The compound of formula I is an essential component for the preparation of its 2-diethylamunoethyl ester, which in the form of the hydrogen oxalate has the generic name naphthidrofuryl. It is currently widely used in the treatment of conditions after cerebral lesions and in geriatrics (Litomericky 5. et al., Farm, horizon 51, 309, 1982). It acts as peripheral and cerebral vasodilators and nootropics.
(Belg. 613, 547 /1962/) a Bull. Soc. Chim. Po oddestilování benzylakoholuf .se získá látka vzorce I destilací ve vakuu při 212.°C / 0,1 kPa.The compound of formula I is still prepared according to Szarvasi E. et al. (Belg. 613, 547 (1962)) and Bull. Soc. Chim. Fr. 1966, 1938; LIPHA Fr. 1,36,3,948 (1964) from diethyl ester of 1- (tetrahydro-2-furyl) -3- (1-n-phenyl) -2,2-propanedicarboxylic acid in benzyl alcohol solution by treatment with potassium hydroxide for eight hours at boiling at 205 ° C followed by acidification. After distilling off the benzyl alcohol f, the compound of formula I is obtained by distillation under vacuum at 212. ° C / 0.1 kPa.
22, 677 (1969), 1- (tetrahydro-2-furyl) -3- (1-naphthyl) -2,2-propanedioic acid diethyl ester was boiled in 15% ethanolic potassium hydroxide solution for 12 hours. After distilling off the ethanol, the residue is dissolved in water, acidified, concentrated and heated at 180 ° C. From the crude product thus obtained, the compound of formula I is isolated by distillation under vacuum at 225 to 226 ° C / 0.4 kPa.
Přípravu látky vzorce I tepelnou dekarboxylací kyseliny l-/tetrahydro-2-furyl/-3-/l-naftyl/-2,2-propandikarboxylové při 140 až 160 °C popisují Valenta V. a spol. (Coli. Czech. Chem. Commun. 52, 2534, 1987/.The preparation of the compound of formula I by thermal decarboxylation of 1- (tetrahydro-2-furyl) -3- (1-naphthyl) -2,2-propanedioic acid at 140-160 ° C is described by Valenta V. et al. (Coli. Czech. Chem. Commun. 52, 2534, 1987).
The original preparation method of Szarvasi et al. requires working with significant amounts of benzyl alcohol at a boiling point of 205 ° C, (its regeneration is heat-intensive or requires vacuum distillation), the reaction time is 8 hours and the isolation of the compound of formula I is carried out by vacuum distillation C. The design is both time-consuming and energy-intensive and requires relatively demanding equipment. Indeed, the Jinjojan-Badaljan method omits the use of benzyl alcohol, but otherwise retains all the disadvantages of the above process. The third method is more advantageous than the previous method. Its disadvantage is the isolation of intermediates.
Uvedené nevýhody odstraňuje způsob příprav 2-/tetrahydro-2-furfuryl/-3-/l-naftyl/ propanové kyseliny vzorce IThe above-mentioned disadvantages are overcome by the process for the preparation of 2- (tetrahydro-2-furfuryl) -3- (1-naphthyl) propanoic acid of the formula I

CS 271 577 Bl ve kterém R značí skupinu methylovou nebo ethylovou, a následující dekarboxylací odpovídajících alkalických solí a/nebo dikarboxylové kyseliny obecného vzorce IIWherein R is methyl or ethyl, followed by decarboxylation of the corresponding alkali salts and / or dicarboxylic acids of formula II
8-θίζ 4COOR1 /II/, ve kterém R1 značí atom sodíku, draslíku nebo vodíku, podle vynálezu, jehož podstata spočívá v tom, Že se látky obecného vzorce II přímo v reakční směsi za varu podrobí dekarboxylaci působením minerální kyseliny a aprotického rozpouštědla.8-θί CO 4 COOR 1 (II), wherein R 1 represents a sodium, potassium or hydrogen atom according to the invention, characterized in that the compounds of the formula II are directly boiled in the reaction mixture by decarboxylation under the action of mineral acid and aprotic solvents.
I. ,The process according to the invention is carried out by subjecting the dilaalkyl ester of the general formula III to a hydrolysis reaction with an aqueous-alkaline solution having a concentration of 5 to 50% by weight of alkali hydroxide, preferably 25% by weight, of potassium hydroxide solution under rapid stirring and reflux. 1 to 5 hours, preferably 2.5 hours, after which an excess of mineral acid to pH 1, preferably hydrochloric acid, and an aprotic solvent, preferably toluene, are added to the reaction mixture, and the reaction mixture is refluxed for 1 to 5 hours. 4 hours, preferably 2.5 hours. The desired acid of formula I is all contained in the organic layer. After draining the lower aqueous acidic layer, this organic acid solution of formula I can further be directly used to prepare the 2-diethylaminoethyl ester and / or other acid ester of formula I.
Způsob podle vynálezu evidentně odstraňuje všechny nevýhody dosavadních postupů, snižuje energetickou a technologickou náročnost, snižuje pracnost, redukuje požadavky na aparaturní vybavení a zásadně zkracuje reakční dobu a celkovou dobu přípravy kyseliny vzorce I. Tyto výhody činí způsob přípravy podle vynálezu neobyčejně výhodný pro provozní technologii a podstatně zlevňuje výrobu. The process according to the invention obviously eliminates all the disadvantages of the prior art processes, reduces energy and technological demands, reduces labor demand, reduces equipment requirements and substantially reduces the reaction time and total preparation time of the acid of formula I. These advantages make the process of the invention extremely advantageous for process technology and significantly reduces production.
The following examples illustrate but do not limit the method according to the invention.
Příklad 1Example 1
Diethylester l-/tetrahydro-2-furyl/-3-/l-naftyl/-2,2-propandikarboxylové kyseliny (vzorce III, kde R značí ethyl) v množství 19,22 g (50 mmol) se při teplotě 80 °C během 20 min připustí do ostře míchaného roztoku hydroxidu draselného, připraveného rozpuštěním 14,0 g (250 mmol) hydroxidu draselného ve 40 ml vody. Tato reakční směs se míchá1- (tetrahydro-2-furyl) -3- (1-naphthyl) -2,2-propanedioic acid diethyl ester (Formula III where R is ethyl) in an amount of 19.22 g (50 mmol) at 80 ° C add 20.0 g (250 mmol) of potassium hydroxide in 40 ml of water over 20 minutes to a sharply stirred solution of potassium hydroxide. The reaction mixture was stirred
After cooling, the non-hydrolyzed fractions were separated by extraction with 3 x 10 ml toluene. The aqueous-alkaline phase was overlaid with 50 ml of toluene and 30 ml (ca. 300 mmol) of concentrated hydrochloric acid was added dropwise with stirring at 60-80 ° C over 15 minutes to a strongly acidic reaction. Thereafter, the temperature of the reaction mixture was raised to reflux for 15 min and the emulsion was stirred for 2.5 h. The aqueous-acid layer was drained and the organic layer was heated to reflux for 15 min. After cooling to 25 ° C, the toluene solution is washed with 3 x 10 ml of water and the acid content of the formula I in the toluene solution is determined by titration. Content: 258 mg / ml. The yield of the desired 2- (tetrahydro-2-furfuryl) -3- (1-naphthyl) -propanoic acid of the formula I is 12.9 g (45.4 mmol) in total, i.e. 91% t. This toluene solution of the acid of formula I It is directly applicable to the synthesis of the 2-diethylaminoethyl ester of the acid of formula I by the phase catalytic transfer method (Cs, АО δ. 268 355). Evaporation of the solvents under reduced pressure (3.3 kPa) from water bath to 80 DEG C. yielded 12.9 g of a viscous residue which crystallized after a long time. The identity of the acid of formula I thus obtained was confirmed by IR and UV spectroscopy, based on 1 H-NMR spectra and by gas and thin layer chromatography.

Takto ве míchá 1,5 h. Fyzikálně-chemické měření (UV, IR a ^H-NMR spektra) potvrdila identitu látky. While stirring, an emulsion of 38.4 g (100 mmol) of diethyl ester, a compound of formula III wherein R is ethyl, in a solution of 28.05 g (500 mmol) of potassium hydroxide in 42 ml of water is heated to reflux for 2 hours. The ethanol released from alkaline hydrolysis is distilled off for 5 hours at a vapor temperature of 78 to 97 ° C, for a total of about 20 ml of distillate. After diluting the slurry with 70 ml of 70 ° C warm water, a solution is formed from which the non-hydrolyzable fractions are extracted at 50 ° C with 3 x 15 ml xylene. 80 ml of xylene are added to the aqueous-alkaline layer and acidified with an excess of 90 ml of 26% hydrochloric acid to pH 1 while stirring. The temperature is exothermic to 60 DEG C. and further heated to reflux for 30 min. The aqueous-acid layer was then drained and extracted with 20 ml and 10 ml of xylene at 50 ° C. The xylene extracts are attached to the organic phase in the reaction vessel. Approximately 15 ml of the turbid azeotrope is distilled off from these combined xylene extracts with stirring at a temperature of 78 to 138 ° C. The solution was stirred at 138 ° C for 15 min. The solution thus obtained can be directly used for further synthesis. Concentration in vacuo afforded a viscous residue having a weight of 29.3 g and a content of 91% acid of the formula I by titration. Thus, the yield of the product is 93.9%. Physico-chemical measurements (UV, IR and 1 H-NMR spectra) confirmed the identity of the substance.
