Abstract
Gegenstand der Erfindung ist ein Verfahren zur Herstellung von 1,3-Dipherioxypropan-2~on-~derivaten und deren therapeutisch verträglichen Salze, welche in Arzneimitteln gegebenenfalls zusammen mit pharmazeutisch zulässigen Verdünnungs- oder Trägermitteln, insbesondere in einer Einzeldosis von 10 bis 500 mg Wirkstoff, angewandt werden» Die Wirksamkeit der neuen Substanzen beruht auf ihrem starken hypolxpämischen Effekt.
Description
Verfahren zur Herstellung von neuen 1,3-Dipbenoxypropan-
-2~on-derivaten
Anwendungsgebiet der Erfindung:
Die Erfindung betrifft ein Verfahren zur Herstellung von neuen 1,3-Diphenoxypropan~2-on-derivaten.
Charakteristik der bekannten technischen Lösungen^
Der Grundkörper, das 1,3-Diphenoxypropan-2-on, ist 1951 von J. IJunch-Petersen/Acta Chem. ^x 519-528 (1951)/ und später von Mc Baizer / J. Org. Chem. 2jj, 670-671 (1960)/ beschrieben worden. Über symmetrisch gleichartig substituierte Verbindungen, wie z.B. das 1,3~Di-(4'-methylenphenoxy)-propan- -2-on und das 1, 3-Di-(4'-nitrophenoxy)-propan»2-ons berichtete J. Hill/J. Chem. Soc. 1970» 462-464/. Die Herstellung und Verwendung von. symmetrisch mehrfach substituierten 1,3-Di-phenoxyprop£tn-2~onen geht aus den Patentschriften"
US-PS 3 655 893 (Chloronitrophenyl ethers as nematocides) und US-PS 3 666 814 (Ghlornitrophenyl ethers as herbicides) hervor.
Kürzlich wurde über hypolipämisch wirksame,.symmetrisch gleichartig substituierte 1,3-Diphenoxypropanone berichtet, von denen die Verbindungen 1,3-Di-C4'-methylphenoxy)-propan- ' 2-on und 1,3~Di(4t~chlorphenoxy)-propan-2-on die höchste Aktivität besitzen sollen. /C. Piantadosi, I. H. Hall, S. D. V/yrik und K. S. Ishaq, J. Med. Chem. 1£, 222-229 (1976)/.
Wie noch gezeigt wird. (Tab. 3)> haben diese Verbindungen an normal gefütterten, normolopämischen, männlichen, 190 bis 230 g schweren Wistar-Ratten keine hypolipämische Wirkung
Ziel der Erfindung:
Mit der Erfindung sollen verbesserte Pharmazeutika mit starker hypolipämischer Wirkung zur Verfügung gestellt werden.
Darlegung des Wesens der Erfindung;
Die Erfindung beruht auf der überraschenden Erkenntnis, daß ungleich substituierte 1,3~Diphenoxypropan-2-one, dann eine starke hypolipämische Wirkung entfalten, wenn einer der Substituenten eine Carbuxylgruppe darstellt, die gegebenenfalls salzartig oder auch in veresterter Form vorliegen kann.
Der Erfindung liegt die Aufgabe zugrunde, ein Verfahren zur Herstellung von'Verbindungen der allgemeinen Formel I und ihrer therapeutisch verträglichen Salze zu schaffen.
In der Formel I bedeuten
R1: -Cl, -Br,'-CH(CH3)2, -C(CH3K;
R : Wasserstoff, das Kation eines therapeutisch verträglichen Salzes oder ein gerader oder verzweigter, gesättigter oder ungesättigter Alkylrest mit bis zu drei Kohlenstoffatomen, der gegebenenfalls endständig eine Methoxygruppe trägt; · ' .
A: Einfachbindung, -CH = CH-J-CH2-CH2.
Erfindungsgemäß wird diese Aufgabe dadurch gelöst, daß man
a: Verbindungen der allgemeinen Formel (II), in der R , R und A die obige Bedeutung haben, einer Oxidation mit Chromsäure oder Dimethylsulfoxid unterzieht;
bi Verbindungen der allgemeinen Formel (II), in der R und
A die obige Bedeutung haben und R eine Methylgruppe darstellt, nach Oxidation zur Ketoverbindung einer Verseifung unterzieht und gegebenenfalls in die entsprechend physiologisch verträglichen Salze überführt;
c: in Verbindungen der allgemeinen Formel (II), in der R
und A die obige Bedeutung haben und R eine Benzylgruppe darstellt, nach Oxidation zur Ketoverbindung die Benzylgruppe hydrogenolytisch mit Palladium/Kohle abspaltet und die resultierende Carbonsäure gegebenenfalls in ein therapeutisch verträgliches Salz überführt.
Die Verbindungen der allgemeinen Formel (II)lassen sich, wie bereits angedeutet, mit verschiedenen Oxidationsmitteln in die entsprechenden Ketone überführen. Z.B." gelingt die Oxidation in acetonischem Milieu mitr wäßrigen, schwefelsauren Chromtrioxidlösungen (Jones-Reagenz). In guter Ausbeute verläuft die Oxidation auch in wasserfreien, inerten organischen Lösungsmitteln, wie z.B, in Äther mit Dimethylsulfoxid 5 Dicyclohexylcarbodiimid und Trifluoressigsäure unter Zusatz von Pyridin. Ebenso eignen sich Mischungen von

Dimethylsuifoxid und Trifluoressigsäureanhydrid in halogenierten Kohlenwasserstoffen, wie z.B. in Methylenchlorid. Von wesentlicher Bedeutung ist für eine gezielte Oxidation das Einhalten bestimmter Reaktionskriterien, wie sie in den Beispielen näher angegeben werden. . . .
Verbindungen der allgemeinen Formel (II) lassen sich bequem nach Verfahren herstellen, wie sie bereits in der DOS 24 60 689 näher beschrieben wurden. So lassen sich Verbindungen der allgemeinen Formel (III), in der R ein Chloroder Bromatom, bzw. eine Isopropyl» oder tertiäre Butylgruppe darstellt, mit einem Glycidäther der allgemeinen Formel (IV)s in der A eine Einfachbindung, eine Vinylen- oder Äthyl-

engruppe bedeutet und R ein gerader oder verzweigter, gesättigter oder ungesättigter Alkylrest mit 1-3 Kohlenstoffatomen ist, der gegebenenfalls endständig eine-Methoxygruppe trägt, in organisch-protischen Lösungsmitteln, vorzugsweise Alkoholen, die dem Alkylrest entsprechen, in Gegenwart von Basen, vorteilhaft Alkalihydroxiden oder Lewissäuren, beispielsweise Bortrifluoridätherat·, umsetzen.
Ebenso können auch Verbindungen der allgemeinen Formel (V),
in der A und R die in der allgemeinen Formel (IV) angegebene Bedeutung besitzen, mit einem Glycidäther der allgemeinen Formel (VI), in der R die in der allgemeinen Formel (III)'angegebene Bedeutung besitzt, in gleicher V/eise umgesetzt werden.
Die Verbindungen der allgemeinen Formel (I) und ihre Salze werden bevorzugt oral verabreicht. Gewöhnlich beträgt die orale Tagesdosis bei Erwachsenen 0.1 - 10 g, vorzugsweise 0*5 -3g.
Die Wirkstoffe können in üblicher Form zur oralen Verabreichung konfektioniert v/erden, z.B. in Kapseln, in flüssiges." Form, als Tabletten oder als Pulver. Durch Vermischen
mit festen, pulverförmigen Trägerstoffen wie Lactose, Saccharose, Sorbit, Mannit, Kartoffel« oder Maisstärke, Cellulosederivaten oder Gelatine, gegebenenfalls unter Zusatz von Gleitmitteln, wie z.B. Magnesium- oder Calciumstearat oder Polyäthylenglycolen, können sie zu Dregeekernen oder Tabletten verarbeitet werden.
Als weitere Verabreichungsformen eignen sich Steckkapseln, z.B, aus Hartgelatine, sowie geschlossene Weichgelatinekapseln mit einem Weichmacher, wie z.B. Glycerin. Die Steckkapseln enthalten den Wirkstoff vorzugsweise als Granulat, z.B. in Mischung mit,Füllstoffen wie Lactose, Saccharose, Mannit, Stärken, wie z.B„ Kartoffelstärke, Maisstärke oder Amylopectin, Cellulosederivaten, Gelatine oder hochdispersen Kieselsäuren. In weichen Kapseln ist der Wirkstoff vorzugsweise in geeigneten Flüssigkeiten, z.B. in Pflanzenöl oder Polyäthylenglycolen, gelöst oder suspendiert.
Ausführungsbeispiele:
Im folgenden wird die Erfindung anhand von Ausführungsbei- ' spielen näher erläutert.

a) Herstellung; des Ausgangsproduktes
3-(4*-Methoxycarbonylphenoxy)-1-(4'-chlorphenoxy)-propan-2-ol .
Zur Lösung von 2*24 g (0.04 Mol) Kaiiumhydroxid in 300 ml absolutem Methanol v/erden 3o.60g (0.24 Mol) 4'-Hydroxybenzoe· säuremethylester gegeben und die Mischung langsam zum Sieden
erhitzt. Im.Verlaufe von 1 Stde. gibt man 36.92 g (0.2 Mol) 3-(4'-Chlorphenoxy)-1,2-epoxypropan hinzu und erhitzt 21 Stdn. am Rückfluß. Anschließend wird das Lösungsmittel i. Vak. entfernt, der Rückstand in Äther aufgenommen und mit V/asser· bis zur neutralen Reaktion gewaschen. Nach dem Einengen der über Natriumsulfat getrockneten organischen Phase verbleiben ca. 50 g Rohprodukt, welches aus Tetrachlorkohlenstoff umkristallisiert wird

b) Erfindungsgemäße Herstellung; von

mittels Chromsäureoxidation
Die Lösung von 33.67 g (0.1 Mol) 3-(4'-Methoxycarbonylphenoxy)~1-(4'-chlorphenoxy)-propan-2-ol in 350 ml Aceton (über Kaliumpermanganat destilliert) wird unter Eiskühlung mit 100 ml Jones-Reagenz (hergestellt aus 40 g Chromtrioxid, 20 ml konz. Schwefelsäure und 80 ml Wasser) derart versetzt, daß die Reaktionstemperatur nicht über +10 0C ansteigt. An~ schließend wird bei Raumtemperatur solange gerührt, bis im Dünnschichtchromatograram keine Ausgangsverbindung mehr nachzuweisen ist. Nach dem Versetzen mit der doppelten Menge an Wasser wird mehrmals mit Chloroform extrahiert. Die organische Phase wird zuerst mit Natriumbicarbonatlösung und dann mit Wasser gewaschen, schließlich über Magnesiumsulfat getrocknet und i. Vak. eingeengt. Farblose. Kristalle vom Pp. 100 0C (Essigester/Petroläther); Ausbeute 24.45 g (73.0 %).

mittels PIvüSQ-Oxidation ' ' '
Unter Ausschluß von Feuchtigkeit wird die Lösung von 33.67 g (0.1 Mol) 3-(4'-Methoxycarbonylphenoxy)-1-(4'-chlorphenoxy)-propan-2~ol in 500 ml absolutem Äther bei Raumtemperatur mit. 40 ml Dimethy1sulfoxid, 7 ml Pyridin und 30.95 g (0.15 Mol) Bicyclohexylcarbodiimid versetzt. Beim Zutropfen von 5 ml Trifluoressigsäure setzt die Reaktion ein, ersichtlich am Auftreten eines farblosen Niederschlags; nach 4 Stdn. Rühren bei Raumtemperatur ist die Reaktion laut Dünnschichtehromatogramm beendet. Zur Beseitigung von überschüssigem Dicyclorhexylcarbodiimid wird das Reaktionsgemisch protionsweise mit einer Lösung von 30 g Oxalsäure in Methanol versetzt und 20 Min. gerührt. Nach Abtrennen des Niederschlags wird das FiItrat mit einer Natriumhydrogencarbonatlösung und dann mit Wasser gewaschen. Die organische Phase wird über Natriumsulfat getrocknet und i. Vak. eingeengt. Farblose Kristalle vom Fp. 100 0C (Essigester/Petroläther); Ausbeute 26.80 g (80.1 %)«
Im IR-, H-NMR- und Massenspektrum identisch mit dem nach Beispiel 1b) hergestellten Produkt.
3- [ 4' -Z2"-(2-Propinoxycarbonyl) -äthyl/-phenoxy V -1 - (4' bromphenoxy)-propan-2-on
43.35 g (0,1 Mol) 3- ( 4'-/2'-(2-Propinoxycarbonyl)-äthyl7~ phenoxy j ~1-(4'~bromphenoxy)-propan-2-ol v/erden in 500 ml
absolutem Äther gelöst und bei Raumtemperatur mit 40 ml Dime thylsulfoxid, 7 ml Pyridin und 30.95 g (0.15 Mol) Dicyclohexylcarbodiimid versetzt. Nach Zugabe von 5 ml Trifluoressigsaure wird solange gerührt, bis mittels dünnschichtchromatographischer Kontrolle keine Ausgangsverbindung mehr nachzuweisen ist. Zur Beseitigung von überschüssigem Dicyclohexylcarbodiimid wird das Reaktionsgemisch 'portionsweise mit einer Lösung von 30 g Oxalsäure in Methanol versetzt und 20 Min. gerührt. Nach Abtrennen des entstandenen Dicyclohexylharnstoff-Niederschlag.3 wird das Piltrat mit einer Natriumhydrogencarbonatlösung und dann mit Wasser gewaschen, Die · organische Phase wird über Natriumsulfat getrocknet und i. Vak. eingeengt. Das Rohprodukt wird zweimal umkristallisiert. Farblose Kristalle vom Fp. 90-1 0C (Isopropanol); Ausbeute 37.56 g (87.1 %).

Beispiel 3 . '
3-(4'-Carboxyphenoxy)-1-(4f-tert.butylphenoxy)-propan-2-on
a) "Herstellung mittels Chromsäureoxidation . '.
34.44 g (0.1 Mol) 3-(4'-Carboxyphenoxy)-1-(4!-tert.butylphenoxy)-propan-2-ol v/erden in 500 ml Aceton (über Kaliumpermanganat destilliert) gelöst und unter Biskühlung 110 ml Jones-Reagenz /Herstellung siehe Beispiel 1b)_7derart zugetropft, daß die Reaktionstemperatur nicht über +10 0C ansteigt· Anschließend wird weitere 8.Stdn. bei Raumtemperatur gerührt und dann analog Beispiel 1b) aufgearbeitet. Farblose Kristalle vom Pp. I64 0C (Aceton); 11.8 g (34.4 %),

b) Herstellung mittels DMSO-Oxidation
In die Lösung von 14.2 ml (0.2 Mol) wasserfreiem Dimethylsulfoxid in 150 ml Methylenchlorid v/erden bei -70 0C '* ' 20.9 ml (0.15 Mol) Trifluoressigsäureanhydrid in 35 ml Methylenchlorid eingetropft und ca* TO Min. - bis sum Auftreten eines farblosen Niederschlags - gerührt. An-
schließend gibt man eine Lösung von 34.44 g (0.1 Mol) 3_(4·-Carboxyphenoxy)-1 -(4f-tert.butylphenoxy)-propan-2-οΓ in 70 ml Methylenchlorid sowie 13.5 ml Dimethylsulfoxid in > der Weise zu, daß die Temperatur im Reaktionsgemisch nicht über -60 0C ansteigt. Nach 30 Min. werden vorsichtig 40 ml Triäthylamin hinzugefügt und das Reaktionsgemisch nach dem Entfernen des Kältebads auf Raumtemperatur gebracht. Durch Zugabe von V/asser wird das Oxidationsprodukt als farbloser niederschlag erhalten, der sich aus Aceton Umkristallisieren läßt. Farblose Kristalle vom Pp. 164 0C (Aceton); Ausbeute 14.8 g (43.3 %). Im IR-, 1H-KaR- und Massenspektrum identisch mit dem nach -Beispiel 3a) hergestellten Produkt.
3-^'-(2-Carboxyvinyl)-phenox;£7-1 ~(4'-chlorphenoxy)-propan-2-on
In die Lösung von 36.08 g (0.1 Mol) 3-/4 '-(2-IvIe thoxy carbonyl vinyl)-phenoxy_7-1-(4'-chlorphenoxy)-propan-2-on in 1000 ml Methanol tropft man unter Rühren 200 ml wäßrige 2 η Kaliumhydroxidlösung und läßt das Reaktionsgemisch so lange, bei Raumtemperatur stehen, bis dünnschichtchromatographisch keine Ausgangsverbindung mehr nachzuweisen ist. Dann wird i. Vak. eingeengt, der Rückstand mit Wasser aufgenommen und vorsichtig mit 0.5 η Schwefelsäure bis zur schwach sauren Reaktion versetzt. Die ausgefallene Säure v/ird nach dem Waschen über Phosphorpentoxid i. Vak. getrocknet und· anschliessend umkristallisiert. Parblose Kristalle vom Pp. 173-5 0G (Aceton); Ausbeute 10.0 g (28.9 %).

Beispiel 5 -
3-/4 !-(2-Carboxyäthyl)-phenoxyl7-1 -(4·-chlorphenoxy)-propan-2-on
Die Lösung von 44.0 g (0.1 Mol) 3~/4'-(2-Phenylmethoxycarbonyläthyl)-phenoxy l/-1-(4'-chlorphenoxy)~propan-2-on in 1000 ml Aceton wird mit 1 ml Essigsäure versetzt und nach Zugabe von 8 g Palladium/Kohle (10 $ig) bis zur Aufnahme der berechneten Wasserstoffmenge hydriert. Nach dem Abtrennen des Katalysators wird das Piltrat i. Vak. eingeengt, der Rückstand in Chloroform aufgenommen und die Säure durch Zusatz von Petroläther ausgefällt. Farblose Kristalle vom Pp. 114-5'0C (Trichlorethylen)·; Ausbeute 24.00 g (69.4 %).

3-/4t-(2~Isopropoxycarbonyläthyl)-phenox;v_7.-1-(4l-tert. butylphenoxy)-propan-2-on enthaltendes Arzneimittel
250 g 3-/4'-(2-Isopropoxycarbonylethy1)-phenoxy/-1-(4'-tert.butylphenoxy)-propan-2-on werden mit 250 g PoIyäthylenglycol vermischt und nach dem Schererverfahren in eintausend Weichgelatinekapseln abgefüllt, die je 250 mg Wirkstoff enthalten. .
3- ( 4'-Methoxymethoxycarbonylphenoxy)-1-(4 *-chlorphenoxy)-propan-2-on enthaltendes Arzneimittel
100 g 3-(4'-Methoxymethoxycarbonylphenoxy)-1-(4'-chlorphenoxy)-propan-2-on werden mit 16 g Maisstärke und 6 g hochdispersem Siliciumdioxid gut vermischt, anschließend mit einer Lösung von 2 g Stearinsäure, 6 g Acetylcellulose und 6 g Stearin in 70 ml Isopropanol befeuchtet und granuliert« Das getrocknete Granulat wird durch ein Sieb geschlagen und nach dem Vermischen mit 16 g Maisstärke, 16 g Talk und 2 g Magnesiums tearat zu eintausend Drageekernen verpreßt. Nach Überziehen mit einem Sirup von 2 g Lacca, 7«5 g Gummi arabicum, O.15 g Farbstoff, 2 g kolloidalem Siliciumdioxids 25 g Talk und 53«35 g Saccharose werden nach dem Trocknen eintausend 260 mg schwere Dragees mit .je 100 mg Wirkstoff erhalten.
Beispiel 8 ·
3-(4'-Carboxyphenox-y)-1-(4'-tert. butyl phenoxy )~propan-2-on enthaltendes Arzneimittel
Eine fein gepulverte Mischung von 250 g 3-(4'-Carboxyphen- »oxy)-1-(4'-tert.bu1;ylphenoxy)-propan-2-6n, 133 g Maisstärke, 12g Magnesiumstearat und 5 g Gelatine wird durch ein feinmaschiges Sieb geschlagen und anschließend in eintausend Hartgelatinekapseln trocken eingefüllt, die je 250 mg • Wirkstoff enthalten,
Beispiel 9 Pharmakologische Testung
1. Orale Verträglichkeit
Nach oraler Applikation der Testverbindungen an Mäusen vom Stamm MvIR-I, mit einem Gewicht von 15-2Og, wurde die akute Toxizitat bestimmt. Die LD(-0-Werte wurden nach Litchfield-Wilcoxon /J. Pharmacol. Ex. Ther. 99 (194927 berechnet und beziehen sich auf den achten Tag der Behandlung. Der LD^-Wert für CLOPIBRAT /2-(4'-Chlorphenoxy)-2-methylpropionsäureäthylester7betrug in dieser Versuchsreihe ca. 1900 mg/kg. Die erfindungsgemäßen, getesteten Substanzen waren durchweg verträglicher und dem CLOPIBRAT überlegen.
2. Lipidsenkende Y/irkunp;
Die lipidsenkende Wirkung wurde an Gruppen von jeweils 10 normal gefütterten ("ssniff"-Alleinfutter), normolipämisehen männlichen 190-230 g schweren Wistar-Ratten ' (Ivanovas-Kisslegg) geprüft. Die Testverbindungen wurden in einer wäßrigen Lösung von 0.25 % Agar und 0.85 % HaCl aufgenommen und oral verabreicht* Nach Applikation von
4 x 100' mg/kg über einen Zeitraum von drei Tagen wurden die Tiere im Anschluß an einen vierstündigen Putterentzug durch Herzpunktion entblutet /R. Zschocke, H. Enomoto, R. Löser und G. Hofrichter, Proc. 25, Coil. Protides Biol. Fluids, Editor H. Peeters. .
Die Bestimmung der Serumlipide wurde mit einem Technicon Autoanalyzer durchgeführt. Das Gesamtcholesterin (TC) wurde mit dem enzymatischen Färbtest /R, Röschlau et al., U. Klin. Chem. u. Klin. Biochem. 12, 403 (197427 erfaßt. Die quantitative Analyse der Triglyceride (TG) erfolgte nach Eggstein und Kreutz /Klin. Wschr. ^y1, (196627smodifiziert für den Autoanalyzer. Die lipidsenkende Wirkung ist ausgedrückt als die prozentuale Senkung des Gesamtcholesterins und der Triglyceride gegenüber der Kontrolle. Sämtliche getesteten, erfindungsgemäßen Verbindungen waren mindestens in einem Wert dem CLOPIBRAT überlegen.

der R und A die obige Bedeutung besitzen und R eine Benzylgruppe darstellt, nach Oxidation zur KetoVerbindung die Benzylgruppe hydrogenolytisch mit Palladium/Kohle abspaltet und die resultierende Carbonsäure gegebenenfalls in. ein therapeutisch verträgliches Salz überführt.
