Abstract

The invention can be used for fruit nurseries and treating soil, particularly apple outplanting. In comparison to existing techniques based on soaking the root system of stecklings or seedlings in a bacterial suspension of the bacterial strain, the amount of rooted plants is increased and their growth improved.

Description

Vynález se Lýká nového kmene produkčního mikroorganismu Agrobacterium rudiobucLur (Beierinck ct van Dcldcn 1902, Conn 1942) s antagonistickým účinkem vůči Fytotoxickým houbám označeným jako M 11. Kmen produkuje antibiotika a této schopnosti lze využít např. k odstranění či zmírnění vzniku jabloňové půdní únavy. Vzorek kultury tohoto kmene je uložen v Československé sbírce kultur mikroorganismů University 3. E. Purkyně v Brně pod číslem CCM 3996.The present invention relates to a novel strain of the production microorganism Agrobacterium rudiobucLur (Beierinckt van Dcldcn 1902, Conn 1942) having an antagonistic effect on the phytotoxic fungi designated M11. The strain produces antibiotics and this ability can be used, e.g. A sample of the culture of this strain is deposited in the Czechoslovak Collection of Cultures of Microorganisms of University 3. E. Purkyně in Brno under the number CCM 3996.

15; 468, 1970; Brown Μ. E. - 3. appl. Bacteriol. 35: 443, 1972; Přikryl Z. et al. - Biologia Plantarum 27: 159, 1985), či nepřímým účinkem, tj. např. antagonistickým účinkem vůči fytopatogenním mikroorganismům ke zlepšení zdravotního stavu rostlin (Cook R. 3. a Rovira A. D. - Soil Biol, Biochem. 8: 275, 1976; Staněk M. - Soil Biol. Biochem. 8: 269, 1976; Staněk M. - Schippers B. a Gamus W. (ed): 247, Academie Press, London 1979).It is known from prior literature that some soil and rhizospheric microorganisms can be used to inoculate plants with the aim of stimulating growth with a direct effect, eg by producing growth substances (Hussain A. et al. - Folia Microbiol. 15; 468, 1970) Brown E.E. - 3rd Appl. Bacteriol. 35: 443, 1972; by Přikryl Z. et al. - Biologia Plantarum 27: 159, 1985), or by an indirect effect, e.g., an antagonistic effect on phytopathogenic microorganisms to improve plant health (Cook R. 3. and Rovira AD - Soil Biol, Biochem. 8: 275, 1976; Stanek M. - Soil Biol. Biochem. 8: 269, 1976; Stanek M. - Schippers B. and Gamus W. (ed): 247, Academic Press, London 1979).

Nyní bylo zjištěno, že některé bakterie izolované s povrchu kořenů jabloní mohou potlačovat rozvoj fytotoxických hub v rizosféře, podílejících se na vzniku půdní únavy. It has now been found that some bacteria isolated from the surface of apple roots may inhibit the development of phytotoxic fungi in the rhizosphere, contributing to the development of soil fatigue.

Na základě selektivního výběru a účinku UV záření byl vyprodukován nový kmen produkčního mikroorganismu Agrobacterium radiobacter s antagonistickým účinkem vůči Pytotoxickým houbám zejména houbám rodu Penicillíum a Alternaria. Uvedený kmen -bakterií byl izolován metodou omyvů kořenů (Harley 3. L. a Waid 3. S. - Trans. Brit. mycol. Soc. 38: 104, 1955).Based on the selective selection and the effect of UV radiation, a new strain of the producing microorganism Agrobacterium radiobacter with an antagonistic effect on Pytotoxic fungi, especially fungi of the genus Penicillia and Alternaria, was produced. The bacterial strain was isolated by the root wash method (Harley, L. L. and Waid, S.S., Trans. Brit. Mycol. Soc. 38: 104, 1955).

g kořenů jabloňových semenáčů bylo třepáno 10 minut ve 100 ml sterilní vodovodní vody. Voda slita a znovu třepáno ve 100 ml vody. Po desetinásobném omytí byly potom 1 mm úseky nařezaných kořínků kladeny na agarové živné médium se streptomycinem (modifikované médium Smith N. R, a Dawson V. T. - Soil Sci. 58: 467, 1944).The g roots of the apple seedlings were shaken for 10 minutes in 100 ml of sterile tap water. Water is poured and shaken again in 100 ml of water. After washing ten times, 1 mm sections of cut roots were then placed on agar nutrient medium with streptomycin (modified Smith N.R. medium and Dawson V.T. - Soil Sci. 58: 467, 1944).

Izolace byla provedena z kolonií bakterií obrůstajících kořenové úseky. Potom následovala jejich selekce a testy ke stanovení antibiotické aktivity vůči fytotoxickým houbám, též vlivu na růst jabloňových semenáčů.Isolation was performed from colonies of bacteria growing in the root sections. This was followed by their selection and tests to determine antibiotic activity against phytotoxic fungi, as well as the effect on the growth of apple seedlings.

Ke kultivaci izolovaných kmenů bakterií byl použit mnsopeptonový agar nebo bujón. Kultivace probíhala při 28 °C na třepačce.Mnsopepton agar or broth was used to cultivate isolated strains of bacteria. Cultivation was carried out at 28 ° C on a shaker.

Morfologie kmene: G- tyčinky vyskytující se nepravidelně, kolonie - kulaté, hladké, lesklé, bez pigmentu, pohyblivost - peritrichální bičíky.Strain morphology: G-sticks occurring irregularly, colonies - round, smooth, shiny, pigment-free, mobility - peritrichal flagella.

Kmen byl identifikován v Čs. sbírce mikroorganismů Univerzity 3. E. Purkyně v Brně.The strain was identified in Cs. collection of microorganisms of the University of E. Purkyně in Brno.

V laboratorních podmínkách metodou titrace antibiotik byla zjištěna až dvojnásobná antibiotické aktivita u bakterie Agrobacterium radiobacter vůči fytotoxické houbě Penicillium a ve skleníkových podmínkách stimulace růstu jabloňových semenáčků.Under laboratory conditions, antibiotic titration has shown up to twice the antibiotic activity of Agrobacterium radiobacter against the phytotoxic fungus Penicillium, and under greenhouse conditions, stimulation of the growth of apple seedlings.

Vynález lze použít v ovocných školkách a k ozdravění půdy jabloňové výsadby (řízkovanci, semenáče).The invention can be used in nurseries and for the recovery of apple plantation soils (cuttings, seedlings).
