Abstract
Die Erfindung betrifft ein Verfahren zur Herstellung des 11a-Chlor-5-hydroxytetracyclin-6,12-hemiketals durch Chlorieren von 5-Hydroxytetracyclin. Ziel der Erfindung ist ein Verfahren in technologisch einfach auszufuehrender Arbeitsweise und unter Verwendung eines kostenguenstigeren Chlorierungsmittels als N-Chlorsuccinimid. Es wurden gefunden, dass die Alkalisalze von N-Monochlor-arylsufonamiden exzellente Chlorierungsmittel zur Chlorierung des 5-Hydroxy-tetracyclins in der 11a-Stellung unter Bildung des 6,12-Hemiketals in der Enolform sind.The invention relates to a process for preparing the 11a-chloro-5-hydroxytetracycline-6,12-hemiketal by chlorinating 5-hydroxytetracycline. Für die Herstellung des Ha-Chlor-S-hydroxy-tetracyclin-e^-hemiketals wurden bereits mehrere Verfahren vorgeschlagen (U.S. P. 2984686, DE-OS 2164840, DD-AP 113527). Bei allen vorgeschlagenen Verfahren wird 5-Hydroxy-tetracyclin in einem reaktionsinerten mit Wasser mischbaren Lösungsmittel chloriert, wobei folgenede Chlorierungsmittel verwendbar sein sollen: Chlor, N-Chlor-niedrigalkansäureamide, Chlordicarbonsäureimide, N-Niedrigalkanoylanilide, Chlordimethylhydantoin, Pyridinperchlorld-hydrochlorid und niedrigere Alkylhypochlorite. Nach dem Verfahren des U.S. P. 2984686, Chlorierung des 5-Hydroxytetracyclins als Base bei Temperaturen von 0 bis 500C, wurden nur relativ niedrige Ausbeuten (~40% d. Th.)-erhalten, da ein Teil des.Chlorierungsproduktes in der wenig stabilen Ketoform anfällt. Das Verfahren der DD-AP 113527 chloriert dagegen bei noch tieferer Temperatur (-400C) und stärkerer Pufferung des Reaktionsgemisches mit Triethylamin auf pH = 6,5 bis 7,7, besonders 7,3 bis 7,5 und fällt das Reaktionsprodukt aus der Reaktionsmischung als Salz der Enolform mit Mineralsäure in ~78%d. Die Reaktion verläuft damit unter sehr müden Bedingungen in sehr kurzer Reaktionszeit nur schwach exotherm und nahezu quantitativ zu einer Lösung des 11a-Chlor-5-hydrosxytetracyclin-6,12-hemiketals. Der Vorteil der Verwendung der N-Chlor-arylsulfonsäureamidsalze in dieser Reaktion äußerte sich auch darin, daß lediglich ein einmaliges Abkühlen der Lösung bzw. Suspension des 5-Hydroxytetracyclins in z.B. wasserhaltigem Aceton auf -10...-400C erforderlich ist und bei der Zugabe des Chlorierungsmittels nicht weiter gekühlt werden mußte. Dabei stieg die Temperatur dann auf-15 ..+50C an. Interessant waren die Ergebnisse der pH-Messung im Reaktionsgemisch. Bei Verwendung der für die Erzielung guter Ausbeuten erforderlichen Menge an N-Monochlor-p-toluolsulfonamidnatrium, von 1,1 bis 1,3 Mol pro Mol 5-Hydroxyteträcyclin ergab sich ein pH-Wert von 5,1 bis 6,3. Die pH-Messung bei Temperaturen unter -100C stellt ein noch nicht befriedigend gelöstes Meßproblem dar. Die Isolierung des Produktes kann auf übliche Art und Weise erfolgen, z. B. durch Fällung mit Salzsäure als Hydrochlorid, wobei nicht wasserfrei gearbeitet werden muß. Dabei fällt ein Teil des aus dem Chloramin T stammenden Natriums als Natriumchlorid aus und verbessert entscheidend die Filtrierbarkeit des Produktes. Bei der Weiterverarbeitung, z. B. durch die bekannte Dehydratisierung mit Fluorwasserstoff zum Ha-Chlor-metacyclin stört das Natriumchlorid nicht, es werden Ausbeuten an Ha-Chlor-metacyclin von >95% d.,Th. erhalten. ·A decisive advance over the state of the art also exists in this! that no measurement of the pH value is necessary.
