral proteins can be easily released from the surface, for example by the effect of dilute neutral salt solutions. The number of phospholipid molecules bound to a protein molecule depends both on the size of the integral protein molecule and on the composition and structure of its surface, which is in contact with the lipid bilayer. Phospholipids bound to a protein molecule are constantly exchanged at a rate comparable to the rate of lateral diffusion of these molecules in the membrane space, and their composition significantly affects the function of the given complex. Lipoproteins, which are the most important transport form of blood plasma lipids, are, like biomembranes, formed by a core /containing triacylglycerols and cholesterol esters/ and a shell formed by a phospholipid layer, to which special protein molecules, the so-called apoproteins, are bound. Here too, the binding is mainly carried out by hydrophobic interactions between the hydrophobic chains of phospholipids and the hydrophobic side chains of apolipoproteins /whose structure is said to be amphiphilic/. The hydrophilic side chains of apoproteins protrude into the aqueous phase and enable the transport of lipids in blood plasma. The binding of all components in lipoprotein particles is relatively loose, because during the movement of lipoproteins in blood plasma, individual molecules of apoproteins, but also lipids, are easily exchanged for others. The existence of the above-described phospholipoprotein complexes in biomembranes and in lipoproteins was derived mainly from the physical properties of native systems. As far as we know, such complexes have not yet been isolated due to their low stability. Interactions of lipids with peptides and proteins are also significantly applied in some other physiological processes, for example in the myelination of nerve fibers, when, among other things, the binding of lysophosphatidylcholine to the myelin basic protein takes place, in the formation of antiatelectatic lipoproteins, which line the inner surface of the pulmonary alveoli, and in the formation of ionophores, complexes of lipids with lower peptides that enable the transport of ions across membranes, etc. In recent years, attention has also been paid to the importance of the interaction of lipids and proteins in foods, where the formation of the complex is attributed to a significant role in digestion and in the resorption of nutrients from the digestive tract. In this context, the interactions of neutral /amphoteric/ phospholipids /phosphatidylcholine, lysophosphatidylcholine/ with some food proteins and their fractions, especially with milk and soy proteins, have been studied. These works also showed that the formation of complexes is mainly due to hydrophobic interactions between the acyl chains of phospholipids and the hydrophobic groups of the protein, and that the polar part of the phospholipids remains relatively free. Po 2 hod. míchání, když původně kyselá směs přešla na neutrální, byly získané fosfolipoproteiny prosety v sítovém mlýnu a použity jako nutriční faktory při výrobě dietetického pečivá a jako emulgátory v emulgovaných tucích. Podle zatím získaných výsledků lze počítat s jejich využitím pro přípravu disperzních enterálních dietních směsí pro aplikaci v medicíně, které se zavádějí jako plnohodnotný doplněk parenterální výživy. V dermatologii se pravděpodobně uplatní především komplexy s albuminy a kolageny jako přísady do krémů a lotionů a jako farmaka pro léčení ran a popálenin. V humánní výživě je lze aplikovat jako aditiva s vysokou nutriční hodnotou do nových typů dietetik a směsi s roborujícím účinkem, zejména pro sportovce, rekonvalescenty, nemocné se zhoubnými nádory jakož i pro fyziologicky stárnoucí populaci.
