Enzymelektrode zur NAD(,P)-BestimmungEnzyme electrode for NAD (, P) determination Anwendungsgebiet der ErfindungField of application of the invention

The invention relates to an enzyme electrode for determining the concentration of Nicotinsäureadenin-dinucleotide (NAD) and Kicotinsäureadenin-dinucleotide phosphate (NADP) (collectively referred to as NAD (P)) and allows the determination of the concentration of substrates lTAD (P) H-dependent dehydrogenases and the dehydrogenase activity. Since there are approximately 200 different dehydrogenases (DH) in vivo, the enzyme electrode is particularly suitable for use in the clinical laboratory. It is also suitable for NAD (P) measurements in microbiology and food processing.

The common method for determining the NAD (P) concentration is the optical test. Ziel der ErfindungObject of the invention

Darlegung des Wesens der ErfindungExplanation of the essence of the invention

The object of the invention is to obtain by the combination of the enzymes glucose oxidase (GOD) and glucose dehydrogenase (GDH) a signal which depends on the NAD (P) concentration. The object is achieved with an enzyme electrode in that the two enzymes are arranged together in a layer in front of a conventional oxygen electrode and the measuring solution is added a defined amount of glucose. In the absence of NAD (P), the glucose oxidation causes a local consumption which is proportional to the amount of glucose added (Principle of the Glucose Electrode (Scheller P. et al., Z. Med.Lab.Diagn. J8, 312 (1977)). When NAD (P) is added to this solution, part of the glucose is converted by the GDH in front of the electrode to gluconic acid, so that a decrease in the signal for glucose occurs, which depends on the NAD (P) amount.

GOD Glukose * O2 Glukonsäure + ^2O2 GOD glucose * O 2 gluconic acid + ^ 2 O 2

GDH Glukose + NAD(P) Glukonsäure + NAD(P)HGDH glucose + NAD (P) gluconic acid + NAD (P) H

The enzyme electrode according to the invention thus operates without the use of a cofactor. It allows both the continuous and the pointwise determination of the NAD (P) concentration with simple devices (pOg-meters). Thus, the determination of NAD (P) consumption, ie e of NAD (P) H formation is possible.

In order to determine substrates of NAD (P) H-dependent DH, it is expedient that the enzyme layer for GOD and GDH is additionally provided with the specific DH for the substrate.

The enzyme electrode prepared in this way is submerged in the stirred measuring solution of 0.1 M phosphate buffer, pH 7.6, and is connected to a pO «meter with recorder.

After the basal current has stopped, an amount of glucose is added so that the concentration in the measurement solution is 0.5 mM. After 2 minutes, the steady state current value is reached and 8 times the addition of 0.25 mM NAD each (uptake) the calibration curve). In each case, a reduction of the glucose signal occurs, whereby after the steep drop immediately after NAD addition, the transition to a constant region occurs. The measured value results as an intersection of the linear extrapolation of both regions. To measure the IDH activity, a solution containing 0.5 mM glucose and 1 mM NAD and 0.1 M lactate is introduced into the measuring cell. After the current has a constant value, 20 μl of the LDH-containing serum sample is added. The enzyme reaction leads to an increase in the current-time curve, since the NAD concentration is reduced. The LDH activity can thus be calculated from the calibration curve.

A photopolymerized enzyme layer containing only GOD is applied to the O 2 electrode as in Example 1.

Finally, a second polyacrylamide layer of 20 / U thickness, containing 30 U GDH and 20 U alcohol dehydrogenase per cm 2 , placed over the GOD layer and fixed with a dialysis tube on the end face of the electrode sheath. This electrode immersed in 5 ml tempered stock containing 0.5 mM glucose, 1 mM EiAD in 0.1 M phosphate buffer pH 7.6. , After the electricity has stabilized; are added 100 / Ul of G A'thanol-containing sample. In the enzyme reaction, an amount of NAD that is equimolar to the concentration of ethanol is consumed and therefore the current is increased. This increase gives the ethanol concentration from the calibration curve recorded under identical conditions.
